Abstract
Se propone un procedimiento de obtención y purificación del 2-(2-metil-2-nitrovinil)- furano en un paso de reacción a partir de furfural y nitroetano utilizando isobutilamina como catalizador y carbon activado como adsorbente, lograndose en todos los casos un producto con calidad farmacéutica. Esta tecnología reduce apreciablemente el impacto ambiental al eliminarse por absorción en agua los residuales gaseosos del sistema. La composición farmacéutica que se propone entre los objetos de la presente invención contiene 2-(2-metil-2-nitrovinil)-furano y presenta potente acción frente a ectoparasites a bajas concentraciones.
Description
PROCESS FOR THE PREPARATION OF 2- (2-METHYL-2-NITROVINYL) FURAN AND ITS USE FOR THE TREATMENT OF DISEASES CAUSED BY ECTOPARASITES .
Campo de Ia Técnica
La presente invención se relaciona con Ia rama de Ia salud animal y humana con un producto que presenta potente actividad frente a los ectoparásitos, el cual ha sido evaluado ya con resultados muy satisfactorios in vitro y en animales de experimentación. Específicamente, Ia invención describe un compuesto sencillo, de base furánica, obtenido a partir del furfural en un paso de reacción, que presenta un grupo nitro y uno metilo unido a un doble enlace exocíclico.
Arte previo
No existe información publicada hasta Ia fecha sobre un producto de acción frente a ectoparásitos basado en este compuesto como principio activo. El 2-(2-metil-2-nitrovinil)-furano tiene un bajo costo de producción, es fácilmente biodegradable y por no haberse aplicado anteriormente no debe presentar resistencia a los ectoparásitos.
Se reportan los pirazoles como los principios activos que mas patentes han generado, pero no prevalece una estructura química si no que existe una gran variedad de principios activos para estos fines.
Descripción detallada de Ia invención
El presente procedimiento se caracteriza por obtener de forma reproducibles altos rendimientos y purezas que sobrepasan el 99 %, Io cual permite su utilización en Ia medicina, aspecto este nunca antes reportado. El nuevo procedimiento de obtención del 2-(2-metil-2-nitrovinil)-furano consiste en poner a reaccionar el nitroetano y el furfural en relaciones molares de 1 :1 y 1 :1.5 durante un tiempo de contacto entre 0.5 y 1.5 horas y un tiempo de reacción entre 3 a 5 horas en un rango de temperatura entre 110 y 130 0C, utilizando entre 0.05 y 0.1 mol de una amina por mol de furfural como catalizador con velocidad de agitación entre 100 y 300 rpm.
El condensador del equipo está conectado a trampas de absorción que contienen agua para poder absorber los gases nitrosos y evitar de esta forma Ia contaminación de Ia atmósfera.
La mezcla reaccionante obtenida en estas condiciones se somete a un proceso de enfriamiento entre 1 y 10 0C y se separa el sólido mediante centrifugación. El producto crudo se purifica disolviéndolo en un volumen de un solvente polar que se encuentra entre 4 y 5 mi por g de 2-(2-metil-2-nitrovinil)-furano crudo y entre 3 y 4 g de carbón activado por g de 2-(2-metil-2-nitrovinil)-furano crudo, durante un tiempo entre 10 y 30 minutos. Posteriormente se filtra el carbón y se deja precipitar a una temperatura entre 0 y 10 0C. El producto puro se filtra, se seca durante 1.5 a 3.5 horas a una temperatura que oscila entre 25 y 30 0C y se almacena a un temperatura entre 10 y 20 0C en frasco ámbar. En los estudios toxicológicos realizados con el producto se comprobó que Ia DL50 obtenida en ratas Wistard/Lppt por vía oral, tuvo diferencias entre machos y hembras, siendo estos últimos mas susceptible con DL50 de 561 mg/kg y 448 mg/kg de peso vivo respectivamente. Para los ratones se demostró que Ia DL50 del producto es de 120 mg/kg, de peso corporal, extendiéndose las observaciones en ambas pruebas hasta 14 días. Las evaluaciones a largo plazo en estudios subcrónicos (30 días) por vía oral en ratas no mostró ningún efecto indeseable que nos indicara peligrosidad del mismo, puesto que Ia dosis mayor (40 % de Ia DL50 para hembras) a los 90 días de tratamiento solo mostró algún daño hepático y los niveles inferiores (20 y 10 %) no mostraron ninguna alteración. Los estudios especiales de mutagenicidad para el test de AMES no mostraron efecto mutagénico que Io invaliden como producto a emplear en el campo biológico. La determinación de la irritabilidad oftálmica puso de manifiesto que el producto en forma pura es altamente irritante sobre las estructuras oculares. Por otra parte las concentraciones inferiores al 5 % no manifiestan esta agresividad. El producto resulta irritante para las mucosas y Ia piel en forma pura por Io que es recomendable tomar medidas en Ia industria, del ingrediente farmacéutico activo y al elaborar las diferentes formas farmacéuticas. El producto tiene una absorción muy pobre por Ia piel, cuestión que esta asociada al vehículo empleado; en base hidrófila el paso es muy pobre, aspecto este que se determinó empleando técnicas de cromatografía gaseosa. De todas estas investigaciones se puede inferir que el producto se puede catalogar de moderamente tóxico y que no existen criterios que Io invaliden en el aspecto terapéutico.
Para evaluar Ia efectividad contra los ácaros e insectos de diferentes especies de animales, se realizaron experimentos in vitro e in vivo, en animales infestados naturalmente.
EJEMPLOS DE REALIZACIÓN
EJEMPLO 1.
Obtención de purezas en diferentes síntesis llevadas a cabo en un reactor de 2 litros variando parámetros dentro de los rasgos reportados. Tabla 1.
La determinación de Ia pureza se llevó a cabo por HPLC y por Espectrofotometría UV-VIS
EJEMPLO 2
El 2-(2-metil-2-nitrovinil)-furano puro fue disuelto en una solución de aceite/acetona realizando diferentes concentraciones con el que se impregnaron papeles de filtro Watman No.1. Para Ia evaluación se utilizaron larvas de Boophilus microplus de una cepa sensible a organofosforados mantenidas en el Laboratorio de Parasitología del Centro de Bioactivos Químicos de Ia Universidad Central "Marta Abreu" de Las Villas, Cuba (CBQ) y Ia Estación Nacional Experimental de Parasitología perteneciente al Instituto de Medicina Veterinaria de Cuba (ENEP). Para cada concentración utilizada se realizaron 3 réplicas utilizando Ia técnica descrita por Stone y Hayduck (1962), versión FAO (1988), realizándose Ia lectura a las 24 horas, utilizando el criterio de muertas y vivas para medir Ia efectividad y Ia determinación de Ia CL50 por el método Probit. Los resultados se exponen en Ia tabla 2.

EJEMPLO 3 El 2-(2-metil-2-nitrovinil)-furano formulado con Tween 80 + XiIoI y llevado a concentraciones de 0,08 %, 0,17 % y 0,35 %. La efectividad garrapaticida se verificó sobre hembras repletas de B. microplus. Para el desarrollo de estos experimentos se utilizaron hembras adultas sensibles a garrapaticidas organofosforados con el que fueron bañadas por Ia técnica de mininmersión 30 teleoginas de B. microplus mantenidas en el CBQ, controlando el peso de las teleoginas, el peso de los huevos, Ia eclosión, el estimado de reproducción y el control sobre Ia reproducción (ERD) según Ia técnica de Drunmond (1971). La incubación se realizó a 30 ± 10C con 90 ±5 % de humedad relativa, además se añadió un control con Tween 80 + XiIoI + agua destilada. Los resultados se demuestran en Ia tabla 3. Tabla 3
El 2-(2~met¡l-2-nitrovinil)-furano fue formulado en petrolato sólido a diferentes concentraciones. Para Ia evaluación se seleccionaron conejos de Ia raza Nueva Zelanda Blanco de 5 meses de edad con manifestaciones clínicas de sarna auricular, corroborándose en el laboratorio Ia presencia de Psoroptes cuniculli. Las muestras de Ia piel lesionada se tomaron antes de Ia aplicación del tratamiento y transcurridos 6 días posteriores al mismo. Para Ia toma, traslado y procesamiento de las mismas se siguieron las indicaciones de Liebisch et al. (1978). Se realizó el conteo de huevos, larvas y adultos. Para este experimento se utilizaron 12 conejos divididos en 4 grupos, los que fueron tratados con las formulaciones de Ia siguiente manera: grupo 1 = 2-(2-metil-2-nitrovinil)-furano 1 %, grupo 2 = 2-(2-metil-2- nitrov¡nil)-furano 0.5%, grupo 3 = petrolato sólido, grupo 4 = control sin tratar. Para evaluar Ia efectividad se utilizaron los resultados del número de ácaros vivos, huevos, larvas y adultos encontrados antes y después del tratamiento, expresados en rangos de invasión, así como el comportamiento clínico de los animales. Los resultados obtenidos aparecen en Ia tabla 4. Tabla 4.
El 2-(2-mθt¡l-2-n¡trovinil)-furano fue disuelto en una solución de acetona al 1 % con Ia que se impregnaron papeles de filtro Watman No.1. Para Ia evaluación del efecto por inhalación se utilizaron 40 insectos adultos de Musca domestica mantenidos en el Instituto de Investigaciones Agrícolas y Forestales (INIFAT). Los insectos fueron confinados en una cámara, separados mediante una malla metálica. Las evaluaciones se hicieron con intervalos de 1 hora para ver el efecto knock down y a las 24 horas para anotar Ia mortalidad total. Los resultados obtenidos fueron los siguientes: Tabla 5.

Claims
1.- Procedimiento para Ia obtención del 2-(2-met¡l-2~nitrovin¡l)-furano caracterizado por comprender los siguientes pasos:
• reacción del nitroetano y el furfural en una relación molar de 1 :1 a 1 :1.5 durante un tiempo de contacto entre 0.5 y 1.5 horas y un tiempo de reacción de 3 a 5 horas en un rango de temperatura entre 110 y 130 0C;
• adición de una amina primaria como catalizador en cantidades entre 0.05 y 0.1 mol por mol de furfural, con velocidad de agitación entre 100 y 300 rpm;
» Ia mezcla reaccionante se deja enfriar entre 1 y 10 0C y el crudo así obtenido es separado por métodos de separación sólido-líquido;
• purificación del crudo con carbón activado y solventes polares a una temperatura entre 25 y 35 0C, con un tiempo de contacto entre 10 y 30 minutos;
• separación del carbón activado;
• enfriamiento de Ia solución purificada entre 0 y 10 0C y recuperación de los cristales por centrifugación o filtración a vacío;
• secado del producto final a una temperatura entre 25 y 30 0C durante un tiempo de 1.5 a 3.5 horas;
• almacenamiento del producto final a una temperatura entre 10 y 20 0C en frasco ámbar.
2.- Procedimiento para Ia obtención del 2-(2-metil-2-nitrov¡nil)-furano según Ia reivindicación 1 donde Ia amina primaria empleada como catalizador es preferentemente Ia isobutilamina.
3.- Procedimiento para Ia obtención del 2-(2-metil-2-nitrovinil)-furano según Ia reivindicación 1 donde Ia separación sólido-líquido se efectúa preferentemente por centrifugación o filtración a vacío.
4.- Procedimiento para Ia obtención del 2-(2-metil-2-nitrovinil)-furano según Ia reivindicación 1 donde el solvente polar utilizado en Ia purificación del crudo es preferentemente etanol.
5.- 2-(2-metil-2-n¡trovinil)-furano obtenido según el procedimiento de las reivindicaciones 1 a Ia 4 con una pureza de al menos un 98%.
6.- Composición farmacéutica caracterizada por contener como principio activo 2- (2-metil-2-nitrovinil)-furano según el procedimiento descrito en Ia reivindicación 1 para Ia manufactura de medicamentos veterinarios y humanos.
7.- Uso del 2-(2-metil-2-nitrovinil)-furano según Ia reivindicación 6 para el tratamiento de los ectoparásitos.
