Description
本發明係關於應用於牙齒以提供美白及其他益處之新穎薄膜組成物。
現今使用之大多數牙齒美白系統含有過氧化物或其他漂白材料。以過氧化物為基質之產品可能呈現調配困難，因為過氧化氫之揮發性與反應性，且許多國家嚴格地規定牙齒美白產品中之過氧化物量。
亦有一需求，以保護牙齒免受細菌及牙菌斑形成，以及提供活性劑之持續傳遞，例如氟化物、抗菌劑以及再礦化劑。

我們已發現包含丙烯酸酯/辛基丙烯醯胺共聚物之調配物係適合應用於牙齒、其容易施加以及其相較於其他聚合物系統更持久(甚至在面對刷牙及酸的考驗)，因此使其成為用於傳遞及持續施加美白粒子及/或活性劑至牙齒以及用於保護牙齒免於染色(staining)以及生物薄膜及牙菌斑形成(其除此之外可能導致齲齒及牙齦炎)之合適載劑。

如全文所用，範圍係為了描述在範圍中之各個及每個數值而使用作為簡略表達。範圍中之任何數值可選擇作為範圍之終點。此外，本文所引述之所有文獻藉此藉由其全體之參照而併入。在本揭示內容中之定義與所引述文獻之定義衝突的事件中，以本揭示內容支配。
除非另有指明，應瞭解以下及說明書其他地方所表示之所有百分比及量係指重量百分比。所提供之量係以材料之有效重量(active weight)為基準。
測試調配物係如下製備：
E7-乙基纖維素(Ethocel® E7(Dow))
HPC-羥丙基纖維素
A/O-丙烯酸酯/辛基丙烯醯胺共聚物(DERMACRYL ® 79)
EPO-甲基丙烯酸丁酯-(2-二甲基胺基乙基)甲基丙烯酸酯-甲基丙烯酸甲酯共聚物
RL 100-甲基丙烯酸銨共聚物
在機械及酸考驗下移除之塗料百分比：珐琅板(Enamel blocks)係藉由先暴露於胜肽20 uM胜肽(DE 62)以及隨後使用於10mM磷酸鹽緩衝液中之0.5%紅氧化鐡分散液著色而染色。各個經染色之珐琅板以1毫升之配方(formula)處理2分鐘並允許風乾。對於濕式擦拭(wet swabbing)，使用水濕潤聚苯乙烯發泡體拭子(Styrofoam swab)並且使用於擦拭測試。對於刷淨考驗，製備1：2：：牙膏：水漿料並使用於刷淨。牙膏係商購牙膏(COLGATE Max Fresh ®)。刷淨各個板15秒中。對於酸考驗，經處理之板暴露於酸(pH=3.8)5分鐘並再次以牙膏刷淨15秒。
在37C下唾液暴露5小時而移除之塗料百分比：為了模擬口腔內條件，各經處理(塗布)之板係以1毫升經淨化(clarified)全唾液處理以及在37ºC下溫育5小時。後處理，對於不同考驗的測試，再測式各板，並且記錄移除塗料之百分比。因此下表中，移除百分比越低，塗料更加持久且有效。
各板係連續地暴露於所有的上述三個考驗。僅包含丙烯酸酯/辛基丙烯醯胺共聚物於20%下之調配物(C)對於刷淨、酸及唾液而不受影響，雖然包含丙烯酸酯/辛基丙烯醯胺共聚物於10%下之調配物(B)亦差不多地進行。
色斑(stain)測試
：此測試係設計於確認若經塗布之表面引來色斑。牛的珐琅板係以原型溶液(prototype solutions)塗布，以及在乾燥時係在40ºC下暴露於茶-咖啡色斑(1：2比率)30分鐘。美白程度係在暴露於該染色溶液之前及之後量測。
在40ºC下，來自A及F之塗料在熱溫度條件下
並不存在以及塗料無任何機械力而完全移除。使用EPO(D以及E)，30-50%之塗料在處理後脫離，再次無任何機械力，以及殘留之塗料係更深色，其指示出來自茶-咖啡染色溶液之被塗料吸收之色斑。具僅具有丙烯酸酯/辛基丙烯醯胺共聚物之原型B及C在熱溫度條件下仍存在，以及此外此些原型並不會吸收色斑。因此，調配物係有用於防止染色。
進一步顯示乙基纖維素及羥丙基纖維素的存在(其用作為分散劑)以及在乙基纖維素情況下(作為額外的薄膜成形劑)，增進丙烯酸酯/辛基丙烯醯胺共聚物調配物之保護免被染色的效果，其可能係藉由增加薄膜之疏水性。如上可知，使用調配物A，乙基纖維素及羥丙基纖維素本身係完全不受影響，因為其並不會附著至牙齒，因此此係令人驚訝的協乘性。在本實驗中之控制組係一裸珐琅板；更負的L值指出增加的染色。
亦評估乾燥時間及穩定性。穩定性係使用光學離
心作為對物理穩定性之預測模式而評估。將調配物注入小的離心管。將樣品水平地置於旋轉板上並以高速(約2400 rpm)自旋(spun)/離心約3天。自光源發射之光傳導通過光析管(cell)以及藉由偵測器紀錄經傳送之光。經傳送之光的百分比隨著時間描繪，以及有一已建立之分離度(如藉由增加光穿透率而指出)與架(shelf)上穩定性之間的關聯性。3或更低的穩定性分數被認為是可接受的。某些經測試之調配物具有對應於約6-12個月儲存穩定性(shelf stability)之穩定性。
製備含有不同量分散劑之調配物，如表3a、3b及3c中提供，單位為毫升/10毫升總體積：

成分相對分離度量測之穩定性係隨後以光學離心模式(optical centrifugation model)評估，以預測在3個月、6個月及1年時之穩定性，如表5中所示，其中數字反應出分離度、更低數字指出更大的穩定性、以及3或更低被認為是穩定的：
調配物10顯示良好的穩定性，就如同調配物4、6及9。展現更短乾燥時間之調配物傾向具有更佳的穩定性，大概是因為兩者參數係關於分散度。
Claims (7)
- 一種形成牙齒薄膜之組成物，其包括i.丙烯酸酯/辛基丙烯醯胺共聚物、ii.一或多種烷基纖維素醚，其包含乙基纖維素以及羥丙基纖維素，且乙基纖維素對羥丙基纖維素之比率係2：1至30：1、以及iii.口腔可接受溶劑，其中該丙烯酸酯/辛基丙烯醯胺共聚物係以5-30重量%之量存在，且該口腔可接受溶劑係乙醇。
- 如申請專利範圍第1項之組成物，其中該丙烯酸酯/辛基丙烯醯胺共聚物係2-丙烯酸，2-甲基-、2-甲基丙基酯，具有2-丙烯酸及N-(1,1,3,3-四甲基丁基)-2-丙烯醯胺之聚合物。
- 如申請專利範圍第1項之組成物，其中該丙烯酸酯/辛基丙烯醯胺共聚物係以大於10重量%之量存在。
- 如上述申請專利範圍第1項之組成物，進一步包含美白材料。
- 如申請專利範圍第4項之組成物，其中該美白材料係選自二氧化鈦、氧化鋅、氫氧基磷灰石及其組合。
- 如上述申請專利範圍第1項之組成物，其包含一或多種抗菌劑、氟離子源(fluoride ion source)、鈣源或鹼性胺基酸。
- 一種套件，其包含如申請專利範圍第1至6項中之任一項之組成物以及一用於施加該組成物至牙齒之施用器。
