Abstract

Die Erfindung betrifft ein Verfahren zur Isolierung von multipotenten Stammzellen aus Zahngewebe, bei dem die Stammzellen aus einem Gewebeverbund gewonnen und anschließend kultiviert werden. Die Erfindung betrifft ferner mittels des erfindungsgemäßen Verfahrens isolierte Stammzellen sowie Knochenzellen und neuronale Zellen, die mittels des erfindungsgemäßen Verfahrens hergestellt wurden. Die Erfindung betrifft auch ein Verfahren zur Herstellung einer Bank von Stammzellen, bei dem die Zellen mittels des erfindungsgemäßen Verfahrens gelagert werden. Erfindungsgemäß werden die Zellen eines kissenartigen Weichgewebes, das unmittelbar an der apikalen Seite eines extrahierten unreifen Zahns unterhalb der Papille lokalisierbar ist, im Gewebeverbund kryokonserviert, wobei der Gewebeverbund erst nach dem Auftauen zum Gewinnen der Stammzellen aufgelöst wird. Die Ergebnisse zeigen, dass auch nach der Kryokonservierung des Ursprungsgewebes Stamm-/Progenitorzellen isoliert werden können und diese Stamm-/Progenitorzellen auf osteogene Stimulierung reagieren. Darüber hinaus fällt die Reaktion der Zellen nach Kryokonservierung höher aus als ohne Kryokonservierung.
