Description
本發明有關含有L-麥角硫因(ergothioneine)或其鹽及槲皮素或其配糖體之組成物。又,本發明有關經口組成物之微生物增殖抑制方法。本發明又有關槲皮素或其配糖體之微生物增殖抑制作用增強劑等。
市售之飲食品以及醫藥品等之經口組成物中,抑制微生物增殖及提高保存性非常重要。因此,有效抑制微生物增殖且安全性高的源自天然之成分為有用。
槲皮素係類黃酮之一種,其直接或以配糖體包含於洋蔥等植物中。非專利文獻1中記載槲皮素可抑制大腸桿菌及金黃色葡萄球菌之增殖。
麥角硫因係含硫胺基酸之一種,天然以L型存在。已知L-麥角硫因具有抗氧化作用等之有用生理活性。例如專利文獻1中記載源自金頂側耳之麥角硫因作用於人類不死化角化細胞(HaCaT細胞)時,人類β-防禦素-1(hBD-1)及人類β-防禦素-3(hBD-3)之mRNA表現量增加。

[用以解決課題之手段]
本發明人等為了解決上述課題而積極研究之結果,發現L-麥角硫因增強了槲皮素或其配糖體之微生物增殖抑制作用。藉由併用L-麥角硫因或其鹽與槲皮素或其配糖體,可有效抑制微生物增殖。
即,雖不限定於以下,但本發明有關以下之組成物、經口組成物之微生物增殖抑制方法及槲皮素或其配糖體之微生物增殖抑制作用增強劑。
[1] 一種組成物,其含有(A) L-麥角硫因或其鹽,及(B)槲皮素或其配糖體。
[2] 如上述[1]之組成物,其中(B)槲皮素或其配糖體係選自由異槲皮素(isoquercetin)、盧丁(rutin)、酵素處理盧丁及槲皮素(quercetin)所成之群中之至少1種。

依據本發明可提供有效抑制經口組成物等中之微生物增殖的技術。
(組成物)
本發明之組成物含有(A) L-麥角硫因或其鹽及(B)槲皮素或其配糖體。
槲皮素或其配糖體具有微生物增殖抑制作用(抗菌作用)。將L-麥角硫因或其鹽與槲皮素或其配糖體組合時,可增強槲皮素或其配糖體之微生物增殖抑制作用。因此,藉由組合使用L-麥角硫因或其鹽與槲皮素或其配糖體,可有效抑制微生物增殖。
L-麥角硫因係胺基酸之一種。作為L-麥角硫因之鹽只要為藥理學上可接受之鹽或飲食品可接受之鹽則未特別限制,可為酸性鹽及鹼性鹽之任一者。作為酸性鹽,舉例為例如鹽酸鹽、硫酸鹽、硝酸鹽、磷酸鹽等之無機酸鹽;乙酸鹽、檸檬酸鹽、馬來酸鹽、蘋果酸鹽、草酸鹽、乳酸鹽、琥珀酸鹽、富馬酸鹽、丙酸鹽等之有機酸鹽等。作為鹼性鹽舉例為例如鈉鹽、鉀鹽等之鹼金屬鹽;鈣鹽、鎂鹽等之鹼土類金屬鹽等。
作為L-麥角硫因或其鹽,較佳使用L-麥角硫因。
L-麥角硫因或其鹽可使用化學合成品,亦可使用自天然物萃取及純化者。L-麥角硫因大多含於側耳科(Pleurotaceae)側耳屬之蕈菇的金頂側耳(Golden/Yellow Oyster Mushroom)(學名:Pleurotus cornucopiae var. citrinopileatus)。L-麥角硫因亦含於白蘑菇、褐色菇、波特貝拉(Portabella)蘑菇等之雙孢蘑菇(學名:Agaricus bisporus)、袖珍菇(Grey Oyster Mushroom)(學名:Pleurotus ostreatus)、椎茸(學名:Lentinula edodes)、舞菇(學名:Grifola frondosa)、靈芝(學名:Ganoderma lucidum)、猴頭菇(學名:Hericium erinaceus)、柳松茸(學名:Agrocybe aegerita)、雞油菌(學名:Cantharellus cibarius)、牛肝菌(學名:Boletus edulis)、羊肚菌(學名:Morchella esculenta)等之蕈菇中。自天然物獲得L-麥角硫因時,較佳自金頂側耳萃取等。L-麥角硫因或其鹽亦可藉微生物發酵而製造。亦可使用包含以微生物發酵製作之L-麥角硫因或其鹽之萃取物或自該等純化者。自天然物等之萃取及純化可藉由習知方法實施。L-麥角硫因或其鹽亦可經單離。
組成物中之L-麥角硫因或其鹽之含量未特別限制,但例如以L-麥角硫因換算,較佳為0.00001重量%以上,更佳為0.0001重量%以上,且可為50重量%以下,可為10重量%以下,較佳為0.1重量%以下,更佳為0.01重量%以下。一態樣中,組成物中之L-麥角硫因或其鹽之含量,以L-麥角硫因換算,可為0.00001~50重量%,可為0.00001~10重量%,較佳為0.00001~0.1重量%,更佳為0.0001~0.01重量%。L-麥角硫因或其鹽之含量可藉由高速液相層析(HPLC)法測定。
本說明書中,關於L-麥角硫因或其鹽,以L-麥角硫因換算之量或類似表現係指L-麥角硫因或鹽之作為L-麥角硫因之量。具體而言,於L-麥角硫因時意指其量,於L-麥角硫因之鹽時,意指以該鹽的莫耳數乘以L-麥角硫因的分子量而得之值。
本說明書中,所謂槲皮素係指屬於多酚之一種的黃醇酮之化合物的槲皮素。本發明中,所謂槲皮素配糖體意指上述槲皮素之配糖體,具體而言為於槲皮素之3位羥基及/或4’位之羥基鍵結糖苷鍵之一連串化合物之總稱。槲皮素配糖體可為1種化合物亦可為2種以上之化合物。作為與槲皮素鍵結之糖(單糖)可舉例為葡萄糖、鼠李糖、半乳糖、葡萄糖醛酸等,較佳為葡萄糖、鼠李糖。與槲皮素鍵結之糖亦可經修飾(如丙二醯化)。
一態樣中,作為槲皮素配糖體,較佳為於槲皮素之3位羥基糖苷鍵結有1個以上之糖的化合物。於槲皮素之3位羥基糖苷鍵結有1個以上之糖的槲皮素配糖體係以下述通式表示之化合物。下述通式中之(X)n表示糖鏈。X表示糖(單糖),n為1以上之整數。
與槲皮素糖苷鍵結之以X表示之構成糖鏈的糖舉例為例如葡萄糖、鼠李糖、半乳糖、葡萄糖醛酸等,較佳為葡萄糖、鼠李糖。且,n若為1以上,則未特別限制,但較佳為1~16,更佳為1~8。n為2以上時,X部分可由一種糖而成,亦可由複數種糖而成。若換言之,n為2以上時,(X)n可為由1種糖所成之糖鏈或由複數種糖所成之糖鏈。
本發明中之槲皮素配糖體亦包含既有的槲皮素配糖體以酵素等處理而進行糖轉移者。本發明中所稱之槲皮素配糖體具體舉例為蘆丁(槲皮素3-盧丁糖苷)(quercetin 3-lutinoside)、酵素處理蘆丁(蘆丁之酵素處理物)、槲皮素(槲皮素-3-O-鼠李糖苷)、異槲皮苷(槲皮素-3-葡萄糖苷)、槲皮素3,4’-二葡萄糖苷、槲皮素4’-葡萄糖苷、槲皮素3-(6-丙二醯基葡萄糖苷)、金絲桃苷(槲皮素3-半乳糖苷)、槲皮素-3-O-葡糖苷酸等。酵素處理蘆丁係於異槲皮素之葡萄糖苷部分進而鍵結糖鏈之槲皮素配糖體。作為酵素處理蘆丁之較佳例,舉例為將蘆丁進行酵素處理而將鼠李糖鏈部分去除之異槲皮素以糖轉移酵素處理所得之於異槲皮素鍵結有由1~7個葡萄糖所成之糖鏈者,及以其混合物為主成分者。一態樣中,作為槲皮素或其配糖體較佳為槲皮素配糖體,更佳為於槲皮素之3位之羥基糖苷鍵結有1個以上之糖的槲皮素配糖體,更佳為於選自由異槲皮素、蘆丁、酵素處理蘆丁及槲皮素(quercetin)所成之群之至少1種,又更佳為異槲皮素、酵素處理蘆丁。作為槲皮素配糖體,特佳為於槲皮素之3位之羥基糖苷鍵結有1~8個葡萄糖之化合物(例如異槲皮苷、於異槲皮素鍵結有1~7個葡萄糖之配糖體),最佳為異槲皮素。
本發明中,關於用以獲得槲皮素或其配糖體之來源、製法未特別限制。例如,作為含有大量槲皮素或其配糖體之植物,已知有蕎麥、槐、刺山柑、蘋果、茶、洋蔥、葡萄、西蘭花、宿葵、覆盆子、越橘、蔓越莓、仙人掌、葉菜類、柑橘類類等,由該等植物可獲得槲皮素或其配糖體。
本發明中使用之槲皮素或其配糖體,只要可發揮本發明之效果,則可使用將源自上述植物等之天然物的萃取物經濃縮、純化等操作而提高槲皮素或其配糖體之含量者,例如可使用含有槲皮素或其配糖體之萃取物之濃縮物或純化物。作為濃縮方法或純化方法,可採用已知方法。本發明中,亦可使用經純化或單離之槲皮素或其配糖體。槲皮素或其配糖體亦可使用化學合成品。
組成物中之槲皮素或其配糖體之含量,例如以異槲皮素換算,較佳為0.0001重量%以上,更佳為0.001重量%以上,且為50重量%以下,可為10重量%以下,較佳為1重量%以下,更佳為0.1重量%以下。一態樣中,組成物中之槲皮素或其配糖體之含量,以異槲皮素換算,可為0.0001~50重量%,可為0.0001~10重量%,較佳為0.0001~1重量%,更佳為0.001~0.1重量%。
槲皮素或其配糖體之含量可藉由高速液相層析(HPLC)法測定。
本說明書中,關於槲皮素或其配糖體,以異槲皮素換算之量或類似表現意指槲皮素或其配糖體作為異槲皮素之量。具體而言,化合物為異槲皮素時表示其量,化合物為槲皮素或異槲皮素以外之槲皮素配糖體時,意指對該化合物之莫耳數乘以異槲皮素的分子量所得之值。
本發明中,相對於(B)槲皮素或其配糖體之含量(以異槲皮素換算)的(A)L-麥角硫因或其鹽之含量(以L-麥角硫因換算)之重量比(L-麥角硫因換算之(A)/異槲皮素換算之(B))較佳為0.01以上,更佳為0.02以上,又更佳為0.025以上,且,較佳為1.2以下,更佳為1.1以下,又更佳為1以下。相對於(B)槲皮素或其配糖體之異槲皮素換算之含量的(A)L-麥角硫因或其鹽之L-麥角硫因換算之含量的重量比(L-麥角硫因換算之(A)/異槲皮素換算之(B))若為上述範圍,則有效地增強槲皮素或其配糖體之微生物增殖抑制作用。一態樣中,上述重量比(L-麥角硫因換算之(A)/異槲皮素換算之(B))較佳為0.01~1.2,更佳為0.02~1.2,又更佳為0.025~1.2,再更佳為0.025~1.1,特佳為0.025~1。本發明之組成物較佳以上述重量比包含(A)L-麥角硫因或其鹽及(B)槲皮素或其配糖體。
本發明之組成物由於包含(A)L-麥角硫因或其鹽及(B)槲皮素或其配糖體,故可抑制微生物增殖。本發明之組成物可作為微生物之增殖受抑制之的組成物(較佳為經口組成物)而有用。
作為學術定義,微生物不僅包含細菌(bacteria),亦包含菌類(蕈菇、黴菌、酵母菌等)、病毒、微細藻類等,但本說明書中,所謂微生物係指細菌(bacteria)。
本發明中,作為微生物,較佳為大腸桿菌。藉由組合L-麥角硫因或其鹽與槲皮素或其配糖體,可發揮對大腸桿菌之優良增殖抑制作用。
本發明之組成物可作成例如飲食品、醫藥品、準醫藥、飼料等之形態。本發明之組成物本身可為飲食品、醫藥品、準醫藥、飼料等,亦可調配其而使用之材料或製劑等。一態樣中,本發明之組成物為飲食品、醫藥品、準醫藥、飼料等時,組成物中之L-麥角硫因或其鹽之含量,以L-麥角硫因換算,較佳為0.0001~0.1重量%,更佳為0.001~0.01重量%。組成物中之槲皮素或其配苷體之含量,以異槲皮素換算,較佳為0.001~1重量%,更佳為0.01~0.1重量%。
一態樣中,本發明之組成物較佳為經口組成物。作為經口組成物舉例為飲食品、經口用醫藥品、準醫藥、飼料等,較佳為飲食品或經口用醫藥,更佳為飲食品。
一態樣中,本發明之組成物作為微生物增殖受抑制之經口組成物,較佳為大腸桿菌增殖受抑制之經口組成物而有用。一態樣中,本發明之組成物例如作為微生物增殖受抑制之飲食品、經口醫藥品、經口準醫藥、飼料等而有用。
本發明之組成物,只要不損及本發明之效果,除了L-麥角硫因或其鹽、槲皮素或其配糖體以外,可含有任意添加劑、任意成分。該等添加劑及成分可根據組成物形態等進行選擇,可使用一般於飲食品、醫藥品、準醫藥、飼料等可使用者。本發明之組成物作成飲食品、醫藥品、準醫藥、飼料等時,其製造方法未特別限制,可藉由一般方法製造。
本發明之組成物形態未特別限制,可為固體狀(粉末狀、顆粒狀、片狀等)、液狀、糊狀等。
例如本發明之組成物作成飲食品時,於L-麥角硫因或其鹽及槲皮素或其配糖體中調配飲食品可使用之成分(例如,食品材料、根據需要使用之食品添加劑),可作成各種飲食品。飲食品未特別限制,例如可舉例為一般飲食品、健康食品、健康飲料、機能性顯示食品、特定保健用食品、健康輔助食品、病人用飲食品等。上述健康食品、機能性顯示食品、特定保健用食品、健康輔助食品等可作成細粒劑、錠劑、顆粒劑、散劑、膠囊劑、咀嚼劑、糖漿劑、液劑、流動食品等各種製劑形態。
本發明之組成物作成醫藥品或準醫藥時,例如於L-麥角硫因或其鹽及槲皮素或其配糖體中調配藥理學上可接受之載體,根據需要添加之添加劑等,製成各種劑型之醫藥品或準醫藥。此等載體、添加劑等只要為可使用於醫藥品或準醫藥、藥理學上可接受者即可,舉例為例如賦形劑、結合劑、崩解劑、潤滑劑、抗氧化劑、著色劑等之1或2種以上。作為醫藥品或準醫藥之投予形態,舉例為經口或非經口投予之形態,但較佳為經口投予形態。本發明之組成物作成醫藥品或準醫藥時,較佳作成經口醫藥品或準醫藥。作為用於經口投予之劑型舉例為液劑、錠劑、散劑、細粒劑、顆粒劑、糖衣錠、膠囊劑、懸浮劑、乳劑、咀嚼劑等。作為非經口投予之劑型舉例為注射劑、點滴劑等。
本發明之組成物作成飼料時,只要於飼料中調配L-麥角硫因或其鹽及槲皮素或其配糖體即可。飼料中亦包含飼料添加劑。作為飼料,舉例為例如用於牛、豬、雞、羊、馬等之家畜用飼料;用於兔子、大鼠、小鼠等之小動物用飼料;用於狗、貓、小鳥等的寵物食品等。
本發明之組成物作成經口組成物時,攝取或投予本發明之組成物的對象(亦可稱為投予對象)未特別限制。較佳為人類或非人類哺乳動物,更佳為人類。
本發明之組成物作成經口組成物時,本發明之組成物的攝取量(投予量)未特別限制,但例如於人類(成人)之情況,以L-麥角硫因換算每天之攝取量較佳為2~50mg,更佳為5~25mg,又更佳為5~20mg,特佳為10~20mg。
(A)L-麥角硫因或其鹽與(B)槲皮素或其配糖體之組合,可作為用以抑制微生物增殖之有效成分使用。一態樣中,本發明之組成物可較佳地使用於例如抑制微生物增殖。一態樣中,本發明之組成物可為微生物增殖抑制用組成物。
L-麥角硫因或其鹽及槲皮素或其配糖體含於天然物及飲食品中係有食用經驗的化合物。本發明之組成物可較佳地使用於例如抑制經口組成物中之微生物增殖。一態樣中,本發明之組成物可較佳地使用作為經口組成物之微生物增殖抑制用組成物。微生物增殖抑制用組成物亦可稱為微生物增殖抑制劑。一態樣中,本發明之組成物可較佳地使用作為大腸桿菌增殖抑制用組成物。
一態樣中,本發明之組成物例如可添加於經口組成物中使用。本發明之組成物使用於抑制經口組成物中之微生物增殖時,只要將本發明之組成物添加於經口組成物中即可。一態樣中,本發明之組成物可作為食品添加劑使用。添加本發明之組成物的經口組成物未特別限制,可舉例為飲食品、經口醫藥品、經口準醫藥、飼料等,較佳為飲食品。一態樣中,本發明之組成物作為微生物增殖抑制用組成物而添加於飲食品等中而使用時,組成物中之(A)L-麥角硫因或其鹽及(B)槲皮素或其配糖體之合計含量例如為0.01~99重量%,較佳為0.1~90重量%。
本發明之組成物基於獲得微生物增殖抑制效果之目的而添加於經口組成物時,其添加量為經口組成物中L-麥角硫因或其鹽之含量,以L-麥角硫因換算,較佳為0.00001~0.1重量%,更佳為0.0001~0.01重量%之量。又,槲皮素或其配糖體之添加量為經口組成物中之槲皮素或其配糖體含量,以異槲皮素換算,較佳為0.001~1重量%,更佳為0.01~0.1重量%之量。基於獲得微生物增殖抑制效果之觀點,較佳添加L-麥角硫因或其鹽及槲皮素或其配糖體之含量成為上述量之組成物。
(微生物增殖抑制方法)
本發明又包含經口組成物之微生物增殖抑制方法,係將(A)L-麥角硫因或其鹽與(B)槲皮素或其配糖體添加於經口組成物中。
本發明之方法中,(A)L-麥角硫因或其鹽及(B)槲皮素或其配糖體,該等之較佳態樣與上述本發明之組成物相同。
例如,槲皮素或其配糖體較佳為選自由異槲皮素、蘆丁、酵素處理蘆丁及槲皮素所成之群中之至少1種。
本發明之方法中,微生物及其較佳態樣與上述者相同,較佳為大腸桿菌。本發明之方法較佳為經口組成物中之大腸桿菌增殖抑制方法。經口組成物未特別限制,可舉例為上述飲食品、經口醫藥品、經口準醫藥、飼料等。

L-麥角硫因或其鹽之添加量,較佳係經口組物中之L-麥角硫因或其鹽之含量,以L-麥角硫因換算,較佳成為0.00001~0.1重量%,更佳為0.0001~0.01重量%之量。槲皮素或其配糖體之添加量,較佳係經口組物中之槲皮素或其配糖體含量以異槲皮素換算,較佳成為0.001~1重量%,更佳為0.01~0.1重量%之量。
本發明之方法中,(A)L-麥角硫因或其鹽與(B)槲皮素或其配糖體亦可分別添加於經口組成物中,但亦可將包含該等之組成物添加於經口組成物中。例如,亦可使用上述本發明之組成物,將(A)L-麥角硫因或其鹽與(B)槲皮素或其配糖體添加於經口組成物中。

[實施例]
以下,藉由實施例更詳細說明本發明,但不限定本發明之範圍。
<實施例1>
調查L-麥角硫因(以下有時簡稱為麥角硫因)及槲皮素配糖體之除菌(微生物增殖抑制)效果。
槲皮素配糖體係使用異槲皮素(槲皮素3-葡萄糖苷,由Cayman化學公司製)。
試驗菌係使用大腸桿菌(Escherichia coli NBRC 3972)。
(試驗液菌之調製)
將大腸桿菌接種於普通瓊脂培養基中,在30~35℃培養24小時。培養後,以生理食鹽水調製為菌數為10
8CFU/mL,作為試驗菌液。
(試驗試料之調製)
將磷酸緩衝生理食鹽水及麥角硫因混合,調製試驗試料1(麥角硫因濃度100ppm(0.01重量%))。
將磷酸緩衝生理食鹽水及異槲皮素混合,調製試驗試料2(異槲皮素濃度100ppm(0.01重量%))。
將磷酸緩衝生理食鹽水、麥角硫因及異槲皮素混合,調製試驗試料3(麥角硫因濃度2.5ppm(0.00025重量%,異槲皮素濃度100ppm(0.01%重量)。將磷酸緩衝生理食鹽水、麥角硫因及異槲皮素混合,調製試驗試料4(麥角硫因濃度100ppm(0.01重量%),異槲皮素濃度100 ppm(0.01重量%)。

接種1小時後,以添加有卵磷脂聚山梨酯80之大豆‧酪蛋白‧消化液體培養基製備經接種試驗菌液之試驗試料之10倍稀釋度。將所得之包含大腸桿菌之稀釋液接種於添加有卵磷脂聚山梨酯80之大豆‧酪蛋白‧消化瓊脂培養基中,於30~35℃培養3~5天。培養後,計數所形成之菌落並計算生菌數(n=3)。
從生菌數(n=3之平均值),以下述式求出除菌效果(%)。

圖1顯示L-麥角硫因(試驗試料1)、異槲皮素(試驗試料2)、L-麥角硫因與異槲皮素之組合(試驗試料3~4)之除菌效果的圖表。單獨使用L-麥角硫因未見到對大腸桿菌之除菌效果。單獨使用異槲皮素時雖抑制大腸桿菌增殖,但藉由L-麥角硫因與異槲皮素組合所得之大腸桿菌的增殖抑制效果係比基於單獨使用各成分所得之效果所預測的相加效果更顯著優異的相乘效果。可知藉由併用L-麥角硫因與異槲皮素,獲得對微生物之優異除菌效果,即獲得優異之微生物增殖抑制效果。
[圖1]係顯示L-麥角硫因(試驗試料1)、異槲皮素(試驗試料2)、L-麥角硫因與異槲皮素之組合(試驗試料3~4)之除菌效果之圖表。
Claims (11)
- 一種組成物,其含有(A)L-麥角硫因(ergothioneine)或其鹽,及(B)槲皮素或其配糖體。
- 如請求項1之組成物,其中(B)槲皮素或其配糖體係選自由異槲皮素(isoquercetin)、盧丁(rutin)、酵素處理盧丁及槲皮素(quercetin)所成之群中之至少1種。
- 如請求項1或2之組成物,其中相對於(B)槲皮素或其配糖體之含量(異槲皮素換算)的(A)L-麥角硫因或其鹽之含量(L-麥角硫因換算)之重量比(L-麥角硫因換算之(A)/異槲皮素換算之(B))為0.01~1.2。
- 如請求項1或2之組成物,其係微生物增殖抑制用組成物。
- 如請求項4之組成物,其係經口組成物的微生物增殖抑制用組成物。
- 如請求項4之組成物,其中微生物為大腸桿菌。
- 一種經口組成物之微生物增殖抑制方法,其係於經口組成物中添加(A)L-麥角硫因或其鹽,及(B)槲皮素或其配糖體。
- 如請求項7之方法,其中(B)槲皮素或其配糖體係選自由異槲皮素、盧丁、酵素處理盧丁及槲皮素所成之群中之至少1種。
- 如請求項7或8之方法,其中係以經口組成物中之相對於(B)槲皮素或其配糖體之含量(異槲皮素換算)的(A)L-麥角硫因或其鹽之含量(L-麥角硫因換算)之重量比(L-麥角硫因換算之(A)/異槲皮素換算之(B))成為0.01~1.2之方式,將(A)L-麥角硫因或其鹽及(B)槲皮素或其配糖體添加於經口組成物中。
- 如請求項7或8之方法,其中微生物為大腸桿菌。
- 一種槲皮素或其配糖體之微生物增殖抑制作用增強劑,其包含L-麥角硫因或其鹽。
