本発明は、 医農薬などの中間体として有用な新規アミノフエノール誘導体お よび、 その製造方法に関する。 背 景 技 術

(式中、 Ri、 nおよび mは式 (1) と同じ。 R5は水素原子、 炭素数 1〜4のアル キル基、 炭素数 3〜6のシクロアルキル基、 または一 (CH2) p〇R6 (pは 2 または 3の整数、 R6は炭素数 1〜4のアルキル基を表わす。)で表わされるァニ リド誘導体は、 医薬、 農薬などの合成中間体として有用であり、 例えば EP0 968999にある様に、抗不整脈剤として有用であることが報告されている。 このァニリド誘導体の製造方法としては、 式 (5)
(式中、 Riおよび nは式 ( 3 ) と同じ。 ) で表されるカルボン酸を直接縮合す る方法が知られている。 しかしながら、 この方法では、 式 (4 ) で表わされる 目的のァニリド誘導体の他に、 カルボン酸(6 ) が式 (5)のァニリン誘導体の脂 肪族第二アミン部位にアミ ド結合した生成物が副生し、 目的物を収率よく得る ことが困難であった。 そこで、 副反応を抑制するために、 t一ブトキシカルボ ニル基の様な保護基を導入した後、 アミ ド化する方法も知られているが、 保護 化剤が高価であること、 および保護、 脱保護工程が必要となり、 製造コストや 設備面で問題があった。
また、 式 (6 ) で表される非常に高価なカルボン酸を原料として用いており、 この点からも製造コストとして問題があつた。 発 明 の 開 示
本発明の目的は、 式 (4 ) で表わされるァニリ ド誘導体を収率よく製造する に好適な式 (1 ) で表される新規製造中間体、 およびその安価な製造方法を提 供することにある。
本発明者らは、 上記課題を解決すべく鋭意検討した結果、 式 (4 ) で表され るァニリド誘導体のアミ ド結合を先に導入し、 続いて脂肪族第二アミン部分を 導入することによって、 保護基を用いることなく、 目的の化合物を収率よく得 る方法を見出し、 本製造法に関わる新規中間体の発明に至った。 また、 式 (3 ) で表される安価で入手容易なカルボン酸エステルと、 式 (2 ) で表されるァ
'ミノフ Iノール誘導体を、 縮合剤を用いることなく反応させることにより、 式
( 1 ) で表される新規中間体を安価かつ収率よく得る方法を見出すに至った すなわち、 本発明のアミノフヱノール誘導体は、 式 (1 )
(式中、 Riは炭素数 1〜4のアルキル基、 nは 2または 3の整数を表わし、 R2 は水素原子、 または一 (C H2) mR3 (mは 2または 3の整数、 R 3は水酸基、 ハロゲン原子、メタンスルホニルォキシ基または p—トルエンスルホニルォキシ 基を表わす。 ) で表わされる化合物である。
上記式 (1 ) の化合物の好ましい態様としては以下の化合物を挙げることが できる。
1 . Riがメチル基であり、 nは 2の整数を表わし、 R2が水素原子である化合物
2. Riがメチル基であり、 n、 mはそれぞれ 2の整数を表わし、 R3が水酸基、 塩素原子、 臭素原子、 ヨウ素原子、 メタンスルホニルォキシ基または p—トルェ ンスルホニルォキシ基である化合物。

発明を実施するための最良の形態
以下、 本発明を詳細に説明する。
上記各式において、 炭素数 1〜4のアルキル基とはメチル基、 ェチル基、 n- プロピル基、 イソプロピル基、 n-ブチル基、 イソブチル基、 sec-ブチル基、 tert- ブチル基をいう。 ハロゲン原子とはフッ素原子、 塩素原子、 臭素原子、 ヨウ素 原子をいう。

(a)法の縮合反応はエステルとァミンの混合物を加熱し、 生成するアルコール を留去させることで実施される。 反応は、 無溶媒あるいは溶媒存在下行うこと ができ、 そのとき使用する溶媒は、 反応の進行を妨げない溶媒であればどのよ うなものであってもよく、 単独あるいは混合して用いられる。例えばベンゼン、 トルエン、 キシレン、 ジメチルホルムアミ ドのような非プロトン性溶媒が好ま しい。 最も好ましくは、 無溶媒で行うことである。 反応温度は、 アルコールが 留去できる温度であれば、 特に限定されるものではないが 5 CTC以上が好まし い。 また、 本反応は窒素気流下で行うのが好ましく、 減圧条件下でも反応が実 施される。 また、 例えば、 水、 グリコールの添加、 あるいは、 塩化アンモニゥ 厶、 ナトリウムメトキシド、 ナトリウムアミ ド、 ブチルリチウムなどの塩基の 添加によって反応が促進され、 例えば、 ヒドロキノン、 フエノチアジン、 2, 6—ジ一 t—プチル一 4一メチルフエノールなどの酸化防止剤の添加によって、 副反応が抑制される。
• (b)法の縮合反応は通常のアミ ド結合形成反応で行われる。 例えば、 ジシクロ へキシルカルボジィミ ド、 Ν,Ν'—カルボ二ルジィミダゾール、 ジフエ二ルリン
酸アジド、 シァノリン酸ェチルなどのアミ ド結合形成試薬を用いて行うことが できる。 または、 カルボキシル基を活性エステル、 酸無水物、 混合酸無水物ま たは酸塩化物等に誘導してァミノ基と反応するなどの一般的な方法によって実 施できる。 いずれの場合にあっても、 例えばトリェチルァミン、 N-メチルピぺ リジン等の塩基の添加によって反応が促進される。
(c)法は、 例えばトリェチルァミン、 N-メチルビペリジン等塩基の存在下、 ス ルホン酸塩化物をァミノ基と反応する一般的な方法で実施される。
(d)法の縮合反応は、例えば炭酸ナトリウム、水酸化ナ卜リゥム等の無機塩基、 金属水素化物、 ナトリウム、 リチウム、 カリウム等の金属またはナトリウムメ トキシド、 力リウム tert-ブトキシド等のアルコキシドのような塩基の存在下で 反応する一般的な方法で実施できる。 また、 式 (7 ) において Y 1が水酸基で表 される化合物を用いて反応する場合は、 トリフエニルホスフィンとジェチルァ ゾジカルボキシレート等を用いて行う光延反応など、 一般的な方法で実施でき る。
(e)法は、 例えば炭酸力リゥム等の無機塩基、 水素化ナトリゥム等の金属水素 化物等の塩基、 ヨウ化テトラプチルアンモニゥム等の四級アンモニゥム塩等の 存在下で反応する一般的な方法で実施される。 四級アンモニゥム塩を用いる場 合、 用いる四級アンモニゥム塩の量は触媒量であっても構わない。
(f)法のハロゲン化は、 例えば (1 ) 塩化水素、 臭化水素、 ヨウ化水素等ハロ ゲン化水素を用いる反応、 (2 ) 三塩ィ匕リン、 五塩ィヒリン、 三臭ィ匕リン、 五臭 化リン等のハロゲン化リンによる反応、 (3 ) ホスホン酸トリフヱニル一ハロ ゲン化アルキル、 トリフヱニルホスフィン一四ハロゲン化炭素および関連化合 物による反応、 (4 ) メタンスルホニルクロリ ド、 p—トルエンスルホニルクロ リド等のスルホン酸ハロゲンィ匕物、 およびスルホン酸エステルによる反応、 (
5 ) 塩化チォニル、 臭化チォニル等ハロゲン化チォニルによる反応等の一般的 なハロゲン化の方法で実施される。
(g)法は、 例えば LiCl、 LiBr、 NaBr等の金属塩またはテトラプチルアンモニ -ゥムクロリ ド等四級アンモニゥム塩のを用いてハロゲン置換反応する一般的な 方法で実施される。
( )法は、 例えば炭酸ナトリゥム、 水酸化ナトリゥム、 等の無機塩基、 金属水 素化物、 ナトリウム、 リチウム、 カリウム等の金属およびナトリウムメトキシ ド、 カリウム tert-ブトキシド等のアルコ一ル金属等で、 式 (7)において Y 1が水 酸基である化合物を金属塩としたのち、式 ( 8)で表される化合物と反応する一般 的な方法で実施される。
式(1 )で表される化合物は、反応後、濾取あるいは抽出等の通常の操作により 単離することができ、 必要に応じて再結晶、 クロマトグラフィー等の操作によ り精製することができる。
かくして得られる式 ( 1 ) (式中、 Riは炭素数 1〜4のアルキル基、 nは 2ま たは 3の整数を表わし、 R2は一 (C H2) mR3 (mは 2または 3の整数、 R 3 は水酸基、 ハロゲン原子、 メタンスルホニルォキシ基または p—トルエンスルホ 二ルォキシ基を表わす。 )のアミノフヱノール誘導体は、 式 (9)
(式中、 R5は式(4 ) と同じ。;)で表される一級ァミンと反応させることにより 収率よく式 (4) で表される抗不整脈剤へと誘導することができる。
以下、 本発明を実施例を挙げて説明する。
実施例 1
N-(4-ヒドロキシフエニル) -3-メトキシプロパンアミ ドの製造
4-ァミノフエノール 54.6g、 3-メトキシプロピオン酸メチル 118.1gを混合し、 窒素気流下 140°Cでメタノ一ルを留去しながら 24時間反応した。 100°Cまで冷却 した後、 反応液にトルエン 500mlを加え室温まで冷却した。 析出結晶を濾取し た後、 トルエン 50mlで洗浄し、 N-(4-ヒドロキシフエニル) -3-メトキシプロパン アミド 83.9g (収率 85.9%)を得た。
融点 108.5〜110.5°C
•NMR(400MHz,DMSO-d6) δ ppm: 2.48(2H, t)、 3.24(3H, s)、 3.59(2H, t)、 6·67(2Η, d)、 7.36(2H,d)、 9.13(1H, s)、 9.66(1H, s)
実施例 2
N-[4-(2-ヒドロキシエトキシ)フエニル] -3-メ トキシプロパンアミ ドの製造
N-(4-ヒドロキシフエニル) -3-メトキシプロパンアミ ド 25.7g、 1,3-ジォキソラ ン -2-オン 28.5g、 ヨウ化テトラプチルアンモニゥム lO.Ogを混合し、 1 5 0 °Cから 160°Cで 5時間反応した。反応液を少量の酢酸ェチルに溶かし、 これをシ リカゲルカラムクロマトグラフィ一で精製した。シリ力ゲルは 300メッシュ相当 のもの 750gを使用し、展開はへキサン:酢酸ェチル =1:1の混合溶媒で行った。 目 的フラクションを集め濃縮乾固して N-[4-(2-ヒドロキシエトキシ)フ: ニル] -3- メトキシプロパンアミド 47.2g (収率 75.0%)を得た。 融点 99〜: L00°C NMR(90MHz,CDCl3) δ pm: 2.60(2H, 、 3.41(3H, s)、 3.73(2H, t)、
3.86-4.09(4H, m)、 7.12(2H,め、 7.45(2H, d)
実施例 3
N-[4-(2-メタンスルホニルォキシエトキシ)フエニル] -3-メ トキシプロパンアミ ドの製造
N-[4-(2-ヒドロキシエトキシ)フエニル] -3-メトキシプロパンアミ ド 47.9gお よびトリェチルァミン 60.7gを 750mlのクロ口ホルムに溶かした。 攪拌下これ を氷冷し内温 10°Cでメタンスルホニルクロライド 45.8g を滴下した。 その後、 室温として 1時間反応した。 反応液を希水酸化ナトリゥム水溶液、 次いで水で 洗った。
クロ口ホルム層を無水硫酸マグネシウムで脱水してから濃縮乾固した。
結晶として N-[4-(2-メタンスルホニルォキシエトキシ)フ Xニル] -3-メトキシプ 口パンアミ ド 59.1g (収率 93.0%)を得た。
融点 116〜117°C

実施例 4
. N- [4- [2-(p-トルエンスルホニルォキシ)エトキシ]フヱニル] -3-メトキシプロパン アミ ド
実施例 3と同様の方法により実施例 4の化合物を得た。
融点 102〜: L04°C
NMR(400MHz,DMSO-d6) δ ppm: 2.29(3H, s)、 2.49-2.52(2H, m)、 3.24(3H, s)、
3.60(2H, t)、 4.10-4.12(2H, m)、 4.30-4.32(2H, m)、 6.77(2H, d)、

実施例 5
N-[4-(2-クロ口エトキシ)フエニル] -3-メトキシプロパンアミ ドの製造
N-[4-[2-(p-トルエンスルホニルォキシ)ェトキシ]フェニル ]-3-メトキシプロパ ンアミ ド 59.0g、 塩化リチウム 12.5gを 200mlの DMFに溶かし 65〜70°Cで 3 時間反応した。 反応後、 反応液を 2500mlの氷水中に注ぎ、 析出した結晶を濾過 し、 次に水で洗った。 これを真空乾燥して N-[4-(2-クロ口エトキシ)フヱニル ]-3-メトキシプロパンアミ ド 31.5g (収率 81.8%)を得た。 融点 99.5〜: 100°C NMR(400MHz,DMSO-d6) δ ppm: 2.51(2H, t)、 3.24(3H, s)、 3.61(2H, 、 3.91(2H,t)、 4.20(2H, t)、 6.89(2H, 、 7.51(2H, d)、 9.81(1H, s) 産業上の利用分野
以上のように、 本発明により、 医薬農薬の合成中間体として有用な式 (1 ) で表されるァミノフヱノール誘導体を安価に提供することができる。
Claims
1. 式 (1)
RiO(CH2)nCONH
(式中、 Riは炭素数 1〜4のアルキル基、 nは 2または 3の整数を表わし、 R2 は水素原子、 または一 (CH2) mRs (mは 2または 3の整数、 R3は水酸基、 ハロゲン原子、メタンスルホニルォキシ基または p_トルエンスルホニルォキシ 基を表わす。 )で表わされるアミノフヱノール誘導体。
2. Riがメチル基であり、 nは 2の整数を表わし、 R2が水素原子である請求 項 1記載のァミノフエノール誘導体。
3. Riがメチル基であり、 n、 mはそれぞれ 2の整数を表わし、 R3が水酸基 、 塩素原子、 臭素原子、 ヨウ素原子、 メタンスルホニルォキシ基または p_トル エンスルホニルォキシ基である請求項 1記載のアミノフ Xノール誘導体。
4. 式 (2)
(式中、 R2は水素原子、 または一 (CH2) mRs (mは 2または 3の整数、 Rs は水酸基、 ハロゲン原子、 メタンスルホニルォキシ基、 p—トルエンスルホニル ォキシ基または _N (C2H5) COOC (CH3) 3を表わす。 ) で表わされるァ -ミノフエノール誘導体と、
(式中、 Riは炭素数 1〜4のアルキル基、 nは 2または 3の整数を表わし、 R4 は炭素数 1〜4のアルキル基を表す。 ) で表されるカルボン酸エステルを縮合 剤を用いることなく反応させ、 式 (1)
(式中、 Riは炭素数 1〜4のアルキル基、 nは 2または 3の整数を表わし、 R2 は水素原子、 または— (CH2) mR3 (mは 2または 3の整数、 R3は水酸基、 ハロゲン原子、 メタンスルホニルォキシ基、 p—トルエンスルホニルォキシ基ま たは _N (C2H5) COOC (CH3) 3を表わす。 )で表わされるアミノフエノ ール誘導体を製造する方法。
5. R2が水素原子もしくは、 R3が水酸基または一 N (C2H5) COOC (C H3) 3である請求項 4記載のァミノフヱノール誘導体を製造する方法。
