発明の分野
本発明は、 高濃度の炭素源に耐性を有するモルティ エ レラ (Mort ierella ) 属に属する微生物、 該微生物を用いた醱酵法によるァラ キ ド ン酸、 ジホモー ア 一 リ ノ レ ン酸及び Z又はエイ コサペンタエン 明
酸及びそれらを含有する脂質の製造方法並びに該微生物を含む動物 田
用飼料に関する。
背景技術
ァラキ ドン酸 ( 5 、 8 、 1 1 、 1 4 一エイ コサテ トラェ ン酸) や 、 ジホモー ア 一 リ ノ レ ン酸 ( 8、 1 1 、 1 4 —エイ コサ ト リ ェン酸 ) 、 エイ コサペンタエン酸 ( 5 、 8 、 1 1 、 1 4 、 1 7 —エイ コサ ペンタエン酸) は、 プロスタ グラ ン ジ ン、 ロイ コ ト リ ェン、 ト ロ ン ボキサン等のエイ コサノ ィ ドの前駆体となり得るほか、 その脂肪酸 自身のもつ生理活性からも注目されている。 例えばエイ コサペンタ ェン酸は血栓防止作用あるいは脂質低下作用に基づいて健康食品や 医薬品と して市販されている。
またァラキ ドン酸は、 近年、 ドコサへキサェ ン酸と同 じ く 母乳中 に含まれており、 乳児の発育に役立つとの報告がある ( 「Advances in Polyunsaturated Fatty Acid Research 」 , Elsevier Science Publishers, 1993, pp.261-264 ) 。 さ らに、 胎児の身長や脳の発 育における重要性が報告されている (Proc. Natl. Acad. Sci. USA , 90, 1073-1077 (1993), Lancet, 344, 1319-1322 (1994) ) 。
このよ う なァラキ ドン酸、 ジホモー ア 一 リ ノ レ ン酸又はエイ コサ
ペンタエン酸の製造方法と しては、 モルティ エレラ (Mortierella
) 属に属する微生物を使用 して、 従来より も高収率で得る方法が開 発されている (特公平 7 _ 3 4 7 5 2、 特開平 6 — 1 5 3 9 7 0、 特開平 8 - 2 1 4 8 9 3 , Chapter 4 in Industrial Applications of Single Cell Oil, ed. by D. J. Kyle and C. しかしながらこ こで使用されているモルティ エレラ ( Mor t i erel 1 a_) 属に属する微生物は、 いずれも高グルコース濃度に対する耐性 が低く 、 実際の通気攪拌培養での工業生産ではグルコースの濃度が 2重量%〜 4重量%となるよう にグルコースの流加培養を実施して いるのが現状であり、 製造工程が煩雑になつてしま う という問題点 があった。 また従来ァラキ ドン酸、 ジホモー ア リ ノ レン酸又はェ ィ コサペンタエン酸に使用されているモルティ エレラ (Mortierell a ) 属モルティ エレラ亜属 (Subgenus Mortierella) に属する微生 物は、 ア ー リ ノ レン酸の製造に用いられているモルティ エレラ r t i erel la ) 属マイ ク ロムコール亜属 ( Subgenus Mi coromucor) に 属する微生物と比較して、 明らかに菌の生育度 (培地あたりの乾燥 菌体重量) が低く 、 その結果、 培地当たりで得られる高度不飽和脂 肪酸を含有する脂質の生産量は低いものであった (例えば、 モルテ ィ エレラ亜属に属する Mortierella alpinaの生育度が 2 2. 5 g/ L に対して、 マイ ク ロムコ一ノレ亜属に属する Mortierel la ramann iana var. angul ispora の生育度は 7 9 g/L に達している (Chap ter 5, Chapter 7 in Industrial Applications of Single Cell 0 i 1 , ed. by D. モルティ エレラ属モルティ エレラ亜属に属する微生物のァラキ ド ン酸の生産性に関して、 これまでに、 例えば特開平 6 — 1 5 3 9 7 0の実施例では、 初発グルコース濃度 2 %、 培養 7 日間で 4. 0 9 g/L のァラキ ドン酸が生産される こ とが、 また特開平 8 — 2 1 4 8
9 3の実施例では、 初発グルコース濃度 4. 3 %、 培養 3 日間で 2
. 3 g/L のァラキ ドン酸が生産されるこ と力《、 それぞれ開示されて いる。
. 炭素源の濃度を高めた培地用いた例と しては、 「Chapter 4 in I ndustr ial Applications of Single Cell Oil, ed. by D. J. Kyle and C. Rat ledge, American Oil Chemists' Society, Illinois, 1 992 」 の実験結果では、 初発デキス ト ロ一ス濃度 9. 8 %、 培養 7 日間で 1 . 5 g/L 、 初発デキス ト ロース濃度 9. 8 %、 培養 1 6 日 間で 9. 1 /L のァラキ ドン酸が生産されているが、 初発デキス 卜 ロース濃度が高いため特に菌の生育が悪く 、 培養 7 日 目でのァラキ ドン酸生産量はわずか 1 . 5 g/L と低く 、 生産量を確保するために 培養日数を 1 6 日間と している。
また、 W 0 9 6 / 2 1 0 3 7の実施例では、 初発グルコ一ス濃度
1 0 % 、 培養 8 日間で 5. 3 g/L のァラキ ドン酸が生産されている が、 高グルコース濃度に対する菌の生育阻害を抑制する目的で、 pH コ ン ト ロール、 塩類添加などの手法を駆使しており、 煩雑な操作が 必要である。
このよ う に、 微生物を用いた醱酵法による物質の生産では、 一般 に生産性を高めよう とする場合には微生物の生育量を増やすか、 微 生物当たりの生産量を増やすなどの方法が取られる。 微生物生育の ための栄養源となる炭素源の濃度を高める こ とは微生物の生育量を 増やすこ とにつながり、 出来るだけ高い炭素源の濃度での培養が好 ま しい。 しかし一方で、 炭素源の濃度を高く するこ とは、 微生物の
生育にとって浸透圧の関係で過酷な条件となり、 炭素源の濃度を高 く する こ とは微生物の生育を抑制する こ とになつてしま う。
前述の、 初発のグルコース濃度を高めた場合には菌の生育が悪く なり 7 日間の培養ではァラキ ドン酸の生産量は 1. 5 g / L程度し か得られ例や、 初発のダルコ一ス濃度を高めるために pHコ ン ト ロ一 ルゃ塩類添加などを行って培地の調整を行っている例は、 微生物を 用いた醱酵法による物質生産の難しさを示している例と見るこ と も できる。
従って、 高濃度の炭素源に対する耐性があり、 初発のグルコース 濃度を高めた培地中でも十分な生育度を有する微生物を見出し、 こ れを用いてァラキ ドン酸、 ジホモー ア 一 リ ノ レ ン酸又はエイ コサぺ ンタエン酸を簡便な操作で効率良く 、 かつ大量に製造する方法を開 発するこ とが望まれていた。 発明の開示
従って本発明は、 高濃度の炭素源に耐性を有するモルティ エ レラ (Mortierella ) 属に属する微生物を利用 して、 ァラキ ドン酸、 ジ ホモ一 ァ 一 リ ノ レン酸及び/又はエイ コサペンタエン酸又はそれら を含有する脂質の効率的な製造方法、 並びに高濃度の炭素源に耐性 を有するモルティ エレラ (Mortierella ) 属に属する微生物、 及び 該微生物を含有する動物用飼料を提供しょう とする ものである。
本発明者等は、 上記の目的を達成するために種々研究した結果、 高濃度の炭素源に対する耐性があり、 培養開始時の炭素源濃度を高 めた培地中でも菌の生育度が高く 、 その結果と してァラキ ド ン酸、 ジホモー ァ 一 リ ノ レン酸及び Z又はエイ コサペンタエン酸又はそれ らを含有する脂質の生産性が高ま ったモルティ エレラ属に属する微 生物を見いだし本発明を完成した。
より具体的には、 PH制御、 無機塩類添加などの煩雑な操作を駆使 せず、 炭素源と してグルコース、 窒素源と して酵母エキスなどを用 いた単純な培地で、 ァラキ ドン酸、 ジホモー ア 一 リ ノ レン酸及び Z 又はエイ コサペンタエン酸の生産に従来使用されているモルティ エ レラ属モルティ エレラ亜属に属する微生物 ( Mortierel la alpina 1 FO 8568)では菌の生育度が低下する炭素源濃度である 4重量%以上 、 より特徴的には、 ほとんど菌の生育が認められない炭素源濃度で ある 8重量%以上、 さ らには 1 1 %以上であっても菌の生育度が高 く 、 その結果と してァラキ ドン酸、 ジホモ一 ァ 一 リ ノ レン酸及び/ 又はエイ コサペンタエン酸又はそれらを含有する脂質の生産性が高 ま ったモルティ エレラ属に属する新しい微生物を見いだし本発明を 兀成した。
従って本発明は、 高濃度の炭素源に耐性を有するモルティ エレラ (Mortierella ) 属に属する微生物を培養開始時の炭素源濃度が 4 重量%以上の培地中で培養して、 ァラキ ドン酸又はこれを含有する 脂質を生成せしめ、 そ してァラキ ドン酸又はこれを含有する脂質を 採取する こ とを特徴とするァラキ ドン酸又はこれを含有する脂質の 製造方法を提供する。
即ち、 本発明によれば、 高濃度の炭素源に耐性を有するモルティ エレラ (Mortierel la)属に属する微生物を、 培養開始時の炭素源濃 度が 4重量%以上の培地中で培養するこ とにより、 8 日間の培養で 7 gZL以上のァラキ ドン酸を製造するこ とが出来る。 培養開始時 の炭素源濃度をさ らに高めるとと もに培養中に炭素源を流加するこ とによりァラキ ドン酸の生産性を更に向上させるこ とが出来る。 例 えば、 培養開始時の炭素源濃度を 8重量%以上とすると約 1 0 g / L以上のァラキ ドン酸を製造でき、 培養開始時の炭素源濃度を 1 1 重量%以上とする こ とにより、 約 1 4 g/L以上のァラキ ドン酸を
製造できる。 これは、 これまで知られていたモルティ エレラ属の微 生物を用いたァラキ ドン酸の生産性 5. 3 g/L (W096/21037)の 約 3倍の生産性である。
本発明はまた、 高濃度の炭素源に耐性を有するモルティ エレラ ( Mortierella ) 属に属する微生物を、 Δ 5不飽和化酵素阻害剤を含 有する培地中で培養して、 ジホモー ァ 一 リ ノ レ ン酸又はこれを含有 する脂質を生成せしめ、 そ してジホモー ア 一 リ ノ レ ン酸又はこれを 含有する脂質を採取するこ とを特徴とする ジホモ— ア ー リ ノ レ ン酸 又はこれを含有する脂質の製造方法を提供する。
本発明はまた、 高濃度の炭素源に耐性を有するモルティ エレラ ( Mortierella ) 属に属する微生物を、 2 0 °C以下で培養して、 エイ コサペンタエン酸又はこれを含有する脂質を生成せしめ、 そ してェ ィ コサペンタエン酸又はこれを含有する脂質を採取するこ とを特徴 とするエイ コサペンタエン酸又はこれを含有する脂質の製造方法を 提供する。
本発明はまた、 高濃度の炭素源に耐性を有するモルティ エレラ ( Mortierella ) 属に属する微生物を含んでなる動物用飼料を提供す る。
本発明はまた、 高濃度の炭素源に耐性を有するモルティ エ レラ属 S A M 2 1 9 7株 (F E RM B P— 6 2 6 1 ) を提供する。 発明の実施の形態
本発明で使用される高濃度の炭素源に耐性を有するモルティ エレ ラ (Mortierella ) 属に属する微生物とは、 モルティ エレラ (Mor_^ ierella ) 属に属する微生物であって、 培養開始時の炭素源濃度が 4 %以上、 好ま し く は 8 %以上、 より好ま し く は 1 1 %以上で、 培 養開始時の窒素源濃度が 2 %以上である液体培地を用いて通気撹拌
培養により常法に従って培養を 2〜 1 2 日間、 好ま し く は 5〜 1 2 日間、 より好ま し く は 5〜 1 0 日間行ったとき、 培地あたり、 ァラ キ ド ン酸を 7 gZL以上生産できる能力を有する微生物である。
このような特徴を有するモルティ エレラ (Mortierella ) 属に属 する微生物を用いるこ とによって、 培養条件や培地への添加物によ つては、 ァラキ ドン酸の他、 ジホモー ア 一 リ ノ レン酸又はエイ コサ ペンタエン酸も従来より高収率で得るこ とができる。
なおモルティ エレラ (Mortierella ) 属には、 モルティ エレラ亜 属 ( Subgenus Mortierella) とマイ ク ロ ム コ 一ノレ亜属 (Subgenus— M icoromucor) があるこ とが知られており、 ァラキ ドン酸、 ジホモ一 Ί リ ノ レ ン酸及び Z又はエイ コサペンタエン酸の製造にはモルテ ィ エ レラ亜属 (Subgenus Mortierella) に属する微生物が望ま しい またモルティ エレラ亜属 (Subgenus Mortierella) には、 ァノレピ ナ区 (Section Alpina ) 、 フ ィ グロフ イ ラ区 (Section Hygroph i la ) 、 モノレテ ィ エ レラ区 (Section Mortierella ) 、 シュマッカ リ区 (Section Schmuckeri ) 、 シ ンプレ ッ ク ス区 (Section Sim plex) 、 スピノサ区 (Section Spinosa) 、 スチロスポラ区 (Sect i on Stylospora ) 等があるこ とが知られており、 ァラキ ド ン酸、 ジホモ一 Ί ― リ ノ レン酸及び/又はエイ コサペンタエン酸の製造に はアルピナ区 (Section 一 Αΐρίϋ) に属する微生物が望ま しい。 またアルピナ区 (Section Alpina ) にはアルピナ (alpina) 種 やア リ アセァ (alliacea) 種等があるこ と力く、 フ ィ グロフ イ ラ区 ( Section Hygrophila ) にはエロ ンガ、夕 (elongata) 種ゃ ミ ヌティ ッ シマ ( minutiss ima ) 種等があるこ と力く、 モルティ エレラ区 ( S e ct ion Mortierella) にはポ リ セフ ァラ (po cephala ) 種等力くあ るこ と力く、 スチロスポラ区 (Section Stylospora ) にはノく一ティ
シラタ (verticillata) 種等があることが知られている。
具体的な菌株と しては、 例えば本発明者らによって土壌から分離 され、 ブダぺブ ト条約に基き工業技術院生命工学工業技術研究所 ( 茨城県つく ば巿東 1 丁目 1番 3号) に、 1 9 9 7年 3 月 3 日に、 菌 株 Mortierella sp. S A M 2 1 9 7株 ( F E RM B P— 6 2 6 1 号) と して国際寄託された微生物を使用することができる。
Mortierella sp. S AM 2 1 9 7株は下記の菌学的性質を有す る。

多数の胞子のう胞子を含む胞子のうは亜球形で、 直径は約 1 5 /
m、 胞子のう胞子は、 単細胞で楕円形、 無色、 長さ 3〜 5 z m、 幅 1 . 5〜 2. 5 / m。 数個の胞子のう胞子を含む胞子のう は不規則 な形態をとる。 楕円形、 亜球形あるいは不規則な形態の壁の薄い厚 膜胞子を形成する。
上記の結果から、 S AM 2 1 9 7株は、 Mortierella 属 Mortiere 11a 亜属に属する ものと同定された。
本発明の高濃度の炭素源に耐性を有するモルティ エ レラ ( Mortie rella ) 属に属する微生物は、 以下に示す培養条件及び培養方法で 培養するこ とができる。
本発明に使用される菌株を培養する為には、 その菌株の胞子、 菌 糸、 又は予め培養して得られた前培養液を、 液体培地又は固体培地 に接種し培養する。 液体培地の場合に、 炭素源と してはグルコース 、 フ ラ ク ト 一ス、 キシロース、 サ ッ カ ロース、 マル ト ース、 可溶性 デンプン、 糖蜜、 グ リ セロール、 マ ンニ ト ール、 ク ェ ン酸、 コー ン スターチ等の一般的に使用されている ものがいずれも使用できるが
、 特にグルコース、 フ ラ ク ト 一ス、 マル ト ース、 グリ セロール、 ク ェン酸、 コー ンスターチが好ま しい。
窒素源と してはペプ ト ン、 酵母エキス、 麦芽エキス、 肉エキス、 カザミ ノ酸、 コーンスティ 一プリ カ一、 大豆タ ンパク等の天然窒素 源の他に、 尿素等の有機窒素源、 ならびに硝酸ナ ト リ ウム、 硝酸ァ ンモニゥム、 硫酸ア ンモニゥム等の無機窒素源等の一般的に使用さ れている ものを用いる こ とができる。 この他必要に応じて微量栄養 源と して用いる、 リ ン酸カ リ ウム、 リ ン酸二水素カ リ ウム等の リ ン 酸塩、 硫酸ア ンモニゥム、 硫酸ナ ト リ ウム、 硫酸マグネシウム、 硫 酸鉄、 硫酸銅、 塩化マグネ シウ ム、 塩化カ リ ウム等の無機塩及びビ タ ミ ン等全て一般的に使用されている ものがいずれも使用できる。 また本発明においては、 培地中に目的とするァラキ ドン酸、 ジホ
モー Ί リ ノ レ ン酸及び/又はエイ コサペンタエン酸の基質を添加 する こ とにより、 該高度不飽和脂肪酸の蓄積を促進するこ と もでき る。 この基質と して、 例えば、 へキサデカ ン若し く はォク タデカ ン のごとき炭化水素 ; ォレイ ン酸若し く はリ ノ 一ル酸のごとき脂肪酸 又はその塩、 例えばナ ト リ ウム塩若し く はカ リ ウム塩、 又は脂肪酸 エステル、 例えばェチルエステル、 グ リ セ リ ン脂肪酸エステル、 ソ ルビタ ン脂肪酸エステル ; 又はオリ 一ブ油、 大豆油、 なたね油、 綿 実油若し く はヤシ油のごとき油脂類を単独で、 又は組み合わせて使 用できる。 し力、し、 これらに限られる ものではない。 基質の総添加 量は培地に対して 0 . 0 0 1 〜 1 0重量%、 好ま し く は 0 . 5 〜 1 0重量%である。 またこれらの基質を唯一の炭素源と して培養して もよ く 、 この場合には必要に応じてさ らに添加量を増加させる。 上記の炭素源、 窒素源、 無機塩類、 ビタ ミ ン及び Ζ又は基質等の 培地成分は、 培養開始前の培地及び/又は培養中の培養液に添加す る こ とができる。 これらの成分は一度に添加するこ と もでき、 又は 連続的に、 若し く は複数回に分けて経時的に添加する こ と もできる 。 これらの培地成分は各々単独で、 又は予め混合して添加するこ と ができる。 これらの培地成分の内、 無機塩類、 ビタ ミ ン類及び Ζ又 は基質等の培地成分は微生物の生育を害しない濃度であれば特に制 限はな く 、 一般的に使用される範囲で用いればよい。
一方、 炭素源は、 培養開始時の培地中の炭素源濃度を 4重量%以 上、 好ま し く は 8 重量%以上、 より好ま し く は 1 1 重量%以上とす る。 これによつて培養操作の簡素化をはかる こ とができるよう にな り、 さ らに培養中の培養液に添加する場合にも、 炭素源を 2重量% 以上、 好ま し く は 4重量%以上、 より好ま し く は 6 重量%以上の濃 度で流加するこ とが可能となる こ とから、 更に効率良く 菌の増殖並 びに目的とする高度不飽和脂肪酸の生産を行う こ とができる。
また窒素源の総添加量は、 0 . 0 1 〜 1 0重量%、 好ま し く は 0 . 1 〜 1 0重量%の濃度とするのが良い。 また培養開始時の培地中 の窒素源濃度は 2重量%以上とするこ とができ、 また培養途中の培 養液に窒素源を流加するこ と もできる。
培養温度は 5 〜 4 0 °C、 好ま し く は 2 0 〜 3 0 °Cと し、 また 2 0 〜 3 0 °Cにて培養して菌体を増殖せしめた後 5 〜 2 0 °Cにて培養を 続けて目的とする高度不飽和脂肪酸を生産せしめる。 このよ う な温 度管理により、 生成脂肪酸中の高度不飽和脂肪酸の比率を上昇せし める こ とができる。 培地の pHも特に制御する必要はな く 、 通常用い る pHの範囲である 4 〜 1 0 、 好ま し く は 6 〜 9 と して通気攪拌培養 、 振盪培養、 又は静置培養を行う。 特に液体培地を用いた通気撹拌 培養は、 目的とする高度不飽和脂肪酸を効率よ く 製造するのに好ま しい。 培養は通常 2〜 1 2 日間、 好ま し く は 5 〜 1 2 日間、 より好 ま し く は 5 〜 1 0 日間行う。
培養温度を培養開始時よりあるいは培養途中より 2 0 °C以下にす るこ とでエイ コサペンタエン酸を産生する こ とができる。 また、 ェ ィ コサペンタエン酸の前駆体である α リ ノ レン酸あるいは α ― リ ノ レン酸を含有する脂質 (例えば、 亜麻仁油、 シ ソ油など) を培地 に添加すればエイ コサペンタエン酸が効率的に産生される し、 培養 温度を 2 0 °C以下にするこ とで、 さ らに生産性が向上する。
固体培養で培養する場合は、 固形物重量に対して 5 0 〜 1 0 0重 量%の水を加えたふすま、 もみがら、 米ぬか等を用い、 5 〜 4 0 °C 、 好ま し く は前記の温度において、 3 〜 1 4 日間培養を行う。 この 場合に必要に応じて培地中に窒素源、 無機塩類、 微量栄養源を加え る こ とができる。
またジホモ一 ァ リ ノ レ ン酸の生産量を増加せしめるためには、 本発明の微生物を Δ 5不飽和化酵素阻害剤の存在下で培養するのが
好ま しい。 Δ 5不飽和化酵素阻害剤と しては、 セサ ミ ン、 セサ ミ ノ —ル、 ェピセサ ミ ン、 ェピセサ ミ ノ 一ル、 セサモ リ ン、 2 — ( 3, 4 ー メ チ レ ンジォキシフ ヱニル) 一 6 — ( 3 — メ ト キ シー 4 ー ヒ ド ロキシフ エニル) 一 3, 7 — ジォキサビシク ロ [ 3. 3. 0 ] ォク タ ン、 2, 6 — ビス一 ( 3 — メ トキシ一 4 — ヒ ドロキ シフ ヱニル) - 3 , 7 — ジォキサビシク ロ [ 3. 3. 0 ] オク タ ン又は 2 — ( 3 , 4 ー メ チ レ ン ジォキシフ エニル) ー 6 — ( 3 — メ ト キシー 4 — ヒ ドロキシフ エ ノ キシ) 一 3, 7 — ジォキサ ビシ ク ロ [ 3 . 3. 0 ] オク タ ン等の リ グナ ン類化合物 ; ピぺロニルブ ト キサイ ド ; ク ルク ミ ン ; 胡麻油 ; 落花生油 ; 香辛性植物、 例えばタラ ゴン (Tarragon ) 、 イ ノ ン ド種子 (Dill Seed ) 、 パセ リ (Parsley ) 、 ゥ コ ン ( Turmeric) 、 ナツメ グ (Nutmeg) 等からの抽出物 ; 五加皮の抽出物 ; 桐木の抽出物 ; 白果樹皮の抽出物 ; ヒハツの抽出物 ; 細辛の抽出 物 ; 胡麻油、 胡麻油製造過程の副産物 (例えば胡麻種子の脱脂粕、 胡麻油の脱臭スカム等) 、 胡麻種子等からの抽出物などを使用する こ とができる。
なお、 胡麻油、 胡麻油製造過程の副産物 (例えば胡麻種子の脱脂 粕、 胡麻油の脱臭スカム等) 、 胡麻種子等からの抽出物は、 胡麻油 とは実質的に非混和性であり且つ Δ 5不飽和化酵素阻害活性を有す る成分を抽出 · 溶解するこ とのできる種々 の有機溶剤を用いて調製 するこ とができる。 このような有機溶剤と して、 例えばアセ ト ン、 メ チソレエチルケ ト ン、 ジェチルケ ト ン、 メ タ ノ ール、 エタ ノ ール等 を挙げるこ とができる。 また溶媒抽出だけでなく水蒸気蒸留や分子 蒸留等によつて分離されたものも抽出物に含まれる。 また上記香辛 性植物からの抽出物は、 常用の溶剤 (例えばジク ロ ロメ タ ン、 エタ ノ ール、 メ タ ノ ール、 ェチルエーテル等) を用いて調製する こ とが できる。
上記 Δ 5不飽和化酵素阻害剤の培地への添加量はおよそ次の通り である。 胡麻油又は落花生油、 あるいはこの両者の総添加量は培地 に対して 0 . 0 0 1 〜 1 0重量%、 好ま し く は 0 . 5 〜 1 0重量% である。 上記胡麻油、 胡麻油製造過程の副産物 (例えば胡麻種子の 脱脂柏、 胡麻油の脱臭スカム等) 、 胡麻種子等からの抽出物を添加 する場合、 その添加量は培地に対して 3 X 1 0 — 3〜 3 X 1 0 '重量 %である。
また、 セサ ミ ン、 セサ ミ ノ ール、 ェピセサ ミ ン、 ェピセサ ミ ノ ー ル等の リ グナン類化合物を添加する場合、 その添加量 (これらの 2 種類以上を組み合わせて使用する場合はその合計量) は、 培地に対 して 1 X 1 0 — 3〜 1 X 1 0 — 1重量%である。 これらの添加物類は生 産微生物を接種する前又はその直後の培地に加えてもよ く 、 又は培 養を開始した直後の培地に加えてもよ く 、 あるいは両時点で加えて もよい。 培養開始後の添加は 1 回でもよ く 、 又は複数回に分けて間 欠的に添加してもよい。
このよう にして培養して、 菌体内にァラキ ドン酸、 ジホモ一 ァ ー リ ノ レン酸及び/又はエイ コサペンタエン酸を大量に含有する脂質 が生成蓄積される。
目的とする高度不飽和脂肪酸は、 培養によって脂質を製造する途 中の培養液若し く はその殺菌した培養液、 または培養終了後の培養 液若し く はその殺菌した培養液、 またはそれぞれから集菌した培養 菌体若し く はその乾燥物から常法に従って得るこ とができる。 液体 培地を使用 した場合には、 培養菌体から、 例えば、 次のよう に して 目的とする高度不飽和脂肪酸の採取を行う。
培養終了後、 培養液より遠心分離及び/又は濾過等の常用の固液 分離手段により培養菌体を得る。 該菌体は好ま し く は、 水洗、 破砕 、 乾燥する。 乾燥は、 凍結乾燥、 風乾等によって行う こ とができる
。 乾燥菌体は、 好ま し く は窒素気流下で有機溶媒によって抽出処理 する。 有機溶媒と してはエーテル、 へキサン、 メ タ ノ ール、 ェタノ —ル、 ク ロ口ホルム、 ジク ロロメ タ ン、 石油エーテル等を用いるこ とができ、 又メ タノ 一ルと石油エーテルの交互抽出やク ロ 口ホルム 一メ タ ノ ール一水の一層系の溶媒を用いた抽出によっても良好な結 果を得る こ とができ、 好ま し く はへキサンを用いて抽出する。
抽出物から減圧下で有機溶媒を留去する こ とによ り、 高濃度の高 度不飽和脂肪酸を含有した脂質が得られる。 また、 上記の方法に代 えて湿菌体を用いて抽出を行う こ とができる。 この場合にはメ タノ —ル、 エタノ ール等の水に対して相溶性の溶媒、 又はこれらと水及 び/又は他の溶媒とからなる水に対して相溶性の混合溶媒を使用す る。 その他の手順は上記と同様である。
上記のよう にして得られた脂質中には、 各種高度不飽和脂肪酸が 脂質化合物、 例えば脂肪の構成成分と して含まれている。 これらを 直接分離するこ と もできるが、 低級アルコールとのエステル、 例え ばジホモ- ァ - リ ノ レン酸メ チル、 ァラキ ドン酸メ チル、 エイ コサ ペンタエン酸メ チル等と して分離するのが好ま しい。 このようなェ ステルにするこ とにより、 他の脂質成分から容易に分離するこ とが でき、 また、 培養中に生成する他の脂肪酸、 例えばパル ミ チン酸、 ォレイ ン酸等 (これらも、 高度不飽和脂肪酸のエステル化に際して エステル化される) から容易に分離する こ とができる。 例えば、 高 度不—飽和脂肪酸のメ チルエステルを得るには、 前記の抽出脂質を無 水メ タノ ール一塩酸 5〜 1 0 %、 B F 一メ タノ ール 1 0〜 5 0 % 等により、 室温にて 1 〜 2 4 時間処理するのが好ま しい。
前記の処理液から高度不飽和脂肪酸メ チルエステルを回収するに はへキサン、 エーテル、 酢酸ェチル等の有機溶媒で抽出するのが好 ま しい。 次に、 この抽出液を無水硫酸ナ ト リ ウム等により乾燥し、
有機溶媒を好ま し く は減圧下で留去する こ とにより主と して脂肪酸 エステルからなる混合物が得られる。 こ う して得られる脂肪酸エス テルの混合物には、 目的とする高度不飽和脂肪酸メ チルエステルの 他に、 パルミ チン酸メ チルエステル、 ステア リ ン酸メ チルエステル 、 ォレイ ン酸メ チルエステル、 ァ - リ ノ レン酸メ チルエステル等の 脂肪酸メ チルエステルが含まれている。
これらの脂肪酸メ チルエステル混合物から高度不飽和脂肪酸メ チ ルエステル、 すなわち本来の目的と してのジホモ ァ - リ ノ レン酸 メ チルエステル、 ァラキ ドン酸メ チルエステル、 エイ コサペンタエ ン酸メ チルエステルを単離するには、 カラムク ロマ ト グラフ ィ 一、 低温結晶化法、 尿素包接法、 液々向流分配ク 口マ ト グラフ ィ 一等を 単独で、 又は組み合わせて使用するこ とができる。
こ う して単離された各種高度不飽和脂肪酸メ チルから高度不飽和 脂肪酸を得るには、 アルカ リで加水分解した後、 エーテル、 酢酸ェ チル等の有機溶媒で抽出すればよい。 また、 高度不飽和脂肪酸をそのメ チルエステルを経ないで採取す るには、 前記の抽出脂質をアルカ リ分解 (例えば 5 %水酸化ナ ト リ ゥムにより室温にて 2 〜 3 時間) した後、 この分解液から、 脂肪酸 の抽出 ' 精製に常用されている方法により抽出 ' 精製するこ とがで きる。
本発明の高濃度の炭素源に耐性を有するモルティ エレラ ( Mor t i e r e l la ) 属に属する微生物を培養するこ とによって得られる脂質は 、 種々の動物用飼料又は食品などの製品に利用する こ とができる。 脂質を製品に利用するにあたっては、 培養菌体から採取した脂質ま たはそれを精製して得られる脂質を使用する こ とができるが、 該脂 質を菌体培養によって製造する途中の培養液若し く はその殺菌した 培養液、 または培養終了後の培養液若し く はその殺菌した培養液、
またはそれぞれから集菌した培養菌体若し く はその乾燥物、 または 培養液も し く は菌体から該油脂を採取した後の該油脂を含有する残 渣も使用するこ とができる。
本発明は、 本発明の微生物の菌体を配合した動物用飼料に関する 。 本発明の動物用飼料と しては、 ドッ グフー ドやキヤ ッ トフ一 ドな どのぺッ トフ一 ド、 鶏などの家禽のための飼料、 豚や牛などの家畜 のための飼料、 養魚用飼料などが挙げられる。 また、 脂質を産生、 蓄積した微生物の菌体は、 脂質が菌体内に保護されているため酸化 されにく く 、 また加熱殺菌にも安定であるため好ま し く 、 培養菌体 から脂質を採取した後の残渣も、 本発明の脂質の他に蛋白質や灰分 、 炭水化物などを含んでいるため好ま しい。 なお動物用飼料は、 本 発明の微生物菌体のほか、 他の原料と組み合わせて加工するこ と も できる。
さ らに、 本発明の微生物の菌体及びそれから得られる脂質は動物 用飼料添加物と しても使用できる。
さ らに、 脂質を産生、 蓄積した培養菌体または培養液を含んでな る微小餌料生物用餌料と して用いること もできる。 従来、 魚貝類や 甲殻類の養殖において、 種苗 (稚仔魚) 生産には、 微小餌料生物 ( シォ ミ ズッボヮムシ、 ブライ ンシュ リ ンプなどの動物プラ ンク ト ン ) が用いられており、 稚仔魚の養殖には先ずこれらの微小生物を養 殖する必要がある。 これらの微小生物を養殖する場合には、 後にそ れを餌料と して摂取する稚仔魚の栄養要求性を考えて微小餌料生物 に与える餌料が決められる。 脂質を含有する培養菌体または培養液 を微小餌料生物に与えるこ とにより、 ァラキ ドン酸、 ジホモー ア 一 リ ノ レン酸及び/又はエイ コサペンタエン酸を含有し、 稚仔魚の栄 養要求性を満足できる微小餌料生物が得られる。
この他、 上記の微小餌料生物を含有する魚介類用餌料とするこ と
もできる。
本発明で得られる脂質は、 ァラキ ドン酸、 ジホモ一 ア ー リ ノ レ ン 酸及び/又はエイ コサペンタエン酸を強化した家禽卵の生産に利用 する こ と、 並びにァラキ ドン酸、 ジホモ一 ア ー リ ノ レン酸及び 7又 はエイ コサペンタエン酸を強化した卵黄油の製造に利用する こ と も できる。 ァラキ ドン酸、 ジホモー ア リ ノ レ ン酸及び/又はエイ コ サペンタエン酸を強化した家禽卵は、 上述の動物飼料を採卵用家禽 、 特に鶏に与え飼育するこ とによって産生される。 また該家禽卵、 特に卵黄から常法に したがって油脂を抽出する こ とによって、 ァラ キ ドン酸、 ジホモー ア リ ノ レン酸及び Z又はエイ コサペンタエン 酸を強化した卵黄油が得られる。 またこの卵黄油を乳児用調製乳、 未熟児用調製乳、 幼児用食品、 妊産婦用食品に添加する こ と もでき る。
脂質、 好ま し く は ト リ グリ セ リ ド油を含有する乳児用調製乳、 未 熟児用調製乳、 幼児用食品、 妊産婦用食品、 老人用食品、 栄養補助 食品、 健康食品等の食品に本発明で得られる脂質を利用する こ と も できる。 本発明の食品は、 ァラキ ドン酸、 ジホモー ア — リ ノ レ ン酸 及び Z又はエイ コサペンタエン酸を補う こ とを目的と し、 健康維持 等に用いられる。 その形態は、 固形、 あるいは液状の食品ない しは 嗜好品のいずれであってもよい。
例えば油脂を含む食品と して肉、 魚、 ナッ ツ等の油脂を含む天然 食品、 中華料理、 ラーメ ン、 スープ等の調理時に油脂を加える食品 、 天ぶら、 フ ライ、 油揚げ、 チ ャ ーハ ン、 ドーナッ ツ、 かりん糖等 の熱媒体と して油脂を用いた食品、 バタ一、 マ一ガリ ン、 マヨネ一 ズ、 ドレ ッ シ ング、 チ ョ コ レー ト、 即席ラ ーメ ン、 キ ャ ラ メ ル、 ビ スケッ ト、 ク ッキ一、 ケーキ、 アイスク リ ーム等の油脂食品又は加 ェ時に油脂を加えた加工食品、 おかき、 ハー ドビスケッ ト、 あんパ
ン等の加工仕上げ時に油脂を噴霧又は塗布した食品等を挙げるこ ど ができるが、 油脂を含む食品に限定しているわけではな く 、 例えば ノ ン、 めん類、 ごはん、 菓子類 (キャ ンデー、 チューイ ンガム、 グ ミ 、 錠菓、 和菓子) 、 豆腐およびその加工品などの農産食品、 清酒 、 薬用酒、 みりん、 食酢、 醤油、 味噌などの発酵食品、 ヨーグル ト 、 ハム、 ベーコ ン、 ソ一セージなどの畜農食品、 かまぼこ、 揚げ天 、 はんぺんなどの水産食品、 果汁飲料、 清涼飲料、 スポーツ飲料、 アルコール飲料、 茶などの飲料等を挙げる こ とができる。
食品に用いる場合には、 所定量の脂質を、 食品原料とと もに配合 し、 一般の製造法により加工製造する こ とができる。 その配合量は 剤形、 食品の形態性状により異なるが、 一般には食品全量に対して
0 . 0 0 1 〜 5 0重量%が好ま しいが特に限定される ものではない 本発明で得られる脂質はまた、 脂質を含有する機能性食品とする こ と もできる。 機能性食品は、 ァラキ ドン酸、 ジホモー ア ー リ ノ レ ン酸及び/又はエイ コサペンタエン酸が有する生理活性機能を発揮 する こ とを目的と し、 機能低下した状態を健康状態に戻し維持する ための、 あるいは機能低下を予防するための食品である。
形態と しては散剤、 顆粒剤、 錠剤、 カプセル剤、 ト ローチ、 内用 液剤、 懸濁剤、 乳剤、 シロ ップ剤、 ドリ ンク剤、 経腸栄養剤等の形 態であっても、 また例えば蛋白質 (蛋白質源と してはア ミ ノ酸バラ ンスのとれた栄養価の高い乳蛋白質、 大豆蛋白質、 卵アルブミ ン等 の蛋白質が最も広く 使用されるが、 これらの分解物、 卵白のオ リ ゴ ペプチ ド、 大豆加水分解物等の他、 ア ミ ノ酸単体の混合物も使用さ れる) 、 糖類、 脂肪、 微量元素、 ビタ ミ ン類、 乳化剤、 香料等に本 発明の脂肪酸が配合された自然流動食、 半消化態栄養食および成分 栄養食等の形態、 あるいは前記飲食品の形態であってもよい。
機能性食品、 栄養補助食品は、 脂質を用いて、 散剤、 顆粒剤、 錠 剤、 カプセル剤、 ト ローチ、 内用液剤、 懸濁剤、 乳剤、 シ ロ ッ プ剤 、 ド リ ンク剤、 自然流動食、 半消化態栄養食、 成分栄養食、 経腸栄 養剤等の形態を有する飲食品と して製造する こ とができる。 この際 、 脂質と と もにいずれの栄養成分あるいは機能性成分を配合しても よい。 また医師の指示に基づ く 栄養士の管理下に、 病院給食の調理 の際に本発明で得られる脂質を加え、 その場で調整した食事を、 ァ ラ キ ド ン酸、 ジホモー ア 一 リ ノ レ ン酸及び/又はエイ コサペンタエ ン酸の供給を必要と している患者に与えるこ と もできる。 実施例
次に、 実施例によ り この発明を具体的に説明する。
実施例 1 . 高濃度の炭素源に対する耐性の評価
( 1 ) グルコース 1 %、 酵母エキス 0 . 5 %、 ( 2 ) グルコース 2 %、 酵母エキス 1 %、 ( 3 ) グルコース 3 %、 酵母エキス 1 . 5 %、 ( 4 ) グルコース 4 %、 酵母エキス 2 %、 ( 5 ) グルコース 5 %、 酵母エキス 2. 5 %、 ( 6 ) グルコース 8 %、 酵母エキス 1 . 6 %を含む培地 (pH6. 3 ) 1 0 mlを 5 0 mlエルレ ンマイヤ一フ ラ スコ にそれぞれ入れ、 1 2 0 °Cで 2 0分間殺菌した。
モルティ エレラ属 S AM 2 1 9 7株 ( F E RM B P— 6 2 6 1 ) を Czapek寒天培地 ( 0. 2 %NaN03 、 0. 1 %K2HP0" 0 . 0 5 % gS04 · H20 、 0. 0 5 %KC1 、 0 . 0 0 1 %FeS04 · 7H20、 3 %Sucrose 、 2 %寒天、 pH 6. 0 ) スラ ン ト に植菌し、 胞子形成ス ラ ン トを調製した。 胞子形成スラ ン ト に滅菌水 8 mlを人れ、 攪拌し た後、 胞子溶液を 1 0 0 pi 1 植菌した。
レシプロ シ ェーカー ( 1 1 0 rpm ) によ り 2 8 。Cで 6 日間培養 し た。 培養後、 濾過により菌体を回収し、 十分水洗した後、 1 0 5 °C
で 2 時間乾燥させ、 乾燥菌体重量を求め生育度 ( g Z L ) を算出 し た。 得られた乾燥菌体 2 0 mgに塩化メ チ レ ン 2 ml、 無水メ タ ノ ール 一塩酸 ( 1 0 2 mlを加え、 5 0 °Cで 3 時間処理する こ とによつ てメ チルエステル化し、 n—へキサン 4 ml、 水 l mlを加えて、 2 回 抽出 し溶剤を遠心エバポレーター ( 4 0 °C、 1 時間) で留去した後 、 得られた脂肪酸メ チルエステルをガスク ロマ ト グラフィ一で分析 した。 表 1 にその結果を示す。

初発グルコース濃度を高める程生育度が向上しており、 高濃度の 炭素源に対し耐性を有している こ とがわかり、 また初発グルコース 濃度が 4 %以上での、 ァラキ ドン酸の生産性の向上が確認された。
実施例 2 , 初発グルコース濃度を高めた場合のアサキ ド ン酸の 生産量の評価
( 1 ) グルコース 6 % 、 酵母エキス 2 % 、 大豆油 0 . 2 %及びァ デカノ 一ル 0 . 0 1 %を含む培地 ( pH 6. 3 ) ( 2 ) グルコース 8 % 、 酵母エキス 2 % 、 大豆油 0. 2 %及びアデカノ 一ル 0. 0 1 %を含む培地 (pH 6. 3 ) 、 ( 3 ) グルコース 1 1 % 、 酵母ェキ
ス 2 % 、 大豆油 0. 2 %及びアデ力ノ ール 0. 0 1 %を含む培地 ( pH 6 . 3 ) 5 L を 1 0 L ジ ャ 一フ ア ーメ ンターに入れ、 1 2 0 °C で 3 0 分間殺菌した。 モルティ エ レラ属 S A M 2 1 9 7株 ( F E R M B P - 6 2 6 1 ) の前培養液 1 0 0 mlを植菌し、 通気量 1 vvm 、 攪拌数 3 0 0 rpm 、 培養温度 2 8 °Cで、 8 日間の通気攪拌培養を 行った。
培養 2 日 目、 培養 3 日目、 培養 4 日目 に 2 . 0 %グルコ ースを添 加した。 培養終了後、 実施例 1 と同様の操作により、 培地 1 L当り
( 1 ) 4 2. 3 g、 ( 2 ) 6 0 . 5 g、 ( 3 ) 8 3. l gの乾燥菌 体を得た。 実施例 1 と同様に、 乾燥菌体 2 0 mgから加水分解、 メ チ ルエステル化、 及び抽出を行い、 得られた脂肪酸メ チルエステルを ガスク ロマ トグラフ ィ 一で分析した。 ァラキ ドン酸の全脂肪酸に対 する割合 (% ) は ( 1 ) 4 0. 7、 ( 2 ) 3 9. 4、 ( 3 ) 4 1 .
7 であり、 ァラキ ドン酸の生産量 (培地あたりのァラキ ドン酸量) は ( 1 ) 7 . 1 g / L . ( 2 ) 9. 8 g / L、 ( 3 ) 1 4. 3 g / Lであった。 初発グルコース濃度を高める程ァラキ ドン酸の生産量 は向上し、 初発グルコース濃度 1 1 %では 1 4. 3 g / Lにまで向 上した。
実施例 3. 実施例 4 および実施例 5 のためのコ ン ト ロール グルコース 6 %及び酵母エキス 2 %を含む培地 (pH 6. 0 ) 1 0 0 mlを 5 0 0 mlエルレンマイヤ一フラスコに入れ、 1 2 0 °Cで 2 0 分間殺菌した。 実施例 1 で用いたモルティ エレラ属 S AM 2 1 9 7 株 ( F E RM B P— 6 2 6 1 ) の胞子溶液 1 0 0 〃 1 を植菌し、 レシプロ シヱ一力一 ( 1 1 0 r pm ) により 2 8 °Cで 8 日間振盪培養 した。 培養後、 濾過により菌体を回収し、 十分水洗した後、 凍結乾 燥し、 乾燥菌体 3 1 2 O mgを得た。 この菌体より、 ク ロ口ホルム— メ タノ ール一水の一相系の溶媒を用いる Bligh & Dyerの抽出によつ
て油脂を抽出 し、 6 2 4 mgの油脂が得られた。
この油脂を無水メ タノ ール一塩酸 ( 1 0 %) を用いて 5 0 °Cにて 3 時間処理する こ とによってメ チルエステル化し.、 エーテルで抽出 して、 得られた脂肪酸メ チルの組成をガスク 口マ トグラフ ィ 一で分 析した結果、 パルミ チン酸メ チル 1 3 . 3 %、 ステア リ ン酸メ チル 6 . 1 %、 ォレイ ン酸メ チル 1 5 . 7 リ ノ ール酸メ チノレ 7 . 2 ァ 一 リ ノ レ ン酸メ チル 4 . 3 % . ジホモ一 ア ー リ ノ レ ン酸メ チ ル 4 . 7 %、 ァラキ ドン酸メ チル 4 2 . 1 %であり、 エイ コサペン タエン酸は検出されなかった。 これを培地当りの生産量に換算する と ジホモ一 ア ー リ ノ レン酸は 0 . 2 9 g / L、 ァラキ ドン酸は 2 . 6 3 g Z Lであった。
この混合物脂肪酸メ チルについて、 さ らにカラムク ロマ トグラフ ィ 一によって分離し、 ア ー リ ノ レン酸メ チル、 ジホモ一 ア ー リ ノ レ ン酸メ チル、 ァラキ ドン酸メ チル画分をそれぞれ分取し、 口一タ リ —エバポレーターによって溶剤を留去した結果、 精製されたア ー リ ノ レン酸メ チル、 ジホモ一 ア ー リ ノ レン酸メ チル、 ァラキ ドン酸メ チルをそれぞれ 2 4 . 1 m , 2 6 . 4 mg、 及び 2 3 6 . 4 mg 得た 実施例 4 . A 5不飽和化酵素阻害剤添加によるジホモー ア ー リ ノ レン酸の生産
グルコース 6 %及び酵母エキス 2 %及び胡麻油 2 %を含む培地 ( pH 6— . 0 ) 2 mlを 1 0 mlエルレンマイヤ一フラスコに入れ、 1 2 0 °Cで 2 0分間殺菌した。 実施例 1 で用いたモルティ エレラ属 S A M 2 1 9 7株 ( F E R M B P — 6 2 6 1 ) の胞子溶液 5 0 n 1 を植 菌し、 レシプロ シヱ一力一 ( 1 5 0 rpm ) により 2 8。Cで 8 日間振 盪培養した。 培養終了後、 実施例 1 と同様に濾過、 水洗、 乾燥、 加 水分解、 メ チルエステル化、 及び抽出を行い、 得られた脂肪酸メ チ
ルエステルをガスク ロマ トグラフ ィ ーで分析した。 その結果、 培地 あたり乾燥菌体が 3 1 . 2 g / L、 ジホモー ア リ ノ レ ン酸が 1 . 4 1 g / L . ァラキ ドン酸が 0. 8 7 g / Lの生産が認められた。 この結果から明らかなよう に、 Δ 5不飽和化酵素阻害作用を有する 胡麻油を培地に添加するこ とにより、 ァラキ ドン酸の生産が押さえ られ、 ジホモ- ァ - リ ノ レ ン酸が大量に生産された。
実施例 5 . 低温培養によるエイ コサペンタエン酸の生産
グルコース 6 %及び酵母エキス 2 %を含む培地 (pH 6. 0 ) 2 mlを 1 0 mlエルレ ンマイ ヤーフ ラ ス コ に入れ、 1 2 0 °Cで 2 0分間 殺菌した。 実施例 1 で用いたモルティ エレラ属 S AM 2 1 9 7株 ( F E RM B P— 6 2 6 1 ) の胞子溶液 5 0 n 1 を植菌し、 レ シプ ロ シヱ一カー ( 1 5 0 r pm ) により 2 8 °Cで 2 日間振盪培養した後 、 培養温度を 1 2 °Cに下げさ らに 6 日間培養した、 培養終了後、 実 施例 1 と同様に濾過、 水洗、 乾燥、 加水分解、 メ チルエステル化、 及び抽出を行い、 得られた脂肪酸メ チルエステルをガスク ロマ トグ ラ フ ィ 一で分析した。
その結果、 パル ミ チ ン酸メ チル 1 1 . 7 %、 ステア リ ン酸メ チル 5 . 8 %、 ォ レイ ン酸メ チル 1 2. 1 %、 リ ノ ール酸メ チノレ 6 . 4 %、 ア ー リ ノ レ ン酸メ チル 4. 7 %、 ジホモー ア 一 リ ノ レ ン酸メ チ ル 4. 9 %、 ァラキ ドン酸メ チル 3 8. 1 %、 エイ コサペンタエン 酸メ チル 6. 1 %であり、 エイ コサペンタエン酸が低温培養により 生産できるこ とが確かめられた。

Claims
1 . 高濃度の炭素源に耐性を有するモルティ エ レラ (Mortierel a_) 属に属する微生物を培養開始時の炭素源濃度が 4重量%以上の 培地中で培養して、 ァラキ ドン酸又はこれを含有する脂質を生成せ しめ、 そ してァラキ ドン酸を採取するこ とを特徴とするァラキ ドン 酸の製造方法。
2. 高濃度の炭素源に耐性を有するモルティ エ レラ (Mortierell a_) 属に属する微生物を培養開始時の炭素源濃度が 4重量%以上の 培地中で培養して、 ァラキ ドン酸を含有する脂質を生成せしめ、 そ してァラキ ド ン酸を含有する脂質を採取することを特徴とするァラ キ ドン酸を含有する脂質の製造方法。
3. 上記モルテ ィ エレラ (Mortierel la ) 属に属する微生物が、 モルティ エ レラ亜属 (Subgenus Mortierel la) に属する微生物であ る請求項 1又は 2記載の製造方法。
4. 上言己モノレティ エ レラ亜属 (Subgenus Mortierel la) に属する 微生物が、 ア リ アセァ (alliacea) 種に属する微生物である請求項 3記載の製造方法。
5. 上言己モノレティ エレラ亜属 (Subgenus Mortierel la) (こ属する 微生物が、 アルピナ (alpina) 種に属する微生物である請求項 3記 載の製造方法。
6—. 上言己モノレティ エレラ亜属 (Subgenus Mortierel la) (こ属する 微生物が、 モルティ エレラ属 S AM 2 1 9 7株 (F E RM B P— 6 2 6 1 ) である請求項 3記載の製造方法。
7. 培養開始時の炭素源濃度が 8重量%以上である こ とを特徴と する請求項 1 乃至 6のいずれかに記載のァラキ ドン酸又はこれを含 有する脂質の製造方法。
8. 高濃度の炭素源に耐性を有するモルティ エレラ (Mortierell a_) 属に属する微生物を、 培養開始時の炭素源濃度が 4重量%以上 であり且つ Δ 5不飽和化酵素阻害剤を含有する培地中で培養して、 ジホモー ア 一 リ ノ レン酸又はこれを含有する脂質を生成せしめ、 そ してジホモー ァ 一 リ ノ レ ン酸を採取するこ とを特徴とする ジホモー ア ー リ ノ レ ン酸の製造方法。
9. 高濃度の炭素源に耐性を有するモルティ エレラ (Mortierell 属に属する微生物を、 培養開始時の炭素源濃度が 4重量%以上 であり且つ Δ 5不飽和化酵素阻害剤を含有する培地中で培養して、 ジホモー ァ 一 リ ノ レ ン酸を含有する脂質を生成せしめ、 そ してジホ モー ア 一 リ ノ レ ン酸を含有する脂質を採取するこ とを特徴とする ジ ホモ一 ア ー リ ノ レ ン酸を含有する脂質の製造方法。
1 0. 上記モルティ エレラ (Mortierella ) 属に属する微生物が 、 モルテ ィ エ レラ亜属 (Subgenus Mortierella) に属する微生物で ある請求項 8又は 9記載の製造方法。
1 1 . 上 ΐ己モノレティ エレラ亜属 (Subgenus Mortierella) (こ属す る微生物が、 ア リ アセァ (alliacea) 種に属する微生物である請求 項 1 0記載の製造方法。
1 2. 上記モルティ エレラ亜属 (Subgenus Mortierella) に属す る微生物が、 アルピナ (alpina) 種に属する微生物である請求項 1 0記載の製造方法。
1 —3. 上言己モノレティ エレラ亜属 (Subgenus Mortierella) (こ属す る微生物が、 モルテ ィ エ レラ属 S AM 2 1 9 7株 ( F E RM B P - 6 2 6 1 ) である請求項 1 0記載の製造方法。
1 4. 培養開始時の炭素源濃度が 8重量%以上であるこ とを特徴 とする請求項 8乃至 1 3のいずれかに記載のジホモー ア 一 リ ノ レン 酸又はこれを含有する脂質の製造方法。
1 5. 高濃度の炭素源に耐性を有するモルティ エ レラ (Mortiere 1 ) 属に属する微生物を、 培養開始時の炭素源濃度が 4重量%以 上の培地中で 2 0 °C以下で培養して、 エイ コサペンタエン酸又はこ れを含有する脂質を生成せしめ、 そ してエイ コサペンタエン酸を採 取するこ とを特徴とするエイ コサペンタエン酸の製造方法。
1 6. 高濃度の炭素源に耐性を有するモルティ エ レラ (Mortiere 11a ) 属に属する微生物を、 培養開始時の炭素源濃度が 4重量%以 上の培地中で 2 0 °C以下で培養して、 エイ コサペンタエン酸を含有 する脂質を生成せしめ、 そ してエイ コサペンタエン酸を含有する脂 質を採取するこ とを特徴とするエイ コサペンタエン酸を含有する脂 質の製造方法。
1 7. 上記モルティ エ レラ (Mortierella ) 属に属する微生物が 、 モルティ エレラ亜属 (Subgenus Mortierella) に属する微生物で ある請求項 1 5又は 1 6記載の製造方法。
1 8. 上記モルティ エレラ亜属 (Subgenus Mortierella) に属す る微生物が、 ア リ アセァ (alliacea) 種に属する微生物である請求 項 1 7記載の製造方法。
1 9. 上記モルティ エレラ亜属 (Subgenus Mortierella) に属す る微生物が、 アルピナ (alpina) 種に属する微生物である請求項 1 7記載の製造方法。
2 0. 上記モルティ エレラ亜属 (Subgenus Mortierella) に属す る微—生物が、 モルティ エ レラ属 S AM 2 1 9 7株 ( F E RM B P - 6 2 6 1 ) である請求項 1 7記載の製造方法。
2 1 . 培養開始時の炭素源濃度が 8重量%以上であるこ とを特徴 とする請求項 1 5乃至 2 0のいずれかに記載のエイ コサペンタエン 酸又はこれを含有する脂質の製造方法。
