Abstract

Die Erfindung betrifft ein Verfahren zum direkten Nachweis von Target-Nukleotidsequenzen, Target-Aminosäuresequenzen oder Target-Antigenen durch Amplifikation von DNS unter Verwendung einer DNS-Polymerase und eines DNS-doppelstrangdestabilisierenden Enzyms, wobei das Target-Molekül in situ oder in Lösung vorliegt oder direkt oder über eine Fangsonde an eine feste Phase immobilisiert wird, eine zur Strangverdrängung befähigte DNS-Polymerase, dNTPs und ein DNS-doppelstrangdestabilisierendes Enzym zugegeben werden und das entstehende Amplifikat direkt detektiert und über die Detektion das Target nachgewiesen wird oder für den Nachweis nichtamplifizierbarer Targetmoleküle zunächst eine Matrix-DNS tragende Kopplungssonde zugegeben wird und anschließend eine zur Strangverdrängung befähigte DNS-Polymerase, dNTPs und ein DNS-doppelstrangdestabilisierendes Enzym zugegeben werden und das entstehende Amplifikat direkt detektiert und über die Detektion das Target nachgewiesen wird.
