Description
Oppfinnelsens bakgrunn
Foreliggende oppfinnelse gjelder fremgangsmåte for fremstilling av et renset rekombinant human-IL-3-analogprotein som omfatter Met<3>huIL-3 (Pro<8>Asp15Asp<70>) med gunstige kliniske og toksikologiske egenskaper sammenlignet med huIL-3-proteiner fremstilt i E. coli. Videre gjelder oppfinnelsen isolert DNA-sekvens, rekombinant ekspresjonsvektor og gjærvertscelle for anvendelse ved fremgangsmåten.
Differensiering og proliferasjon av hematopoietiske celler reguleres av utskilte glykoproteiner kjent under fellesbetegnelsen kolonistimulerende faktorer (CSF). I systemer hos mus og menneske omfatter disse proteiner granulocytt-makrofagkolonistimulerende faktor (GM-CSF), som fremmer produksjon av granulocytter og makrofager i normal benmarg og som videre ser ut til å regulere aktiviteten av modne, diffe-rensierte granulocytter og makrofager. Andre CSF omfatter makrofag-CSF (M-CSF eller CSF-1), som induserer selektiv proliferasjon av makrofager og granulocytt-CSF (G-CSF), som induserer utvikling av granulocyttforløpere fra forstadier i benmarg. Ytterligere en CSF, først isolert fra musesystemer og senere med humane celler som kilde, er blitt kalt IL-3 eller multi-CSF.
Muse-IL-3 ble opprinnelig identifisert av Ihle et al., J. Immunol. 126:2184 (1981), som en faktor som induserte ekspresjon av et T-celleassosiert enzym, 20a-hydroksysteroid-dehydrogenase. Faktoren ble renset til homogenitet og vist å regulere vekst og differensiering av en rekke subklasser av tidlige hematopoietiske og lymfoide forløperceller. cDNA-kloner som tilsvarte muse-IL-3, ble først isolert av Fung et al., Nature 307:233 (1984), og Yokota et al., Proe. Nati. Acad. Sei. USA 81:1070 (1984). Genomiske DNA-homologer av muse-IL-3-sekvensen fra gibbonape og menneske ble beskrevet av Yang et al., Cell 47:3 (1986). Den humane sekvens beskrevet av Yang et al. omfattet en serinrest i posisjon 8 i den modne proteinsekvens. Etter dette funn rapporterte tre grupper isolering av Pro<8>huIL-3-cDNA, omfattende Dorssers et al., Gene 55:115 (1987); Otsuka et al., J. Immunol. 140:2288 (1988); og Gillis et al., Behring Inst. Mitt. 83:1 (1988).
En undersøkelse av individer for å bestemme frekven- sen av Pro<8->allelet ble beskrevet av Gillis et al., se ovenfor. I dette arbeidet ble polymerasekjedereaksjonen benyttet til amplifisering av DNA-sekvensene som flankerer posisjon 8-locuset. Radioaktivt merkede oligonukleotidprober komplementære til Ser<8->og Pro<8->formene ble så benyttet til analyse av amplifisert DNA isolert fra 13 genetisk ubeslektede individer. Amplifisert DNA ble immobilisert på nitrocellulose og analysert ved hybridisering under betingelser med økende stringens. Resultatene viste at alle de 13 undersøkte personer var positive for DNA som koder for Pro<8->versjonen av huIL-3. Tre var i tillegg positive for Ser<8>huIL-3 i dette locus (dvs. at de var heterozygoter), noe som indikerer et moderat poly-morfinivå for det humane IL-3-gen i den analyserte populasjon.
Prekliniske og kliniske undersøkelser av humane IL-3-proteiner fremstilt av forskjellige produsenter har avslørt overraskende forskjeller i klinisk anvendelighet og toksisitet. Utvikling av ikke-giftige, tolererbare former av humant IL-3 til terapeutisk bruk er viktig, da rekombinant, humant IL-3 synes å være det første cytokin som kan stimulere granu-lopoiese, erytropoiese og, mest viktig, trombopoiese in vivo.
Alvorlig giftighet av rIL-3 ble beskrevet av Valent et al., ved "First International Symposium: Cytokines in Hemo-poiesis, Oncology and AIDS", holdt i Hannover, Forbundsrepu-blikken Tyskland, 14.-17. juni 1989. Et huIL-3(Ser<8>)-preparat fremstilt i E. coli ble vurdert in vivo ved tilførsel til rhesusaper (n = 10) tre ganger daglig ved forskjellige doser (0, 11, 33 og 100 ug/kg/dag, s.c.) i 14 dager med påfølgende tilførsel av GM-CSF (dagene 14-28, 5 ug/kg tre ganger daglig). Alle aper responderte på rhuIL-3 med en 2-3 gangers økning av hvite blodceller (WBC) ved dag 14 grunnet økning av eosinofiler og basofiler. Intracellulært histamin (IH) i WBC ble vist å øke kontinuerlig etter IL-3-behandling frem til dagene 8-10. Dette overskudd av IH ble fulgt av en økning av hista-minverdier i plasma og et neslefeberlignende utslett som tydet på infiltrering av mastceller og lymfocytter i dermis, observert i hudbiopsiprøver. De kliniske bivirkninger av dette rhuIL-3-preparat ble tilskrevet aktivering av basofiler og mastceller og de histaminproduserende virkninger av dette cytokin.
I motsetning til dette ga analogproteinet fremstilt ifølge foreliggende oppfinnelse ingen påvisbar neslefeber eller stimulering av mastcelle- eller lymfocyttinfiltrering i dermis ved tilførsel til cynomolgusaper eller humane pasienter. Resultatene av disse toksikologiske og kliniske under-søkelser gis i eksemplene.
Sammendrag av oppfinnelsen
Foreliggende oppfinnelse tilveiebringer en fremgangsmåte for fremstilling av et renset rekombinant human-IL-3-analogprotein som omfatter Met<3>huIL-3 (Pro<8>Asp15Asp70),
og fremgangsmåten er kjennetegnet ved at en gjærvertscelle som inneholder en ekspresjonvektor omfattende en DNA-sekvens som koder for human-IL-3-analogproteinet, dyrkes under betingelser som fremmer ekspresjon av proteinet.
Ved tilførsel til primater gir ikke slike analogproteiner påvisbar neslefeber eller mastcelle- og lymfocyttinfiltrasjon i dermis.
Videre tilveiebringer oppfinnelsen en isolert DNA-sekvens kjennetegnet ved at den koder for et human-IL-3-ana-logprotein fremstilt ifølge krav 1, en rekombinant ekspresjonsvektor kjennetegnet ved at den omfatter en DNA-sekvens ifølge krav 3 og en gjærvertscelle kjennetegnet ved at den inneholder en ekspresjonsvektor ifølge krav 4.
Detaljert beskrivelse av oppfinnelsen
huIL-3-analogproteinene fremstilt ifølge foreliggende oppfinnelse omfatter utvalgte, forkortede huIL-3-analogproteiner med små variasjoner i den N-terminale ende sammenlignet med det native protein. Slike forkortede proteiner ble påvist som bestanddeler i proteinblandinger fremkommet ved ekspresjon av cDNA som koder for modne huIL-3-proteiner i S. cerevisiae, antagelig som et resultat av proteolyse av gjærekspresjons-verten. Disse blandinger kan inneholde ett eller flere humane IL-3(Pro<8>Asp15Asp<70>)-analogproteiner med N-terminal ende ved Met<3>, Thr4 eller Thr6 i den følgende aminosyresekvens (de alternative N-terminale ender er understreket):
Begrepet rhuIL-3 (Pro<8>Asp<15>Asp70)betegner et rekombinant, humant interleukin-3-polypeptid med en prolinrest i posisjon 8 og asparaginsyrerester i posisjonene 15 og 70, hvor rest 1 viser til den første aminosyre i det modne, native eller villtypehumane interleukin-3. Aminosyresekvensen til et slikt polypeptid er vist ovenfor. De forskjellige N-terminale arter angis ved å benevne den N-terminale rest foran begrepet "rhuIL-3", f.eks. Met<3>rhuIL-3 (Pro<8>Asp<15>Asp<70>). Analogproteinene fremstilt ifølge foreliggende oppfinnelse har en asparagin-syrerest (Asp) som erstatning for hver asparaginrest i N-glykosyleringssetene som finnes i den native, humane IL-3-sekvens for å forhindre N-glykosylering av gjærverten i disse seter.
Et DNA-segment som koder for humant IL-3, ble isolert fra et cDNA-bibliotek som beskrevet i den eksperimentelle av-deling nedenfor. Setespesifikk oligonukleotidmutagenese ble så utført for å fremskaffe et cDNA som koder for humant IL-3 (Pro<8>Asp<15>Asp<70>). Det resulterende endrede cDNA ble benyttet til fremstilling av en gjærekspresjonsvektor som ble benyttet til transformering av en passende gjærekspresjonsstamme som ble dyrket i kultur under betingelser som fremmer derepressering av gjærpromotoren. Det resulterende protein ble så renset ved en kombinasjon av reversfase-høytytende væskekromatografi (RPHPLC) og ionebytterkromatografi slik at et renset produkt ble erholdt. Analyse av dette produkt ved hjelp av et analyse-system for proliferasjon av human benmarg og ved analyse av binding til human IL-3-reseptor bekreftet ekspresjon av et analogprodukt egnet til farmasøytisk bruk i mennesket. N-terminal sekvensering av produktet viste en blanding av analogproteiner med lette variasjoner fra parti til parti. For å oppnå et preparat med i det vesentlige en enkelt art, Met<3>huIL-3 (Pro<8>Asp<15>Asp<70>), ble en ny ekspresjonsvektor fremstilt hvor sekvensen som koder for ledersekvensen for sekresjon i gjær, ble direkte satt inn i samme leseramme slik at et konstrukt som mangler kodonene for de første to aminosyrer i det modne protein, mangler.
Preparater som inneholder analogproteiner fremstilt ifølge oppfinnelsen, består fortrinnsvis i det vesentlige av Met<3>huIL-3 (Pro<8>Asp<15>Asp<70>). Hvis en ekspresjonsvektor som koder for det modne protein i sin hele lengde benyttes, erholdes imidlertid en blanding som består av Ala<1->huIL-3 (Pro<8>Asp15Asp70)blandet med ett eller flere proteiner valgt fra den foregående gruppe, typisk bestående av: (a) fra tilnærmet 30 til tilnærmet 40 % Ala^uIL-3 (Pro<8>Asp<15>Asp<70>), (b) fra tilnærmet 10 til tilnærmet 30 % Met<3>huIL-3 (Pro<8>Asp<15>Asp70), og (c) en restfraksjon som i det vesentlige består av en blanding av Thr<4->og Thr<6>huIL-3 (Pro<8>Asp15Asp70).
Preparater som inneholder analogproteiner fremstilt ifølge oppfinnelsen viser fortrinnsvis en spesifikk biologisk aktivitet på fra tilnærmet 1,8 til tilnærmet 7,0 x IO<7>U/mg i analysesystemet med human benmarg, fortrinnsvis 4,0 til tilnærmet 7,0 x IO<7>U/mg, og en bindingsaffinitet til humane IL-3-reseptorer fra monocytter, uttrykt som en inhiberingskonstant, på fra tilnærmet 2,0 til tilnærmet 8,0 x1010NT<1>, fortrinnsvis fra tilnærmet 4,0 til tilnærmet 8,0 x 10<10>M"1.Det innses at mindre fraksjoner (mindre enn 1 mol%) av rhuIL-3-proteiner med andre N-terminale ender også kan være til stede i blandingene ifølge foreliggende oppfinnelse.
1. Analyser av biologisk aktivitet for huIL- 3
Analyser benyttet til måling av biologisk aktivitet for huIL-3 er beskrevet nedenfor.
A. Proliferasjonsanalyse med human benmarg Nyisolerte humane benmargsceller preinkuberes i 2 timer ved 37 °C og 5 % C02i vevsdyrkingsflasker med 2 x IO<6>celler pr. ml forvarmet og forgasset, serumfritt RPMI 1640-medium (Gibco, Chagrin Falls, OH, USA) med 50 enheter/ml peni-cillin, 50 pg/ml streptomycin og 300 ug/ml fersk L-glutamin (heretter kalt "analysemedium"). Etter preinkubasjonen fjernes ikke-adherente celler ved forsiktig pipettering av mediet over flaskens overflate. Ikke-adherente celler oppsamles ved sentrifugering ved 1000 rpm i 10 minutter ved 4 °C, resuspenderes i et lite volum analysemedium med 10 % føtalt, bovint serum (FBS) og telles ved hjelp av trypanblått for viabilitets-analyse og Turks farge for gjenvinning av hvite celler. Cellene oppbevares ved tilnærmet 4 °C i analysemedium med 10 % FBS inntil de tilsettes analyseskålene. 50 ul analysemedium tilsettes hver brønn i en 96-brønners flatbunnet vevsdyrkingsplate. 50 pl prøve fortynnet i analysemedium tilsettes den første brønn i hver rad, og serie-fortynninger utføres over hver rad på vanlig måte. 1,25 x 10<4>benmargsceller i et volum på 100 ul tilsettes så hver brønn. Platene inkuberes i 4 dager ved 37 °C og 5 % C02i en plastikkboks med sterilt, destillert H20 for å forhindre uttørking. På dag 4 tilsettes 25 ul analysemedium med 5 % FBS og 80 uCi/ml [<3>H]-thymidin (80 Ci/mmol) til hver brønn, og platen inkuberes i 5 timer ved 37 °C og 5 % C02i en plastikkboks. Etter inku-beringen høstes cellene på glassfiberfiltre, vaskes og analyseres for inkorporert radioaktivitet med en scintillasjons-teller. Enheter av huIL-3-aktivitet beregnes ved referanse til den mengde huIL-3 som induserer 50 % av maksimal thymidininkorporering. Hvis f.eks. en 100 ul prøve gir halvparten av maksimal thymidininkorporering ved en fortynning på 1:20, defineres 1 enhet som aktiviteten i 1/20 av 100 ul, eller 5 pl. Prøven vil således inneholde 1000 delt på 5, eller 200 enheter pr. ml (U/ml) med huIL-3-aktivitet.
B. Analyse av binding til human IL- 3- reseptor
I en 96-brønners, rundbunnet mikrotiterplate frem-stilles ti 2-gangers fortynninger av hver prøve i RPMI 1640-medium med utgangskonsentrasjon 0,075 pg/ml (50 pl hver). For hver analyseplate settes 50 pl aliquoter av rhuIL-3-standarder (1,5 pg/ml) og negative kontroller (bindingsmedium) opp i triplikat. I forbindelse med denne analysen betyr "bindings medium" RPMI 1640 med 2,5 % (vekt/vol) bovint serumalbumin (BSA), 0,2 % (vekt/vol) natriumazid og 20 mM HEPES, pH 7,2.
50pl12<5>I-merket rhuIL-3-lagerløsning (1 x IO<9>M i bindingsmedium, spesifikk aktivitet 4-8 x 10<15>cpm/mmol), fremstilt som beskrevet i PCT-søknad US 89/02599, tilsettes så hver brønn, fulgt av 50 pl humane monocytter (høstet fra kultur ved sentrifugering, vasket to ganger med RPMI 1640 og resuspendert ved 4 x IO<7>celler/ml i bindingsmedium). Hver plate inkuberes så i 1 time ved 37 "C på en rotatorisk ristemaskin under kraftig sammenblanding. Etter inkubasjonen skilles celler og bundet<125>I-rhuIL-3 fra ubundet<125>I-rhuIL-3 ved en ftalat-olje-sentrifugeringsteknikk utført som følger. Først fjernes 65 pl aliquoter i duplikat fra hver inkubasjonsblanding, og hver aliquot legges på toppen av 200 pl kald ftalat-oljeblanding [1,5 deler dibutylftalat, 1 del bis(-2-etylheksyl)-ftalat (Eastman Kodak Co., Rochester, NY)] i 400 pl polyetylensentri-fugerør og sentrifugeres i 1 minutt i en mikrosentrifuge ved 8 °C. Cellene pluss bundet12<5>I-rhuIL-3 sedimenterer gjennom
ftalat-oljeblandingen mens det mindre tette bindingsmediet med ubundet<125>I-rhuIL-3 gjenblir på overflaten.<125>I-rhuIL-3 bundet til celler måles ved å kutte rørene i to og måle<125>I i topp og bunn. Resultatene av analysen av binding til IL-3-reseptor uttrykkes som inhibering (%) beregnet i henhold til ligning 1 nedenfor:
hvor maks er nivået av binding av12<5>I-IL-3 i nærvær av medium alene, min er nivået av binding i nærvær av 1,5 pg/ml umerket, og test er bindingsnivået i nærvær av fortynninger av analyse-prøver. Resultatene analyseres ved ikke-lineær, minste kvadra-ters tilpasning, ligning 2 nedenfor:

2. Ekspresjon av protein i rekombinante aiærsvsterner
Rekombinante Saccharomyces cerevisiae vertsceller benyttes til ekspresjon av de rekombinante proteiner fremstilt ifølge foreliggende oppfinnelse. Foretrukne ekspresjonsvektorer kan avledes fra pBC102-K22 (ATCC 67255) som inneholder DNA-sekvenser fra pBR322 for seleksjon og replikasjon i E. coli (Ap<r->genet og replikasjonsorigo) og gjær-DNA-sekvenser omfattende en glukoserepresserbar alkoholdehydrogenase 2 (ADH2 )-promotorADH2-promotoren er blitt beskrevet av Russel et al., J. Biol. Chem. 258:2674 (1982) og Beier et al., Nature 300:724 (1982). Plasmid pBC102-K22 inneholder også et Trpl-gen som selekterbar markør og replikasjonsorigo fra 2 u-plasmidet i gjær. Ved siden av promotoren sitter ledersekvensen for gjær-a-faktor som tillater utskillelse av heterologe proteiner fra en gjærvert. Ledersekvensen for a-f aktor er modifisert slik at den nær sin 3' ende inneholder et Asp718-restriksjons-sete (Kpnl og Asp 718 er isoskisomere) for å lette fusjon av denne sekvens til fremmede gener. En sekvens som koder for aminosyrene Glu-Ala-Glu-Ala, ble utelatt for å tillate effek-tiv prosessering av utskilt protein, som beskrevet av Brake et al., Proe. Nati. Acad. Sei. USA 81:4642 (1984).
Alternative ekspresjonsvektorer er gjærvektorer som inneholder en a-faktorpromotor, f.eks. pYaHuGM (ATCC 53157), som inneholder villtypen av det humane GM-CSF-gen. Andre er kjent for fagfolk. Oppbyggingen av pYaHuGM er beskrevet i den publiserte europeiske patentsøknad nr. 183.350 (8530682.7), beskrivelsen i hvilken inkorporeres heri ved referanse.
Valg av passende gjærstammer for transformering vil avgjøres av de selekterbare markørers natur og andre egenskaper ved vektoren. Passende S. cerevisiae-stammer for transformering med ekspresjonsvektorer avledet fra pBC102-K22 eller pYaHuGM omfatter stamme X2181-1B, tilgjengelig fra Yeast Genetic Stock Center, Berkeley, CA, USA (se nedenfor) med genotypen atrpl gall adel his2; J17 (ATCC 52683; ahis2 adel trpl metl4 ura3); og IL166-5B (ATCC 46183; ahisl trp 1). En spesielt foretrukket ekspresjonsstamme for bruk med pBC102-K22, XV2181 er en diploid dannet ved kryssing av to haploide stammer, X2181-1B, tilgjengelig fra Yeast Genetic Stock Center, Department of Biophysics and Medical Physics, University of California, Berkeley, CA 94702, USA; og XV617-1-3B, tilgjengelig fra Department of Genetics, University of Washington, Seattle, WA 98105, USA, eller Immunex Corporation, 51 University Street, Seattle, WA 98101, USA. En egnet trans-formeringsmetode er den beskrevet av Hinnen at al., Proe. Nati. Acad. Sei. USA 75:1929 (1978), som selekterer for Trp<+->transformanter i et selektivt medium som består av 0,67 % gjærnitrogenbase, 0,5 % kasaminosyrer, 2 % glukose, 10 pg/ml adenin og 20 pg/ml uracil.
Vertsstammer transformert med vektorer som inneholder ADH2- eller a- faktorpromotorer dyrkes for ekspresjon i et rikt medium som består av 1 % gjærekstrakt, 2 % pepton og 1 % glukose tilsatt 80 pg/ml adenin og 89 pg/ml uracil. Derepressering av ADH2-promotoren oppnås når glukosen i mediet er opp-brukt. Urensede gjærsupernatanter høstes ved filtrering og fryses eller lagres ved 4 °C forut for videre rensing.
3. Rensing av IL- 3
Rekombinant huIL-3 oppnådd ved fermentering av gjærstammer kan renses ved en enkelt eller på hverandre følgende reversfase-HPLC-trinn på en preparativ HPLC-kolonne ved metoder analoge til dem beskrevet av Urdal et al., J. Chromatog. 296:171 (1984) og Grabstein et al., J. Exp. Med. 163:1405 (1986). En foretrukket rensingsprotokoll er beskrevet i eksempel 2 nedenfor.

Isolering av cDNA som koder for huIL- 3
Lymfocytter fra perifert blod ble isolert fra "buffy coats" fremstilt fra helblod (Portland Red Cross, Portland, Oregon, USA) ved Ficoll-hypaque-tetthetssentrifugering. T-celler ble isolert ved rosettering med 2-aminoetyltiouronium-bromidbehandlede røde blodceller fra sau. Celler ble dyrket i flasker på 175 cm<2>med 5 x IO<6>celler/ml i 18 timer i 100 ml RPMI, 10 % føtalt kalveserum, 50 uM (3-merkaptoetanol, 1 % fytohemagglutinin (PHA) og 10 ng/ml forbol-12-myristat-13-acetat (PMA). RNA ble ekstrahert ved guanidin-CsCl-metoden og poly A<+->RNA fremstilt ved oligo-dT-cellulosekromatografi (Maniatis et al., Molecular Cloning: A Laboratory Manual, Cold Spring Harbor, 1982). cDNA ble fremstilt fra poly A<*->RNA i det vesentlige som beskrevet av Gubler og Hoffman, Gene, 25:263-269 (1983). cDNA ble gjort dobbelttrådet med DNA-polymerase I, buttendet med T4-DNA-polymerase, metylert med EcoRI-metylase for beskyttelse av EcoRI-kløvingsseter i cDNA og ligert til EcoRI-linkere. Disse konstrukter ble kuttet med EcoRI for å fjerne alle unntatt én kopi av linkeren i hver ende av cDNA, ligert til EcoRI-kuttede og defosforylerte armer av bakteriofag AgtlO (Huynh et al., DNA Cloning: A Practical Approach, Glover, red. IRL Press, s. 49-78), pakket med Å-fagekstrakter (Stratagene, San Diego, CA, USA) i henhold til produsentens instruksjoner. 500.000 rekombinanter ble utsådd på E. coll-stamme C600hfl" og analysert ved standard "plakk"-hybridi-seringsteknikker ved bruk av følgende prober. ;To oligonukleotider ble syntetisert med sekvenser komplementære til utvalgte 5'- og 3'-sekvenser i huIL-3-genet. Den 5' probe som er komplementær til en sekvens som koder for en del av ledersekvensen til huIL-3, hadde sekvensen 5'-GAG-TTGGAGCAGGAGCA-GGAC-3' . Den 3'-probe, som tilsvarer et område som koder for aminosyrer 123-130 i det modne protein, hadde sekvensen 5'-GATCGCGAGGCTCAAAGTCGT-3'. Syntesemetoden var en standard automatisert triestermetode i det vesentlige lik den beskrevet av Sood et al., Nucl. Acids Res. 4:2557 (1977) og Hirose et al., Tet. Lett. 28:2449 (1978). Etter syntesen ble oligonukleotidene avblokkert og renset ved preparativ gelelek-troforese. For bruk som analyseprober ble oligonukleotidene radioaktivt endemerket med<32>P-ATP og T4-polynukleotidkinase ved bruk av teknikker lik dem beskrevet av Maniatis et al. E. ;coil-stammen benyttet for analyse av biblioteket var C600hfl" ;(Huynh et al., 1985, supra). ;Tretten positive "plakk" ble renset og probet på nytt og uavhengig med de to hybridiseringsprober. Elleve kloner hybridiserte til begge de to oligonukleotidene. cDNA-innskuddene fra en rekke positive rekombinante fager ble subklonet i et EcoRI-kuttet derivat (pGEMBL18) av standardkloningsvektoren pBR322 med en polylinker med et unikt EcoRI-sete, et BamHl-sete og en rekke andre unike restriksjonsseter. Et eksempel på en vektor av denne type, pEMBL, er beskrevet av Dente et al., Nucl. Acids Res. 11:1645 (1983) i hvilken promotorene for SP6-og T7-polymerase flankerer de mange kloningsseter.Nukleotid-sekvensen av utvalgte kloner ble bestemt ved hjelp av kjede-termineringsmetoden. Nærmere beskrevet gav delvis EcoRI-kutting av IGT10:IL-3 kloner 2, 3, 4 og 5, fragmenter med størrelser fra 850 basepar til 1000 basepar som ble separat subklonet inn i EcoRI-setet i pGEMBL18. Innskuddene i pGEMBL:rhuIL-3-subklonene ble sekvensert ved bruk av en uni-versell primer som bindes ved siden av det multiple klonings-sete i pGEMBL18, og syntetiske oligonukleotidprimere avledet fra huIL-3-sekvensen. ;Eksempel 1 ;Modifisering av N- glykosvleringsseter kodet for av huIL- 3- cDNA og sammensetning av ekspresjonsvektor for rhuIL- 3 ( Pro8Asp15-Asp70);De to asparaginknyttede glykosyleringsseter som finnes i det naturlige protein (Asn<15>og Asn<70>), ble endret ved å forandre kodonene i disse posisjoner til kodoner som koder for asparaginsyre. Dette hindrer N-bundet glykosylering (ofte hyperglykosylering) av det utskilte protein av gjærcellene, slik at et mer homogent produkt erholdes. I tillegg ble et 514 basepar stort DNA-fragment avledet fra den enkelttrådete bakteriofag fl med replikasjonsorigo og den intergeniske region innsatt ved Nrul-setet i pBR-322-DNA-sekvensene. Nærvær av f1-replikasjonsorigo tillater dannelse av enkelttrådete kopier av vektoren ved transforma-sjon inn i passende (hannlige) stammer av E. coil og superinfeksjon med bakteriofag fl. Denne evne enkler DNA-sekvensering av vektoren og tillater muligheten av in vitro-mutagenese. ;Gjærekspresjonsvektoren pIXY120 ble kuttet med restriksjonsenzymene Asp718, som kløver nær den 3' ende av a-faktorlederpeptidet (nukleotid 237), og BamHl, som kløver i polylinkeren. Det store vektorfragment ble renset og ligert til følgende DNA-fragmenter: (1) et huIL-3-cDNA-fragment avledet fra plasmid GEMBL18:huIL-3 fra Clal-setet (nukleotid 58 i modent huIL-3) til BamHl-setet (3' for huIL-3-cDNA i en polylinker); og (2) følgende syntetiske oligonukleotidlinker ;A: ; ;
Oligonukleotid A gjendanner sekvensen som koder for den C-terminale ende av a-faktorlederpeptidet og fuserer det i leserammen med oktapeptidet DYKDDDDK, som i sin tur fuseres til den N-terminale ende av modent rhuIL-3. Denne fusjon til rhuIL-3-proteinet tillater påvisning med antistoff spesifikt for oktapeptidet og ble opprinnelig benyttet for måling av ekspresjon og rensing av rhuIL-3. Dette oligonukleotid koder også for en aminosyreforandring i posisjon 15 (Asn<15>til Asp<15>) for å endre dette N-knyttede glykosyleringssete. De under-strekte nukleotider i oligonukleotid A representerer forandringer fra villtype-cDNA-sekvensen. Bare A til G- og C til T-endringene ved hhv. nukleotid 43 og 45 (telt fra kodonet som tilsvarer det N-terminale alanin i det modne hulL-3-molekyl) resulterer i en aminosyreendring (Asp<15>). De øvrige baseend-ringer introduserer nyttige restriksjonsseter (Ahall og PvuII) uten å endre aminosyresekvensen. Det resulterende plasmid ble kalt pIXY139 og inneholder et rhuIL-3-cDNA med én gjenværende konsensussekvens for N-bundet glykosylering (Asn<70>). ;Plasmid pIXY139 ble benyttet for utførelse av oligo-nukleotidstyrt mutagenese for å fjerne den andre konsensussekvens for N-bundet glykosylering ved endring av Asn<70>tilAsp<70>. In vitro-mutagenesen ble utført ved en metode lik den beskrevet av Walder og Walder, Gene 42:133 (1986). Gjærtransformanter ble selektert ved dyrking i medium som manglet tryptofan, slått sammen i puljer og DNA ekstrahert som beskrevet av Holm et al., Gene 42:169 (1986). Dette DNA som inneholdt en blanding av villtype- og mutert plasmid-DNA, ble benyttet til transformering av E. coli RR1 til ampicillinresistens. De oppnådde kolonier ble analysert ved hybridisering til radioaktivt merket oligonukleotid B ved bruk av gjengse teknikker. Plasmider som inneholdt DNA som koder for huIL-3 Asp<70>, ble identifisert ved hybridisering til radioaktivt merket oligonukleotid B under stringente betingelser og bekreftet ved nukleotidsekvensering. ;Det resulterende gjærekspresjonsplasmid ble kalt pIXY138 og inneholdt huIL-3-genet som koder for Asp<15->og Asp<70->aminosyreendringene og oktapeptidet DYKDDDDK i den N-terminale ende. Det endelige gjærekspresjonsplasmid er identisk med pIXY138, bortsett fra at det mangler nukleotidsekvensene som koder for oktapeptidet og følgelig gir modent rhuIL-3 som produkt. ;Det endelige gjærekspresjonsplasmid ble fremstilt som beskrevet nedenfor. Gjærekspresjonsvektoren pIXY120 ble kuttet med restriksjonsenzymene Asp718 og BamHl, som beskrevet ovenfor. Oligonukleotid C gjendanner den 3' ende av a-faktorlederpeptidet fra Asp718-setet (aminosyrene Pro-Leu-Asp-Lys-Arg) og de N-terminale 7 aminosyrer i huIL-3 til Ahall-setet. Det resulterende plasmid ble kalt pIXY151. I gjær tillater denne vektor glukoseregulert ekspresjon og utskillelse av rhuIL-3 (Pro<8>Asp<15>Asp<70>). For å tilveiebringe relativt homogene preparater som i det vesentlige består av Met<3>rhuIL-3 (Pro<8->Asp15Asp<70>), ble en lignende ekspresjonsvektor kalt pIXY290 fremstilt ved å bruke et oligonukleotid identisk med oligonukleotid C, men uten kodonene som spesifiserer Ala- og Pro-restene til stede i det native, humane IL-3-protein. Ved transfeksjon inn i en egnet vert, som så dyrkes under passende betingelser, er denne vektor i stand til å dirigere ekspresjon av rhuIL-3-preparater som i det vesentlige består av Met<3>rhuIL-3(Pro<8>Asp15Asp<70>)-produktet (se eksempel 3 nedenfor). ;Eksempel 2 ;Ekspresjon og rensing av rhuIL- 3( Pro8Asp15Asp70)- blanding ;Vertsstammen XV2181, en diploid S. cerevlsiae-stamme ble dannet ved krysning av XV617-1-3B [a, his6, leu2-l, trpl-1, ura3, ste5] erholdt fra University of Washington, Depart ment of Genetics Yeast Strain Bank, Seattle, WA, USA, og X2181-1B [a, trpl-1, gall, adel, his2] erholdt fra Yeast Genetic Stock Center, University of California, Berkeley, CA, USA. Vertsstammen transformeres med ekspresjonsplasmidet ifølge fremgangsmåten til Sherman et al., Laboratory Course Manual for Methods i Yeast Genetics, Cold Spring Harbor Laboratory, 1986. ;Gjær med ekspresjonsplasmid pIXY151 (se eksempel 1 ovenfor) oppbevares på YNB-trp-agarskåler lagret ved 4 °C. En forkultur startes ved inokulering av flere isolerte rekombinante gjærkulturer i 1 liter YNB-trp-medium (6,7 g/l gjærnitrogenbase, 5 g/l kasaminosyrer, 40 mg/l adenin, 160 mg/l uracil og 200 mg/l tyrosin) og dyrkes over natten i to 2-liters flasker ved 30 °C under kraftig risting. Om morgenen er kulturen mettet, i stasjonær fase, ved en OD600på 2 til 7. De på forhånd rensede og steriliserte fermentorer (tre maskiner med 10 liters arbeidsvolum) fylles til 80 % av sin arbeidskapasitet med SD-2-medium (4,0 g/l ammoniumsulfat, ;3,2 g/l monobasisk kaliumfosfat, 3,0 g/l gjærekstrakt, 1,0 g/l sitronsyre, 0,1 g/l natriumklorid, 5 ml/l 2 % kalsiumklorid, 2,5 ml/l vitamin 101-løsning, 0,5 ml/l sporelementløsning, ;0,5 ml/l 20 % magnesiumsulfat, 2,0 ml/l glukose) og holdes ved 30 °C med røring ved 500-600 rpm og 10-16 l/minutt luftgjen-nomstrømning. Forkulturen tilsettes. Etter 2 timers dyrking begynnes næringstilførsel i form av 50 % glukose med en hastighet hvorved 50 g/l tilsettes over et tidsrom på 10-12 timer. Næringstilførselen endres så til 50 % etanol tilsatt med 30-40 ml/time inntil høsting. ;Det totale tidsforløp for fermenteringen er tilnærmet 20 timer, hvoretter den optiske tetthet (600 nm) varierer mellom 30 og 45. Fermentorene kjøles så ned til 20 °C, pH i gjærbrygget justeres til 8,0 ved tilsetning av 5 M NaOH, og det resulterende materiale filtreres gjennom et Millipore Pellicon-filtersystem utstyrt med en 0,45 pm filterkassett og oppsamles i en steril 10 liters glassballong. ;Det rhuIL-3 som forefinnes i gjærbrygget, påsettes en 5 cm x 30 cm kolonne pakket med 15-20 pm C-4 reversfasekisel (Vydac, Separations Group, Hesperia, CA, USA) ved å pumpe det filtrerte gjærbrygg direkte på kolonnen. Kolonnen ekvilibreres med 0,1 % trifluoreddiksyre i vann (løsemiddel A) forut for påsetting av gjærbrygget og vaskes med dette løsemiddel etter påsettingen av brygget på søylen inntil den optiske absorbans av det eluerte materiale nærmer seg baselinjeverdier. Etter dette etableres en gradient av 0,1 % trifluoreddiksyre i acetonitril (løsemiddel B) fra 0 % B til 100 % B ved en endringshastighet på 2 % B pr. minutt og en strømningshastig-het på 100 ml/minutt. En pause på 5 minutter når gradienten når 30 % B, er programmert inn i gradientkontrollenheten. 20 minutter etter start av gradienten oppsamles fraksjoner på 1 minutt. Aliquoter av fraksjonene analyseres for proteininnhold ved en fluorescaminanalyse med bovint serumalbumin (BSA) som standard. rhuIL-3 viste seg å elueres i fraksjoner 8, 9 og 10. Fraksjonene med rhuIL-3 fra første trinn lagres ved 4 °C i polyetylenflasker inntil tilnærmet 1 g totalprotein erholdes. Til det sammenslåtte protein tilsettes to volumer 0,1 % trifluoreddiksyre i vann. Denne løsning pumpes så på en ny 5 cm x 30 cm kolonne pakket med 15-20 u C-18-kisel (Vydac, Separations Group, Hesperia, CA, USA) som er ekvilibrert med løsemiddel A (0,1 % TFA i vann). Etter påsetting av materialet vaskes kolonnen med løsemiddel A, hvoretter en gradient identisk med den beskrevet ovenfor etableres med en hastighets-forandring på 1 % løsemiddel B pr. minutt og med en strøm-ningshastighet på 100 ml/minutt. Fraksjoner oppsamles hvert 0,4 minutt 24 minutter inn i gradienten. Aliquoter av fraksjonene analyseres for proteininnhold ved fluorescaminanalysen med BSA som standard. ;Toppfraksjoner fra C-18-kolonnen slås sammen og ;1/10 volum 0,5 M p-alanin, pH 3,6, tilsettes. En prøve tas ut, hvoretter de sammenslåtte fraksjoner påsettes en 10 ml "S-Sepharose"-kolonne (1 cm x 10 cm, Pharmacia) ved en strøm-ningshastighet på 5 ml/minutt. Etter påsetting av prøven vaskes kolonnen med 50 ml 10 mM tris, pH 7,4, og IL-3 elueres med en lineær gradient fra 10 mM tris, pH 7,4, til 200 mM tris, pH 7,4 (200 ml totalt gradientvolum). Toppf raks joner med rhuIL-3 slås så sammen og dialyseres mot 100 mM tris, pH 7,4, over natten ved 4 "C og sterilfiltreres. ;De biologiske aktiviteter (enheter/mg) og bindings-affinitetene til tre produksjonspartier med rhuIL-3, fremstilt i det vesentlige som beskrevet ovenfor, ble bestemt. Bindings-affiniteten ble fastsatt ved bestemmelse av inhiberings-konstanter (Kx) med rensede proteiner. Resultatene er gitt i tabell I nedenfor. I tabell I er alle verdier uttrykt som gjennomsnittet for prøver analysert i triplikat i to separate analyser. ;rhuIL-3 fra disse tre partier (AZZ-0001-0003) ble også gjort gjenstand for proteinsekvensering ved Edman-degra-dering og den integrerte verdi (pmol) av hver PTH-aminosyre-rest sammenlignet med den kjente proteinsekvens. Eksempel 3 ;Fermentering og rensing av Met3rhuIL- 3 ( Pro8Asp15Asp70) ;Ekspresjonsvektoren pIXY290 (se eksempel 1 ovenfor) ble transfektert inn i S. cerevisiae-produksjonsstammen XV2181 i det vesentlige som beskrevet for pIXY151 i eksempel 2. ;Et inokulum ble fremstilt ved inokulering av 0,5 ml av et frosset glyserolforråd av produksjonsstammen i en 6 liters flaske med (a) 500 ml av et gjærdyrkingsmedium som inneholdt pr. liter sterilt H20: 20,0 g (NH4)2S04, 5,0 g KH2P04, 1,0 g MgS04-7 H20, 0,1 g CaCl2-2 H20, 10,0 g kaseinhydrolysat, 20,0 g glukose og 20,0 g galaktose; (b) 0,5 ml av en sporsalt-blanding som inneholdt pr. liter sterilt H20: 5,0 g borsyre, 2,0 g kobbersulfat, 10,0 g jern(III)klorid, 10,0 g mangan-sulfat, 0,5 g natriummolybdat, 1,0 g sinksulfat, 0,5 g kobolt-klorid og 100 ml 6 N HC1; og (c) 0,5 ml av en vitaminløsning som inneholdt pr. liter sterilt H20: 1,0 ml av en 0,2 % løsning av biotin, 1,0 g kalsiumpantotenat, 25,0 g myo-inositol, 5,0 g niacin, 0,4 g pyridoxin-HCl, 0,1 g folsyre og 0,5 g cholin-klorid. Denne inokuleringskulturen ble inkubert ved 30 °C i en rotatorisk ristemaskin ved 250 rpm i tilnærmet 24 timer. ;En 10 liters fermentor ble forberedt til fermentering ved å fylles med 7,5 1 deionisert H20 med 50 g monobasisk kaliumfosfat, 200 g ammoniumsulfat, 10 g MgS04* 7 H20, 1,0 g CaCl2-2 H20 og 2 ml av et overflateaktivt stoff med antiskum-dannende virkning. Fermentoren ble så sterilisert ved opp-varming til 121 °C i 30 minutter under røring. Etter sterili-seringen ble fermentoren tilsatt 35 ml 1 % (vekt/vol) tiamin-HC1, 500 ml 10 % kaseinhydrolysat, 100 ml 50 % glukose og 25 ml hver av vitamin- og sporsaltløsningene beskrevet ovenfor. pH i fermentoren ble justert til 5,5 ved tilsetning av 30 % NH40H og oppløst 02ført til 100 % ved 29 °C. Innholdet i inokulum ble så tilsatt fermentoren og mating startet med tilnærmet 2,0 ml pr. minutt av en matingsløsning som bestod av (a) 3600 ml 50 % glukose; (b) 625 ml av en gjærmatingssaltløs-ning som inneholdt (pr. liter H20): 250 g ammoniumsulf at, 125 g monobasisk kaliumfosf at og 25 g MgS04- 7 H20; (c) 900 ml 50 % EtOH; (d) 60 ml 1 % tiamin-HCl; (e) 450 ml 20 % gjærekstrakt; (f) 450 ml 20 % pepton; (g) 900 ml 20 % kaseinhydrolysat; og (h) 25 ml hver av vitamin- og sporsaltløsningene beskrevet ovenfor. Matingshastigheten ble holdt ved dette nivå i tilnærmet 20 timer og så justert til 3,25 ml pr. minutt i tilnærmet 20 timer, hvoretter kultursupernatanten ble høstet i det vesentlige som beskrevet i eksempel 2 ovenfor.
rhuIL-3 i gjærbrygget ble først redusert ved jus-tering til 1 % p-merkaptoetanol ved pH 7,4 fulgt av inkubering i 1 time ved romtemperatur. Den resulterende blanding ble på-satt en 5 cm x 30 cm kolonne pakket med 15-20 um C-4-reversfasekisel (Vydac, Separations Group, Hesperia, CA, USA) ved å pumpe det filtrerte gjærbrygg direkte på kolonnen. Kolonnen ble ekvilibrert med 0,1 % trifluoreddiksyre i vann (løsemiddel A) før påsetting av gjærbrygget, og ble vasket med dette løse-
middel etter avsluttet påsetting av brygget på søylen til den optiske absorbans av det eluerte materiale nærmet seg baselinjeverdier. På dette tidspunkt ble en gradient med 0,1 % trifluoreddiksyre i acetonitril (løsemiddel B) etablert fra 0 % B til 100 % B ved en endringshastighet på 2 % B pr. minutt og en strømningshastighet på 100 ml/minutt.
Toppfraksjoner fra C-4-kolonnen ble slått sammen og justert til 50 mM natriumacetat, pH 4,7. Denne løsning ble så satt direkte på en kolonne med "Mono S"-ionebytterharpiks ekvilibrert med 50 mM natriumacetat, pH 4,7, og rhuIL-3 eluert med en lineær gradient på 1 M natriumklorid over 20 kolonne-volumer. Det resulterende rhuIL-3 var i det vesentlige 100 % Met<3>rhuIL-3 (Pro8Asp<15>Asp<70>) og tilsvarte rhuIL-3-produksjonspartiene beskrevet i eksempel 2 ovenfor med hensyn til biologisk aktivitet.
Eksempel 4
Intravenøs tilførsel av rhuIL- 3 ( Pro8 Asp15 Asp70)- preparater til synomolqusaper
Tre grupper med synomolgusaper ( Macaca fascicularis), tre hanner og tre hunner i hver gruppe, mottok hver materiale fra en av rhuIL-3-partiene fremstilt som beskrevet i eksempel 2 ved intravenøs tilførsel én gang daglig i 30 på hverandre følgende dager ved dosenivåer på 1 ug/kg (gruppe 2, lavdose), 10 pg/kg (gruppe 3, middels dose) og 100 pg/kg (gruppe 4, høy-dose). En fjerde gruppe med tre hanner og tre hunner mottok 0,9 % natriumklorid (saltvann) og fungerte som kontrollgruppe (gruppe 1, kontroll). Ett dyr av hvert kjønn fra hver gruppe ble beholdt i en 17 dagers restitusjonsperiode. Kriterier evaluert for virkning av stoffet omfattet overlevelse, kliniske tegn, fysisk, elektrokardiografisk og oftalmoskopisk under-søkelse, forandringer i kroppstemperatur, kroppsvekt, klinisk patologi, organvektmålinger og synlig og mikroskopisk patologi.
Alle dyr overlevde til sitt planlagte avlivningstids-punkt. Behandlingsrelaterte kliniske tegn omfattet anoreksi. Ingen neslekjeder ble påvist. Ikke-behandlingsrelaterte effek-ter ble bemerket med hensyn til kroppsvekt, fysisk undersøkel- se, elektrokardiografisk evaluering eller oftalmoskopisk undersøkelse. Klinisk-patologiske parametrer som ble vurdert, omfattet hematologi og myeloid/erytroid(M/E)-forhold evaluert fra lårbensutstrykninger. Serumkjemi og urinanalyse ble også utført. Ingen bemerkelsesverdige serumkjemiparametrer ble observert for middels- og høydosedyrene ved sammenligning med samtidige kontrollverdier eller respektive verdier forut for behandling. Urinanalyseverdier erholdt under behandlingen var ikke bemerkelsesverdige.
Intravenøs injeksjon av analog-rhuIL-3 gav en dra-matiskøkning i leukocyttallet hos middels- og høydosehanner og -hunner. Det gjennomsnittlige lymfocyttall økte også hos cellebehandlede hanner og høydosehunner. Økningen av segmen-terte nøytrofiler, eosinofiler og basofiler hos middels- og høydosedyrene fulgte generelt de økte leukocytt- og lymfocytt-tall. En økning ble også observert i det gjennomsnittlige blodplatetall hos høydosehannene og behandlede hunner i løpet av behandlingstiden. De mest bemerkelsesverdige doserelaterte økninger opptrådte gradvis i løpet av den første og annen uke med en generell nedgang i retning av nivåene før behandling i løpet av restitusjonsfasen.
Undersøkelser av synlig patologi viste abnormaliteter i milt, brissel og binyrer hos dyr som mottok de høyeste doser. Ingen dyr viste imidlertid de samme symptomer, og i ett tilfelle ble abnormaliteten diagnostisert som en forut eksi-sterende tilstand. En økning av gjennomsnittlig relativ brisselvekt ble bemerket hos lav- og middelsdosehanner ved avlivning og hos én lavdose- og én middelsdosehunn ved avlivning etter restitusjonsperioden. Mikroskopiske funn bemerket hos dyrene som ble avlivet ved avsluttet tilførsel, var få og hovedsakelig minimale til lette med hensyn til alvorlighets-grad. Alle histopatologiske funn ble betraktet som tilfeldige og uten opphav i medikamenttilførselen.
Eksempel 5
Klinisk evaluering av IL- 3- preparater
rhuIL-3 fra ett eller flere av produksjonspartiene fremstilt som beskrevet i eksempel 2, ble tilført pasienter med fremskredet progressiv neoplasma eller benmargssvikt i 15 dager som daglige, subkutane injeksjoner. Regelmessige, rutinemessige hematologiske og biokjemiske undersøkelser ble utført og benmargsutsugninger og -biopsier erholdt forut for og etter behandlingssyklusen. I alt 14 pasienter ble behandlet med 60, 125, 250 og 500 ug/m<2>. Ingen pasient avbrøt behandlingen grunnet toksisitet. Hos alle de ni evaluerbare pasienter ble vesentlige økninger i WBC, absolutt segmentert granulocytt-eosinofil- og monocyttall så vel som i retikulo-cytt- og blodplatetall observert. De hematologiske endringer, særlig økningen i blodplatetall, opptrådte i det vesentlige i løpet av annen uke med behandling med rhuIL-3 og fortsatte i 1-2 uker etter avsluttet terapi. Den gjennomsnittlige økning i blodplatetall var 179.500/ul (63.000-448.000). Hos to pasienter med vedvarende trombocytopeni på hhv. 3000/pl og 22.500/ pl økte blodplatetallet til hhv. 104.000/pl og 88.000/pl i et lengre tidsrom etter behandling med 60 pg/m<2>. Toksiske virkninger omfattet feber hos tre pasienter (annengrads), hodepine hos én pasient (annengrads) og lokal erytem på injeksjons-stedet (førstegrads) hos én pasient. Neslefeber eller mastcelle- eller lymfocyttinfiltrasjon i dermis ble ikke observert.
Claims (6)
1. Fremgangsmåte for fremstilling av et renset rekombinant human-IL-3-analogprotein som omfatter Met<3>huIL-3 (Pro<8>Asp<15>Asp<70>),
karakterisert vedat en gjærvertscelle som inneholder en ekspresjonvektor omfattende en DNA-sekvens som koder for human-IL-3-analogproteinet, dyrkes under betingelser som fremmer ekspresjon av proteinet.
2. Fremgangsmåte ifølge krav 1,karakterisert vedat en gjærvertscelle av arten Sacciiaromyces cerevisiae dyrkes under betingelser som fremmer ekspresjon av proteinet.
3. Isolert DNA-sekvens,
karakterisert vedat den koder for et human-IL-3-analogprotein fremstilt ifølge krav 1.
4. Rekombinant ekspresjonsvektor,karakterisert vedat den omfatter en DNA-sekvens ifølge krav 3.
5. Gjærvertscelle,
karakterisert vedat den inneholder en ekspresjonsvektor ifølge krav 4.
