Description
Foreliggende oppfinnelse angår amidinofenoksyalkoksyfenyl-derivatene og thioanaloger som heri definert, som er spesielt nyttige som selektive Leukotriene B4 (LTB4 )-reseptor-antagonister, farmasøytiske sammensetninger inneholdende nevnte forbindelser og anvendelse av disse forbindelsene for fremstilling av farmasøytiske preparater.
EP-A-150118 beskriver strukturelt divergente monokarboksami-der, tiokarboksamider, karboksamidinene med formel I, hvor amidene er avledet fra aminosyrer, og i hvilken den andre benzenringen bærer acylgruppen -C(=0)=R4, hvor R4 er hydroksy, laverealkyl eller laverealkoksy. Forbindelsene nevnt i EP-A-150118 er angitt å være antagonister av leukotriener B4, C4, D4 og E4. Der er ingen indikasjon på noen effekter på interleukinaktivitet.
Leukotrien B4 (LTB4) er en viktig inflammatorisk mediator som er et kraftig kjemotaktisk stoff og aktivator av polymorfonukleære leucocyter (PMN) og monocyter. Det modulerer fremstillingen og påvirker andre viktige inflammatoriske mediatorer for eksempel interleukin-1 og gammainterferon. LTB4 er implisert i patogenesen i flere inflammatoriske sykdommer som rheumatoid arthritis, inflammatorisk tarmlidelse, psoriasis, ikke-steroidal-antiinflammatorisk medikamentindusert gastropati, voksen respirasjonssvikt syndrom (ARDS), myocardialt infarkt, allergisk rhinitis, hemodialysisindusert neutropenia og senfase astma.
Det er et sterkt behov i teknikken for å finne kraftige antagonister til LTB4 på humane PMN, spesielt slike som er oralt aktive. Det er funnet at forbindelsen ifølge oppfinnelsen viser klar LTB4-antagonistaktivitet på humane PMN og er oralt aktive.

hvor fenylringene i formel I uavhengig av hverandre kan være substituert med en eller flere substituenter valgt blant halogen, laverealkyl og laverealkoksy; eller et farmasøytisk akseptabelt salt av dette.
Siden forbindelsene ifølge oppfinnelsen har et basisk senter, kan de danne syreaddisjonssalter, spesielt farmasøytisk akseptable salter. Disse blir dannet for eksempel med uorganiske syrer som mineralsyrer, for eksempel svovelsyre, en fosfor eller hydrohalidsyre eller med organiske karboksylsyrer, som (C1-C4)alkankarboksylsyrer som for eksempel er ikke-substituert eller substituert med halogen, for eksempel eddiksyre, slik som mettede eller umettede dikarboksylsyrer, for eksempel oksalsyre, malonsyre, ravsyre, maleinsyre, fumarsyre, ftalsyre eller tereftalsyre, slik som hydro-karboksylsyrer, for eksempel ascorbinsyre, glycolsyre, melkesyre, malinsyre, tartarsyre eller sitronsyre, slik som aminosyrer, for eksempel aspartinsyre eller glutaminsyre, benzosyre eller med organiske sulfonsyrer, som (C1-C4 )alkan-eller arylsulfonsyre som er ikke-substituert eller substituert, for eksempel med halogen, for eksempel metan- eller toluensulfonsyre. Foretrukne salter er dannet med saltsyre, metansulfonsyre og maleinsyre.
Fenylringene i formel I, så vel som aromatiske radikaler referert til tidligere og senere, er vanligvis ikke-substituert eller videre substituert slik som monosubstituert eller polysubstituert, for eksempel disubstituert eller sekundært trtisubstituert, spesielt for eksempel med en substituent valgt fra gruppen bestående av halogen, trifluormetyl , laverealkyl, laverealkenyl, laverealkynyl, hydroksy, laverealkoksy, laverealkenyloksy, fenyl-laverealkoksy, laverealkanoyloksy, laverealkenoyloksy og fenyl-laverealkanoyloksy. Det er foretrukket at- fenylringene i formel I ikke har noen ytterligere substituent.
Foretrukne posisjoner til de følgende strukturelle elementene i de korresponderende fenylringene i formel I er: posisjon 4 (para) eller 5 (meter) for -CO-Rj: , posisjon 2 (ortho) for R2 og posisjon 4 (para) for -C(-NE)-NH-R3. Substituenten R4 kan være lokalisert på et av nitrogenene i -C(=NE)NH2 gruppen.
De generelle definisjonene brukt over og under har hvis ikke annet er spesifisert følgende betydning: Prefikset "lavere" betyr at korresponderende grupper og forbindelser i hvert tilfelle inneholder færre enn 7 og fortrinnsvis ikke mer enn 4 karbonatomer.
Halogen er spesielt fluor, klor og brom og omfatter videre lod.
Laverealkyl er spesielt C^-Cyalkyl og er for eksempel metyl, etyl, n-propyl, isopropyl, sec-butyl, tert-butyl og omfatter videre korresponderende pentyl, heksyl og heptylradikaler. Ci-C<4->alkyl er foretrukket.
Fenyl-laverealkyl er spesielt fenyl-Ci-C4-alkyl og foretrukket er benzyl, 1- og 2-fenetyl, mens fenyl-laverealkenyl og fenyl-laverealkynyl spesielt er fenyl C2~C5alkenyl og-alkynyl, spesielt 2-fenyl-vinyl, 3-fenylallyl og 3-fenylpro-pargyl.
C3-Cy-cykloalkyl er for eksempel cyklopropyl, cyklobutyl, cyklopentyl, cykloheksyl og cykloheptyl. Cyklopentyl og cykloheksyl er foretrukket.
Laverealkylen, for eksempel i amino som er disubstituert med laverealkylen er spesielt C2-C6-alkylen og er for eksempel butylen, pentylen, eller 2,6-butylen. Foretrukket er C4-C5-alkylen, spesielt pentylen.
Laverealkylen X2 er spesielt C2-Cg-alkylen, fortrinnsvis rettlkjedet og er for eksempel etylen, propylen, butylen, pentylen, heksylen, heptylen og også oktylen. C4-C7-alkylen er foretrukket, spesielt pentylen og også butylen, heksylen eller heptylen.
Laverealkylen som er avbrutt av et fenylradikal (X2) er spesielt laverealkylen-fenylen-laverealkylen eller laverealkylen-naftylen-laverealkylen som C2"C4-alkylen-fenylen-C2-C4-alkylen eller C2-C4-alkylen-naftylen-C2-C4-alkylen, spesielt rettkjedet og er for eksempel metylen-fenylen-metylen, 1,2-etylen-fenylen-1,2-etylen, som 1,2-etylen-l,4-fenylen-1,2-etylen, 1,3-propylen-fenylen-1,3-propylen, som 1,3-propylen-1,4-fenylen-l,3-propylen eller butylen-fenylen-butylen-radikaler, også et korresponderende 1,2-etylen-nsftylen-1,2-etylenradlkal. C2~C4-alkylen-fenylen-C2-C4-alkylen eller C2-C3~alkylen-naftylen-C2~C3-alkylen er foretrukket, spesielt 1,2-etylen-l,4-fenylen-l,2-etylen.
Laverealkoksy er spesielt C^-Cy-alkoksy og er for eksempel metoksy, etoksy, n-propyloksy, isopropyloksy, n-butyloksy, isobutyloksy, sec-butyloksy, tert-butyloksy og omfatter videre korresponderende pentyloksy, heksyloksy og heptyl-oksyradikaler. C1-C4~alkoksy er foretrukket.
Alkoksykarbonyl er spesielt C2-Ci2~alk0ksykarbonyl °S er f°r eksempel metoksy-, etoksy-, propyloksy-, pivaloyloksy- eller oktyloksy-karbonyl. C2-Cg-alkoksykarbonyl er foretrukket.
Alkenyloksykarbonyl er spesielt C3-Ci2-alkenyloksykarbonyl» for eksempel allyloksykarbonyl. Foretrukket er C3-C5-alkenyloksykarbonyl.
C3-C7~cykloalkoksykarbonyl er fortrinnsvis cyklopentyloksy-karbonyl eller cykloheksyloksykarbonyl.
C^-Cy-alkansulfonyl er for eksempel metan-, etan-, n-propan-eller isopropansulfonyl. C1-C4-alkansulfonyl er foretrukket.
Fenyl-C^-C7~alkansulfonyl er for eksempel benzyl- eller 1-eller 2-fenyletansulfonyl. Fenyl-C1-C4-alkansulfon er foretrukket.
Fluorenyl er for eksempel 1-, 2-, 3-, 4- eller 5-fluorenyl.
Laverealkanoyl er spesielt C1-C7-alkanoyl og er for eksempel formyl, acetyl, propionyl, butyryl, isobutyryl eller pivavolyl. C2~C5-alkanoyl er foretrukket. Fenyl-laverealkanoyl er spesielt fenyl-Cg-Cy-alkanoyl og er for eksempel fenylacetyl eller 2- eller 3-fenylpropionyl. Fenyl-C2~C4-alkanoyl er foretrukket.
Forbindelsen ifølge oppfinnelsen viser verdifulle farmakologiske egenskaper hos pattedyr, og er spesielt nyttige som selektive Leukotrien B4 (LTB4) reseptor antagonister, for eksempel for behandling av en tilstand eller sybdrom hos pattedyr som påvirkes av de selektive antagonistene til LTB4-reseptorer, som leddgift, inflammatorisk magelidelse, psoriasis, ikke-steroidal antiinflammatorisk medikamentindusert gastropati, voksent respirasjonssviktsyndrom (ARDS), myocardialt infarkt, allergisk rhinitis, hemodialysisindusert neutropeni og senfase astma. Forbindelsen ifølge oppfinnelsen er også nyttig som analgesica for behandlingen av smerte av en hvilken som helst opprinnelse og for behandlingen av osteoarthritis.
De ovenfor nevnte egenskapene kan demonstreres i in vitro og in vivo tester, best ved hjelp av pattedyr, for eksempel rotter. Nevnte forbindelser kan bli anvendt in vitro i form av oppløsninger, for eksempel vandige oppløsninger, og in vivo enten enteralt eller parantalt, helst oralt, for eksempel som en suspensjon eller i vandig oppløsning. Dosen in vitro kan variere mellom 0,5 ng/ml og 100 ng/ml. Doseringen in vivo kan variere avhengig av administrasjonsveien, mellom ca 1 og ca 1000 mg/kg pr. dag.
Fordelaktige effekter blir evaluert i farmakologiske tester som er velkjente i teknikken, for eksempel som illustrert her.
Reseptorbinding med [<3>H]-LTB4 til intakte humane neutrofile: Neutrofile (PMN) ble fremstilt fra ikke-koagulert humant venøst blod. Blodet ble fordelt i 50 ml polypropylenrør inneholdende 15 ml BESPAN (Dupont, Wilmington, DE), og blandet. Rørene ble satt i henstand ved romtemperatur i 40 minutter inntil det meste av de røde blodcellene er sedi-mentert. Supernatanten blir fjernet og sentrifugert i 5 til 10 minutter ved 400 g. De gjenværende pelletene blir fortynnet i 70 ml forsfatbuffret saltvannsoppløsning uten kalslummagnesium (PBS uten metaller; GIBCO, Grand Island, NY) og 35 ml av denne suspensjonen ble plassert i to poly-propylenrør inneholdende 15 ml Ficoll-Paque (Sigma, St. Louis, MO). Gradientene ble så sentrifugert i 15 minutter ved 420 g. Det mononukleære cellelaget ble kastet og den gjenværende pelleten av røde blodceller blir resuspendert i 10 ml PBS uten metaller. 20 ml filtrert deionisert vann ble tilsatt suspensjonen i ca 20 sekunder fulgt av det samme volumet buffer med to ganger den normale konsentrasjonen. Cellesuspensjonen blir blandet og sentrifugert i 5 minutter ved 200 g, fulgt av en vasking med buffer og endelig resuspensjon.
Binding av [<3>H]LTB4 til LTB4 reseptorer ble målt i intakte humane polymorfonukleære leukocyter, som beskrevet av Gorman og Lin (Gorman, R. og Lin, A Methods Enzymol. 141:372-378, 1987). Intakte human neutrofile blir suspendert i "Hank's Balanced Salt Solutions (HBSS) ved en konsentrasjon på 3 x 10^ celler/testrør. En prøve av cellesuspensjonen (300 pl) ble tilsatt til tre rør inneholdende 50>j1 [3H]LTB<4> (spesifikk aktivitet 32 Ci/mmol, DFuPont-NEN, Boston, MA) med en endelig konsentrasjon på 0,5 nM, 100pl buffer og 50>j1 medikament eller buffer. Ikke-spesifikk binding blir bestemt i nærvær av 300 nM LTB4. Reaksjonen ble startet ved tilsetning av cellesuspensjon og fortsatt ved 0°C i 20 minutter. Bundet radioaktivitet ble resultert ved vakuumfiltrering gjennom Whatman GF/C glassfibere ved hjelp av en Brandel cellehøster og ubundet radioaktivitet ble fjernet med 2 x 5 ml vaskin med iskald saltvannsoppløsning. Filtrene ble plassert i poly-etylen scintillasjons mini-glass til hvilke det ble tilsatt 3,5 ml formula-989 scintillasjonsblanding (NEN). Etter utjevning, blir radioaktivitetbestemmelser og databeregninger utført ved hjelp av ikke-lineær regresjonsanalyse på RS/1.
LTB4-indusert PMN aggregering
Humane PMN ble fremstilt som beskrevet ovenfor. Neutrofil aggregering blir anslått ved måling av intensitetenav lys som passerer gjennom en suspensjon av celler (Craddock et al., J. Clin. Invest. 60:260-264, 1977) ved hjelp av et Payton to kanal aggregometer (modell 300BD). Kuvetter inneholdende 0,25 ml cellesuspensjon (25 x IO<6> celler/ml) i PBS uten kalsium og magnesium blir inkubert med 5pg/ml cytochalasin B i 2 minutter ved 37" C. 5pl av 2 uM LTB4 i PBS (20 nM endelig konsentrasjon) ble tilsatt og den følgende aggregeringen målt i 3 til 5 minutter, den tiden som er nødvendig for optimal respons. Forbindelsene blir oppløst i 0.01M DMSO og så fortynnet i PBS til 0.001M. 5ul av oppløst forbindelse ble tilsatt sammen med cytochalasin B og celler som beskrevet over. Etter preinkuberingstiden blir 5ul av 2uM LTB4 tilsatt og aggregeringen blir målt. Prosent hemming av aggregeringen ble beregnet ved sammenligning av maksimalhøydene i nærvær og fravær av forbindelsen. Prosent hemming blir plottet som funksjon av logreritmen til konsentrasjonen av forbindelsen og IC50 bestemt direkte fra kurven.
LTB4-indusert neutropenia hos rotter
Hann Sprague Dawley rotter (CDBR; Charles River, Wilmington, MA.) (250-300) gram ble fastet over natten før eksperimentet. Minst 6 dyr ble brukt pr. behandlingsgruppe. Rottene blir gitt en bærer eller en forbindelse enten intravenøst eller oralt og ved bestemte intervaller etter doseringen blir neutrofilantallet bestemt fra blodprøver oppnådd rett før og 20 sekunder etter intravenøs infusjon av 200 ng LTB4. I studiene blir forbindelsene administrert oralt, blir medikamentet gitt med sonde. Når medikamentet blir administrert intravenøst, blir rottene først bedøvd med 50 mg/kg i.p. natriumpentabarbital. Den jugulære venen blir eksponert og renset for omgivende vev. Etter 3,4 og 18 timer etter administrasjon av forbindelse eller bærer gitt etter disse veiene, blir blodprøver tatt (0,3 ml blod i 1,5 ml poly-propylenmikrosentrifugerør inneholdende 0,01 ml 7, 5% EDTA). Blodets neutrofilantall blir bestemt ved hjelp av et Technicon H-l hematologi instrument. Antagonisme av LTB4-indusert neutropenia respons for forbindelsene som er testet blir beregnet.
Analgesisk aktivitet kan bli vist for eksempel i Randall-Selitto testen for analgesia, for eksempel som beskrevet i Arch. Int. Pharmacodyn. Ther. 111, 409 (1957).
Antiinflammatorisk aktivitet kan bli vist ved måling av hemmingen av ødemet og hemmingen av innstrømmingen av polymorfonukleære (PMN) og mononukleære leukocyter (monocyter og makrofager) etter oral administrasjon i rottemodellen i hvilken pleurit først er indusert ved injeksjon av carra-geenin i brysthinnehulen, for eksempel ifølge A.P. Almeida et al., J. Pharmacol. Exp. Therap. 214, 74 (1980), spesielt under den sene fasen av den corrageenin-induserte pleurit.
Bronchiale effekter som anti-asmatisk aktivitet kan bli vist i antigenindusert bronchokonstriksjonstest for hamster, for eksempel som beskrevet av Anderson et al, Br. J. Pharmacol. 1983, 78, 67-74.
Trinitrobenzensulfonsyreindusert kronisk colittest i rotter, for eksempel som beskrevet av Wallace et al, Gastroenterology 1989, 96, 29-36 kan bli brukt for å evaluere forbindelsenes effekt for infallmatoriske tarmlidelser.

En forbindelse med formel Ila hvor er aktivert karboksy, (Z]i) er for eksempel et anhydrid av dette, omfattende et blandet anhydrid, som for eksempel et syrehalid, for eksempel klorid, eller et anhydrid med en maursyreester, en aktivert karboksylsyreester som cyanometyl, (4-)nitrofenyl, poly-halogenofenyl, for eksempel pentaklorfenylester.
Reaksjonen blir for eksempel utført med et reagens som passer for introduksjon av R3, for eksempel et amin med formelen H-Rl. Reaksjonen med forbindelsene med formel Ila hvor Z^ karboksy eller et salt av dette for eksempel finner sted under vannfrie betingelser, for eksempel med azeotropisk fjerning av vann fra reaksjonen, eller ved behandling med et passende kondenserende middel, for eksempel N,N'-dicyklo-heksyl-karbodiimid. Reaksjonen med et aktivert karboksy-derivat kan med fordel bli utført i nærvær av en base. Fremgangsmåte b) :
Alkoksy-iminokarbonyl er for eksempel C-^-C^alkoksy-iminokarbonyl som metoksy- eller etoksy-iminokarbonyl, mens halogeno-iminokarbonyl er for eksempel kloriminokarbonyl.
Fortrinnsvis blir de forbindelsene med formel Illa benyttet hvor Z2 er Ci-C4-alkoksy-iminokarbonyl som metoksy- eller etoksy-iminokarbonyl. Dersom forbindelsen med formel I eller IA inneholder umettede radikaler, som laverealkylgrupper, kan disse bli omdannet til mettede radikaler med en fremgangsmåte kjent per se. Således kan flerverdige bindinger blir hydrogenert ved katalytisk hydrogenering i nærvær av hydrogeneringskatalystor, passende for dette formålet er for eksempel nikkel, som Raney nikkel og edelmetaller eller deres derivater, for eksempel oksyder, som palladium eller palladiumoksyd, som hvis ønsket kan være påført bærermaterialer, for eksempel til karbon eller kalsiumkarbonat. Avhengig av valget av startmaterialer og fremgangsmåter, kan de nye forbindelsene være i form av en av de mulige isomerene eller blandinger av disse, for eksempel som helt rene geometriske (cis eller trans) isomerer, optiske isomerer (antipoder), racemater eller blandinger av disse. De nevnte mulige isomerer og blandinger av disse er innen rammene av oppfinnelsen.
En hvilken som helst resulterende blanding av isomerer kan bli separert på basis av de fysisk-kjemiske forskjellene til bestanddelene, til rene geometriske eller optiske isomerer, diastereoisomerer, racemater for eksempel ved kromatografi og/eller fraksjonen krystallisering.
Hvilke som helst resulterende racemater av endelige produkter eller mellomprodukter kan bli oppløst i optiske antipoder ved kjente fremgangsmåter, for eksempel ved separering av diasteroisomere salter av disse, oppnådd med en optisk aktiv syre eller base, og frigir de optisk aktive sure eller basiske forbindelsene. Amidinet (hvor R3 er hydrogen) kan således bli adskilt i sine optiske antipoder for eksempel ved fraksjonen krystallisering av salter dannet med en optisk aktiv syre.
Til sist er forbindelsene ifølge oppfinnelsen enten oppnådd i fri form eller som salter av disse.
I lys av det nære slektskapet mellom de frie forbindelsene og forbindelsene i form av deres salter, menes det også et korresponderende salt når det blir referert til en forbindelse i denne sammenhengen, forutsatt at dette er mulig eller passende under forholdene.
Forbindelsene, inkludert deres salter, kan også bli oppnådd i form av sine hydrater eller omfatter andre oppløsningsmidler som er benyttet for krystall iseringen.

Disse blir fremstilt på en måte kjent per se, for eksempe ved hjelp av konvsnesjonell blanding, granulering, belegging, oppløsing eller fryse-tørkingsprosesser. Således kan farmasøytiske produkter for oral bli oppnådd ved blanding av den aktive forbindelsene ved faste excipienter, hvis det er formålstjenelig å granulere en blanding som blir oppnådd, og bearbeide blandingen eller kornene hvis ønsket eller nødvendig, etter tilsetning av passende hjelpestoffer til tablettene eller kjernene til belagte tabletter.
De farmakologisk aktive forbindelsene ifølge oppfinnelsen er nyttige i fremstillingen av farmasøytiske sammensetninger bestående av en effektiv mengde av disse i konjunksjon eller blanding med excipienter eller bærere som passer for enten enteral eller parental anvendelse. Foretrukket er tabletter og gelatinkapsler bestående av det aktive stoffet sammen med
a) fortynnere, for eksempel lactose, dekstrose, sucrose, manitol, sorbitol, cellulose og/eller glycin; b) smørende
stoffer, for eksempel silika, talkum, stearinsyre, dets magnesium eller kalsiumsalt og/eller polyetylenglycol; for
tabletter også c) bindemidler, for eksempel magnesium-aluminiumsilikat, stivelsepasta, gelatin, tragacanth, metylcellulose, natriumkarboksymetylcellulose og/eller polyvinylpyrrolidon; hvis ønsket d) disintegranter, for eksempel stivelse, agar, alginsyre eller dets natriumsalt, eller brusende blandinger; og/eller e) absorbanter, farge-midler, smaksstoffer og søtningsstoffer. Injiserbare sammensetninger er fortrinnsvis vandige isotone oppløsninger eller suspensjoner, og stikkpiller blir helst fremstilt av fettemulsjoner eller suspensjoner. Nevnte sammensetninger kan bli sterilisert og/eller inneholde adjuvanter, som konser-veringsmidler, stabiliserende midler, fuktende eller emulgeringsstoffer, oppløselighetsfremmere, salter for regulering av det osmotiske trykket og/eller buffere. Kjernene til belagte tabletter blir utstyrt med passende, helst enteriske belegg, ved hjelp av blant annet konsentrerte sukkeroppløsninger som eventuelt inneholder arabisk gummi, talk, polyvinylpyrrolidon, polyetylenglycol og/eller titandioksyd, lakkoppløsninger i passende organiske opp-løsningsmidler eller oppløsningsmiddelblandinger eller, for fremstilling av enteriske "belegg, oppløsninger av passende celluloseprodukter som acetylcelluloseftalat eller hydroksy-propylmetylcelluloseftalat. Fargestoffer eller pigmenter kan bli tilsatt tablettene eller beleggene av belagte tabletter, for eksempel for å identifisere eller indikere forskjellige doser av aktive forbindelser. I tillegg kan de også inneholde andre verdifulle terapeutiske stoffer. Nevnte sammensetninger blir fremstilt ifølge konvensjonell blanding, granulering eller beleggingsmetoder, og inneholder fra 0,1 til 75?6, fortrinnsvis fra 1 til 50% av det aktive stoffet.
Passende formuleringer for transdermal anvendelse omfatter en effektiv mengde av forbindelsen ifølge oppfinnelsen med en bærer. Fordelaktige bærere omfatter absorberbare farmasøytisk akseptable oppløsningsmidler for å assistere passeringen
gjennom vertens hud. Karakteristiske transdermale inn-retninger er i form av en bandasje inneholdende en støttende innretning, et reservoar inneholdende forbindelsen eventuelt med bærere, eventuelt en hastighetskontrollerende barriere for å levere forbindelsen til vertens hud ved en kontrollert og forhåndsbestemt hastighet over en lang tidsperiode og hjelpemidler til å sikre innretningen mot huden.
I sammenheng med andre aktive ingrtedienser, kan en forbindelse ifølge oppfinnelsen bli administrert enten samtidig, før eller etter den andre aktive ingrediensen, enten separat med samme eller forskjellig administrasjonsvei eller sammen i samme farmasøytiske formulering.
Oppfinnelsen angår videre spesielt en fremgangsmåte for behandling av en tilstand eller syndrom som responderer på den selektive antagonismen av LTB4~resptor, som rheumatoid arthritis, inflammatorisk tarmlidelse, psoriasis, ikke-steroidal-antiinflammatoriskmedikamentindusert gastropati, voksen åndenødssyndrom (ARDS), myocardial infarkt, allergisk rhinitis, hemadialysis-indusert neutropeni og senfase astma.
Doseringen av aktiv forbindelse som blir administrert er avhengig av arten varmblodig dyr (pattedyr), kroppsvekt, alder og individuell tilstand og administrasjonsformen. En doseringsenhet for oral administrasjon til et pattedyr på ca 70 kg kan inneholde for eksempel mellom 1 og 1000 mg/kg pr. dag av det aktive stoffet.
De følgende eksemplene er ment som illustrasjoner for oppfinnelsen og må ikke bli ansett som begrensninger av denne* Temperaturen er gitt i °C. Hvis ikke annet er nevnt, blir alle avdampingene utført under redusert trykk, fortrinnsvis mellom 15 og 100 mm Hg. Struktruen til endelige produkter, mellomprodukter og startmaterialer blir bekreftet ved standard analytiske fremgangsmåter, for eksempel mikroanalyse og spektroskopiske karakteristikker (for eksempel MS, IR, NMR). Eksempel 1:
En omrørt oppløsning ved 0"C av etyl 4-[5-[2-metoksy-4-[N,N-bis( 1-metyletyl )-aminokarbonyl]fenoksy]pentoksy]benzenkarboksimidoat (420 mg, 0,87 mmol) i 20 ml vannfri etanol blir behandlet med vannfri hydrogenkloridgass i 5 minutter. Etter oppvarming til romtemperatur, blir den resulterende oppløsning konsentrert under vakuum, gjenoppløst i kloroform (20 ml) og konsentrert igjen. Det resulterende hydroklorid-salt blir så oppløst i etanol (40 ml), overført til et trykkrør og behandlet med vannfri ammoniak i 10 minutter ved 0°C. Trykkrøret ble forseglet og oppvarmet til 100°C i 45 minutter. Etter avkjøling og konsentrering under vakuum, blir det resulterende materialet renset ved kromatografi på silikagel (15 g) med 3 til 2056 metanol/diklormetan som elueringmiddel for å gi 4-[5-[4-(aminoiminometyl)fenoksy]-pentoksy]-3-metoksy-N,N-bis(1-metyletyl )benzamid monohydroklorid som et fargeløst skum:

Startmaterialet, etyl 4-[5-[2-metoksy-4-[N,N-bis(l-metyletyl )aminokarbonyl] -f enoksy] pentoksy]benzenkarboksimidoat, kan bli fremstilt for eksempel som følger: En omrørt oppløsning ved 0° av 4-hydroksybenzonitril (50,7 g, 426 mmol) i 1400 ml diklormetan og 75 ml vannfri etanol blir behandlet med vannfri hydrogenkloridgass (110 g) i 1,5 time. Denne oppløsningen ble omrørt ved romtemperatur i 64 timer og det resulterende faststoff oppsamlet og vasket med 500 ml dietyleter og 2 x 1000 ml etylacetat. Det gjenværende faststoff (60,4 g) ble oppløst i 1200 ml vann og det resterende faststoffet filtrert. Til filtratet ble det tilsatt en oppløsning av natriumhydroksyd (12,57 g) i 150 ml vann. Det resulterende hvite faststoffet ble filtrert og vasket med vann for å gi etyl 4-hydroksybenzenkarboksimidoat.
En omrørt oppløsning av etyl 4-hydroksybenzenkarboksimidoat (32,0 g, 194 mmol) i 250 ml vannfri N,N-dimetylformamid blir behandlet med cesiumkarbonat (75,7 g, 232 mmol) og 1,5-dibrompentan (89,1 g, 387 mmol) og varmet ved 70° C i 1,5 time. Etter avkjøling til romtemperatur, blir reaksjonsblandingen filtrert og det resulterende filtratet konsentrert under vakuum (< 1 mtorr) for å gi en gul olje (85,7 g). Dette materialet ble renset ved kromatografi på silikagel (850 g) med 10 til b0% etylacetat/heksan som elueringsmiddel for å gi etyl 4-[5-bromopentoksy]benzenkarboksimidoat som en fargeløs olje.
En omrørt oppløsning av 4-hydroksy-3-metoksy-N,N-bis(1-metyletyl )benzenkarboksamid (319 mg, 1,27 mmol) i 3,5 ml vannfri N,N-dimetylformamid ble behandlet med cesiumkarbonat (435 mg, 1,33 mmol) og etyl 4-[5-bromopentoksy]benzenkarboksimidoat (400 mg, 1,27 mmol) og oppvarmet til 70'C i 1,0 time. Etter avkjøling til romtemperatur ble reaksjonsblandingen skilt mellom etylacetat og vann og den organiske fasen vasket med saltvann, tørket over natriumsulfat og konsentrert under vakuum for å gi en gul olje. Dette materialet blir renset ved kromatografi på silikagel (22 g) med 30 til 75% etylacetat/heksan som elueringsmiddel for å gi etyl 4-[5-[2-metoksy-4-[N,N-bis(l-metyletyl)-aminokarbonyl]-fenoksy]pentoksy]benzenkarboksimidoat som en fargeløs olje.

Reaksjonsblandingen ble oppvarmet under nitrogen i ytterligere 3 dager inntil TLC viser at intet klormellomprodukt er til stede. Badet ble fjernet og røringen stoppet. Reaksjonsblandingen ble filtrert gjennom en filterkrukke og oppi en annen 840 1 reaktor. Filtratet ble omrørt og avkjølt på isbad til 10°C i 3 timer og produktet filtrert, vasket med dietyleter (4 x 11) og tørket under vakuum (50°C, 3 mm Hg) for å gi 4-[5-(4-cyanofenoksy )-pentoksy]-3-metoksybenzoat, smeltepunkt 105-107'C, HPLC indikerte en renhet på 99,8*. Metyl 4-[5-(4-cyanofenoksy )-pentoksy]-3-metoksybenzoat (3600 g, 9,74 mol) ble oppløst i tetrahydrofuran (36 1) under oppvarming til 30°C. Oppløsningen ble så avkjølt til 20°C og en oppløsning av tetrabutylammoniumhydroksyd (40* i vann, 7,1 1, 1,64 m) ble tilsatt i løpet av 20 minutter. Reaksjonsblandingen ble så omrørt ved en intern temperatur på 25"C i 7 timer og over natten ved romtemperatur (23°C). Etter avkjøling til 10°C med isbad, ble saltsyre (1.0N, 14,4 1) tilsatt i løpet av 1 time. Etter omrøring og avkjøling til 10°C i 3 timer, ble produktet filtrert, vasket med vann (24 1) og tørket under vakuum (76°C, 3 mm Hg) for å gi 4-[5-(4-cyanofenoksy)pentoksy]-3-metoksybenzosyre, smeltepunkt 159 til 161°C. HPLC indikerer 99,1* renhet,. En annen syreavling ble oppnådd fra filtratene etter fortynning med vann (20 1). Produktet ble filtrert, vasket med vann (4 x 31) og tørket under vakuum for å gi 4-[5-(4-cyanofenoksy)pentoksy]-3-metoksybenzosyre, smeltepunkt 158-160°C, (HPLC indikerte 98,3* renhet).
4-[5-(4-cyanofenoksy)pentoksy]-3-metoksybenzosyre (3600 g, 10,13 mol) ble suspendert i diklormetan (36 1) og til dette ble det tilsatt thionylklorid (1345 g, 11,30 mol) dråpevis i løpet av 20 minutter fulgt av dimetylformamid (74,4 g, 10,0 mol). Reaksjonsblandingen ble omrørt ved romtemperatur i 21 timer; etter 6 timer er fullstendig oppløsning oppnådd. Oppløsningen ble konsentrert under vakuum (50°C, 3 mm Hg) for å gi 4-[5-(4-cyanofenoksy)pentoksy]-3-metoksybenzoyl-klorid som et faststoff, som så ble gjenoppløst i diklormetan (36 1) uten ytterligere rensing og avkjølt til 5°C på isbad. Diisopropylamin (3077 g, 30,40 mol) ble dråpevis tilsatt i løpet av 1 time. Reaksjonsblandingen ble omrørt ved 20° C i 3 timer og så ble N,N-dimetyletylendiamin (80,3 g, 0,91 mol) tilsatt. Reaksjonen ble fulgt med TLC (2:1 etylacetat/heksan, silika plater) inntil det ikke ble detektert mer biprodukt (bis-anhydris av 4-[5-(4-cyano-fenoksy)pentoksy]-3-metoksybenzosyre). Reaksjonsblandingen
ble så omrørt over natten ved romtemperatur. Vann (18 1) ble tilsatt, omrørt og lagene separert. De organiske lagene ble vasket med saltsyre (1,0 N, 23 1), vann (18 1), ammonium-hydroksyd (0,5 N, 2 x 23 1), vann (18 1) og sal tvanns-oppløsning (18 1). Den separerte diklormetanoppløsningen ble tørket over vannfri magnesiumsulfat, filtrert og konsentrert under vakuum (50°C, 3 ml Hg) for å gi 4-[5-(4-cyanofenoksy)-pentoksy]-3-metoksy-N,N-bis(1-metyletyl)-benzamid som et faststoff. Faststoffet ble oppløst i 2 B etanol (21,51) ved oppvarming og oppløsningen ble tilsatt vann (8,6 1). Produktet ble tillatt å krystallisere over natten i isbad. Produktet ble filtrert, vasket med EtOH/H20 (5:2, 2 x 2 1) og tørket under vakuum (54'C, 3 mm Hg) for å gi 3700 g (83,2*) av 4-[5-(4-cyanofenoksy )pentoksy]-3-metoksy-N,N-b i s(1-metyletyl )-benzamid, smeltepunkt 90-92''C. HPLC indikerte en renhet på mer enn 99,7*.
Etanol (2 B, 6,0 1) ble avkjølt i isbad til 0°C og mettet med vannfri hydrogenklorid over en periode på 7,5 time. Oppløs-ningen ble satt på is over natten og hydrogenklorid til-setningen ble fortsatt ytterligere 1 time ved 0°C. 4-[5-(4-cyanof enoksy jpentoksy] - 3-metoksy-N, N-bis (1-metyletyl )-benzamid (4200 g, 9,57 mol) ble tilsatt hurtig i løpet av 30 minutter og en eksoterm reaksjon hevet temperaturen i blandingen til 10°C. Hydrogenkloridtilsetning ble fortsatt i 6 timer og den purpurfargede oppløsningen omrørt over natten ved 20"C. Reaksjonsblandingen ble fulgt med TLC (etylacetat/- heksan, 2:1) ved hjelå av silikaplater. Totalt 4 til 5 dager er nødvendig for fullførelse av denne reaksjonen med periodisk gjenmetting med hydrogenklorid. Den tykke purpur oppløsningen ble overført til en 840 1 reaktor og gnidd med vannfri dietyleter (2 x 30 1), med en fjerning av eter-oppløsningen etter hver vask. Reaktoren ble satt under vakuum (3 mm Hg) i 0,5 time for å fjerne det meste av hydrogen-kloridet for å etterlate etyl 4-[5-[3-metoksy-4-[N,N-bis(l-metyletyl)aminokarbonyl]fenoksy]pentoksy]benzenkarboksimidoat monohydroklorid som en tykk purpurfarget olje.
Etanol 2 B (42 1) ble tilsatt reaktoren og etyl-4-[5-[3-metoksy-4-[N,N-bis( 1-metyletyl ) aminokarbony 1] f enoksy] - pentoksy]benzenkarbokimidoatmonohydroklorid ble oppløst. Oppløsningen ble avkjølt til 5°C og vannfri ammoniakk ble boblet inn i oppløsningen i 7,5 timer inntil metting var oppnådd. Etter omrøring over natten i isbad ble ammoniakk-tilsetningen fortsatt i ytterligere 7,5 timer ved 5°C. Reaksjonsblandingen ble fulgt ved TLC (CH2Cl2/MeOH, 9:1) ved hjelp av silikaplater. Totalt 4 dager er nødvendig for fullførelsen av reaksjonen med periodisk remetting med ammoniakkgass. Reaksjonen ble filtrert og filtratet delt i to like porsjoner. Konsentrering under vakuum (55°C, 3 mm Hg) gir en oljeaktig viskøs suspensjon som ble fylt i en 840 1 reaktor. Dette ble blandet med saltsyre (12 N, 560 ml) fortynnet med vann (44 1). På dette tidspunkt oppløses forbindelsen (pH er 1,62). Oppløsningen ble vasket med dietyleter (3 x 18 liter) og pH justert til 12,6 med en 6 N . natriumhydroksydoppløsning (3,0 1). Den oljeaktige suspensjonen krystalliserer etter omrøring over natten ved 20°C. Produktet ble filtrert, vasket med vann (5x4 liter) og tørket under vakuum (60°C, 3 mm Hg) for å gi produktet. Den andre halvdelen av reaksjonsblandingen ble bearbeidet på en tilsvarende måte for å gi 4-[5-[4-(aminoiminometyl)fenoksy]-pentoksy]-3-metoksy-N,N-bis(1-metyletyl)-benzamid, smeltepunkt 112-114°C. HPLC Indikerte en renhet på 98,5*.
Eksempel 8:
En omrørt oppløsning av 4-[5-[4-(aminoiminometyl)fenoksy]-pentoksy]-3-metoksy-N,N-bis(1-metyletyl )benzamid monohydroklorid (200 mg, 0,41 mmol) i 4,0 ml vannfri metylenklorid ble behandlet ved 0°C først med trietylamin (0,068 ml, 0,48 mmol) og så med metylisocyanat (0,030 ml, 0,49 mmol). Oppløsningen ble omrørt i 30 minutter, konsentrert under vakuum og det resulterende faststoffet ble skilt mellom etylacetat og vann. Det organiske laget ble vasket med vann og saltvann, tørket over natriumsulfat og konsentrert. Det resulterende materialet ble renset ved kromatografi på silikagel (15 g) ved 70 til 100* etylacetat/heksan som elueringsmiddel for å gi 4-[5-[4-[imino(metylaminokarbonyl-amino )metyl]fenoksy]pentoks]-3-metoksy-N,N-bis(1-metyletyl)-benzamid som et fargeløst skum:

En fremgangsmåte analogt med den beskrevet for eksempel 8, kan de følgende forbindelsene bli fremstilt: 4-{ [5-[4-(imino[3,4-dimetoksyfenyl)karbonylamino]metyl]-fenoksy]pentoksy]-3-metoksy-N,N-bis(l-metyletyl)benzamid ble oppnådd fra 4-[5-[4-(aminoiminometyl)fenoksy]pentoksy]-3-metoksy-N,N-bis(1-metyletyl)benzamidmonohydroklorid (400 mg) og 3,4-dimetoksybenzoylklorid (170 mg) som et fargeløst skum: CHN beregnet for C35<H>45N307; 4-[5-[4-[imino[[[(l -me ty le tyl ) am i no] karbonyl] amino )metyl] - fenoksy]pentoksy]-3-metoksy-N,N-bis(1-metyletyl)benzamid ble oppnådd fra 4-[5-[4(aminoiminometyl)fenoksy]pentoksy]-3-metoksy-N,N-bis(1-metyletyl)benzamidnomohydroklorid (300 mg) og isopropylisocyanat (54 mg) som et fargeløst skum: CHN beregnet for C3<qH>44<N>405; 4 -[5 -[4-[(etyl amino ) karbonyl] amino] iminometyl] f enoksy ] pentoksy]-3-metoksy-N ,N-bis( 1-metyletyl )benzamid ble oppnådd fra 4-[5-[4-(aminoiminometyl)f enoksy]pentoksy]-3-metoksy-N ,N-bis(1-metyletyl)benzamidmonohydroklorid (300 mg) og etyliso-cyanat (45 mg) sin et fargeløst skum: CHN beregnet for C29<H>42<N>4O5; 4-[5-[4-[imino[[( f enol amino ) karbonyl ] amino] metyl] fenoksy] - pentoksy]-3-metoksy-N,N-bis(1-metyletyl)benzamid (240 mg) ble oppnådd fra 4-[5[4-(aminoiminometyl)fenoksy]pentoksy]-3-metoksy-N,N-bis(1-metyletyl )benzamidmonohydroklorid (300 mg) og fenolisocyanat (76 mg) som et fargeløst skum: CHN beregnet for C33<H>42<N>4O5;
Eksempel 10:
a) Fremstilling av 10.000 tabletter hver inneholdene 20 mg av den aktive ingrediens, for eksempel 4-[5-[4-(aminoiminometyl ) f enoksy] pentoksy] -3-metoksy-N,N-bis (1-metyletyl )benzamid (Z )-2-butendioat (1:1):
Alle pulverene ble ført gjennom en sikt med åpninger på 0,6 mm. Det medisinske stoffet, laktose, magnesiumstearat og halvparten av stivelsen ble blandet med en passende blander. Den andre halvdelen stivelse ble suspendert i 65 ml vann og suspensjonen tilsatt den kokende oppløsningen av polyetylen-glykol i 250 ml vann. Den dannede pastaen ble tilsatt pulverene, som ble granulert hvis nødvendig med mer vann. Granulatet ble tørket over natten ved 35°C, knust på en sikt med 1,2 mm åpninger og presset til tabletter, ved hjelp av konkave stansere med delt overdel.
Eksempel 11:
Analogt ble tabletter fremstilt, inneholdene fra 10-100 mg av en av de andre forbindelsene beskrevet og eksemplifisert her. b) Fremstilling av 1000 kapsler hver inneholdene 40 mg av det aktive stoffet, for eksempel 4-[5-[4-(aminoiminometyl)-fenoksy]pentoksy]-3-N,N-bis(1-metyletyl)benzamld (Z )-2-butendioat (1:1):
Alle pulvrene ble ført gjennom en sikt med åpninger på 0,6 mm. Det medisinske stoffet ble plassert i en passende blander og blandet først med magnesiumstearat, så med laktosen og stivelsen til det ble homogent. Nr 2 hard gelatinkapsler ble fylt med 300 mg av nevnte blanding, ved hjelp av en kapsel-fy11ingsmaskin.
Analogt ble det fremstilt kapsler inneholdene 10-100 mg av de andre forbindelsene beskrevet og eksemplifisert her.

som hver er ikke-substituert eller substituert med fenyl eller fluorenyl, eller er C3-C7-cykloalkoksykarbonyl som er ikke-substituert eller mono- eller polysubstituert med laverealkyl, eller er laverealkanoyl eller fenyl laverealkanoyl, eller er benzoyl, eller er C^-Cy-alkansulfonyl, fenyl-Ci-C7-alkansulfonyl, eller fenylsulfonyl eller er aminokarbonyl som er substituert med laverealkyl, fenyl-laverealkyl eller fenyl; X} og X3 er 0;
X2 er laverealkylen,
hvor fenylringene i formel I uavhengig av hverandre kan være substituert med en eller flere substituenter valgt blant halogen, laverealkyl og laverealkoksy; eller et farmasøytisk akseptabelt salt av dette.

Anvendelse av en forbindelse med formel I, for fremstillingen av et medikament for behandling av rheumatoid arthritis, inflammatorisk tarmlidelse, psoriasis, ikke-steroidalanti-inflammatorisk medikamentindusert gastropati, voksen åndenødsyndorm (ARDS), myokardialt infarkt, allergis rhinitis, hemodialysis-indusert neutropenia og senfase-astma.

Anvendelse av en forbindelse med formel I, for fremstilling av et medikament for behandling av en tilstand eller et syndrom som responderer på den selektive antagoniseringen av LTB4~reseptorer som omfatter administrering til et pattedyr som trenger dette, en effektiv LTB4~reseptor-antagoniserende mengde av en forbindelse ifølge krav 1, eller et farmasøytisk akseptabelt salt av en forbindelse derav og en bærer.

Farmasøytisk sammensetning som passer for antagonisering av LTB4-reseptorer i pattedyr, karakterisert ved at den inneholder en effektiv LTB4~reseptor-antagoniserende mengde av en forbindelse ifølge krav 1, eller et farmasøytisk akseptabelt salt av denne og en bærer.
