Definitions

  • the present invention relates to a feed additive for use as a substitute for antibiotics, which contains inulin extracted from Jerusalem artichoke, and more particularly, to feed composition as a substitute for antibiotics, which contains an inuloprebiotics, which is prepared by mixing inulin, extracted from Jerusalem artichoke, with an excipient.
  • Antibiotics have been primarily used for the prevention and treatment of diseases, but the use of antibiotics as antibacterial growth promoters in feed is being gradually prohibited. Thus, there is a need to develop novel materials, which are natural materials and, at the same time, have the effect of killing harmful bacteria, like antibiotics.
  • inulin which is composed of polysaccharides and accounts for more than 50% of Jerusalem artichoke.
  • Said inulin are fructose polymers linked by /3-2,1 bonds with the chemical structure of a non-reducing terminal ⁇ -2,1 glucosidic bond.
  • inulin is not degraded by gastric juice and digestive enzymes of human beings and livestock, more than about 80% of inulin reaches the large intestine and is fermented by intestinal microorganisms, and it has a bifidogenic effect of selectively stimulating the growth of bifidobacteria, known as beneficial intestinal microorganisms, and at the same time, inhibiting the i growth of harmful microorganisms.

  • inulin has been reported to have the effects of alleviating obesity, promoting bowel movement, reducing blood insulin and neutral lipid levels and inhibiting the development of colon cancer.

  • Prebiotics are novel materials receiving attention as feed additives and functional food materials capable of substituting for probiotics and antibiotics.
  • the term "prebiotics” refers to non-degradable dietary indegredients, which are not degraded in the stomach and the small intestine and move into the large intestine to selectively stimulate the growth of microorganisms such as bifidobacterium bifidum in the colon or stimulate a limited number of microorganisms, thus providing a beneficial effect to human beings and livestock.
  • Such prebiotics microbiologically produce nutrients, such as antibiotics, vitamins and growth promoting factors, in the large intestine, to improve gastrointestinal functions, increase the bioavailability of minerals and the amount of feces, prevent diarrhea, and enhance immune functions, thus promoting the growth of animals (Cummings, et ah, Brit. J. Nutr., 87:S145, 2002; Patterson et al., Poult. ScL, 82:627, 2003; George et al., Brit. Med. J., 318:999, 1999).
  • the present inventors have made many efforts to develop a feed additive for use as a substitute for antibiotics and, as a result, have prepared an inuloprebiotics-containing feed additive as a substitute for antibiotics using inulin extracted from Jerusalem artichoke, and have found that, when the feed additive is fed to livestock, it has a bifidogenic effect of inhibiting the growth of harmful microorganisms in the intestines, thereby completing the present invention.
  • the present invention provides an antibiotic-free feed composition, which comprises an inuloprebiotics containing 100 parts by weight of an excipient selected from the group consisting of barley bran, wheat middling, limestone powder, and maize powder; and 0.05-10 parts by weight of inulin extracted from Jerusalem artichoke.

  • 0.05-10 parts by weight of inulin, extracted from Jerusalem artichoke was mixed with 100 parts by weight of barley bran which is an excipient.
  • the prepared inuloprebiotics were added to feed in an amount of 0.1-0.5 parts by weight based on 100 parts by weight of the feed, thus preparing a feed composition. It was observed that, when the feed composition was fed to animals, it had the effect of substituting for antibiotics.
  • Example 1 Inulin extraction
  • Jerusalem artichoke was washed, crushed or cut thinly, and steamed in an autoclave for 30 minutes.
  • the steamed Jerusalem artichoke was mixed with water (80 "C) at a ratio of 1 ::5 and, and the mixture was stirred for 20 minutes.
  • the stirred mixture solution was filtered through cotton cloth, and the filtrate was centrifuged at 3,000 rpm. The precipitate was collected and dried, thus extracting inulin.
  • Example 1 Using the inulin extracted in Example 1, the growth rates of Bifidobacterium bifidum, known as typical beneficial intestinal bacteria, and Clostridium perfringens, known as harmful bacteria, were measured in liquid media. For this purpose, in a microbial culture medium containing 0.5 wt% of said inulin, Bifidobacterium bifidum and Clostridium perfringens were incubated at 37 ° C for 48 hours. The growth rates of the microorganisms were measured by comparing the OD value of the bacteria, grown in an inulin-free liquid medium, with the OD value of the bacteria grown in the inulin-containing liquid medium.
  • the case of using the microbial culture medium containing 0.5 wt% of inulin showed the effect of promoting the growth of Bifidobacterium bifidum after 15 hours from the start of the incubation, unlike the case of using the inulin-free culture medium.
  • the case of using microbial culture medium containing 0.5 wt% of inulin showed the effect of inhibiting the growth of the Clostridium perfringens strain after 15 hours from the start of the incubation. This suggests that the Jerusalem artichoke- derived inulin had the "prebiotic" effect of promoting the growth of Bifidobacteria.
