Abstract

L'invention porte sur un procédé destiné à réduire le mauvais appariement de nucléotides dans une hybridation entre une sonde oligonucléotidique et un ADN cible, le procédé comprenant les opérations consistant à prendre au moins une sonde oligonucléotidique d'environ 7 à environ 25 nucléotides, à prendre un échantillon comprenant de l'ADN cible, dans lequel l'ADN est préparé de manière à comprendre des fragments allant jusqu'à environ 100 bases, faire incuber l'ADN avec la au moins une sonde oligonucléotidique dans des conditions appropriées pour permettre une hybridation entre la sonde et l'ADN cible, éliminer l'ADN non lié et détecter l'ADN hybridé à la au moins une sonde oligonucléotidique, procédé suivant lequel l'un ou l'autre de l'ADN et de la au moins une sonde oligonucléotidique, ou les deux comprennent un ou plusieurs nucléotides modifiés, et suivant lequel l'ADN de l'échantillon est facultativement marqué par une fraction détectable.
