Abstract
Description
The invention relates to agriculture, in particular to veterinary medicine, and can be used for the prevention and treatment of staphylococcosis of animals and birds, accelerate wound healing and nonspecific immunostimulation.
A known method of producing a staphylococcal toxoid vaccine by growing followed by destruction of staphylococci, separation of the culture fluid, detoxification of it with formalin, sorption of the target product on aluminum hydroxide and its sterilization by autoclaving (RF Patent No. 2139091, MKI 6 A 61 K 39/085. Method for producing staphylococcus toxoid vaccines, Bull. No. 28, 1999). In the known method, the destruction is carried out by heating microbial cells to 85-100 ° C for 1.0-1.5 hours, formalin is added to the culture fluid twice from the calculation of 0.3-0.4% and 0.5-0.6% formalin after 7-10 days.
The disadvantage of this method is to obtain an toxoid vaccine of low quality (reactogenicity increases after a short storage time).
The aim of the invention is to improve the quality of the target product.
1:3-4.The goal is achieved in a method for producing a staphylococcal toxoid vaccine by growing staphylococci followed by their destruction, detoxification of the mixture with formalin and its sterilization by adding 1-1.5% glycerol solution to the target product in a volume ratio of 1: 3- 4.
В патентной и научно-технической литературе неизвестны технические решения, аналогичные заявляемому, что позволяет сделать вывод о соответствии предложения условию патентоспособности ″новизна″.In the patent and scientific and technical literature unknown technical solutions similar to the claimed, which allows us to conclude that the proposal meets the condition of patentability ″ novelty ″.
Only a combination of the claimed modes in a certain sequence allows to achieve the effect indicated in the purpose of the invention, i.e. the claimed method satisfies the condition of patentability "inventive step", in particular the addition of glycerol at certain ratios of components allowed to obtain an unobvious positive effect - a decrease in the reactogenicity of the target product without the use of chemical preservatives.
Способ используют для получения стафилококковой анатоксин-вакцины, используемой для профилактики стафилококкоза птиц, плотоядных, а также для профилактики и лечения маститов, пневмоний, ускорения заживления ран и неспецифической иммуностимуляции, поэтому заявленное предложение соответствует условию патентоспособности ″промышленная применимость″.The method is used to obtain staphylococcal toxoid vaccine used for the prevention of staphylococcosis of birds, carnivores, as well as for the prevention and treatment of mastitis, pneumonia, accelerate wound healing and nonspecific immunostimulation, therefore, the claimed proposal meets the patentability condition ″ industrial applicability ″.
The invention is illustrated by the following examples.
Example 1. Staphylococci are grown, as in the known method, on a nutrient medium for 10 days. Then autoclaving is carried out at 1.0 atm for 30 minutes. Detoxification is carried out by adding formalin to staphylococcus suspension in two stages: until it is concentrated in a mixture of 0.3% and incubated at 37 ° C for 7 days, followed by formalin to its final concentration of 0.5% and incubation of the mixture at a temperature 44 ° C for 5 days. At the end of the detoxification process, a 1% glycerol solution in a volume ratio of 1: 3 is added to a suspension of staphylococci and packaged in bottles, closed with rubber stoppers and wrapped in aluminum caps, autoclaved at 0.6 atm for 20 minutes.
