Claims (3)
- REIVINDICAÇÕES- lâ Processo para a obtenção de células hospedeiras recombinantes e sua progénie, caracterizado pelo facto de as células obtidas serem células de microorganismos, plantas ou animais contendo, além de sequências de controlo apropriadas, ADN heterólogo codificando proteínas ou do grupo que compreende proteínas que são funcionais, sozinhas ou em cooperação com uma ou mais proteínas adicionais, para catalizar uma operação de oxi dação no processo biológico para a transformação de colesterol em hidrocortisona, operação essa que é escolhida do grupo que consiste em- a transformação de colesterol em pregnolona;- a transformação de pregnolona em progesterona;- a transformação de progesterona em 17OC-hidroxi-progesterona;- a transformação de 17o<-hidroxi-progesterona em cortexolona;- a transformação de cortexolona em hidrocortisona, ou do grupo que compreende as proteínas naturais do processo bio lógico para a transformação de colesterol em hidrocortisona e são- enzima de clivagem da cadeia lateral (P^^qSCC);- adrenodoxina (ADX);- adrenodoxina redutase (ADR);- 3^-hidroxi-esteróide desidrogenase/isomerase (3^3-HED);- esteróide-170C-hidroxilase (P.cn17);- NADPH citocrómio Ρ^θ redutase (RED);- esteróide-21-hidroxilase (Ρ4^θΟ21); e- esteróide-11 β -hidroxilase (Ρ^θΐΐ) , codificando o ADN pelo menos duas proteínas expressáveis que catalisam pelo menos duas operações de oxidação separadas.-
- 2â 23- 2ã Processo para a obtenção de células hospedeiras recombinantes, de acordo com a reivindicação 1, caracte rizado pelo facto de o ADN que codifica sequência ser originado pela espécie bovina.- 3â Processo para a obtenção de células recombinantes, de acordo com as reivindicações 1 ou 2, caracteriza do pelo facto de o ADN heterólogo codificar pelo menos as prote_í nas Ρ45017βί θ P450C21‘- 42 Processo para a obtenção de células hospedeiras recombinantes e da sua progénie, de acordo com qualquer das reivindicações 1 a 3, caracterizado pelo facto de o hospede^ ro ser um microrganismo.- 5ã Processo para a obtenção de células hospedeiras recombinantes e da sua progénie, de acordo com a reivin dicação 4, caracterizado pelo facto de o hospedeiro ser uma espé cie de Saccharomyces, Kluyveromyces ou Bacillus ou ser Escherichia coli.- 6§ 24- 6â Processo para a preparação de uma mistura de proteínas por meios de células hospedeiras recombinantes compreendendo cultivarem-se células do hospedeiro recombinante num meio nutritivo sob condições que permitem que as proteínas se formem e se acumulem na cultura, caracterizado pelo facto de o hospedeiro recombinante ser como se define em uma qualquer das reivindicações 1 a 5.- 7S Processo para a oxidação selectiva múlt_i pia de um composto in vitro, compreendendo a incubação do compos^ to a ser oxidado na presença de pelo menos duas proteínas sob condições que permitem a referida oxidação e a acumulação do com posto oxidado no líquido de cultura, seguida pela recuperação do composto oxidado, caracterizado pelo facto de as proteínas terem sido produzidas pelo processo de acordo com a reivindicação 6.- 8â Processo para a oxidação múltipla selectiva de um composto, compreendendo cultivar-se células hospedeiras recombinantes na presença do mencionado composto sob condições em que a pretendida oxidação ocorre e o composto oxidado se acumula no caldo de cultura, seguida pela recuperação do composto oxidado, caracterizado pelo facto de as células hospedeiras recombinantes serem as células de acordo com qualquer das reivin dicações 1 a 5.- 93 Processo de acordo com as reivindicações 7 ou 8, caracterizado pelo facto de se realizarem duas operações de oxidação escolhidas do grupo que consiste na clivagem da cadeia lateral de um composto de esterol, oxidação e isomerização do grupo 3-hidroxi-5(6)-desidro de maneira a obter-se um grupo
- 3-oxo-4(5)-desiro, introdução de um grupo 17 -hidroxilo, um gru po 21-hidroxilo ou um grupo 11/3-hidroxilo.- 10â Processo de acordo com as reivindicações 7 ou 8, caracterizado pelo facto de a oxidação ter como resultado a introdução simultânea de um grupo C21-hidroxilo assim como de um grupo Cl7-hidroxilo e pregnenolona ou progesterona.- lis Processo para a obtenção de uma cassete de expressão, operável num hospedeiro recombinante de acordo com qualquer das reivindicações 1 a 5, contendo pelo menos dois genes que codificam as proteínas como se definiram na reivindicação 1.- 12ã Processo para a obtenção de uma cassete de expressão, de acordo com a reivindicação 11, caracterizado pe lo facto de o ADN heterólogo codificar as enzimas P45Q-17alfa e P45QC21 e a cassete de expressão ser pGB!7alfa/C21-l.133Processo para a preparação de composições farmacêuticas, caracterizado pelo facto de se incorporar co mo ingrediente activo um composto que foi preparado de acordo com qualquer das reivindicações 9 a 12, nas substâncias veiculares e/ou auxiliares farmaceuticamente aceitáveis.A requerente reivindica a prioridade do pedido de patente Europeia apresentado em 23 de Setembro de 1988, sob o nO. 88202080.3.
