Description
Denne oppfinnelse angår fremstilling av nye forbindelser som kombinerer hemmende virkning mot angiotensinomdannende enzym med diuretisk virkning.
Det er kjent forbindelser med evne til å undertrykke eller hindre overføring av angiotensin I til angiotensin II, en egenskap som gjør dem anvendelige ved behandling av forhøyet blodtrykk. Eksempler på slike forbindelser er de forbindelser som beskrives i US patentskrift nr. 4.105.776, og som har den generelle formel: hvor R er hydrogen eller alkyl med 1-7 carbonatomer, fortrinnsvis methyl, m er 1 eller 2 og n er 0 eller 1, og de forbindelser som beskrives i US patentskrift nr. 4.256.761, og som har den generelle formel:

Dertil er det i US patentsøknad nr. 148.089 av 12. mai 1980 beskrevet forbindelser som oppviser inhiberende aktivitet på angiotensin-omdannende enzym, og som har den følgende struktur:

Det er kjent tallrike diuretika i faget, blant dem benzensulfonamidene som er beskrevet i US patentskrift nr. 3.058.882, og som har den generelle formel

Alkylgruppene som sådanne og alkylgruppene i alkyl-delen i aralkyl, cycloalkylalkyl, polycycloalkylalkyl, hetero-arylalkyl o.l. samt i alkoxy, alkylmercapto, alkanoyl, carbalkoxy, alkylamino og dialkylamino kan være rettkjedede eller forgrenede og er fortrinnsvis alkylgrupper inneholdende fra 1 til 20 carbonatomer. Halogengruppene innbefatter fluor, klor, brom og jod.
De nye forbindelser lar seg lett fremstille under anvendelse av kjente utgangsmaterialer og fremgangsmåter. Det vil forstås av fagfolk på området at forbindelsene som har et asymmetrisk carbonatom, kan foreligge i både venstredreiende og høyredreiende form, og begge former innbefattes innenfor rammen av oppfinnelsen.
De nye forbindelser kan fremstilles ved omsetning av
et acylerende derivat av en syre med formelen
med en forbindelse med formelen eller de nye forbindelser kan alternativt fremstilles ved omsetning av et acylerende derivat av en syre med den generelle formel med en forbindelse med formelen eller
fortrinnsvis med carbodiimider tilstede, slik det vanligvis benyttes ved peptidsynteser, f.eks. N,N-dicyclohexylcarbodiimid. Det benyttes vanligvis et reaksjonsoppløsningsmiddel for å muliggjøre mer intim kontakt mellom reaktantene. Egnede oppløs-ningsmidler er ethere av ethylenglycol og diethylenglycol, tetrahydrofuran, dioxan, alkylketoner såsom aceton og methyliso-butylketon, og lignende oppløsningsmidler. Reaksjonstemperaturen er ikke av avgjørende betydning, da reaksjonen forløper glatt ved romtemperatur og sogar ved lavere temperaturer. Bruk av høyere temperatur under reaksjonen, f.eks. reaksjonsblandingens tilbakeløpstemperatur, vil korte inn reaksjonstiden, men vil
kunne føre til dannelse av uønskede biprodukter. Vanligvis tillates reaksjonen å forløpe ved romtemperatur inntil den er fullført. Produktene oppsamles ved hjelp av vanlige metoder og renses f.eks. ved omkrystallisering fra egnede oppløsnings-midler.
De forbindelser hvor frie syregrupper er tilstede, f.eks. forbindelser hvor Y er OH, kan lett danne salter med egnede kationer, fortrinnsvis med slike som er farmasøytisk aksepterbare, f.eks. såvel énverdige som flerverdige metallioner, såsom natrium, kalium, calcium, magnesium, jern og lignende, samt ammoniumioner avledet fra ammoniakk og primære, sekundære og tertiære aminer.
De følgende eksempler illustrerer oppfinnelsen.
Eksempel 1
Dette eksempel illustrerer fremstillingen av mellom-produkter benyttet ved fremstillingen av de nye forbindelser.
A. N- Cyclopentylglycin- t- butyl- ester
Til en oppløsning inneholdende 75 g (0,88 mol) cyclo-pentylamin og 100 g (1,0 mol) triethylamin i 500 ml ether, ble det ved 0°C tilsatt langsomt 136 g (0,70 mol) t-butyl-bromacetat i 200 ml ether. Etter omrøring ved romtemperatur i 6 timer ble blandingen filtrert, vasket med vann og konsentrert i vakuum. Residuet ble surgjort med 5%-ig vandig HC1, vasket med~ether, gjort basisk med 5%-ig vandig I^CO^ og ekstrahert med ether. Etheroppløsningen ble så vasket med vann, tørret over MgSO^og konsentrert i vakuum, hvorved man fikk 9 0 g (65%) N-cyclopentyl-glycin-t-butylester i form av en olje som krystalliserte ved henstand.
B. t-butylesteren av N-(3-acetylthio-2-methylpropanoyl)-N-cyclopentylglycin
Til en oppløsning inneholdende 60 g (0,30 mol) N-cyclopentylglycin-t-butyl-ester og 75 ml (0,54 mol) triethylamin i 200 ml methylenklorid ble det ved 0°C tilsatt dråpevis en oppløsning av 54 g (0,30 mol) 3-acetylthio-2-methylpropanoyl-klorid i 250 ml methylenklorid. Etter at reaksjonsblandingen var blitt omrørt ved romtemperatur natten over ble den filtrert og konsentrert i vakuum. Residuet ble tatt opp i ethylacetat og vasket én gang med mettet vandig NaHCO^, én gang med saltopp-løsning, tre ganger med 5%-ig vandig HCl, én gang med saltopp-løsning og fem ganger med mettet vandig NaHCO-^. Den organiske fase ble tørret over MgSO^, filtrert og konsentrert i vakuum, hvorved man fikk 80,1 g (78%) N-(3-acetylthio-2-methylpropanoyl)-N-cyclopentylglycin-t-butyl-ester i form av en olje som krystalliserte ved henstand.
C. N-( 3- acetylthio- 2- methylpropanoyl)- N- cyclopentyl- glycin
En blanding inneholdende 80,1 g (0,23 mol) N-(3-acetylthio-2-methyl-propanoyl)-N-cyclopentyl-glycin-t-butyl-ester,
37,9 g (0,35 mol) trimethylsilylklorid og 52,5 g (0,35 mol) natriumjodid i 300 ml acetonitril ble oppvarmet ved 45-50°C i 30 minutter. Etter tilsetning av 50 ml vann ble blandingen konsentrert i vakuum. Residuet ble gjort basisk med 400 ml mettet vandig NaHCO^, vasket tre ganger med ethylacetat, surgjort med konsentrert HCl og ekstrahert tre ganger med ethylacetat. Disse siste organiske ekstrakter ble slått sammen, tørret over MgSO^, filtrert og konsentrert i vakuum, hvorved man fikk 60 g
(91%) av det urene produkt. Etter rensning ved høytrykks væskekromatografering (HPLC) ethylacetat/hexan/eddiksyre (60/40/2) fikk man N-(3-acetylthio-2-methylpropanoyl)-N-cyclopentyl-glycin i form av en olje.
D. N-( 3- mercapto- 2- methylpropanoyl)- N- cyclopentyl- glycin
Gjennom en blanding inneholdende 10,4 g (36 mmol) N-(3-acetylthio-2-methylpropanoyl)-N-cycldpentyl-glycin ble det boblet ammoniakkgass i en time. Etter 20 minutter ble blandingen konsentrert i vakuum, og residuet ble tatt opp i ethylacetat og vasket tre ganger med 5%-ig vandig natriumbisulfat. Oppløsningen ble så tørret over MgSO^, filtrert, konsentrert i vakuum og renset ved høytrykks væskekromatografering ethylacetat/toluen/ hexan/eddiksyre (50/25/50/2) , hvorved man fikk 7,9 g (90%) N-(3-mercapto-2-methylpropanoyl)-N-cyclopentyl-glycin i form av en olje.
E. N-(3-mercapto-2-methylpropanoyl)-N-cyclopentyl-glycin-methylester
En blanding inneholdende 19,0 g (77 mmol) N-(3-mercapto-2-methylpropanoyl)-N-cyclopentyl-glycin og 2,0 g (10,5 mmol) p-toluensulfonsyre i 500 ml methanol ble kokt med tilbake-løpskjøling natten over. Reaksjonsblandingen ble konsentrert i vakuum, og residuet ble tatt opp i ether. Det ble vasket en gang med mettet vandig natriumbicarbonat og én gang med saltopp- løsning. Den organiske fase ble tørret over MgSO^, filtrert og konsentrert i vakuum, hvorved man fikk N-(3-mercapto-2-methyl-propanoyl)-N-cyclopentyl-glycin-methylester i form av en olje.
F. N-(3-mercapto-2-methylpropanoyl)-N-cyclopentylglycin-t-butyl- ester
Inn i en oppløsning inneholdende 50 g (0,14 mol) N-(3-acetylthio-2-methylpropanoyl)-N-cyclopentylglycin-t-butyl-ester i 700 ml methanol ble det boblet nitrogengass i 10 minutter og deretter ammoniakkgass i 30 minutter. Etter 30 minutter ble blandingen konsentrert i vakuum og residuet tatt opp i ethylacetat. Det ble vasket én gang med 3%-ig vandig HCl og én gang med vann, tørret over MgSO^, filtrert og konsentrert i vakuum, hvorved man fikk et oljeaktig produkt. Rensning ved høytrykks væskekromatografering 15% ethylacetat i hexaner ga 30 g (70%) N-(3-mercapto-2-methyl-propanoyl)-N-cyclopentylglycin-t-butyl-ester i form av en olje.
Eksempel 2
N-[ 3- [2,3-diklor-4-(2-methylen-1-oxobutyl)-fenoxy]-acetylthio-2- methylpropanoyl]- N- cyclopentyl- glycin- methylester
Til en oppløsning inneholdende 10,5 g (40,0 mmol) N-(3-mercapto-2-methylpropanoyl)-N-cyclopentyl-glycin-methylester og 12,1 g (39,9 mmol) ethacrynsyre i 200 ml glyme ble det ved 0°C tilsatt en oppløsning av 8,24 g (39,9 mmol) N,N-dicyclo-hexylcarbodiimid li 20 ml. glyme. Oppløsningen ble tillatt å oppvarmes til romtemperatur og ble omrørt natten over. Blandingen ble filtrert, og filtratet ble konsentrert i vakuum. Rensning av produktet ved høytrykks væskekromatografering hexan/ethylacetat/ aceton (70/20/1 5) ga N-[3-[2,3-diklor-4-(2-methylen-l-oxobutyl) - fenoxy] -acetylthio-2-methylpropanoyl]-N-cyclopentyl-glycin-methylester i form av en viskøs olje.
Eksempel 3
N-J^3-[ 2, 3-diklor-4- ( 2-methylen-1 -oxobutyl) -f enoxy] -acetylthio-2- methylpropanoyl]- N- cyclopentyl- glycin- t- butyl- ester
Til en oppløsning inneholdende 19,2 g (63,8 mmol) N-(3-mercapto-2-methylpropanoyl)-N-cyclopentylglycin-6-butyl-ester og 19,3 g (63,7 mmol) ethacrynsyre i 200 ml methylenklorid ble det ved 0°C tilsatt en oppløsning av 13,2 g (64,0 mmol) N,N-dicyclohexylcarbodiimid i 20 ml methylenklorid. Oppløsningen ble tillatt å oppvarmes til romtemperatur og ble omrørt natten over. Oppløsningen ble filtrert, og filtratet, ble konsentrert i vakuum. Rensning av produktet ved høytrykks væskekromatografering 10% ethylacetat i hexaner ga N-[3-[2,3-diklor-4-(2-methylen-1-oxobutyl)-fenoxy]-acetylthio-2-methylpropanoyl]-N-cyclopentyl-glycin-t-butyl-ester i form av en viskøs olje.
Eksempel 4
N-[3-[2,3-diklor-4-(2-methylen-l-oxobutyl)-fenoxy]-acetylthio-2- methylpropanoyl] - N- cyclopentyl- glycin
Til en oppløsning inneholdende 5,2 g (8,7 mmol) N-^3-[2,3-diklor-4-(2-methylen-l-oxobutyl)-fenoxyj-acetylthio-2-methylpropanoylJ-N-cyclopentyl-glycin-t-butyl-ester i 100 ml methylenklorid ble det satt 9,0 ml (117 mmol) trifluoreddiksyre. Etter 12 timer ble reaksjonsblandingen konsentrert forsiktig i vakuum, og residuet ble kromatografert ved høytrykks væskekro-matograf ering methylenklorid/ether/eddiksyre (88/10/2,5) hvorved man fikk 2,4 g (52%) N-[3-[2 ,3-diklor-4-(2-methylen-l-oxobutyl) -fenoxy]-acetylthio-2-methylpropanoyl] -N-cyclopentyl-glycin i form av et glass.
Eksempel 5
N-£3-(4-klor-2- £(2-furanylmethyl)-amino]-5-sulfamoyl)-benzoyl-thio- 2- methylpropanoylj- N- cyclopentyl- glycint- butyl- ester
Til en oppløsning inneholdende 11,5 g (38,2 mmol) N-(3-mercapto-2-methylpropanoyl)-N-cyclopentylglycin-t-butyl-ester og 12,6 g (38,1 mmol) furosemid i 100 ml tetrahydrofuran og 35 ml aceton ved 0°C ble det tilsatt 7,84 g (38,0 mmol) N,N-dicyclohexylcarbodiimid i 20 ml tetrahydrofuran. Oppløs-ningen ble tillatt å oppvarmes langsomt til romtemperatur og ble omrørt natten over. Oppløsningen ble filtrert, og filtratet ble konsentrert i vakuum. Rensning av produktet ved høytrykks væskekromatografering hexan/ethylacetat/aceton (4/1/1) ga N-[*3- (4-klor-2- £( 2-f uranylmethyl) -amino] -5-sulf amoyl) -benzoylthio- 2-methylpropanoylJ-N-cyclopentyl-glycin-t-butyl-ester i form av et glass.
Eksempel 6
N-^3-(4-klor-2- [(2-furanylmethyl)-amino]-5-sulfamoyl)-benzoyl-thio- 2- methylpropanoyl]- N- cyclopentyl- glycin
Inn i en oppløsning av -10°C inneholdende 7,72 g
(7,69 mmol) N-[3-(4-klor-2- [(2-furanylmethyl)-amino)-5-sulfamoyl)-benzoylthio-2-methylpropanoyl] -N-cyclopentyl-glycin-t-butyl-ester og 624 ml (7,72 mmol) pyridin i 100 ml methylenklorid ble det boblet isobutylen i 5 minutter. Til denne oppløsning ble det så dråpevis satt 2,03 ml (14,3 mmol) trimethylsilyljodid. Etter 2 timer ble reaksjonen stanset.ved tilsetning av 1,0 ml vann. Oppløsningen ble konsentrert i vakuum, og residuet ble tatt opp i en blanding av kloroform og methylenklorid. Det utfelte faste stoff ble frafiltrert og omkrystallisert 2 ganger fra en blanding av acetonitril, ethanol og vann, hvorved man f ikk 1,2 g (28%) N-[3-(4-klor-2-[(2-furanylmethyl)-amino]-5-sulfamoyl)-benzoylthio-2-methylpropanoyl]-N-cyclopentyl-glycin i form av et gult fast stoff. Smeltepunkt 204-206°C.
På samme måte ble mercaptanforbindelsene i rekke A nedenfor acylert med syrene i rekke B for dannelse av forbindelsene oppført i rekke C etter spaltning av deres t-butyl-estere.
5-sulfamyl-4-klor-2-[(2<1->furanylmethyl)-amino]-benzoesyre omsettes med mercaptanforbindelse #^1 for dannelse av det tilsvarende acylerte mercaptan. På tilsvarende måte dannes acylerte mercaptaner ved omsetning av 5-sulfamyl-2-[(2<1->furanylmethyl)-amino]-4-fenoxybenzoesyre med mercaptanforbindelse M2 og omsetning av 2,3-diklor-4-(2-methylen-l-oxobutyl)-fenoxy-eddiksyre med forbindelse # 5.
