Abstract

Beschrieben ist ein Verfahren zur Identifikation von 5-Methylcytosin-Positionen in genomischer DNA, welches dadurch gekennzeichnet ist, daß folgende Verfahrensschritte ausgeführt werden: a) die genomische DNA einer Zelle, einer Zellinie, eines Gewebes oder eines Individuums wird chemisch so behandelt, daß Cytosin und 5-Methylcytosin unterschiedlich reagieren und sich in der Duplex ein unterschiedliches Basenpaarungsverhalten der beiden Produkte ergibt, b) derselbe Nukleinsäure-Abschnitt mittels einer Polymerasereaktion amplifiziert wird, c) der gleiche Nukleinsäure-Abschnitt mindestens einer weiteren Zelle, Zellinie, Gewebes oder Individuums oder einer beliebigen Referenz-DNA entsprechend den Punkten a) und b) behandelt wird, d) aus den mindestens zwei Amplifikaten der Punkte b) und c) Heteroduplexes gebildet werden, e) durch eine Reaktion, die spezifisch ist für nicht komplementäre Basenpaare, eine aufzeigbare Markierung in die Heteroduplex eingeführt wird.
