明 細 書 角膜障害治療剤
5 技術分野
本発明はラク トフエ リ ンおよび Zまたはラク トパーォキシ ダーゼを有効成分と して含む角膜障害治療剤に関する。
背景技術
- ラク トフヱ リ ンおよびラク トパーォキシダーゼはヒ トゃ牛 0 などの乳や涙液に含まれている蛋白質であり、 これらが抗菌 作用ゃリ ンパ球の増殖作用などの薬理作用を有することは知 られている (特開平 2 — 4 8 5 3 4号公報参照) 。
しかしながら、 これら化合物の眼科領域についての報告は ほとんどなされていない。
5 本発明者等はラク トフ.エ リ ンおよびラグトパーォキシダー ゼの新しい薬理作用を見つけ、 眼科領域へ応用するこ とを種 々検討した結果、 ごれらの化合物が優れた角膜実質細胞増殖 促進作用を有し、 角膜障害治療剤と して有用であることを見 い出した。
0 発明の開示
本発明は、 ラク トフヱ リ ンおよび Zまたはラク トパーォキ シダーゼを有効成分と して含む角膜障害治療剤を提供するも のである。
ラク ト フエ リ ンおよびラク トパ一ォキシダ一ゼは通常ヒ ト5 や牛などの動物の分泌物たとえば乳や涙液から得られる。 し
たがって、 これらは安全性の点では全く問題ない。
本発明者等はラク トフヱリ ンおよびラク トパ一ォキシダー ゼの新しい薬理作用を見出すべく研究し、 眼科領域について の応用を鋭意検討した結果、 これらの化合物が優れた角膜実 質細胞増殖促進作用を有し、 角膜障害治療剤となることを見 い出した。
ラグ トフエ リ ンおよびラク 卜パーォキシダーゼが角膜実質 細胞に対し増殖促進効果があるかどうかを調べるため、 i n' v i t r o でのゥサギ培養角膜実質細胞に対する作用と i n v i v o でのアルカ リバーン角膜炎に対する作用を調べた。
詳細なデータは薬理試験の項で示すが、 ラク トフユ リ ンぉ よびラク トパーォキシダーゼは有意に角膜実質細胞の増殖を 促進した。
この結果はラク トフエ リ ンおよびラク トパ一ォキシダ一ゼ が角膜潰瘍、 角膜炎、 眼手術等により引き起こされる各種の 角膜障害に対する治療剤と して有用なものであることを示し ている。
ラク トフエ リンおよび/またはラク トパーォキシダーゼは 経口投与、 非経口投与のいずれでも投与することができるが 点眼剤と して投与することが好ま しい。
本発明の角膜障害治療剤においては、 ラク トフ二 リ ンおよ びラク トパーォキシダーゼをそれぞれ単独に用いても、 また はこれらを併用してもよい。
ラク トフエ リ ンおよび/またはラク トパーォキシダーゼの 投与量は症状や年齢、 剤型等によって決められるが、 点眼剤
の例でいえば 0 . 0 1〜 3 . 0 % (重量 重量) の濃度が好 ま しい。
ラク トフエ リ ンおよび/またはラク トパーォキシダーゼの 製剤化は公知の方法を用いて行なわれる。 たとえば点眼剤は ラク トフエ リ ンおよびノまたはラク トバーオキシダ一ゼに必 要に応じて等張化剤、 緩衝剤、 防腐剤、 p H調整剤等の通常 用いられる添加剤を加えて調製すれば良い。
発明を実施するための最良の形態
以下に処方の例をいく つか示す。
処方例

薬理試験
ラク トフエ リ ンおよびラク トパーォキシダーゼが角膜実質
細胞に対し増殖促進効果があるかどうかを調べるため、 in vitro でのゥサギ培養角膜実質細胞に対する作用と vivo でのアル力 リバーン角膜炎に対する作用を調べた。
1) ゥサギ培養角膜実質細胞に対する作用
ゥサギ培養角膜実質細胞を用い、 3H—チ ミ ジンの細胞内 への取り込み量を指標と してラク トフエ リ ンおよびラク トパ 一ォキシダーゼの増殖促進効果を調べた。
(実験方法)
1 04 個のゥサギ角膜実質細胞を、 5容量%の牛胎仔血清 を含有させた T C - 1 9 9培養液 (GIBCD 社製) に浮遊させ これを 9 6ゥヱル平底カルチヤ一プレー ト中でラク トフヱ リ ンも しく はラク トパーォキシダーゼとともに炭酸ガスィ ンキ ュベータ一中で C O n 5 %で 3 7 °Cで 2 4時間培養した。 そ の後、 3H—チミ ジン (AMERSHAM社製) を添加し、 さ らに 3 7 °Cで 2 4時間培養を行なった後、 細胞中に取り込まれた
3H—チ ミ ジンの放射活性を液体シンチレ一シ ョ ンカウ ンタ 一を用いて測定した。
(結果)
得られた結果を表 1に示す。

表 1からわかるように、 ラク トフエ リ ンおよびラク トパー ォキシダーゼは濃度依存的に角膜実質細胞の増殖を有意に促 iiした o
2) アル力 リバー ン角膜炎に対する作用
i n v i v oでの効果を確認するため、 ゥサギでのアルカ リバ 一ン角膜炎に対するラク トフエ リ ンおよびラク トパーォキシ ダーゼの効果を調べた。
(実験方法)
1 Nの水酸化ナ ト リ ウム水溶液に浸した円形口紙 (直径 5 mm) を 1 . '5分間ゥサギの角膜上に置き、 角膜炎を惹起せし め、 生理食塩液にて洗浄を行なった。 角膜炎惹起直後から 1
時間間隔で 1 日 1 0回、 生理食塩液に溶解した 0 . 5重量% のラク トフヱ リ ンまたはラク トパーォキシダーゼ点眼剤を点 眼した。 1 3 日間連続して点眼した後、 角膜実質細胞の病理 組織学的検査を行なつた。
なお、 コ ン ドロールと して生理食塩液を点眼した。
(結果)
下記のスコァ表に従って評価を下した。 .

得られた結果を表 2に示した。 なお、 各々のスコアは 8眼 のスコアの合計で表した。

この結果は、 i n v i v o試験においてラク トフヱ リ ンまたは ラク トパーォキシダーゼが角膜実質細胞の再生を有意に促進 していることを明らかに示している。
産業上の利用可能性
本発明は、 ラク トフヱ リ ンおよび Zまたはラク トパーォキ
シダーゼを有効成分と して含む優れた角膜障害治療剤を提供 する ものである。
Claims
1. ラク トフエ リ ンおよびラク トパーォキシダ一ゼを有効成 分と して含む角膜障害治療剤。
2. ラク トフユ リ ンを有効成分として含む角膜障害治療剤。
3. ラク トパーォキシダーゼを有効成分と して含む角膜障害 治療剤。
4. 該有効成分を点眼剤の形態で含む請求の範囲 1〜 3の内 いずれか 1の角膜障害治療剤。 '
5. 該有効成分の濃度が 0. 0 1〜 3. 0 % (重量 Z重量) の範囲にある請求の範囲 4による角膜障害治療剤。
6. ラク トフエ リ ンおよびラク トパーォキシダーゼを有効成 分と して投与することからなる角膜障害治療方法。
7. ラク トフエ リ ンを有効成分と して投与することからなる 角膜障害治療方法。
8. ラク トパーォキシダーゼを有効成分と して投与すること からなる角膜障害治療方法。
9. 該有効成分を点眼投与する請求の範囲 6〜 8の内いずれ か 1の角膜障害治療方法。
1 0. ラク トフエ リ ンおよび/またはラク トパーォキシダー ゼの濃度が 0. 0 1〜 3. 0 % (重量 重量) の範囲にあ る点眼剤を投与することからなる角膜障害治療方法。
