Abstract

On obtient des polypeptides sur des ribosomes dans un système de translation exempt de cellules contenant un ARN matriciel et, à titre de substrats, ATP, GTP ainsi que des acides aminés. Le processus de translation est basé sur l'emploi d'ARN matriciel modifié de 15 à 75 % par un agent d'alkylation, au niveau de 2'-hydroxyles de restes de ribose. Les produits de translation contenus dans le système comprennent des polypeptides, AMP, GDP, de la pyrophosphate ainsi que de la phosphate non organique. La consommation en substrats pendant le fonctionnement du système exempt de cellules, est compensée par l'alimentation de ce dernier en ATP, GTP et acides aminés à titre de substrats, afin de maintenir la concentration initiale en substrats.
