Abstract
Description
The invention relates to agriculture, in particular, to animal husbandry, the field of preventive wellness measures aimed at improving the quality of milk and productivity of highly dairy cows and can be used on livestock complexes, on small commodity farms with various technologies for keeping animals.
- 2015. - №2. - С. 22-23).On the Russian market, drugs for single-run cows are known, in particular, bactericidal antibiotics Orbenin DC and Orbenin EDC (Pfizer, USA). Multimast DC (Bimeda Animal Health, Ireland) and Nafpenzal DC (Intervet, Netherlands) are also widely used. Bayokloks (Bayer, Germany) also proved to be quite good (see Semen L.K. Sensitivity of microflora isolated from the secret of the udder of patients with mastitis of cows to drugs of prolonged action / L.K.Semen, T.G. Voroshilova, E.V. Remizova // Russian Veterinary Journal (farm animals) .- 2015. - No. 2. - S. 22-23).
In Russia, a number of drugs are also known that are used to treat and prevent mastitis in cows during the dry period, in which the antibiotic of the tetracycline group, doxycycline, is used as the active substance. So, the complex preparation Doksimast contains doxycycline, emulsion wax, aluminum stearate and liquid paraffin. The composition of the drug Neodoximast also includes doxycycline, neomycin sulfate, emulsion wax, aluminum stearate and vaseline oil. The administration of Doksimast prevented the development of mastitis in 94.4% of cows. The use of Neodoximast for cows at the end of the run, like Orbenin DC, provides a preventive effect in 90% of animals (see Klimov N.T. Doksimast - a drug for the prevention of mastitis in dry cows / N.T. Klimov // Modern problems of veterinary reproductive health of animals : Materials of the International Scientific and Practical Conference dedicated to the centenary of the birth of Professor V.A. Akatov, Voronezh: Sources, 2009. - S. 207-209). With good results from the use of these drugs, animals become addicted to them and it is necessary to develop newer and more effective drugs.
The objective of the invention is to develop a method for the prevention of mastitis in highly productive cows while improving the quality of milk without the use of expensive imported medications, while expanding the range and methods of administering drugs that do not require large labor costs and funds, for the manufacture of the drug and its use, simple in execution, but allowing significantly to improve the quality and quantity of products received from lactating cows while maintaining and improving animal health, without violating the accepted system emy prevention and treatment.
The problem is solved in that a pharmacological composition consisting of doxycycline as an active substance, silicon dimethylglycerolates, silicon glycerolates as a base, hydroxyethyl cellulose as a stabilizer and distilled water as a solvent was developed and experimentally tested, while the preparation consists of the following components:
20.02.2010 г.).Silicon dimethylglycerolates are synthesized in a 1-molar excess of glycerol (CH 3 ) 2 Si (C 3 H 7 O 3 ) 2 ° C 3 H 8 O 3 . The presence of silicon dimethylglycerolates in the composition (2.5 wt.%) Not only has a reparative and regenerative effect, but also allows, due to transcutaneous activity, to reduce the dose of active active substances in them while maintaining high efficiency, replacing dimethyl sulfoxide (see RF patent No. 2282046, C07F 7/18, publ. 02.20.2010).
The presence of silicon glycerolates in the composition (2.5 wt.%) Also has a reparative, regenerative effect and allows them to reduce the dose of active active ingredients while maintaining high efficacy, replacing DMSO (see RF Patent No. 2255939, C07F 7/04, A61K 47/30, published on July 10, 2005).
Before research on cows, an experimental preclinical study of the toxicological properties of the pharmacological composition for the prevention of mastitis in cows in the dry and postpartum periods in laboratory animals was conducted.
В результате по параметрам острой токсичности, заявляемая композиция может быть отнесена к четвертому классу опасности (малоопасные вещества).To establish LD 50, according to GOST 12.1.007-76, the maximum toxic dose during oral administration was 6,000 mg / kg and did not cause death in any animal, nor did it have a significant effect on the behavior and general condition of animals, growth rate, and vegetative functions, indicating a low toxicity of the studied drug. As a result, according to the parameters of acute toxicity, the claimed composition can be assigned to the fourth hazard class (low hazard substances).
Chronic toxicity was determined by subcutaneous administration of the drug. The results of hematological studies, leukocyte formula and biochemical analysis showed the absence of significant differences after application of the composition in comparison with control animals, which suggests that the claimed composition does not cause deviations in the growth rate of animals, hematopoiesis disorders, does not affect the functional state of internal organs and does not cause changes in metabolic processes.
The application of the test substance at a concentration of 2.5 mg / kg to the shaved areas of the skin of laboratory animals in no case led to the formation of erythema, scab and the occurrence of edema. Thus, the pharmacological composition does not cause a skin-irritating effect and does not have allergenic properties in the doses studied.
Scientific and industrial experience on the study of the effectiveness of the pharmacological composition was carried out on the basis of the pedigree factory of the Sverdlovsk region CJSC "Agrofirm" Patrushi ". The organization contains 1055 highly productive cows of black and motley breed with an average annual milk productivity of 8088 kg. According to the results of obstetric examination, two groups of analogue cows of 2-3 lactation were formed with a gestation period of 225-230 days. There were 10 cows in each group. The experimental animals in the early dry period, one day before start-up, were administered intracisternally to all lobes of the mammary gland using a syringe with a pharmacological composition in a dose of 5 ml. The introduction was carried out once after the last milking of the cows. Before the introduction of the pharmacological composition, milk was removed from all lobes of the udder, after administration, the nipples of the udder were massaged from the bottom up.
Перед запуском и после родов животных исследовали с помощью быстрого маститного теста «Кенотест» (CID LINES, Бельгия), производили отбор материала: крови для иммуногематологических исследований и содержимого молочной железы для микробиологического исследования, так же проводили исследование молока по показателю соматических клеток (таблица 1).Before starting and after childbirth, animals were examined using the Kenotest rapid mastitis test (CID LINES, Belgium), material was selected: blood for immunohematological studies and breast contents for microbiological examination, milk was also examined for somatic cells (table 1 )
DC (Orbenin DC)).The control group cows were run according to the technology adopted at the farm (Orbenin DC - Pfizer, USA). Orbenin DC is an antibacterial drug in the form of a suspension for intracisternal administration, containing 500 mg of cloxacillin in the form of a salt of benzathic acid in each individual package as an active ingredient, and mineral oil as an excipient. The drug is used once for the prevention and treatment of mastitis in all animals after the last milking before being transferred to the dry period (see Instructions for the use of the drug Orbenin DC (Orbenin DC)).
Изучение результатов гематологических исследований и лейкоцитарной формулы в послеродовой период коров, показало отсутствие достоверных отличий после применения фармакологической композиции в сравнении с показателями до запуска и с контрольными животными, что свидетельствует об отсутствии отрицательного влияния фармакологической композиций для одномоментного запуска коров на гемопоэз и свертываемость крови (таблица 2).A study of the results of hematological studies and the leukocyte formula in the postpartum period of cows showed the absence of significant differences after the use of the pharmacological composition in comparison with the pre-launch and control animals, which indicates the absence of a negative effect of the pharmacological compositions for the simultaneous launch of cows on hematopoiesis and blood coagulability (table 2).
е. (таблица 3).When conducting immunological studies of blood of cows before starting, there was a decrease in the number of T and B lymphocytes in the control group to 33.0 ± 1.4% and 15.9 ± 0.8%, and in the experimental group - 39.3 ± 3.2 % and 19.2 ± 2.8%, respectively, this decrease indicates a low value of immunocompetent cells and a decrease in cell-mediated immunity as a result of the development of depression of the immune system in subclinical mastitis. The number of circulating immune complexes in the control group of cows prior to launch was significantly increased to 171.8 ± 9.9 cu, while in the experimental group the increase was insignificant and amounted to 122.9 ± 7.3 cu (table 3).
This increase may be due to a violation of the factors of humoral immunity in the inflammatory reaction and an increase in the production of immunoglobulins. In the postpartum period in the experimental group, where a new pharmacological composition was used, the immune system parameters were within the normative values, while in the control group, as before, a decrease in T-lymphocytes, B-lymphocytes and an increased value of circulating immune complexes were detected.
Проведенные исследования показывают, что разработанная фармакологическая композиция оказывает положительное влияние на иммунологические показатели крови животных в сравнении с используемым препаратом в контрольной группе (таблица 3).Studies show that the developed pharmacological composition has a positive effect on the immunological parameters of the blood of animals in comparison with the drug used in the control group (table 3).
При этом количество пораженных долей в контрольной группе составило 10, а в опытной - 13.Prior to launch, in the control and experimental groups, subclinical mastitis was detected in 7 and 8 animals, respectively. Moreover, the number of affected shares in the control group was 10, and in the experimental group - 13.
When the pharmacological composition and control drug were used during the launch, in both groups in the postpartum period, the cows experienced a decrease in diagnosed mastitis.
При этом в опытной группе уменьшалось количество пораженных субклиническим маститом долей молочной железы на 10% по сравнению с контрольной группой (таблица 4).Thus, the use of the pharmacological composition allowed to reduce the level of subclinical mastitis in cows by 2 times, while in the control group the level of diagnosed mastitis decreased by 1.8 times. Moreover, in the experimental group, the number of breast fractions affected by subclinical mastitis decreased by 10% compared with the control group (table 4).
После применения фармакологической композиции у коров опытной группы в послеродовой период встречалось поражение субклиническим маститом одной доли молочной железы, в то время как у животных контрольной группы встречались поражения в одной и двух долях молочной железы (таблица 5).After applying the pharmacological composition in the cows of the experimental group in the postpartum period, subclinical mastitis of one lobe of the mammary gland was observed, while in animals of the control group lesions were found in one and two lobes of the mammary gland (Table 5).
The studies took into account the degree of damage to the lobes of the mammary gland of cows by subclinical mastitis. When diagnosing subclinical mastitis in the postpartum period, a mild (+) lesion of the lobes of the mammary gland was observed in the experimental group, while before starting 5 and 2 lobes of the mammary gland had moderate (++) and severe (+++) lesions, respectively.
В контрольной группе у животных в послеродовой период диагностировано легкое (+) и среднее (++) поражение долей молочной железы субклиническим маститом (таблица 6).In the control group, animals in the postpartum period were diagnosed with mild (+) and moderate (++) lesions of the breast lobes by subclinical mastitis (table 6).
Полученные данные показывают положительное влияние заявляемой фармакологической композиции на качество молока по показателю соматических клеток (таблица 7).During the period of the scientific and production experiment, we studied the dynamics of changes in somatic cells (SC) in the milk of cows. In the first study of the amount of SC in cow's milk 30 days after birth, it was found that the number of milk samples with a SC content of up to 200 thousand / ml in the experimental group was 10.0% higher compared to the control group, while samples containing SC from 201 up to 400 thousand / ml, were at the same level and amounted to 30%. From the second month of lactation, there was a gradual decrease in the number of milk samples with an SC content of 401 to 600 thousand / ml by 30.0% (from 40.0% to 10.0%). Moreover, in the first month of lactation, the average somatic cell content in the experimental and control groups was 323 thousand / ml and 346 thousand / ml. In the second month of lactation in the experimental group, the average number of somatic cells was 69 thousand / ml less than in the control, and amounted to 187 thousand / ml. The data obtained show a positive effect of the claimed pharmacological composition on the quality of milk in terms of somatic cells (table 7).
Так, количество проб молока с S. aureus и S. epidermalis сократилось в 2,3 и 4 раз, соответственно (таблица 8).The use of the pharmacological composition as a means of prevention and treatment of mastitis in the dry and postpartum period allowed to reduce the amount of bacteria and fungi sown from the contents of the mammary gland, which is an unobvious effect of the proposed method. So, the number of milk samples with S. aureus and S. epidermalis decreased by 2.3 and 4 times, respectively (table 8).
The calculation of the economic efficiency of the claimed pharmacological composition was carried out on the prevented damage by increasing productivity and product quality.

9.The data for calculating the economic efficiency of the use of the pharmacological composition for the prevention of diseases of the mammary gland of cows in the dry period in ZAO Agrofirma Patrusi are presented in table 9.
The use of a pharmacological composition is economically feasible. The economic effect per ruble of costs calculated on prevented damage due to the reduction of days with reduced productivity and reduced quality of milk is 1.7 rubles.
Thus, the obtained data indicate the effectiveness of the use of the pharmacological composition for simultaneously launching cows due to the use of silicon dimethylglycerolates and glycerolates as a basis, which have a reparative and regenerating effect, which allows, due to transcutaneous activity, to reduce the dose of doxycycline as an active substance, while maintaining its high effectiveness of action. As a result, the data obtained allow us to expand the range of drugs available in the Russian Federation for the treatment and prevention of mastitis in cows during the launch period, and also to use them to replace imports with goods produced in Russia.
As a result, this allows import substitution and the expansion of the country's range of domestic drugs and agents for the treatment and prevention of mastitis in cows during the start-up period.
