Claims (61)
1. Линкерное звено, содержащее:
ядро, содержащее 2-20 лизиновых (K) остатков, один или несколько спейсеров и первый конъюгирующий фрагмент, при этом:
любые два из остатков K прилегают друг к другу или разделены одним из спейсеров;
каждый из спейсеров независимо содержит (1) один или несколько не-K-аминокислотных остатков или (2) пегилированную аминокислоту, содержащую от 2 до 12 звеньев этиленгликоля (EG);
по меньшей мере один спейсер связан с N-концом первого остатка K, начиная с N-конца или с С-концом последнего остатка K, начиная с N-конца; и
первый конъюгирующий фрагмент содержит карбоксильную группу или аминогруппу и конъюгирующую группу, выбранную из азидной, алкиновой, тетразиновой, циклооктеновой и циклооктиновой групп, при этом:
если спейсер связан с N-концом первого остатка K, то первый конъюгирующий фрагмент содержит карбоксильную группу и связан со спейсером путем образования амидной связи с альфа-аминогруппой спейсера; или
если спейсер связан с С-концом последнего остатка K, то первый конъюгирующий фрагмент содержит аминогруппу и связан со спейсером путем образования амидной связи с карбоксильной группой спейсера; и
множество связывающих ветвей, при этом:
каждая из связывающих ветвей представляет собой пептид, содержащий от 2 до 12 не-K-аминокислотных остатков, или цепь полиэтиленгликоля (PEG), содержащую от 2 до 24 звеньев EG,
каждая из связывающих ветвей содержит реакционноспособную группу на одном конце и функциональную группу на другом конце и связана с остатками K ядра путем образования амидной связи между реакционноспособной группой связывающей ветви и аминогруппой остатка K ядра, и
функциональная группа выбрана из аминовой, карбоксильной, гидроксильной, трет-бутилдиметилсилиловой (TBDMS), N-гидроксисукцинимидиловой (NHS), малеимидной, винилсульфоновой, моносульфоновой, метилсульфонилбензотиазоловой, йодной, йодацетамидной, азидной, алкиновой, циклооктиновой, тетразиновой и циклооктеновой групп, при условии, что если конъюгирующая группа представляет собой азидную, алкиновую или циклооктиновую группу, то функциональная группа представляет собой тетразиновую или циклооктеновую группу, а если конъюгирующая группа представляет собой тетразиновую или циклооктеновую группу, то функциональная группа представляет собой азидную, алкиновую или циклооктиновую группу.
2. Линкерное звено по п. 1, в котором спейсер содержит один или несколько глициновых (G) остатков и/или сериновых (S) остатков.
3. Линкерное звено по п. 1, в котором реакционноспособная группа представляет собой сукцинимидиловый сложный эфир (SE), тетрафторфениловый сложный эфир (TFP) или карбоксильную группу.
4. Линкерное звено по п. 1, в котором ядро содержит два спейсера, связанных с N-концом первого остатка K и с С-концом последнего остатка K, соответственно, и ядро дополнительно содержит второй конъюгирующий фрагмент, содержащий карбоксильную группу или аминогруппу и конъюгирующую группу, выбранную из азидной, алкиновой, тетразиновой, циклооктеновой и циклооктиновой групп, при этом:
один из первого и второго конъюгирующих фрагментов связан с N-концом спейсера, соединенного с N-концом первого остатка K; и
другой из первого и второго конъюгирующих фрагментов связан с С-концом спейсера, соединенного с С-концом последнего остатка K.
5. Линкерное звено по п. 4, в котором:
функциональная группа представляет собой малеимидную, винилсульфоновую, моносульфоновую, йодную или йодацетамидную группу;
конъюгирующая группа одного из первого и второго конъюгирующих фрагментов представляет собой азидную, алкиновую или циклооктиновую группу; и
конъюгирующая группа другого из первого и второго конъюгирующих фрагментов представляет собой тетразиновую или циклооктеновую группу.
6. Линкерное звено по п. 1, в котором пептид связывающей ветви содержит от 5 до 10 аминокислотных остатков, независимо выбранных из остатков G, S, глутаминовой кислоты (Е) и аргинина (R).
7. Линкерное звено по п. 1, в котором:
циклооктеновая группа представляет собой норборненовую или транс-циклооктеновую (ТСО) группу;
циклооктиновая группа представляет собой дибензоциклооктиновую (DIBO), дифторциклооктиновую (DIFO), бициклонониновую (BCN) или дибензоазациклооктиновую (DIBAC) группу; или
тетразиновая группа представляет собой 1,2,3,4-тетразиновую, 1,2,3,5-тетразиновую или 1,2,4,5-тетразиновую группу или их производные.
8. Линкерное звено по п. 1, в котором ядро дополнительно содержит множество первых элементов, соответственно, связанных с множеством связывающих ветвей путем образования амидной или сложноэфирной связи между ними или путем тиол-малеимидной реакции, реакции SN2, реакции катализируемого медью азид-алкинового циклоприсоединения (CuAAC), промотируемой напряжением азид-алкиновой клик-реакции (SPAAC) или реакции Дильса-Альдера с обратными электронными требованиями (iEDDA).
9. Линкерное звено по п. 8, в котором ядро дополнительно содержит второй элемент, связанный с конъюгирующей группой путем реакции CuAAC, реакции SPAAC или реакции iEDDA.
10. Линкерное звено по п. 9, в котором:
каждый первый элемент представляет собой фрагмент первого антитела, специфичный к первому антигену клеточной поверхности; и
второй элемент представляет собой цитотоксическое лекарственное средство или фрагмент второго антитела, специфичный ко второму антигену клеточной поверхности.
11. Линкерное звено по п. 10, в котором первый антиген клеточной поверхности выбран из CD5, CD19, CD20, CD22, CD23, CD27, CD30, CD33, CD34, CD37, CD38, CD43, CD72a, CD78, CD79a, CD79b, CD86, CD134, CD137, CD138 и CD319; или вторым антигеном клеточной поверхности является CD3 или CD16a.
12. Линкерное звено по п. 9, в котором:
каждый из первых элементов представляет собой пептидный гормон, первый фактор роста или фрагмент первого антитела, специфичный к опухоль-ассоциированному антигену; и
второй элемент представляет собой цитотоксическое лекарственное средство, агонист толл-подобных рецепторов, хелатор, образующий комплекс с радиоактивным нуклидом, цитокин или фрагмент второго антитела, специфичный ко второму фактору роста, антигену клеточной поверхности, гаптену или цитокину.
13. Линкерное звено по п. 12, в котором:
первый фактор роста выбран из эпидермального фактора роста (EGF), мутантного EGF, эпирегулина, гепарин-связывающего эпидермального фактора роста (HB-EGF), фактора роста эндотелия сосудов A (VEGF-A), основного фактора роста фибробластов (bFGF) и фактора роста гепатоцитов (HGF);
второй фактор роста выбран из EGF, мутантного EGF, VEGF-A, bFGF и HGF;
пептидный гормон выбран из секретина, холецистокинина (CCK), соматостатина, октреотида и тиреотропного гормона (TSH);
опухоль-ассоциированный антиген выбран из рецептора эпидермального фактора роста (HER1, HER2, HER3, HER4), углеводного антигена 19-9 (СА 19-9), углеводного антигена 125 (СА 125), карциноэмбрионального антигена (СЕА), муцина 1 (MUC 1), ганглиозида GD2, меланом-ассоциированного антигена (MAGE), простат-специфического мембранного антигена (PSMA), антигена стволовых клеток простаты (PSCA), мезотелина, родственного муцину Tn, сиалированного Tn, глобо-Н, стадиеспецифического эмбрионального антигена-4 (SSEA-4) и адгезивной молекулы эпителиальных клеток (ЕрСАМ); или
антиген клеточной поверхности выбран из CD3, CD16a, CD28, CD134, цитотоксического Т-лимфоцит-ассоциированного белка 4 (CTLA-4), белка 1 программируемой смерти клеток (PD-1) и первого лиганда белка 1 программируемой смерти клеток (PD-L1).
14. Линкерное звено по п. 5, в котором ядро дополнительно содержит:
множество первых элементов, соответственно, связанных с множеством связывающих ветвей путем образования амидной либо сложноэфирной связи между ними или путем тиол-малеимидной реакции либо реакции SN2; и
второй элемент и третий элемент, соответственно связанные с конъюгирующими группами первого и второго конъюгирующих фрагментов, причем один из второго и третьего элементов связан с одной конъюгирующей группой путем реакции iEDDA, а другой из второго и третьего элементов связан с другой конъюгирующей группой путем реакции SPAAC или реакции CuAAC.
15. Молекулярная конструкция, содержащая первое и второе линкерные звенья, независимо охарактеризованные в любом из пп. 1-8, в которой первое и второе линкерные звенья связаны реакцией между конъюгирующими группами первого и второго линкерных звеньев путем реакции CuAAC, реакции SPAAC или реакции iEDDA.
16. Молекулярная конструкция по п. 15, дополнительно содержащая множество первых элементов и множество вторых элементов, соответственно связанных со множеством связывающих ветвей первого линкерного звена и второго линкерного звена путем образования амидной либо сложноэфирной связи между ними или путем тиол-малеимидной реакции, реакции SN2, реакции CuAAC, реакции SPAAC либо реакции iEDDA.
17. Молекулярная конструкция по п. 16, в которой:
каждый первый элемент представляет собой фрагмент первого антитела, специфичный к первому антигену клеточной поверхности; и
каждый второй элемент представляет собой цитотоксическое лекарственное средство или фрагмент второго антитела, специфичный ко второму антигену клеточной поверхности.
18. Молекулярная конструкция по п. 17, в которой:
первый антиген клеточной поверхности выбран из CD5, CD19, CD20, CD22, CD23, CD27, CD30, CD33, CD34, CD37, CD38, CD43, CD72a, CD78, CD79a, CD79b, CD86, CD134, CD137, CD138 и CD319; или
вторым антигеном клеточной поверхности является CD3 или CD16a.
19. Молекулярная конструкция по п. 16, в которой:
каждый из первых элементов представляет собой пептидный гормон, первый фактор роста или фрагмент первого антитела, специфичный к опухоль-ассоциированному антигену; и
каждый из вторых элементов представляет собой цитотоксическое лекарственное средство, агонист толл-подобных рецепторов, хелатор, образующий комплекс с радиоактивным нуклидом, цитокин или фрагмент второго антитела, специфичный ко второму фактору роста, антигену клеточной поверхности, гаптену или цитокину.
20. Молекулярная конструкция по п. 19, в которой:
первый фактор роста выбран из эпидермального фактора роста (EGF), мутантного EGF, эпирегулина, гепарин-связывающего эпидермального фактора роста (HB-EGF), фактора роста эндотелия сосудов A (VEGF-A), основного фактора роста фибробластов (bFGF) и фактора роста гепатоцитов (HGF);
второй фактор роста выбран из EGF, мутантного EGF, VEGF-A, bFGF и HGF;
пептидный гормон выбран из секретина, холецистокинина (CCK), соматостатина, октреотида и тиреотропного гормона (TSH);
опухоль-ассоциированный антиген выбран из рецептора эпидермального фактора роста (HER1, HER2, HER3, HER4), углеводного антигена 19-9 (СА 19-9), углеводного антигена 125 (СА 125), карциноэмбрионального антигена (СЕА), муцина 1 (MUC 1), ганглиозида GD2, меланома-ассоциированного антигена (MAGE), простат-специфического мембранного антигена (PSMA), антигена стволовых клеток простаты (PSCA), мезотелина, родственного муцину Tn, сиалированного Tn, глобо-Н, стадиеспецифического эмбрионального антигена-4 (SSEA-4) и адгезивной молекулы эпителиальных клеток (ЕрСАМ); или
антиген клеточной поверхности выбран из CD3, CD16a, CD28, CD134, цитотоксического Т-лимфоцит-ассоциированного белка 4 (CTLA-4), белка 1 программируемой смерти клеток (PD-1) и первого лиганда белка 1 программируемой смерти клеток (PD-L1).
