Abstract

En nukleotidsekvens kodende for karbamoylfosfatsyntetase II til Plasmodium falciparum er beskrevet. Karbamoylfosfatsymetase II katalyserer det første rate- begrensede trinnet i de novo pyrimidin biosyntesereaksjonsveien. P.falciparum er avhengig av pyrimidin syntese de novo på grunn av dets manglende evne til overføring av pyrimidiner. Modne humane røde blodceller er ikke et anerkjent krav på et pyridimidinnukleotid. Dette enzymet representerer følgelig et primært kjemoterapeutisk lokus. Anvendelse av sekvensen kodende for karbamoylfosfat- syntetase II i rekombinant produksjon av karbamoylfosfatsyntetase II og anti- sensmolekyler, ribozymer og andre geninaktiveringsmidler konstruert fra denne sekvensen, er også beskrevet.
