Claims (89)
1. Способ отбора образца, содержащего T-клетки, для адоптивной иммунотерапии, где указанный способ включает определение уровня экспрессии любого из гранзима B, гранзима K, перфорина, CD107a, IFNg, IL-2, TNF или любой комбинации перечисленных, лимфоцитами в образце и отбор образца для адоптивной иммунотерапии, если по меньшей мере пороговый уровень лимфоцитов в образце экспрессирует любой из гранзима B, гранзима K, перфорина, CD107a, IFNg, IL-2, TNF или любую комбинацию перечисленных.
2. Способ отбора образца, содержащего T-клетки, для включения в банк клеток, где указанный способ включает определение уровня экспрессии любого из гранзима B, гранзима K, перфорина, CD107a, IFNg, IL-2, TNF или любой комбинации перечисленных, лимфоцитами в образце и отбор образца для включения в банк клеток, если по меньшей мере пороговый уровень лимфоцитов в образце экспрессирует любой из гранзима B, гранзима K, перфорина, CD107a, IFNg, IL-2, TNF или любую комбинацию перечисленных.
3. Способ отбора образца, содержащего T-клетки, из библиотеки образцов для адоптивной иммунотерапии, где указанный способ включает скрининг библиотеки образцов на экспрессию любого из гранзима B, гранзима K, перфорина, CD107a, IFNg, IL-2, TNF или любой комбинации перечисленных, лимфоцитами в образцах, и отбор образца из библиотеки образцов для адоптивной иммунотерапии, где по меньшей мере пороговый уровень лимфоцитов в образце экспрессирует любой из гранзима B, гранзима K, перфорина, CD107a, IFNg, IL-2, TNF или любую комбинацию перечисленных.
4. Способ отбора образцов, содержащих T-клетки, из библиотеки образцов для включения в банк клеток, где указанный способ включает скрининг библиотеки образцов на экспрессию любого из гранзима B, гранзима K, перфорина, CD107a, IFNg, IL-2, TNF или любой комбинации перечисленных, лимфоцитами в образцах, и отбор образцов для их включения в банк клеток, где по меньшей мере пороговый уровень лимфоцитов в образце экспрессирует любой из гранзима B, гранзима K, перфорина, CD107a, IFNg, IL-2, TNF или любую комбинацию перечисленных.
5. Способ отбора образца, содержащего T-клетки для размножения ex vivo, где указанный способ включает
(a) индуцирование пролиферации T-клеток из части образца,
(b) определение уровня экспрессии любого из гранзима B, гранзима K, перфорина, CD107a, IFNg, IL-2, TNF или любой комбинации перечисленных, лимфоцитами в части образца; и
(c) отбор образца для размножения, если по меньшей мере пороговый уровень лимфоцитов экспрессирует любой из гранзима B, гранзима K, перфорина, CD107a, IFNg, IL-2, TNF или любую комбинацию перечисленных.
6. Способ по п. 5, также включающий отбор образца для включения в банк клеток.
7. Способ по п. 5, также включающий отбор образца для адоптивной иммунотерапии.
8. Способ по любому из пп. 1-7, где пороговый уровень для CD107a составляет 0,5%.
9. Способ по любому из пп. 1-7, где пороговый уровень для CD107a составляет 1%.
10. Способ по любому из пп. 1-7, где пороговый уровень для CD107a составляет 2%.
11. Способ по любому из пп. 1-7, где пороговый уровень для IFNg составляет по меньшей мере 0,5%.
12. Способ по п. 11, где пороговый уровень для IFNg составляет по меньшей мере 1%.
13. Способ по п. 11, где пороговый уровень для IFNg составляет по меньшей мере 2%.
14. Способ по любому из пп. 1-7, где пороговый уровень для IL-2 составляет по меньшей мере 0,5%.
15. Способ по п. 14, где пороговый уровень для IL-2 составляет по меньшей мере 1%.
16. Способ по п. 14, где пороговый уровень для IL-2 составляет по меньшей мере 2%.
17. Способ по п. 14, где пороговый уровень для IL-2 составляет по меньшей мере 10%.
18. Способ по любому из пп. 1-7, где пороговый уровень для TNF составляет по меньшей мере 0,5%.
19. Способ по п. 18, где пороговый уровень для TNF составляет по меньшей мере 1%.
20. Способ по п. 19, где пороговый уровень для TNF составляет по меньшей мере 2%.
21. Способ по любому из пп. 1-7, где пороговый уровень для гранзима B составляет по меньшей мере 5%.
22. Способ по п. 21, где пороговый уровень для гранзима B составляет по меньшей мере 10%.
23. Способ по п. 21, где пороговый уровень для гранзима B составляет по меньшей мере 15%.
24. Способ по п. 21, где пороговый уровень для гранзима B составляет по меньшей мере 20%.
25. Способ по любому из пп. 1-7, где пороговый уровень для гранзима K составляет по меньшей мере 3%.
26. Способ по п. 25, где пороговый уровень для гранзима K составляет по меньшей мере 5%.
27. Способ по п. 25, где пороговый уровень для гранзима K составляет по меньшей мере 8%.
28. Способ по п. 25, где пороговый уровень для гранзима K составляет по меньшей мере 10%.
29. Способ по п. 25, где пороговый уровень для гранзима K составляет по меньшей мере 15%.
30. Способ по любому из пп. 1-7, где пороговый уровень для перфорина составляет по меньшей мере 2,0%.
31. Способ по п. 30, где пороговый уровень для перфорина составляет по меньшей мере 5%.
32. Способ по п. 30, где пороговый уровень для перфорина составляет по меньшей мере 8%.
33. Способ по п. 30, где пороговый уровень для перфорина составляет по меньшей мере 10%.
34. Способ по любому из пп. 1-7, где экспрессию любого из гранзима B, гранзима K, перфорина, CD107a, IFNg, IL-2, TNF или комбинации перечисленных определяют с помощью клеточного сортинга с активацией флуоресценции (FACS) или ELISpot.
35. Способ по любому из пп. 1-7, где T-клетки инкубируют с антигенпрезентирующими клетками для индуцирования пролиферации T-клеток до определения уровня экспрессии гранзима B, гранзима K, перфорина, CD107a, IFNg, IL-2 или TNF лимфоцитами.
36. Способ по любому из пп. 1-7, также включающий определение уровня экспрессии CD8 клетками в образце и отбор образца, если по меньшей мере 20% клеток в образце экспрессируют CD8.
37. Способ по п. 36, где образец отбирают, если по меньшей мере 25% клеток экспрессируют CD8.
38. Способ по п. 36, где образец отбирают, если по меньшей мере 30% клеток экспрессируют CD8.
39. Способ по любому из пп. 1-7, также включающий определение уровня экспрессии CD4 клетками в образце и отбор образца, если по меньшей мере 10% клеток экспрессируют CD4.
40. Способ по п. 39, где образец отбирают, если по меньшей мере 15% клеток в образце экспрессируют CD4.
41. Способ по п. 39, где образец отбирают, если по меньшей мере 25% клеток в образце экспрессируют CD4.
42. Способ по любому из пп. 1-7, где отобранные T-клетки хранят в банке клеток после отбора.
43. Способ по любому из пп. 1-7, где T-клетки экспрессируют TCR, специфичный к пептиду вируса Эпштейна-Барра, презентированному на МНС класса I.
44. Способ по п. 43, где пептид EBV содержит пептид LMP1.
45. Способ по п. 43, где пептид EBV содержит пептид LMP2A.
46. Способ по п. 43, где пептид EBV содержит пептид EBNA1.
47. Способ по любому из пп. 1-7, также включающий введение отобранных T-клеток индивидууму, нуждающемуся в этом.
48. Способ по п. 47, где индивидуум, нуждающийся в этом, страдает раком.
49. Способ по п. 48, где раком является рак, ассоциированный с вирусом Эпштейна-Барра.
50. Способ по п. 49, где индивидуум имеет карциному носоглотки, NK/T-клеточную лимфому, карциному желудка, ассоциированную с вирусом Эпштейна-Барра, или лейомиосаркому, ассоциированную с вирусом Эпштейна-Барра.
51. Способ по п. 50, где индивидуум, нуждающийся в этом, имеет лимфопролиферативное расстройство после трансплантации.
52. Способ по п. 51, где индивидуум имеет аутоиммунное расстройство.

61. Банк клеточных образцов, включающих T-клетки, где банк клеток обогащен образцами, в которых по меньшей мере пороговый уровень лимфоцитов в образце экспрессирует биомаркер, где пороговый уровень лимфоцитов, экспрессирующих биомаркер, представляет собой уровень, при котором:
(a) по меньшей мере 1% лимфоцитов в образце экспрессируют CD107a;
(b) по меньшей мере 1% лимфоцитов в образце экспрессируют IFNg;
(с) по меньшей мере 1% лимфоцитов в образце экспрессируют IL-2;
(d) по меньшей мере 1% лимфоцитов в образце экспрессируют TNF;
(e) по меньшей мере 5% лимфоцитов в образце экспрессируют гранзим B;
(f) по меньшей мере 3% лимфоцитов в образце экспрессируют гранзим K;
(h) по меньшей мере 2% лимфоцитов в образце экспрессируют перфорин;
(i) по меньшей мере 20% лимфоцитов в образце экспрессируют CD8;
(j) по меньшей мере 10% лимфоцитов в образце экспрессируют CD4;
(k) по меньшей мере 5% лимфоцитов в образце экспрессируют PD-1;
(l) по меньшей мере 8% лимфоцитов в образце экспрессируют TIM-3;
(m) по меньшей мере 3% лимфоцитов в образце экспрессируют LAG-3; или
(n) по меньшей мере 8% лимфоцитов в образце экспрессируют CTLA-4.
62. Банк клеток по п. 61, где банк клеток обогащают образцами, в которых по меньшей мере пороговый уровень лимфоцитов в образце экспрессирует два или более биомаркеров.

65. Банк клеток по любому из пп. 61-64, где по меньшей мере 25% образцов в банке клеток имеет по меньшей мере пороговый уровень лимфоцитов в образце, экспрессирующих биомаркер.
66. Банк клеток по любому из пп. 61-64, где по меньшей мере 50% образцов в банке клеток имеет по меньшей мере пороговый уровень лимфоцитов в образце, экспрессирующих биомаркер.
67. Банк клеток по любому из пп. 61-64, где по меньшей мере 75% образцов в банке клеток имеет по меньшей мере пороговый уровень лимфоцитов в образце, экспрессирующих биомаркер.
68. Банк клеток по любому из пп. 61-64, где по меньшей мере 100% образцов в банке клеток имеет по меньшей мере пороговый уровень лимфоцитов в образце, экспрессирующих биомаркер.
69. Банк клеток по любому из пп. 61-64, где T-клетками являются CD8-T-клетки.
70. Банк клеток по п. 69, где CD8-T-клетками являются цитотоксические T-лимфоциты (CTL).
71. Банк клеток по п. 70, где CTL экспрессируют IFNg, TNF, IL-2 и CD107.
72. Банк клеток по любому из пп. 61-64, где T-клетками являются CD4-T-клетки.
