Abstract

Die Erfindung betrifft ein Verfahren und ein Testbesteck zur quantitativen Beurteilung der oxidativen Modifikation proteinhaltiger Stoffe bzw. des oxidativen Stresses in lebenden Organismen mit Hilfe der Messung der antiradikalen Eigenschaften ihrer proteinhaltigen Komponenten, z.B. Körperflüssigkeiten bzw. Gewebe. Das Wesen der Erfindung besteht darin, daß die Proben nach einer Abtrennung von niedermolekularen Antioxidantien, z.B. mittels der Gelchromatographie, in einem freiradikalgenerierenden System untersucht werden und die Meßergebnisse mit denen einer Probe gleicher Zusammensetzung von Aminosäuren, jedoch ohne oxidative Streßeinwirkung (Normwert), verglichen werden. Das Verfahren der Messung der antiradikalen Aktivität von Proteinen basiert auf dem erfindungsgemäßen Merkmal, daß einige nicht oxidativ modifizierte Aminosäuren als Bestandteile der Proteine diese Eigenschaft nicht besitzen, sie aber während der oxidativen Belastung erwerben.
