Abstract

La présente invention concerne un processus de production d'une glycoprotéine comprenant une chaîne de sucre de type mammifère. Le processus de production se caractérise en ce qu'il consiste à transférer et à exprimer l'alpha-1,2-mannosidase dans le mutant du gène de la voie de biosynthèse de la chaîne sucre d'une levure de méthylotrophe sous le contrôle d'un promoteur puissant, à mettre en culture dans un milieu approprié, les cellules de levure de méthylotrophe dans lesquelles un gène étranger a été transfecté, puis à récolter dans le milieu, la glycoprotéine comprenant la chaîne de sucre de type mammifère. A l'aide d'une levure de méthylotrophe récemment cultivée portant une mutation de la chaîne de sucre, une chaîne de sucre neutre identique à une chaîne de sucre de type à forte teneur en mannose produite par des cellules de mammifère telles que des cellules humaines ou une glycoprotéine comprenant une telle chaîne de sucre neutre peut être produite, en grande quantité avec une grand niveau de pureté. Le fait de transférer un gène dans une voie de biosynthèse de la chaîne de sucre de type mammifère pour former le mutant décrit ci-avant, permet de produire efficacement une chaîne de sucre de type mammifère telle qu'une chaîne de sucre hybride ou une chaîne de sucre complexe ou bien encore une protéine comprenant une chaîne de sucre de type mammifère.
