Description

COMPLEJO VEGETAL ANTIPOLUCION, PROCEDIMIENTO PARA SU OBTENCIÓN Y USO DEL MISMO EN LA PREPARACIÓN DE COMPOSICIONES FARMACÉUTICAS Y/O COSMÉTICASANTIPOLUTION VEGETABLE COMPLEX, PROCEDURE FOR OBTAINING AND USING THE SAME IN THE PREPARATION OF PHARMACEUTICAL AND / OR COSMETIC COMPOSITIONS
This invention relates to an anti-pollution plant complex, a method for obtaining it and the use thereof in the preparation of a pharmaceutical and / or cosmetic composition provided with protective action of the skin, hair, nails, mucous membranes and respiratory tract, individually or collectively, against the harmful effects of air pollution and UV solar radiation and meteorological changes.
The anti-pollution vegetable complex of this invention has, in particular, antioxidant action, chelating action, protective and / or stabilizing action of cell membranes, anti-cytotoxic action and moisturizing action, so it can be used as an active component in pharmaceutical and / or cosmetic compositions. to protect and prevent the action of environmental pollutants, as an anti-aging asset and to protect against meteorological changes and improve skin, hair and skin attachments. The complex according to the invention is characterized by inhibiting free radicals, chelating metals, stabilizing cell membranes against external aggressions and protecting against climatic changes.
The skin is an organ of vital importance, complex structure and multifunctional, whose surface reaches almost 2 m 2 . It occupies a border or interface position, limiting our body from the external environment and performing numerous functions, especially protection, acting as a barrier, maintaining the internal environment and protecting the body from physical, chemical and microbiological aggressions, maintaining body homeothermia and protecting from ultraviolet radiation. Industrial processes, transportation, fuel combustion and solid waste disposal are the main sources of pollution. This is a problem that affects more and more individuals every day and causing harmful effects both internally and externally.
Environmental pollutants, whether gases, particles or heavy metals, cause measurable effects on living things and materials, causing irritation to the skin and mucous membranes and respiratory problems. At the cutaneous level, the effects are manifested primarily in the outermost layers of the skin, being able to arrive in extreme cases to the appearance of necrosis of these tissues. Solar radiation is a fundamental energy source, vital for living organisms. However, an excess of it entails a series of short-term (burns) and long-term (aging, elastosis, cancer) negative effects. UV radiation is a generator of reactive substances (free radicals), which alone and / or in conjunction with external and xenobiotic agents can trigger lipid oxidation processes. These processes take place mainly on cell membranes, triggering chain reactions and generating more reactive species, which causes a destabilization of these membranes.
The skin acts as an interface between the body and the environment, is always in contact with the oxygen in the air, exposed to UV radiation and the influence of environmental agents (gases and heavy metals) and chemical agents.
The increase in the rate of chemical agents and pollutants present, together with the decrease in the ozone layer makes the skin more and more subject to more aggressions and if this We add that with the passage of time the skin decreases its thickness, while losing the flexibility and stiffness that characterizes it, making it necessary to use effective assets in this field. Therefore, it would be desirable to be able to have a product that exhibits demonstrated effectiveness on the different types of compounds involved in air pollution: heavy metals and polluting gases. Cosmetic compositions are already known from the state of the art comprising active ingredients of animal, synthetic or biosynthetic origin. In this sense, the active product according to the present invention can be anticipated here is entirely of plant origin.
Thus, in US Patent 5,571,503 cosmetic formulations with protective action against environmental contaminants, UVA, UVB and with sebum controlling action are described. However, this document does not describe or suggest that the formulations described therein are active against heavy metals.
However, the active components described therein (lipid derivatives) are completely different from those used in the present invention, as will be indicated in greater detail below. On the other hand, the compositions of the present invention require a lower active ingredient concentration than that mentioned in said document, to achieve an activity demonstrated in vitro, as will be discussed below. Patent FR 92 02611 describes a cosmetic formulation that contains between 9 and 12 jointly active components that appear to have activity against atmospheric aggressions, but nowhere in that document is evidence provided confirming the effectiveness of such formulations. Thus, FR 92 02611 requires the use of a total of 9-12 active ingredients to claim an unconfirmed anti-pollution action. In addition, the active product of the present invention is water soluble, of total solubility and very easy incorporation, and allows the formulation of all types of cosmetic and / or pharmaceutical products, including single phase aqueous gels, which does not allow the combination of 9- 12 ingredients since some of them are water soluble and some fat soluble.
The same occurs in US 5,571,503 where a multitude of ingredients are used to achieve the purpose recommended there. On the other hand, this reference is limited to an emulsion cosmetic formula.
Consequently, the state of the art mentioned above refers to active products only against heavy metals or against gases, but not against both types with only. It has been shown that the harmful effects of environmental pollution are combined in the skin with those of UN solar radiation and other climatic environmental aggressions, increasing these effects.
It has also been shown that, in the mechanism of the toxic action of heavy metals, the generation of free radicals and the destabilization of biological membranes are involved. Therefore, action against free radicals and against dermal dehydration is also important. In this sense, the product of this invention combines the anti-pollution action with the moisturizing and anti-free radical actions. The aforementioned patents do not describe or suggest all of these actions in any of the cases.
Now and according to the present invention, an active complex of plant origin is provided which has antioxidant action, chelating action, protective and / or stabilizing action of cell membranes, anti-cytotoxic action and moisturizing action, which can be used as active ingredient to protect and prevent the action of environmental pollutants, as an anti-aging active and to protect against meteorological changes and improve skin, hair and skin attachments. It should be understood that such examples are offered for purposes only. illustrative and in no way limiting the scope of the invention. E 1
Preparation of the Vegetable Antipollution Complex In the first place, the fruits are washed with deionized water (apple, pear, orange, bitter orange, grapefruit, lemon), crushing and subsequent elimination of the juices. Subsequently, its extraction is carried out at a frankly acidic pH (1.5-3.0) and temperatures above 60 ° C, which must not exceed 100 ° C in any case. Finally, the acid extractive solution is filtered and / or centrifuged. In some cases it can be concentrated first, filtered and then centrifuged. This extract (which we will call A) is kept under constant stirring, at a temperature below 60 QC and pH below 4 and phase B and phase C are added as described in its preparation.
To obtain phase B, the milling and disintegration of plants containing sapogenins (Licorice root and / or Horse Chestnut Seeds and / or Whole Centella Plant and / or Ginseng Root and / or Eleuterococcus Root) and / or plants that contain flavonoids (Fruits of Thistle and / or Ginkgo Leaves and / or Fruits of Rutáceas and / or Spring flowers and / or Flowers of hawthorn). Subsequently, the extraction is carried out by maceration using a hydroalcoholic extraction solvent containing between 50-70% by weight of ethyl alcohol, maintaining a plant-solvent extraction ratio of 1: 2 or 1: 2.5 depending on the case. , at a temperature not exceeding 50 ° C. The extraction process is extended for 24 hours. After separating the extraction liquids, this same extraction process is repeated. Finally, the extraction liquids are joined, making a first filtration on fabrics to remove any suspended particles and then a filtration on filter presses to obtain a clear and transparent solution whose pH should be around 5. This phase, which we will call B, is added to extract A by stirring and temperature below 60 ° C.
A a temperatura inferior a 40 C.For the preparation of phase C, one or more of the fruits of cereals and / or legumes are started. First, a cleaning is carried out to remove foreign particles, dust particles and foreign seeds and then it is subjected to conditioning at temperatures below 65QC. Then, it is milled by roller mill or other system, until the particle diameter is obtained between 250-20Tm (flour). Subsequently, the extraction is carried out by maceration using a slightly added aqueous solvent (pH 4-5). Finally, the extraction liquids are filtered until a completely transparent solution is obtained. This phase, which we will call C, is added by mechanical agitation to extract A at a temperature below 40 C.
Once phases B and C are added to extract A, stirring is continued until it is completely homogenized and neutralized to obtain the anti-pollution plant complex.

Para evaluar la eficacia del complejo vegetal antipolución, se procedió a realizar una serie de testsTo evaluate the effectiveness of the anti-pollution plant complex, a series of tests were carried out
Cin-vitro and C ± n-vivo to first know the effect on the cutaneous tissue of environmental pollutants on the chosen model systems and then evaluate the protective effect of this plant complex. For the realization of the Cin-vitro studies, the model systems of liposomes and cell cultures of fibroblasts were chosen.
En este caso fueron liposomas multilamelares que contiene principal- mente, ácido oleico (41%), ácido palmítico (22%) y ácido esteárico ( 18%) .Liposomes are used as an alternative model for the study of lipid peroxidation through the use of artificial membranes. Due to the great difficulty of working with natural membranes, models have been developed to simulate the effect of reactive oxygen species on biological structures, quantify the deterioration produced and study the protection mechanisms. Liposomes are microscopic vesicles of one or more concentric bilayers, which contain aqueous compartments inside. The lipid composition of these vesicles is selected based on the type of study to be performed. In this case they were multilamellar liposomes that contain principal- mind, oleic acid (41%), palmitic acid (22%) and stearic acid (18%).
El TBA reacciona específicamente con el MDA dando un producto coloreado cuyo máximo de absorbancia se encuentra a 530 nm.In the first stage, the effect of different heavy metal salts (Pb, Zn, Cd, Hg) on the lipid peroxidation of cell membranes has been determined by the thiobarbituric acid assay and then the inhibition of the lipid peroxidation due to the presence of the antipollution plant complex (250Tg / mL). The use of 2-thiobarbituric acid (TBA) to study the evolution of lipid oxidation, is based on the ability of this acid to react with substances that originate during the degradation of hydroperoxides. These compounds are primary products of oxidation. lipid, which end up causing malonic dialdehyde (MDA). The TBA reacts specifically with the MDA giving a colored product whose maximum absorbance is at 530 nm.
The results obtained in this first stage demonstrate that lipid lipoperoxidation induced by heavy metals (Pb, Hg, Zn, Cd) is appreciable at long incubation times (72h). The anti-pollution vegetable complex exerts a protective action on lipid peroxidation that reaches up to 40%. In the second stage, the cytotoxicity of the aforementioned metals in cell cultures of fibroblasts has been studied and the protective effect of the antipollution plant complex has been evaluated through cell viability, morphological changes and lactate dehydrogenase (LDH) levels in the culture medium.
The fibroblast cell cultures used at this stage correspond to the Foreskin cell line (ATCC CRL-1635) corresponding to human facial skin fibroblasts that came from Boehringer Mannheim. Cell viability has been determined by the method of exclusion of the blue Trypan and the cell count has been carried out in a Neubauer chamber. Morphological changes have been followed by electron microscopy in different periods of time. LDH levels have been determined spectrophotometrically, using an LDH kit (LDH MPR 1, Boehringer Mannheim). Lactate dehydrogenase is a ubiquitous cytosolic enzyme in all cell types and when there is cell damage or death a permeabilization of the cytoplasmic membrane occurs so that cytosolic enzymes are released to the extracellular environment and therefore an increase in their extracellular levels .
The results obtained in this second stage demonstrate in the first place the total absence of toxicity of the antipollution plant complex. This anti-pollution plant complex (250T1) exerts a protective effect that reaches values of 78% in cell count and 67% by measuring the activity of LDH (0.3M Zn) and 20% in cell count in the case of Cd 0.2mM and 11% in cell count for 0.05mM Hg.
The results obtained with the gases show the cytotoxic effect on cell cultures and their activating effect on lipoperoxidation processes. The anti-pollution plant complex effectively protects against this toxic action.
In order to verify the "in-vivo" efficacy of the antipollution plant complex, "non-invasive" biophysical testing techniques were carried out. The hydration of the skin surface was measured, using two cosmetic formulations, one of which contained 2% of the active substance, while the other was a control or placebo formulation. Through a corneometer (CM 825 Courage & Khazaka) the capacitance values of the skin surface were recorded. This method is based on the dielectric constant of water, usually of 81 capacitance units, while the rest of the substances have values below 7 units, so that the measured capacitance changes are a measurable function of water levels, and therefore of skin hydration, the experiment was designed as an experimental, randomized and double blind study. The results obtained show that the use of cosmetic and pharmaceutical formulations containing anti-pollution plant complex increases skin hydration.
