Definitions
- the present invention concerns milk serum (lactoserum) comprising aqueous solutions supplemented with poorly water soluble or water insoluble and/or poorly water wetable or water non-wetable and possibly also oxidation sensitive ingredients such as hydrophobic vitamins and coenzymes, in particular ubiquinones, especially coenzyme QlO.
- Vitamins comprising preparations are e.g. vitamins supplemented energy drinks .
- Such drinks comprise e.g. water soluble vitamins and water-soluble derivatives of fat-soluble vitamins and minerals.
- a disadvantage of such drinks is that - due to the instability of many vitamins in contact with humidity and/or oxygen and/or light, necessitating to add a surplus of them to ensure the declared amount at the end of the marked storage time.
- alternative or usual preparations are in the form of water disintegrating tablets, in particular effervescent tablets, or, especially for water insoluble substances, in the form of capsules .
- Coenzyme QlO is an essential component of cellular energy metabolism, in particular respiration, and thus is a very desirable ingredient in preparations intended for active people or in preparations intended as nutritional supplements to improve the health status and the vitality of people in need thereof.
- the amount of substance actually consumed is unpredictable and may to a high extent depend on the time elapsed between the disintegration of the tablet and the actual time of consumption, the more so since many people prefer to take few sips several times during a certain time than a whole glass at once.
- reference to substances that in water are poorly soluble also includes substances that are insoluble in water (water insoluble substances) , substances that are poorly wetted by water (poorly water wetable substances) and substances that are not wetted by water (not water wetable substances) .
- Poorly water soluble means that the respective substance has a solubility in water at 5°C of less than 10 mg/100 ml, in general less than 7 mg/100 ml.
- the invention is much advantageous in connection with substances having a solubility of less than 5 mg/100 ml or even an as low solubility as less than 3 or even 2 mg/100 ml. It has now surprisingly be found that milk serum (lactoserum) is a very efficient medium to improve the solubility or dispersibility and the wetability of lipophilic/hydrophobic substances .
- milk serum or lactoserum respectively, is not used as synonym to whey but means the liquid resulting after removal of the proteins and the fats from milk or whey.
- Milk serum suitable for use in the present invention may be non-acidic, acidic (e.g. obtained from sour-whey) or acidified. While whey in general still comprises about 20% of the originally present milk proteins, the milk serum (lactoserum) in general has a milk-fat content of less than 100 mg/100 g, usually less than about 50 mg/100 g, and a milk-protein content, determined as nitrogen content, of less than 100 mg/100 g, usually less than about 20 mg/100 g. Milk serum, however, still contains about 90 % of the water-soluble vitamins, minerals and trace elements present in full milk and therefore is a very desirable basis or additive for nutritional supplementation.
- lactoserum lactoserum
- milk serum wherein the lactose that usually is present in an amount of about 90 % of the lactose originally present in milk is destroid, e.g. by cleavage by means of lactase ( ⁇ -galactosidase) .
- the milk serum may optionally be acidified by lactic acid. While in the scope of the present invention the use of milk serum with less than about 100 mg/lOOg milk-fat and less than about 100 mg/100 g milk-proteins is possible and - since milk serum is e.g.
- milk serum or whey i.e. milk derived products having higher fat and/or protein content, such as a whey having - in general - about 0.5 % by weight proteins and about 0.1 % by weight fats.
- the milk serum suitable for being used in the scope of this invention can be produced by ultrafiltration of milk or whey, the lactose can be cleaved by means of lactase, and optionally acidified e.g. by previous fermentation or by addition of acid , in particular lactic acid.
- Such milk serum can be used as produced, concentrated, dried or rediluted/redissolved.
- Whey suitable for being used in the scope of the present invention can be obtained by centrifugation and optionally acidified by fermentation or addition of acid (see above) .
- the aqueous composition is manifested by the features that it comprises at least one in water poorly soluble substance and milk serum or a milk serum comprising substance such as whey. Milk serum or a milk serum comprising substance are also referred to as "milk serum solid comprising”.
- Such in water poorly soluble substances often do not only strongly adhere to container surfaces but are also oxidation sensitive limiting their storability in contact with water and/or air and/or light, it is preferred that such substances are stored separately, preferably separate from any water comprising medium.
- Such substances are e.g. fat-soluble vitamins and/or coenzymes.
- coenzyme QlO One very desirable and critical coenzyme is coenzyme QlO.
- the aqueous solution of milk serum solids can either be milk serum or whey.
- Milk serum can be water diluted or non-diluted milk serum as produced or milk serum concentrate, i.e. milk serum with a lower water content than the serum as produced, or it can be rediluted milk serum concentrate or redissolved dried milk serum, whereby the rediluting or the redissolving can be performed with an amount of water leading to a concentration as found in milk serum as produced, or to a lower or higher concentration.
- a preferred aqueous solution of milk serum solids is selected from milk serum as produced and/or rediluted concentrated milk serum.
- Whey can be water diluted or non-diluted whey as produced or whey concentrate, i.e. whey with a lower water content than the whey as produced, or it can be rediluted whey concentrate or redissolved dried whey, whereby the rediluting or the redissolving can be performed with an amount of water leading to a concentration as found in whey as produced, or to a lower or higher concentration.
- a preferred aqueous solution of whey solids is selected from whey as produced and/or rediluted concentrated whey.
- the component comprising the oxidation sensitive component (s) , the second component is a solid formulation, in particular in the form of a tablet or as granules.
- a tablet especially a fast disintegrating gas liberating tablet known as effervescent tablets.
- Such tablets have the advantage that they have a density ensuring that they go to the bottom of the aqueous solution comprising container thereby ensuring that finely powdered poorly wetable substances are not prevented from entering the water bulk due to the high surface tension of the water. Due to the gas liberation, in particular the CO2 liberation, the powdered substances are transported upwards within the aqueous phase and they are thereby distributed within the water phase, making additional mixing by shaking unnecessary.
- effervescent granules may be used, whereby their density and granule size preferably are chosen such that the above described benefit of effervescent tablets is also obtained.
- Powders are also possible provided that their composition is such that they penetrate the water surface or that dissolution or dispersion is obtained upon moderate shaking.
- the milk serum or whey is preferably present in dried form.
- Such a preparation can either be provided separately, namely as a supplement to tap water or mineral water, or as one component of a two component composition, wherein the other component can be water or an aqueous solution comprising water soluble substances and/or part of the milk serum.
- any two composition preparations it is also possible to provide a liquid component comprising the poorly water soluble, and in particular also oxidation and/or light sensitive substances with or without milk serum.
- the number of acceptable solvents is limited by the solubility of the substances and the approval of the solvent for oral uptake.
- milk serum solids comprising suspensions/solutions can be envisaged. Nevertheless, in view of storage stability, solid preparations are much preferred.
- solid or concentrated liquid formulations comprising light and/or air sensitive substances with or without milk serum or whey
- they are packed in light impermeable and preferably also air impermeable one portion packages, in particular in fully metalized, e.g. aluminized, blister packages.
- fully metalized e.g. aluminized, blister packages.
- effervescent tablets such tablets are placed between two metalized foils and sealed in a dry atmosphere.
- compositions of the present invention can be confectioned as functional beverages, i.e. as vitamins and minerals supplemented drinks, as sports drinks etc.
- the amount of essential substances is in an amount suitable to supplement a usual daily intake.
- the drinks can also be designed such that they are suitable for prevention of a deficiency or for therapy of a deficiency of essential substances.
- an object of the present invention is the use of milk serum for enhancing the solubility and/or the dispersibility and/or the re-dipersibility of at least one in water poorly soluble substance in particular substances with the solubility and reactivity/sensitivity of ubiquinone, or a method for enhancing the solubility and/or the dispersibility and/or the re-dipersibility of at least one in water poorly soluble substance, in particular substances with the solubility and reactivity of ubiquinone, by adding milk serum (or milk serum comprising products) to the substance or to the aqueous medium prior to combining the aqueous medium and the poorly water soluble, in particular also oxidation and/or light sensitive substance.
- a further object of the present invention is a container comprising the inventive two component preparation, said container comprising two compartments and an opening means, said compartments being separated by a wall that can be destroyed/opened by said opening means, said container comprising in a first compartment water or an aqueous solution and in a second compartment a preparation comprising at least one in water poorly soluble substance, said container comprising in the first and/or in the second compartment milk serum, and said two compartments being united due to the operation of the opening means upon the first use of the container such that the contents of the two compartments are combined.
- a preferred kind of container is e.g. described in more detail in PCT/CH2.005/000596 filed 26 October 2005.
- Suitable two-compartment containers for use with respective compositions are known in the art, e.g. from WO 98/40289, GB-A-2 321 231, GB-A-2 364 699, WO 2005/009862 and PCT/CH2005/000596.
- a method is preferred, wherein the bottle body is sealed with a breakable or cuttable foil such as an aluminized or non-metalized plastics foil, e.g. a PET foil, onto which a tablet of the present invention, sealed into a blister, is placed and fixed, e.g. by crimping.
- a breakable or cuttable foil such as an aluminized or non-metalized plastics foil, e.g. a PET foil, onto which a tablet of the present invention, sealed into a blister, is placed and fixed, e.g. by crimping.

- coenzyme QlO 30 to 35 mg/500 ml is preferred, for caffeine 80 to 90 mg/500 ml and for L- carnitin 250 to 350 mg/500 ml.
- the carbohydrate content is enhanced by addition of sugars (including maltodextrin) .
- sugars including maltodextrin
- the sugars, in particular fructose and dextrose are added in amounts of about 3 g per 500 ml, maltodextrin in an amount of about 1 g per 500 ml.
- a milk serum with a solid concentration as produced is suitable.
- Such milk serum preferably is acidified and lactose-free and may be obtained by ultrafiltration through a membrane with a molecular weight cut-off of 10000 kDa, e.g.
- It may be used as produced or be a rediluted milk serum concentrate or a redissolved dried milk serum, whereby, however, the dried milk serum due to the energy needed for full drying is less preferred.
- a suitable milk serum basis has the following product specification (per 100 ml) :
- Carbohydrates 3.0 g (all sugars, namely 1.5 g dextrose, 1.5 g galactose, 0.0 g lactose)
- Fats below 100 mg, preferably below 50 mg
- Lactic acid 0.05 - 0.3 g
- a suitable effervescent tablet may be pleasantly colored to get an optical effect during the dissolution.
- the tablet may be homogeneously colored or be a two or more phase tablet having different colors. Since several desirable ingredients are colored themselves, but resulting in an unpleasant color, colorants are preferably added at least to that phase of the tablet comprising colored ingredients. In the case of two or more phase tablets, the several phases may be layered or homogeneously mixed with each other.
- a tablet suitable for use in the scope of the present invention can be produced according to any method suitable for the production of vitamins, especially oxygen and light sensitive vitamins comprising effervescent tablets, by preparing a pre-mix comprising all ingredients and then pressing said premix according to methods well known to the skilled person.
- the carbon dioxide generated by each tablet in water or in aqueous solution can be regulated by the carbonate content and the acid content and the acidity of the acid component, advantageously is above 1.5 g/liter, preferably in the range of 1.5. to 3.0 g per liter, and most preferred in the range of 1.7 - 1.9 g/liter.
- Such milk serum comprising basic drink, i.e. without carbohydrate or fat content influencing substances, has the following nutritional values (per 100 ml) : Energy value: about 51 kJ or 12 kcal
- milk serum concentrations as low as about 1.5 % are still able to dissolve or homogeneously disperse the coenzyme QlO.
- Example 2 The solubility of QlO powder in different aqueous solutions was tested. For each solution, the identical procedure was applied. The results are listed in Table 2. As can be seen from Samples I and J, the adhesion of the QlO powder at the bottle walls is markedly reduced by the milk serum addition.
- Example 3 The effect of additional fat or albumin on the solubility of QlO powder and an effervescent tablet (see Example 4) was tested.
- the Process applied for samples P and Q and for samples R and S were identical. Samples P and Q were produced by heating to 60 0 C, adding the fat, cooling to 35°C, and then adding QlO. Samples R and S were produced by heating to 60 0 C, adding the protein, cooling to 10 0 C, and then adding QlO, The results are listed in Table 3. They show the benefit obtained by adding the coenzyme QlO by means of an effervescent tablet.
- a drink rich in vitamins, minerals and trace elements is produced by providing 470 ml of a solution comprising a milk serum basis in a concentration as produced, with a solids content of 3.2 +/- 0.2 %, a glucose content of 1.5 %, a galactose content of 1.5 %, a fat content of ⁇ 100 mg, a protein content of ⁇ 100 mg, a lactic acid content of 0.1 %, a minerals content of 0.2 %, and a pH of 4.5 +/-0.2 %. in a sterile container filled under sterile conditions and sealed with a breakable or cuttable foil and providing an effervescent tablet with a weight of 5.7 g and having the following composition:
- the tablet is sealed in an aluminized blister and then placed onto the seal of the bottle neck and crimped. Finally, the bottle is closed with a bottle cover including an opening means for destroying the bottle neck sealing foil, thereby liberating the tablet such that it can fall (or is pushed) into the solution within the bottle body upon the first intended use of the bottle (see also PCT/CH2005/000596 for a more detailed description) .
- the bottle cover preferably is a "sports cap", i.e. a cover with a re-closable drinking opening and said cover is unremovably fixed to the neck of the bottle body.
