(従来法12)の方法は、ヒドロキシアリール化合物のオルト位に選択的にホルミル化できる方法であるが、置換基を持つ目的物を得る場合、1段階の反応で達成できず、置換基の保護、ホルミル化反応、置換基の脱保護と3段階の反応が必要となり、工業的な見地から有利な方法と言い難い。さらに、この方法は、置換基がハロゲンであるヒドロキシアリール化合物や、ベンジル部位を有するヒドロキシアリール化合物については、リチオ化反応が制御できず副生成物が生じる問題がある。

R2が水素原子、フッ素原子、塩素原子、臭素原子、ヨウ素原子、メチル基、エチル基、i-プロピル基、t-ブチル基、トリフルオロメチル基又はペンタフルオロエチル基であり、R3及びR4がそれぞれ独立に水素原子、フッ素原子、塩素原子、臭素原子、ヨウ素原子、メチル基、エチル基、i-プロピル基、t-ブチル基、トリフルオロメチル基、メトキシ基、エトキシ基、i-プロポキシ基又はt-ブトキシ基である〔1〕記載の製造方法。
R2とR3とが一緒になって-CR5=CR6-CR7=CR8-であり、R5及びR7がそれぞれ独立に水素原子、フッ素原子、塩素原子、臭素原子、ヨウ素原子、メチル基、エチル基、i-プロピル基、t-ブチル基、トリフルオロメチル基、メトキシ基、エトキシ基、i-プロポキシ基又はt-ブトキシ基であり、R6が水素原子、フッ素原子、塩素原子、臭素原子、ヨウ素原子、メチル基、エチル基、i-プロピル基、t-ブチル基、トリフルオロメチル基又はペンタフルオロエチル基であり、R8が水素原子又はフッ素原子である〔1〕に記載の製造方法。

前記式(i)中の好ましいR11は、水素原子、フッ素原子、塩素原子、臭素原子、ヨウ素原子、メチル基、エチル基、n-プロピル基、i-プロピル基、n-ブチル基、トリフルオロメチル基、ペンタフルオロエチル基である。
前記式(i)中のより好ましいR11は、水素原子、フッ素原子、メチル基及びトリフルオロメチル基である。
前記式(a)中のR9は、C1-3アルキル基又は、同じ窒素原子上の2つのR9が一緒になって=CH-R10である。
前記式(a)中のR9を具体的に説明する。該C1-3アルキル基は、メチル基、エチル基、n-プロピル基、i-プロピル基等が挙げられる。
前記式(a)中の好ましいR9は、メチル基、エチル基である。
前記式(a)中の同じ窒素原子上の2つのR9が一緒になって=CH-R10である場合のより好ましいR10は、フェニル基、2-フルオロフェニル基、2-メチルフェニル基、2-メトキシフェニル基、2-i-プロポキシフェニル基である。
前記式(a)で表される好ましいアミン化合物は、N,N,N’,N’-テトラメチルエチレンジアミン、N,N,N’,N’’,N’’-ペンタメチルジエチレントリアミンであり、より好ましいアミン化合物は、N,N,N’,N’-テトラメチルエチレンジアミンである。
2’-(フェニルメトキシ)-1,1’-ビフェニル-2-オール(5)の合成は、Organic & Biomolecular Chemistry (2006年),4(19)巻,3639‐3647.記載の方法に従って製造できる。合成法を(反応式3)にて示す。
実施例1
2-ヒドロキシ-2’-(フェニルメトキシ)-1,1’-ビフェニル-3-カルバルデヒド(9)の合成法と(反応式5)を下記に示す。
次にアミン化合物であるN,N,N’,N’-テトラメチルエチレンジアミン(0.84g,7.2mmol)を加え、テトラヒドロフラン(1.1mL)に溶解させたヒドロキシアリール化合物(5)(1.0g,3.6mmol)の溶液を、反応溶液中に加えた。反応温度を64~66℃にて、21時間撹拌し反応を行った。反応終了後は、15~20℃に冷却し、2.5mol/Lに調製した希釈塩酸を用いて反応をクエンチして、有機層を分液し、テトラヒドロフランにて再抽出した有機層と合わせ、溶媒留去し粗生成物を得た。次にカラムクロマトグラフィーを用いて精製し、目的化合物の2-ヒドロキシアリールアルデヒド化合物(9)(0.88g,収率80%,化学純度99.5%)を淡黄色オイル状で得た。

2-ヒドロキシ-2’-メトキシ-1,1’-ビフェニル-3-カルバルデヒド(10)の合成法と(反応式6)を下記に示す。
次にアミン化合物であるN,N,N’,N’-テトラメチルエチレンジアミン(3.5g,30mmol)を加え、テトラヒドロフラン(3.4mL)に溶解させたヒドロキシアリール化合物(8)(3.0g,15mmol)の溶液を、反応溶液中に加えた。反応温度を64~66℃にて、20時間撹拌し反応を行った。反応終了後は、15~20℃に冷却し、2.5mol/Lに調製した希釈塩酸を用いて反応をクエンチして、有機層を分液し、テトラヒドロフランにて再抽出した有機層と合わせ、溶媒留去し粗生成物を得た。次にカラムクロマトグラフィーを用いて精製し、目的化合物の2-ヒドロキシアリールアルデヒド化合物(10)(2.88g,収率84%,化学純度97%)を淡黄色結晶で得た。

撹拌機器と還流管を備えたフラスコを窒素置換した後、塩化マグネシウム(52g,0.55mol)を、パラホルムアルデヒド(25g,0.82mol)を、テトラヒドロフラン(495mL)を加え、63~66℃にて撹拌した。
次にアミン化合物であるN,N,N’,N’-テトラメチルエチレンジアミン(64g,0.55mol)を加え、テトラヒドロフラン(93mL)に溶解させたヒドロキシアリール化合物(8)(55g,0.27mol)の溶液を、反応溶液中に加えた。反応温度を64~66℃にて、12時間撹拌し反応を行った。反応液の一部をサンプリングして、HPLC分析により、目的生成物の2-ヒドロキシアリールアルデヒド化合物(10)の類縁分析を行い、転化率を求めた。下記の表1に、結果を示す。

2-ヒドロキシ-2’-メトキシ-1,1’-ビフェニル-3-カルバルデヒド(10)の合成法
実施例3で使用したN,N,N’,N’-テトラメチルエチレンジアミンをトリエチルアミンに切り替えて行った比較例1について示す。
撹拌機器と還流管を備えたフラスコを窒素置換した後、塩化マグネシウム(3.8g,40mmol)を、パラホルムアルデヒド(1.8g,60mmol)を、テトラヒドロフラン(40mL)を加え、63~66℃にて撹拌した。
次にアミン化合物であるトリエチルアミン(4.0g,40mmol)を加え、テトラヒドロフラン(5mL)に溶解させたヒドロキシアリール化合物(8)(4.0g,20mmol)の溶液を、反応溶液中に加えた。反応温度を64~66℃にて、5時間撹拌し反応を行った。反応液の一部をサンプリングして、HPLC分析により、目的生成物の2-ヒドロキシアリールアルデヒド化合物(10)の類縁分析を行い、転化率を求めた。下記の表2に、結果を示す。

二核の2-ヒドロキシアリールアルデヒド化合物である3,3’-(1-メチルエチリデン)ビス(6-ヒドロキシベンズアルデヒド)(12)の合成法を(反応式7)に示す。
次にアミン化合物であるN,N,N’,N’-テトラメチルエチレンジアミン(3.1g,26mmol)を加え、市販品のヒドロキシアリール化合物である4,4’-(1-メチルエチリデン)ビスフェノール(11)(1.5g,6.6mmol)を、反応溶液中に加えた。反応温度を78~81℃にて、18時間撹拌し反応を行った。反応液の一部をサンプリングして、HPLC分析により転化率は、78%であった。反応終了後は、15~20℃に冷却し、2mol/Lに調製した希釈塩酸を用いて反応をクエンチした後、ジクロロメタンを加えて有機層を分液した。水層をジクロロメタンで再抽出した有機層と合わせ、溶媒留去し粗生成物を得た。次にカラムクロマトグラフィーを用いて精製し、目的化合物の二核の2-ヒドロキシアリールアルデヒド化合物(12)(1.4g,収率73%)を微黄透明オイル状で得た。この得られた化合物は数時間後に微黄白色固体となった。

2-ヒドロキシ-2’-メトキシ-1,1’-ビフェニル-3-カルバルデヒド(10)の合成法と(反応式8)を下記に示す。
次に市販のアミン化合物であるN,N,N’,N’’,N’’-ペンタメチルジエチレントリアミン(2.3g,20mmol)を加え、テトラヒドロフラン(2g)に溶解させたヒドロキシアリール化合物(8)(2.0g,10mmol)の溶液を、反応溶液中に加えた。反応温度を64~66℃にて、30時間撹拌して反応を行った。反応溶液の一部をサンプリングして、HPLC分析により転化率は、85%であった。

2-ヒドロキシ-2’-(9-アントリルメトキシ)-1,1’-ビフェニル-3-カルバルデヒド(23)の合成について、下記の(反応式9)に記載する。「TMEDA」は、N,N,N’,N’-テトラメチルエチレンジアミンを意味する。
2’-(9-アントリルメトキシ)-1,1’-ビフェニル-2-オール(22)の合成:
2,2-ビフェノール(4.10g、22mmol)、炭酸カリウム(3.04g、22mmol)、ヨウ化カリウム(3.66g、22mmol)及びアセトン26mLを、反応容器に投入し、撹拌しながら、オイルバスを60℃まで加熱した。この懸濁液に9-クロロメチルアントラセン(21)(5.01g、22mmol)を投入し、60℃で20時間撹拌した。HPLCで反応転化率を確認した後、飽和塩化アンモニウム水溶液を添加して反応を停止させた。分液後、水層を酢酸エチルで抽出し、合わせて濃縮することで、粗生成物を得た。この粗生成物をシリカゲルカラムクロマトグラフィーで精製しヒドロキシアリール化合物(22)(3.01g)を白色結晶として、収率36.3%で得た。

パラホルムアルデヒド(0.72g、31mmol)、無水塩化マグネシウム(1.5g、31mmol)、N,N,N’,N’-テトラメチルエチレンジアミン(2.4mL、16mmol)及びアセトニトリル(27mL)を反応容器に投入し、温度を85℃に保った。この混合溶液に出発原料(22)(3.0g、8mmol)を投入し、アセトニトリル(3.0g)で洗いこんだ。85℃で17時間撹拌した後、2mol/Lの塩酸を添加して、反応を停止させた。中和後、アセトニトリルを留去した水層を酢酸エチルで抽出し、有機層を水(100mL)で3回、洗浄した後、酢酸エチルを留去することで、淡褐色の結晶を得た。この粗生成物の結晶をイソプロピルアルコールで懸濁洗浄することで、淡黄色粉末としてヒドロキシアリールアルデヒド(23)(1.86g)を、収率57.5%、HPLC相対面積百分率89.2%で得た。

2,2-ビフェノール(3.72g、20mmol)、炭酸カリウム(1.38g、10mmol)、ヨウ化カリウム(3.72g、20mmol)及びアセトン23.5mLを、反応容器に投入し、撹拌しながら、オイルバスを60℃まで加熱した。この懸濁液にp-メチルベンジルクロリド(31)(2.81g、20mmol)を滴下し、60℃で17時間撹拌した。HPLCで反応転化率を確認した後、飽和塩化アンモニウム水溶液を添加して反応を停止させた。分液後、水層を酢酸エチルで抽出し、合わせて濃縮することで、粗生成物を得た。シリカゲルカラムクロマトグラフィー(ヘキサン/酢酸エチル=9/1、Rf値=0.25)で精製し、ヒドロキシアリール化合物(32)(4.30g)を白色結晶として、収率74.0%で得た。

パラホルムアルデヒド(0.93g、31mmol)、無水塩化マグネシウム(2.0g、31mmol)、N,N,N’,N’-テトラメチルエチレンジアミン(TMEDA)(3.1mL、21mmol)及びテトラヒドロフラン(30mL)を反応容器に投入し、温度を65℃に保った。この混合溶液に出発原料(32)(3.0g、10mmol)を投入し、テトラヒドロフラン(3.0g)で洗いこんだ。65℃で17時間撹拌した後、2mol/Lの塩酸を添加して、反応を停止させた。分液後、テトラヒドロフランで水層を抽出し、有機層を合わせて濃縮した。濃縮物を酢酸エチル(200mL)に溶解させ、水(100mL)で3回、洗浄した後、酢酸エチルを留去することで、淡褐色の結晶を得た。この結晶をイソプロピルアルコールで懸濁洗浄することで、白色粉末としてヒドロキシアリールアルデヒド(33)(2.21g)を、収率67.4%で得た。

2’-メシチルメトキシ-1,1’-ビフェニル-2-オール(42)の合成:
2,2-ビフェノール(3.72g、20mmol)、炭酸カリウム(2.76g、20mmol)、ヨウ化カリウム(3.32g、20mmol)及びアセトン24mLを、反応容器に投入し、撹拌しながら、オイルバスを60℃まで加熱した。この懸濁液に2,4,6-トリメチルベンジルクロリド(41)(3.37g、20mmol)を投入し、60℃で17時間撹拌した。HPLCで反応転化率を確認した後、飽和塩化アンモニウム水溶液を添加して反応を停止させた。分液後、水層を酢酸エチルで抽出し、合わせて濃縮することで、粗生成物を得た。この粗生成物を2-プロパノールで再結晶することで、ヒドロキシアリール化合物(42)(5.93g)を白色結晶として、収率93.1%で得た。

パラホルムアルデヒド(0.90g、30mmol)、無水塩化マグネシウム(1.9g、20mmol)、N,N,N’,N’-テトラメチルエチレンジアミン(TMEDA)(3.0mL、20mmol)及びテトラヒドロフラン(32mL)を反応容器に投入し、温度を65℃に保った。この混合溶液に出発原料(42)(3.2g、10mmol)を投入し、テトラヒドロフラン(3.2g)で洗いこんだ。70℃で17時間撹拌した後、2mol/Lの塩酸を添加して、反応を停止させた。中和後、アセトニトリルを留去した水層を酢酸エチルで抽出し、有機層を水(100mL)で3回、洗浄した後、酢酸エチルを留去することで、粗生成物を得た。この粗生成物をイソプロピルアルコールで懸濁洗浄することで、白色結晶としてヒドロキシアリールアルデヒド(43)(1.57g)を、収率45.4%、HPLC相対面積百分率99.2%で得た。

2,2-ビフェノール(3.7g、20mmol)、炭酸カリウム(2.8g、20mmol)及びアセトン24mLを、反応容器に投入し、撹拌しながら、オイルバスを60℃まで加熱した。この懸濁液に2-(ブロモメチル)ナフタレン(51)(4.42g、20mmol)を投入し、60℃で17時間撹拌した。HPLCで反応転化率を確認した後、飽和塩化アンモニウム水溶液を添加して反応を停止させた。分液後、水層を酢酸エチルで抽出し、合わせて濃縮することで、粗生成物を得た。この粗生成物を2-プロパノールで再結晶することで、ヒドロキシアリール化合物(52)(4.75g)を白色結晶として、収率72.8%で得た。

パラホルムアルデヒド(0.90g、30mmol)、無水塩化マグネシウム(1.9g、20mmol)、N,N,N’,N’-テトラメチルエチレンジアミン(TMEDA)(3.0mL、20mmol)及びテトラヒドロフラン(33mL)を反応容器に投入し、温度を65℃に保った。この混合溶液に出発原料(52)(3.26g、10.0mmol)を投入し、テトラヒドロフラン(3.3g)で洗いこんだ。70℃で15時間撹拌した後、2mol/Lの塩酸を添加して、反応を停止させた。中和後、アセトニトリルを留去した水層を酢酸エチルで抽出し、有機層を水(100mL)で3回、洗浄した後、酢酸エチルを留去することで、粗生成物を得た。この粗生成物をイソプロピルアルコールで懸濁洗浄することで、白色結晶としてヒドロキシアリールアルデヒド(53)(2.63g)を、収率74.1%、HPLC相対面積百分率99.0%で得た。

2,2-ビフェノール(3.72g、20mmol)、炭酸カリウム(2.76g、20mmol)、ヨウ化カリウム(3.32g、20mmol)及びアセトン24mLを、反応容器に投入し、撹拌しながら、オイルバスを60℃まで加熱した。この懸濁液に4-ブロモベンジルブロミド(61)(4.99g、20mmol)を投入し、60℃で17時間撹拌した。HPLCで反応転化率を確認した後、飽和塩化アンモニウム水溶液を添加して反応を停止させた。分液後、水層を酢酸エチルで抽出し、合わせて濃縮することで、粗生成物を得た。この粗生成物を2-プロパノールで再結晶することで、ヒドロキシアリール化合物(62)(4.78g)を白色結晶として、収率67.3%で得た。

パラホルムアルデヒド(0.98g、33mmol)、無水塩化マグネシウム(2.1g、22mmol)、N,N,N’,N’-テトラメチルエチレンジアミン(TMEDA)(3.3mL、22mmol)及びテトラヒドロフラン(36mL)を反応容器に投入し、温度を65℃に保った。この混合溶液に出発原料(62)(3.2g、10mmol)を投入し、テトラヒドロフラン(3.6g)で洗いこんだ。70℃で15時間撹拌した後、2mol/Lの塩酸を添加して、反応を停止させた。中和後、アセトニトリルを留去した水層を酢酸エチルで抽出し、有機層を水(100mL)で3回、洗浄した後、酢酸エチルを留去することで、粗生成物の結晶を得た。この粗生成物をイソプロピルアルコールで懸濁洗浄することで、白色結晶としてヒドロキシアリールアルデヒド(63)(2.64g)を、収率63.3%、HPLC相対面積百分率98.9%で得た。

2,2-ビフェノール(3.7g、20mmol)、炭酸カリウム(2.8g、20mmol)及びアセトン24mLを、反応容器に投入し、撹拌しながら、オイルバスを60℃まで加熱した。この懸濁液に2-メトキシベンジルクロリド(71)(3.13g、20mmol)を投入し、60℃で17時間撹拌した。HPLCで反応転化率を確認した後、飽和塩化アンモニウム水溶液を添加して反応を停止させた。分液後、水層を酢酸エチルで抽出し、合わせて濃縮することで、粗生成物を得た。この粗生成物を2-プロパノールで再結晶することで、ヒドロキシアリール化合物(72)(5.58g)を白色結晶として、収率91.0%で得た。

パラホルムアルデヒド(0.90g、30mmol)、無水塩化マグネシウム(1.9g、20mmol)、N,N,N’,N’-テトラメチルエチレンジアミン(TMEDA)(3.0mL、20mmol)及びテトラヒドロフラン(31mL)を反応容器に投入し、温度を65℃に保った。この混合溶液に出発原料(72)(3.26g、10.0mmol)を投入し、テトラヒドロフラン(3.1g)で洗いこんだ。70℃で15時間撹拌した後、2mol/Lの塩酸を添加して、反応を停止させた。中和後、アセトニトリルを留去した水層を酢酸エチルで抽出し、有機層を水(100mL)で3回、洗浄した後、酢酸エチルを留去することで、粗生成物の結晶を得た。この粗生成物をイソプロピルアルコールで懸濁洗浄することで、白色結晶としてヒドロキシアリールアルデヒド(73)(2.08g)を、収率62.1%、HPLC相対面積百分率99.2%で得た。

2’-(2-フルオロベンジルオキシ)-1,1’-ビフェニル-2-オール(82)の合成:
2,2-ビフェノール(24.21g、130mmol)、炭酸カリウム(8.98g、65mmol)、ヨウ化カリウム(4.32g、26mmol)及びアセトン120gを、反応容器に仕込み、20~25℃にて撹拌した。この反応溶液に2-フルオロベンジルブロミド(81)(24.57g、130mmol)を仕込んだ後、反応温度60℃にて23時間撹拌した。20~25℃に冷却して、飽和塩化アンモニウム水溶液を反応溶液に添加して反応を停止し、分液した後、水層をクロロホルムで抽出して、有機層を合わせて濃縮後、シリカゲルカラムクロマトグラフィー(ヘキサン:酢酸エチル=80:20)で精製することで、ヒドロキシアリール化合物(82)(26.55g)を淡茶色結晶として、収率69.4%で得た。

2-ヒドロキシ-2’-(2-フルオロベンジルオキシ)-1,1’-ビフェニル-3-カルバルデヒド(83)の合成:
撹拌機器と還流管を備えたフラスコを窒素置換した後、パラホルムアルデヒド(4.62g、154mmol)、無水塩化マグネシウム(10.2g、107mmol)、出発原料(82)(15.1g、51.2mmol)及びアセトニトリル(100mL)を加え、20~25℃にて撹拌した。この反応溶液にN,N,N’,N’-テトラメチルエチレンジアミン(TMEDA)(11.84g、102mmol)を滴下した。この反応溶液を反応温度75~80℃にて、16時間撹拌した後、15~20℃に冷却して2mol/Lの塩酸を添加して、反応を停止した。反応溶液を中和した後、アセトニトリルを留去した水層を酢酸エチルで抽出し、有機層を合わせた後、溶媒を留去して粗生成物を得た。この粗生成物をクロロホルム-ヘキサン-酢酸エチルの系にて再結晶化することで、淡白黄色結晶としてヒドロキシアリールアルデヒド(83)(10.3g)を、収率62.7%で得た。

2,2-ビフェノール(18.62g、100mmol)、炭酸カリウム(6.91g、50.0mmol)、ヨウ化カリウム(3.32g、20.0mmol)及びアセトン90gを、反応容器に仕込み、20~25℃にて撹拌した。この反応溶液に2-フェニルベンジルブロミド(91)(24.71g、100mmol)を仕込んだ後、反応温度55℃にて27時間撹拌した。20~25℃に冷却して、飽和塩化アンモニウム水溶液を反応溶液に添加して反応を停止し、分液した後、水層をクロロホルムで抽出し、有機層を合わせて濃縮後、酢酸エチル-ジイソプロピルエーテル-ヘキサンの系で、再結晶化を行って精製することで、ヒドロキシアリール化合物(92)(34.38g)を黄色オイル状にて、収率97.6%で得た。
1H-NMR(300MHz,CDCl3)δ;4.97(s,2H,-OCH2-),6.30(s,1H,-OH),6.84~6.90(m,1H),6.96~7.05(m,2H),7.06~7.14(m,1H),7.19~7.43(m,13H).
2-ヒドロキシ-2’-(2-フェニルベンジルオキシ)-1,1’-ビフェニル-3-カルバルデヒド(93)の合成:
撹拌機器と還流管を備えたフラスコを窒素置換した後、パラホルムアルデヒド(4.61g、154mmol)、無水塩化マグネシウム(9.73g、102mmol)、出発原料(92)(18g、51mmol)及びアセトニトリル(144g)を加え、20~25℃にて撹拌した。この反応溶液に、N,N,N’,N’-テトラメチルエチレンジアミン(TMEDA)(11.86g、102mmol)を滴下した。この反応溶液を反応温度80℃にて、24時間撹拌した後、20~25℃に冷却して2mol/Lの塩酸を添加して、反応を停止した。反応溶液を中和した後、クロロホルムで抽出し、有機溶媒を留去して粗生成物を得た。この粗生成物をクロロホルム-ヘキサン-酢酸エチルの系にて再結晶化することで、白色結晶としてヒドロキシアリールアルデヒド(93)(10g)を、収率54%で得た。
1H-NMR(300MHz,CDCl3)δ;4.96(s,2H,-OCH2-),6.81~6.86(m,1H),6.98~7.08(m,2H),7.20~7.43(m,11H),7.50~7.57(m,2H),9.95(s,1H,-CHO),11.33(s,1H,-OH).
2’-(2,4,6-トリ-i-プロピルベンジルオキシ)-1,1’-ビフェニル-2-オール(102)の合成:
2,2-ビフェノール(7.36g、39.5mmol)、炭酸カリウム(2.84g、20.5mmol)、ヨウ化カリウム(1.32g、7.95mmol)及びアセトン35gを、反応容器に仕込み、20~25℃にて撹拌した。この反応溶液に2,4,6-トリ-i-プロピルベンジルクロリド(101)(9.99g、39.5mmol)を仕込んだ後、反応温度55℃にて39時間撹拌した。20~25℃に冷却して、飽和塩化アンモニウム水溶液を反応溶液に添加して反応を停止し、分液した後、水層を酢酸エチルで抽出し、合わせた有機層を硫酸マグネシウムにて乾燥し濃縮することで、ヒドロキシアリール化合物(102)(13.9g)を淡橙白色結晶として、収率87.2%で得た。
1H-NMR(300MHz,CDCl3)δ;1.10(d,J=6.9Hz,12H),1.24(d,J=6.9Hz,6H),2.86(qq,J=6.9Hz,1H),3.04(qq,J=6.9Hz,2H),5.06(s,2H),6.45(s,1H),6.87~7.01(m,4H),7.14~7.29(m,4H),7.34~7.46(m,2H).
2-ヒドロキシ-2’-(2,4,6-トリ-i-プロピルベンジルオキシ)-1,1’-ビフェニル-3-カルバルデヒド(103)の合成:
撹拌機器と還流管を備えたフラスコを窒素置換した後、パラホルムアルデヒド(3.13g、104mmol)、無水塩化マグネシウム(6.62g、69.6mmol)、出発原料(102)(14.0g、34.8mmol)及びアセトニトリル(100mL)を加え、20~25℃にて撹拌した。この反応溶液に、N,N,N’,N’-テトラメチルエチレンジアミン(TMEDA)(8.08g、69.6mmol)を滴下した。この反応溶液を反応温度80℃にて、37時間撹拌した後、20~25℃に冷却して2mol/Lの塩酸を添加して、反応を停止した。反応溶液を中和した後、ジクロロメタンで抽出し、有機溶媒を留去して粗生成物を得た。この粗生成物をクロロホルム-ヘキサン-酢酸エチルの系にて再結晶化することで、白色結晶としてヒドロキシアリールアルデヒド(103)(11.8g)を、収率78.5%で得た。
1H-NMR(300MHz,CDCl3)δ;1.10(d,J=6.9Hz,12H),1.22(d,J=6.9Hz,6H),2.84(qq,J=6.9Hz,1H),3.13(qq,J=6.9Hz,2H),4.99(s,2H),6.90~6.99(m,3H),7.04~7.12(m,1H),7.20~7.31(m,2H),7.39~7.49(m,3H),9.85(s,1H),11.20(s,1H).

- 式(1)
R2とR3とは一緒になって-CR5=CR6-CR7=CR8-を形成していても良く、このときR5、R6、R7及びR8はそれぞれ独立に水素原子又は置換基Aであり、
置換基Aは、ハロゲン原子、C1-4アルキル基、C1-4ハロアルキル基、C1-4アルコキシ基、C6-12アリールオキシ基、C6-22アリール基(該アリールオキシ基及び該アリール基は、無置換であるか、又はC1-4アルキル基、C1-4ハロアルキル基、C1-4アルコキシ基、C1-4アルキルカルボニルオキシ基、C6-12アリール基(該アリール基は、無置換であるか、又はC1-4アルキル基、C1-4ハロアルキル基及びC1-4アルコキシ基から選ばれる同一又は相異なった1以上の置換基で置換されている。)、t-ブチルジメチルシリル基、C6-22アリールメチルオキシ基(該アリールメチルオキシ基は、無置換であるか、又はC1-4アルキル基、C1-4アルコキシ基、ハロゲン原子及びフェニル基から選ばれる同一又は相異なった1以上の置換基で置換されている。)及びハロゲン原子から選ばれる同一又は相異なった1以上の置換基で置換されている。)、ニトロ基又はシアノ基から選ばれる置換基である。)で表されるヒドロキシアリール化合物を、式(a)
R10はC6-12アリール基(該アリール基は、無置換であるか、又はC1-4アルキル基、C1-4アルコキシ基及びハロゲン原子から選ばれる同一又は相異なった1以上の置換基で置換されている。)であり、
nは0又は1から10までの整数である。)で表されるアミン化合物存在下、無水塩化マグネシウム及びパラホルムアルデヒドと反応させることにより、ヒドロキシアリール化合物のオルト位選択的にホルミル化することを特徴とする下記式(2)
- R1が水素原子、フェニル基(該フェニル基は、無置換であるか、又はC1-4アルキル基、C1-4ハロアルキル基、C1-4アルコキシ基、C1-4アルキルカルボニルオキシ基、C6-12アリール基(該アリール基は、無置換であるか、又はC1-4アルキル基、C1-4ハロアルキル基及びC1-4アルコキシ基から選ばれる同一又は相異なった1以上の置換基で置換されている。)、C6-22アリールメチルオキシ基(該アリールメチルオキシ基は、無置換であるか、又はC1-4アルキル基、C1-4アルコキシ基、ハロゲン原子及びフェニル基から選ばれる同一又は相異なった1以上の置換基で置換されている。)及びハロゲン原子から選ばれる同一又は相異なった1以上の置換基で置換されている。)又はナフチル基(該ナフチル基は、無置換であるか、又はC1-4アルキル基、C1-4ハロアルキル基、C1-4アルコキシ基、C1-4アルキルカルボニルオキシ基、C6-12アリール基(該アリール基は、無置換であるか、又はC1-4アルキル基、C1-4ハロアルキル基及びC1-4アルコキシ基から選ばれる同一又は相異なった1以上の置換基で置換されている。)、t-ブチルジメチルシリル基、C6-22アリールメチルオキシ基(該アリールメチルオキシ基は、無置換であるか、又はC1-4アルキル基、C1-4アルコキシ基、ハロゲン原子及びフェニル基から選ばれる同一又は相異なった1以上の置換基で置換されている。)及びハロゲン原子から選ばれる同一又は相異なった1以上の置換基で置換されており、光学活性又は光学不活性である。)であり、
R2が水素原子、フッ素原子、塩素原子、臭素原子、ヨウ素原子、メチル基、エチル基、i-プロピル基、t-ブチル基、トリフルオロメチル基又はペンタフルオロエチル基であり、R3及びR4がそれぞれ独立に水素原子、フッ素原子、塩素原子、臭素原子、ヨウ素原子、メチル基、エチル基、i-プロピル基、t-ブチル基、トリフルオロメチル基、メトキシ基、エトキシ基、i-プロポキシ基又はt-ブトキシ基である請求項1に記載の製造方法。 - R1が水素原子、フェニル基(該フェニル基は、無置換であるか、又はC1-4アルキル基、C1-4ハロアルキル基、C1-4アルコキシ基、C1-4アルキルカルボニルオキシ基、C6-12アリール基(該アリール基は、無置換であるか、又はC1-4アルキル基、C1-4ハロアルキル基及びC1-4アルコキシ基から選ばれる同一又は相異なった1以上の置換基で置換されている。)、C6-22アリールメチルオキシ基(該アリールメチルオキシ基は、無置換であるか、又はC1-4アルキル基、C1-4アルコキシ基、ハロゲン原子及びフェニル基から選ばれる同一又は相異なった1以上の置換基で置換されている。)及びハロゲン原子から選ばれる同一又は相異なった1以上の置換基で置換されている。)又はナフチル基(該ナフチル基は、無置換であるか、又はC1-4アルキル基、C1-4ハロアルキル基、C1-4アルコキシ基、C1-4アルキルカルボニルオキシ基、C6-12アリール基(該アリール基は、無置換であるか、又はC1-4アルキル基、C1-4ハロアルキル基及びC1-4アルコキシ基から選ばれる同一又は相異なった1以上の置換基で置換されている。)、t-ブチルジメチルシリル基、C6-22アリールメチルオキシ基(該アリールメチルオキシ基は、無置換であるか、又はC1-4アルキル基、C1-4アルコキシ基、ハロゲン原子及びフェニル基から選ばれる同一又は相異なった1以上の置換基で置換されている。)及びハロゲン原子から選ばれる同一又は相異なった1以上の置換基で置換されており、光学活性又は光学不活性である。)であり、
R2とR3とが一緒になって-CR5=CR6-CR7=CR8-であり、R5及びR7がそれぞれ独立に水素原子、フッ素原子、塩素原子、臭素原子、ヨウ素原子、メチル基、エチル基、i-プロピル基、t-ブチル基、トリフルオロメチル基、メトキシ基、エトキシ基、i-プロポキシ基又はt-ブトキシ基であり、R6が水素原子、フッ素原子、塩素原子、臭素原子、ヨウ素原子、メチル基、エチル基、i-プロピル基、t-ブチル基、トリフルオロメチル基又はペンタフルオロエチル基であり、R8が水素原子又はフッ素原子である請求項1に記載の製造方法。 - R9がメチル基、エチル基又は同じ窒素原子上の2つのR9が一緒になって=CH-R10を表し、R10がフェニル基、2-フルオロフェニル基、2-メチルフェニル基、2-メトキシフェニル基又は2-i-プロポキシフェニル基である請求項1に記載の製造方法。
