[0002] 近年、腫瘍免疫学において、免疫細胞による腫瘍抗原認識機構がかなり解明され てきた。それによると、まず、抗原提示細胞である榭状細胞(dendritic cellsまたは D C)は、細胞内で、腫瘍が発現するタンパク質を分解する際に生じた 8〜: 10個のァミノ 酸力 なる抗原ペプチドを、主要組織適合性抗原複合体(major histocompatibility c omplexまたは MHC ;ヒトでは、 human leukocyte antigenまたは HLA)と共に細胞 表面に提示する。細胞障害性 T細胞(cytotoxic T lymphocyteまたは CTL)は、榭状 細胞表面の HLAクラス Iに結合した抗原ペプチドを認識し、活性化'増殖し、腫瘍内 に侵入し、抗原ペプチドが由来するタンパク質を有する腫瘍細胞に対し細胞障害を 生じる( Arch. Surg. (1990) 126: 200〜205 )。
[0003] この機構を利用して、腫瘍の治療方法として癌ワクチンが開発されてきた。例えば、 腫瘍特異的タンパク質由来の抗原ペプチドを細胞表面に提示する樹状細胞を in vitr oで作製し、増殖させ、腫瘍患者に投与したり、その樹状細胞によって教育された細 胞障害性 T細胞を投与したりすることにより、腫瘍患者の体内で腫瘍免疫を誘導させ る。あるいは、腫瘍特異的タンパク質を腫瘍患者に投与し、患者の体内で、腫瘍免疫 機構の全過程を誘導させるのである( Science (1991) 254: 1643 - 1647; Exp. Med. (1 996) 183: 1185 - 1192; J. Immunol. (1999) 163: 4994- 5004; Proc.Natl.Acad.Sci.US A (1995) 92: 432 -436; Science (1995) 269: 1281— 1284; J.Exp.Med. (1997) 186: 7 85— 793)。

例えば、メラノーマ抗原 gplOOペプチドをメラノーマ患者に皮下投与し、インターロイ キン 2を静脈内投与することにより、 42%の患者で腫瘍の縮小が認められている( Nature Medicine (1998) 4: 321)
発明の開示
発明が解決しょうとする課題
[0005] しかし、腫瘍免疫を効率的に誘導することのできる腫瘍特異的タンパク質は、一部 の腫瘍において、ほんの少数の例が知られているだけである。
[0006] そこで、本発明は、腫瘍免疫を効率的に誘導できるペプチド、そのペプチドを含有 する組成物、そのペプチドを提示した抗原提示細胞、この抗原提示細胞によって刺 激された τ細胞、およびこれらのペプチドや細胞を利用した癌ワクチン、及びそれら を用いた腫瘍患者の治療方法を提供することを目的としてなされた。
課題を解決するための手段
[0007] 本発明者らは、転写因子である SOX6が、ダリオ一マに発現していることを既に見 いだしていたが(Ueda et al" Oncogene 23, 1420-1427, 2004)、他の腫瘍においても 発現していることを見出した(実施例 2参照)。この知見に基づき、 SOX6の様々な部 分ペプチドに対し、 HLAクラス I拘束性に細胞障害性 T細胞に認識されるかどうか調 ベたところ、樹状細胞に添加したとき、効率よく細胞障害性 T細胞によって認識される レ、くつかの部分ペプチドを同定することができ、本発明の完成に至った。
[0008] 本発明にかかるペプチドは、配列番号:!〜 3のいずれかの配列を有する。これらの ペプチドを提示した抗原提示細胞、及びこの抗原提示細胞によって誘導され、 SOX 6を発現する細胞を認識する T細胞も、本発明の技術的範囲に属する。この T細胞は 細胞障害性 T細胞であることが好ましぐ SOX6を発現する細胞は、ダリオ一マ、肝癌 細胞、肺癌細胞、膝癌細胞、食道癌細胞、メラノーマ細胞、前立腺癌細胞、乳癌細 胞、腎癌細胞、または白血病細胞であることが好ましい。
[0009] さらに、本発明に力、かる癌ワクチンは、配列番号:!〜 3から選択される一つ以上のぺ プチド、上記抗原提示細胞、上記 T細胞のうち、少なくとも一つを含有する。この癌ヮ クチンは、 SOX6を発現する癌細胞に対する癌ワクチンであり、特に細胞ダリオ一マ、 肝癌、肺癌、陴癌、食道癌、メラノーマ、前立腺癌、乳癌、腎癌、または白血病に対 する癌ワクチンであることが好ましレ、。

ウェブサイト上のテータベースである Biolnformatics & Molecular Analysis Section ( BIMAS)の HLA Peptide Binding predictions Programを用い、 SOX6遺伝子がコード
するアミノ酸配列から、 HLAクラス Iと結合性が高レ、、 9〜: 10アミノ酸残基のペプチド 配列を特定した。 日本人の HLAクラス Iのタイプは、 ^1し八ー八2と11し八ー八24が多く 、これらで日本人全体の約 80%をカバーする。なお、 日本人に多い HLA— Aの遺 伝子型として、 HLA—A2は A0201及び A0206、 HLA— A24は A2402が挙げら れ、本明細書では、細胞の遺伝子型として A0201、 A2402と記載した場合、記載し たアレルを少なくとも一つ有することを意味する。
[0016] ここでは、 A24に対する結合性についてスクリーニングした。表 1に、このスクリー二 ングによって得られたスコアの高いペプチド配列の例を示す。
[0018] これらのペプチドを実際に合成し、抗原として抗原提示細胞である樹状細胞表面に 投与したとき、 HLAクラス I分子に結合することにより細胞表面に提示され、細胞障害 性 T細胞に認識されることで特異的な細胞障害性 T細胞を誘導できるペプチドを同 定するため、樹状細胞と共培養された細胞障害性 T細胞のヒト ·ダリオ一マ細胞に対 する反応性を調べた。その結果、配列番号 1〜3のペプチドを細胞表面に提示する 樹状細胞が効率よく細胞障害性 T細胞を刺激し、また各ペプチドの刺激により樹立さ れた細胞障害性 T細胞が SOX6を発現するダリオ一マ細胞を効率よく認識することが 明らかになった。
[0019] 以下の実施例には、これらの実験結果を示すことにより、配列番号:!〜 3のペプチド を抗原提示細胞に添加すると、それらのペプチドが HLA上に提示され、共培養した T細胞を刺激することにより抗原特異的な T細胞を誘導し、この抗原特異的な T細胞 は SOX6を発現するダリオ一マ細胞と反応する力、、または SOX6を発現する細胞に 対し細胞障害性を有することを示す。このこと力ら、配列番号 1〜3のペプチド、その ペプチドを細胞表面に提示した抗原提示細胞、及びその抗原提示細胞の刺激によ つて樹立された細胞障害性 T細胞は、ダリオ一マに対する癌ワクチンとして有用であ ること力わ力る。
[0020] = =癌ワクチンの投与方法 = =
現在、癌ワクチンとして、腫瘍特異的癌抗原、癌抗原提示抗原提示細胞、または癌 抗原反応性細胞障害性 τ細胞を腫瘍患者に投与する方法が開発されている。本発 明においては、 SOX6の部分ペプチドを用いているので、治療(予防も含まれる)対 象となる腫瘍は、 SOX6が発現している腫瘍であれば何でもよぐ特に SOX6を高レ ベルに発現しているダリオ一マ、肝癌、肺癌、膝癌、食道癌、メラノーマ、前立腺癌、 乳癌、腎癌、または白血病も治療対象の一つとなる。主な治療対象は、こうした腫瘍 を有するヒト患者であるが、腫瘍を有するヒト以外の脊椎動物でもかまわない。
[0021] 治療対象となる腫瘍を有する患者に対し投与する癌ワクチンは、腫瘍特異的癌抗 原となりうる配列番号 1〜3をもつペプチドを含有してもよい。この場合、あらかじめ患 者の HLAクラス Iのタイプを調べ、それに適したペプチドを用いる。ここでは、患者の HLAクラス Iタイプが A24である場合に、配列番号:!〜 3のペプチドを投与する。投与 するペプチドは、一種類であっても複数種類であってもよい。投与部位に関しては、 皮内投与、皮下投与、静脈内投与、腹腔内投与などが考えられ、特に限定されること はない。また、投与する際には、免疫誘導能を高めるアジュバントなどとともにぺプチ ドを投与してもよい。また、投与されるペプチドは、生体内で分解されに《するような 修飾が施されていてもよい。
[0022] また、上記癌ワクチンは、配列番号:!〜 3を有するペプチドを提示した抗原提示細 胞を含有してもよい。ここで、細胞表面に提示されているペプチドは、配列番号:!〜 3 を有するペプチドそのものでもよぐ糖やリン酸などで修飾されてもよい。抗原提示細 胞としては、樹状細胞の他にマクロファージ、 B細胞等が考えられるが、抗原提示能 の高さなどから、樹状細胞が好ましい。以下、樹状細胞の単離方法の例を記述する。
[0023] まず、脊椎動物個体の末梢血から単核球を単離する。この単核球は、治療対象と なる個体自身から分離することが好ましいが、他の個体から単離してもよい。また、こ の単核球は CD14陽性であることが好ましレ、。単離した単核球を、 GM-CSFと IL-4 で 7 日前後培養すると、未熟な樹状細胞に分化誘導することができる。このようにして 分化誘導された樹状細胞は、抗原提示分子である MHC分子を高発現している。こ の未熟な樹状細胞の HLAクラス Iタイプを調べ、 A24である場合は、配列番号:!〜 3
のペプチドを添加する。添カ卩するのは、人工合成ペプチドに限らず、ペプチドを発現 させた細胞の抽出物(extractや lysate)や精製物などでもよレ、。こうして得られた抗原 提示榭状細胞を、腫瘍を有する個体に投与する。投与部位に関しては、皮内投与、 皮下投与、静脈内投与、リンパ節内投与などが考えられ、特に限定されることはない 、生理的な樹状細胞の抗原提示が、樹状細胞投与部位の所属リンパ節にて行な われることを考えると、リンパ節内への直接投与が好ましい。
[0024] また、上記癌ワクチンは、配列番号:!〜 3由来のペプチドを提示した抗原提示細胞 の刺激によって樹立された T細胞を含有してもよい。配列番号:!〜 3由来のペプチド を提示した抗原提示細胞に対し、 T細胞を共培養し、抗原提示細胞で刺激する。こ のようにして樹立された T細胞を腫瘍を有する個体に投与してもよレ、。ここでの T細胞 は、細胞障害性 T細胞が好ましいが、ヘルパー T細胞等でもよい。投与部位に関して は、皮内投与、皮下投与、静脈内投与、腫瘍内投与などが考えられ、特に限定され ることはないが、細胞障害性 T細胞の場合、抗原を発現する細胞を直接攻撃できるた め、腫瘍内投与が好ましい。
実施例
[0025] <実施例 1 >抗原を提示した樹状細胞による細胞障害性 T細胞の活性化
= =実験初日 = =
HLAクラス 1のタイプが A24 (遺伝子型は A2402)の健常人末梢血から、以下のよ うに単核球を分離した。まず、へパリン 5mlで洗浄したシリンジで、末梢血を 50ml採 血した。等量の Lymphoprep (Fresenius kabi Norge AS, Axis-Shield PoC AS, Oslo, N orway)を転倒混和し、遠心(20°C、 2000 rpm、 35分間、ブレーキなし)し、中間層を 吸引して採取した。これに PBSを加えて再混濁し、遠心(20°C、 2000 rpm、 10分間) する操作を 3回繰り返して、得られた単核球を洗浄した。
[0026] この単核球を初代細胞培養用培養皿 (FALCON MULTIWELL PRIMARIA 24 well) に 5xl06個/ゥエルの密度で播種し、 37°C、 5% CO存在下で 4時間培養した。培養

液は AIM-V(GIBCO)と RPMI-1640を 1:1に混合したもの(基本培養液)を用いた。培養 皿の底面に接着した細胞を回収し、 4x105個/ゥエルの密度で新しい 24ゥエルの培 養皿に播種し、 GM-CSF(10ng/ml), IL_4(lng/ml)添加した基本培養液を用いて、 37
°C、 5%CO存在下で 7日間培養し、榭状細胞を分化させた。

培養 7日目に分化した樹状細胞を抗原提示細胞(antigen presenting cells;APCs) として用いた。得られた樹状細胞を irradiate (60Gy)したのち、合成'精製したペプチド (配列番号:!〜 3及び 5の SOX6由来のペプチド)を ΙΟ μ Μ添加し、 37°C、 5% CO存
2 在下で 2時間培養して細胞に結合させ、 PBSで 2回洗浄し、刺激細胞(stimulator eel Is)とした。なお、コントローノレとして、 SOX6由来のペプチドのかわりにサイトメガロウイ ルス (CMV)由来のペプチド(配列番号 4 : QYDPVAALFF、 Provenzano et al, Tra nsfusion 43, ppl567_1574参照)を用いた実験を行った。
[0028] 一方、この日(培養 7日目当日)、実験 15日目に樹状細胞を得るために、初日と同 じ操作で単核球を培養した。
[0029] その過程で、単核球を 4時間培養後、培養皿に接着せず、培養上清に浮遊してい る細胞を回収し、 lxlO7個の細胞に対し IMag anti-human CD8 particules -DM (BD Biosciences社) 100 μ ΐを、 4°C、 30分間反応させた。磁石を用いて CD8陽性細胞を 吸着'回収し、応答細胞(responder cells)とした。
[0030] このようにして得られた応答細胞 2x105個を、上記刺激細胞のゥエルに添加し、共 培養した。なお、培養液として、 IL-l a (10 unit/ml), IL_2(20 unit/ml), IL_4(1 ng/ml), IL-6(125 unit/ml), IL_12(1 ng/ml)を添加した基本培養液を用いた。

培養 7日目力 培養し、分化させた樹状細胞を用いて、上記と同様の操作で刺激 細胞を調整した。
また、培養 7日目より刺激細胞と共培養した応答細胞を回収し、新たに調整した刺 激細胞と、同様に共培養した。ただし、培養液は培養 7日目に用いた上記培養液に おいて IL— 12を含まない培養液を用いた。
なお、実験 22日目に樹状細胞を得るための単核球の培養を、同様にして、新たに 合めておいた。

実験 15日目と同様に、刺激細胞と共培養を続けている応答細胞を回収し、新たに

なお、樹状細胞を得るための単核球の培養を始めた力 24ゥエルの培養皿ではな く、 96ゥエルの培養皿を用い、 4x104個/ゥエルに調整して播種し、培養した。

2時間培養して細胞に結合させ、 PBSで 2回洗浄し、ターゲット細胞として用いた。 共培養を続けている応答細胞を回収し、エフェクター細胞として 2xl04個に調整し た細胞を、こうして新たに調整したターゲット細胞のゥエルに添加し、共培養した。

上記共培養力も培養上清を回収し、以下のように IFN-γ ELISAを行った。
[003ΰ」 目 ij日に、一次?九体、 anti— human IFN— γ monoclonal Ab, Endogen)を 0. 5 μ g/ mlに PBSで希釈し、 98ゥエル'プレート(Nunc Maxisorp)に 1ゥエルあたり 100 β 1を添カロ し、 4°Cでー晚放置してプレートをコートした。実験当日、 1ゥエルあたり 300 /ilの 5% FCS含有 PBSで室温 1時間処理し、ブロッキングした。ゥエルを 0. 2%Tween-20 含有 PBSで 3回洗浄した後、共培養から回収した培養上清 50 / lを添加し、ピオチン 化 2次抗体(Endogen、 5%FCS含有 PBSで 0. 5mg/mlに調整したもの)を 50 /i 1添 加し、室温で 2時間放置した。ゥエルを 0. 2%Tween— 20含有 PBSで 3回洗浄した 後、ストレプトアビジン結合 HRP (ExtraAvidine peroxidase conjugate, SIGMA E-288 6、 5%FCS含有 PBSで 4000倍希釈したもの)を 100 μΐ添加し、 室温で 30分放置し た。ウエノレを 0. 2%Tween_20含有 PBSで 3回洗浄した後、 ΤΜΒを基質にして発 色させた(TMBは、 TMB tablet 1錠を DMSOlmlに溶解しリン酸クェン酸バッファー 9mlを加えて調整した)。 25μ^Ό1Ν H SOを添加し、反応を停止した。プレートリ

ーダ一で 450nmの吸光度を測定し、既知の濃度の IFN- γで予め作製した標準曲 線から IFN-γの濃度を算出し、結果を図 1に示した。

HLA—A24に結合し得ると予想された SOX6由来ペプチド(表 1参照)のうち、 S〇
X6— 360、 SOX6— 504、および SOX6— 628を添カロされた樹状細月包との共培養に より刺激された細胞障害性 T細胞(エフェクター細胞)は、各々の抗原ペプチドを提示 した榭状細胞をターゲットとして共培養した時、様々なコントロールに比して、 IFN- γ 放出量が増加した(図 1)。このことは、 SOX6ペプチドに由来するペプチド(S〇X6 _ 360、 SOX6 - 504,および S〇X6 _ 628)を抗原提示糸田胞に添カロすると、それらの ペプチドを提示する抗原提示細胞が得られ、得られた抗原提示細胞によって T細胞 を刺激することにより各ペプチドを特異的に認識する T細胞が誘導されたことを示す

[1] S〇X6の発現解析
SOX6を発現している細胞を明らかにすることにより、具体的にどのような細胞が、 本発明の方法の対象となるかを調べた。

ヒト正常組織 (成人脳及び精巣、及びヒト胎児脳)、ダリオ一マ細胞株 (GI— 1、 U87 、及び T98G)、患者から単離されたダリオ一マ組織 (ダリオブラストーマ組織、及びァ スト口サイトーマ組織)、ヒト腫瘍細胞株(888mel、 928mel、 586mel、 LK2、 PC9、 LU99、 RERF— LC MA、 TE8、 PK56、 PK1、 PC3、 MDA231、 RCC6、 RCC 8、 KU7、及び Molt4)における SOX6の mRNAの発現を定量的 PCRにより解析し た。
[0039] RNAを単離する材料としての細胞は、以下のように入手した。まず、ダリオ一マ細 月包株 SF丄 2bと Marcusは Health science Research Resources Bank (Osaka, Japan)力、ら 購入した。 U-87-MGと T98G (glioma), GI-1 (gliosarcoma), 888mel, 928melと 586mel (melanoma) Molt4 (leukemia and lymphoma), PC9, LU99, LK2, RERF— LC—MA (lung cancer), RCCb and RCC8 (renal cell cancer), TE8 (esophageal cancer), PK1と PK5 9 (pancreas cancer)、 MDA231 (breast cancer)は American Type Culture Collection (Manassas, VA)より購入した。また、ダリオ一マ組織は、慶應義塾大学倫理委員会承 認 (No. 12-21-2)の説明文に対して同意の得られた手術患者の腫瘍検体より得た。こ
れらの細胞株、または組織から、 RNeasy (キアゲン社)を用いて全 RNAを単離した。 また、ヒト正常組織の全 RNAは CLONTECH Laboratories, Inc.より購入した。これら の全 RNA(こメ し、 AMV Reverse transcriptase XL (Takara Bio In , Ohtsujapan)及 びオリゴ dTプライマーを用いて逆転写反応を行レ、、 cDNAを合成した。
[0040] これらの cDNAに対し、以下のように定量的 PCRを行レ、、 SOX6の mRNAの発現 について検討した。 SOX6に対するプライマーは
forward primer, 5'_GATGCCATCAACTCCACAGC- 3,(配列番号 6) reverse primer, 5'-GCTGCAGAGCCATTCATTGC-3' (配列番号 7)
を用い、 内部コントロールとしての j3 _actinに対するプライマーは
forward primer, 5 -GGCACCCAGCACAATGAAG-3' (配列番号 8)
reverse primer, 5'_GCCGATCCACACGGAGTACT- 3,(配列番号 9)
を用いた。定量的 PCRの条件は 95°C10分、その後、変性を 95°Cで 30秒,ァニーリ ングを 60°Cで 1分の過程を 50サイクル行レ、、最後に伸長反応を 72°Cで 1分間行った 。結果を図 2Aに示した。なお、各組織での SOX6の発現量は、それぞれの組織にお ける βァクチンの発現量で標準化した。
[0041] = =ウェスタン'ブロッテイングによる解析 = =
次に、タンパク質レベルでの SOX6の発現をウェスタン'ブロッテイングによって調 ベた。まず、上記と同様にして得られた細胞を回収し、破砕バッファー (最終濃度 20m M HEPES、最終濃度 0.25Mショ糖を純水 950mlに加え、 PH 7.5に調整した)と共に Dou nce型ホモジナイザーにいれ、氷上にて細胞を破砕した。破碎液を 1000gで 7分間遠 心し、上清を破棄し、核や未破砕細胞を含む沈殿を回収した。この沈殿に、 SDSゲ ル用ローデイングバッファーを加えて溶解し、電気泳動用サンプルとした。これらのサ ンプノレを、 SDS- PAGEを用レ、、各レーンに 20 x gずつ泳動した。メンブレンにトラン スファーし、 10 μ gZmlに調製した抗 SOX6抗体(CHEMICON社)によって SOX6タ ンパク質を認識し、 AP結合ャギ抗ゥサギ IgG (Fc)抗体(2000倍希釈、 Cappel, Auro ra, Ohio)を反 せた後、 Nitro り lue tetrazolium (Boehrmger Mannheim, mbH, e rmany)と 5-bromo-4-cnloro-3-indolyl phosphate (Sigmaし hemical Co., St. Louis, M〇)の混合液にて発色させた。結果を図 2Bに示した。

SOX6は、これまでに発現が報告された、ヒト胎児脳、成人精巣、ダリオ一マ細胞株 、ダリオ一マ組織のみならず、メラノーマ、小細胞肺癌、非小細胞肺癌、扁平上皮肺 癌、食道癌、陴癌、前立腺癌、乳癌、腎癌、慢性骨髄性白血病、 T細胞性白血病由 来の細胞株においても、発現が検出された。 (図 2A)。
[0043] 一方、正常組織における SOX6タンパク質の発現は、精巣以外では検出されなか つた(図 2B)。更に、 SOX6は、グリオ一マ細胞株(Marcus、 U87、 KNS_42、 SF1 26)だけでなぐ肺癌細胞株 (PC9)、肝細胞癌株 (HepG2)、膝臓癌株(Panc-1) におレ、ては、タンパク質レベルでも発現が検出された。
[0044] 以上より、 SOX6は、ヒト成人正常組織においては精巣しか発現が検出されないも のの、腫瘍細胞においては、広く発現が検出された。このことは、 SOX6由来のぺプ チドを用いた癌ワクチン力 ダリオ一マだけでなぐ多種の腫瘍に効果があることを示 してレ、る。さらに、正常組織では、ほとんど発現が検出されないため、この癌ワクチン は副作用が少ないと考えられる。

この日まで、実施例 1と同様にしてエフェクター細胞を調整した。なお、単核球を得 る健常人の HLAクラス 1のタイプを A24 (遺伝子型は A2402)とし、添加したぺプチ ドとして配列番号 1〜3のペプチドを用い、コントロールのペプチドとして CMV由来の ペプチド(配列番号 4: QYDPVAALFF)を用いた。

[0048] なお、コントローノレとして、ヒト 'ダリオ一マ細胞 KNS— 81の代わりに、 CIR—24糸田 胞 (HLA— A24が発現しており、 SOX6は発現していない細胞)とダリオ一マ細胞 U8 7 (HLA—A2と SOX6が共に発現しており、 HLA—A24は発現していない細胞)を 用いて、実験を行った。

上記共培養から培養上清を回収し、 IFN- T/ ELISAを行った。図 3に結果を示す。

SOX6由来の部分ペプチド(SOX6 - 504、および SOX6 _ 628)を用いて誘導さ れた細胞障害性 T細胞(エフェクター細胞)は、ターゲット細胞であるダリオ一マ細胞( KNS— 81)に反応して、コントロールに比して細胞数比依存的に IFN- γ放出量の増 加を示した(図 3)。このことは、 SOX6— 504または SOX6— 628を提示している抗 原提示細胞を用いて刺激することにより樹立した T細胞は、 SOX6と HLA— 24を発 現しているダリオ一マ細胞 (KNS— 81)を特異的に認識し得ることを示す。樹立した T細胞は U87 (HLA— A201と SOX6が共に発現しているが HLA— A24は発現し ていない細胞)に反応しないことから、これらの SOX6特異的 T細胞は HLA—A24 拘束性に抗原を認識することが示された。
[0051] [3]エフェクター細胞の細胞傷害活性による、 SOX6発現細胞に対する反応性の 評価

この日まで、実施例 1と同様にしてエフェクター細胞を調整した。なお、単核球を得 る人の HLAクラス 1のタイプを A24 (A2402)とし、添加したペプチドとして配列番号 1及び 2のペプチドを用いた。なお、本実施例では、単核球を得るための末梢血には 、健常人末梢血 (結果は図 4)だけでなぐダリオ一マ患者末梢血 (結果は図 5)も用い た。
[0052] ターゲット細胞として、 HLA— A, B欠損ヒト B細胞株 CIRに HLA— A2402の cDN Aを導入した CIR— A2402、 CIR— A24株に SOX6遺伝子を導入した CIR— A240 2_ S〇X6、 CIRに HLA—A0201の cDNAと S〇X6遺伝子を導入した CIR—A02
01— S〇X6、 HLA— A24と SOX6を共に発現しているダリオ一マ細胞(Marcus、 SF 126、 KNS— 42)、 HLA— A24と SOX6を共に発現している他の癌細胞(肝細胞癌 由来 HepG2、肺癌由来 PC9)を準備した。なお、 CIR— A2402— SOX6細胞及び CIR—A0201— SOX6細胞で SOX6が発現していることは、上記ウェスタン'ブロッ ティングによって確認、した(図 2B)。
[0053] これらの細胞 5X106個に対して 500 μ ΐの Fetal Bovine Serum (FBS)、 51Cr (l . 85 MBqZ50 μ l)を50 μ l添カロし、 37°C、 5%C〇2下で 60分振蕩培養した。その後洗 浄を 3回行レ、、 5xl03Zl00 x lに調整して 96穴プレート(COSTER 3595—96 well)の 各ゥエルに 100 μ 1ずつ添加した。
[0054] この51 Crでラベルしたターゲット細胞に対し、ゥエルあたり、エフェクター細胞を 3x1 05個 (E/T ratio=60)、 1. 5xl。5個 (E/T ratio=30)、 7. 5X104個 (E/T ratio=15)のそ れぞれを加え、 37°C、 5% CO存在下で 4時間培養した。

(なお、 csr(cpm spontaneous release)はエフェクター細胞を含まない培地を添加した ゥエルの Cpmであり、 cmr(cpm maximal release)はエフェクター細胞を含まない 0· 1 % Triton Xを添加したゥエルの cpmである。)

CIR— A24— SOX6、グリオ一マ細胞由来 Marcus、グリオ一マ細胞由来 SF126、 ダリオ一マ細胞由来 KNS— 42、肝癌由来 HepG2、肺癌由来 PC9は、 HLA—A24 と SOX6を発現している細胞である。図 4 (健常人由来の末梢血リンパ球から CTLを 誘導した場合)及び図 5 (ダリオ一マ患者由来の末梢血リンパ球から CTLを誘導した 場合)に示したように、 3〇 6 _ 504または30 6 _ 628を添加され樹状細胞との共 培養により刺激された細胞障害性 T細胞(エフヱクタ一細胞)である CTL (SOX6 - 5 04)と CTL (SOX6 _628)は、これらの HLA—A24と SOX6を発現している細胞を E/T ratio依存性に溶解した。
[0057] 一方、 SOX6を発現しているが HLA—A24を発現していない細胞 CIR—A0201
- SOX6や、 HLA— A24を発現してレ、るが SOX6を発現してレ、なレ、細胞 CIR— A2 402に対する溶解性が低レ、こと力ら、 CTL (SOX6 - 504)と CTL (SOX6— 628)は 、 HLA拘束性に SOX6を発現する細胞を広ぐ特異的に傷害し得ることが示された。
[0058] [4]エフェクター細胞のターゲット細胞に対する特異性
SOX6由来のペプチドの刺激により樹立したエフェクター細胞の、 SOX6と無関係 なペプチドを表面に有する細胞に対する細胞障害活性を調べることにより、エフエタ ター細胞のターゲット細胞に対する特異性を調べた。
[0059] 基本的には、実施例 2と同様に実験を行ったが、ターゲット細胞として、
(1) HLA-A24と SOX6が共に発現しているダリオ一マ細胞株 Marcus
(2) HLA-A24を発現してレ、て SOX6を発現してレ、なレ、CIR_A2402に、配列番号 1を有するペプチド SOX6-504を結合させた CIR-A2402 (SOX6-504)または配 列番号 2を有するペプチド SOX6-628を結合させた CIR_A2402 (S〇X6_628)
(3)非特異的細胞障害を測定するためのコントロールとして、 CIR-A2402に対し、 C MV由来の無関係なペプチド(配列番号 4)を結合させた CIR-A2402 (CMV)
(4)無処理の CIR-A2402
を用いた。実施例 3—1に記載の手順にて、上記細胞を51 Crでラベルし、 5xl03/10
0 μ 1に調整して 96穴プレートの各ゥエルに 100 μ 1ずつ添加した。
[0060] エフェクター細胞として、ペプチド SOX6-504の刺激により樹立した CTL (SOX6- 504)とペプチド SOX6-628の刺激により樹立した CTL (SOX6-628)を準備した。 エフェクター細胞はゥエルあたり lxlO5個(ET ratio = 20 : lを、 96穴プレート上のタ 一ゲット細胞に添加した。これらを 37°C、 5% CO存在下で 4時間培養後、各ゥエル

の cpm (cpm experimental release)を十測し、特異的溶角军率 (Percentage of specific 1 ysis)を上記式力も算出した。図 6に結果を示す。
[0061] CTL (SOX6-504)及び CTL (SOX6-628)は、 HLA-A24と SOX6が共に発現 してレ、る Marcusや、 SOX6由来のペプチド SOX6- 504または SOX6-628を結合さ せた CIR-A2402に対する溶解率は、無関係なペプチドであるペプチドを結合させ た CIR- A2402や無処理の CIR- A2402に対する溶解率に比べて高かった。このこ と力、ら、 CTL (SOX6-504)及び CTL (SOX6-628)は、それぞれ SOX6-504及び
SOX6-628を提示した細胞に対し、特異的に細胞障害を起こすことが示された。
[0062] このように、本実施例では、 SOX6由来のペプチドで誘導されたエフェクター細胞 のターゲット細胞に対する特異性が高いことが示され、従って、 SOX6由来のぺプチ ドが癌ワクチンとして用いられた時、副作用が低いと考えられる。

以上より、 SOX6に由来する配列 1〜3を有するペプチドを用いて、それらの抗原ぺ プチドを提示した抗原提示細胞を作製することができ、さらに、その抗原提示細胞に よって刺激することにより、各ペプチドのみならず SOX6を発現した細胞を認識する、 特異的 T細胞が誘導される。
産業上の利用可能性
[0064] 本発明によって、腫瘍免疫を効率的に誘導できるペプチド、そのペプチドを含有す る組成物、そのペプチドを提示した抗原提示細胞、この抗原提示細胞によって刺激 された T細胞、およびこれらのペプチドや細胞を利用した癌ワクチン、及びそれらを 用いた腫瘍患者の治療方法を提供することが可能になった。

請求の範囲
[I] 配列番号:!〜 3のいずれかの配列を有するペプチド
[2] 配列番号:!〜 3のいずれかの配列を有するペプチドを提示した抗原提示細胞。
[3] 樹状細胞であることを特徴とする請求項 2の抗原提示細胞
[4] 請求項 2または 3に記載の抗原提示細胞によって誘導され、 SOX6を発現する細胞 を認識する T細胞。
[5] 細胞障害性 T細胞であることを特徴とする請求項 4の T細胞。
[6] SOX6を発現する細胞がダリオ一マであることを特徴とする請求項 4の T細胞。
[7] SOX6を発現する細胞が肝癌細胞、肺癌細胞、膝癌細胞、食道癌細胞、メラノーマ
、前立腺癌細胞、乳癌細胞、腎癌細胞、または白血病細胞であることを特徴とする請 求項 4の T細胞。
[8] SOX6を発現する細胞が肝癌細胞、肺癌細胞、または膝癌細胞であることを特徴と する請求項 4の T細胞。
[9] 配列番号:!〜 3から選択される一つ以上のペプチドを含有する癌ワクチン。
[10] 請求項 2または 3に記載の抗原提示細胞を含有する癌ワクチン。
[II] 請求項 4〜8のいずれかに記載の T細胞を含有する癌ワクチン。
[12] SOX6を発現する癌細胞に対する癌ワクチンであることを特徴とする請求項 9〜 11 のレ、ずれかに記載の癌ワクチン。
[13] ダリオ一マに対する癌ワクチンであることを特徴とする請求項 9〜: 11のいずれかに 記載の癌ワクチン。
[14] 肝癌、肺癌、陴癌、食道癌、メラノーマ、前立腺癌、乳癌、腎癌、または白血病に対 する癌ワクチンであることを特徴とする請求項 9〜: 11のいずれかに記載の癌ワクチン
[15] 肝癌、肺癌、または陴癌に対する癌ワクチンであることを特徴とする請求項 9〜11 のレ、ずれかに記載の癌ワクチン。
[16] ヒト以外の脊椎動物に対し、請求項 9〜: 15のいずれかに記載の癌ワクチンを用いた 腫瘍の治療方法。
