Description
Foreliggende oppfinnelse omhandler en ny fremgangsmåte for framstilling av alfa-L-aspartyl-L-fenylalanin alkylestere av formel I
Hvor R representere en alkylgruppe med fra 1 til 5 karbon-atomer som starter fra den korresponderende alfa-halo-acyl-alfa-L-aspartyl-L-fenylalanin alkylestere (11) i henhold til følgende reaksjonsskjerna:

R er som ovenfor definert;
Ar er fenylgruppen;
R<1>representere hydrogen eller en C^-C^lineær eller forgrenet alkylgruppe;
Hal representerer klor, brom eller jod;
Y er en forbindelse valgt fra gruppen bestående av tiourea, cyansyresalter, tiocyansyresalter, di ti okarbanatsal ter, guanidin, 1,2-etanditiol, systamin, etanolamin, atylendiamin, 1,2-diaminobenzen, 1,2-dimerkaptobenzen, pyrokatekol, o-amino-fenol.
I en foretrukket utførelse av fremgangsmåten ifølge oppfinnelsen representerer R<1>hydrogen eller en metylgruppe; Hal
er klor eller brom; Y er tioyrea.
I en spesielt foretrukket form fremskaffer foreliggende oppfinnelse en fremgangsmåte for fremstilling av alfa-L-aspartyl-L-f enylalanin metylester (I, hvor R = CH^) eller aspar tan.
Aspartan er et velkjent søtningsstoff mye brukt som diete-tisk søtningsmiddel p.g.a. sin smak som er svært lik den til sakkarose.
Dipeptider av formel (I) er kjent a være fremstilt ved å starte fra L-asparaginsyrederivater. Ifølge en kjent metode utføres fremgangsmåten ved å starte fra L-asparaginsyre hvor både -NH2og ( 2> - og alfa-karboksylgruppene har blitt blokkert ved hjelp av f.eks. benzyloksykarbonyl eller formyl eller 4-ni trofenylestergrupper: ved reaksjon med en L-fenyl-alaninester fjernes 4-nitrofenylgruppen og det blokkerte dipeptid erholdes hvorfra de andre blokker1ngsgruppene kan fjernes ved slike konvensjonelle teknikker som hydrogenolyse eller hydrolyse.
Imidlertid er selektiv es ter1 f1 ser1ng av (i -karboksylsyre-gruppen av L-asparaginsyre vanskelig å utføre og dessuten gir den lavt utbytte; på den annen side er det for å produ-sere aspartan nødvendig at kondensasjon finner sted kun på alfa-kaboksylsyregruppen.
I henhold til en annen kjent metode benyttes et N-blokkert L-asparaginsyreanhydrid som startmateriale imidlertid gir kondesasjon av nevnte anhydrid med aminosyrer eller ammo-syreestere (d.v.s. L-fenylalaninmetylester) fulgt av fjerning av den blokkerende gruppen, alltid endeprodukter som inneholder små men ikke neglisjerbare mengder av alfa-L-alpartyl-L-fenylalanin og av (2-bezyl-3,6-diokso-piperasin-5-y1)eddiksyre (de såkalte "diketopiperasin").
Dette er også tilfelle for deformuleringen av N-formyl-alfa-L-aspartyl-L-fenylalaninestere med hydrasin eller acylhydra-
sin som beskrevet i nyere patentansøkninger.
Ulempene med fremgangsmåtene ifølge tidligere teknikk over-vinnes med metoden til foreliggende oppfinnelse som tillater lett å framstille med høyt utbytte alfa-L-asparty1-L-feny1-alanin alkylestere som erkarakterisert vedmeget god renhet .
Ifølge oppfinnelsen fremstilles N-alfa-haloacylesterne (I) ved reaksjon mellom di tilsvarende N-alfa-haloacyl-L-aspara-ginsyreanhydrider (III) med L-fenylalanin alkylestere (IV)
hvor R, R 1 , Ar og Hal er som ovenfor definert. I sin tur erholdes anhydridene (III) fra L-asparaginsyre og alfa-halo-asidklorider (eller bromider) R'- CH(Hal) - C0C1 (eller R'-CH(Hal) - COBr), hvor R<1>og Hal er som ovenfor definert i henhold til konvensjonelle N-asyleringsmetoder fulgt av behandling med et dehydrerende middel fortrinnsvis eddiksyre-anhydrid. Alternativt fremstilles forbindelsene (III) direk-te ved å reagere L-asparaginsyre med alfa-haloasidanhydrider av f ormelen (V)
hvor R1 og Hal er som ovenfor definert. Denne reaksjonen som representerer en foretrukket utførelsesform i framstilling av anhydridene (III) utføres fortrinnsvis i inerte løsnins-midler såsom aromatiske hydrokarboner (benzen, toluen) eller haloalkaner (CCl^, CHCl^, tetrakloroetan) ved temperaturer i området fra 0°C til 100°C fortrinnsvis fra 30°C til 60°C. Alfa-haloasidanhydridene (V) er fortrinnsvis benyttet i lite overskudd; generelt 2,1 - 2,3 mol av (V) pr. mol av L-asparaginsyre som benyttes. Separasjonen av N-alfa-haloasyl-L-asparaginsyreanhydrid (III) fra alfa-halo-asidet R'- CH(Hal) - COOH dannet som biprodukt oppnåes med letthet f.eks. med krystallisering fra etylacetat.
Den følgende acylering av L-fenyalaninalkylestere (IV) med (III) utføres fortrinnsvis i inerte løsningsmidler (fortrinnsvis lavere estere såsom etylacetat eller aromatisk hydrokarboner eller lavere ketoner eller kloroalkaner) ved temperaturer i området fra -20°C til +50°C fortrinnsvis fra 0 C til + 20 C. Et spesielt rent produkt med spesielt godt utbytte erholdes når en lavere alkansyre, fortrinnsvis eddiksyre, tilsettes til reaksjonsblandingen.
Alternativt kan alfa-haloacyl-alfa-L-aspartyl-L-fenylalanin alkylestrene (II) framstilles ifølge oppfinnelsen ved å starte fra L-asparaginderivater d.v.s. fra derivater av et kommersielt tilgjengelig naturlig produkt med C> -karboksyl-syregruppen allerede blokkert.
Ifølge oppfinnelsen kondenseres en N-alfa-haloacyl-L-asparagin (VI) med en L-fenylalanin alkylester (IV) for å gi di-peptidderivat (VII) hvorav amidogruppen selektivt transfor-meres til en karboksylsyregruppe som vist i følgende reak-sjonskjerna:
hvor R, R', Ar og Hal er som ovenfor definert mens Y representerer OH eller et aktiverende atom eller gruppe; mer spesielt representerer Y hydroxy, alkoxykarbonyloxy, o-nitro-fenoxy eller en O-N-suksinimidogruppe.
Både alfa-haloacylgruppen og Y kan lett innføres i L-aspara-ginmolekylet og med stort utbytte hvor de resulterende derivater generelt er krystallinske forbindelser som lett kan isoleres i ren form.
Kondensasjonen av L-asparaginderivåtet (VI) med en L-fenylalanin alkylester (IV) blir passende utført under forskjel-lige betingelser avhengig av Y-gruppens natur.
Når Y = OH kan kondensasjonen med forbindelse (IV) med for-del utføres med disykiohexylkarbodiimid i nærvær av N-hydroxy-suksinimid i reaksjonsmedia såsom dimetylformamid. Alternativt kan samme kondensasjon også utføres ved å bruke pivaloylklorid i nærvær av tertiære baser såsom pyridin og N-etylpiperidin i løsningsmidler såsom metylenklorid eller dimetylformamid. På den annen side når Y = o-nitrofenyl kan kondensasjonen utføres ved enkel behandling med forbindelse (IV) ved romtemperatur f.eks. i acetonitri1/pyridin som løsningsmiddel.
Reaksjonsblandingen konsentreres under vakuum, fortynnes så med vann og det resulterende dipeptid (VII) gjenvinnes ved filtrering. Dipeptidet kan derpå vaskes med en natriumkarbo-natoppløsning med fortynnet saltsyre og med vann og det kan benyttes som sådan for neste steg eller det kan eventuelt rekrystalliseres med f.eks. metanol.
Konversjonen avasparaginyl-dipeptid (VII) i asprtyl-dipeptid (II) utføres med nitrosoniumsa1 ter såsom nitrosy1 tetrafluo-borat eller sulfat eller med uorganiske eller organiske nitriter i nærvær av mmerals<y>rer .
Reaksjonen utføres fortrinnsvis ved å reagere forbindelse (VII) med nitrosylsvovelsyre i eddiksyre eller i acetonitril eller dimetylformamid ved romtemperatur i omkring 4 timer under sterk omrøring. Forbindelse (II) kan utfelles ved tilsetning av isvann og gjenvinnes ved filtrering.
Reaksjonen av alfa-haloacyl-alf a-L,-aspar tyl-L-fenylalanin alkylsetrene (II) med Y utføres med temperaturer i område fra 20°C til 80°C fortrinnsvis i protiske løsningsmidler ved pH-verdier mellom 0 og 8. Reaksjonstiden avhenger av den spesielle forbindelse Y brukt til å fjerne alfa-haloacylgruppen og på reaksjonstempera turen. Som ovenfor nevnt opp-nås nevnte fjerning på en spesielt lett måte ved reaksjon av (II) med tiourea og dekomponering av det resulterende iso-tioroniumsalt i henhold til skjemaet:
hvor R, R', Ar og Hal har de ovenfor definerte betydninger.
Gjenvinningen av isotiouroniumsaltet (VIII) er ikke nødven-dig siden framstillingen av de endelige estere (I) fra halo-acylderivatene (II) viser seg å være meget enkel uten å iso-lere disse salter ved foretrukket behandling i protiske løs-ningsmidler valgt fra gruppen bestående av lavere alkoholer, vann og blandinger av disse ved pH-verdier i område fra 0
til 8 og ved temperaturer i område fra 20°C til 80°C.
Mot slutten av reaksjonen filtreres pseuditiohydantom dannet som biprodukt fra og de endelige estere (I) isoleres på en i og for seg kjent måte.
De følgende ikke-begrensende eksempler er gjengitt som illu-strasjon av fremgangsmåten i henhold til foreliggende oppfinnelse .
Eksempel 1
a) N- Kloroacety1- L- asparaginsyreanhydrid ( III, hvor R' = H, Hal = Cl)
200 g (1,05 mol) av monokloroeddiksyreanhydrid og 67 g (0,5 mol) av L-asparaginsyre oppvarmes til 40°C i 8 timer. Råproduktet vaskes to ganger med 150 ml toluen oppvarmes så til 50°C med 100 ml etylacetat. Ved avkjøling til 0°C erholdes 69 g ( 12%) av et produkt med smeltepunkt 157 - 159°C

NMR-spektrum (i hexadeuterodimetylsulfoxyd, v. TMS; signaler er gitt som cf ) : 2,6-4,5, multiplet (AB-delen av et ABX-system) , 2H : CH_2-CH; 4,2, singlet, 2H: CH2-C1;
4,8, multiplet, 1H: CH-CH2;
9,15, dublet, 1H: NH.
b) N- Kloroacetyl- alfa- L- aspartyl- L- fenylalanin metylester
( II, hvor R = CH3, R' = H, Hal = Cl)
50 g (0,26 mol) N-kloroacetyl-L-asparaginsyreanhydrid til-føres over ca 30 min. til den omrørte oppløsning av 46,5 g (0,26 mol) L-fenyl-alanm metylester i 300 ml acetonitril ved en temperatur på omkring 0°C. Omrøringen fortsettes i 1 time ved samme temperatur hvorpå blandingen oppvarmes til 40°C i 2 timer. Etter oppbevaring ved 10°C i 15 timer filtreres det krystsallinske produkt og vaskes med 150 ml avkjølt CH^CN: 81 g (84%) av N-kloroacetyl-aspartam erholdes, smeltepunkt 135-139°C (ca. 60% alfaisomer og 40% betaisomer).
Analyse: for C,rHn nClN _0, , (MW = 370,5)

c) alfa- L- aspartyl- L- fenylalanin metylestes ( aspartam) ( I, hvor R = CH3J
En oppløsning av 40 g (0,107 mol) N-kloro-acetyl-aspartan og 8,5 g (0,111 mol) tiourea i 100 ml metanol kokes under til-bakeløp i 6 timer og avkjøles så til +5°C og psuedotio-hydantoinet filtreres på sug. Filtratet inndampes til tørr-het og residiet oppvarmes til +30°C med en blanding av 13 ml 30% HC1 og 70 ml vann. Oppløsningen avkjøles til 10°C i omkring 6 timer hvorpå det krystallinske produkt filtreres på sug og vaskes med avkjølt vann (20 ml). Et rent aspartam hydroklorid erholdes med et totalt utbytte på 34%.
Eksempel 2
alfa-L-Aspartyl-L-fenylalanin metylester (aspartam) (I, hvor
R = CH,)
150 g N-kloroacetyl-L-asparaginsyreanhydrid tilsettes raskt ved -5°C til oppløsningen av 140 g metyl L-fenylalaninat i 400 ml dikloroetan pluss 200 ml eddiksyre. Blandingen omrør-es i 1 time ved -5°C; temperaturen ble så hevet over 2 timer til 15°C derpå over 30 min. til 40°C. Dikloroetan fordampes under vakuum; 500 ml metanol og 60 g tiourea ble tilsatt og blandingen kokes under tilbakeløp i 3 timer. Etter avkjøling til 20°C tilsettes 200 ml metanol inne-holdende 11 vektprosent HC1 hvorpå oppløsningen avkjøles til 0°C over 1 time, filtreres og inndampes til tørrhet in vacuo. Residiet rekrystalliseres fra 600 ml vann inneholdene 70 g NaCl: et meget rent aspartam hydroklorid erholdes med 48% utbytte.
Eksempel 3
a) N- kloroacetyl- L- asparaginsyreanhydrid ( III, hvor R' = H, Hal = Cl)
Til en blanding av 210 g (1,23 mol) monokloroeddiksyreanhydrid og 100 ml CHC13tilsettes 67 g (0,5 mol) L-asparagin-
syre. Etter omrøring i 6 timer ved 40-45°C tilsettes 400
ml CHCl^og den varme suspensjonen filtreres på sug. Produktet behandles med 200 ml CHCl^ for å fjerne kloroeddik-syren i så stor grad som mulig, filtreres på sug, vaskes på filteret med noe CHCl^og tørkes under vakuum ved 40°C: 88 g av et meget rent produkt erholdes, smaltepunkt 157-159°C.
Potensiometrisk titrering (morfolm): 98%.
Utbytte: 91,9%.
Fra moderluten kan monokloroeddiksyre lett gjenvinnes.
b) N- kloroacetyl- alfa- L- aspartyl- L- fenylalanin metylester
( II, hvor R = CH3, R<1>= H Hal = Cl)
Til en blanding av 60 g (0,32 mol) N-kloroacetyl-asparagin-syreanhydr id , 100 ml etylacetat og 80 ml eddiksyre omrørt ved 5-10°C tilsettes oppløsningen av 58 g (o,32 mol) L-fenylalanin metylester i 200 ml etylacetat over ca. 1 time. Blandingen omrøres i 4 timer ved 5-10°C og så i 2 timer ved romtemperatur. Presipitatet pumpefi 1 treres: 113 g av produktet erholdes, smeltepunkt 147-149°C (96% utbytte); potensiometrisk assay (NaOH 0,1 N) 98%; ved HPLC 80% alfaisomer og 20% betaisomer isentifiseres.
c ) alf a- L- aspar ty 1- L- f eny lal am n metylester ( aspartam)
( I, hvor R = CH3)
En blanding av 100 g (0,27 mol) metylester av N-kloroacety1-alfa-L-aspartyl-L-fenylalanin (80% alfa - 20% beta), 21 g (0,21 mol) tiourea og 300 ml metanol kokes under tilbakeløp i 5 timer. Etter avkjøling til +5°C filtreres det presipiterte tiohydandoin på sug. Filtratet inndampes in vacuo til et lite volum og behandles med 60 ml eddiksyre, 140 ml vann og 30 ml 30% HC1. De gjenværende metanol fordampes in vacuo og oppløsningen kimes ved 40 °C med aspartam hydroklorid for derpå å sakte avkjøles til +5°C og oppbevares over natten ved denne temperatur. Presipitatet gjenvinnes ved avsug og vaskes med 30 ml isavkjølt vann. Fra dette hydroklorid erholdes 40 g av et meget rent aspartam ved konvensjonelle metoder. Utbytte er 50% fra råproduktet (62,5% av alfaisomer). Smeltepunkt 240-248°C; ( c< ) 2 0=
15,5 (4% maursyre); potensiometrisk assay: a) med tetrabutylamoniumhydroksyd = 99,5%; b)med HC1C>4 = 99,5%
(tørrprodukt); "diketopiperasin" < 0,3%; pseudotiohydantoin < 0,3%; "beta"aspartam: fraværende.
Fra moderluten gjenvinnes ved kraftig hydrolyse 18,5 g av L-fenylalanin.
Eksempel 4
a) alfa- bromoproponioyl- L- asparaginy1- L- fenyalanin metylester ( VII, hvor R = R' = CH , Hal = Br)
26,7 g (0,1 mol) fint pulverisert alfa-bromopropiony1-L-asparagin (erholdt fra L-asparagin og alfa-bromopropionyl-klorid i henhold til Schotten-Baumann) ble oppløst i 100 ml dimetylformamid ved tilsetning av 8 ml pyridin og 28 ml N-etyl-piperidin. Oppløsningen ble raskt avkjølt til -30°C under sterk omrøring og den resulterende suspensjonen ble behandlet med 12 g pivaloylklorid oppløst i 20 ml dimetylformamid. Den klare oppløsning som ble erholdt ble behandlet etter ca. 30 min. med 22 g L-fenylalanin metylester hydroklorid. Blandingen ble holdt ved -30°C i 30 min. og så spontant hevet til romtemperatur over ca. 2 timer. Oppløs-ningen konsentrert under vakuum ble helt over i 1 liter vann og suspensjonen ble holdt under omrøring i noen timer. Det presipiterte produkt ble filtrert, vasket med vann og tørket under vakuum for å gi 30 g, 70% utbytte.
Etter sur hydrolyse av produktet i et lukket kammer i 22 timer i 6M HC1 ved 110°C gav analysen i aminosyrer et fenylalanin:asparaginsyre forhold = 1:0,97 med et 99,2% utbytte.
b) alfa- L- aspartyl- L- fenylalanin metylester ( I, hvor R =
CH^)
21,4 g (0,05 mol) alfa-brompropionyl-L-asparaginyl-L-fenylalanin metylester oppløst i 150 ml dimetylformamid ble behandlet med 15 ml 6M HC1 oppløsning i dioxan og så med 10,5 g n-butylnitritt tilsatt over ca. 20 min. Etter 6-8 timer ved romtemperatur under konstant omrøring ble blandingen konsentrert til tørrhet og residiet tatt opp i 100 ml abso-lutt metanol. 3,8 g (0,05 mol) tiourea ble tilsatt den resulterende blanding og det ble så kokt under tilbakeløp i 20 min. Den metanoliske suspensjon av isotiouroniumbromid der-ved erholdt (VIII, hvor R = R'= CH3og Hal = brom) ble konsentrert til tørrhet og residiet tatt opp i 100 ml vann hvor pH av oppløsningen ble bragt til 7-8 med NaHCC>3, hvorpå blandingen så ble oppvarmet til 40°C i 2 timer. Etter avkjøling til romtemperatur bel blandingen surgjort til pH 4,5 med 2M HCl og så konsentrert under vakuum. 11,47 g aspartam i hovedsak identisk til den erholdt i tidligere eksempler ble framstilt.
Eksempel 5
a) N-kloroacety1-alfa-L-asparty1-L-fenylalanin etylester
(II, hvor R = C.H-, R' = H Hal = Cl)

5 g av N-kloroacety1-L-asparaginsyreanhydrid ble tilsatt over 10 min. under omrøring til 4,8 g etyl L-fenylalanin i
50 ml CH3CN, ved ca. 0°C. Blandingen ble omrørt i 30
min. ved 0°C så for 2 timer ved 40°C og filtrert på sug. Etter vask med 20 ml isavkjølt CH3CN, ble 7,5 g av tittel-forbindelsen erholdt, smeltepun kt 118-120 C.
b) alfa-L-aspartyl-L-fenylalanin etylester (I, hvor R =
Til oppløsningen av produktet erholdt i a) i 30 ml metanol
ble det tilsatt 1,7 g tiourea. Etter koking på tilbakeløp i 5 timer ble blandingen avkjølt til 0°c og pseudotiohydan-
toinet ble fjernet ved pumpefi 1 trering. Moderluten ble inn-dampet til tørrhet og residiet ble behandlet ved 30°C med 2 ml 30% HC1 i 10 ml vann. Etter avkjøling til +5°C ble det krystalinske produkt filtrert på sug. TittelforbindeIs-en ble erholdt (som hydroklorid) med 38% utbytte.
Analyse: for c15H2iclN2°5(MW = 344-5)
Utregnet %: C = 52,25; H = 6,09; N = 8,13
Funnet % : C = 52,03; H = 6,19; N = 8,03.
På samme måte ble isobutylesteren av alfa-L-asparty1-L-fenylalanin erholdt.
Eksempel 6
Ved å gå fram som beskrevet i eksempel 2 men ved å bruke natriumtiosyanat i stedet for tiourea ble aspartam erholdt med et totalt utbytte på 22%.
Eksempel 7
Prosedyren fra eksempel 1 ble gjentatt ved å bruke - i stedet for tiourea - ekvimolare mengder av følgende forbindelser: kaliumcyanat; natriumditiokarbamat; guanidin; cyctamin; etylendiamin; pyrokatekol.
Aspartam ble erholdt med utbytte fra 10-15%.
Eksempel 8
Fremgangsmåten fra eksempel 2 ble gjentatt med det unntak at alfa-haloacylgruppen var alfa-bromo-hexanoylgruppen (R' = n-butyl). Aspartam av meget god renhet ble erholdt ; 43% utbytte.
Claims (8)
1. Fremgangsmåte for fremstilling av alkylestere av alfa-L-aspartyl-L-fenylalanin av formel (I)
hvor R er en C-^-C^ alkylgruppe, karakterisert ved at alfa-haloacyl-alfa-L-aspartyl-L-fenylalanin alkylestere av formel (II)
hvor
R er som definert ovenfor;
Ar er fenylgruppen;
R' er hydrogen eller en C^ -C^ , lineær eller forgrenet alkylgruppe;
Hal er klor,brom eller jod,
reageres med en forbindelse valgt fra gruppen bestående av tiourea, cyansyresalter, tiocyansyresalter, ditiokarbamat-salter, guanidm, 1,2-etanditiol, cyctamin, etanolamin, atylendiamin, 1,2-diaminobenzen, 1,2-dimerkaptobenzen, pyrokatekol, o-amino-fenol.
2. Fremgangsmåte ifølge krav 1, karakterisert ved at reaksjonen utføres i protiske løsningsmidler.
3. Fremgangsmåte ifølge krav 2, karakterisert ved at løsningsmidlet er valgt fra gruppen bestående av lavere alkoholer, vann og blandinger derav.
4. Fremgangsmåte ifølge kravene 1 til 3, karakterisert ved at reaksjonen utføres ved pH-verdi i området fra. 0 til 8.
5. Fremgangsmåte ifølge kravene 1 til 4, karakterisert ved at reaksjonstemperaturen ligger i området 20°C til 80°C.
6. Fremgangsmåte ifølge kravene 1 til 5, karakterisert ved at alfa-haloacyl-alfa-L-aspartyl-L-fenylalanin alkylestere (II) reageres med tioyrea.
7. Som nye produkter alfa-haloacy1-alfa-L-asparty1-L-fenylalanin alkylestere av formel (II)
karakterisert ved at R, R', Ar og Hal er som definert ovenfor.
8. Nye produkter ifølge krav 7, karakterisert ved at R er en metylgruppe.
