Abstract
本發明之課題係提供可促進膽固醇代謝,且可治療、改善或預防伴隨血中之膽固醇增加的各式各樣的症狀之新穎藥劑。
解決手段為本發明者等新發現牛蒡苷元具有促進自膽固醇至膽汁酸的轉換的效果。本發明提供一種膽汁酸合成促進劑、膽汁酸合成促進用組成物及膽汁酸合成促進用食品組成物,其含有作為有效成分之牛蒡苷元及/或牛蒡苷。
Description
本發明係關於一種膽汁酸合成促進劑、膽汁酸合成促進用組成物及膽汁酸合成促進用食品組成物,其可於肝臟促進自膽固醇至膽汁酸的合成。
膽固醇係與活體之各式各樣生理現象有關的重要成分。然而,已知一旦血液內的膽固醇量成為過剩時,會引起例如動脈硬化症(arteriosclerosis)等之生活型態疾病(lifestyle disease)。膽固醇於血液內與蛋白質形成複合體,而呈脂蛋白(lipoprotein)而存在。此脂蛋白中,據說低密度脂蛋白膽固醇(LDL膽固醇(Low density lipoprotein cholesterol))會促進動脈硬化。因此,一般認為藉由使血液內的脂質減少,尤其是藉由使LDL膽固醇減少,可改善動脈硬化症。
膽固醇係於肝臟中被轉換成膽汁酸,並自腸排泄出。膽汁酸合成係用以將多餘膽固醇排出至體外的主要機制。一般認為藉由促進此種膽固醇向膽汁酸的轉換,可能治療、改善或預防由於血中膽固醇增加所產生的各式各樣的症狀。
膽固醇係於肝臟,藉由CYP7A1及CYP8B1等之膽汁酸合成酵素而被轉換成膽汁酸(非專利文獻1)。膽汁酸被分泌於腸道後,一部分被排泄。殘餘的膽汁酸係自腸被再吸收,並返回肝臟。膽汁酸係藉由肝臟與腸道之間進行循環而被回收。自膽固醇至膽汁酸的轉換係受到經再吸收而返回肝臟的膽汁酸控
制。返回肝臟的膽汁酸與為核內受體的FXR(類法尼醇X受體(farnesoid X receptor))結合而將其活性化。經活性化的FXR誘導為其他核內受體的SHP(小異源二聚體伴侣受體(small heterodimer partner))表現。SHP具有抑制Cyp7a1及Cyp8b1等之膽汁酸合成酵素基因表現的作用。又,FXR係於腸道促進FGF15(纖維母細胞生長因子15(Fibroblast growth factor 15))的表現。FGF15係藉由與肝臟之受體FGF4結合而活性化,而抑制為膽汁酸合成酵素基因的Cyp7a1及Cyp8b1的表現。
就用以促進脂質代謝的藥劑而言,例如於專利文獻1已揭示一種以磷蝦蛋白質(Krill Protein)作為有效成分之脂質代謝改善劑。專利文獻1記載之脂質代謝改善劑係藉由膽固醇吸收阻礙或膽汁酸排泄效果而使血中膽固醇降低。

本發明係以提供一種新穎藥劑為目的,該藥劑可促進膽固醇代謝,且可治療、改善或預防伴隨血中之膽固醇增加的各式各樣的症狀。
本發明者等發現,測量投予牛蒡苷元(arctigenin)的瘦素(leptin)受體缺損模型之糞中膽汁酸量的結果,與對照組比較,膽汁酸量增加。又,本發明者等發現,於牛蒡苷元投予組(AG組),與對照組比較,除了正向控制為膽汁酸合成酵素的Cyp8b1,同時負向控制具有抑制膽汁酸合成的作用,如肝臟之SHP、FXR及FGF4R、以及小腸之FGF15及SHP。即,本發明者等新發現牛蒡苷元具有促進自膽固醇至膽汁酸的轉換的效果,而完成本發明。
本發明提供一種膽汁酸合成促進劑,其含有作為有效成分之牛蒡苷元(arctigenin)及/或牛蒡苷(arctiin)。
又本發明提供一種膽汁酸合成促進用組成物,其含有作為有效成分之牛蒡苷元及/或牛蒡苷。
又本發明提供一種膽汁酸合成促進用組成物,其於上述膽汁酸合成促進用組成物,含有作為牛蒡(Arctium lappa)、牛蒡子(Arctii Fructus)、牛蒡芽(burdock sprout)或連翹(Forsythia)或此等之提取物之牛蒡苷元及/或牛蒡苷。
又本發明提供一種膽汁酸合成促進用食品組成物,其含有作為有效成分之牛蒡苷元及/或牛蒡苷。
又本發明提供一種膽汁酸合成促進用食品組成物,其於上述膽汁酸合成促進用食品組成物,含有作為牛蒡、牛蒡子、牛蒡芽或連翹或此等之提取物之牛蒡苷元及/或牛蒡苷。
又本發明提供一種膽汁酸合成酵素之表現促進劑,其含有作為有效成分之牛蒡苷元及/或牛蒡苷。
又本發明提供一種膽汁酸排出促進劑,其含有作為有效成分之牛蒡苷元及/或牛蒡苷。
又本發明提供一種肝臟中的膽固醇代謝促進劑,其含有作為有效成分之牛蒡苷元及/或牛蒡苷。
本發明因具有促進膽汁酸合成的作用,可促進膽固醇代謝,治療、改善或預防伴隨血中之膽固醇增加的各式各樣的症狀。

[用以實施發明之形態]
本發明提供一種膽汁酸合成促進劑及膽汁酸合成促進用組成物,其含有作為有效成分之牛蒡苷元及/或牛蒡苷。
膽汁酸係於肝臟內自膽固醇所合成之具有膽烷酸(cholanic acid)骨架類固醇衍生物的總稱。於本發明,膽汁酸包含例如膽酸(cholic acid)、鵝去氧膽酸(chenodeoxycholic acid)、去氧膽酸(deoxycholic acid)及石膽酸(lithocholic acid)等之非結合膽汁酸;牛膽酸(taurocholic acid)、甘膽酸(glycocholic acid)、甘胺鵝去氧膽酸(glycochenodeoxycholic acid)、牛磺鵝去氧膽酸(taurochenodeoxycholic acid)、甘胺去氧膽酸(glycodeoxycholic acid)、牛磺去氧膽酸(taurodeoxycholic acid)、甘胺石膽酸(glycolicolic acid)及牛磺石膽酸(taurolithocholic acid)等之結合膽汁酸;以及此等之鹽及共軛物(conjugate)。又,本發明中的膽汁酸不僅包含於人類發現的膽汁酸,亦包含於哺乳動物、鳥類、爬蟲類、兩生類及魚類等之膽汁中發現的各種膽汁酸。
於本說明書,「膽汁酸的合成」係指自肝臟中的膽固醇至膽汁酸的合成、轉換或代謝。於本說明書,「促進膽汁酸合成」係包含使膽汁酸的合成開始、加速、使加速及增加等。膽汁酸合成被促進係可藉由肝臟中的膽汁酸量的增加、肝臟中的膽固醇的減少、糞中的膽汁酸量的增加及膽汁酸合成酵素的表現量的增加等來決定。
本發明之膽汁酸合成促進劑及膽汁酸合成促進用組成物,其含有作為有效成分之牛蒡苷元及/或牛蒡苷。牛蒡苷元及牛蒡苷係牛蒡等之植物中所含的二苯基丙烷類(diphenylpropanoid)(木脂素(lignan)類)之一者。牛蒡苷為牛蒡苷元的前驅體,已知於活體內被代謝成為牛蒡苷元。作為牛蒡苷元及/或牛蒡苷,可使用化學合成的牛蒡苷元及/或牛蒡苷,亦可使用自植物單離的牛蒡苷元及/或牛蒡苷。又,作為牛蒡苷元及/或牛蒡苷,可使用包含牛蒡苷元及/或牛蒡苷的植
物本身或該植物之提取物。包含牛蒡苷元及/或牛蒡苷的植物係包含例如牛蒡(芽‧葉‧根莖‧牛蒡子)、金鐘連翹(Forsythia x intermedia)(花‧葉‧果實‧根莖)、朝鮮連翹(Forsythia koreana)(花‧葉‧果實‧根莖)、連翹(花‧葉‧果實‧根莖)、支那連翹(Forsythia viridissima)(花‧葉‧果實‧根莖)、紅花(safflower)、矢車菊(Centaurea cyanus)、翼薊(Cirsium vulgare)、聖薊(blessed thistle)(薊屬(Cnicus))、大薊(cardon)、大翅薊(Onopordum acanthium)、伊利里亞大翅薊(Onopordum illyricum)、芝麻、番仔藤(Ipomoea cairica)、金牛草(Polygala chinensis)、黃金絡石(Trachelospermum asiaticum)、亞洲絡石(T.asiaticum var.intermedium)、台灣絡石(Trachelospermum foetidum)、細梗絡石(Trachelospermum gracilipes)、白花藤(Trachelospermum jasminoides)、絡石藤(Trachelospermum jasminoides var.pubescens)、南嶺蕘花(Wikstroemia indica)、紅蓼(Persicaria orientalis)、山櫻(Cerasus jamssakura)、阿拉伯芥(Arabidopsis thaliana)、莧菜(Amaranth)、胡桃(Walnut)、燕麥(Avena sativa)、斯佩耳特小麥(Triticum spelta)、軟質小麥(soft wheat)、墨西哥柏木(Cupressus lusitanica)及日本極樹(Torreya nucifera)。其中,牛蒡(尤其牛蒡子及牛蒡芽)及連翹(尤其葉)因牛蒡苷元及/或牛蒡苷的含量高而較佳。使用植物本身的情形,可使用生的或乾燥切碎者、或使用乾燥呈粉末者。
使用植物之提取物作為牛蒡苷元及/或牛蒡苷的情形,提取物例如可藉由以下之方法自植物調製。於本發明所使用的提取物係例如可自含牛蒡苷元及/或牛蒡苷的植物,藉由酵素轉換步驟及利用有機溶媒的提取步驟的2階段進行提取。
酵素轉換步驟係利用內存於植物的酵素β-葡萄糖苷酶(β-glucosidase),將該植物所含的牛蒡苷酵素轉換成牛蒡苷元的步驟。具體而言,藉由將植物乾燥栽切者保持於適當溫度而使內在的β-葡萄糖苷酶作用,使進行自牛
蒡苷至牛蒡苷元的反應。例如,藉由於裁切的植物中添加水等之任意溶液,於30℃附近的溫度(20~50℃)之間攪拌等,可將植物保持於任意溫度。
利用有機溶媒的提取步驟係使用任意適當的有機溶媒,自植物提取牛蒡苷元及牛蒡苷的步驟。即,藉由利用上述酵素轉換步驟而牛蒡苷元成為高含量的狀態,添加適當溶媒,而自植物提取提取物(萃取)的步驟。例如,於植物中添加適當溶媒,適當時間加熱攪拌而提取提取物。又,加熱攪拌以外,亦可使用加熱回流、滴液式提取、浸漬式提取或加壓式提取法等之本項技術領域中具通常知識者公知之任意提取法,而提取提取物。
因牛蒡苷元為水難溶性,藉由添加有機溶媒,可提升牛蒡苷元的產率。有機溶媒係可使用任意之有機溶媒。例如,可使用甲醇、乙醇及丙醇等之醇,以及使用丙酮。考慮安全性之面向時,較佳使用30%量之乙醇作為有機溶媒。自提取物餾除溶媒時,獲得糊狀濃縮物,進一步將此濃縮物乾燥時,可獲得乾燥物。
本發明之膽汁酸合成促進劑及膽汁酸合成促進用組成物可為任意形態之製劑。膽汁酸合成促進劑及膽汁酸合成促進用組成物係可作為經口投予製劑,例如糖衣錠、口腔錠、包衣錠及咀嚼錠等之錠劑;菱形劑;丸劑;散劑;包含硬膠囊劑及軟膠囊劑的膠囊劑;顆粒劑;以及懸浮劑、乳劑、糖漿劑及酏劑等之液劑等。
又,本發明之膽汁酸合成促進劑及膽汁酸合成促進用組成物係可為靜脈注射、皮下注射、腹腔內注射、肌肉內注射、經皮投予、經鼻投予、經肺投予、經腸投予、口腔內投予及經黏膜投予等之非經口投予製劑。本發明之膽汁
酸合成促進劑及膽汁酸合成促進用組成物係可例如,注射劑、經皮吸收帶、噴霧劑及栓劑等。
又,本發明之膽汁酸合成促進劑及膽汁酸合成促進用組成物可呈外用劑被提供。本發明之外用劑可為醫藥品及化妝品等。本發明之外用劑可為用以適用於皮膚、頭皮、毛髪、黏膜及爪等的外用劑。外用劑包含例如霜劑、軟膏劑、液劑、凝膠劑、洗劑、乳液劑、噴霧劑、棒劑、片狀遮覆(sheet mask)劑、固形劑、泡沫劑、油劑及貼布劑等之塗布劑;泥敷劑(cataplasm)、硬膏劑(plaster)、帶劑及貼布劑等之貼附劑;以及噴霧劑等。
又,本發明之膽汁酸合成促進劑及膽汁酸合成促進用組成物可為適合食用的形態,例如固形狀、液狀、顆粒狀、粒狀、粉末狀、膠囊狀、霜狀及糊狀等。
本發明之膽汁酸合成促進用組成物可為用以使用於醫藥品、化妝品及食品等的組成物。本發明之膽汁酸合成促進用組成物可進一步含有醫藥品、化妝品及食品中通常使用的任意成分。例如,本發明之膽汁酸合成促進用組成物亦可進一步含有藥學上可容許的基劑、載體、賦形劑、結合劑、崩解劑、潤滑劑及著色劑等。
於膽汁酸合成促進用組成物使用的載體及賦形劑之例,包含乳糖、葡萄糖、白糖、甘露醇、糊精、馬鈴薯澱粉、玉米澱粉、碳酸鈣、磷酸鈣、硫酸鈣及結晶纖維素等。
結合劑之例包含澱粉、明膠、糖漿、黃蓍膠、聚乙烯醇、聚乙烯醚、聚乙烯吡咯啶酮、羥丙基纖維素、甲基纖維素、乙基纖維素及羧甲基纖維素等。
崩解劑之例包含澱粉、瓊脂、明膠粉末、結晶纖維素、碳酸鈣、碳酸氫鈉、海藻酸鈉、羧甲基纖維素鈉及羧甲基纖維素鈣等。
潤滑劑之例包含硬脂酸鎂、氫化植物油、滑石及聚乙二醇(macrogol)等。著色劑可使用被容許添加於醫藥品、化妝品及食品的任意著色劑。
又,膽汁酸合成促進用組成物,因應必要,可以白糖、明膠、純化蟲膠、明膠、甘油、山梨糖醇、乙基纖維素、羥丙基纖維素、羥丙基甲基纖維素、聚乙烯吡咯啶酮、鄰苯二甲酸纖維素乙酸酯、羥丙基甲基纖維素鄰苯二甲酸酯、甲基丙烯酸甲酯及甲基丙烯酸聚合物等以一層以上的層進行被膜。
又,膽汁酸合成促進用組成物,因應必要,可添加pH調節劑、緩衝劑、安定化劑、保存劑、防腐劑、稀釋劑、塗覆劑、甘味劑、香料及助溶劑等。
又,本發明提供含有本發明之膽汁酸合成促進用組成物的醫藥品。本發明之醫藥品可為用以促進自膽固醇至膽汁酸的合成的醫藥品。又,本發明之醫藥品可為用以治療、改善及預防與血中膽固醇增加有關的各種症狀的醫藥品,例如可為用以治療、改善及預防脂質異常症、動脈硬化、狹心症、心肌梗塞、中風、腦梗塞及腦出血之醫藥品。
又,本發明提供膽汁酸合成促進用食品組成物,其含有作為有效成分之牛蒡苷元及/或牛蒡苷。本發明之膽汁酸合成促進用食品組成物係可與上述膽汁酸合成促進劑及膽汁酸合成促進用組成物同樣地被構成。本發明之食品組成物可為用以改善或預防脂質異常症、動脈硬化、狹心症、心肌梗塞、中風、腦梗塞及腦出血等之疾病及狀態之食品組成物。
於本說明書,「食品組成物」不僅包含一般的食品及飲品,亦包含病患用食品、健康食品、機能性食品、特定保健用食品、營養補充食品及補充
品等。一般的食品及飲品例如包含各種飲料、各種食品、加工食品、液狀食品(湯等)、調味料、營養飲料及甜點類等。於本說明書,「加工食品」係指對天然食材(動物及植物等)施予加工及/或調理者,例如肉加工品、蔬菜加工品、果實加工品、冷凍食品、殺菌食品(retort food)、罐頭食品、瓶裝食品及即食食品(instant food)等。本發明之食品組成物可為附有促進膽汁酸合成的意旨或促進膽固醇代謝的意旨食品。又,本發明之食品組成物可以封入袋及容器等的形態被提供。於本發明,使用的袋及容器可為通常使用於食品的任意袋及容器。
本發明之膽汁酸合成促進劑、膽汁酸合成促進用組成物及膽汁酸合成促進用食品組成物中的牛蒡苷元及/或牛蒡苷的含量,只要可發揮促進膽汁酸合成的效果的量即可,可因應適用的對象、目的及投予方法(攝取方法)加以適當設定。例如使對人類經口攝取的情形,較佳可包含牛蒡苷元及/或牛蒡苷每1日的攝取量成為10~2000mg。
又,本發明提供膽汁酸合成酵素之表現促進劑,其含有作為有效成分之牛蒡苷元及/或牛蒡苷。膽汁酸合成酵素包含例如CYP7A1及CYP8B1等。牛蒡苷元係如於後述實施例所示,具有促進膽汁酸合成酵素的表現的作用。又,牛蒡苷元係具有抑制負向控制膽汁酸合成酵素的表現之作用,如SHP、FXR、FGF4R及FGF15等之各種因子的表現的作用。
於本說明書,基因的「促進表現」係意指例如使源自基因的轉錄產物的量增加及使源自基因的蛋白質的量增加等。又,於本說明書,「抑制表現」係意指例如使源自基因的轉錄產物的量減少及使源自基因的蛋白質的量減少等。
本發明之膽汁酸合成酵素的表現促進劑係可含有牛蒡、牛蒡子、牛蒡芽或連翹或此等之提取物作為牛蒡苷元及/或牛蒡苷。又,本發明之膽汁酸合成酵素的表現促進劑係可與上述本發明之膽汁酸合成促進劑、膽汁酸合成促進用組成物及膽汁酸合成促進用食品組成物同樣地被構成。
本發明之膽汁酸合成酵素的表現促進係可藉由促進膽汁酸合成酵素的表現,促進自膽固醇至膽汁酸的合成。因此,本發明之膽汁酸合成酵素的表現促進劑係可改善膽固醇代謝,治療、改善或預防伴隨血中膽固醇增加的各式各樣症狀。
又,本發明提供膽汁酸排出促進劑,其含有作為有效成分之牛蒡苷元及/或牛蒡苷。於本說明書,「膽汁酸排出」係指膽汁酸向體外的排出,例如膽汁酸自腸道被排泄至糞便中。
本發明之膽汁酸排出促進劑可含有牛蒡、牛蒡子、牛蒡芽或連翹或此等之提取物作為牛蒡苷元及/或牛蒡苷。又,本發明之膽汁酸排出促進劑可與上述本發明之膽汁酸合成促進劑、膽汁酸合成促進用組成物及膽汁酸合成促進用食品組成物同樣地被構成。本發明之膽汁酸排出促進劑可改善膽固醇代謝,治療、改善或預防伴隨血中之膽固醇增加的各式各樣症狀。
又,本發明提供肝臟中的膽固醇代謝促進劑,其含有作為有效成分之牛蒡苷元及/或牛蒡苷。於本說明書,「膽固醇代謝」係指將膽固醇變換為其他化合物,例如將膽固醇變換為膽汁酸。
本發明之肝臟中的膽固醇代謝促進劑可含有牛蒡、牛蒡子、牛蒡芽或連翹或此等之提取物作為牛蒡苷元及/或牛蒡苷。又,本發明之肝臟中的膽固醇代謝促進劑係可與上述本發明之膽汁酸合成促進劑、膽汁酸合成促進用組
成物及膽汁酸合成促進用食品組成物同樣地被構成。本發明之肝臟中的膽固醇代謝促進劑可改善膽固醇代謝,且可治療、改善或預防伴隨血中之膽固醇增加的各式各樣的症狀。
[實施例]
以下呈示實施例,進一步詳細說明本發明之實施形態,但本發明並未被限定於以下實施例。
(酵素活性之測定)
牛蒡子中之β-葡萄糖苷酶活性係以下列方法測定。利用威利氏粉碎機(Willey mill)將產地、批次相異的牛蒡子粉碎,以10mL的水稀釋此牛蒡子粉碎品0.1g,作為試料溶液。
作為基質溶液,於p-硝基苯基-β-D-哌喃糖苷(p-nitorphenyl-β-D-glucopyranoside)0.15g中添加水,定量為25mL,而調製20mmol/Lp-硝基苯基-β-D-哌喃糖苷水溶液。於0.1mol/L乙酸緩衝液1mL中添加20mmol/L p-硝基苯基-β-D-哌喃糖苷水溶液0.5mL,調製反應混液,於37℃進行約5分鐘預備加熱。
於反應混液中添加試料溶液0.5mL,於37℃使反應15分鐘後,添加2mL為反應停止液的0.2mol/L碳酸鈉水溶液而使反應停止。測定此液之400nm中的吸光度,自無酵素反應進行的空白溶液的變化量,利用下式求得酵素活性。
酵素活性(U/g)=(試料溶液之吸光度-空白溶液之吸光度)×4mL×1/18.1(p-硝基酚之上述測定條件下的毫莫耳分子吸光係數:cm2/μmol)×1/光路長(cm)×1/反應時間(分)×1/0.5mL×1/試料溶液濃度(g/mL)。
(實施例1 牛蒡子提取物之製造1)
作為本發明之膽汁酸合成促進用組成物之一實施例,自牛蒡子提取萃取物(提取物)。將牛蒡子(酵素活性8.23U/g)裁切,將通過9.5mm篩者進一步通過0.85mm的篩子,確認有75%殘留。將此牛蒡子細切80kg添加於保溫於29~33℃的水560L而攪拌30分鐘。接著,添加乙醇265L而升溫至85℃,進一步加熱回流60分鐘。將此溶液離心分離,獲得牛蒡子提取液。將此操作重複2次,合併獲得的提取液,減壓濃縮,相對於提取物固體含量,添加糊精25%量,進行噴霧乾燥。牛蒡苷元及牛蒡苷含量係各自為6.2%及7.1%,獲得牛蒡苷元/牛蒡苷(重量比)=0.89之牛蒡子提取物粉末(含有糊精20%)。
(實施例2 牛蒡子提取物之製造2)
自牛蒡子提取萃取物(提取物)作為本發明之膽汁酸合成促進用組成物之一實施例。將牛蒡子(酵素活性8.23U/g)裁切,將通過9.5mm的篩子者進一步通過0.85mm的篩子,確認有75%殘留。將此牛蒡子細切80kg添加至保溫於30~33℃的水560L,攪拌30分鐘後,添加乙醇265L並升溫至85℃,進一步加熱回流30分鐘。將此溶液離心分離,獲得牛蒡子提取液。合併重複此操作2次而獲得的提取液,減壓濃縮,相對於提取物固體含量,添加糊精25%量,噴霧乾燥。牛蒡苷元及牛蒡苷含量係各自為6.0%及6.8%,獲得牛蒡苷元/牛蒡苷(重量比)=0.87之牛蒡子提取物粉末(含有糊精20%)。
(實施例3 牛蒡子提取物之製造3)
自牛蒡子提取萃取物(提取物)作為本發明之膽汁酸合成促進用組成物之一實施例。將通過9.5mm的篩子者進一步通過0.85mm的篩子,確認有75%殘留。將此牛蒡子細切80kg添加至保溫於30~32℃的水560L,攪拌40分鐘後,於60分鐘後添加乙醇258L並升溫至85℃,進一步加熱回流30分鐘。將此液離心分離,獲得牛蒡子提取液。合併重複此操作2次而獲得的提取液,減壓濃縮,相對於提取物固體含量,添加糊精25%量,噴霧乾燥。牛蒡苷元及牛蒡苷含量係各自為6.2%及6.7%,獲得牛蒡苷元/牛蒡苷(重量比)=0.93之牛蒡子提取物粉末(含有糊精20%)。
(實施例4 牛蒡子提取物之製造4)
自牛蒡子提取萃取物(提取物)作為本發明之膽汁酸合成促進用組成物之一實施例。將牛蒡子(酵素活性7.82U/g)裁切,將通過9.5mm的篩子者進一步通過0.85mm的篩子,確認有75%殘留。將此牛蒡子細切80kg添加至保溫於30~32℃的水560L,攪拌30分鐘後,添加乙醇253L並升溫至85℃,進一步加熱回流40分鐘。將此液離心分離,得到獲得的提取液。合併重複此操作2次而獲得的提取液,減壓濃縮,相對於提取物固體含量,添加糊精25%量,噴霧乾燥。牛蒡苷元及牛蒡苷含量係各自為6.4%及7.2%,獲得牛蒡苷元/牛蒡苷(重量比)=0.89之牛蒡子提取物粉末(含有糊精20%)。
(實施例5 支那連翹葉提取物之製造1)
自支那連翹葉提取萃取物(提取物)作為本發明之膽汁酸合成促進用組成物之一實施例。於含有牛蒡苷2.53%及牛蒡苷元0.76%的連翹葉切碎50g中添加水350mL,於37℃保溫30分鐘後,添加乙醇150mL並加熱提取30分鐘。將此溶液使
用100網孔篩進行固液分離,藉由進行冷凍乾燥,獲得牛蒡苷元含量為5.62%之支那連翹葉提取物18.62g。
(實施例6 支那連翹葉提取物之製造2)
自支那連翹葉提取萃取物(提取物)作為本發明之膽汁酸合成促進用組成物之一實施例。於含有牛蒡苷7.38%及牛蒡苷元0.78%的連翹葉切碎720g中添加水5L,於37℃保溫30分鐘後,添加乙醇2.16L並加熱提取30分鐘。將此溶液使用100網孔篩進行固液分離,藉由進行冷凍乾燥,獲得牛蒡苷元含量為9.55%之支那連翹葉提取物343.07g。
(實施例7 牛蒡子提取物粉末摻合顆粒劑)
使用牛蒡子提取物製造顆粒劑作為本發明之膽汁酸合成促進用組成物之一實施例。按照「日本藥典」製劑總則、顆粒劑的項目,製造顆粒劑。即,取下述(1)~(3)之成分,製成顆粒狀。將其各1.5g填充於鋁層壓膜,獲得每1包含有0.5g牛蒡子提取物粉末的顆粒劑。
(實施例8 牛蒡子提取物粉末摻合顆粒劑)
使用牛蒡子提取物製造顆粒劑作為本發明之膽汁酸合成促進用組成物之一實施例。按照「日本藥典」製劑總則、顆粒劑的項目,製造顆粒劑。即,取下述(1)~(3)之成分,製成顆粒狀。將其各3.0g填充於鋁層壓膜,獲得每1包含有2g牛蒡子提取物粉末的顆粒劑。
(實施例9 牛蒡子提取物粉末摻合錠劑)
使用牛蒡子提取物製造錠劑作為本發明之膽汁酸合成促進用組成物之一實施例。按照「日局」製劑總則、錠劑的項目,製作錠劑。即,取下述(1)~(6)之成分,獲得錠劑。
(實施例10 牛蒡苷元對膽汁酸合成的效果)
〔實驗動物〕
使用為瘦素受體欠損模型的5週齡雄性db/db小鼠(BKS.Cg-+Leprdb/+Leprdb/Jcl,購自日本CLEA公司),1週的預備飼育後開始實驗。動物於溫度23.0±5℃、濕度50.0±10%、12小時的明暗週期的環境下飼育。動物實驗獲得慶應義塾大學動物實驗委員會的承認,基於慶應義塾大學動物實驗規定來實施。
〔實驗方法〕
預備飼育後,將小鼠分成(a)對照組及(b)牛蒡苷元投予組(AG組)之2組(各組n=8),對照組使於9週自由攝取精製飼料(AIN-93G),於AG組使於9週自由攝取牛蒡苷元(>98.0%)0.2%摻合的精製飼料。投予結束後,進行18小時的斷食,於麻醉下自腹部大靜脈採血,採取肝臟及小腸。
〔糞中膽汁酸量之測定〕
投予期間結束時,自各組的飼育籠回收糞便,測定糞中膽汁酸量。將聚集的糞便乾燥粉末10mg懸浮於90%乙醇0.2mL,於65℃培育1小時,合併離心分離後的上清液及沉澱物的洗液而回收。將溶媒蒸發乾固後於殘渣中添加90%乙醇0.5mL而溶解者作為測定用樣品。於測定使用超效能液相層析串聯質譜儀(Ultra-performance liquid chromatography tandem mass spectrometry(UPLC-MS/MS、Waters公司製))。圖1為呈示對照組及AG組中的糞中膽汁酸量(μmol/L)的圖表。AG組與對照組比較,糞便中的總膽汁酸量較多。
〔肝臟中總膽固醇之測定〕
將自各組獲得的肝臟均質化,使用脂質提取套組(Lipid Extraction Kit(Chloroform-Free)、Cell Biolabs,Inc.),而提取總脂質。自脂質提取液,使用膽固醇E-Test Wako(和光純藥)而測定總膽固醇值。圖2係呈示對照組及AG組中的肝臟中總膽固醇量(mg/dL)的圖表。AG組與對照組比較,肝臟中的總膽固醇量較少。
〔肝臟膽固醇代謝關連基因的表現量〕
於各組,自獲得的臟器進行總RNA的提取及cDNA合成後,藉由qPCR,比較為肝臟膽固醇代謝關連基因的肝臟之Cyp8b1、SHP、FXR及FGF4R、以及小腸的FGF15及SHP的表現量。使用18S rRNA作為對照組。qPCR用的引子係使用18S(F:TTCTGGCCAACGGTCTAGACAAC(序列識別號1)、R:CCAGTGGTCTTGGTGTGCTGA(序列識別號2))、Cyp8b1(F:GGAAGCCAAGAAGTCGTTCA(序列識別號3)、R:GACGCAGACTCTCCTCCATC(序列識別號4))、SHP(F:
CAAGGAGTATGCGTACCTGAAG(序列識別號5)、R:GGCTCCAAGACTTCACACAGT(序列識別號6))、FXR(F:CAAAATGACTCAGGAGGAGTACG(序列識別號7)、R:GCCTCTCTGTCCTTGATGTATTG(序列識別號8))、FGF4R(F:GAGGCATGCAGTATCTGG(序列識別號9)、R:CAAAGTCAGCGATCTTCATCACA(序列識別號10))、FGF15(F:CCAGTGGTCTTGGTGTGCTGA(序列識別號11)、R:GGCCTGGATGAAGATGATATGGA(序列識別號12))。
圖3係呈示對照組及AG組中的肝臟之Cyp8b1、SHP、FXR及FGF4R的相對mRNA表現量。圖3中的值係表示6隻的平均±標準誤差(Mean±S.E.M)。司徒頓t檢驗(Student’s t-test)的結果,p值較0.05小的情形,附加「*」,較0.01小的情形,附加「**」。AG組,與對照組比較,Cyp8b1的表現量顯著地高,SHP、FXR及FGF4R的表現量顯著地低。
圖4係呈示對照組及AG組中的小腸之FGF15及SHP的相對mRNA表現量。AG組,與對照組比較,小腸之FGF15及SHP的表現量為低。
由此等之結果,牛蒡苷元,促進肝臟的Cyp8b1的表現,另一方面,呈現抑制肝臟的SHP、FXR及FGF4R、以及小腸的FGF15及SHP的表現。據此,強烈暗示牛蒡苷元具有促進肝臟中的膽固醇代謝、促進膽汁酸的合成及排出的作用。
[產業上之利用可能性]
本發明因藉由促進膽汁酸合成,可促進膽固醇的代謝,可適合利用於用以治療、改善或預防伴隨膽固醇增加的各種症狀的醫藥及食品等。
Claims (8)
- 一種牛蒡苷元(arctigenin)及/或牛蒡苷(arctiin)用於製造藥劑之用途,前述藥劑係經由促進膽汁酸合成,用以治療、改善及/或預防選自由狹心症、腦梗塞及腦出血所組成之群的疾病。
- 如請求項1記載之用途,其中,前述牛蒡苷元及/或牛蒡苷係藉由以下至少一者來促進膽汁酸的合成:促進肝臟的Cyp8b1的表現;抑制肝臟的SHP、FXR及/或FGF4R的表現;以及抑制小腸的FGF15及/或SHP的表現。
- 如請求項1或2記載之用途,其中,前述牛蒡苷元及/或牛蒡苷係來自牛蒡(Arctium lappa)、牛蒡子(Arctii Fructus)、牛蒡芽(burdock sprout)或連翹(Forsythia)或上述四者之提取物。
- 一種牛蒡苷元及/或牛蒡苷用於製造食品組成物之用途,前述食品組成物係經由促進膽汁酸合成,用以改善及/或預防選自由狹心症、腦梗塞及腦出血所組成之群的疾病。
- 如請求項4記載之用途,其中,前述牛蒡苷元及/或牛蒡苷係藉由以下至少一者來促進膽汁酸的合成:促進肝臟的Cyp8b1的表現;抑制肝臟的SHP、FXR及/或FGF4R的表現;以及抑制小腸的FGF15及/或SHP的表現。
