技術分野
[0001 ] 本発明は、 樹状細胞免疫受容体 (DG I R)遺伝子を欠損 (ノックアウト(K0) ) した関節症等を含む自己免疫疾患や骨疾患のモデル動物に関する。 さらに本 発明は、 当該モデル動物を用いた疾患治療/予防薬のスクリーニング方法、 さらには当該スクリーニング方法によって得られる自己免疫疾患又は骨疾患 の診断及び/又は治療にも関する。
背景技術
[0002] 関節リウマチ(RA)は、 全身性の慢性炎症性疾患であり、 主に多くの関節に おける滑膜に影響し、 滑膜の炎症は軟骨破壊、 骨侵食、 ひいては関節変形及 び関節機能の喪失をもたらす。 この疾患は自己免疫性であり、 滑膜表層及び 関節周辺腔液への T細胞、 B細胞、 マクロファージ及び好中球の浸潤を特徴 とする。
[0003] 本発明者等は、 以前に胚の遺伝子操作技術を用いて関節リゥマチについて の 2種類のマウスモデルを確立した。 これらは、 ヒト T細胞白血病ウィルス I型トランスジエニック (HTLV-卜 Tg) マウス (特許文献 1 ) 及び I L - 1受 容体アンタゴニスト欠損(I L-1 Ra-/_)マウス (特許文献 2 ) であり、 自己免疫と 関節炎を自発的に発症する。 これらの動物における感染した関節の病理組織 はヒトの関節リウマチに極めて類似していた。 本発明者等は、 マイクロアレ ィを用いてこれら 2つの関節炎モデルマウスの遺伝子発現を包括的に解析し たところ、 HTLV- I Tgマウスと I L_1 Ra+マウスとの間で遺伝子発現に強い相関 があることを見いだした。 即ち、 これら 2つのマウスモデルは病因学的に明 らかに相違しているにもかかわらず、 組織病理学的のみならず遺伝子発現パ ターンも互いに類似していた。

[0005] 樹状細胞 (DG)は主要な抗原提示細胞 (APG)であり、 免疫系の調節において 中枢的な役割を担う。 近年、 幾つかの GLR類が樹状細胞の表面に発現されると のキャラクタリゼ一シヨンがなされた。 I型の GLR類メンバ一である MMR (CD2 06)及び DEG-205 (GD205)は、 その N末端に複数のカルシウム依存性細胞外糖鎖 認識ドメイン (GRD)を有する。 樹状細胞上に発現する GLRの第 2のファミリ一 は、 G末端に唯一の GRDを持つ I I型タンパク質であり、 それらは DG-SIGN (CD 209) 、 Langerin (GD207)、 GLEG- 1、 Dectin-1 (S_GR) 、 Dectin- 2、 DLEG、 及 び DGIRを含む。

発明が解決しょうとする課題
[0008] 即ち、 本発明は、 自己免疫疾患である関節リウマチについて確立された 2 つのマウスモデルにおいて共通して発現が亢進された樹状細胞免疫受容体 (DG IR)に着目してなされたものであり、 関節炎を含む自己免疫疾患の発症におけ る DGIRの役割を解明すべく DGIR欠損(K0)マウス (DGIR-/-マウス) を作製し、 該欠損マウスの疾患動物モデルとしての有用性を実証し、 当該欠損マウスを 用いた自己免疫疾患の治療あるいは予防に有効な薬剤のスクリーニング法を 提供し、 さらには当該スクリーニング法によって同定されうる自己免疫疾患 の治療■予防剤を提供することを目的とする。

本発明に係る DGI R欠損 (K0)マウス (DGI R+マウスと略記) では、 コラーゲ ン誘導関節炎 (GI A)に対する感受性が亢進しており、 当該マウスは関節炎を 含む自己免疫疾患の疾患モデル動物として有用である。 また、 加齢した DGI R 欠損マウスが、 腱、 靱帯と骨の付着部における炎症 (付着部炎) や唾液腺炎 等の自己免疫様疾患を自発的に発症することから、 免疫系における DGI Rの調 節的役割が初めて明らかにされ、 本発明の疾患モデルマウスは、 関節リウマ チ (RA)、 強直性脊椎炎 (AS)、 特発性全身性骨硬化症 (D I SH) , あるいはシェ グレン症候群 (SS)等の自己免疫疾患モデルなどとして極めて有用である。 また、 本発明の疾患モデル動物は、 野生型に比較して踵関節における石灰 化領域の増加及び大腿骨における骨量の減少が見られるので、 骨疾患のモデ ル動物としても使用しうる。
さらに、 DGI Rを欠損した本発明のモデル動物においては抗腫瘍免疫効果の 向上が観察された。 DGI Rは樹状細胞の抑制性の機能調節に関与していること から、 DGI Rタンパク質又はその等価物は、 ガン免疫療法における賦活剤とな りうる。
本発明のモデル動物を用いることによって、 DGIRの生理学的機能の解析、 自己免疫発症機構の解析が可能になる。 また、 RAや AS、 DISHあるいは骨粗鬆 症といった骨■軟骨疾患の発症メカニズムを解明し、 より有効な治療/診断 の方法及び薬剤を開発することができる。

発明を実施するための最良の形態
本発明の疾患モデル動物は、 齧歯目動物、 中でもマウスであるのが好まし し、。 当該モデル動物では、 関節炎等の自己免疫疾患が自然発症するため、 自
己免疫疾患のモデル動物として有用である。 本発明のモデル動物を用いて研 究できる自己免疫疾患としては、 例えば、 関節リウマチ (RA)、 強直性脊椎炎 (AS) , シエグレン症候群 (SS)、 又は特発性全身性骨硬化症 (D I SH)等が挙げ られるが、 これらに限られるわけではない。 また、 前記 ASや D I SH等の自己免 疫性骨疾患のみならず、 骨粗鬆症等の代謝関連骨疾患のモデルとしても使用 できる。
[0014] 本発明のスクリーニング方法は、 例えば、 前記の疾患モデル動物に被検物 質を投与し、 又は、 当該動物由来の組織、 器官、 もしくは細胞を被検物質と 接触させ、 当該動物又は組織、 器官もしくは細胞における骨髄幹細胞由来細 胞の分化及び増殖を測定及び評価することを含み、 骨髄幹細胞由来細胞の分 化及び増殖促進又は抑制する物質を同定することができる。 あるいは、 例え ば、 前記の疾患モデル動物に被検物質を投与し、 コラーゲン誘導関節炎の発 症率及び関節炎スコアを測定、 評価すること含み、 関節炎の発症抑制物質を 同定することができる。
[0015] 本発明のスクリーニング方法は、 前記疾患モデル動物で得られた測定及び 評価結果を野生型の同種動物で得られた結果と比較することをさらに含むの が好ましい。
前記のスクリーニング方法を実施することにより、 関節リウマチ (RA)、 強 直性脊椎炎 (AS)、 乾癬性関節炎 (PsA)、 シエグレン症候群 (SS)、 及び特発 性全身性骨硬化症 (D I SH)を含む自己免疫疾患又は骨粗鬆症の予防及び治療に 有効な物質を同定することができる。
[0016] また、 樹状細胞免疫受容体 (DGI R)遺伝子を欠損した本発明の疾患モデル動 物において自己免疫疾患が自然発症又は増悪化したことから、 当該疾患の予 防又は治療において DGI Rタンパク質が有効であることが予測される。 従って 、 本発明は、 樹状細胞免疫受容体 (DGI R)タンパク質をコードする遺伝子の発 現を促進する物質を含有してなる自己免疫疾患又は骨粗鬆症の予防■治療剤 を提供する。
さらに、 樹状細胞免疫受容体 (DGI R)タンパク質を欠損した動物において自
己免疫疾患の発症がみられることから、 当該 DGI Rタンパク質をコードする遺 伝子又はその部分配列をコードする DNAは自己免疫疾患又は骨粗鬆症の診断薬 として有効である。 即ち、 本発明は、 当該 DNAを含有する自己免疫疾患又は骨 粗鬆症の診断薬も提供する。
[0017] また、 樹状細胞免疫受容体 (DGI R)タンパク質と同一又は実質的に同一のァ ミノ酸配列を有するぺプチドもしくはその部分べプチドまたはその塩は、 候 補物質とともに培養して、 当該候補物質の有無における DGI Rタンパク質の活 性を比較することにより、 DGI Rタンパク質の活性を促進する物質、 即ち自己 免疫疾患又は骨粗鬆症の予防■治療剤をスクリーニングすることができ、 本 発明はそのためのキットを提供する。
[0018] さらに、 樹状細胞免疫受容体 (DGI R)タンパク質をコードする遺伝子と同一 又は実質的に同一の塩基配列またはその部分配列をコードする DNAをプローブ として使用し、 DGI Rをコードする DNA の有無あるいはその発現量などを測定 することにより、 自己免疫疾患又は骨粗鬆症に対する易罹患性を診断するこ とができ、 本発明はそのためのキットを提供する。
[0019] 本発明は、 樹状細胞免疫受容体 (DGI R) が自己免疫疾患の発症ゃ增悪に関 連していることを初めて明らかにした。 即ち、 樹状細胞免疫受容体 (DGI R) は免疫系の抑制性調節因子の一つであり、 生理状態において免疫反応を抑制 する役割を果たしている。 従って、 逆に樹状細胞免疫受容体 (DGI R) タンパ ク質 (特に可溶性細胞外蛋白部分) と同一又は実質的に同一のアミノ酸配列 を有するぺプチドもしくはその部分べプチドまたはその塩を投与することに より、 免疫系の抑制シグナルが除かれ、 免疫系が活性化されることが期待さ れる。 この結果、 種々のウィルス、 細菌、 カビ、 原虫などの感染症の治療薬 としてこれらを用いることができる。
また、 近年、 ガン免疫療法において、 樹状細胞に腫瘍細胞や抗原を体外で 取り込ませ活性化■成熟させた状態で体内に投与する方法が効果的な免疫反 応の惹起につながるとして注目を集めており、 この方法は自己の樹状細胞を 用いるために副作用の危険性が低下するという利点もある。 従って、 DGI Aタ
ンパク質のぺプチド又はその等価物類はガン免疫療法における賦活剤として も作用し得る。
実施例
[0020] 以下、 具体例に則して本発明を詳細に説明するが、 本発明はこれら具体例 に限定されるものではない。
実験方法
マウス:
遺伝子発現プロフアイリング実験には、 既述した HTLV-卜 Tgマウス及び I L-1 Ra- の 2つのマウスモデルを用いた。 HTLV-卜 Tgマウスは、 HTLV- I ゲノムの LT R-env-pX-LTR領域を(G3H/Hen x C57BL/6J) ド,胚に注入することにより作製し 、 BALB/cAマウス(GLEA Japan. , Tokyo, Japan)に 20世代戻し交配した。 これ らのマウスは 4週齢で関節炎を自然発症し、 3ヶ月齢で 60%、 6ヶ月齢で 80%が発 症する。 以下の実施例では、 重篤な関節炎 (スコア 3) を発症した HTLV -Tg (TS) (雌、 6〜9週齢)を用いた。 I L-1 Ra-/_マウスは、 相同組換えにより作製した I L_1 Ra+マウスを BALB/cA マウスに 8世代戻し交配した。 これらのマウスは 5週 齢で関節炎を自然発症し、 8週齢及び 13週齢までに、 各々 80% 及び約 100%のマ ウスが発症する。 以下の実施例では、 重篤な関節炎 (スコア 3) を発症した I L _1 Ra-/_マウス(KS) (雄、 13 週齢)を用いた。 また、 野生型同腹子 (WT)をコント ロールとして用いた。
[0021 ] 関節炎の重篤度は、 各足について、 発赤及び腫脹の程度を次のように 0から 3のスコアでランク付けした:スコア 0 =正常、 スコア 1 =関節の軽い腫脹及び /又は足躕の発赤、 スコア 2 =顕著な関節腫脹、 スコア 3 =関節の重篤な腫脹 及び強直。 なお、 全てのマウスは、 東京大学医科学研究所のクリーンルーム で、 特定病原体未感染 (SPF) 環境下で飼育した。 また、 全ての実験は動物実 験に関する倫理ガイ ドライン及び遺伝子操作に関する安全基準に従って実施 した。
[0022] ノ一ザンブロットハイブリダイゼ一ション解析:
関節組織を即座に液体窒素中で凍結し、 -80°Cで保存した。 凍結組織は phys
cotron (Microtech, Chiba, Japan)でホモジナイズし、 グァニジンチオシァ ネ一ト-フエノール -クロロホルム抽出法により全 RNAを調整し、 オリゴ (dT)_ セルロースカラム用いて poly (A)+ RNA を精製した。 各標本について 4、 5匹の マウスの R N Aをプールした。 Poly (A)+ RNA を 1.3%変性ァガロースゲルで 電気泳動し、 ナイロン膜 (Gene Screen Plus, NEN Life Science, Boston, M A, USA) に転写した。 ハイブリダィゼ一シヨンは、 メガプライム DNAラベリン グシステム(Amersham, Arlington Heights, IL, USA) 及び32 P-dGTP (3,000 Ci/ mmol; NEN Life Science, Boston, MA, USA)を用いて標識した32 P-標識 DNAプ ローブで 42°C、 一晩のプロ トコルで実施した。 放射活性は、 BAS-2000システ ム(Fuji Photo Film Co. , Tokyo, Japan)を用いて測定した。 マウス DGIRプロ —ブは、 関節炎を発症した HTLV- 1 _Tgマウスの関節由来 c DNAから PGRにより増 幅した。 PGRプライマ一は次のものを使用した。

DGIRを含むゲノム DNAの配列を、 Celera Genomics (Rockvi Me, MD, USA) のマウスゲノムデータベースから得、 5.5-kbフラグメントからなる 5' ホモ口 ジ一領域及び 2.5-kbフラグメントからなる 3' ホモロジ一領域を ES細胞 (E14. 1) 由来のゲノム DNAから PGRによって増幅した。 PGRプライマ一の組は次の通 りである。

[0024] ターゲテイングベクターは、 ITIMを含む細胞内ドメイン及び膜貫通領域を
含有する第 1及び第 2ェキソンをコードするゲノムフラグメントを、 PGK1プロ モーター制御下にネオマイシン耐性遺伝子 (Neo) を発現するフラグメントで 置換することにより構築した。 また、 ネガティブセレクションのために MG1プ 口モータ一制御下にジフテリア毒素 (DT) を発現するフラグメントを 3' 末端 に連結した。 タ一ゲティングベクタ一を ES細胞にエレク トロポレーシヨンで 導入し、 G418で選択した。 672個のネオマイシン耐性 ESクローンをピックアツ プし、 5' プローブを用いたサザンプロットハイブリダィゼ一シヨン解析によ つて 660クローンをスクリ一ニングした。
スクリーニングに用いた 5' プローブは以下のプライマーで増幅した。

サザンブロットハイブリダイゼ一ション解析によって 96. 2%のク口一ンが判 定され、 2つのターゲティング ESクローンを同定した (ターゲティング効率 : 0. 3%) 。 これらのタ一ゲッティングクローンは 3' 及び Neoプローブにより 確認した。
3' プローブは以下のプライマーで増幅した。

Neoプローブはネオマイシン耐性遺伝子の Nar卜 Xba l フラグメントとした。 核 型分析の結果、 両クローンの約 80%の細胞は正常な 40染色体を有していた。 キ メラマウスを凝集法によって作製し、 雄のキメラマウスを野生型の G57BL/6J 雌マウスと交配させ、 被毛色によって生殖細胞系列への伝播を判定した。 DGI
R変異についてのヘテロ接合マウス同士を交配し、 同型接合マウスを産出した
。 以下の実施例では (129/Sv X C57BL/6) F,遺伝背景の DGI R欠損マウス及び同 腹子を用いた。

[0026] 組織病理検査:
12ヶ月齢の野生型マウス及び DGIR欠損マウスをエーテル麻酔し、 中性緩衝 1 0%ホルマリン溶液で還流固定した。 脳、 心臓、 大動脈、 肺、 気管、 塍臓、 肝 臓、 脾臓、 腋窩リンパ節、 顎下腺及び耳下腺、 腸、 胃、 腎臓、 及び生殖器を 含む主要な器官及び組織を取り出し、 組織病理学的検査に供した。 足、 膝、 手首及び胸椎を含む関節組織は 10%蟻酸を用いて脱灰してからパラフィン包埋 した。 各組織について 2~3國の切片を作製し、 へマトキシリン及びェォジン を用いて染色した。

各細胞を F I TG、 PE又はビォチン修飾の下記モノクロ一ナル抗体 (mAb)で染色 し、 フローサイ トメ トリ一解析を行った。 GD11c (HL3)、 l-Ab (AF6-120.1), C D3 (145-2C11), CD45R/B220 (RA3- 6B2)、 CD4 (RM4-5) 、 CD8 (53-6.7) 、 CD6 2L (MEL-14) 、 CD44 (IM-7) に特異的なハムスター、 マウス又はラットの mAb は、 BD PharMingen (San Diego, CA, USA)から購入した。 マウスの GD80 (RMM P2)、 CD86 (RMMP1) に特異的なラット mAbは Immunotech (Marseille, France) から購入した。 PE修飾ストレブトアビジン(PharMingen)はビォチン修飾抗体 の二次染色に用いた。 細胞表面の染色は標準的なプロ トコルに従って行い、 解析は FAGSGal ibur 及び Gel I Quest (Becton Dickenson, Franklin Lakes, NJ , USA)または FlowJo (Tree Star, Inc. , Ashland, OR) ソフトウェアを用い て行った。

抗原特異的な応答を調べるために、 野生型マウスから Thy1.2陽性 T 細胞を 調整し、 野生型マウス及び DGIR欠損マウスの脾臓から GD11c陽性樹状細胞を、 CD90 (Thy1.2)マイクロビーズ又は GD11cマイクロビーズ(Miは enyiBiotech, B ergisch Gladbach, Germany)を用いた磁気細胞分離により精製した。 精製し た Thy 1.2陽性 T 細胞(2x105 eel Is)及び GD11c陽性樹状細胞(2 x104 eel Is)を 、 抗 GD3 抗体(145-2G11 ; BD PharMingen)を添加して 3日間共培養し、 トリチ —ムチミジン (0.25 ; uGi/mに Amersham, Buckinghamshire, England)を 6時間 取り込ませた。 次に、 細胞は Micro 96 cel l harvester (Skatron, Lier, Norw ay)ガラスファイバ一濾紙上に回収し、 Micro Beta (Pharmacia Biotech, Pis cataway, NJ)を用いてトリチ一ムチミジン放射活性を測定した。
同種異系混合白血球反応を調べるために、 BALB/cマウスから GD90 (Thy1.2 ) マイクロビーズを用いて Thy1.2陽性 T細胞を磁気細胞ソーティングにより精 製した。 また、 野生型マウス及び DGIR欠損マウスの骨髄細胞から樹状細胞を 誘導した。 樹状細胞はマイ トマイシン G処理してから、 Thy1.2陽性 T細胞と 3日 間共培養し、 トリチームチミジンを 6時間取り込ませてその放射活性を測定し た。
[0029] コラーゲン誘導関節炎:
5 mg/ml熱殺傷 M. tuberculosis (H37RA; Difco) を追加したフロイント完全 アジュバント (Difco Laboratories, Detroit, Ml) に 1 mg/ml トリ I I型コ ラ一ゲン (I IG; Sigma-Aldrich, St. Louis, M0) を懸濁し、 マウス尾底部の 皮下複数箇所に免疫した。 関節炎の誘導は追加免疫なしで実施した。 各関節 は、 数日ごとに腫脹及び発赤について検査し、 各手足について重篤度を 0~3に ランク付けした (スコア 0 =変化なし;スコア 1 =関節の軽い腫脹及び/又は 足躕の発赤;スコア 2 =関節の顕著な腫脹;スコア 3 =関節の重篤な腫脹及び 強直) 。 関節炎の組織学的評価のために、 関節組織を 10%中性緩衝ホルマリン 溶液で固定し、 5%蟻酸で脱灰後パラフィン包埋した。 各組織について 4 國の 切片を作製し、 へマトキシリン及びェォジンを用いて染色した。

コラーゲン特異的抗体の測定には 10 mg/ml トリ I IGを、 血清中の抗体レベル の測定には、 ゥサギ抗マウス IgM (2 mg /ml; Zymed) 、 IgG (8.7 mg /ml; Zy med) 、 または IgE (2 mg /ml; PharMingen) を、 自己抗体の測定には、 熱変 性ゥサギ lgM、 IgG (50 mg /ml; Zymed) 、 またはトリ I IG (20 mg/ml; Sigma ) をそれぞれ Falcon 3912 Micro Test 111フレキシブルアツセィプレート (B D Bioscience, Oxnard, CA) に 4°Cで一晚コ一ティングした。 PBSで洗浄後、 希釈した血清サンプルを室温で 1時間ィンキュベ一トした。 PBS-Tweenで洗浄 後、 アルカリホスファタ一ゼ (AP) 修飾ャギ抗マウス lgM、 IgG, IgE, IgGU lgG2a、 lgG2b及び lgG4を室温で 1時間インキュベートし、 AP活性を Substrate Phosphatase SI GMA104 (Sigma-Aldr ich) を用いて ELISAマイクロリーダ一 (M TP- 120; Colona, Tokyo, Japan) により測定した。 抗核抗体の測定のために は、 マウス核抗原コ一ティングプレート (Alpha Diagnostic, Inc. , San Ant onio, TX) を用いて希釈したマウス血清をインキュベートし、 西洋わさびべ ルォキシダーゼ (HRP) 修飾ャギ抗マウス IgGを反応させた後、 TMB基質を用い て HRP活性を測定した。
[0031] コラーゲン特異的増殖応答及びサイ トカイン産生:
I IG/GFA免疫 7日後にリンパ節を回収し、 単一細胞懸濁液を調製した。 リン パ節細胞を熱変性 I IGの非存在下あるいは存在下で 3日間培養し、 トリチーム チミジン (0.25 mCi/ml) を 6時間取り込ませてトリチームチミジン放射活性 を測定した。 サイ ト力イン産生の測定には、 コラーゲン特異的増殖応答にお ける培養上清を 3日後に収集し、 BD OptiEIA EL ISA Set (BD PharMingen) を 用いて IFN-g、 IL-4、 及び IL-10レベルを測定した。
[0032] 骨髄細胞由来樹状細胞の調製:
骨髄細胞由来樹状細胞は大腿骨及び脛骨の骨髄から調製した。 簡潔には、 2 x105 eel Is/mlの骨髄細胞を 20 ng/mlの組換えマウス GM-GSF (PeproTech, Lon don, UK) を添加した 10% FCS- RPM卜 1640で培養した。 3曰目に 20 ng/mlの GM- GSFを含む倍量の新鮮な培地を加え、 6日及び 8日目に、 培地上清の半分を回収
して遠心分離し、 細胞を 10 ng/mlの GM-GSFを含む新鮮な培地に再懸濁し、 元 のプレー卜に戻した。 10曰目の非接着性細胞を骨髄細胞由来樹状細胞とした 。 さらに、 骨髄細胞由来樹状細胞の完全な成熟のために、 10日目の非接着性 細胞を回収し、 10 ng/mlの GM-GSF及び 1 mg/ml LPS (Sigma) を含有する新鮮 培地中に再懸濁した。 この細胞を更に 2日間培養し、 成熟樹状細胞とした。
[0033] ウェスタンブロット解析:
骨髄細胞は大腿骨及び脛骨から調整し、 20 ng/ml 組換えマウス GM-GSFを添 加した無血清 RPM卜 1640培地で一晚培養した。 これらの細胞を洗浄し、 GM-GSF を添加しない無血清培地に再懸濁した。 6時間飢餓状態にした後、 様々な濃 度の GM-GSFとフォスファタ一ゼ阻害剤カクテル 11 (Sigma) で処理し、 全細胞 溶解液を抗リン酸化 STAT5抗体 (Cel l Signal ing, Denvers, MA) および抗 STA T5抗体 (Santa Cruz Biotechnology, Inc. , Santa Cruz, CA) を用しゝた免疫 ブロッテイングにより解析した。 検出抗体は西洋わさびペルォキシダーゼ (H RP) 修飾抗ゥサギ IgG (Cel l Signal ing) を用いた。 蛍光シグナルは EGLブラ ス検出溶液と Typhoon 9000 (Amersham Biosciences, Buckinghamshire, UK) を用いて検出した。

関節リウマチ(RA)関連遺伝子としての DC I Rの同定:
2つのモデルマウス(HTLV- 1 Tg及び I L_1 Ra- マウス)の関節における遺伝子 発現を正常マウス関節と比較して、 関節炎を発症した関節において発現が亢 進する遺伝子を特定した。 マイクロアレイ解析によると、 DGIR遺伝子の mRNA 発現は、 野生型関節に比較して IL-1Ra-/_マウスでは 2.2倍、 そして HTLV-I Tgマ ウスでは 3.8倍に亢進し (図 1 a) 、 この発現亢進はノーザンプロットハイブ リダイゼ一ション解析でも確認された(図 1 b ) 。

DGIR欠損マウスの作製:
DGIR欠損マウス (DGIR-/-マウス) は、 通常の遺伝子タ一ゲティング法によつ て作製した。 DGIR遺伝子のェキソン 1及び 2をネオマイシン耐性遺伝子で置換 することにより、 IT I Mを含む細胞質領域及び膜貫通ドメインの大部分をコ一 ドするゲノム配列を欠損させた (図 2 a) 。 5' プローブを用いたサザンプロ ットハイプリダイゼ一シヨン解析によってタ一ゲッティング ESク口一ンをス クリーニングし (図 2 b) 、 3' プローブ (図 2 c) 及び Neoプローブ (図 2 d ) を用いてタ一ゲッティングァリルを確認した。 マウスにおける DGIR領域 のタ一ゲティングは、 ゲノムサザンプロット解析によって確認した (図 2 e ) 。 DGIR マウスの交配により DGIR- マウスを作製し、 ノーザンプロットハイ ブリダィゼ一シヨン解析によって脾臓における DGIR mRNAの発現が欠損してい ることを確認した (図 2 f ) 。
[0037] DGIR- マウスは繁殖力があり、 メンデル比率で産仔し、 特定病原体未感染の 飼育条件下では若年においては明確な表現型異常を示さなかった。 また、 DGI R-/-マウスの胸腺及び脾臓の細胞においても明らかな異常は見られなかった ( 図 3) 。 T細胞反応に対する DGIR欠損の影響を評価するために、 樹状細胞存在 下において抗 GD3抗体で刺激した際の T細胞の増殖反応及び同種異系混合リン パ球反応 (MLR) を測定した。 この結果、 DGIR欠損樹状細胞と野生型樹状細胞 との間に有意な差は検出されなかった (図 4) 。

加齢 DG I R欠損マウスにおける自己免疫疾患の自然発症:
DGIR欠損マウスを組織学的に検査した結果、 6-12ヶ月齢の DGIR欠損マウス 1 0匹ののうち 7匹の顎下腺に、 唾液腺間質へのリンパ球の蓄積及び小導管上皮 への単核細胞の浸潤、 それに伴う小導管の破壊を特徴とする唾液腺炎が観察 された (表 1 ) 。
[0039] また、 4ヶ月齢において 11%の DGIR欠損マウスが後肢の関節異常を発症した 。 発赤及び複数関節の腫脹を伴う関節炎に始まり、 6ヶ月齢までに 28%のマウ
スが関節炎を発症した。 関節炎の発症率、 重症度は加齢とともに増大し、 関 節の変形及び強直を引き起こした。 その殆どは後肢の足関節で顕著であった
。 発症には性差があり、 12ヶ月齢において 44%の雄マウスで発症したが、 雌 マウスでは 6%であった。 関節の組織学的解析により、 DGIR欠損マウスにおい て腱及び靱帯と骨との付着部への炎症性細胞の浸潤、 顆粒化組織の侵襲によ る骨破壊が観察された。 付着部炎は、 検査した 23匹の DGIR欠損マウスのうち 9 匹で観察された (表 1 ) 。 これらの結果から、 DGIR遺伝子の欠損が自己免疫 を誘導し、 遅始性関節炎及び唾液腺炎を自然発症させることが示された。

DGIR欠損マウスにおける自己免疫の自然発症:
DGIR欠損が自己免疫の発症に影響を与えることが示唆されたので、 DGIR欠 損マウスにおける自己抗体の産生を ELISAで測定した。 血清中の lgM、 IgG, lg E及び以下の自己抗体、 抗核抗体 (ANA) 、 リウマチ因子 (RF ; IgM及び IgG型 ) 、 及び抗 IIG IgGの産生を測定した (図 5) 。 12ヶ月齢の DGIR欠損マウスの 血清免疫グロブリンレベルに増加傾向が見られたが、 IgEレベル以外に有意差 はなかった。 しかしながら、 加齢 DGIR欠損マウスは抗核抗体 (ANA) 、 リウマ チ因子 (RF) 、 及び抗 IIG抗体などの自己抗体を野生型マウスと比較して高い レベルで産生した。 この結果は DGIR欠損が自己抗体の産生に関与しているこ とを示唆している。

DC I R欠損マウスにおける樹状細胞の増殖と GD4陽性 T細胞の活性化:
DGIR欠損マウスにおいて DGIR発現細胞である GD11c陽性細胞の割合を調べた 結果、 GD11c陽性細胞の割合は若年の生理的条件下では正常であつたが、 12月 齢以上では顕著に増加していた。 また、 GD11c陽性細胞の大部分は卜 Abを発現 していた。 さらに、 DGIR欠損マウスの GD4陽性 T細胞は、 活性化 T細胞の特徴で ある GD62Lの発現が低く、 GD44の発現が高い表現型を示した。 GD8陽性 T細胞で は顕著な変化は見られなかった (図 6) 。

DGIR欠損マウスにおけるコラーゲン誘導関節炎の増悪化:
本発明の関節リウマチのモデルマウスにおいて DG I Rの発現が亢進していた ので、 コラーゲン誘導関節炎 (GIA) の発症に対する DGIR欠損の影響を試験し た。 GIAは (129/Sv X C57BL/6) F,雑種背景の若年 DGIR欠損マウスに、 250 mg の熱殺傷 Mycobacterium tuberculosisを添加したフロイント完全アジュバン 卜と 100 mgのトリ I I型コラーゲン (I IG) を免疫することにより誘導した。 通 常の免疫手法では、 マウスは初回免疫から 21日後に I IG/GFAで追加免疫を行う 。 し力、し、 今回の実験では初回免疫から 20日後に、 同腹子の 0%に対して、 DGI R欠損マウスでは 39%に疾患が発症した (図 7) 。 追加免疫の後では、 DGIR欠 損マウスにおける発症率の増大を観察することが困難になることが示唆され たので、 追加免疫を行わずに経過観察を行った。 図 7に示すように、 DGIR欠 損マウスにおける関節炎発症率は、 コントロールの同腹子と比較して顕著に 増大した。 また、 重症度も野生型マウスに比較して有意に増悪化した。 各群 の発症率及び重症度は独立した 2回の実験データの合計である。
[0044] GIAの組織学的解析において、 コントロールマウスの関節では極めて少量の 細胞浸潤のみを示し、 滑膜の増殖や骨破壊は観察されなかった。 それに対し 、 DGIR欠損マウスでは、 このようなマイルドな免疫条件下でも炎症性細胞の 浸潤、 滑膜の増殖、 及び骨破壊を伴う典型的な関節炎を示した。 この結果は 、 DC I Rが G I Aの発症を抑制することを示唆している。

次に、 I IGを免疫した DGIR欠損マウスにおける I IG特異的抗体の産生を測定 した。 I IGに対する抗体レベルは関節炎の発症とよく相関することが知られて いる。 図 7 a、 bに示すように、 I IGに特異的な lgG、 lgG1及び lgG3レベルは 、 野生型同腹子に比較して DGIR欠損マウスで有意に増大した。 一方、 DGIR欠 損マウスにおける lgM、 lgG2a及び lgG2bレベルは正常であった。 即ち DGIRは GI Aにおける抗原特異的な lgG1及び lgG3サブクラスの抗体産生に関与していると 考えられる。

DC I R欠損マウスにおける T細胞の抗原特異的増殖反応の亢進:
次に、 DGIR欠損マウス及びそれらのコントロールである同腹子からの T細胞 の抗原特異的増殖反応を試験した。 I IGに対する T細胞の反応は、 トリ I IG/GF Aでの初回免疫から 7日後に測定した。 DGIR欠損マウスにおける抗原特異的 T 細胞の増殖反応は野生型マウスと比較して有意に増大し (図 7 c) 、 DGIR欠 損マウスにおいて T細胞のプライミングが促進していることが示唆された。 I I G刺激後のリンパ節細胞からの IFN-g産生は、 DGIR欠損マウス及びコント口一 ルマウスで同様に観察された。 これに対して、 IL-4 及び IL-10 レベルは DGIR 欠損リンパ節細胞で有意に増大した (図 7 d ) 。 これらの結果は、 DGIR欠損 マウスにおいて Th2サイ トカイン産生が選択的に促進したことを示し、 I IG特 異的な lgG1及び lgG3サブクラス抗体の産生が促進した観察結果と合致するも のである。

DGIR欠損骨髄細胞 (BMG) の GM-GSFに対する反応性の亢進:
DGIRは主に樹状細胞に発現するので、 DGIR欠損骨髄細胞の樹状細胞への分 化を評価した。 DGIR欠損骨髄細胞を GM-GSFを添加して培養した場合、 非接着 性骨髄由来細胞の分化は、 DGIR欠損骨髄細胞において有意に亢進した (図 1 O a) 。 培養 8日目の非接着細胞のフローサイ トメ トリ一解析により、 GD11C 陽性骨髄細胞由来樹状細胞 (BMDG) が DGIR欠損骨髄細胞の培養において顕著 に増加することが示された (図 1 0 b) 。 これらの GD11c陽性細胞の殆どは卜 Abを発現したが、 活性化マーカーである GD80及び GD86 は発現していなかった 。 培養 10日目の BMDGを LPSで 48時間パルスすると、 DGIR欠損マウス及び野生型 マウスの樹状細胞は卜 Ab、 GD80及び GD86を高発現し、 同レベルの成熟が観察さ れた (図 1 0 c) 。
[0049] 次に、 DGIR欠損マウスの骨髄細胞における GM-GSF刺激による STAT5のリン酸 化を解析した。 図 1 O dに示すように、 GM-GSFによって誘導される STAT5の活 性化は、 DGIR欠損骨髄細胞において顕著に亢進しており、 DGIR欠損骨髄細胞 の GM-GSFに対する反応性の亢進と合致した。 したがって、 DGIR欠損骨髄細胞 の in vitroにおける樹状細胞分化の亢進は骨髄細胞の GM-GSFシグナルに対す る感受性が増大したことによると考えられる。 この観察結果と一致して、 in vivoにおける樹状細胞の割合も、 生理的条件下の若年マウスでは正常である 力 免疫や加齢に伴い増加している。
[0050] 以上詳細に述べたように、 本発明では、 DGIR欠損 (DGIR-/-) マウスを作製し、 DGI Rが自己免疫性唾液腺炎および関節症に関与することを示した。
[0051] 本発明によって、 加齢 DGIR欠損マウスが特徴的な自己免疫疾患を自然発症 することが示された。 12ヶ月齢において、 DGIR欠損マウスの血清免疫グロブ リンレベルが増加し、 野生型マウスと比較して抗核抗体、 リウマチ因子及び 抗 IIG抗体などの自己抗体を高レベルで産生した。 また、 組織学的解析により DGIR欠損マウスの唾液腺及び付着部における病理学的変化が観察された。 唾 液腺へのリンパ球浸潤は、 唾液腺が標的となる代表的な慢性自己免疫疾患で あるシエグレン症候群 (SS) の典型的な兆候である。 幾つかのマウス系統、 N
0D、 MRL/ l pr及び NZB/W Πは、 SSのモデルとして広く受け入れられている。 関 節リウマチ、 複合関節組織疾患 (MGTD) 、 強直性脊椎炎 (AS) 及び脊椎関節 症 (SpA) を含む他の自己免疫疾患を持つ患者では限局性唾液腺炎も高い割合 で見られ、 その発症はリウマチ因子の産生に深く関わっている。 これらの結 果は、 DGI Rが免疫系の恒常性の維持に重要であり、 DGI Rの欠損が自己免疫の 発症を引き起こすことを示唆している。
[0052] 加齢 DGI R欠損マウスでは、 遅発性の強直性関節症の自然発症も観察された 。 これらのマウスの組織病理学的解析により、 炎症性細胞の浸潤を伴う骨及 び付着部の浸食性の破壊が観察された。 しかしながら、 DGI R欠損マウスの病 理組織は、 HTLV-卜 Tgマウス、 I L_1 Ra_K0マウス、 及び GI Aマウス等の他の関節 リウマチモデルマウスの病理像とは明らかに相違していた。 ヒ卜の関節リウ マチと類似する特徴的な滑膜炎を発症する他のモデルマウスとは異なり、 DGI R欠損マウスでは付着部炎が第一の異常であり、 滑膜炎は希に観察されるだけ であった。 滑膜炎が関節リウマチの特徴であるのに対し、 付着部炎は、 強直 性脊椎炎 (AS) 、 脊椎関節症 (SpA) 及び乾癬性関節炎 (PsA) 等の炎症性リ ゥマチ疾患群に特徴的な兆候である。 したがって、 DGI Rの欠損が AS、 SpA 及 び PsAに類似した特徴的な炎症性関節炎の発症を引き起こすことが示唆された
[0053] 本発明では、 DGI R欠損マウスにおいて GI Aが増悪化したことが示された。 I I 型コラーゲンで免疫した後、 DGI R欠損マウスの血清中の抗原特異的 l gG1及び I gG3のレベルが非常に増加した。 また、 抗原特異的な T細胞応答が亢進し、 DGI R欠損マウスにおいて I I Gに対する免疫応答が亢進していることが明らかとな つた。 この時、 DGI R欠損マウスにおける樹状細胞の割合は顕著に増加し、 活 性化していた。 樹状細胞の割合の増加は、 加齢した DGI R欠損マウスでも観察 されたが、 生理的条件下の若年マウスにおいては正常であった。 樹状細胞は 細胞表面の抗原を提示し、 様々な免疫調節性のサイ トカインを産生すること によって T細胞を活性化するのに重要な役割を果たしているが、 本発明によつ て得られた観察結果は、 DGI R欠損マウスにおける樹状細胞の機能亢進が自己
免疫の発症の原因となることを示唆している。
[0054] 樹状細胞は GM-GSFにより骨髄幹細胞から分化するので、 I IG/GFAによる刺激 で産生される GM-GSF力《GIAにおける DGの増殖に関与していると考えられる。 本 発明によって、 DGIR欠損による GM-GSFシグナル伝達の感受性の亢進が、 in vi troでの骨髄由来樹状細胞の増殖及び分化を促進することが示された (図 1 0 ) 。 DGIR欠損骨髄細胞における GM-GSF刺激による STAT5のリン酸化の亢進は、 DGIRが GM-GSFシグナル伝達を抑制的に調節していることを示唆している。 ヒ ト DC I Rが SHP- 1及び SHP- 2をリクルートすることが報告されている。 SHP-1は SH 2ドメイン含有チ口シン脱リン酸化酵素で GM-GSFを含むサイ トカインシグナル 伝達を負に制御する。 さらに、 SHP-1欠損の motheatenマウスは全身性の自己 免疫と重篤な炎症を示す。 マウス DGIRの ITIMが機能的であり、 そのアミノ酸 配列がヒト DGIRの ITIMと同一であることから、 マウス DGIRも SHP-1及び SHP-2 をリクルートして GM-GSFシグナル伝達を調節するものと推察される。
[0055] GIAの際、 DGIR欠損マウスでは I IG特異的な lgG1および lgG3サブクラスの抗 体産生が亢進していることが明らかとなった。 DBA/1系統のマウスではコラ一 ゲン特異的な |gG2aクラスの抗体が GIAの発症に関与することが報告されてい るが、 抗原特異的な lgG2a抗体のレベルは (129xB6) Π雑種背景の DGI R欠損マ ウスでは特に増加していなかった。 DGIR欠損マウスで IL-4、 IL-10の産生が顕 著に亢進していることを示したが、 IL-4及び IL-10は、 lgG1及び lgG3のクラス スィッチに関与することが知られている。 これらの結果から DGI Rが Th2反応の 調節に関与していることが示唆される。
[0056] 樹状細胞は Th1及び Th2エフェクター T細胞の分化に関係している。 細胞内寄 生性病原体に晒された樹状細胞は Th1反応を促進するが、 ある種の寄生虫は樹 状細胞による Th2細胞分化を促進する。 調節性 T細胞 (Treg) の分化も樹状細 胞によって調節され、 特定の病原体が Tregの産生を誘導することが知られて いる。 したがって、 樹状細胞と病原体との相互作用はエフェクター T細胞の特 定のサブセットを誘導するのに重要であり、 DGIRは特定の T細胞分化に関与す る樹状細胞受容体の一つである可能性がある。
[0057] 最近のゲノムワイ ド連鎖解析は、 多くの自己免疫疾患の感受性領域を明ら かにした。 特に、 DGIR遺伝子が存在するマウス 6F2サイ トバンド、 同一染色体 であるラット 4q42、 及びヒト 12p13領域は、 関節炎、 全身性紅斑性狼瘡 (SLE ) 、 自発性糖尿病、 ァテローム性動脈硬化症、 脳脊髄炎、 喘息又は気道反応 性、 及びアレルギーをなど幾つかの炎症性疾患と関連している。 DGIRの欠損 は自己免疫様疾患を引き起こすので、 DGIRはそのような炎症性疾患の感受性 遺伝子の一つであることも示唆された。
[0058] (実施例 1 1 )
DGIR欠損マウスにおける骨代謝の変化:
骨代謝における DGIRの機能を調べるため、 12ヶ月齢の DGIR欠損マウスを用 し、、 X線解析、 組織切片による解析、 pQGT解析を行った。
まず、 DGIR欠損マウスにおいては、 ォスのみで踵関節に強直が観察された
X線解析により当該欠損マウスの骨格を野生型マウスの骨格と比較したとこ ろ、 図 1 1に示すように野生型マウスと同様であり、 骨格形成は正常であつ た。
次いで、 欠損マウス及び野生型マウスの強直発症部 (踵) の X線解析をした ところ、 図 1 2に示すように、 欠損マウスでは踵関節部において関節破壊が 起こり、 石灰化領域の増加が見られた。
[0059] DGIR K0マウス踵関節において、 石灰化領域を調べるために Von Kossa染色 、 軟骨組織を調べるために Toluidine blue染色を行った。 その結果、 踵関節 部において関節軟骨の増殖がみられ、 その軟骨が骨に置き換わっていること が明らかになった (図 1 3) 。
次いで、 大腿骨での骨量に異常があるかどうかを明らかにするため、 12ケ 月齢の個体で pQGTを行った (図 1 4) 。 その結果、 DGIR K0マウスでは骨密度 、 骨塩量共に減少が見られ、 骨量が減少していた (図 1 5) 。
即ち、 DGIR遺伝子の機能として踵関節では軟骨細胞の増加を伴い骨、 石灰 化町域の増加が見られ、 大腿骨では骨量が減少していることが明らかになつ
た。 異常の結果から DGIRは骨代謝や軟骨細胞、 骨芽細胞あるいは破骨細胞の 分化や増殖の制御に関わっていることが示唆された。 このことから DG I Rは AS やり I SHだけでなく骨粗鬆症の良いモデルマウスになると考えられる。
[0060] DGIR欠損マウスにおける抗腫瘍免疫効果の測定:
( 1 ) 移植ガン実験における抗腫瘍免疫効果の評価
抑制性レセプターである DGIRの欠損により抗腫瘍免疫効果の向上が見られ るか否かを、 可移植性腫瘍細胞を用いて検討した。
可移植性腫瘍細胞として MethAを用いた場合、 1 x106cel l/headの MethAを DGI R欠損マウス及び野生型マウスに接種し、 腫瘍形成の差を観察した。 その結果 、 発症率に差は見られなかったものの、 腫瘍の大きさでは DGIR-/-マウスにお いて優位に抑制効果が観察された (図 1 6) 。
次いで、 可移植性腫瘍細胞株として、 繊維肉腫である BALB/3T3にガン抗原 として知られているヒト MUG1を発現させた BALB/3T3 APR-MUC1 (BALB/c背景) を用いた。 BALB/3T3 APR-MUC1 clone16は東北大学加齢研究所により供与され 、 10% FBS (SIGMA), 50Uペニシリン (明治製菓) , 50mg/mlストレブトマイシ ン (明治製菓) 500mg/ml G418 (ナカライテスク) 含有 RPM卜 1640 (SIGMA)で 培養し、 1週間に 2-3回の頻度で継代培養した。
BALB/3T3 APR-MUC1 clone16を 1 x 106cel l/head接種したところ触診できる程 度 (約 4國程度まで) の腫瘍が形成された。 この腫瘍の発症率で抗腫瘍免疫効 果を評価したところ、 DGIR-/-マウスでは腫瘍退縮が有意に促進された (図 1
[0061] (2) DGIR-/-マウスの樹状細胞における細胞傷害活性の評価
腫瘍細胞株 BALB/3T3 APR-MUC1 clone16を用いた細胞傷害性 T細胞 (GTL)の 誘導は以下のように行った。 腫瘍細胞を 8000radの r線照射により不活化し、 1 x107cel Is/headで 1日目と 1 0日目にマウス腹腔内に接種した。 免疫下 5日 後にマウスの脾臓を採取し、 脾臓細胞を調製した。 得られた脾臓細胞を 10we I Iプレー卜で不活化した腫瘍細胞と 1 x107: 1 x106の割合になるよう共培養し 、 3日間の再刺激を行った。
標的細胞を lOO ^Giクロム酸ナトリウム、 51Gr (GEヘルスケアバイオサイェ ンス) で 1時間、 37°Cで標識した後、 RPM卜 1640 (10% FBS, 10mM 2-Mercapto ethanol) に懸濁した。 再刺激した脾臓細胞から Lymphocyte-M(GEDARLANE)で リンパ球を分離し、 96wel l round-bottomプレート ( IWAKI)で標識した標的細 胞と共培養した。 4時間後、 放出された51 Grを放射能活性として Micro Bete ( Pharmacia)で検出することによって細胞傷害活性を測定した。
BALB/3T3 APR-MUC1 clone16を用いて細胞傷害活性を測定したところ、 DCIR -/-マウスで細胞傷害活性が亢進していた。 このとき、 ネガティブコント口一 ルの細胞として用いた MethAに対する傷害活性はみられなかった (図 1 8) 。 従って、 DGIR- -マウスにおいて、 特異的な細胞傷害活性が有意に亢進してい ることがわかった。
Claims
請求の範囲
[I ] 樹状細胞免疫受容体 (DG I R)タンパク質をコードする遺伝子の一部又は全部が 染色体上で欠損したことを特徴とする非ヒト疾患モデル動物。
[2] 齧歯目動物であることを特徴とする請求項 1に記載の疾患モデル動物。
[3] マウスであることを特徴とする請求項 2に記載の疾患モデル動物。
[4] 疾患が自己免疫疾患であることを特徴とする請求項 1から 3のいずれかに記 載の疾患モデル動物。
[5] 疾患が、 関節リウマチ(RA)、 強直性脊椎炎 (AS)、 乾癬性関節炎 (PsA)、 シェグ レン症候群 (SS)、 又は特発性全身性骨硬化症 (D I SH)であることを特徴とする 請求項 4に記載の疾患モデル動物。
[6] 疾患が骨粗鬆症であることを特徴とする請求項 1から 3のいずれかに記載の 疾患モデル動物。
[7] 請求項 1から 6のいずれかに記載の疾患モデル動物に被検物質を投与し、 又 は、 該動物由来の組織、 器官、 もしくは細胞を被検物質と接触させ、 当該動 物又は組織、 器官もしくは細胞における骨髄幹細胞由来細胞の分化及び増殖 を測定及び評価することを含む、 骨髄幹細胞由来細胞の分化及び増殖促進又 は抑制する物質のスクリーニング方法。
[8] 請求項 1から 4のいずれかに記載の疾患モデル動物に被検物質を投与し、 関 節炎又は関節症若しくは骨粗鬆症の発症率及び重症度を測定、 評価すること 含む、 関節炎又は関節症若しくは骨粗鬆症の発症抑制物質のスクリーニング 方法。
[9] 請求項 1から 4のいずれかに記載の疾患モデル動物で得られた測定及び評価 結果を野生型の同種動物で得られた結果と比較することをさらに含むこと特 徵とする、 請求項 7又は 8に記載のスクリーニング方法。
[10] 請求項 6から 9のいずれかに記載のスクリーニング方法で同定される物質を 含有してなる自己免疫疾患又は骨粗鬆症の予防■治療剤。
[I I ] 樹状細胞免疫受容体 (DG I R)タンパク質をコードする遺伝子又はその部分配列 をコードする D N Aを含有してなる自己免疫疾患又は骨粗鬆症の診断薬。
[12] 自己免疫疾患が、 関節リウマチ(RA)、 強直性脊椎炎 (AS)、 乾癬性関節炎 (PsA) 、 シエグレン症候群 (SS)、 又は特発性全身性骨硬化症 (D I SH)である請求項 1 1に記載の診断薬。
[13] 樹状細胞免疫受容体 (DGI R)タンパク質と同一又は実質的に同一のアミノ酸配 列を有するぺプチドもしくはその部分べプチドまたはその塩、 及び候補物質 を含有することを特徴とする自己免疫疾患又は骨粗鬆症の予防■治療剤のス クリーニング用キット。
[14] 樹状細胞免疫受容体 (DGI R)タンパク質をコードする遺伝子と同一又は実質的 に同一の塩基配列またはその部分配列をコードする D N Aを含有することを 特徴とする自己免疫疾患又は骨粗鬆症の診断用キット。
[15] 樹状細胞免疫受容体 (DGI R)タンパク質と同一又は実質的に同一のアミノ酸配 列を有するぺプチドもしくはその部分べプチドまたはその塩からなる、 ウイ ルス感染症、 細菌感染症、 原虫感染症、 真菌感染症を含む感染症の治療薬。
[16] 樹状細胞免疫受容体 (DGI R)タンパク質と同一又は実質的に同一のアミノ酸配 列を有するぺプチドもしくはその部分べプチドまたはその塩からなる、 ガン 免疫療法における賦活剤。
