技術分野
[0001] 本発明は、自動分析装置や分注装置に用いられる液面検知装置に関する。
背景技術
[0002] 免疫学的凝集反応等を用いて血液、体液等の検体に関する分析を行う自動分析 装置では、反応容器内の試料や試薬の分注量が規定量に対してばらつくと、分析結 果に大きく影響し、検査分析の信頼性を損なう。そのため、分注量を高い精度で測定 し、分注した多数の試料から、分注量が適正範囲に入った試料だけを選び出す技術 を確立する必要がある。分注量を測定する方法のひとつとして、液面の高さを検知す る方法があり、その手段としては、電極を用いた電気伝導度測定法によるものや超音 波測定法によるもの等が知られて 、る。
[0003] しかし、容器の口径が小さい場合、電極を用いた電気伝導度測定法による液面検 知手段では、電極を挿入するのが困難であり、また、電極が液面に接触することによ り液量に誤差が生じるという問題がある。また、超音波測定法による液面検知手段で は、超音波センサーから出射された超音波が拡散して、一部の波が容器の開口縁に 照射され、この開口縁で反射した超音波が超音波センサーで検知されて、正確な測 定値が得られな 、と 、う問題がある。

[0010] 本発明の液面検知装置によれば、容器開口端と液面が撮像範囲に入るような所定 位置力 容器内を撮像することにより液面の位置を検知するので、光を透過しない金 属製の容器や、マトリックス状に配列形成された容器内の液面の位置を検知すること が可能となる。その結果、分注量が適正範囲内の試料だけを選び出すことができる ため、分析精度を向上させることができる。

[0013] 以下、本発明の液面検知装置の好適な実施の形態について詳しく説明する。
図 1は、本発明の液面検知装置 10を適用した自動分析装置の分析工程を示す図 である。ここで例示される自動分析装置は、免疫学的凝集反応を用いて被検血液の 抗原抗体反応の分析を行うための装置である。まず、自動分析装置の分析工程の概 略を説明する。
[0014] 被検者より採取された血液は試験管 41に収容され、これら試験管 41はラック 42に 収容されて、ベルトコンペャ等の搬送手段 43によって血液分注位置にある血液分注 装置 44、及び、希釈液分注位置にある希釈液分注装置 44aまで搬送され、血液及 び希釈液が各々試料容器 45に分注される。分注後の希釈された血液が入った試料 容器 45は試料 ·試薬分注位置 46へ搬送される。

図 2—1はマイクロプレート 12の平面図、図 2— 2は図 2—1の A— A線断面図である 。図 2—1及び図 2— 2に示すように、マイクロプレート 12には縦列 12個、横列 10個 の反応容器 13がマトリックス状に配列形成されており、この反応容器 13の底面の一 部には傾斜面が形成されている。

が経過するまで、希釈検体と試薬を分注した複数枚のマイクロプレート 12を反応ライ ン内に収納しておき、抗原抗体反応により検体中の血球粒子、ラテックス粒子等を凝 集させる。凝集反応が完了したマイクロプレート 12は搬送機構 53によって搬送され、 測光位置 54で停止し、 CCDカメラ 57によって、各反応容器 13の底面に形成された 凝集パターンの相違が検出される。
[0018] 図 1に示すように、液面検知装置 10は、分析工程中における試料'試薬分注位置 4 6に配置されており、本実施形態では、マイクロプレート 12上の各反応容器 13内に 分注された試料と試薬との混合液 (以下、省略して「混合液」 t ヽぅ)の液面位置を検 知する。
図 3は、本発明の液面検知装置 10の概略構成図であり、説明の便宜上、マイクロ プレート 12上に形成された反応容器 13のひとつを拡大して示している。また、図 4は 撮像手段 20によって撮像される撮像範囲の一例を示す図である。
[0019] 図 3に示すように、液面検知装置 10は、撮像装置 20と画像処理装置 30とを具える 。本実施形態では、撮像装置 20として CCDカメラが用いられる。画像処理装置 30は 、撮像された画像における容器開口端カゝら液面までの距離が、予め設定された閾値 内にある力否かを判定する判定部 31と、後述する判定用の基準画像を格納する記 憶装置 32とを具える。
[0020] 図 3及び図 4に示すように、 CCDカメラ 20は、反応容器 13の容器開口端 14と混合 液の液面 15とが撮像範囲 21に入る所定の位置に設置され、この位置から反応容器 13内を撮像する。
図 3及び図 4では、 CCDカメラ 20は、反応容器 13の斜め上方に設置されており、 その撮影光軸 16は反応容器の中心軸 Oに対して傾き、且つ、交差した状態に設定 されている。反応容器 13との距離及び角度はカメラの分解能、液体の色等を考慮し て適宜決定される。
[0021] CCDカメラ 20によって撮像された画像データは、画像処理装置 30に入力され、適 宜処理が施された後、判定部 31に送られる。ここで、説明の便宜上、撮像された画 像を画像 Cとする。判定部 31での処理は、例えば以下のように行うことができる。

記憶装置 32には、液量過多閾値 d (l—x)の量の液体が収容された反応容器 13 内を撮像した基準画像 Aと、液量過少閾値 d (1 +x)の量の液体が収容された反応 容器 13内を撮像した基準画像 Bとが予め格納されている。基準画像 A, Bにおける 撮像位置は上記の画像 Cにおける撮像位置と同一(すなわち、反応容器 13に対して 同じ距離'角度)である。
[0023] 画像 C上での開口端から液面までの距離と、基準画像 A上での開口端から液面ま での距離とを比較し、画像 C上での開口端カゝら液面までの距離が液量過多閾値を超 えていないかどうかを判定する。さらに、画像 C上での開口端から液面までの距離と、 基準画像 B上での開口端から液面までの距離とを比較し、画像 C上での開口端から 液面までの距離力 液量過少閾値を超えて ヽな 、かどうかを判定する。
画像 C上での開口端力も液面までの距離が、上記閾値の範囲内にある場合には、 分析データとして採用される。もし液量過多閾値より小さい場合は液量過多と判定さ れ、液量過少閾値より大きい場合には液量過少と判定される。これらはエラーとして 出力され、分析データから除外される。
[0024] 上記の一連の処理をマイクロプレート 12上のすべての反応容器 13につ!/、て行う。
ひとつの反応容器 13を撮像するごとに、各反応容器の開口端 14と液面 15とが撮像 範囲 21に入る所定位置に CCDカメラ 20が来るように、 CCDカメラ 20又はマイクロプ レート 12を移動させて両者を固定し、この位置から各反応容器 13内部を撮像する。 この場合、撮像する毎に、各反応容器 13に対し、 CCDカメラ 20を同じ位置、すなわ ち、同じ角度、同じ距離の位置に固定する必要がある。
撮像方法の一例としては、縦列 12個の反応容器 13を 1個撮像する毎に CCDカメラ 20を縦方向に移動させ、この縦列の液面をすベて撮影した後、マイクロプレート 12を 横方向に移動させて、再び次の縦列を撮像していく方法がある力 他の方法を用い てもよい。
[0025] なお、判定部 31での処理は本実施形態に限定されるものではなぐ発明の内容を 逸脱しな 、範囲で変更することができる。
[0026] また、本実施形態では、反応容器 13内の試料と試薬の混合液の液面位置を検知 したが、試薬を分注する前の、試料が分注された段階で液面検知を行ってもよい。ま た、試料と試薬の分注順序を反対にした場合には、試料を分注する前の、試薬が分 注された段階で液面検知を行ってもよい。さらに、検体としての血液や希釈液が分注 される試料容器 45における液面の検知にも使用できる。
[0027] また、マイクロプレート 12上に形成された反応容器 13の形状'個数は、本実施形態 で用いたものに限定されず、例えば、反応容器 13の底面の垂直断面形状が半円状 、矩形状、 V字状等のものでも適用することができる。

[1] 容器に収容された液体の液面を検知する液面検知装置であって、
容器開口端と液面とが撮像範囲に入る所定の位置カゝら前記容器内を撮像し、 撮像された画像における容器開口端カゝら液面までの距離が、予め設定された閾値 内にある力否かを判定することを特徴とする液面検知装置。
