隨著醫學的進步,由細菌或病毒所造成的疾病對人類的威脅逐漸下降,使得人類的平均壽命不斷的延長。根據政府的統計資料,台灣人的平均年齡已突破80歲。
然而,隨著人類的平均壽命延長,各種因人體老化而發生的疾病如老年痴呆症、心肌肥大等也開始出現在老年人身上,並影響老年人的日常生活。再者,隨著年齡增長,人的膚質逐漸變差,皺紋也開始出現,對於愛美人士而言也是個大問題。因此,如何延緩人體老化的速度已逐漸成為現代人所重視的問題。
本發明係提供一種餘甘子萃取物用於製備提升粒線體功能以對抗老化之組合物的用途,藉此增加BHI(Bioenergetic Healthy Index),進而延緩人體老化的速度。
本發明揭露一種餘甘子萃取物用於製備提升粒線體功能以對抗老化之組合物的用途,包含食用一組合物以增加BHI(Bioenergetic Healthy Index)。組合物包含一餘甘子萃取物、一溶劑及一胎盤粉,該溶劑為琉璃苣
油,該胎盤粉為羊胎盤粉,該餘甘子萃取物的有效劑量為50毫克(mg)至100毫克(mg),該溶劑的有效劑量為300毫克(mg)至600毫克(mg),該胎盤粉的有效劑量為150毫克(mg)至300毫克(mg),且該餘甘子萃取物、該溶劑與該胎盤粉的有效劑量比為0.8-1.2:4.8-7.2:2.4-3.6。
根據上述本發明所揭露的餘甘子萃取物用於製備提升粒線體功能以對抗老化之組合物的用途,食用餘甘子萃取物可增加血液中的GDF11濃度。如此一來,人體細胞在GDF11的調節下,人體老化的速度得到減緩。
此外,食用餘甘子萃取物亦可增加血液中的脫氫異雄固酮(DHEA-S)濃度。如此一來,人體細胞在GDF11及DHEA-S的共同調節下,人體老化的速度進一步得到減緩。
以上之關於本揭露內容之說明及以下之實施方式之說明係用以示範與解釋本發明之精神與原理,並且提供本發明之專利申請範圍更進一步之解釋。

內容足以使任何熟習相關技藝者了解本發明之技術內容並據以實施,且根據本說明書所揭露之內容、申請專利範圍及圖式,任何熟習相關技藝者可輕易地理解本發明相關之目的及優點。以下之實施例係進一步詳細說明本發明之觀點,但非以任何觀點限制本發明之範疇。
餘甘子(例如Phyllanthus Emblica或Emblica Officinale),又稱餘柚子、油柑、庵摩勒(Amalaka)、馬六甲樹(Pokok Melaka)、印度醋栗(Indian Gooseberry),屬於大戟科餘甘子屬(Emblica)之落葉亞喬木,分佈於自印度至馬來西亞地區及中國南部,一般認為印度為原產地。
本發明使用之餘甘子萃取物之取得方式例如以二氧化碳作為超臨界流體萃取餘甘子果實,或者是以甲醇、乙醇、丙酮、乙酸乙酯、重量百分濃度0.1至5%的氯化鈉水溶液、氯化鉀水溶液、氯化鈣水溶液、氯化鎂水溶液或重量百分濃度0.1至5%的氯化鈉乙醇溶液、氯化鉀乙醇溶液、氯化鈣乙醇溶液、氯化鎂乙醇溶液作為溶劑萃取餘甘子果實而得到一初萃液。接著,將初萃液過濾純化後得到本發明所使用的餘甘子萃取物。餘甘子萃取物可使用噴霧乾燥(Spray dry)或真空乾燥進行乾燥程序而得到易於保存的餘甘子萃取物粉末。
本發明實施例使用的餘甘子萃取物含有重量百分比35%至55%之Emblicanin-A與Emblicanin-B混合物、重量百分比4%至15%之Ponigluconin、重量百分比10%至20%之Pedunculagin、重量百分比5%至15%之Rutin與重量百分比10%至30%之Gallo-ellagitannoids。
Emblicanin-A(2,3-di-O-galloyl-4,6-(S)-hexahydroxy diphenoyl-2-keto-glucono-lactone)之結構如式一所示。
Emblicanin-B(2,3,4,6-bis-(S)-hexahydroxydiphenoyl-2-keto-glucono-lactone)之結構如式二所示。
Ponigluconin(2,3-di-O-galloyl-4,6-(S)-hexahydroxyd iphenoyl gluconic acid)之結構如式三所示。
Pedunculagin(2,3,4,6-bis-(S)-hexahydroxydiphenoyl-D-glucose)之結構如式四所示。
Rutin(3’,4’,5,7-tctrahydroxyflavono-1,3-O-rhamnogl ucoside)之結構如式五所示。
本發明實施例之實驗使用的餘甘子萃取物係將餘甘子(Emblica Officinalis)的果實浸於重量百分濃度1%之氯化鈉水溶液,再以65℃至75℃的蒸汽浴(Steam bath)加熱一小時,接著進行過濾,接著再冷凍3天,接著透過噴霧乾燥(Spray dry)程序得到餘甘子萃取物粉末。乾燥後的餘甘子萃取物型態為棕色粉末(Brown powder)。餘甘子萃取物含有重量百分比27%之Emblicanin-A、重量百分比23%之Emblicanin-B、重量百分比8%之Ponigluconin、重量百分比14%之Pedunculagin、重量百分比10%之Rutin與重量百分比10%至30%之Gallo-ellagitannoids。本發明之餘甘子萃取物不以Emblica Officinalis之萃取物為限,其他具有不同學名但具有相似成分之餘甘子之萃取物也具有相同之效果。本發明之餘甘子萃取物不以氯化鈉水溶液進行萃取為限,其他以水或其他溶劑進行萃取但具有相似成分之餘甘子之萃取物也具有相同之效果。
圖1為本發明實施例使用的餘甘子萃取物之紅外線吸收光譜。圖2為本發明實施例使用的餘甘子萃取物之高效能液相層析圖譜。如圖1所示,本發明實施例使用的餘甘子萃取物之紅外線吸收光譜於3403.6公分-1(cm-1)、2931.6公分-1(cm-1)、1385.0公分-1(cm-1)、1318.6公分-1(cm-1)、1623.5公分-1(cm-1)、1451.3公分-1(cm-1)、1352.1公分-1(cm-1)、1218.4公分-1(cm-1)、1148.6公分-1(cm-1)、1035.7公分-1(cm-1)、3403.6公分-1(cm-1)具有特徵吸收峰。如圖2所示,本發明實施例使用的餘甘子萃取物之高效能液相層析圖譜(HPLC Chromatogram)於1.620分鐘、2.148
分鐘、3.265分鐘與4.370分鐘具有特徵峰信號。圖1與圖2所示的餘甘子萃取物為本發明實施例使用的餘甘子萃取物,其僅用於說明本發明實施例的實驗結果,並非用以限制本發明。本發明其他實施例中,餘甘子萃取物之紅外線吸收光譜可於3403.6±5公分-1(cm-1)、2931.6±5公分-1(cm-1)、1385.0±5公分-1(cm-1)、1318.6±5公分-1(cm-1)、1623.5±5公分-1(cm-1)、1451.3±5公分-1(cm-1)、1352.1±5公分-1(cm-1)、1218.4±5公分-1(cm-1)、1148.6±5公分-1(cm-1)、1035.7±5公分-1(cm-1)、3403.6±5公分-1(cm-1)具有特徵吸收峰;餘甘子萃取物之高效能液相層析圖譜(HPLC Chromatogram)可於1.620±0.5分鐘、2.148±0.5分鐘、3.265±0.5分鐘與4.370±0.5分鐘具有特徵峰信號。
本發明的實施例揭露一種提升粒線體功能以對抗老化的方法,包含食用一組合物以增加血液中的GDF11(Growth Differentiation Factor 11)濃度。組合物包含一餘甘子萃取物。餘甘子萃取物具有增加血液中的GDF11濃度之效果,藉此達成延緩人體老化的功效。再者,餘甘子萃取物更具有增加BHI(Bioenergetic Healthy Index)之效果,藉此達成延緩人體老化的功效。此外,餘甘子萃取物更具有增加血液中的脫氫異雄固酮(Dehydroepiandrosterone Sulfate,DHEA-S)濃度之效果,藉此達成延緩人體老化的功效。
GDF11(Growth Differentiation Factor 11)是人體內一種特殊蛋白質,其具有調節胚胎分化的功能。根據2014年的文獻,年輕動物血液中的GDF11具有部份逆轉年長動物的大腦、心臟與肌肉老年化的功能,因此GDF11被認為具有抗老化之效果。
BHI(Bioenergetic Healthy Index)為使用海馬生物能量儀(Seahorse Bioscience XFe Analyzer)偵測細胞中粒線體的能量代謝數據,並以粒線體的能量代謝數據作為參數所計算出的能量代謝評估指標。BHI=[粒線體用於產生ATP的耗氧量×粒線體的潛力]/[粒線體膜的
氫離子洩漏量×非粒線體的耗氧量]。細胞的BHI越高代表的是細胞中粒線體的功能越強,而功能越強的粒線體通常是越年輕的粒線體。因此,BHI被用作評估粒線體老化程度的指標。
脫氫異雄固酮(DHEA)是由腎上腺所分泌的賀爾蒙。由於DHEA是很多不同的賀爾蒙的上游物質,其可被轉化為雄性賀爾蒙、雌性激素、黃體素及腎上腺皮質素,使得DHEA又被稱為「荷爾蒙之母」。在人體內,DHEA通常以硫化物(DHEA-S)的形式穩定存在。血液中的DHEA-S濃度在人的年齡大約達到30歲後開始隨著年齡下降,因此DHEA-S被用作評估老化程度的指標之一。由於DHEA可保護大腦以預防老年失智,因此DHEA也被認為具有抗老化之效果。
為方便以食用的方式由口攝取包含餘甘子萃取物的組合物,組合物可製成例如液體狀、糊狀或凝膠狀的組合物加工品。組合物加工品中可搭配作為添加劑的賦形劑或呈味劑,以提升風味與方便食用。
賦形劑例如為小麥澱粉、米澱粉、玉米澱粉、馬鈴薯澱粉、糊精、環糊精等澱粉類;結晶纖維素類;乳糖、葡萄糖、砂糖、還原麥芽糖、飴糖、果寡糖、乳化寡糖等糖類;山梨糖醇、赤藻糖醇、木糖醇、乳糖醇、甘露醇等糖醇類。
呈味劑例如為龍眼萃取物、荔枝萃取物、柚子萃取物等各種果汁萃取物;蘋果汁、橘子汁、檸檬汁等各種果汁;桃子香料、梅子香料、酸乳酪香料等各種香料;乙醯磺胺酸鉀、蔗糖素、赤藻糖醇、寡糖類、甘露糖、木糖醇、異構化糖類等各種甜味劑;檸檬酸、蘋果酸、酒石酸、葡萄糖酸等各種酸味劑;綠茶、烏龍茶、巴拿巴茶(Banaba Tea)、杜仲茶、鐵觀音茶、薏苡茶、七葉膽茶、茭白茶、昆布茶等各種茶成分;阿拉比卡(Coffee Arabica)、羅布斯塔(Coffee Robusta)、賴比瑞亞(Coffee Liberica)等各種咖啡成分等。
再者,組合物亦可包覆於膠囊中以方便由口攝取餘甘子萃取物。例如,組合物是以溶液狀、懸浮液狀、凝膠狀或糊狀的形式被包覆於軟膠囊中。
軟膠囊中用於溶解或分散餘甘子萃取物之油脂類例如為琉璃苣油、萼梨油、杏仁油、亞麻仁油、小茴香油、白蘇油、橄欖油、橄欖角鯊烯、甜橙油、胸棘鯛油(Orange Roughy oil)、芝麻油、蒜油、可可脂、南瓜子油、洋甘菊油、胡蘿蔔油、胡瓜油、牛油脂肪酸、夏威夷核果油、越橘子油、糙米胚芽油、大米油、小麥胚芽油、紅花油、牛油樹油脂、液狀牛油樹油脂、紫蘇油、大豆油、月見草油、山茶油、玉米油、菜子油、鋸葉棕萃取油(Saw Palmetto Extract Oil)、薏苡油、桃仁油、洋芹子油、蓖麻油、葵花子油、葡萄子油、澳洲胡桃油、繡線菊油(Meadowfoam oil)、棉子油、花生油、龜油、貂油、蛋黃油、魚油、棕櫚油、棕櫚仁油、木蠟、椰子油、長鏈/中鏈/短鏈之脂肪酸三甘油酯、二酸甘油酯、牛油、豬油、角鯊烯、角鯊烷、姥鮫烷、以及該等油脂類之氫化物等。
此外,著色劑、防腐劑、增黏劑、結合劑、崩解劑、分散劑、穩定劑、膠化劑、抗氧化劑、界面活性劑、防腐劑、pH值調整劑等符合政府單位規定之添加物,亦可依照政府單位規定之劑量標準與加工生產之需求添加於組合物加工品中。
本發明一實施例的組合物除了包含餘甘子萃取物外,更包含琉璃苣油與胎盤粉。
琉璃苣(Borago officinalis)是一種一年生雙子葉草本植物,原產於敘利亞。琉璃苣在夏季開花,其果實為小堅果。琉璃苣油(Borage oil)為琉璃苣的種子中所提煉出的種油。琉璃苣的種油含有17-28%的伽瑪亞麻油酸(順-6,9,12-十八碳三烯酸)、26-38%的脂肪酸、10-11%的棕櫚酸、3.5-4.5%的硬脂酸、16-20%的油酸、35-38%
的亞油酸、3.5-5.5%的花生酸、1.5-3.5%的芥子酸和1.5%的神經酸(順-15-二十四碳單烯酸)。琉璃苣油含有大量伽瑪亞麻油酸(Gamma-Linolenic Acid,GLA),伽瑪亞麻油酸屬於人體必須脂肪酸,其具有保濕、促進細胞再生以及提升棕脂(Brown Fat)活躍度以促進脂肪燃燒的功能。
胎盤粉出自於哺乳動物的胎盤。胎盤粉例如是由羊胎盤、牛胎盤或是豬胎盤等哺乳動物的胎盤所製成。胎盤內含有多種營養素與必需胺基酸。根據研究,其中的羊胎盤更具有益氣補虛,溫中暖下,調補腎虛、羸瘦,養血安神、豐肌澤膚、延年益壽之功效。
本發明以食用的方式由口攝取包含餘甘子萃取物、琉璃苣油與胎盤粉的組合物時,餘甘子萃取物的有效劑量為50毫克(mg)至100毫克(mg),琉璃苣油的有效劑量為300毫克(mg)至600毫克(mg),胎盤粉的有效劑量為150毫克(mg)至300毫克(mg)。於本發明部分實施例的組合物中,餘甘子萃取物與琉璃苣油之間的有效劑量比值為0.083至0.333。本發明另一部分實施例的組合物中,餘甘子萃取物與琉璃苣油之間的有效劑量比值為0.13至0.2。本發明再一部分實施例的組合物中,餘甘子萃取物、琉璃苣油與羊胎盤粉的有效劑量比(餘甘子萃取物重量:琉璃苣油重量:羊胎盤粉重量)為0.8-1.2:4.8-7.2:2.4-3.6。本發明再一部分實施例的組合物中,餘甘子萃取物、琉璃苣油與羊胎盤粉的有效劑量比(餘甘子萃取物重量:琉璃苣油重量:羊胎盤粉重量)為0.9-1.1:5.4-6.6:2.7-3.3。
以下藉由本發明一實施例說明本發明所揭露之組合物用於提升粒線體功能以對抗老化的用途,並且進行實驗測試以說明本發明所揭露之組合物用於抗老化的用途之功效。於本實施例中,組合物所包含的餘甘子萃取物為100毫克(mg),琉璃苣油為600毫克(mg),羊胎盤粉為300毫克(mg)。
實驗方式為先對受測者一至受測者五進行抽血檢驗,取得受測者一至受測者五進行實驗前血液中的GDF11濃度、BHI與DHEA-S濃度。接著,受測者一至受測者五於三個月的受測期間內,每天服用1顆包含餘甘子萃取物100毫克(mg)、琉璃苣油600毫克(mg)以及羊胎盤粉300毫克(mg)的膠囊。接著,於受測時間的最後對受測者一至受測者五進行抽血檢驗,取得受測者一至受測者五進行實驗後血液中的GDF11濃度、BHI與DHEA-S濃度。
受測者一至受測者五在實驗前與實驗後抽血檢驗的量測結果如表一與圖3至圖5所示。圖3為本發明實施例的組合物對不同受試者血液中GDF11濃度之影響的示意圖。圖4為本發明實施例的組合物對不同受試者血液中細胞的BHI之影響的示意圖。圖5為本發明實施例的組合物對不同受試者血液中DHEA-S濃度之影響的示意圖。
如圖3所示,受測者一至受測者五在服用組合物三個月後,血液中的GDF11濃度相較於服用組合物前平均提高了66%,說明
了本發明實施例含有餘甘子萃取物的組合物具有提高血液中GDF11濃度的功能,代表本發明實施例含有餘甘子萃取物的組合物具有抗老化之效果。如圖4所示,受測者一至受測者五在服用組合物三個月後,血液中的細胞的BHI相較於服用組合物前平均提高了35%,說明了本發明實施例含有餘甘子萃取物的組合物具有提高血液中細胞的BHI的功能,代表本發明實施例含有餘甘子萃取物的組合物具有降低粒線體老化程度的效果。如圖5所示,受測者一至受測者五在服用組合物三個月後,血液中的DHEA-S濃度相較於服用組合物前平均提高了12%,說明了本發明實施例含有餘甘子萃取物的組合物具有提高血液中DHEA-S濃度的功能,代表本發明實施例含有餘甘子萃取物的組合物具有抗老化之效果。綜上所述,由於本發明實施例含有餘甘子萃取物的組合物提高了服用者血液中的GDF11濃度、血液中細胞的BHI以及血液中的DHEA-S濃度,因此本發明實施例含有餘甘子萃取物的組合物具有延緩人體老化速度的效果。
根據上述本發明所揭露餘甘子萃取物用於製備提升粒線體功能以對抗老化之組合物的用途,食用餘甘子萃取物增加了血液中的GDF11(Growth Differentiation Factor 11)濃度。如此一來,本發明實施例含有餘甘子萃取物的組合物具有延緩人體老化速度的效果。
再者,食用餘甘子萃取物增加了血液中細胞的BHI(Bioenergetic Healthy Index)。如此一來,本發明實施例含有餘甘子萃取物的組合物具有降低粒線體老化程度的效果。
再者,食用餘甘子萃取物增加了血液中的脫氫異雄固酮(Dehydroepiandrosterone Sulfate,DHEA-S)濃度。如此一來,本發明實施例含有餘甘子萃取物的組合物具有延緩人體老化速度的效果。

Claims (5)
- 一種餘甘子萃取物用於製備提升粒線體功能以對抗老化之組合物的用途,其中:食用一組合物以增加BHI(Bioenergetic Healthy Index),該組合物包含一餘甘子萃取物、一溶劑及一胎盤粉,該溶劑為琉璃苣油,該胎盤粉為羊胎盤粉,該餘甘子萃取物的有效劑量為50毫克(mg)至100毫克(mg),該溶劑的有效劑量為300毫克(mg)至600毫克(mg),該胎盤粉的有效劑量為150毫克(mg)至300毫克(mg),且該餘甘子萃取物、該溶劑與該胎盤粉的有效劑量比為0.8-1.2:4.8-7.2:2.4-3.6。
- 如請求項1所述之餘甘子萃取物用於製備提升粒線體功能以對抗老化之組合物的用途,其中該餘甘子萃取物包含有重量百分比35%至55%之Emblicanin-A與Emblicanin-B混合物、重量百分比4%至15%之Ponigluconin、重量百分比10%至20%之Pedunculagin、重量百分比5%至15%之Rutin與重量百分比10%至30%之Gallo-ellagitannoids。
- 如請求項1所述之餘甘子萃取物用於製備提升粒線體功能以對抗老化之組合物的用途,其中食用該組合物時,該組合物被包裹於膠囊中。
- 如請求項1所述之餘甘子萃取物用於製備提升粒線體功能以對抗老化之組合物的用途,其中該餘甘子萃取物的有效劑量為100毫克(mg),該溶劑的有效劑量為600毫克(mg),以及該胎盤粉的有效劑量為300毫克(mg)。
- 如請求項1至4中任一項所述之餘甘子萃取物用於製備提升粒線體功能以對抗老化之組合物的用途,其中食用該組合物以增加血液中的脫氫異 雄固酮(Dehydroepiandrosterone Sulfate,DHEA-S)濃度或增加血液中的GDF11濃度。
