Abstract
Description
A known method of treatment and prevention of acute lactational mastitis in women using leukocyte suspensions for the treatment of patients with purulent lactational mastitis [Abstract of the dissertation - Yu.A. Spesivtseva, 1995]. The essence of the method consists in the transfusion (transfusion) of leukemia from donors immunized with staphylococcal toxoid. Transfusion was carried out twice with an interval of one week in a volume of from 100 to 600 ml. Yu.A. Spesivtsev concluded that transfusions of leucites from immunized donors containing stimulated phagocytic cells have a deliberately mediated immunocorrective effect on the cellular immunity system of patients with purulent lactation mastitis and are a highly effective means of preventing generalization of infection.
The disadvantages of the considered method are as follows: the presence of donors and the receipt of a large volume of leukocyte suspension; intravenous administration of the drug, which requires fixing the patient and a significant investment of time specialists. In addition, this method is expensive, requiring a high level of qualification of specialists, the availability of expensive equipment and reagents, and therefore it is almost not suitable for the treatment of acute lactation mastitis in cows in industrial production.
Курск, 2003).There is a method of treating mastitis using staphylococcal and streptococcal toxoids, which consists in introducing them intracisternally at a dose of 2.0 ml into the affected lobe (developed by the Scientific Research Institute of Agriculture, Kursk, 2003).
The disadvantage of this method is the possibility of trauma to the channel of the milk tank, a specialist is required to take an analysis, prepare toxoids and administer the resulting preparations. Thus, this method is expensive, time-consuming and not possible for implementation in sows due to the peculiarities of the anatomical structure of the nipple canal.
Известен наиболее близкий по технической сущности и решаемой задаче способ лечения острых лактационных маститов у коров стафилококковым анатоксином, наносимым в виде мази на поверхность молочного зеркала вымени коровы (Патент №2396089 - прототип).Known for the closest in technical essence and the problem being solved is a method of treating acute lactation mastitis in cows with a staphylococcal toxoid applied as an ointment to the surface of the milk mirror of the cow's udder (Patent No. 2396089 - prototype).
The objective of the invention is to increase the effectiveness of the treatment of acute lactational mastitis in sows and reduce the mortality of newborn piglets in industrial production, through the use of drugs with directed anti-staphylococcal effect.
The technical result of the task is achieved by the fact that in the method of treating acute lactational mastitis in sows, it is proposed to apply a thin layer to the skin of the emptied breast expressing skin with a composition containing, for example, dimexide in an amount of 1%, sodium laureth sulfate 85-95%, the rest is staphylococcal toxoid.
A method for the treatment of acute lactational mastitis in sows is as follows:
Before carrying out therapeutic manipulations, the inflamed mammary gland of the sow should be thoroughly washed and emptied. The ointment is applied in a thin layer on the affected packages of the mammary gland (from 1 to 4). The hands of a specialist serving a uterine livestock of pigs, when applying the composition, must be protected with latex gloves, since the active substance of the ointment passes through the skin's epithelial barrier.
The participation of a veterinarian is not required. The composition includes:
1. Staphylococcal toxoid obtained by the factory method [instructions for the use of TSA (antistaphylococcal drug), registration No. PVR 1.03.0154 - 96]. ASP provides stimulation of a specific immune response to various exotoxins and somatic antigens of staphylococci and provides rapid death of the pathogen in the body.
2. Sodium laureth sulfate is a surface-active substance (surfactant) in concentration and with additives included in the composition of liquid soap (in particular, My Sun soap manufactured by Avanta Rossiya OJSC can be used).
3. Dimexide (full name - dimethyl sulfoxide) - non-polar solvent has the ability to penetrate through biological membranes, including through skin barriers and mucous membranes without damaging them. It shows a pronounced local analgesic activity in pain syndromes of various etiologies, and has anti-inflammatory and antimicrobial effects. Dimexide acts as a conductor of the toxoid (antigenic determinant) inside the cell. We made a composition based on TSA with the addition of dimexide, which is prepared as follows: components are placed in a sterile glass vessel with a capacity of 0.5 liter in the sequence: anatoxin is added to the surfactant (you can use liquid soap “My Sun”), it is thoroughly mixed with a wooden spatula. Dimexide is poured into the resulting homogenized mixture slowly, with constant stirring, mixed thoroughly.
Composition Composition:
- Лауретсульфат с добавками (ПАВ) 85% (400 мл);- Laureth sulfate with additives (surfactant) 85% (400 ml);
- Стафилококковый анатоксин 14% (56 мл);- Staphylococcal toxoid 14% (56 ml);
In the case of mastitis in sows, which in most cases is caused by S.aureus, a composition with 14% toxoid content should be used; for other types of staphylococci, we recommend using a composition with 4% toxoid content. The composition is hermetically sealed and ready for use. It is stored in a cool place. The proposed composition has the properties of the activation of a specific immune response, the passage of the active substance of the TSA through intact skin, has analgesic, anti-inflammatory and antimicrobial effects.
The purpose of the study was to study the possibility of treating mastitis with the use of an anti-mastitis drug (AMP) based on staphylococcal toxoid by applying it in the area of the elevated lymph nodes.
3 groups were formed selected on the basis of analogues (live weight, age, origin). The first group included clinically healthy sows of the second, third and fourth farrowing. Three sows were pedigree (DYL, LYL, YLY), the weight of the animals ranged from 200-280 kilograms of live weight, the number of nipples from 12-14, the nipples were not damaged by piglets and could give colostrum to piglets. The second group included sows with clinical signs of mastitis of the second, third and fourth farrowing. Mastitis was of a catarrhal type — the general condition of the animals was unchanged, an increase in body temperature was extremely rare, the mammary gland was compacted, slightly increased in size, and soreness was weakly expressed. The third group consisted of sows with clinical signs of catarrhal mastitis of the second, third, and fourth farrowing, who were treated with staphylococcal toxoid, dimethyl sulfoxide (dimexide) and surfactant in the area of inguinal lymph nodes. Within 3 days, colostrum samples were taken by application to a glass slide, followed by fixation of colostrum cells, which was carried out with formaldehyde vapor. Investigated blood and colostrum. Blood tests were carried out on days 1 and 2 of the colostrum. Colostrum studies were performed on days 1,2 and 3 of the colostrum.
Due to the fact that the humoral immune response is provided by antibodies (AT), or immunoglobulins, we studied three classes of immunoglobulins A, M, G in the blood and colostrum of experimental sows. In addition, we studied the clinical picture of blood and leukocyte formula. According to hematological analysis, the action of a drug containing staphylococcal toxoid leads to an active migration of white blood cells into milk.
As a result of the study, it was found that the treatment of arterial lymph nodes of the AMP based on staphylococcal toxoid promotes more active production of immunoglobulins in the mammary gland and in the blood. An increase in the content of immunoglobulins is a powerful factor in humoral defense, and also contributes to the treatment of mastitis in sows and, as a result, the reduction of diarrhea in piglets. Along with this, the content of leukocytes, which carry out a protective and antimicrobial function, increases in milk.
