BCGワクチンとその利用
技術分野 この出願の発明は、 BCG ワクチンとその利用に関するものである。 さらに詳 しくは、 この出願の発明は、 各種感染症や癌等の予防や治療のための免疫誘導に おける初回抗原刺激に使用する組換え BCG ワクチンと、 この BCG ワクチンを 用いたヒ卜または動物の免疫誘導方法に関するものである。
背景技術 牛型結核菌弱毒 BCG株 (Mycobacterium bovis BCG。 以下 「BCG」 と記載す る) は、 その安全性から、 最も一般的な生バクテリアワクチンとして知られてい る。 一方、 この十数年来め遺伝子組換え技術の開発、 向上に伴い、 ウィルスや細菌 などの微生物に外来性の抗原性タンパク質を発現させるように改変して、 様々な 感染症や癌の予防および治療に対するワクチンベクターとして応用しょうという '研究が盛んに行われて来ている。 BCG についても、 例えばヒ卜免疫不全ウィル ス (HIV) ゃサル免疫免疫不全ウィルス (SIV) を標的とする組換え BCG ヮクチ ンカ報告されて ( る ( J. Immunol. 164:4968-4978, 2000; J. Virool. 71 :2303- 2309, 1997; I nfect. Immun. 57:283-288, 1989) 。 そして、 BCGに HIV遺伝子を 発現させたワクチンは長期に渡って (少なくとも 2年間は) 免疫の誘導が可能と なるが、 このような長期間の免疫誘導能は他の DNA ワクチンではみられない優 れた特徴である。

しかしながら、 従来の組換え BCG ワクチンの場合には、 標的となる感染症や 癌等に対する免疫誘導能の点においては必ずしも十分なものではなかった。 例え ば、 HIV-1 を標的とする組換え BCG ワクチンを用いてモルモッ トに免疫誘導を 行った場合には、 通常の BCGワクチンのヒ卜への使用量 (0.05-0.1 mg) の 50〜 1 00倍の量を投与する必要がある (Proc. Natl. Acad. Sci. USA. 92: 10698-10697, 1995) 。 またサルのモデルを用いた試験では組換え BCG単独の投与では病原性 ウィルスの感染防御について好ましい結果は得られていない。 組換え BCG ワクチンは、 免疫の持続性およびその安全性と供給が容易である という点において優れたワクチン候補であリ、 その有効利用は医療分野において 不可欠の課題である。 この出願の発明は、 以上のとおりの事情に鑑みてなされたものであって、 組換 え BCG ワクチンの有効利用のための新しい手段を提供することを課題としてい る。
発明の開示 この出願は、 前記の課題を解決するためめ第 1め発明として、 外来性の抗原性 タンパク質をコードするポリヌクレオチドを保有する発現ベクターによって形質 転換された組換え BCG ワクチンであって、 複数回の抗原刺激による免疫誘導に おいて初回抗原刺激用として使用されることを特徵とする BCG ワクチンを提供 する。 またこの出願は、 第 2の発明として、 外来性の抗原性タンパク質を複数回刺激 する免疫誘導方法であって、 前記第〗発明の BCG ワクチンによって初回抗原刺 激を行い、 同一の抗原性タンパク質を発現する非 BCG ワクチンによって 1 回以 上の追加抗原刺激を行うことを特徴とする方法を提供する。

この第 2発明の方法においては、 追加抗原刺激用のワクチンが、 組換えワクシ ニァウィルスワクチン、 例えば組換え Dlsワクチンであることを好ましい態様と している。 さらに、 前記第 1発明および第 2発明においては、 抗原性タンパク質が免疫不 全ウィルス由来であること、 さらに詳しくは、 免疫不全ウィルスの抗原性タンパ ク質が HIV遺伝子産物、 例えば Gagであることをそれぞれ好ましい態様として いる。 すなわち、 この発明の方法に従えば、 例えば組換えワクシニアウィルス (例え ば組換え Dls-gagワクチン) の単独投与では得られなウィルスの血中への流出を ほぼ完全に防止することができ、 また CD4 細胞の減少も抑制することが可能と なる。 これによつて、 少なくとも病原性ウィルスの体内での蔓延を抑制し、 感染 性疾患の進行を防止することが可能となる。

発明を実施するための最良の形態 第 1発明は、 外来性の抗原性タンパク質をコードするポリヌクレオチドを保有 する発現ベクターによって形質転換された組換え BCG ワクチンである。 そして この組換え BCG ワクチンは、 複数回の抗原刺激による免疫誘導において、 その 使用が初回抗原刺激用として使用されることを特徴とする。 すなわち、 この出願 の発明者らは、 単独使用では十分な免疫誘導能を持たない組換え BCG ワクチン であっても、 初回抗原刺激 (プライミング) として使用することによって、 その 後の追加抗原刺激 (ブースティング) による特異的免疫誘導を強化することを見 出してこの発明を完成させた。
BCG 株は、 結核の予防接種等に使用されている公知のものを対象とすること ができる。 発現ベクターは、 従来の組換え BCG ワクチンの作成に使用されてい るような、 BCG 用ベクター (例えばプラスミド pS0246等) を使用することが できる。 このベクターのクローニングサイ 卜に、 外来性 (すなわち、 BCG 以 外) の任意の抗原性タンパク質をコードするポリヌクレオチドを挿入することに よって発現ベクターを構築することができる。 なお、 以下の記載では、 外来性の 抗原性タンパク質を 「外来ポリペプチド J 、 これをコードするポリヌクレオチド を 「外来ポリヌクレオチド」 と記載することがある。 また、 外来ポリヌクレオチ ドには、 BCG 株由来の任意のプロ乇一夕一およびターミネータ一配列 (例えば BCG株由来のヒー卜ショックタンパク質 Hspのプロモータ一およびターミネ一 ター配列) 等を連結することによって、 外来ポリペプチドを良好に発現させるよ うにする。 外来ポリヌクレオチドは、 BCG 株以外の抗原性タンパク質をコードするポリ ヌクレオチド (例えば、 cDNA 断片) であり、 外来ポリペプチドは生体内で抗原 抗体反応を惹起するものであれば如何なるものであってもよい。 具体的にはヒ卜 後天性免疫不全症候群 (AIDS ) の原因ウィルスであるヒ卜免疫不全ウィルス ( HIV) の gag前駆体 p55または p24タンパク質、 envタンパク質 gp120また は gp160、 po1前駆体タンパク質、 nef タンパク質、 tatタンパク質等を対象とす
ることができる。 また、 サル免疫不全ウィルス (SIV) 由来の同様の抗原性ポリ ペプチドを使用することもできる。 あるいは、 その他の病原体 (他の病原性ウイ ルスや細菌) 、 もしくは癌細胞の抗原タンパク質をコードするポリヌクレオチド 等を用いることもできる。 外来ポリヌクレオチドの取得方法としては、 外来ポリペプチドをコードするゲ ノ厶遺伝子またはその cDNA がクローン化されたプラスミドからその実質的な 配列であるポリヌクレオチドを適当な制限酵素で切り出すか、 適当な配列のブラ イマ一を用いた polymerase chain reaction ( PCR) により増幅すればよい。 ク ローン化されていない場合は、 その遺伝子を持つ細菌、 動物のゲノム DNA を、 ウィルスの場合はウィルスが感染した動物細胞由来の DNA または RNA を铸型 として、 上記 PCR 法により DNA 断片を増幅することにより得ることができる。 このようにして構築した発現ベクターを、 塩化カルシゥ厶法や電気穿孔法等の 公知に方法で BCG 株に導入し、 形質転換菌の外来ポリペプチドの発現をウェス タンプロットや公知の免疫測定法 (例えば ELISA 法等) によって確認すること によって、 この発明の組換え BCGを作成することができる。 このようにして作成した組換え BCG を、 通常の BCG ワクチンと同様の液状 担体に懸濁するすることのよつて、 組換え BCG ワク ンを作成することができ, このワクチンは、 実際には第 2発明の免疫誘導方法に使用することができる。 第 2発明の方法は、 前記第 1発明の BCG ワクチンによって初回抗原刺激を行 い、 同一の抗原性タンパク質を発現する非 BCG ワクチンによって 1 回以上の追 加抗原刺激を行うことを特徴としている。 追加抗原刺激用のワクチン (ブースターワクチン) は、 組換えワクチンに使用 されている公知のウィルスや細菌 (例えばポリオウイルス、 インフルエンザウイ ルス、 ライノウィルス、 水痘ウィルス、 ワクシニアウィルス、 サルモネラ菌、 リ ステリア菌等) を、 第 1発明の組換え BCG ワクチン (プライ厶ワクチン) と同
一の外来ポリヌクレオチドによって形質転換することによって作成することがで きる。 この発明の方法においては、 この出願の発明者らが先に開発した組換えヮ クシニアウィルス Dls ワクチン (特開 2002-017370号公報) を好ましいブース ターワクチンとしている。 プライ厶ワクチンとブースタ一ワクチンの投与は、 注射または経口投与等の公 知の方法によって行うことができる。 投与量およびスケジュールは、 被検疫個体 の種類 (ヒ卜または動物) 、 体重、 誘導する免疫の種類等によって異なるが、 例 えば、 プライ厶ワクチンを 0.01〜10mg、 ブースターワクチンを 105~ 101 Q PFU 程度とすることができる。 また、 ワクチン接種の間隔は、 3〜12 ヶ月程度とする ことができる。 以下、 実施例を示してこの出願の発明についてさらに詳細かつ具体的に説明す るが、 この出願の発明は以下の例によって限定されるものではない。
実施例 実施例 1
組換え BCGの作成
SIVの gag遺伝子をプラスミド pNL432 (丄 Virol. 59:284-291 , 1986) から単 離し、 この遺伝子 DNAの前後に BCG株由来の hsp60プロモーターおよびター ミネーター配列をそれぞれ連結し、 これを大腸菌一 BCG 株シャ トルベクター pS0246 (FEMS Microbiol. Lett. 135:237-243, 1996) のマルチクローニングサイ 卜に挿入して発現ベクター pSO-SIVgagを構築した (図 1 ) 。
この発現ベクターを、 文献 (Pro. Natl. Acad. Sci. USA. 85:6987-6991 , 1988) の記載に従い、 Gene-pulser ( Bio-Rad 社) を用いて BCG 東京株に導入し、 20 g/mlのカナマイシン含有の Middlebrook 7H10寒天培地 (Difco社) 上で形質 転換菌を選択し、 pSO-SIVgaa を保有する組換え BCG 株 (rBCG-SIVgag) を作

成した。
ウェスタンブロッテイングによって SIV gag 夕ンパク質の産生を確認した結 果、 図 2に示したように、 rBCG-SIVgagの抽出物において 55 kDaタンパク質が 検出された。 一方、 コントロールの rBCG-pS0246 では Gag タンパク質は検出 されなかった。 また SIV Gag タンパク質の濃度は、 rBCG-SIVgag 1 mg 当たり 45 ± 12 ngであり、 この産生レベルは少なくとも 450回の in vitro継代の間維持 されていた。
実施例 2
免疫誘導
実施例 1で作成した組換え BCG株 (rBCG-SIVgag) 、 および組換えワクシニ ァ Dls ( rDls-SIVgag) を用いて力二クイサルに免疫誘導を行った。 なお、 rDls- SlVgag は、 特開 2002-017370号公報の実施例 1 における HIV-1 gagの代わりに SlVgag遺伝子を用いて、 前記公報記載と同様の方法により作製した。
力二クイサル 14匹を以下の 5群に分け、 初回抗原刺激から 0、 47、 54週後の 時点で免疫 (ブースティング) した。
第 1 群 (4匹) : コントロール ( 1 匹はナイーブサル、 3匹は rBCG-pS0246 ( 10 mg) の皮内接種 1 回と rDls-LacZ ( 106PFU) の静脈接種を 2回)
第 2群 ( 2匹) : rBCG-SIVgag ( 10 mg) の皮内接種 1 回
第 3群 (2匹) : rDls-SIVgag ( 106PFU) の静脈接種 2回
第 4群 ( 3匹) : rDls-SIVgag ( 106PFU ) の静脈接種 2回 + rBCG-SIVgag ( 10 mg) の皮内接種 1 回
第 5群 ( 3 匹) : rBCG-SIVgag ( 10 mg ) の皮内接種 1 回 + rDls-SIVgag ( 106PFU) の静脈接種 2回
次いで、 2回目のブースター免疫から 10 週間後に、 病原性ウィルス (SHIV- KS661 : 2000TCID50) を粘膜感染させ (チャレンジ) 、 経時的に血中のウィルス RNAコピー数および CD4細胞数を計測した。
コントロール群のサルに SHIV-KS661 を感染させると、 図 3に示したように, CD4は約 2週間後に 1桁から 2桁に減少し、 それに反してウィルス RNAの血中
コピー数は 108-9レベルに増加し、 速やかにセットポイントに達し、 105-6レベル を推移した。 また、 rBCG-SIVgag 単独 (第 2群: 図 4 ) 、 および rDls-SIVgag 単独 (第 3群: 図 5 ) の場合も、 CD4およびウィルス RNA数の経時的変化はコ ン卜ロールと同様であった。 さらに、 rDls-SIVgag (プライミング) + rBCG- SIVgag (ブースティング) を行ったサル (第 4群) の場合もコントロールや単 独免疫と同様の経時的変化を示した。
これに対して、 第 5群 (rBCG-SIVgag + rDls-SIVgag) のサルの場合には、 図 7に示したように、 ウィルス RNAコピー数は顕著に減少し、 強い Gag特異的免 疫が誘導された。 また、 CD4細胞の減少も有意に抑制された。
以上の結果から、 組換え BCG ワクチン (rBCG-SIVgag) でプライミングを行 い、 組換え Dls ワクチン (rDls-SIVgag) でブースティングを行うことによって、 強い特異的免疫が誘導されることが確認された。
実施例 3
BCGの既往症反応の制御 この発明の免疫誘導方法における BCG既往症反応の影響を検討した。 すなわ ち、 ヒ卜での実際の投与の影響を想定して約 2年前に BCG 東京株 (0.1 mg) を 力二クイサルに接種し、 2年後でも DTH が明らかに誘導されていることを確認 した後、 以下の 3群のサルにワクチン接種を行った。
第 1 群 ( 3 匹) : rBCG-pS0246 ( 80 mg ) の経口接種 2 回十 rDls-LacZ
( 106PFU) の静脈接種を 2回)
第 2群 ( 2 匹) : rBCG-SIVgag ( 80 mg ) の経口接種 2 回 + rDls-SIVgag
( 106PFU) の静脈接種 2回
第 3群 ( 2匹) : rBCG-SIVgag ( 10 mg ) の静脈接種 1 回 + rDls-SIVgag ( 10 mg) の静脈接種 1回
なお、 2回のプライミングは- 48 週および 0週時点で、 2回のブースティング は 27 週および 57週後の時点で行った。 また、 第 3群は、 0週時点でブライミ ング、 57週時点でブースティングを行った。

最後のワクチン接種から約 3 か月後に病原性ウィルス ( SHIV-C2/1 20TCID50) をチャレンジし、 CD4細胞数およびウィルス RNAコピー数を経時的 に計測した。
結果は図 8— 1 0に示したとおりである。 rBCG-SIVgag プライミング (静 脈) + rBCG-SIVgagブースティング (静脈) (第 3群) のウィルス RNA数およ び CD4 数の経時的変化 (図 1 0 ) はコン卜ロール (第 1群) と同様であった。 これに対して、 rBCG-SIVgag プライミング (経口) + rBCG-SIVgag ブースティ ング (静脈) (第 2群) では、 血中ウィルス量の有意な減少と、 CD4 細胞の減 少抑制が観察された。
以上の結杲から、 rBCG-SIVgag を経口接種によってプライミングを行い、 rBCG-SIVgag を静脈接種によってブースティングすることによって、 免疫誘導 のみならず、 防御免疫においても既往症反応の影響を除外することが可能である ことが確認された。
産業上の利用可能性 以上詳しく説明したとおり、 この出願の発明によって、 組換え BCG ワクチン を用いた効果的な免疫誘導が可能となり、 様々な感染症や癌等に対して、 有効な 予防が実現される。
Claims
1 . 外来性の抗原性タンパク質をコードするポリヌクレオチドを保有する発現 ベクタ一によって形質転換された組換え BCG ワクチンであって、 複数回の抗原 刺激による免疫誘導において初回抗原刺激用として使用されることを特徴とする BCGワクチン。
2 . 抗原性タンパク質が免疫不全ウィルス由来である請求項 1 の BCG ヮクチ ン。
3 . 免疫不全ウィルスの抗原性タンパク質が HIV 遺伝子産物である請求項 2 の BCGワクチン。
4 . 外来性の抗原性タンパク質を複数回刺激する免疫誘導方法であって、 請求 項 1の BCG ワクチンによって初回抗原刺激を行い、 同一の抗原性タンパク質を 発現する非 BCG ワクチンによって 1 回以上の追加抗原刺激を行うことを特徴と する方法。
5 . 追加抗原刺激用のワクチンが、 組換えワクシニアウィルスワクチンである 請求項 4の方法。 "
6 . 抗原性タンパク質が免疫不全ウィルス由来である請求項 4の方法。
7 . 免疫不全ウィルスの抗原性タンパク質が HIV 遺伝子産物である請求項 6 の方法。
