Abstract

Gegenstand der vorliegenden Erfindung sind Polymerasenchimären, die aus Aminosäurefragmenten, die Domänen repräsentieren, bestehen und die - im Hinblick auf eine bestimmte Verwendung - vorteilhafte Eigenschaften von natürlich vorkommenden Polymerasen in sich vereinen. Überraschenderweise konnte gezeigt werden, daß die Domänen aus den unterschiedlichen Enzymen in der Chimäre aktiv sind und ein kooperatives Verhalten zeigen. Weiterhin ist Gegenstand der vorliegenden Erfindung ein Verfahren zur Herstellung der erfindungsgemäßen Chimären sowie die Verwendung dieser Chimären bei der Synthese von Nukleinsäuren z.B. während der Polymerase-Ketten-Reaktion. Ein weiterer Gegenstand der vorliegenden Erfindung ist ein Kit, der die erfindungsgemäßen Polymerasenchimären enthält.
