Claims (16)
1. Способ лечения дисфункции или состояния печени, включающий
(a) получение суспензии печеночных стволовых клеток/клеток-предшественников; и
(b) введение этой суспензии в стенки или на стенки билиарного дерева субъекту с дисфункцией или состоянием печени; где значительная часть клеток приживается в стенке билиарного дерева,
где эти клетки созревают, становясь функциональными клетками печени, и мигрируют в печень, тем самым осуществляя лечение дисфункции или состояния печени.
2. Способ по п.1, в котором клетки являются стволовыми клетками билиарного дерева или стволовыми клетками печени, или коммитированными клетками-предшественниками, происходящими от указанных стволовых клеток.
3. Способ по п.1, в котором суспензию объединяют с одним или несколькими биологическими материалами для образования матриксного комплекса.
4. Способ по п.3, в котором один или несколько биологических материалов включают коллагены, молекулы адгезии, протеогликаны, гиалуронаны, цепи гликозаминогликанов, хитозан, альгинат и синтетические, биодеградируемые и биологически совместимые полимеры, или их комбинации.
5. Способ по п.4, в котором факторы роста могут включать одно или несколько из следующих веществ: факторы роста фибробластов (FGF), R-спондин, фактор роста гепатоцитов (HGF), эпидермальный фактор роста (EGF), фактор роста эндотелия сосудов (VEGF), инсулиноподобный фактор роста I (IGF-1), инсулиноподобный фактор роста II (IGF-2), онкостатин-M, фактор, ингибирующий лейкоз (LIF), трансферрин, инсулин, глюкокортикоиды, гормоны роста, эстрогены, андрогены, гормоны щитовидной железы, гормоны гипофиза и комбинации этих веществ.
6. Способ по п.1, в котором суспензию комбинируют с факторами роста, дополнительными клетками, или их комбинациями.
7. Способ по п.1, в котором клетки получают из здорового или функционального участка билиарного дерева субъекта.
8. Способ по п.1, в котором клетки получают из билиарного дерева не-аутологичного донора.
9. Способ по п.1, в котором суспензию вводят путем лапароскопического хирургического вмешательства или путем эндоскопии.
10. Способ по п.1, в котором суспензию клеток вводят путем инъекции, в качестве пластыря с биодеградируемым покрытием, или в губке.
11. Способ восстановления функций печени у субъекта с заболеванием или дисфункцией печени, включающий
(а) получение суспензии печеночных стволовых клеток/клеток-предшественников; и
(b) введение этой суспензии в стенки или на стенки желчных протоков поблизости от печени субъекта; и где значительная часть введенных клеток приживается по меньшей мере в части или на части печени в виде зрелых печеночных клеток in vivo.
