Abstract

Фармацевтическая композиция бупропиона гидробромида с модифицированным высвобождением включает ядро с терапевтически эффективным количеством бупропиона гидробромида и полимерное покрытие с регулируемым высвобождением. Покрытие содержит нерастворимый в воде полимер в количестве от 1% до 12% по весу от веса таблетки и растворимый в воде полимер в количестве от 1,5% до 10% по весу от веса таблетки. Указанное покрытие, по меньшей мере, частично окружает указанное ядро. Композиция высвобождает бупропиона гидробромид в среду растворения, состоящую из 0,1N HCl и 5-40 об.% этанола, со скоростью, меньшей или равной примерно 1,1 скорости высвобождения бупропиона гидробромида из аналогичной фармацевтической композиции с модифицированным высвобождением в среду растворения, состоящую из 0,1N HCl, измеренной в интервале времени, по меньшей мере, от 0 до 2 часов с помощью аппарата USP типа I при 75 об/мин и 37±5°С. Claims (9)

1. Фармацевтическая композиция бупропиона гидробромида с модифицированным высвобождением, включающая
ядро, содержащее терапевтически эффективное количество бупропиона гидробромида, и
полимерное покрытие с регулируемым высвобождением, содержащее нерастворимый в воде полимер в количестве от 1% до 12% по весу от сухого веса таблетки и растворимый в воде полимер в количестве от 1,5% до 10% по весу от сухого веса таблетки, при этом указанное покрытие, по меньшей мере, частично окружает указанное ядро; указанная композиция высвобождает бупропиона гидробромид в первую среду растворения, состоящую из 0,1N HCl и 5-40 об.% этанола, со скоростью, меньшей или равной примерно 1,1 скорости высвобождения бупропиона гидробромида из аналогичной фармацевтической композиции с модифицированным высвобождением во вторую среду растворения, состоящую из 0,1N HCl, измеренной в интервале времени, по меньшей мере, от 0 до 2 ч с помощью аппарата USP типа I при 75 об/мин и 37±5°С.
2. Фармацевтическая композиция с модифицированным высвобождением по п.1, при этом указанная композиция высвобождает бупропиона гидробромид в первую среду растворения, состоящую из 0,1N HCl и 5-40 об.% этанола, со скоростью, меньшей или равной скорости высвобождения бупропиона гидробромида из аналогичной композиции с модифицированным высвобождением во вторую среду растворения, состоящую из 0,1N HCl.
3. Фармацевтическая композиция с модифицированным высвобождением по п.2, при этом первая среда растворения состоит из 0,1N HCl и 5 об.% этанола.
4. Фармацевтическая композиция с модифицированным высвобождением по п.2, при этом первая среда растворения состоит из 0,1N HCl и 40 об.% этанола.
5. Фармацевтическая композиция с модифицированным высвобождением по п.1, при этом период времени составляет от 0 до 16 ч.
6. Фармацевтическая композиция с модифицированным высвобождением по п.1, при этом композиция представляет собой таблетку, содержащую 174 мг, 348 мг или 522 мг бупропиона гидробромида.
7. Фармацевтическая композиция бупропиона гидробромида с модифицированным высвобождением по п.1, при этом указанная композиция высвобождает бупропиона гидробромид в среду растворения, состоящую из 0,1N HCl и 40 об.% этанола со скоростью, меньшей скорости высвобождения бупропиона гидрохлорида из композиции бупропиона гидрохлорида с модифицированным высвобождением в аналогичную среду растворения, состоящую из 0,1N HCl и 40 об.% этанола, измеренной в интервале времени, по меньшей мере, от 0 до 2 ч с помощью аппарата USP типа I при 75 об/мин и 37±0,5°С;
при этом композиция бупропиона гидрохлорида с модифицированным высвобождением включает
ядро, содержащее бупропиона гидрохлорид в количестве, эквимолярном количеству бупропиона гидробромида в композиции бупропиона гидробромида с модифицированным высвобождением, и полимерное покрытие с регулируемым высвобождением, содержащее нерастворимый в воде и растворимый в воде полимеры, аналогичное применяемому в композиции бупропиона гидробромида, при этом покрытие, по меньшей мере, частично окружает ядро композиции бупропиона гидрохлорида с модифицированным высвобождением.
8. Способ уменьшения индуцированного алкоголем сброса дозы бупропиона гидробромида у пациента, включающий введение пациенту фармацевтической композиции с модифицированным высвобождением по п.1.
9. Способ по п.8, в котором композиция представляет собой таблетку, содержащую 174 мг, 348 мг или 522 мг бупропиона гидробромида.
