Abstract
本発明は、DNAやRNAなどの生物構成物質の配列を測定する場合などに使用される検出用基板X6に関するものである。この検出用基板X6は、基体16と、基体16の表面に形成された光反射膜26と、光反射膜26の表面に形成された透光性要素36と、透光性要素36の表面に形成され検体固定化膜46と、を備えている。光反射膜26は、たとえば金属膜66を含んだものとされ、好ましくは誘電体多層膜76をさらに含んだものとされる。透光性要素36は、ガラスなどにより柱状体にしてもよい。

技術分野
[0001] 本発明は、 DNAや RNAなどの生物構成物質の配列を測定する場合などに使用さ れる検出用基板に関する。
背景技術
[0002] DNAマイクロアレーをはじめとする検出用基板は、ガラス基体上にアミノシラン、ァ ミノアルキルシランまたはポリ一 L—リジンなどの検体固定ィ匕膜を設け、この検体固定 化膜上にオリゴヌクレオチドや cDNA等の検体を被着させた構成を有して 、る。この 検出用基板を用いる場合には、予め蛍光修飾した検体 DNAと反応させ、その反応 レベルにより変化する蛍光レベルを検出することによって、核酸配列の分析が行なわ れている。このような検出方法によれば、ガラス基体力も発せられるバックグラウンドの 蛍光は、分解能の低下の原因となる。特に、 DNAマイクロアレーでは、スタンフォー ド大学方式については安価ではある力 検出精度がァフィメトリックス社 (Affimetrix) タイプに比較して劣るため、検出精度向上のための感度向上が必要とされている。
[0003] 検出精度向上のための対策としては、検体固定化膜上に、ガラス基体から発せら れる蛍光を減衰させるためのブロッキング材ゃ消光材を被着したりすることが検討さ れている。
[0004] その一例として、図 12に示した検出用基板 Y1のように、ガラス基体 80の表面に検 体固定ィ匕膜 81を設けるとともに、検出精度向上のために、検体固定ィ匕膜 81を消光 剤によりコートすることが提案されており(たとえば特許文献 1参照)、また図 13に示し た検出用基板 Y2ように、ガラス基体 90上に設けた光反射膜 91の表面に、金属と結 合性の良い官能基を有する検体固定ィ匕膜 92を形成したものも提案されている (たと えば特許文献 2参照)。
[0005] しカゝしながら、図 12に示した検出用基板 Y1で採用されていた検体固定ィ匕膜 81に 消光剤やブロッキング剤をコートする方法は、消光剤やブロッキング剤が透光性の有 機物質のため、バックグラウンドの蛍光を完全に除去できないという問題点を有して
いた。また、図 13に示した検出基板 Y2のような光反射膜 91の表面に金属と結合性 の良い官能基を有する検体固定ィ匕膜 92を形成する方法は、光反射膜 91によりガラ ス基体 90の蛍光を遮断できるものの、 Auや Pt等の高価な貴金属を用いる必要があ るとともに、検体固定ィ匕膜 92として金属と結合性のよい官能基を有する特殊な材料 を用いる必要があり、比較的高価となることや、また Sや Se等を含む毒性物質を用い ることもあるため、取り扱いに注意を要すると 、う問題点を有して 、た。
[0006] 一方、誘電体多層膜を用いてバックグラウンドの蛍光遮断と測定蛍光の反射を試 みることも考えられるが、 40層力 60層と非常に多くの誘電体層が必要なため、誘電 体多層膜における光の吸収も大きくなつて反射率が金属ほど大きくならず、コンタミに よる不具合の発生も多くなる。
[0007] また、検出用基板には、複数の生体物質をアレー状に固定ィ匕した形態のものもある 力 アレーの密度は生体物質を固定ィ匕するスポットの面積に左右される。このため、 生体物質固定ィ匕膜としては、若干の撥水性を示すものが選択されるが、生体物質固 定ィ匕膜での水の接触角度が大きくなりすぎると、生体物質を固定ィ匕する際に用いる 溶液が弾かれてしまい、検出用基板に対して十分な量の生体物質を付着させること ができない。このため、生体物質固定化膜として若干の撥水性を示すものを選択す る場合には、測定蛍光強度が低下するという問題点があった。

発明の開示
発明が解決しょうとする課題
[0009] 本発明は、基体力も発せられるノ ックグラウンドの蛍光を遮断するとともに、蛍光の 強度を適切に高めることを課題としている。
[0010] 本発明の別の課題は、一般的に用いられている安価で安全な生体物質固定ィ匕膜 を反射膜上に緻密かつ強固に保持させることにある。
[0011] 本発明のさらに別の課題は、小さな面積に十分な量の生体物質溶液をスポットする ことによって高密度にアレー化された検出用基板を提供することにある。
課題を解決するための手段
[0012] 本発明の第 1の側面においては、基体上に、光反射膜、透光性要素、および検体 固定ィ匕膜が順次形成されている、検出用基板が提供される。
[0013] 本発明の第 2の側面においては、基体と、前記基体上に形成された撥水性膜と、前 記撥水性膜の表面に立設された複数の透光性柱状体力 なる透光性要素と、前記 透光性柱状体の表面に形成された検体固定化膜と、を備えている、検出用基板が提 供される。この場合に撥水性膜は、たとえば光反射膜を介して前記基体の表面に形 成される。

発明の効果
[0028] 本発明の第 1の側面の検出用基板では、基体上に、光反射膜、透光性要素および 検体固定ィ匕膜が順次設けられている。そのため、光反射膜において基体力ものバッ クグラウンドの蛍光を良好に遮断しつつ、検体固定ィ匕膜に結合した検体力 発せら れる蛍光を高効率で反射させることができるため、蛍光の利得を高くすることができる
。また、光反射膜上に透光性要素を設けていることから、透光性要素に対して検体固 定ィ匕膜を簡便に被着させることができるようになる。
[0029] 本発明の第 2の側面の検出用基板では、撥水性膜上に複数の透光性柱状体を立 設するとともに、透光性柱状体の表面に検体固定化膜を設けている。そのため、撥水 性膜によって生体物質など検体を含む検体溶液の広がりを押さえつつ、複数の透光 性柱状体によって検体固定ィ匕膜の表面積を大きく確保するとともに毛管現象によつ て透光性柱状体の間に検体溶液を入り込ませることができる。これにより、検体溶液 が検出用基板の表面から弾かれることを適切に抑制できるようになるとともに、検体固 定ィ匕膜の表面積を大きく確保できるため、検体溶液を小さいスポットに大量に付着さ せることが可能となる。その結果、検出用基板において、より多くの生体物質などの 検体を高密度に付着させることができるため、検体力 発せられる蛍光の強度を高め ることがでさるよう〖こなる。
[0030] また、光反射膜を介して撥水性膜を基体上に形成すれば、光反射膜にお!、て基体 力ものバックグラウンドの蛍光を良好に遮断するとともに、検体固定ィ匕膜に結合した 検体力 発せられる蛍光を高効率で反射させることができるようになる。
[0031] 本発明の検出用基板において、金属膜を、 Ti、 Cr、 Ni、 Au、 Ag、 Pt、 Rh、 Al、 Ni
Cr合金および Fe Cr Ni合金のうちの少なくとも 1種を主成分とするものとすれ ば、これらの金属が化学的に安定で耐薬品性に優れるため、検体測定プロセスに対 して適切に適用できる。また、例示した金属は、比較的高い可視光領域の反射率物 質のため、生体物質などの検体力 の蛍光を比較的高い効率で反射することができ る。
[0032] また、金属膜を、 Ti、 Cr、 ^^ 0:合金ぉょび 6— 0:—?^合金のぅちの少なくとも1 種を主成分とし、とくに Tiまたは Crを主成分とするものとすれば、金属膜を安価に形 成することができるとともに、金属膜が不動体ィ匕した酸ィ匕膜をもつものとすることがで きるため、金属膜が耐薬品性に優れたものとなる。そのため、金属膜が種々の薬品や 環境に曝されても腐食溶解せず、検体の固定から検体中の被検出成分 (検体 DNA など)の測定までの測定プロセスにおける検出用基板の劣化を有効に防止できる。そ の結果、測定プロセスの条件の変更にも多くの場合耐えられるため、適用範囲が広く
なる。さらに、金属膜の表面に透光性要素や撥水性膜を形成する場合には、透光性 要素や撥水性膜との密着性を高くすることができるようになる。このため、製造工程中 における透光性要素や撥水性膜の脱落を有効に防止できる。
[0033] 本発明の検出用基板において、金属膜の表面の算術平均粗さ Raを 0. 以 下とすれば、検体固定ィヒ膜に結合した生体物質力 発せられる蛍光の散乱反射を 効果的に阻止することができ、蛍光の利得をより高めることができるようになる。また、 金属膜の表面の粗さを先の範囲に設定すれば、検体固定化膜を被着する際に、表 面の凹凸によるコート液の塗布ムラが発生するのを有効に防止できるようになる。
[0034] 本発明の検出用基板において、光反射膜を、金属膜および誘電体多層膜からなる ものとすれば、金属膜が基体力 のバックグラウンドの蛍光を良好に遮断し、かつ、検 体固定ィ匕膜に結合した検体力 発せられる蛍光を高い効率をもって反射させること ができるようになる。その一方で、誘電体多層膜においては、蛍光体から発せられた 蛍光が誘電体多層膜を構成する個々の誘電体膜の表面で反射して相互に干渉し、 また誘電体膜の反射光と金属膜の表面で反射光が相互に干渉する。これにより、金 属膜および誘電体多層膜からなる光反射膜を採用した場合には、反射光を適切に 増強することができるため、蛍光の検出精度をきわめて高くすることができる。
[0035] とくに、誘電体多層膜を構成する個々の誘電体膜の厚みを、検体固定化膜に修飾 される生体物質などの検体力 発せられる蛍光の波長と誘電体膜の屈折率との積の (2n— 1) Z4倍 (n:自然数)とすれば、各誘電体膜や金属膜の表面での反射光の干 渉がより適切に起こるため、蛍光の増幅を非常に良好に行なうことができるようになる 。そのため、本発明の検出用基板では、検体から蛍光の強度を効率良く高めて出射 でさるよう〖こなる。
[0036] また、誘電体多層膜の厚みを 1 μ m以下とすれば、誘電体多層膜での蛍光の吸収 を適切に回避できるために、蛍光の増幅効率の悪ィ匕を抑制することができる。
[0037] 本発明の検出用基板において、誘電体多層膜を、酸ィ匕タンタル膜,酸化チタン膜, フッ化カルシウム膜,酸化ジルコニウム膜および酸化ニオブ膜のうちの少なくとも 1つ の誘電体膜を含むものとすれば、様々な蛍光波長に対応できる反射光増加用の誘 電体多層膜を形成することができる。また、先に例示した誘電体膜は、きわめて精密
に膜厚みコントロールが可能であるため、金属膜やガラス膜などの透光性要素との密 着性が良好となるように表面物性を容易に制御することができる。さらに、先に例示し た誘電体膜は、化学的にも安定なため、生体物質などの検体の反応処理の際にもさ ほど大きなダメージを受けることもな 、。
[0038] 本発明の検出用基板において、透光性要素を、酸化珪素の含有量が 70質量%以 上となるように形成すれば、透明性に優れた透光性要素を安価に形成することが可 能となる。また、従来のガラス上に用いられる検体固定ィ匕膜などの検体固定ィ匕物質を そのまま用いることができるため、特殊な官能基を有する検体固定ィ匕物質を用いる必 要はない。そのため、透光性要素の酸ィ匕珪素の含有量を 70質量%以上とすれば、 材料コストを低減することができるようになる。
[0039] 本発明の検出用基板において、透光性要素の厚みを検体固定ィ匕膜に修飾される 検体力 発せられる蛍光の波長と透光性要素の屈折率の積の 1Z4より小さくすれば 、検体固定ィ匕膜に結合した検体に修飾された蛍光体力 発する蛍光のうち、直接外 部に発せられた蛍光と、透光性柱状体を通過して基体表面で反射された反射光とが 干渉し増幅するのを有効に抑制することができる。そのため、検出用基板から出射さ れる蛍光強度を、検出用基板における異なる位置において均一化することができるよ うになり、蛍光強度の測定値に不連続点が生じるのを有効に防止できる。これにより、 蛍光強度が位置によってばらつくことなぐ安定した測定を行なうことができる。

[0058] また、光反射膜 21は、透光性膜 31との膜密着性を良好なものとするために、たとえ ば Auや Ag等の下地層の上に Tiや Cr等の密着層を形成したような複数の層からなる 構成であってもよい。光反射膜 21はまた、蛍光体の励起のための励起光や蛍光体 から出射される出射光を反射する皮膜であればよぐ基体 11や透光性膜 31との密着 性を確保できる限りは、金属膜以外のものを使用することもできる。
[0059] 光反射膜 21の厚みは、 0. 05〜1 μ mとするのが好ましい。これは、光反射膜 21の 厚みが小さすぎる場合には、たとえば光反射膜 21を金属膜として形成する場合には 、光透過膜 21が光を透過するアイランド構造となってしまう一方で、光反射膜 21の厚 みを 0. 05 m以上とすれば、光を透過しない膜構造とすることができるからであり、 また、光反射膜 21の厚みが 1 μ mを超えると、成膜方法にもよるが、光反射膜 21の 内部応力によりガラス製などの基体 11の破壊による膜剥がれが生じやすくなるため である。好ましくは、光反射膜 21の厚みは、 0. 程度とされる。そうすれば、光反 射膜 21の厚みが 0. 程度の場合には、基体 11との膜密着性も比較的に高ぐ また、基体 11をガラスにより形成する場合に、薬液の浸透による基体 11のガラスのコ ロージヨンが発生しなくなるからである。

形成するのが好ましい。このような表面粗さを有する光反射膜 21では、その表面に凹 凸の少ない透光性膜 31を形成することが可能となる。その結果、透光性膜 31に検体 固定ィ匕膜 41を被着する際に、透光性膜 31の表面の凹凸に起因するコート液の塗布 ムラが発生するのを有効に防止することができる。また、光反射膜 21における表面粗 さを小さくすれば、検体固定化膜 41に結合した生体物質などの検体から発せられる 蛍光が光反射膜 21にお 、て乱反射してしまうのを抑制できるために、蛍光の利得を 高めることができるようになる。
[0061] 透光性膜 31は、検体固定ィ匕膜 41を固着させるためのものであり、光反射膜 21の 表面に膜形成されている。この透光性膜 31は、検体固定ィ匕のための物質が良く固着 される材料、たとえばガラス材料により、蛍光の波長の吸収の少ない透明に形成され ている。透光性膜 31は、その厚みを検体に修飾される蛍光体の発光波長の 1Z4より も小さくなるように形成するのが好ましい。これは、透光性膜 31の厚みが蛍光体の波 長の 1Z2であれば、干渉による吸収が生じる一方で、透光性膜 31の厚みが蛍光体 の波長の 1Z4の厚みであれば、光の干渉による増幅が有るために僅かな膜厚みの ばらつきが蛍光強度の測定のばらつきを大きくするからである。

[0071] また、透光性柱状体 32Aは、その厚み (高さ)が検体固定ィ匕膜 42に固定される検 体から発せられる蛍光の波長と、透光性柱状体 3の屈折率と、の積の 1Z4倍よりも小 さくなるように形成するのが好ましい。この場合には、検体力も発せられる蛍光のうち 、直接外部に発せられた蛍光と、透光性柱状体 32Aを通過して光反射膜 22で反射 された反射光とが干渉して増幅するのを有効に抑制することができる。そのため、検 出用基板 X2における全ての位置において、蛍光強度を均一化にし、蛍光強度の測 定値に不連続点が生じるのを有効に防止できる。その結果、検出用基板 X2における 異なる位置において、蛍光強度がばらつくことを抑制できるようになり、安定した測定 を行なうことができるようになる。

実施例 1
[0125] 本実施例では、本発明に係る第 1の実施の形態の検出用基板 XI (図 1および図 2 参照)において、検体固定ィ匕膜 41を透光性膜 (ガラス膜) 31の全面に形成した検出 用基板について評価した。
[0126] 本実施例で用いる検出用基板は、次のようにして作成した。
[0127] まず、基体 11として、硼珪酸ガラスにより、サイズが 26mm X 76mm X 1. 1mmに 形成されたものを準備した。基体 11は、アルカリ脱脂を行って十分に純水洗した後、 希塩酸にて中和処理を行い、十分に純水洗し、さらに希弗酸液にてガラス表面をライ トエッチングした後、十分に純水洗を行った。その後、アルコール置換した後にアル コール蒸気乾燥を行った。
[0128] 乾燥した基体 11に対しては、スパッタ装置にて光反射膜 (金属膜) 21としての Ti膜 を、透光性膜 (ガラス膜) 31としての SiO膜を順次、所定の厚みに成膜した (下記表

[0132] 作成された検出用基板に対しては、オリゴ DANを固定ィ匕した。オリゴ DANとしては 、 5,末端を Cy3蛍光体にて修飾した 15塩基長のものを用いた。このオリゴ DNAは、 0 . 5 Mとなるように純水に溶解してスポット液とした後に検出用基板にスポットし、湿 潤箱に 25°Cにて 12時間放置することにより検出用基板に固定した。オリゴ DNAを固 定ィ匕した検出用基板は、クェン酸緩衝液にて洗浄した後に純水リンスを行い、ブロー 乾燥させた。
[0133] 一方、比較例として、硼珪酸ガラスに検体固定ィ匕膜を直接形成したものを使用した 。硼珪酸ガラスの処理および検体固定ィ匕膜の形成については、本発明の検出用基 板と同様にして行なった。

[0138] また、表 1の No. 7〜No. 14の結果力も分力るように、透光性膜 (ガラス膜) 31の厚 みが 20〜125nmの場合には、蛍光強度が高ぐ好適な結果が得られた。特に、透 光性膜 (ガラス膜) 31の厚みが 30〜125nmの場合には、蛍光強度について非常に よい結果が得られた。その一方で、透光性膜 (ガラス膜) 31の厚みが Cy3の蛍光波 長 538nmの 4分の 1波長となる 135nm前後の厚みの場合には、光の干渉による部 分的な蛍光強度のばらつきが発生していた。このばらつきは解析上不利なため、透 光性膜 (ガラス膜) 31の厚みは、検体力も発せられる蛍光の波長の 1Z4より小さいこ とがより好ま U、ことがわ力つた。
[0139] さらに、表 1の No. 18〜No. 20の結果力 分力るように、透光性膜 (ガラス膜) 31 の材質については、透光性膜 (ガラス膜) 31をガラスにより形成する場合には、酸ィ匕 珪素の質量が 70%以上のときに蛍光強度について良い結果を示した。その一方で 、酸ィ匕珪素の質量が 63%では蛍光強度が弱力つた。そのため、透光性膜 (ガラス膜) 31の酸ィ匕珪素濃度は、 70質量%以上がより好ましいことがわ力つた。

性膜 (ガラス膜) 36, 37の膜厚みばらつき起因と考えられる蛍光強度のばらつきが発 生した。このため、透光性膜 (ガラス膜) 36, 37の厚みは、測定波長と透光性膜 (ガラ ス膜) 36, 37の屈折率との積の 1Z4よりも小さいほうがより好ましいことがわ力つた。

[0162] 本実施例で用いる検出用基板は、次のようにして作成した。まず、基体 18, 19とし て、実施例 1と同様に硼珪酸ガラス製のものを用い、実施例 1と同様に基体 18, 19の 前処理を行い、必要に応じて、実施例 1と同様にして金属膜 69の形成を行なった。さ らに、必要に応じて、実施例 2と同様にして金属膜 69の上面に誘電体膜多層膜 79と 成る 2種の誘電体膜を光学薄膜用蒸着装置によって交互に成膜した。なお、誘電体 多層膜 79を構成する誘電体膜の厚みは、使用する検体に化学的に修飾される蛍光 体の蛍光波長と誘電体膜の屈折率との積の 1Z4倍となるようにした。
[0163] 次に、基体 18または誘電体膜多層膜 79の上面に撥水性膜 58, 59を形成した。撥 水性膜 58, 59は、撥水性樹脂より形成する場合には、撥水性榭脂を含む原料液体 をスピンコートにより塗布した後に乾燥を行い、必要に応じてキュアを行なって撥水性 榭脂を熱硬化させた。また、撥水性膜 58, 59は、表面を弗化物ガスにてプラズマ処 理して撥水化処理を行うことによつても形成した。さらに、撥水性膜 58, 59としてフッ 化膜を形成する際には誘電体膜多層膜 79と同様に光学薄膜用蒸着装置によって成 膜した。
[0164] 次に、撥水性膜 58, 59の上面に、多数の透光性柱状体 38A, 39Aとしての SiO
2 柱状体を所定の厚みに成膜してガラス膜を形成した。 SiO
2の成膜に当たっては膜が 稠密とならな ヽように加熱温度や成膜速度および真空度にっ ヽて配慮した。
[0165] 成膜の終わった基体 18, 19は RC A洗浄を行って十分に純水洗浄した後、アルコ ール置換し、アルコール蒸気乾燥を行った。

しただけでも、測定された蛍光強度は 400以上と高い値の測定がなされた。なお、撥 水性膜 58の厚みや材料により、測定値は振れているが、本発明の本質に関わる物 ではない。
[0171] また、表 6の No. 13〜21の結果力も分力るように、撥水性膜 59の下地に金属膜 6 9を着膜した場合には、いずれも蛍光強度が 1400以上とより高い値を示した。なお、 金属膜 69の材料により、測定値は振れているが、本発明の本質に関わる物ではない 。また、 No. 1〜12にて撥水性膜 58の効果は明らかなため、撥水性膜 59としては弗 化マグネシウムを代表として記載した。
[0172] さらに、表 6の No. 22〜28の結果力も分力るように、金属膜 69と撥水性膜 59の間 に誘電体反射膜 79を挿入して蛍光強度を測定した場合には、 1500以上の更に高 い蛍光強度が測定された。

[0174] 表 7から分力るように、撥水性膜 58の表面に対する水の接触角度に依っての蛍光 強度の変化が観測された。撥水性膜 58の表面に対する水の接触角が 50°以下とな るように処理した Ti膜の場合、蛍光強度は低いが、接触角が 50°を越えると蛍光強度 が高くなることが分かる。これは、撥水性膜 58を設けることにより、隣接する透光性柱 状体 38Aの間での毛管現象が阻害され、オリゴ DNAを含んだ溶液が広がらな 、た めである。そのため、撥水性膜 58 (59)としては、接触角としては 50°以上であるのが 好ましいことが分かる。

実施例 4
[0181] 本実施例では、本発明に係る検出用基板の第 10の実施の形態における検出用基 板 X10 (図 11参照)について評価した。
[0182] 本実施例で用いる検出用基板は、次のようにして作成した。まず、基体 110として、 実施例 1と同様に硼珪酸ガラス製のものを用い、実施例 1と同様に基体 110の前処 理、光反射膜 610としての金属膜の形成を行なった。光反射膜 (金属膜) 610は、 Cr により形成した。
[0183] 次に、光反射膜 610の上面に弗化マグネシウム力も成る撥水性膜 510を光学薄膜 用蒸着装置によって成膜した。なお、撥水性膜 510の被着の際は成膜条件を調整 することにより、島状(図 11の態様)や膜状に形成した。
[0184] 次に、撥水性膜 510の隙間に透光性柱状体 310Aとして SiOを成膜した。この際、

SiOの成膜に当たっては膜が稠密とならないように加熱温度や成膜速度および真

V、薄片状の透光性柱状体 310Aも形成した。

[I] 基体上に、光反射膜、透光性要素、および検体固定化膜が形成されている、検出 用基板。

前記基体上に形成された撥水性膜と、
前記撥水性膜の表面に立設された複数の透光性柱状体からなる透光性要素と、 前記透光性柱状体の表面に形成された検体固定化膜と、
を備えている、検出用基板。
[3] 前記撥水性膜は、光反射膜を介して前記基体の表面に形成されて!ヽる、請求項 2 記載の検出用基板。
[4] 前記光反射膜は、金属膜を含んでいる、請求項 1または 3に記載の検出用基板。
[5] 前記金属膜は、 Ti、 Cr、 Ni、 Au、 Ag、 Pt、 Rh、 Al、 ^^ー0:合金ぉょび 6— 0:—
Ni合金のうちの少なくとも 1種を主成分として ヽる、請求項 4に記載の検出用基板。
[6] 前記金属膜は、 Ti、 Cr、 Ni—Cr合金および Fe— Cr—Ni合金のうちの少なくとも 1 種を主成分としている、請求項 5に記載の検出用基板。
[7] 前記金属膜は、 Tほたは Crを主成分として 、る、請求項 6に記載の検出用基板。
[8] 前記金属膜は、その表面の算術平均粗さ Raが 0. 05 μ m以下とされて 、る、請求 項 4に記載の検出用基板。
[9] 前記光反射膜は、前記金属膜の表面に形成された反射光増幅用の誘電体多層膜 をさらに含んで!/、る、請求項 4に記載の検出用基板。
[10] 前記誘電体多層膜は、酸ィ匕タンタル膜、酸化チタン膜、フッ化カルシウム膜、フッ化 マグネシウム膜、酸ィ匕ジルコニウム膜および酸ィ匕ニオブ膜のうちの少なくとも 1つの誘 電体膜を含んで 、る、請求項 9に記載の検出用基板。
[II] 前記誘電体多層膜を構成する個々の誘電体膜は、その厚みが前記検体固定化膜 に化学的に修飾される検体から発せられる蛍光の波長と前記誘電体膜の屈折率との 積の(2n— 1) Z4倍 (n:自然数)とされている、請求項 9に記載の検出用基板。
[12] 前記誘電体多層膜は、その厚みが 1 μ m以下である、請求項 9に記載の検出用基 板。
[13] 前記透光性要素は、酸ィ匕珪素の含有量が 70質量%以上とされている、請求項 1ま たは 2に記載の検出用基板
[14] 前記透光性要素は、その厚みが前記検体固定ィ匕膜に修飾される検体力 発せら れる蛍光の波長と透光性要素の屈折率との積の 1Z4より小さくされている、請求項 1 または 2に記載の検出用基板。
[15] 前記透光性要素は、前記光反射膜の表面に立設させた複数の透光性柱状体から 成る、請求項 1に記載の検出用基板。
[16] 前記透光性柱状体は、羽根状である、請求項 2または 15に記載の検出用基板。
[17] 前記透光性柱状体は、温度 25°C、湿度 60%のときの 1平方メートル当たりの表面 吸着水量が 0. 5g以上である、請求項 2または請求項 15に記載の検出用基板。
[18] 前記複数の透光性柱状体は、隣接するもの同士が互いに接している、請求項 2ま たは請求項 15に記載の検出用基板。
[19] 前記複数の透光性柱状体は、隣接するもの同士が互いに間隔をあけて設けられて いる、請求項 2または請求項 15に記載の検出用基板。
[20] 互いに隣接する透光性柱状体の間に存在する撥水性膜を備えている、請求項 19 に記載の検出用基板。
[21] 前記撥水性膜の表面が前記透光性柱状体の上端面よりも低くされている、請求項

[22] 前記撥水性膜と前記透光性柱状体との高さの差は、 30nm以上である、請求項 21 に記載の検出用基板。

[24] 前記凹部は、その内面の縦断面形状が放物線状とされている、請求項 23に記載の 検出用基板。
[25] 前記凹部の開口部の全周を囲むように前記検体固定ィ匕膜の上面に形成された撥 水性膜をさらに備えて 、る、請求項 23に記載の検出用基板。
[26] 前記撥水性膜は、その表面における水の接触角が 50°以上である、請求項 2、請 求項 20または請求項 25に記載の検出用基板。
[27] 前記撥水性膜は、フッ素榭脂、ポリプロピレン榭脂、ポリエチレン榭脂、ポリスチレン 榭脂、ポリイミド榭脂、ポリアセテート榭脂、ポリウレタン榭脂、エポキシ榭脂、フエノー ルノボラック榭脂、および無機フッ化物の少なくとも 1種を主成分としている、請求項 2
、請求項 20または請求項 25に記載の検出用基板。
[28] 前記検体固定化膜は、アミノ基、アルデヒド基、メルカプト基、シラン基およびカルボ キシル基のうちの少なくとも 1種を有している、請求項 1に記載の検出用基板。
