被検体溶液は、 生体分子素子 4 0 0内を通過し、 排出される。 その際、 被検体 溶液が、 上述の生体分子材料 4 1 0に対して何らかの反応を示す物質を含み得る 場合、 生体分子材料 4 1 0は、 被検体溶液中の物質と反応する、 その物質に対し て触媒作用をもたらす、 または、 その物質以外の物質を不活性化する等し得る。 そのような変化を生体分子素子 4 0 0が特定の物質を検知したことを示す信号 (すなわち、 電流、 電圧、 光量、 質量、 熱量等の変化に相当する信号) を検知手 段に通知する。 検知手段は、 生体分子素子 4 0 0からの信号に基づいて、 被検体 溶液に特定の物質が存在することを表示し得る。 被検体溶液が、 上述の生体分子材料 4 1 0に対して反応を示す物質を含み得な い場合、生体分子材料 4 1 0は、特定の物質を検知しなかったことを示す信号(す なわち、 電流、 電圧、 光量、 質量、 熱量等の変化ゼロに相当する信号) を検知手 段に通知する。 検知手段は、 生体分子素子 4 0 0からの信号に基づいて被検体溶 液に特定の物質が存在しないことを表示し得る。 なお、 異なる物質に対して反応する生体分子材料 4 1 0をそれぞれ固定化させ た、 複数の生体分子素子 400を組み合わせることによって、 同時に数種類の物 質を検出することも可能である。 次に具体的な実施例を用いて本発明を詳述するが、 本発明がこれら実施例に限 定されないことに留意されたい。 実施例 1 ;
図 1を参照して説明した本発明による製造方法を用いて、 炭素多孔体を製造し た。 ケージ型シリカ多孔体として 150°Cで作製された K I T-5 (以降では K I T—5— 150と示す) 1 g、 炭素源としてショ糖 0. 75 gを準備し、 これ らを濃硫酸 0. 8 gおよび水 2. 5 gに混合させた。 ここで、 シリカ多孔体中の ケィ素 (S i) と炭素源中の炭素 (C) とのモル比は、 CZS 1 = 1. 5であつ た。次いで、混合物を大気中 100°C、 6時間、オープン中で加熱した。 さらに、 160 まで昇温し、 6時間保持して、 完全に重合させた。 得られた重合体を 5 Oml/分で窒素フローしながら、 877°Cで 6時間保持し、 炭化させた。 5重 量%フッ酸を用いて、 K I T— 5— 150を溶解除去した後、 反応物 I CY (以 降では I C Y— 1と称する)をエタノールで数回洗浄し、 120°Cで乾燥させた。 得られた反応物 I CY_ 1について、 X線回折装置 (S i eme n s D 50 05, B r uc ke r AXS、 UK) を用いて構造解析を行った。 X線回折装 置の動作条件は、 Cu—Ko!線を用いて、 40 kV/50mA、 0. 5° 2 Θ / 分の走査速度であった。 用いた K I T— 5— 150の X線回折パターンと、 I C Y— 1の X線回折パターンとを比較した。 高分解能透過型電子顕微鏡 (J EOL- 3000 Fおよび J EOL-3100 FEF、 日本電子、 J ap an) を用いて、 I C Y— 1の観察を行った。 I CY _1を、 乳鉢を用いて粒子状にし、 それを Cu製格子上に位置する穴の開いた炭 素膜上に分散させて試料を調整した。 透過型電子顕微鏡の動作条件は、 加速電圧 300 kV、 分解能 15万〜 120万倍であった。 比表面積 ·細孔分布測定装置 (Au t o s o r b 1, Quan t ac h r o me、 USA) を用いて窒素吸脱着等温線を測定した。 1。丫ー1を5231:で 圧力 10— 5 h P a以下で 3時間脱気した後、 77 Kにて測定した。 吸脱着等温 線を測定することにより、 細孔有無、 細孔の形状および大きさが分かる。 以上の 結果を以下図 5〜図 9に示し詳述する。 実施例 2 ;

ケージ型シリカ多孔体として 100°Cで作製された K I T— 5 (以降では K I T一 5— 100と示す) を用いた以外は、 実施例 1と同様の製造条件、 同様の製 造プロセスによって処理した。 得られた反応物 I CY (以降では I CY— 1 ' と 示す) について、 X線回折装置を用いた構造解析、 および、 比表面積 ·細孔分布 測定装置を用いた窒素吸脱着等温線測定を行った。 以上の結果を以下図 5および 図 7に示し詳述する。 実施例 3 ;
(以降では I CY— 3と示す) について、 X線回折装置を用いた構造解析、 およ び、 比表面積 ·細孔分布測定装置を用いた窒素吸脱着等温線測定を行った。 次いで、 I CY— 3のタンパク質 (リゾチーム) 吸着特性を調べた。 pHl l のリゾチーム緩衝水溶液 (濃度は 1 から 280 ΠΙΟ 1 の範 囲) 4gに、 2 Omgの I CY— 3を分散させた。 分散液を 20° (:、 速度 160 回 分で攪拌させながら約 96時間保持した。 P及着平衡に達した後、 上澄み液の 紫外光吸収 (281. 5 nm) を、 紫外可視分光光度計 (S h i ma d z uUV -3150, 島津製作所、 J ap an) で測定した。 なお、 参考のために、 既知 の炭素多孔体 CMKの吸着特性と、 I C Y— 3の吸着特性とを比較した。 さらに、 I CY_ 3によって吸着されたタンパク質の安定性を調べるために、 タンパク質の吸着前後における赤外吸収スぺクトルをフーリエ変換赤外分光光度 計 (N i c o l e t Nexu s 670、 The rmo E l e c t r on、 U S A)を用いて測定した。測定波長領域は、 400 cm―1〜 950 cm— 1であった。 以上の結果を以下図 8〜図 11に示し詳述する。 実施例 4 ;
炭素源としてショ糖を 1. 2 g用いた以外は、 実施例 1と同様であるため説明 を省略する。 この場合のシリカ多孔体中のケィ素(S i) と炭素源中の炭素(C) との関係は、 C/S i =2. 4であった。 実施例 4で得られた反応物 I CY (以 降では I CY— 4と示す)について、 X線回折装置を用いた構造解析、比表面積' 細孔分布測定装置を用いた窒素吸脱着等温線測定、 および、 紫外可視分光光度計 を用いたリゾチームの吸着特性測定を行った。 以上の結果を以下図 8〜図 10に 示し詳述する。 比較例 2 ;
炭素源としてショ糖を 2. 0 g用いた以外は、 実施例 1と同様であるため説明 を省略する。 この場合のシリカ多孔体中のケィ素(S i) と炭素源中の炭素(C) との関係は、 CZS i =4. 0であった。 比較例 2で得られた反応物 I CY (以 降では I C Y— 2 'と示す)について、 X線回折装置を用いた構造解析、および、 比表面積 ·細孔分布測定装置を用いた窒素吸脱着等温線測定を行った。 以上の結 果を以下図 8〜図 9に示し詳述する。 図 5は、 (a)焼成温度の異なるケージ型シリカ多孔体 KI T一 5 (KI T— 5 — 150、 KI T一 5— 130および KI T— 5—100) それぞれの X線回折 パターンと、 (b) I CY ( I CY- 1, I CY - 2および I CY— 1 ') それぞ れの X線回折パターンとを示す図である。 図 5 (a) に示されるように、 実施例 1、 3〜4、 および、 比較例 2で用いた 焼成温度 150°Cの K I T一 5— 150、 実施例 2で用いた焼成温度 130°Cの KI T— 5— 130、 および、 比較例 1で用いた焼成温度 100°Cの KI T—5 一 100のいずれも、 20 = 0. 7〜 3の範囲において、 面心立方格子 (空間群 Fm6m) の (111)、 (200) および (220) の回折に相当するピークが 確認された。 なお、 焼成温度が高温になるにしたがって、 回折ピークは低角側に シフトするとともに回折強度は大きくなった。 回折ピーク(111)から格子定数 aQを求めた。それぞれの結果を表 1に示す。 表 1に示されるように、 KIT— 5—100、 KI T_5— 130および KIT 一 5— 150の格子定数 a。は、 それぞれ、 18. 1 nm、 19. 0 nmおよび 2 0. 7 nmであった。焼成温度が高温になるにしたがって、格子定数 aQは長くな つていることが分かった。 用いた KIT一 5の比表面積、 孔容量、 直径 d, (図 2 の直径 に相当)、 および、 径 2 (図 2の径 に相当) について調べた結果も 表 1に示す。 表 1 ;
Figure imgf000017_0001
I CY— 1 I CY- 2および I CY— の格子定数 a。 いず れの i CY_l I CY—2および I CY— 1 ' も残留 KI T— 5は、 1重量% 1. 5重量%程度であることが分かった。 好ましい K I T一 5の格子定数 a。 図 7は、 I CY— 1、 I CY— 2および I CY— 1 ' の (a) 窒素吸脱着等温 線および (b) 細孔分布を示す図である。 図 7 (a) に示されるように、 I CY 一 1および I CY— 2の窒素吸脱着等温線は、 H 2型 ( I U P A C分類における
一方、 I CY- 1 'の窒素吸脱着等温線は、 I I型(I UP AC分類)であり、 ヒステリシスを示さなかった。
D2=a0X (6 B /% V ) 1/3 … (1)
I CY— 1、 I CY— 3、 I CY— 4および I CY— 2, の X線回折 パターンを示す図である。
I CY— 1、 I CY— 3および I CY— 4の格子定数 a Qは、それぞれ 20. 68 nm, 19. 98 nmおよび 19. 48 nmと K I T— 5の格子定数と同様 の値を有していた。 一方、 I CY— 2, の格子定数 a。 I CY—1、 I CY— 3、 I CY— 4および I CY— 2, の窒素吸脱 着等温線を示す図である。 表 2に示されるように、 I CY _1、 I CY— 3および I CY—4の比表面積、孔容量、細孔径0,およびケージ 径132は、それぞれ、 1515m2Zg、 2. 0 cm g, 5. 2nmおよび 15. このことから、 K I T一 5を用いた場合、上記モル比は、関係 8<C/S i<3. 0を満たし、 より好ましくは、 関係 0. 85≤C/S i≤0. 95を満たす。 表 2
Figure imgf000021_0001
ns=Knfflc/ (1 +Kc) … (2)
