Abstract

L'invention concerne un procédé détection de la méthylation d'ADN à des sites CpG par utilisation de réseaux d'acides nucléiques et, de préférence, des microréseaux. L'invention concerne spécifiquement, un procédé de production directe d'un échantillon de référence à partir de l'échantillon à tester et de détection simultanée de méthylation sur un grand nombre de sites CpG. Plus spécifiquement, le procédé inventif consiste à diviser un échantillon d'ADN en deux échantillons (un premier échantillon et un second échantillon), à amplifier le premier échantillon d'ADN par un processus d'amplification d'acide nucléique tel que tous les résidus de méthylcytosine soient amplifiés en tant que résidus cytosine non méthylés, traitant le premier échantillon amplifié et le second échantillon (non amplifié) avec du bisulfite afin de convertir les résidus de cytosine non méthylés dans les deux échantillons en résidus de désoxyuraciles, à marquer le second échantillon converti en bisulfite avec un second marqueur fluorescent et le premier échantillon converti en bisulfite avec un premier marqueur fluorescent, les premier et second marqueurs fluorescents présentent des spectres d'excitation et d'émission de fluorescence ne se recouvrant pas, et à hybrider le premier échantillon et le second échantillon sur un dispositif microréseau comprenant une pluralité de sondes de capture d'oligonucléotides conçues afin d'hybrider les sites îlots CpG de l'échantillon d'ADN comme convertis et non convertis par le bisulfite.
