[0001] 本出願は、 2006年 7月 19日に出願された日本国特許出願 2006— 196943号の優 先権を主張するものであり、この先の出願の開示全体をここに参照のために取り込む 技術分野
[0002] 本発明は、試料観察方法および顕微鏡、特にフォトクロミック分子を含む試料を、第 1 の光および第 2の光により照明して、超解像で観察する試料観察方法および顕微鏡 に関するものである。

[0003] 光学顕微鏡の技術は古ぐ種々のタイプの顕微鏡が開発されてきた。また、近年では 、レーザ技術および電子画像技術をはじめとする周辺技術の進歩により、さらに高機 能の顕微鏡システムが開発されて!ヽる。
[0004] このような背景の中、複数波長の光で試料を照明することにより発する二重共鳴吸収 過程を用いて、得られる画像のコントラストの制御のみならずィ匕学分析も可能にした 高機能な顕微鏡が提案されている (例えば、特開平 8— 184552号公報参照)。
[0005] この顕微鏡は、二重共鳴吸収を用いて特定の分子を選択して、特定の光学遷移に 起因する吸収および蛍光を観察するものである。この原理について、図 7〜図 10を 参照して説明する。図 7は、試料を構成する分子の価電子軌道の電子構造を示すも ので、先ず、図 7に示す基底状態 (SO状態)の分子力 Sもつ価電子軌道の電子を波長 λ 1の光により励起して、図 8に示す第 1電子励起状態 (S1状態)とする。次に、別の 波長 λ 2の光により同様に励起して、図 9に示す第 2電子励起状態 (S2状態)とする。 この励起状態により、分子は蛍光あるいは燐光を発光して、図 10に示すように基底 状態に戻る。
[0006] 二重共鳴吸収過程を用いた顕微鏡法では、図 9の吸収過程や図 10の蛍光や燐光 の発光を用いて、吸収像や発光像を観察する。この顕微鏡法では、最初にレーザ光 等により共鳴波長 λ ΐの光で図 8のように試料を構成する分子を SI状態に励起させ るが、この際、単位体積内での S1状態の分子数は、照射する光の強度が増加する に従って増加する。
[0007] ここで、線吸収係数は、分子一個当りの吸収断面積と単位体積当たりの分子数との 積で与えられるので、図 9のような励起過程においては、続いて照射する共鳴波長え 2に対する線吸収係数は、最初に照射した波長 λ 1の光の強度に依存することにな る。すなわち、波長 λ 2に対する線吸収係数は、波長 λ 1の光の強度で制御できるこ とになる。このことは、波長 λ 1および波長 λ 2の 2波長の光で試料を照射し、波長 λ 2による透過像を撮影すれば、透過像のコントラストは波長 λ 1の光で完全に制御で さることを示している。
[0008] また、図 9の励起状態での蛍光または燐光による脱励起過程が可能である場合には 、その発光強度は S1状態にある分子数に比例する。したがって、蛍光顕微鏡として 利用する場合にも画像コントラストの制御が可能となる。
[0009] さらに、二重共鳴吸収過程を用いた顕微鏡法では、上記の画像コントラストの制御の みならず、化学分析も可能にする。すなわち、図 7に示される最外殻価電子軌道は、 各々の分子に固有なエネルギー準位を持つので、波長 λ 1は分子によって異なるこ とになり、同時に波長 λ 2も分子固有のものとなる。
[0010] ここで、従来の単一波長で照明する場合でも、ある程度特定の分子の吸収像あるい は蛍光像を観察することが可能である力 一般にはいくつかの分子における吸収帯 の波長領域は重複するので、試料の化学組成の正確な同定までは不可能である。
[0011] これに対し、二重共鳴吸収過程を用いた顕微鏡法では、波長 λ 1および波長 λ 2の 2波長により吸収あるいは発光する分子を限定するので、従来法よりも正確な試料の 化学組成の同定が可能となる。また、価電子を励起する場合、分子軸に対して特定 の電場ベクトルをもつ光のみが強く吸収されるので、波長 λ 1および波長 λ 2の偏光 方向を決めて吸収または蛍光像を撮影すれば、同じ分子でも配向方向の同定まで 可能となる。
[0012] また、最近では、二重共鳴吸収過程を用いて回折限界を越える高い空間分解能をも つ蛍光顕微鏡も提案されて ヽる(例えば、特開 2001 - 100102号公報参照)。 [0013] 図 11は、分子における二重共鳴吸収過程の概念図で、基底状態 SOの分子が、波長 λ ΐの光で第 1電子励起状態である S1に励起され、さらに波長 λ 2の光で第 2電子 励起状態である S2に励起されている様子を示している。なお、図 11はある種の分子 の S2からの蛍光が極めて弱!、ことを示して!/、る。
[0014] 図 11に示すような光学的性質を持つ分子の場合には、極めて興味深い現象が起き る。図 12は、図 11と同じく二重共鳴吸収過程の概念図で、横軸の X軸は空間的距離 の広がりを表わし、波長 λ 2の光を照射した空間領域 A1と波長 λ 2の光が照射され な 、空間領域 AOとを示して!/、る。
[0015] 図 12において、空間領域 AOでは波長 λ 1の光の励起により S1状態の分子が多数 生成され、その際に空間領域 AO力 は波長え 3で発光する蛍光が見られる。しかし 、空間領域 A1では、波長え 2の光を照射したため、 S1状態の分子のほとんどが即座 に高位の S2状態に励起されて、 S1状態の分子は存在しなくなる。このような現象は 、幾つかの分子により確認されている。これにより、空間領域 A1では、波長え 3の蛍 光は完全になくなり、し力も S2状態からの蛍光はもともとないので、空間領域 A1では 完全に蛍光自体が抑制され (蛍光抑制効果)、空間領域 AOからのみ蛍光が発するこ とになる。
[0016] このことは、顕微鏡の応用分野から考察すると、極めて重要な意味を持っている。す なわち、従来の走査型レーザ顕微鏡等では、レーザ光を集光レンズによりマイクロビ ームに集光して観察試料上を走査するが、その際のマイクロビームのサイズは、集光 レンズの開口数と波長とで決まる回折限界となり、原理的にそれ以上の空間分解能 は期待できない。
[0017] ところが、図 12の場合には、波長 λ 1と波長 λ 2との 2種類の光を空間的に上手く重 ね合わせて、波長 λ 2の光の照射により蛍光領域を抑制することで、例えば波長 λ 1 の光の照射領域に着目すると、蛍光領域を集光レンズの開口数と波長とで決まる回 折限界よりも狭くでき、実質的に空間分解能を向上させることが可能となる。以下、波 長 λ 1の光をポンプ光、波長 λ 2の光をィレース光とも呼ぶ。したがって、この原理を 利用することで、回折限界を越える二重共鳴吸収過程を用いた超解像顕微鏡、例え ば超解像蛍光顕微鏡を実現することが可能となる。 [0018] さらに、近年では、生物学的に興味深いフォトクロミック分子をプローブとする超解像 技術が提案されている。この分子は、特定の波長の光刺激より、蛍光発光特性をもつ 第 1安定状態から蛍光発光性の無い第 2安定状態に変異する (光異性化)。また、他 の変異の過程として特定の化学結合が切断または接合したり、電荷移動を起こしたり することでも光異性ィ匕が起きる。基本的には、光異性化が起きると分子構造が全く変 化するので、その光学的性質も変化する。
[0019] 例えば、フォトクロミック部位をもつ蛍光タンパク FP595は、第 1安定状態において波 長 595nm近傍の黄色い光を照射すると、これより長波長領域の赤い蛍光を発光す る。ところが、波長 458nm以下の短波長の光を照射すると第 2安定状態に変異して、 もはや黄色い光を照射しても、光学的性質が全く変化して蛍光は発しない。一度、第 2安定状態に変異をすると、その状態は安定で、外部力 エネルギー刺激を強制的 に与えないと短時間内に第 1安定状態には戻らない性質がある。
[0020] フォトクロミック分子をプローブとする超解像技術では、上記のようなフォトクロミック分 子の特性を一種の蛍光抑制効果として応用しょうとするもので、生物用顕微鏡への 応用が期待されている。
[0021] しかしながら、従来報告されているフォトクロミック分子に関する超解像技術では、分 光学的に蛍光抑制効果の発現が弱いために、超解像効果の発現が二重共鳴吸収 過程と比較して弱いと言う問題がある。また、光異性ィ匕が遅いために、蛍光抑制およ び第 1安定状態への復帰が遅ぐ計測時間が力かる問題があった。
[0022] 以下、この点について説明する。図 13 (a)は、従来の二重共鳴吸収過程を用いた超 解像法の場合の分光ダイヤグラム、図 13 (b)は、フォトクロミック分子の蛍光抑制効果 を利用した超解像法の場合の分光ダイヤグラムを示しており、横軸は核間距離を、縦 軸は励起エネルギーを示して 、る。
[0023] 二重共鳴吸収過程を利用する超解像法では、図 13 (a)に示すように、基底状態 (SO )の分子を、光子エネルギー E21のポンプ光照射により蛍光発光可能な第 1励起状 態(S1)に励起し、さらに蛍光抑制させるために、光子エネルギー E22のィレース光 照射により上位の励起状態 (S3)に遷移させて、 S3からは熱緩和により蛍光を抑制し ている。この時の光反応過程は、全てナノ秒以下であり非常に高速である。 [0024] これに対し、フォトクロミック分子は、図 13 (b)に示すように、第 1安定状態(SO)およ び第 2安定状態(S3)に対応する 2つの準安定なローカルミニマムのポテンシャルを 持っている。このため、フォトクロミック分子の蛍光抑制効果を利用する超解像法では 、一般には、第 1安定状態である基底状態 (SO)にあるフォトクロミック分子を、光子ェ ネルギー E31のポンプ光照射により蛍光発光可能な第 1励起状態 (S1)に励起し、さ らに蛍光抑制させるために、第 1安定状態 (SO)から第 2安定状態 (S3)への変異に 不可欠な障壁エネルギー Etを超える光子エネルギー E32のィレース光照射により上 位の第 2励起状態 (S2)に遷移および経由させることで、第 2安定状態 (S3)に変異さ せるようにしている。
[0025] しかし、この場合、第 2安定状態(S3)に変異させるィレース光の強度に関しては、あ まり強くすると、一度は第 1安定状態 (S 1)から第 2安定状態 (S3)に変異したものの、 再びィレース光によって励起されて第 1安定状態(S1)に戻り、再びポンプ光励起に より蛍光が発せられて、残留蛍光成分が生じることになる。
[0026] この現象は、幾らィレース光強度を強くしても、蛍光抑制が確実に誘起できないことを 意味しており、超解像法の結像性能に好ましくない影響を与えることになる。この点に ついて、図 14に示す 2次元点像分布関数を参照して説明する。
[0027] 超解像法では、ガウシアンタイプのポンプ光に、光軸直交平面における断面形状が ドーナッツ形状のィレース光を空間的に同軸上に重ね合わせて試料に照射する。こ の際、二重共鳴吸収過程を用いる場合には、ィレース光強度が十分なポンプ光の辺 縁部においては蛍光が完全に抑制されるので、得られる蛍光スポットパターンすなわ ち 2次元点像分布関数は、図 14に符号 Iで示すように、非常に幅の細いローレンツタ イブ関数なる。
[0028] これに対し、フォトクロミック分子の蛍光抑制効果を利用する場合には、幾らィレース 光強度を強くしても蛍光抑制が確実に誘起されないため、ポンプ光の辺縁部で蛍光 抑制が不完全となる。その結果、得られる 2次元点像分布関数は、図 14に符号 IIで 示すように、先尖のみでスポットサイズの収縮が起こり、裾野では蛍光抑制が空間的 に十分にできていない関数形となる。なお、図 14において、符号 IIIは、ポンプ光の 2 次元点像分布関数を示して 、る。 発明の開示
[0029] したがって、力かる点に鑑みてなされた本発明の目的は、フォトクロミック分子を含む 試料を、蛍光抑制効果を確実に誘起して、残留蛍光成分を生じることなく超解像で 観察できる試料観察方法および顕微鏡を提供することにある。
[0030] 上記目的を達成する第 1の観点に係る試料観察方法の発明は、少なくとも第 1安定 状態 SOおよび第 2安定状態 S3の量子状態を有するフォトクロミック分子を含む試料 を観察するにあたり、
上記フォトクロミック分子を上記第 1安定状態 SOから光応答可能な第 1励起状態 S1 に励起する第 1の光と、上記フォトクロミック分子を上記第 1励起状態 S1から他のエネ ルギー準位の第 2励起状態 S2に励起する第 2の光とを用い、
上記第 1の光および上記第 2の光を一部重ね合わせて上記試料に照射し、上記第 1の光および上記第 2の光が重ね合わされて照射された領域の上記フォトクロミック分 子を上記第 2励起状態 S2を経て上記第 2安定状態 S3に遷移させ、上記第 1の光の みが照射された領域の上記フォトクロミック分子を上記第 1励起状態 S1に遷移させて 、上記試料を観察することを特徴とするものである。
[0031] さらに、上記目的を達成する第 2の観点に係る顕微鏡の発明は、少なくとも第 1安定 状態 SOおよび第 2安定状態 S3の量子状態を有するフォトクロミック分子を含む試料 を観察する顕微鏡であって、
上記フォトクロミック分子を上記第 1安定状態 SOから光応答可能な第 1励起状態 S1 に励起する第 1の光を出射する第 1の光源と、
上記フォトクロミック分子を上記励起状態 S1から他のエネルギー準位の第 2励起状 態 S2に励起する第 2の光を出射する第 2の光源と、
上記第 1の光および上記第 2の光を一部重ね合わせて上記試料に集光して照射す る光学系と、
上記光学系により上記第 1の光および上記第 2の光が重ね合わされて照射される 領域の上記フォトクロミック分子を上記第 2励起状態 S2を経て上記第 2安定状態 S3 に遷移させ、上記第 1の光のみが照射される領域の上記フォトクロミック分子を上記 第 1励起状態 S1に遷移させるように、上記光学系を経て照射される光と上記試料と を相対的に移動させて上記試料を走査する走査部と、
上記第 1励起状態 S1に励起された上記フォトクロミック分子力も発生する光応答を 検出する検出部と、を有することを特徴とするものである。

[0042] 図 1は、本発明の原理を説明するフォトクロミック分子の分光ダイヤグラムを示してい る。本発明では、従来のようにィレース光によりフォトクロミック分子を第 1安定状態か ら第 2安定状態へ直接変異させるのではなぐフォトクロミック分子に 2色の第 1の光お よび第 2の光を照射することにより二重共鳴吸収を誘導させて、フォトクロミック分子を 第 1安定状態から第 2安定状態へ変異させる。
[0043] このため、先ず、第 1安定状態である基底状態 (SO)にあるフォトクロミック分子を、光 子エネルギー E1の第 1の光 (ポンプ光)照射により光応答可能な第 1励起状態 (S 1) に励起遷移させる。この第 1励起状態(S1)に遷移すると、フォトクロミック分子は、通 常は、光応答過程により基底状態(SO)に緩和する。
[0044] 本発明では、第 1励起状態 (S1)が緩和しない間に、光子エネルギー E2の第 2の光( ィレース光)照射により、フォトクロミック分子を第 1励起状態 (S1)から、それよりも高 い量子レベルの第 2励起状態 (S2)に励起遷移させる。この第 2励起状態 (S2)に遷 移させる際の光子エネルギー E2は、第 1安定状態 (SO)から第 2安定状態 (S3)への 変異に不可欠な障壁エネルギー Et以上で、かつ、第 2安定状態(S3)において、フ オトクロミック分子がポンプ光およびィレース光の照射によって二重共鳴吸収を起こさ ないことが条件となる。
[0045] このため、本発明では、フォトクロミック分子が第 1安定状態 (SO)から第 2安定状態( S3)へ変異すると、分子構造が変化して光学的性質も変わることに着目し、ポンプ光 およびィレース光による励起によって、第 2安定状態(S3)では二重共鳴吸収の共鳴 条件力 外れるように、それぞれの光の光子エネルギー E1および E2を設定する。こ のようにすれば、ポンプ光およびィレース光の照射によっては、第 2安定状態(S3)か ら第 1安定状態 (SO)への変異は起きないので、残留蛍光成分は生じない。もちろん 、ポンプ光またはィレース光の単独励起では、第 2安定状態(S3)の分子の励起は不 可能なので、蛍光発光は生じない。
[0046] さらに、本発明の好適実施の形態では、第 2安定状態 (S3)から第 1安定状態 (SO) への変異を確実に阻止するため、選択するフォトクロミック分子がもつ量子状態間の エネルギー間隔を、下記の条件を満たすようにする。
[0047] 1)大前提として、第 1安定状態 (SO)にあるフォトクロミック分子を光応答可能な第 1励 起状態(S1)に励起するポンプ光の光子エネルギー E1は、第 1安定状態(SO)から 第 2安定状態 (S3)への変性が起こらないエネルギーとする。すなわち、第 1安定状 態 (SO)から第 2安定状態 (S3)への変異に要する障壁エネルギー Etよりも小さくする

発明を実施するための最良の形態
[0067] (第 1実施の形態)本実施の形態では、フォトクロミック分子として蛍光タンパク FP595 を含む試料を観察する。蛍光タンパク FP595は、第 1安定状態(SO)で波長 568nm の光(ポンプ光)を照射すると、波長 605nmに蛍光発光ピークをもち、波長 458nm の光 (ィレース光)を照射すると、障壁エネルギーを乗り越えて、第 2安定状態 (S3) に変性し、一度、第 2安定状態 (S3)に変性するとポンプ光を照射しても蛍光を発し な!、が、ィレース光の照射では蛍光を発光する特性を有して 、る。
[0068] 蛍光タンパク FP595を含む試料の場合、ポンプ光波長を勘案すると、 SO状態から光 応答としての蛍光発光可能な第 1励起状態 S1へ励起遷移させるために必要な光子 エネルギー E1は、 2. 2eVとなる。一方、ィレース光単独で第 1安定状態(SO)から障 壁エネルギーを越えて第 2安定状態 (S3)へ変異させるのに必要な光子エネルギー E32 (図 13 (b)参照)は、 2. 7eVである。したがって、少なくとも 2. 7eV以上の光子 エネルギーをもつ光がィレース光として利用できる。
[0069] ここで、ポンプ光とィレース光との光子エネルギーの差は、 0. 5eVである。すなわち、 第 1励起状態 S1を基準にすると、そこ力 0. 5eVの上位のエネルギー準位として第 2励起状態 S2が存在する。一般に、第 2励起状態 S2以上の電子状態はエネルギー 間隔が密に存在し、し力もそれに帰属する振動 ·回転準位も広く分散するので、第 1 励起状態 S1の分子に 0. 5eV以上の光子エネルギーを有する光を照射すると、障壁 エネルギー Etを超えて第 2安定状態 (S3)に変性させることができる。これは、波長換 算すると、 2. 4 mとなる。
[0070] したがって、ィレース光として、 0. 5eV以上で、かつ 2. 2eV (ポンプ光の光子エネル ギー)以下の光子エネルギーを有する光を用いれば、第 1安定状態(SO)の蛍光タン パク FP595を第 1励起状態 S1に励起して、さらに効率的に第 2安定状態 (S3)に変 性させることがきる。言い換えると、波長が 568nmより長く 2. 4 mより短い光源を選 ベば、ィレース光として用いることができる。このような波長帯域で発振できるレーザと しては、例えば、 Ti:サフアイャレーザや、手軽な近赤外の各種半導体レーザが知ら れている。
[0071] 従来の二重共鳴吸収過程のみを用いた超解像顕微鏡法においては、ィレース光源 として先頭値の高いレーザ光が必要であった。そのため、ピコ秒、フェムト秒といった 極短パルスレーザ光を、取り扱 ヽが難し ヽ各種非線形結晶を用いて波長変換を行つ ていた。その結果、顕微鏡システムが非常に大掛かりとなって、生物系の一般ユーザ には使いにくいものとなっていた。
[0072] これに対し、本実施の形態の蛍光タンパク FP595を用いた超解像顕微鏡法では、ィ レース光強度が wクラスでも使えるので、光源の選択の幅が広がるのと同時に、生 体試料へのダメージも大幅に低減できる。
[0073] 図 2は、本実施の形態による超解像顕微鏡の光学系の要部構成図である。この超解 像顕微鏡は、通常のレーザ走査型蛍光顕微鏡を前提としたもので、主に 3つの独立 したユニット、すなわち、光源ユニット 10、スキャンユニット 30および顕微鏡ユニット 5 0からなつている。
[0074] 光源ユニット 10は、例えば波長 568nmのポンプ光を出射するアルゴン 'クリプトンレ 一ザからなる第 1の光源 11と、波長 1064nmのィレース光を出射する Nd: YAGレー ザ力もなる第 2の光源 12と、ィレース光空間変調用の位相板 13と、ィレース光および ポンプ光を同軸上に重ね合わせるためのビームコンパイナ 14とを有している。位相 板 13は、図 3にか拡大平面図を示すように、光軸対称の位置で通過したィレース光 の位相が反転するように調整された光学薄膜を蒸着して構成されている。図 3では、 光軸の周りに独立した 4つの領域を有し、ィレース光波長に対して 4分 1ずつ位相が 異なっている。この位相板 13を通過した光を集光すれば、光軸上で電場が相殺され 中空状のィレース光が生成される。
[0075] スキャンユニット 30は、光源ユニット 10から出射される同じ光学軸を共有するポンプ 光とィレース光とを、ハーフミラー 31を通過させた後、 2枚のガルバノミラー 32および 33により 2次元方向に揺動走査して、後述の顕微鏡ユニット 50に出射するようになつ ているとともに、顕微鏡ユニット 50で検出された蛍光を、往路とは逆の経路を迪つて ハーフミラー 31で分岐し、その分岐された蛍光を投影レンズ 34、ピンホール 35、ノッ チフィルタ 36および 37を経て光電子増倍管 38で受光するようになっている。図 2で は、図面を簡略ィ匕するため、ガルバノミラー 32, 33を同一平面内で揺動可能に示し ている。なお、ノッチフィルタ 36および 37は、蛍光に混入したポンプ光およびィレー ス光を除去するものである。また、ピンホール 35は、共焦点光学系を成す重要な光 学素子で、観察試料内の特定の断層面で発光した蛍光のみを通過させるものである
[0076] 顕微鏡ユニット 50は、いわゆる通常の蛍光顕微鏡で、スキャンユニット 30から入射す るポンプ光およびィレース光をノヽーフミラー 51で反射させて、対物レンズ 52により蛍 光タンパク FP595を含む試料 53上に集光させるとともに、試料 53で発光した蛍光を 、再び対物レンズ 52でコリメートしてハーフミラー 51で反射させることにより、再び、ス キャンユニット 30に戻すと同時に、ハーフミラー 51を通過する蛍光の一部を接眼レン ズ 54に導 、て、蛍光像として目視観察できるようになって!/、る。
[0077] 本実施の形態では、ポンプ光およびィレース光を試料 53に連続的に照射しながら試 料 53を走査して、中空状のィレース光の中央部を蛍光観察領域として超解像で観察 する。したがって、本実施の形態の場合には、ィレース光の照射によって第 2安定状 態に変性される領域が、熱緩和により第 1安定状態に復帰する時間に同期させて、 試料 53を走査する。
[0078] (第 2実施の形態)本発明の第 2実施の形態では、第 1実施の形態の超解像顕微鏡 において、第 1の光源 11からのポンプ光および第 2の光源 12からのィレース光を、蛍 光タンパク FP595が変性したときの寿命の長さに着目して、図 4に示すタイミングで 試料 53にパルス状に照射する。
[0079] すなわち、試料 53の各観察点において、先ず、ポンプ光およびィレース光を同時照 射して、信号検出に必要ない領域にある分子を第 2安定状態に変性させ (超解像記 録)、続いて、熱緩和によって第 2安定状態から第 1安定状態に戻る前に、ポンプ光 を単独照射し、そのポンプ光の単独照射に同期して回折限界以下の狭い領域にあ る第 1安定状態の蛍光タンパク FP595からの蛍光を読み出す。
[0080] このように、本実施の形態によれば、ポンプ光単独照射時の蛍光信号を計測するの で、ィレース光の散乱光が混入せず、良好な SZNで試料 53を超解像で観察するこ とがでさる。
[0081] なお、パルス状のポンプ光およびィレース光は、第 1の光源 11および第 2の光源 12 の駆動を制御して、それらの出力光自体をオン'オフして得る力、あるいは第 1の光源 11および第 2の光源 12からはそれぞれ CWのポンプ光およびィレース光を出射させ 、ビームコンパイナ 14と第 1の光源 11および第 2の光源 12との間の光路中に、それ ぞれ電気光学変調素子や音響光変調素子等からなる強度変調手段を配置して、対 応する強度変調手段の駆動を制御することにより、パルス状のポンプ光およびィレー ス光を得る。 [0082] (第 3実施の形態)図 5は、本発明の第 3実施の形態による超解像顕微鏡の光学系の 要部構成図である。本実施の形態は、図 2に示す構成において、光源ユニット 10に、 蛍光タンパク FP595が第 2安定状態力も第 1安定状態に復帰するのを促進する第 3 の光 (復帰光)を出射する第 3の光源 15と、この第 3の光源 15からの復帰光を、その 光軸をポンプ光ゃィレース光の光軸と一致させてスキャンユニット 30に入射させるビ ームコンパイナ 16とを付カ卩したものである。その他の光学系の構成は、図 2と同様で ある。
[0083] 本実施の形態では、第 1の光源 11からのポンプ光、第 2の光源 12からのィレース光 および第 3の光源 15からの復帰光を、図 6に示すタイミングで試料 53にパルス状に 照射する。すなわち、図 4に示した第 2実施の形態と同様に、試料 53の各観察点に おいて、先ず、ポンプ光およびィレース光を同時照射して、信号検出に必要ない領 域にある分子を第 2安定状態に変性させ (超解像記録)、続いて、熱緩和によって第 2安定状態から第 1安定状態に戻る前に、ポンプ光を単独照射し、そのポンプ光の単 独照射に同期して回折限界以下の狭い領域にある第 1安定状態の蛍光タンパク FP 595からの蛍光を読み出す。その後、超解像計測が終了した当該観察点に、第 3の 光源 15からの復帰光を照射して、第 2安定状態から第 1安定状態への復帰を促進さ せる。
[0084] ここで、パルス状の復帰光は、パルス状のポンプ光およびィレース光を得る場合と同 様に、第 3の光源 15の駆動を制御して、その出力光自体をオン'オフして得る力、あ るいは第 3の光源 15からは CWの復帰光を出射させ、ビームコンパイナ 16と第 3の光 源 15との間の光路中に強度変調手段を配置し、その駆動を制御してパルス状の復 帰光を得る。また、第 3の光源 15から出射させる復帰光は、第 2の光源 12から出射さ れるィレース光よりも短い波長で、かつ蛍光タンパク FP595を第 2安定状態から第 1 安定状態へ復帰するための障壁エネルギー Ec (図 1参照)より大き 、光子エネルギ 一を持つものとする。
[0085] このように、本実施の形態によれば、超解像計測が終了した試料 53の観察点に、第 3の光源 15からの復帰光を照射して、蛍光タンパク FP595の第 2安定状態から第 1 安定状態への復帰を促進させるようにしたので、第 2実施の形態の効果に加えて、第 1安定状態への復帰を早めることができる。その結果、試料 53の走査を早めることが でき、大幅な計測時間の短縮が図れる。
[0086] なお、上記実施の形態では、蛍光タンパク FP595を例にとって説明した力 本発明 は蛍光タンパク FP595を含む試料に限らず、他のフォトクロミック分子を含む試料を 観測する場合も有効に適用することができ、フォトクロミック分子の光学特性に応じて 、第 1の光 (ポンプ光)および第 2の光 (ィレース光)、さらには第 3の光 (復帰光)を適 宜設定すればよい。
産業上の利用可能性
[0087] 本発明によれば、フォトクロミック分子を含む試料を、蛍光抑制効果を確実に誘起し て、残留蛍光成分を生じることなく観察できるので、従来の二重共鳴吸収過程を利用 した超解像顕微鏡法に勝るとも劣らない結像性能を得ることができる。また、二重共 鳴吸収過程を用いた超解像顕微鏡法では、非蛍光緩和の過程が熱緩和のパスのみ であったのに対して、フォトクロミック分子の場合には、フォトクロミック光反応も非蛍光 緩和の過程に加わるので、弱 、強度の第 2の光 (ィレース光)で蛍光抑制を実現する ことができるとともに、生体試料へのダメージも大幅に低減することができる。その結 果、商用顕微鏡装置の構成を考える際に、超解像顕微鏡のもつ高いパフォーマンス をもちつつ、し力もシステム構成が簡単な商品コンセプトを提示することができる。

Claims

請求の範囲
[1] 少なくとも第 1安定状態 SOおよび第 2安定状態 S3の量子状態を有するフォトクロミツ ク分子を含む試料を観察するにあたり、
上記フォトクロミック分子を上記第 1安定状態 SOから光応答可能な第 1励起状態 S1 に励起する第 1の光と、上記フォトクロミック分子を上記第 1励起状態 S1から他のエネ ルギー準位の第 2励起状態 S2に励起する第 2の光とを用い、
上記第 1の光および上記第 2の光を一部重ね合わせて上記試料に照射し、上記第 1の光および上記第 2の光が重ね合わされて照射された領域の上記フォトクロミック分 子を上記第 2励起状態 S2を経て上記第 2安定状態 S3に遷移させ、上記第 1の光の みが照射された領域の上記フォトクロミック分子を上記第 1励起状態 S1に遷移させて 、上記試料を観察することを特徴とする試料観察方法。
[2] 少なくとも第 1安定状態 SOおよび第 2安定状態 S3の量子状態を有するフォトクロミツ ク分子を含む試料を観察する顕微鏡であって、
上記フォトクロミック分子を上記第 1安定状態 SOから光応答可能な第 1励起状態 S1 に励起する第 1の光を出射する第 1の光源と、
上記フォトクロミック分子を上記励起状態 S1から他のエネルギー準位の第 2励起状 態 S2に励起する第 2の光を出射する第 2の光源と、
上記第 1の光および上記第 2の光を一部重ね合わせて上記試料に集光して照射す る光学系と、
上記光学系により上記第 1の光および上記第 2の光が重ね合わされて照射される 領域の上記フォトクロミック分子を上記第 2励起状態 S2を経て上記第 2安定状態 S3 に遷移させ、上記第 1の光のみが照射される領域の上記フォトクロミック分子を上記 第 1励起状態 S1に遷移させるように、上記光学系を経て照射される光と上記試料と を相対的に移動させて上記試料を走査する走査部と、
上記第 1励起状態 S1に励起された上記フォトクロミック分子力も発生する光応答を 検出する検出部と、を有することを特徴とする顕微鏡。
[3] 上記第 1の光の光子エネルギーを El、上記第 2の光の光子エネルギーを E2、上記 フォトクロミック分子を上記第 1安定状態 SOから上記第 2安定状態 S3へ変異させる際 の障壁エネルギーを Et、上記フォトクロミック分子を上記第 2安定状態 S3から上記第
1安定状態 SOへ変異させる際の障壁エネルギーを Ecとするとき、
E2<El <Et<El +E2、かつ El <Ec
を満足することを特徴とする請求項 2に記載の顕微鏡。
[4] 上記第 1の光の照射により上記第 1励起状態 S1に励起された上記フォトクロミック分 子が緩和しない間に、上記第 2の光の照射を開始するように、上記第 1の光および上 記第 2の光の照射タイミングを制御することを特徴とする請求項 2に記載の顕微鏡。
[5] 上記試料の各観察点において、上記第 1の光および上記第 2の光を照射してから、 上記第 1の光を単独照射するように、上記第 1の光および上記第 2の光の照射タイミ ングを制御し、
上記第 1の光の単独照射により上記フォトクロミック分子から発生する光応答を上記 検出部で検出することを特徴とする請求項 2に記載の顕微鏡。
[6] 上記フォトクロミック分子を上記第 2安定状態 S3から上記第 1安定状態 SOに変異さ せる際の障壁エネルギー Ecよりも大きい光子エネルギーをもつ第 3の光を出射する 第 3の光源を有し、上記試料の各観察点において、上記フォトクロミック分子力も発生 する光応答の検出後に、当該観察点に上記光学系を経て上記第 3の光を照射して、 上記第 2安定状態 S3にある上記フォトクロミック分子を上記第 1安定状態 SOに復帰さ せることを特徴とする請求項 2に記載の顕微鏡。
[7] 上記第 1の光の照射により上記第 1励起状態 S1に励起された上記フォトクロミック分 子が緩和しない間に、上記第 2の光の照射を開始するように、上記第 1の光および上 記第 2の光の照射タイミングを制御することを特徴とする請求項 3に記載の顕微鏡。
[8] 上記試料の各観察点において、上記第 1の光および上記第 2の光を照射してから、 上記第 1の光を単独照射するように、上記第 1の光および上記第 2の光の照射タイミ ングを制御し、
上記第 1の光の単独照射により上記フォトクロミック分子から発生する光応答を上記 検出部で検出することを特徴とする請求項 3に記載の顕微鏡。
[9] 上記フォトクロミック分子を上記第 2安定状態 S3から上記第 1安定状態 SOに変異さ せる際の障壁エネルギー Ecよりも大きい光子エネルギーをもつ第 3の光を出射する 第 3の光源を有し、上記試料の各観察点において、上記フォトクロミック分子力も発生 する光応答の検出後に、当該観察点に上記光学系を経て上記第 3の光を照射して、 上記第 2安定状態 S3にある上記フォトクロミック分子を上記第 1安定状態 SOに復帰さ せることを特徴とする請求項 3に記載の顕微鏡。
[10] 上記試料の各観察点において、上記第 1の光および上記第 2の光を照射してから、 上記第 1の光を単独照射するように、上記第 1の光および上記第 2の光の照射タイミ ングを制御し、
上記第 1の光の単独照射により上記フォトクロミック分子から発生する光応答を上記 検出部で検出することを特徴とする請求項 4に記載の顕微鏡。
[11] 上記フォトクロミック分子を上記第 2安定状態 S3から上記第 1安定状態 SOに変異さ せる際の障壁エネルギー Ecよりも大きい光子エネルギーをもつ第 3の光を出射する 第 3の光源を有し、上記試料の各観察点において、上記フォトクロミック分子力も発生 する光応答の検出後に、当該観察点に上記光学系を経て上記第 3の光を照射して、 上記第 2安定状態 S3にある上記フォトクロミック分子を上記第 1安定状態 SOに復帰さ せることを特徴とする請求項 4に記載の顕微鏡。
[12] 上記フォトクロミック分子を上記第 2安定状態 S3から上記第 1安定状態 SOに変異さ せる際の障壁エネルギー Ecよりも大きい光子エネルギーをもつ第 3の光を出射する 第 3の光源を有し、上記試料の各観察点において、上記フォトクロミック分子力も発生 する光応答の検出後に、当該観察点に上記光学系を経て上記第 3の光を照射して、 上記第 2安定状態 S3にある上記フォトクロミック分子を上記第 1安定状態 SOに復帰さ せることを特徴とする請求項 5に記載の顕微鏡。
