光学活性 PPAR活性化化合物中間体及びその製造法

[oooi] 本発明は、高脂血症、動脈硬化症、糖尿病等の疾患の予防及び Z又は治療薬と して有用な PPAR (ペルォキシゾーム増殖剤応答性受容体)活性化化合物の製造中 間体及びその製造法に関する。
背景技術
[0002] PPARは核内受容体ファミリーの一つとして知られており、現在までに 3つのサブタ ィプ(α、 γ、 δ )の存在が知られている(非特許文献 1〜5)。このうち、 PPAR αは 主に肝臓に発現しており、可塑剤及びフイブレート系薬剤、例えば Wyl4643や既に 医薬品として市販されているクロフイブレート、フエノフイブレート、ベザフイブレート、 ゲムフイブロジル等の薬剤により活性ィ匕されることが知られている(非特許文献 6〜7)
[0003] PPAR αの活性ィ匕は、哺乳動物においては、脂肪酸の β酸化を亢進し、血中トリグ リセリドを低下し、ヒトでは低密度リポ蛋白(LDL)コレステロール、超低密度リポ蛋白( VLDL)コレステロール等の血中脂質の低下が起こることが知られており、 PPAR a 活性化剤は高脂血症等の予防及び Z又は治療剤として有用であると考えられている 。また、 PPAR a活性化剤は高密度リポ蛋白(HDL)コレステロールの上昇、血管内 においては細胞接着因子である VCAM— 1の発現を抑制するので、動脈硬化等の 予防及び Z又は治療剤、糖尿病や炎症性疾患、さらには心疾患等の予防及び Z又 は治療に有用であるとも考えられて 、る(非特許文献 8〜 14)。
[0004] 一方、 PPAR γの活性ィ匕は、ヒトにおいて脂肪を増加し、体重の増加や肥満を起こ すという好ましくない作用を有する事が報告されており(非特許文献 15)、最近では Ρ PAR γの拮抗剤によってもインスリン抵抗性改善の可能性を示す報告もされている( 非特許文献 16〜18)。また、 PPAR δの活性ィ匕は、脂質蓄積作用に繋がることが示 唆されて!/、る(非特許文献 19)。従って、 PPAR γ及び δ活性の低!ヽ PPAR a選択 的な活性化剤が、体重増加、肥満を伴わずに、高脂血症、動脈硬化症、糖尿病、糖
尿病合併症、炎症、心疾患等の予防及び Z又は治療剤として有用であると考えられ ている。

発明が解決しょうとする課題
[0008] 本発明は、上記式 (A)で示される化合物の一つである (R) - 2- [3- [ [N- (ベン ズォキサゾールー 2 ィル) N— 3—(4ーメトキシフエノキシ)プロピル]アミノメチル ]フ ノキシ]酪酸 (ィ匕合物 (A— 1) )の製造中間体及びこれを高収率且つ高光学収 率で得る方法を提供することを目的とする。
課題を解決するための手段
[0009] 本発明者らは、上記化合物 (A— 1)の有用な製造法につ!、て鋭意研究を行った結 果、下記反応式 (I)に示すように、光学活性 2—トリフルォロメタンスルホニルォキシ 酪酸エステル (化合物(2) )を用い、これと 3 ヒドロキシベンズアルデヒド (化合物(1)
)とを塩基の存在下に反応させることにより、光学活性べンズアルデヒド誘導体 (化合 物(3))を高収率且つ高光学純度で得ることができることを見出した。そして、当該化 合物(3)を経由する方法(下記反応式 (Π)及び後記参考例参照)によれば、収率及 び光学純度を損なうことなくィ匕合物 (A— 1)が製造でき、化合物(3)が化合物 (A— 1 )の有用な合成中間体となることを見出した。

発明の効果
[0016] 本発明の方法によれば、 PPAR a選択的な活性化剤であり、体重増加、肥満を伴 わずに、高脂血症、動脈硬化症、糖尿病、糖尿病合併症、炎症、心疾患等の予防及 び Z又は治療が可能な (R) - 2- [3- [ [N- (ベンズォキサゾールー 2 ィル) -N — 3— (4 メトキシフエノキシ)プロピル]アミノメチル]フエノキシ]酪酸 (化合物 (A— 1 ) )の有用な製造中間体を高収率で且つ高光学純度で製造できる。
発明を実施するための最良の形態
[0017] 本発明の製造法は、化合物(1)と光学活性な化合物(2)を、塩基の存在下に反応 させて化合物(3)を得るものである。
ここで、化合物(2)及び(3)中の Rは、炭素数 1〜6のアルキル基又は炭素数 7〜8 のァラルキル基を示す力 アルキル基としては、メチル基、ェチル基、 n プロピル基 、イソプロピル基、 n ブチル基、イソブチル基、 t ブチル基等が好ましぐァラルキ ル基としてはべンジル基、フエネチル基等が好まし!/、。
[0018] 塩基としては、例えば水酸化リチウム、水酸化ナトリウム、水酸ィ匕カリウム等の水酸 化アルカリ金属類;炭酸ナトリウム、炭酸カリウム、炭酸セシウム等の炭酸アルカリ金 属類;炭酸水素ナトリウム、炭酸水素カリウム等の炭酸水素アルカリ金属類等の無機 塩基、ピリジン、トリエチルァミン、 N, N ジイソプロピルェチルァミン、 N—メチルモ ルホリン、 N, N ジメチルァ-リン等の有機塩基を使用することができる力 化学収 率の点力も炭酸カリウムを用いることが好まし 、。
[0019] 本反応は、溶媒中で行うことが好ましい。溶煤は、特に制限はないが、例えば塩ィ匕 メチレン、クロ口ホルム、四塩化炭素、 1, 2—ジクロロエタン、クロ口ベンゼン等のハロ ゲン化炭化水素類、ベンゼン、トルエン等の芳香族炭化水素類、テトラヒドロフラン、
ジェチルエーテル、ジォキサン等のエーテル類、アセトン、メチルェチルケトン等のケ トン類、ァセトニトリル、 N, N ジメチルホルムアミド、ジメチルスルホキシド等の非プ 口トン性極性溶媒、酢酸ェチル等が挙げられ、このうちァセトニトリルが好ましい。
[0020] 反応温度は、 0〜100°C、好ましくは 20〜90°Cで、 0. 5〜48時間反応させればよ く、 1〜24時間反応させるのが好ましい。
[0021] 前記特許文献 1には、 2—ヒドロキシカルボン酸エステルのフエニルエーテル化反 応に関し、 2—ヒドロキシカルボン酸エステルの水酸基をメシル化若しくはトシル化等 して水酸基をメタンスルホ二ルォキシ基ゃパラトルエンスルホニルォキシ基等の脱離 基に変換し、炭酸ナトリウム、炭酸カリウム、炭酸セシウム等の無機塩基やトリェチル ァミン、 N, N ジイソプロピルェチルァミン等の有機塩基の存在下、フエノール体と 反応させる方法が記載されている(反応工程式 F— 4)。更に 2 ハロゲン化カルボン 酸エステルのフエニルエーテル化反応に関しても、 2 ハロゲン化カルボン酸エステ ルを炭酸ナトリウム、炭酸カリウム、炭酸セシウム等の無機塩基ゃトリエチルァミン、 N , N ジイソプロピルェチルァミン等の有機塩基の存在下、フエノール体と反応させる 方法が記載されている (反応工程式 A—l)。これらに対し、脱離基としてトリフルォロ メタンスルホ -ルォキシ基を有する光学活性 2—トリフルォロメタンスルホ-ルォキシ 酪酸エステル (化合物(2) )を用いる本発明の方法によれば、極めて高収率で且つ 高光学純度で化合物(3)を得ることができる(後記実施例参照)。そして、当該化合 物(3)は化合物 (A— 1)の製造中間体として有用である。

本工程は、化合物(3)と化合物 (4)を縮合して得られるイミノ体又はイミ-ゥム塩を 還元してァミン化合物(5)を製造する工程である。
縮合反応は、化合物(3)と化合物 (4)を、例えばメタノール、エタノール、イソプロピ ルアルコール、テトラヒドロフラン、ジォキサン、トルエン、ァセトニトリル、 N, N—ジメ チルホルムアミド等の溶媒中、酢酸、塩酸等の酸の存在下又は非存在下で行われる 。反応は通常 20〜100°Cで 1〜12時間反応することにより行うことができる。
[0029] 還元は、水素化ホウ素ナトリウム、トリァセトキシ水素化ホウ素ナトリウム、シァノ水素
化ホウ素ナトリウム等の水素化還元剤を用い、例えば、水、アルコール類 (メタノール 、エタノール、イソプロピルアルコール等)、エーテル類(テトラヒドロフラン、ジォキサ ン、ジェチルエーテル等)、ハロゲン化炭化水素類 (ジクロロメタン、クロ口ホルム等)、 ァセトニトリル、ジメチルホルムアミド等の溶媒中で行うのが好ましい。反応は、通常 0 〜 30°Cで 1〜 12時間程度行うのが好まし 、。

本工程は、化合物(5)と 2 クロ口べンズォキサゾール (6)を塩基の存在下に反応 させて化合物(7)を製造する工程である。
本反応は、化合物(5)と 2 クロ口べンズォキサゾール(6)を、例えば N, N ジメチ ルホルムアミド、ァセトニトリル、テトラヒドロフラン、ジォキサン、クロ口ホルム、酢酸ェ チル等の溶媒中、 20〜100°Cで 1〜12時間反応することにより行うことができる。 塩基としては、例えば、炭酸ナトリウム、炭酸カリウム、炭酸セシウム等の無機塩基 ゃトリエチルァミン、 N, N ジイソプロピルェチルァミン、ピリジン等の有機塩基を用 いることがでさる。

本工程は、化合物(7)のエステルを脱エステル化して化合物 (A— 1)を製造するェ 程である。
脱エステル化反応は、加水分解、加水素分解 (還元)等の常法に従って実施するこ とができる。加水分解は、エステルの加水分解反応に用いられる反応条件のいずれ もが適用でき、例えば、水酸化リチウム、水酸化ナトリウム、水酸ィ匕カリウム、炭酸ナト リウム、炭酸カリウム等の無機塩基;塩酸、硫酸、臭化水素酸等の鉱酸;或いは p ト ルエンスルホン酸等の有機酸等の存在下、水、メタノール、エタノール、プロパノール 等のようなアルコール類、テトラヒドロフラン、ジォキサン等のようなエーテル類、ァセト ン、メチルェチルケトン等のようなケトン類、酢酸等の溶媒又はこれらの混合溶媒中で 行われる。
反応は、通常 0〜100°C、好ましくは 10〜50°Cで行われ、反応時間は通常 0. 5〜 24時間、好ましくは 1〜12時間である。

一テル類;酢酸メチル、酢酸ェチル、酢酸イソプロピル等のエステル類;メタノール、 エタノール、イソプロピルアルコール等のアルコール類; N, N ジメチルホルムアミド 等のアミド類等)中、水素化触媒 (例えば、パラジウム—炭素、パラジウム黒、パラジゥ ム、水酸化パラジウム、白金 炭素、二酸化白金、ラネーニッケル等)の存在下、無 機酸 (例えば、塩酸、硫酸、次亜塩素酸等)又は有機酸 (例えば、酢酸、トリフルォロ 酢酸、ギ酸等)の存在下又は非存在下、常圧又は加圧下の水素雰囲気下で行われ る。
反応は、通常 0〜30°C、好ましくは 10〜25°Cで行われ、反応時間は通常 5分〜 24 時間、好ましくは 1〜12時間である。

[0035] すなわち、 4—メトキシフエノール(c)にアクリロニトリル(d)をトリトン B、トリェチルアミ ン、 N, N ジイソプロピルェチルァミン等の塩基存在下で反応させ、 3— (4—メトキ シフエノキシ)プロピオ-トリル (e)を得、これをテトラヒドロフラン、ジォキサン等の溶媒 中、ボラン'テトラヒドロフラン錯体、ボラン'ジメチルスルフイド錯体、水素化アルミ-ゥ ムリチウム等を用いて還元する力、水素雰囲気下もしくはアンモニア中、ラネー-ッケ ル等の金属触媒を用いて還元することにより、 3—(4ーメトキシフエノキシ)プロピルァ ミン (化合物 (4) )を得ることができる。
[0036] 尚、本発明の各反応における目的物の単離は、必要に応じて、有機合成化学で常 用される精製法、例えば濾過、洗浄、乾燥、再結晶、各種クロマトグラフィー等により 行えばよい。
実施例

製造例 1
トリフルォロメタンスルホ-ルォキシ酪酸 n—ブチルの合成

[0038] (S)— 2—ヒドロキシ酪酸 n—ブチル(1. Og, 99%ee)を塩化メチレン(8mL)に溶 解し、 0°Cにてピリジン(518mg)をカ卩えた。次に 0°Cにてトリフルォロメタンスルホン酸 無水物(1.8g)を滴下し、 10分間攪拌した。反応液を直接的にシリカゲルカラムクロ マトグラフィ一に付し、塩化メチレンで流通後、減圧濃縮し、無色油状物を得た。 (1. 8g, 98.2%)

[0041] 3—ヒドロキシベンズアルデヒド(418mg)のァセトニトリル(10mL)溶液に、炭酸カリ ゥム(473mg)をカ卩え、続いて(S)—2—トリフルォロメタンスルホ-ルォキシ酪酸 n— ブチル(1. Og)を加え室温にて 12時間攪拌した。反応液に酢酸ェチルを加え、水、 飽和食塩水の順に洗浄し、無水硫酸ナトリウムで乾燥した。反応液を濾過し、減圧濃 縮後、残渣をシリカゲルカラムクロマトグラフィー(へキサン Z酢酸ェチル =4Zl)で 精製し、無色油状物を得た。 (904mg, 99.9%)

3- (4—メトキシフエノキシ)プロピオ-トリルの合成
[0044] 4—メトキシフエノール(263. Og)をアクリロニトリル(224. 8g)に溶解後、室温にて トリトン B (18mL)を滴下し、 80°Cにて 48時間撹拌した。反応液をゆっくりと撹拌しつ つ室温に戻し 12時間撹拌した。さらに 6°Cにて放置し、白色プリズム結晶を析出させ た。母液をデカンテーシヨンし、冷トルエン(300mL)を加え結晶をろ取し、得られた 粗結晶を室温'減圧乾燥(3時間)した (粗結晶 232. 6g) 0粗結晶を酢酸ェチル(25 OmL)に 50°Cで溶解し、ゆっくりと n—ヘプタン(250mL)を撹拌しながら加え、 12時 間ゆっくりと撹拌しつつ再結晶した。母液をデカンテーシヨンし、 n—ヘプタン(300m L)をカ卩ぇ結晶をろ取した。 n—ヘプタン (400mL)で結晶を洗浄し、室温にて減圧乾 燥した(1番晶:白色プリズム晶 154. 5g) 0母液に結晶が生じた為、 2番晶として淡黄 色プリズム晶を得た(2番晶:淡黄色プリズム晶 56. lg)。全てのろ液と洗浄 n—ヘプ タンを回収し減圧濃縮した後、残渣にトルエン(500mL)を加え、 1N—水酸化ナトリ ゥム水溶液(100mL X 3回)、水(500mL)、 IN—塩酸水溶液(100mL X 3)、水(5 OOmL)、飽和食塩水(300mL)で洗浄後、無水硫酸ナトリウムで 30分間乾燥し、ろ 過後、ろ液を減圧濃縮した。残渣を酢酸ェチル(150mL)に 50°Cで溶解し、ゆっくり と n—ヘプタン(150mL)を撹拌しながら加え、 12時間ゆっくりと撹拌しつつ再結晶し た。母液をデカンテーシヨンし、 n—ヘプタン(200mL)をカ卩ぇ結晶をろ取した。 n ブタン(300mL)で結晶を洗浄し、室温にて減圧乾燥した(3番晶:白色プリズム晶 59 . 0g)。更に 4番晶として淡黄色プリズム晶を得た (4番晶:19. 0g)。収量合計 288. 6g、収率 76. 9% +粗結晶(黄色プリズム晶、 21. 8g, 5. 8%)

製造例 3
3—(4ーメトキシフエノキシ)プロピルァミンの合成
[0047] アルゴン雰囲気下、 3— (4—メトキシフエノキシ)プロピオ-トリル(5. Og)をテトラヒド 口フラン(20mL)に溶解し、 80°Cにてボラン.テトラヒドロフラン錯体(1. 02mol/L, 30. OmL)を 10分間で滴下した。同温にて 3時間撹拌後、反応液を室温まで戻した 。氷冷下、 4N—水酸ィ匕ナトリウム水溶液(30mL)を 10分間でカ卩えた。 10分後室温 にて 5分間撹拌後、 80°Cにて 12時間撹拌した。室温に戻し、トルエン(lOOmL)をカロ え 1時間撹拌後、不溶物をセライトろ去した。有機層を分取し、水(100mL X 2回)、 飽和食塩水(lOOmL)で洗浄後、無水硫酸ナトリウム(80g)で乾燥し、ろ過後、ろ液 を減圧濃縮し、白色固体を得た。(4. Og, 79. 0%)

室温にて攪拌しながら(R) -2- (3 ホルミルフエノキシ)酪酸 n—ブチル(50mg)の メタノール溶液 2mLをカ卩ぇ 80°Cにて 12時間攪拌した。室温にて水素化ホウ素ナトリ ゥム(7mg)の水溶液 0.5mLを加え、 30分攪拌した。反応溶液を減圧濃縮後、クロ 口ホルムを加え有機層を水で洗浄した。無水硫酸ナトリウムで乾燥し、減圧濃縮した 後、淡黄色油状物 8 lmgを得た。このまま全量を次の反応に用いた。

[0053] (R)— 2— [3— [N— [3— (4—メトキシフエノキシ)プロピル]アミノメチル]フエノキシ ]酪酸 n—ブチル(81mg)をァセトニトリル(5mL)に溶解させ、室温にてトリエチルァ ミン(29mg)を滴下した。さらに 2 クロ口べンズォキサゾール(35mg)を加え 80°Cで 12時間攪拌した。水を加えた後、酢酸ェチルで抽出を行い、有機層を飽和食塩水 で洗浄した。無水硫酸ナトリウムで乾燥し、減圧濃縮した後、残渣をシリカゲルクロマ トグラフィー(へキサン Z酢酸ェチル = 5Z2)で精製し無色油状物を得た。 (83mg, 81%)

[0056] (R)— 2— [3— [[N— (ベンズォキサゾール一 2—ィル) N— 3— (4—メトキシフエ ノキシ)プロピル]アミノメチル]フエノキシ]酪酸 n—ブチル(83mg)をエタノール 3mL に溶解させ、 4N 水酸ィ匕ナトリウム水溶液 (0. lmL)を滴下した。室温にて 1時間攪 拌した後減圧濃縮し、 1N—水酸ィ匕ナトリウム水溶液を加え、ジェチルエーテルで洗 浄した。水層に 1N—塩酸をカ卩えて pHlに調整し、クロ口ホルムを用いて抽出した。有 機層を無水硫酸ナトリウムで乾燥し、減圧濃縮した後、残渣をプレパラティブクロマト グラフィー(クロ口ホルム Zメタノール =10Zl)で精製し無色固体を得た。 (72mg, 9 6. 1%)

以上より、出発原料の(S)—2—ヒドロキシ酪酸!1ーブチル(99%66)から1^^活 性化化合物である化合物 (A— 1) (参考例 3化合物)が高光学純度 (99%ee)で得ら れた。すなわち、上記反応工程において、(S)— 2—ヒドロキシ酪酸 n—ブチルの光 学純度は高く維持されており、本発明の方法においては、化合物 (A— 1)の製造中 間体である実施例 1ィ匕合物が、高収率で且つ高光学純度で製造できることは明らか である。

〔式中、 Rは炭素数 1〜6のアルキル基又は炭素数 7〜8のァラルキル基を示す。〕 で表される光学活性 2 -トリフルォロメタンスルホ-ルォキシ酪酸エステルを塩基の存 在下に反応させることを特徴とする下記式 (3):
