Abstract

Die Erfindung betrifft ein Verfahren zur Aufarbeitung von Proteinen in Gelen, indem Gelfragmente mit Proteinen aus dem Gel isoliert, einem Proteaseverdau unterworfen und die entstandenen Spaltpeptide identifiziert werden, wobei die isolierten Protein-Gelfragmente in die entsprechende Anzahl Reaktionsgefäße einer ersten Mikrotiterplatte (Filterplatte) überführt werden, wobei die Böden der Reaktionsgefäße der Mikrotiterplatte als hydrophobe Filtermembran ausgestaltet sind, die Protein-Gelfragmente gewaschen und mit der Proteaselösung inkubiert werden, wobei nach jedem Schritt die jeweilige Lösung durch die Filtermembran abgesaugt wird. Anschließend wird die erste Mikrotiterplatte über einer zweiten Mikrotiterplatte (Auffangplatte) postiert, wobei die Reaktionsgefäße der zweiten Mikrotiterplatte konische Böden aufweisen und aus einem Material bestehen, das keine Peptide adsorbiert, und dann die an den Filtermembranen der ersten Mikrotiterplatte gebundenen Peptide mittels hydrophober Waschlösung unter Anlegen von Druck in die Reaktionsgefäße der zweiten Mikrotiterplatte eluiert und aufkonzentriert werden.
