제1도는 포유동물의 부신피질에서 발생되는, 콜레스테롤에서 히드록코르티손으로의 변환에 있어서 연속적 단계에 속하는 단백질에 대한 개략도이고, 제2도는 발현 카세트 pGB17α-5의 물질적 지도를 도시하며, 제3도는 발현 카세트 pGBC21-9의 물질적 지도를 도시한다.
Claims (13)
- 콜레스테롤을 히드로코르티손으로 변화시키기 위한 생물학적 경로가, 콜레스테롤을 프레그넨올론으로 변환시키는 단계; 프로그넨올론을 프로게스테론으로 변환시키는 단계; 프로게스테론을 17α-히드록시프로게스테론으로 변환시키는 단계; 17α-히드록시 프로게스테론을 코르텍솔론으로 변환시키는 단계; 코르텍솔론을 히드로코르티손으로 변환시키는 단계; 로 구성되는 군으로부터 선택되고, 적당한 지배서열 이외에도, 산화단계에 촉매작용을 하며 단독으로나 하나 또는 그 이상의 추가적 단백질과 공동으로 기능적인 단백질로 이루어지는군, 또는 콜레스테롤을 히드로코르티손으로 변환시키기 위한 생물학적 경로의 단백질인 곁사슬 분해효소(P4 5 0SCC); 아드레노독신(ADX); 아드레노독신 환원효소(ADR);3β-히드록시스테로이드 탈수소효소/이소메라제(3β-HSD); 스테로이드-17α-히드록실라아제(P4 5 0C21); 및 스테로이드-11β-히드록실라아제(P4 5 011β);로 이루어지는 군으로부터 단백질을 코딩하는 이형구조 DNA가 함유된 미생물 식물 또는 동물세포로 이루어지며, 상기 이형구조 DNA는 최소한 2개의 별도의 산화단계를 촉매하는 최소한 2개의 발현 가능 단백질을 코드화하는 것을 특징으로 하는 재조합 숙주세포 및 이것의 자세포.
- 제1항에 있어서, 상기 이형구조 DNA 코드서열이 소에서 유래하는 것을 특징으로 하는 재조합 숙주세포.
- 제1항 또는 2항에 있어서, 상기 이형구조 DNA가 P4 5 017α효소를 코드화하는 것을 특징으로 하는 재조합 숙주세포.
- 제1항 내지 3항중의 어느 한 항에 있어서, 상기 숙주가 미생물인 것을 특징으로 하는 재조합 숙주세포 또는 이것의 자세포.
- 제4항에 있어서,상기 숙주가 삭카로마이세스, 클뤼베로마이세스 또는 바실루스 종이거나 대장균인 것을 특징으로 하는 재조합 숙주세포 또는 이것의 자세포.
- 단백질이 형성되어 배양물에 축적될 수 있도록 하는 조건하에 재조합 숙주의 세포를 영야배지에서 배양하는 것으로 이루어지는 재조합 숙주 세포에 의한 단백질 호합물의 제조에 있어서, 상기 재조합 숙주세포가 제1항 내지 5항중의 어느 한 항에 정의된 바와같은 것을 특징으로 하는 방법.
- 화합물을 체외에서 선택적 다중 산화시키기 위하여, 산화될 화합물을 배양액내에서 산화된 화합물이 상기 산화 및 축적될 수 있는 조건에서 적어도 2개의 단백질의 존재하에 산화되도록 인큐베이션 하는 단계; 및 산화된 화합물을 회수하는 단계로 이루어지며, 상기 단백질이 제6항의 방법에 의해 제조된 것을 특징으로 하는 방법.
- 화합물의 선택적 다중 산화를 위하여, 목적 산화반응이 발생하고 산화된 화합물이 배양조에 축적되는 조건하에서 상기 화합물의 존재하에 재조합 숙주세포를 배양하는 단계; 및 산화된 화합물을 회수하는 단계로 이루어지며, 재조합 숙주세포가 제1항 내지 5항중의 어느 한 항의 세포인 것을 특징으로 하는 방법.
- 제7항 또는 8항에 있어서, 스테롤 화합물의 곁사슬 절단, 3-히드록시,5(6)-데히드로기의 3-옥소, 4(5)-데히드로기에로의 산화 및 이성질화, 17α-히드록실기, 21-히드록실기 또는 11β-히드록실기의 도입으로 이루어지는 군으로부터 선택된 최소한 2개의 산화 단계가 수행되는 것을 특징으로 하는 방법.
- 제7항 또는 8항에 있어서, 산화작용이 C17α-히드록실기 뿐만아니라 C21-히드록실기를 동시에 프레그넨올론이나 프로게스테론으로 도입하는 것을 특징으로 하는 방법.
- 제1항 내지 5항중의 어느 한 항에 따른 재조합 숙주내에서 조작가능하고 제1항에 정의된 단백질을 코딩하는 유전자가 적어도2개 함유된 것을 특징으로 하는 발현카세트.
- 제11항에 있어서, 상기 이종구조 DNA가 P4 5 0-17알파P4 5 0C21 효소를 코드화하고 상기 발현 카세트가 pGB17알파/C21-1 인 것을 특징으로 하는 발현카세트.
- 활성화합물을 함유하고 있으며 이 화합물이 제9 내지 12항중의 어느 한 항에 따라 제조되는 것을 특징으로 하는 약학 제조물.※ 참고사항 : 최초출원 내용에 의하여 공개하는 것임.
