Description
Foreliggende oppfinnelse vedrører 2,6-C-dimetyl-
.1o
tyrosin -D-aminosyre^-e-amxnokapronsyre- og y-aminosmørsyre-derivater av metioninenkefalin, og fremgangsmåte for deres fremstilling.
I 1975 ble et pentapeptid, metioninenkefalin, rap-portert av Hughes et al., Nature, 258, 577 (1975). Dette peptid finnes i mange områder i hjernen hvor det synes å
virke som en neurotransmitter for neumodulator i et sentralt smerteundertrykkende system. Det naturlige peptid bindes stereospesifikt til partielt rensede hjerneopiat-reseptor-steder, se f.eks. Bradbury et al., Nature, 260, 793 (1976).
Det naturlige peptid er også meget aktivt i biobestemmelser
for opiat aktivitet, men utviser bare svak, forsvinnende analgetisk aktivitet når det injiseres direkte i hjernen hos rotte, se f.eks. Belluzi et al., Nature, 260, 625 (1976).
For å overvinne mangelen på invivo aktivitet, har en rekke forskere foretatt flere modifikasjoner i metioninenkefalin-strukturen, slik som erstatning av glycindelen i 2-stillingen med en D-aminosyre, N-metylering av 1-tyrosin-delen, erstatning av 4-fenylalanindlen med f.eks. metyl eller halogen, modifisering av C-terminusen, osv. for å fremstille enkefalinderivater med forskjellige egenskaper og virknings-grader .
Ifølge foreliggende oppfinnelse tilveiebringes nye enkefalinderivater og deres farmasøytisk akseptable salter som har virkningsgrad tilnærmet den for morfin som analgeti-
ske midler ved både oral og parenteral administrasjon.
Foreliggende oppfinnelse tilveiebringer nye derivater av metioninenkefalin, 2,6-C-dimetyltyrosyl-D-^metionyl-glycyl-fenylalanyl-Y-aminosmørsyre og derivater og 2,6-C-dimetyltyrosyl-D-metionyl-glycyl-fenylalanyl-kapronsyre og derivater og farmasøytisk akseptable salter derav, forbindelsene er nyttige som analgetiske midler.
Enkefalinderivatene ifølge oppfinnelsen representeres ved formelen:
hvor X er metionin, alanin eller nor-leusin; m er 3 eller 5;
og R er hydrogen, lavere-alkyl eller -NR2R3; hvor R2og R^
er like eller forskjellige, valgt fra gruppen bestående av hydrogen eller lavere-alkyl; og hvor (<*>) refererer til den viste forbindelse, dens speilbilde eller en blanding av racemater; eller farmasøytisk akseptable salter derav.: I den ovenfor angitte formel betegner "C-diMe" di-metylsubstitusjonen på den aromatiske ring i tyrosin. Tid-ligere substitusjoner i tyrosin har vært N-substitusjoner, ;og man har funnet at C-substitusjonen uventet øker aktiviteten til forbindelsene. ;Alle aminosyrerester identifisert heri foreligger i den naturlige eller L-konfigurasjonen med mindre annet er spe-sifisert, med unntagelse av glycin som ikke har noe symmetri-senter, og man har benyttet standardforkortelser for aminosyre-restene. ;Dimetyltyrosinresten kan være i L-, D- eller DL-konfigurasjonen. ;Betegnelsen "lavere-alkyl" refererer til C^-Cg;rette eller forgrenede alkylradikaler, dvs. metyl, etyl, n-propyl, iso-propyl, n-butyl, tert-butyl, n-hexyl osv. ;Den benyttede betegnelsen "farmasøytisk akseptable salter" refererer til de ikke-toksiske syreaddisjonssaltene som generelt fremstilles enten in situ eller ved omsetning av en forbindelse ifølge oppfinnelsen, med den ønskede organiske eller uorganiske syre ifølge velkjente metoder. Repre-sentative salter er hydrokloridet, hydrobromidet, sulfatet, bisulfatet, acetatet, oxalatet, valeratet, oleatet, leuratet, boratet, benzoatet, laktatet, fosfoatet, tosylatet, sitratet, maleatet, fumaratet, succinatet, tartratet, tosylatet, nap-sylatet osv. ;Den analgetisk aktivitet til forbindelsen ifølge oppfinnelsen ble først påvist ved varmplåteprøven pg PBQ-vridningsprøven med mus. ;Varmpl a t e - metode. / Undersøkelsesmetodikk: ben benytted^ metode var en modifikasjon av den som er beskrevet av Eddy og Leimbach. ;To grupper av fjorten mus ble bragt inn i forsøksrommet om-trent en halv time før undersøkelse. Musene ble anbragt enkeltvis under en begrensende sylinder (600 ml "Pyrex") på ;en varm plate ("Pyro-magnilistør") med temperaturen styrt av en temperaturregulator ("YSI modell 72") ved 55° 0,5°C. Reaksjonstiden for hver mus med henblikk på å slikke en fot eller hoppe ble målt ("Lab-chron timer") tre ganger ved tyve minutters intervaller. Mus som ikke reagerte innen 15.sekunder ble utelukket. Ti mus ble gitt en dose av test-legemiddelet, og ti ble gitt 0,9% saltoppløsning intravenøst tyve minutter etter måling av den siste reaksjonstid: Dyrene ble testet som før, ti, tredve og seksti minutter etter denne injeksjon. Mus som ikke reagerte i løpet av 30 sekunder ble fjernet fra varmeplaten, og gitt en reaksjonstid på tredve sekunder. Analgesi ble ansett som demonstrert hos en mus dersom dens post-legemiddelreaksjonstid var større enn midde-let for kontrollgruppen pluss standard avvik. Antalll dyr som viste analgesi i legemiddelgruppen ble sammenlignet med den samme verdi for kontrollgruppen ved hjelp av Fisher<*>s nøyak-tige sannsynlighetstest. ED5g-verdier ble beregnet ved Log-probit-analyse under anvendelse av den prosentandel mus som viste analgesi i hver legemiddelgruppe minus den samme målte verdi for kontrollprøven.
PBQ- indusert vridning hos mus.
Metode: Grupper på minst ti CD-1 hahnmus (Charles River), med en vekt på 18-25 g fikk sulte natten over og ble forbehandlet intragastrisk (i.g.) med et bærermateriale (1-2 dråper av en 1:1 blanding av propylenglykol:"Tween 80" pr. 10 ml 0,9% NaCl) 10 ml/kg eller legemiddelet enten i oppløs-ning eller som en homogenisert suspensjon. PBQ ble admini-strert i.p. 30 minutter senere som en 0,025% oppløsning i 5% etylalkohol. Med utgangspunkt fem minutter etter PBQ-administrasjonen, ble hver mus plassert enkeltvis i et glass-kår og antall vridninger ble opptelt i løpet av den etterføl-gende ti minutters periode. En vridning ble ansett som å
bestå av en kombinasjon eller rekkefølge av krumning av ryggen, pelvisk rotasjon og strekking av bakben.(1). For å
oppnå maksimum sensitivitet ble signifikant analgesi antatt å foreligge når et enkelt dyrs vridningsfrekvens var 50% eller mindre enn kontrollgruppens midlere frekvens (2). Be-stemmelser av EDcjQ-verdien og hellingen på dose-responslinjen ble foretatt ut fra kvantedata under anvendelse av Berksons logistiske metode (3). ED^Q-verdien ble definert som den dose som minsket vridning med 50% eller mer av kontrollgruppe-middelet hos 50% av forsøksdyrene.
Forbindelsene som fremstilles ifølge oppfinnelsen kan administreres enten oralt eller parenteralt for å lindre smerter hos pasienter (pattedyr) i orale doseringer på 0,5-5mg/kg legemsvekt og 0,05-3 mg/kg legemsvekt daglig, fortrin-nsvis i oppdelte doser, dvs. hver fjerde-sjette time etter behov for å lindre smerten.
Følgende eksempler illustrerer oppfinnelsen ytterligere.
EKSEMPEL 1
Fremstilling av tert- butyloksykarboriyl- fenylala-nyr- metyl- e- amino- caproat.
Til en kald oppløsning av 2,6 5 g (10.0 mmol) tert-butyloksykarbonyl (Boe) -fenylalanin og 2,00 g (11,0. mmol) metylkaproathydroklorid i 25 ml metylenklorid, ble det tilsatt 1,25 ml (11,2 mmol) dicykloheksylkarbodiimid og 1,25 ml (1,10 mmol) N-metylmorfolin. Reaksjonsblandingen ble pmrørt i tredve minutter ved romtemperatur, og deretter avkjølt i flere dager.
Den i reaksjonen dannede dicykloheksylurea-forbindelsen ble filtrert og filtratet ekstrahert med vann flere ganger, tørket over vannfritt magnesiumsulfat og til slutt filtrert og inndampet. Den resterende olje stivnet raskt, og dette :ga et. kvantitativt produktutbytte. Tynnsjikekromato-grafi [kloroform:metanol:eddiksyre:vann (83:15:1:lv/v/v/v)]-Rf 0,85. Strukturen ble bekreftet ved NMR.
EKSEMPEL 2.
Fremstilling av tert- butyloksykarbonyl- fenylalanylmetyl- y- amlnobutyrat.
Tittelforbindelsen ble fremstilt på samme måte som
i eksempel 1 fra 2,65 g Boc-Phe, 1,69 g (11,0 mmol) metyl-y-aminobutyrathydrokloridsalt, 1,25 ml N-metylmorfolin og 2,27 g (11,6 mmol) dicykloheksylkarbodiimid i 25 ml metylenklorid, omsatt i 24 timer for tilveiebringelse av et kvantitativt produktutbytte (tynnsjiktskromatografi i det ovenfor angitte oppløsningsmiddelsystem, Rj 0,84).
EKSEMPEL 3.
Frems ti Iling av fe ny la 1 any 1 - me ty 1 - r e - aml nok apr o a t-hydroklorid.
Tert-butyloksykarbonyl-fenylalanyl-metyl-e-amino-kaproat (10,0 mmol) ble oppløst i 37 ml iseddik 6g behandlet med 18 ml 5,6 N saltsyre i dioxan i 20 minutter ved romtemperatur. Oppløsningsmidlene ble inndampet og vannfri eter tilsatt for dannelse av et bunnfall. Det kalde bunnfall ble.filtrert og vasket med eter, deretter tørket i vakuum ved 65°C i tre timer, og dette ga produktet i en mengde på 2,79 g (84,8% utbytte) .
EKSEMPEL 4.
Fremstilling av fenylalanyl- metyl- y- aminobutyrat-bydroklorid.
Fremgangsmåten i eksempel 3 ble fulgt under anvendelse av identiske mengder stoffer for oppnåelse av 2,85 g produkt (98,4% utbytte).
EKSEMPEL 5.
Fremstilling av Boc- glycyl- fenylalanyl- metyl-e- aminokaproat.
Til 2,45 g (7,45 mmol) av dipeptidet i eksempel 2 og 2,04 g (8,57 mmol,.15% overskudd)Boc-Gly-2,4,5-triklor-fenylester (OCP) i 15 ml dimetylformamid, ble det tilsatt 0,85 ml (7,65 mmol) N-metylmorfolin og reaksjonsblandingen ble hensatt ved romtemperatur i .48 timer. Tilsetning av 175 ml kald, 5% kaliumbisulfat ga en gummiaktig bunnfall som ble ekstrahert (3 x 50 ml) i etylacetat. De kombinerte organiske lag ble vasket med vann, deretter saltoppløsning, og tørket over vannfritt magnesiumsulfat, og etter filtrering inndampet, og dette ga en olje.
Produktet ble renset ved lavtrykks-væske-kromato-grafi under anvendelse av siliciumdioksydgel, og eluering med en 100% kloroform til 100% etylacetat lineær gradient. Vekten av det oljeaktive produkt var 3,17 g (94,6% utbytte). Tynnsjiktskromatografi [kloroform:etylacetat (1:1 y/v)]
Rf 0,09.
EKSEMPEL 6.
Fremstilling av Boc- glycyl- fenylaranyl- metyl- Y-am i no but yr a t.
Den ovennevnte forbindelse ble fremstilt fra 2,55 g (8,49 mmol) av dipeptidet i eksempel 4, 3,47 g (9,77 mmol,
15% overskudd) Boc-Gly-OCP og 1,1 ml (9,9 mmol) N-metylmorfolin i 17 ml dimetylformamid ved å følge metoden i eksempel 5. Ved opparbeidelse som i. eksempel 5 ble produktet renset ved lågtrykks-væskekromatografi under'anvendelse av. 95% kloroform/5% "Skellysolve B" til 100%, etylacetat-lineær gradient på en 25 x 100 mm siliciumdioksydgel-kolonne for oppnåelse av 2,52 g (70,4%) produkt. Tynnsjiktskromatografi [kloform: metanol:eddiksyre:vann (83:15:1:1 v/v/v/v] Rf 0,74;. Tynnsjiktskromatografi [kloroform:etylacetat (1:1 v/v)] Rf 0,06.
EKSEMPEL 7.
Fremstillirig av Glycyl- fenylalanyl- metyl- £- amino-kaproathydroklorid.
Forbindelsen i eksempel 5 (3,17 g, 7,05 mmol) ble oppløst i 26 ml eddiksyre og behandlet ved romtemperatur med 13 ml eddiksyre og behandlet ved romtemperatur méd 13: ml 5,7 M saltsyre/dioxan i 25 minutter. Oppløsningsmidlene ble inndampet og det resulterende krystallinske, faste stoff bel vasket med eter og deretter tørket i vakuum véd 60°C
i 2,5 time, og dette ga 2,68 g (98,6% utbytte). Tynnsjiktskromatografi [kloroform:metanol:eddiksyre:vann (83:15:1:1, v/v/v/v)] ga en flekk, Rf 0,09.
EKSEMPEL 8.
Fremstilling av glycyl- fenylalanylmetyl- y-ami no bu tyr a thydr ok1or id.
Boc-peptidet i eksempel 6 (2,52 g, 5,99 jnmol) ble oppløst i 22 ml eddiksyre og behandlet ifølge metoden i eksempel 7 med 10,7 ml 5,6 N saltsyre i dioxan. Etter opparbeidelse og tørking i vakuum ved 55°C i tre timer, ble det oppnådd et produkt i et utbytte på 98,6% (2,14 g). Elementær analyse viste at det var et 1/4 hydrat. TLC [metanol:eddik: syre:vann:kloroform (15:1:1:83 v/v/v/v)], Rf 0,07 (homogen).
EKSEMPEL 9.
Fremstilling av Boe-( DL)- 2, 6, C- dimétyltyrosyl-( D) -
métionyl glycyl- fenylaTanyl- métyl- Y- aminobtyrat..
En oppløsning av (DL) Boe-2,6-C-dimetyltyrosyl-(D)-metionin (l,37g, 3,05 mmol) i .18.>5 ml dimetylformamid; og 0,72 ml (6,48 mmol) N-metylmorfin bie'avkjølt til -60°C og 0,42 ml (4,20 mmol, 5% overskudd) iso-butylklorformeat ble.tilsatt
på en gang. Oppløsningen ble omrørt i 10 minutter mens temperaturen steg til -50°C, hvoretter 1,21 g (3,35 mmol) av tripeptidhydrokloridet i eksempel 8 i 9 ml dimetylformamid ble tilsatt i løpet av 20 minutter, mens temperaturen ble holdt ved under -20°C. Réaksjonsblandingen ble omrørt ved ca. -10°C i 30 minutter, og fikk deretter oppvarmes til romtemperatur og holdt ved romtemperatur i 20.timer.
Reaksjonsblandingen ble deretter tilsatt til 300 ml kald..5% kaliumbisulfat, og dette ga et hvitt bunnfall som ble filtrert, vasket med vann og tørket til 1,97 g (86,8% utbytte) produkt, Tynnsjiktskromatografi [kloroformrmétanol: eddiksyre:vann (83:15:1:1 v/v/v/v)] - to flekker med lik intensitet (diasteromerér) ved R^ 0,70 og 0,76.
EKSEMPEL 10.
Fremstilling av Boe-( DL)- 2, 6- C- dimetyltyrosyl-( D)-me tiony1- glycy1- fe ny1alany1- méty1- e- amihokaproat.
Tittelforbindelsen ble fremstilt fra 1,29 g (3,35 mmol) av tripeptidhydrokloridet i eksempel 7, og de samme molare overskudd av Boe-(DL)-2,6-C-dimetyltyrosyl-(D) metip-nin, N-metylmorfolin og iso-butylkloroformeat og oppløsnings-middel ble tilsatt på samme måte som i eksempel 9. Opparbeidelse som i eksempel 9 ga 2,19 g produkt(93,2%) . > T<y>nnsj ikts-kromatograf i i oppløsningsmiddelsystemet i eksempel 9 ga to flekker med lik intensitet, Rf 0,64 og 0,72.
EKSEMPEL 11.
Fremstilling av ( DL) 2, 6- D- dimetyltyrosyl—( D)-metionyl- glycyl- fenylalanyl- metyl- Y- amihobutyrat-hydroklorid.
Det beskyttede peptid i eksempel 9 (1,97 g, 2,65 mmol) ble behandlet i 20 minutter ved romtemperatur i 10 ml eddiksyre og 5 ml 5 M saltsyre i dioxan, og oppløsningsmidlene ble inndampet. Tilsetning av eter og triturering ga et pul-ver som ble filtrert, vasket med eter og tørket i vakuum ved 60°C i 3,5 time, og dette ga 1,81 g (96,8% utbytte) produkt som et hydroklorid 1-1/2 H90, molekylvekt 707,28 [a] 25+5,0° 7

(c 0,1, metanol). Elementæranalyse og NMR-analyse bekreftet strukturen.
EKSEMPEL 12.
Fremstilling av ( DL)- 2, 6- C- dimetyltyrosyl-( D)-metionyl-<g>l<y>c<y>l- fen<y>laiån<y>l- metylkaproathydroklorid.
Det beskyttede pentapeptid i eksempel 10 (2,19 g, 2,84 mmol) ble behandlet som ovenfor med saltsyre i dioxan, og dette ga etter tørking ved 60°C i 2-1/2 time i vakuum 1,89 g (91,9%) produkt som et hydroklorid 1-1/2 H20.;

Farmasøytiske preparater omfatter som aktiv bestanddel minst en av forbindelsene med formel I i forbindelse med en farmasøytisk bærer eller fortynningsmiddel. I tilfelle av kapsler, tabletter og piller, kan doseringsfor- mene også omfatte buffermidler. Tabletter og piller kan i tillegg fremstilles med enteriske belegg.
Flytende doseringsformer for oral administrasjon omfatter farmasøytisk akseptable emulsjoner, oppløsninger, suspensjoner, siruper og eleksirer inneholdende inerte fortynningsmidler som vanlig benyttet, slik som vann. Foruten inerte fortynningsmidler, kan slike preparater også inneholde hjelpestoffer slik som fuktemidler, emulgerings- og suspen-sjonsmidler, samt søtnings-, smak- og parfymeringsmidler.
Preparater for parenteralt administrasjon omfatter sterile vanndige eller ikke-vanndige oppløsninger, suspensjoner eller emulsjoner. Eksempler på ikke-vanndige oppløsnings-midler eller bærere er propylenglykol, vegetabilske oljer, slik som olivenolje, og injiserbare", organiske estere, slik som etyloleat. Slike doseringsformer kan også inneholde hjelpestoffer slik som preserverings-, fukte-, emulgerings-og dispergeringsmidler. De kan steriliseres ved.f.eks. filtrering gjennom et bakterie-tilbakeholdende filter, ved in-korporering av steriliseringsmidler i preparater,; De kan også fremstilles i form av sterile, faste preparater som kan opp-løses i sterilt vann, eller et annet sterilt, injiserbart me-dium umiddelbart før bruk.
Doseringen av aktiv bestanddel i preparatene kan varieres, men det er nødvendig at mengden av den aktive bestanddel er slik at en egnet doseringsform oppnås. Den valgte dose avhenger av den ønskede terapeutiske effekt, administrasjonsmåte og behandlingens varighet. Generelt administreres doseringsnivåer på mellom 0,5 og 5 mg/kg legemsvekt daglig til pattedyr for oppnåelse av' effektiv smertelindring.
Claims (10)
1. Enkefalinderivat, karakterisert ved formelen:
hvor C er metionin, alanin eller nor-leucin; m er .3 eller 5; og R er valgt fra-gruppen bestående av hydrogen, .lavere alkyl eller -NR2R3; hvor R2 og R3 er like eller forskjellige fra gruppen bestående av hydrogen og laverealkyl; og hvor
(-) tegner den viste forbindelse, dens speilbilde eller en blanding av racemater; samt farmasøytisk akseptable salter deravi
2. Forbindelse ifølge krav 1, karakterisert ved at m er 3.
3. Forbindelse ifølge krav 1, karakterisert ved at m er 3 og R er. hydrogen.
4. Forbindelse ifølge krav 2, karakterisert ved R er laverealkyl.
5. Forbindelse ifølge krav 4, karakterisert ved at den utgjøres, av (DL)-2,6-C-dimetyltyrosyl- (D)metionyl-glycyl-feny;ialanyl-metyl-Y-aminobutyrat eller et farmasøytisk akseptabelt salt derav.
6. Forbindelse ifølge krav 2, karakterisert ved at R er NR2 R3 .
7. Forbindelse ifølge krav 1, karakterisert ved at m er 5 .
8. Forbindelse ifølge krav 7, karakterisert ved at R er hydrogen.
9. Forbindelse ifølge krav 1, karakterisert ved at den utgjøres av: (DL)-2,6-C-dimetyltyro syl-CDjmetionyl-glycyl-fenylalanyl-metyl e-aminokaproat eller et farmasøytisk akseptabelt salt derat.
