Description
Oppfinnelsen vedrører analogifremgangsmåte for fremstilling av terapeutisk aktive forbindelser som er derivater av hydroksaminsyre. Videre vedrører oppfinnelsen nye forbindelser som er mellomprodukter i fremgangsmåten.
Forbindelsene fremstilt ifølge foréliggende oppfinnelse virker som inhibitorer for metalloproteaser involvert i vevsnedbrytning, slik som kollagenase, som initierer kollagen-nedbrytning, stromelysin (protoglykanase), gelatinase og kollagenase (IV). Det foreligger bevismateriale som trekker kollagenase inn som ett av nøkkelenzymene i~nedbrytningen av leddbrusk og -ben ved reumatoid artritt (Arthritis and Rheumatism, 20, 1231-1239, 1977). Sterke inhibitorer for kollagenase og andre metalloproteaser involvert i vevsnedbrytning er anvendbare ved behandlingen av reumatoid artritt og beslektede sykdommer hvor kollagenolytisk aktivitet er viktig. Inhibitorer for metalloproteaser av denne type kan derfor brukes ved behandling eller forhindring av tilstander som omfatter vevsnedbrytning; de er derfor anvendbare ved behandlingen av artropati, dermatologiske tilstander, benresorp-sjon, inflammatoriske sykdommer og tumorinvasjon, og i beford-ringen av sårheling. Nærmere bestemt kan forbindelser ifølge foreliggende oppfinnelse være anvendbare ved behandlingen av slike osteopenier som osteoporose, reumatoid artritt, osteo-artritt, periodontitt, gingivitt, hornhinnesårdannelse og tumorinvasj on.
Det er blitt beskrevet en rekke småpeptidlignende forbindelser som inhiberer metalloproteaser. Kanskje de mest bemerkelsesverdige av disse er de som vedrører angiotensin-omdannende enzym (ACE), hvor slike midler virker til å blokkere omdannelsen av dekapeptidet angiotensin I til angiotensin II som er et sterkt pressorstoff. Forbindelse av denne type er beskrevet i EP-A-0012401.
Visse hydroksaminsyrer er blitt foreslått som kollagenaseinhibitorer, slik som i US-A-4599361 og EP-A-0236872. Andre hydroksaminsyrer er blitt fremstilt som ACE-inhibitorer, f.eks. i US-A-4105789, mens ytterligere andre ér blitt beskrevet som enkefalinaseinhibitorer, slik som i US-A-4496540.
I EP-A-0012401 beskrives antihypertensivforbindelser med formelen:

I US-A-4496540 beskrives forbindelser med den generelle formel:

Det ville være ønskelig å forbedre oppløseligheten av kjente kollagenaseinhibitorer og/eller stromelysininhibitorer (enten som den frie base eller som saltet) og dessuten har det også vært søkt etter økninger i aktivitet. Det er imidlertid ingen enkel sak å forutsi hvilke variasjoner i kjente forbindelser som ville være ønskelige for å -øke eller til og med bibeholde aktivitet; visse modifikasjoner av kjente hydroks-aminsyrederivater er blitt funnet å føre til aktivitetstap.
Ifølge et første aspekt av oppfinnelsen er det tilveiebrakt en analogifremgangsmåte for fremstilling av en terapeutisk aktiv forbindelse med den generelle formel I:

Som her brukt, henviser uttrykket "C^-Cg-alkyl" til en alkylrest med rett eller forgrenet kjede som har fra ett til seks karbonatomer, inkludert f.eks. metyl, etyl, propyl, isopropyl, butyl, t-butyl, pentyl og heksyl, og beslektede uttrykk (slik som "C1-C6-alkoksy") skal forstås tilsvarende.
Uttrykket "Ci-Cg-alkenyl" henviser til en alkylrest med rett eller forgrenet kjede som har 1-6 karbonatomer og i tillegg har en dobbeltbinding, enten av E- eller Z-stereokjemi. Uttrykket "aminosyresidekjede" betyr en karakteris-tisk sidekjede bundet til -CH(NH2) (COOH)-resten i de følgende R- eller S-aminosyrer: glysin, alanin, valin, leucin, isoleucin, fenylalanin, tyrosin, tryptofan, serin, treonin, cystein, metionin, asparagin, glutamin, lysin, histidin, arginin, glutaminsyre og asparaginsyre.
Det er flere chirale sentrer i forbindelsene ifølge oppfinnelsen pga. tilstedeværelsen av asymmetriske karbonatomer. Tilstedeværelsen av flere asymmetriske karbonatomer gir opphav til flere diastereomerer med den passende R- eller S-stereokjemi i hvert chirale sentrum. Den generelle formel I og, der det passer slik, alle andre formler i denne beskrivelse skal forstås å omfatte alle slike stereoispmerer og blandinger (f.eks. racemiske blandinger) derav. Forbindelser hvor det chirale sentrum ved siden av substituenten R<3> har S-stereokjemi, er foretrukket.
Særlig foretrukne forbindelser er: [4-(N-hydroksyamino)-2R-isobutylsuccinyl]-L-fenylalaninrN-CS-aminometylpyridinJamid og [4-(N-hydroksyamino)-2R-isobutyl-3S-metylsuccinyl]-L-fenylalanin-N-[4-(2-aminoetyl) - morfolino]amid på grunn av deres gode kollagenaseinhiberende og protoglykanaseinhiberende virkninger, og den først nevnte er den mest foretrukne.

Som nevnte ovenfor, er forbindelser med den generelle formel I anvendbare i human- eller veterinærmedisin ettersom de er aktive inhibitorer for metalloproteaser involvert i vevsnedbrytning.
Forbindelser med den generelle formel I kan således brukes i en fremgangsmåte for behandling av sykdom som omfatter vevsnedbrytning, særlig reumatoid artritt, og/eller i en fremgangsmåte for befordring av sårheling, idet fremgangsmåten i begge tilfellene omfatter administrering til en menneske-eller dyrepasient av en effektiv mengde av en forbindelse med den generelle formel I.
Styrken av forbindelser med den generelle formel I når det gjelder å virke som inhibitorer for kollagenase (en metalloprotease involvert i vevsnedbrytning) ble bestemt ved hjelp av fremgangsmåten til Cawston and Barrett (Anal. Biochem., 99_, 340-345, 1979), og deres styrke når det gjelder å virke som inhibitorer for stromelysin ble bestemt ved å bruke fremgangsmåten ifølge Cawston et al (Biochenu J., 195, 159-165, 1981), hvorav begge teknikker vil bli beskrevet mer fullstendig nedenunder.
Forbindelser med den generelle formel I kan formuleres for administrering på hvilken som helst måte og dette vil være avhengig av sykdommen som skal behandles. De kan være i form av tabletter, kapsler, pulvere, granuler, pastiller, flytende preparater eller gelpreparater, slik som orale, topiske eller sterile, parenterale oppløsninger eller suspen-sjoner.
Tabletter og kapsler for oral administrering kan være i enhetsdosepresentasjonsform og kan inneholde vanlige hjelpe-stoffer, slik som bindemidler, f.eks. sirup, akasie, gelatin, sorbitol, tragant eller polyvinylpyrrolidon; fyllstoffer, som f.eks. laktose, sukker, maisstivelse, kalsiumfosfat, sorbitol eller glysin; tabletteringssmøremidler, f.eks. magnesiumstea-rat, talkum, polyetylenglykol eller silika; desintegrasjons-midler, f.eks. potetstivelse; eller akseptable fuktemidler, slik som natriumlaurylsulfat. Tablettene kan belegges i henhold til fremgangsmåter som er godt kjent innen normal farma-søytisk praksis.
Orale, flytende preparater kan være i form av f.eks. vann- eller oljesuspensjoner, -oppløsninger, -emulsjoner, -siruper eller -eliksirer, eller kan presenteres som et tørt produkt for rekondisjonering med vann eller annen egnet opp-løsning før bruk. Slike flytende preparater kan inneholde vanlige additiver, slik som oppslemmingsmidler, f.eks. sorbitol, sirup, metylcellulose, glukosesirup, gelatin, hydrogener-te, spiselige fettstoffer; emulgeringsmidler, f.eks. lecitin, sorbitanmonooleat eller akasie; ikke-vandige oppløsninger (som kan omfatte spiselige oljer), f.eks. mandelolje, fraksjonert kokosnøttolje, oljeestere, slik som glyserol, propylenglykol eller etylalkohol; preserveringsmidler, f.eks. metyl- eller propyl-p-hydroksybenzoat eller sorbinsyre, og om ønsket, vanlige smaksstoffer eller fargestoffer.
Doseringsenheten som brukes ved oral administrering kan inneholde fra ca. 1 til 250 mg, fortrinnsvis fra ca. 25 til 250 mg, av en forbindelse med den generelle formel I. En passende daglig dose for et pattedyr kan variere bredt, avhengig av pasientens tilstand og vil til sist avhenge av legen eller veterinærens bedømmelse. En dose av en forbindelse med den generelle formel I på ca. 0,1 til 300 mg/kg kroppsvekt, særlig fra ca. 1 til 100 mg/kg kroppsvekt, kan imidlertid være passende.
For topisk påføring på huden kan legemidlet lages som en krem, lotion eller salve. Krem- eller salvepreparater som kan brukes for legemidlet, er vanlige preparater som er godt kjent innen teknikken, f.eks. som beskrevet i standardlære-bøker innen farmasøytika, slik som "British Pharmacopoeia".
For topiske anvendelser på øyet kan legemidlet lages som en oppløsning eller suspensjon i en egnet, steril, vandig eller ikke-vandig bærer. Additiver, f.eks. buffere, slik som natriummetabisulfitt eller dinatriumedeat; preserveringsmidler inkludert bakteriedrepende og soppdrepende midler, slik som fenylkvikksølvacetat eller -nitrat, benzalkoniumklorid eller klorheksidin, og fortykningsmidler, slik som hypromellose, kan også være inkludert.
Den anvendte dosering for den topiske administrering vil selvsagt avhenge av størrelsen på området som skal behandles. For øynene vil hver dose vanligvis være i området fra 10 til 100 mg av forbindelsen med den generelle formel I.
Den aktive bestanddel kan også administreres parente-ralt i et sterilt medium. Legemidlet kan, avhengig av den anvendte bærer og konsentrasjonen, enten være oppslemmet eller oppløst i bæreren. Slike adjuvanser som et lokalanestetikum, preserveringsmidler og buffermidler, kan fordelaktig være opp-løst i bæreren.
For bruk ved behandlingen av reumatoid artritt kan forbindelsene ifølge denne oppfinnelse administreres ad oral vei eller ved injeksjon intraartikulert i det angrepne ledd. Den daglige dosering for et pattedyr på 70 kg vil være i området 10 mg til 1 g av en forbindelse med den generelle formel

Kollaqenaseinhiberingsaktivitet
Styrken til forbindelser med den generelle formel I når det gjelder å virke som inhibitorer for kollagenase (en metalloprotease involvert i vevsnedbrytning), ble bestemt ved fremgangsmåten til Cawston and Barrett, (Anal. Biochem., 99. 340-345, 1979), hvorved en 1 mM oppløsning av inhibitoren som testes, eller fortynninger derav, ble inkubert ved 37°C i 16 timer med kollagen og kollagenase (bufret med 25 mM hepes, pH 7,5, inneholdende 5 mM CaCl2, 0,05 % Brij 35 og 0,02 % NaN3). Kollagenet ble acetylert <14>C-kollagen fremstilt ved fremgangsmåten til Cawston and Murphy (Methods in Enzymology, 80, 711, 1981). Prøvene ble sentrifugert for å sedimentere uoppløst kollagen og en aliquot av den radioaktive supernatant fjernet for analyse på en scintillasjonsteller som et mål på hydrolyse. Kollagenaseaktiviteten i nærvær av 1 mM inhibitor, eller en fortynning derav, ble sammenlignet med aktivitet i en kontroll som var uten inhibitor, og resultatene rapportert nedenunder som den inhibitorkonsentrasjon som bevirker 50 % inhibering av kollagenasen (IC50).
Stromelysininhiberinqsaktivitet
Styrken til forbindelser med den generelle formel I når det gjelder å virke som inhibitorer for stromelysin, ble bestemt ved å bruke fremgangsmåten til Cawston et al (Biochem. J., 195, 159-165, 1981), hvorved en 1 mM oppløsning av inhibitoren -som testes, eller fortynninger derav, ble inkubert ved 37°C i 16 timer med stromelysin og <14>C-acetylatkasein (bufret med 25 mM hepes, pH 7,5, inneholdende 5 mM CaCl2, 0,05 % Brij 35 og 0,02 % NaN3). Kaseinet ble <14>C-acetylert i henhold til fremgangsmåten beskrevet i Cawston et al (Biochem. J., 195, 159-165, 1981). Stromelysinaktiviteten i nærvær av 1 mM, eller en fortynning derav, ble sammenlignet med aktivitet i en kontroll uten inhibitor, og resultatene rapportert nedenunder som den inhibitorkonsentrasjon som bevirker 50 % inhibering av stromelysinet (IC50).

succinyl1- L- fenylalanin- metylester
Benzyl-(2-benzyloksykarbonyl-5-metyl-3R-tert. - butoksykarbonyl)-heksanoat (52 g, 115 mmol) ble omrørt ved værelsestemperatur med 5 % vann i 250 ml TFA i 1,5 time. Etter denne tiden ble TFA avdampet under redusert trykk og så ble resten azeotropbehandlet med toluen (3 x 250 ml).
Den urene syre fra denne reaksjonen ble oppløst i DCM/DMF (4:1) og så ble HOBT (16 g, 118 mmol), NMM (12 g,
118 mmol) og WSCDI (22 g, 115 mmol) tilsatt ved værelsestemperatur. Etter 20 minutter ble en ytterligere ekvivalent av NMM (12 g, 118 mmol) tilsatt etterfulgt av L-fenylalaninmetyl-esterhydroklorid (23 g, 107 mmol). Denne oppløsningen ble omrørt over natten og så oppkonsentrert under vakuum. Olje-resten ble oppløst i DCM og så vasket med 10 % sitronsyre (2 x 250 ml), med 10 % natriumbikarbonat (2 x 250 ml) og én gang med mettet saltoppløsning (250 ml). Det organiske lag ble tørket (natriumsulfat), filtrert og så ble oppløsningsmidlet fjernet under redusert trykk, hvorved man fikk tittelforbindelsen som en olje (50,9 g, 79 %).

Produktet ovenfor (50,9 g, 91 mmol) ble oppløst i 100 ml etanol og omrørt ved værelsestemperatur med aktivkull-pellets i 1 time. 10 % palladium-på-karbon (20 g) i etylacetat ble oppslemmet i den etanoliske oppløsning. 20 ml cykloheksen i 100 ml etanol ble tilsatt og blandingen ble brakt til koking under tilbakeløpskjøling i 5 timer. Reaksjonsblandingen ble filtrert for å fjerne katalysatoren og så ble oppløsnings-midlene avdampet under redusert trykk, hvorved man fikk en gul olje (29,8 g). Denne oljen ble tatt opp i 500 ml xylen og varmet opp under tilbakeløpskjøling i 10 minutter. Xylenet ble fjernet under redusert trykk, hvorved man fikk det urene materialet som en olje (26,5 g).
(c) r 4-( N- benzyloksvamino)- 2R- isobutylsuccinyl" l - L- f enyl-alanin- metylester
Den urene syre (26,5 g, 79 mmol) ble oppløst i DCM/ DMF (4:1, 500 ml), så ble NMM (9,6 g, 95 mmol), HOBT (12,8 g, 95 mmol) og WSCDI (18,2 g, 95 mmol) tilsatt og blandingen ble omrørt ved værelsestemperatur inntil tic indikerte fullstendig omdannelse til den aktiverte ester (ca. 10 minutter). Til denne oppløsningen som inneholdt den aktive ester, ble det tilsatt benzylhydroksylaminhydroklorid (15,2 g, 95 mmol) og en ytterligere ekvivalent av NMM (9,6 g, 95 mmol) i oppløsnings-middelblandingen (80 ml). Etter omrøring ved værelsestemperatur over natten ble DCM (250 ml) tilsatt og blandingen ble så vasket med sitronsyre (2 x 250 ml), 10 % natriumbikarbonat-oppløsning (2 x 250 ml) og saltoppløsning (250 ml), og så til sist tørket over natriumsulfat. Oppløsningen ble filtrert og oppløsningsmidlet fjernet under redusert trykk, hvorved man fikk en olje (27,2 g) som ble renset ved kolonnekromatografi under anvendelse av eter som elueringsmiddel, hvorved man fikk tittelforbindelsen (11 g, 23,7 mmol, 30 %).

[4-(N-benzyloksyamino)-2R-isobutylsuccinyl]-L-fenylalanin-metylester (9,5 g, 21 mmol) ble oppløst i 200 ml metanol, og litiumhydroksidoppløsning (0,5 N, 84 ml, 42 mmol) ble tilsatt med omrøring ved værelsestemperatur. Etter at omsetningen var fullstendig, bedømt ut fra tic, ble metanolen fjernet ved avdamping og den gjenværende vannfase ble surgjort til pH 1 med sitronsyre. Det utfelte faststoffet ble frafiltrert og tørket mens filtratet ble ekstrahert med 500 ml DCM og tørket over natriumsulfat. Oppløsningsmiddelfjerning fra den organiske fase ga en olje (5,38 g) som kunne rekrystalliseres fra diisopropylester og metanol, hvorved man fikk materialet som var identisk med faststoffet som utfeltes under surgjøring. Disse to mengdene ble slått sammen, hvorved man fikk tittelforbindelsen (6,40 g, 15 mmol, 71 %).

[4-(N-benzyloksyamino)-2R-isobutylsuccinyl]-L-fenylalanin (200 mg, 0,47 mmol) ble oppløst i 6 ml THF og avkjølt i is. Trietylamin (52 mg, 0,522 mmol) ble tilsatt sammen med etylklorformiat (50 mg, 0,47 mmol) og etter 10 minutter ble 2-(2-aminoetyl)-N-metylpyrrolidin (67 mg, 0,522 mmol) i THF
(1 ml) tilsatt. Etter 3 timer ved værelsestemperatur ble reaksjonsblandingen fortynnet med etylacetat, så ble det vasket med natriumbikarbonatoppløsning og saltoppløsning, og så tørket over natriumsulfat. Oppløsningsmiddelfjerning under redusert trykk ga det urene benzylhydroksamat (220 mg, 0, 41 mmol).
Det urene materialet ovenfor ble oppløst i cykloheksen/etanol (10 % oppløsning, 5 ml), 10 % palladium-på-karbon (50 mg) ble tilsatt og blandingen ble så kokt under tilbakeløpskjøling inntil utgangsmaterialet hadde forsvunnet i henhold til tic (ca. 30 minutter). Katalysatoren ble fjernet ved filtrering og oppløsningsmidlet fjernet under redusert trykk, hvorved man fikk en olje som kunne utkrystalliseres ved tilsetning av heksan. Det erholdte produkt (150 mg, 0,34 mmol, 72 %) ble samlet opp ved filtrering.

(a) Fenylalanin-N-f 2-(2-amin oetyl)- 1- metylpyrrol] amid Til en oppløsning av N-(benzyloksykarbonyl)-fenylalanin (8,00 g, 26,7 mmol), 2-(2-aminoetyl)-1-metylpyrrol (3,65 g, 29,4 mmol), HOBT (3,98 g, 29,4 mmol) og NMM (2,97 g, 29,4 mmol) i DMF avkjølt til 0°C ble det tilsatt en oppløsning av DCC (6,07 g, 29,4 mmol) i THF. Etter at tilsetningen var fullført ble reaksjonsblandingen hensatt under omrøring ved værelsestemperatur over natten. Oppløsningsmidlene ble fjernet under redusert trykk og det gjenværende faststoffet ble tatt opp i etylacetat og vasket med vann, 10 % natriumbikarbonat og saltoppløsning, og så tørket over natriumsulfat. Filtrering og oppløsningsmiddelfjerning ga det urene benzyloksy-beskyttede materialet som ble omdannet til det frie amin ved oppløsning i etanol/cykloheksen (10 % oppløsning, 150 ml), tilsetning av 10 % palladium-på-karbon og koking under tilbakeløpskjøling i 2 timer. Denne blandingen ble filtrert for å fjerne katalysatoren, etanolen ble fjernet under redusert trykk, resten ble oppløst i etylacetat og ekstrahert med sitronsyre. Det vandige lag ble gjort basisk til pH 12 med natrium-hydroksidoppløsning og deretter ekstrahert med etylacetat, hvorved man fikk tittelforbindelsen (4,11 g, 15 mmol, 57 %). (b) [ 4- benzyloksy- 3- benzyloksykarbonyl- 2R- isobutylsuccinyl1-L-fenylalanin-N-[2-(2- aminoetyl)- 1- metylpyrrol ] amid
Til en oppløsning av fenylalanin-N-[2-(2-aminoetyl)-1-metylpyrrol]amid (2,00 g, 7,4 mmol), 4-tert.-butyl-2R-isobutyl-succinat (3,23 g, 8,1 mmol), HOBT (1,09 g, 8,1 mmol) og NMM (1,82 g, 8,1 mmol) i DMF avkjølt til 0°C ble det tilsatt DCC (1,67 g, 8,1 mmol) i THF. Den resulterende blanding fikk varmes opp til værelsestemperatur og så ble om-røring fortsatt i 72 timer. Det utfelte DCU ble samlet opp ved filtrering og oppløsningsmidlene ble så fjernet under redusert trykk. Den gjenværende olje ble oppløst i etylacetat, vasket med 10 % natriumbikarbonat, vann og saltoppløsning, og så tørket over natriumsulfat. Oppløsningsmiddelfjerning under redusert trykk ga det urene produkt (5,48 g) som en olje. Dette materialet ble renset ved kolonnekromatografi under anvendelse av 40 % etylacetat i heksan som elueringsmiddel, hvorved man fikk den ønskede isomer (1,45 g, 2,2 mmol, 30 %) fullstendig atskilt fra [4-benzyloksy-3-benzyloksykarbonyl-2S-isobutylsuccinyl]-L-fenylalanin-N-[2-(2-aminoetyl)-1-metylpyrrol ]amidisomeren (1,04 g, 1,6 mmol, 22 %).

[4-benzyloksy-3-benzyloksykarbonyl-2R-isobutylsuccinyl]-L-fenylalanin-N-[2-(2-aminoetyl)-1-metylpyrrol]amid (1,45 g, 2,3 mmol) ble oppløst i etanol/cykloheksen (10 % opp-løsning), 10 % palladium-på-karbon (0,14 g) ble tilsatt og blandingen ble kokt under tilbakeløpskjøling i 2 timer. Etter denne tiden viste tic fullstendig forbruk av utgangsmateriale slik at reaksjonsblandingen ble avkjølt, filtrert gjennom celitt og oppløsningsmidlet fjernet under vakuum. Den resulterende olje ble tatt opp i toluen og kokt under tilbake-løpskjøling i 2 timer, oppløsningsmiddelfjerning ga så det urene materialet (1,09 g, 2,6 mmol) som ble brukt uten ytterligere rensing.

[ 4-hydroksy-2R-is.obutylsuccinyl ] -L-fenylalanin-N- [ 2-(2-aminoetyl)-1-metylpyrrol]amid (0,68 g, 1,6 mmol), benzylhydroksylamin (0,28 g, 1,8 mmol), NMM (0,18 g, 1,8 mmol) og HOBT (0,24 g, 1,8 mmol) ble oppløst i DMF og avkjølt til 0°C. DCC (0,36 g, 1,8 mmol) i THF ble dråpevis tilsatt og da tilsetningen var fullstendig, fikk blandingen omrøres over natten. Det utfelte DCU ble fjernet ved filtrering, opp-løsningsmidlene ble fjernet fra filtratet under redusert trykk, resten oppløst i etylacetat (50 ml) og vasket med 10 % natriumbikarbonat (2 x 50 ml), vann (50 ml) og saltoppløsning (50 ml), så ble det tørket over natriumsulfat. Filtrering og
fjerning av etylacetatet ga det urene produkt (1,25 g) som en olje. Ytterligere rensing ble oppnådd ved kolonnekromatografi under anvendelse av 2 % metanol/DCM som elueringsmiddel, hvorved man fikk tittelforbindelsen (0,71 g, 1,34 mmol, 83 %).

a) Fenylalanin- N-[ 4-( 2- aminoetyl) morfolino] amid
Til en oppløsning av N-(benzyloksykarbonyl)-fenylalanin (29,91 g, 100 mmol), 4-(2-aminoetyl)-morfolin (14,32 g, 110 mmol), HOBT (14,86 g, 110 mmol) og NMM (11,13 g, 110 mmol) i DGM/DMF (4:1) ble det tilsatt DCC (6,07 g, 29,4 mmol). Etter at tilsetningen var fullført, ble reaksjonsblandingen hensatt for omrøring ved værelsestemperatur over natten. Opp-løsningsmidlene ble fjernet under redusert trykk og det gjenværende faststoffet ble tatt opp i etylacetat og vasket med vann, 10 % natriumbikarbonat og saltoppløsning. Etylacetat laget ble så ekstrahert med 3 % saltsyre, den vandige oppløsning fraskilt og gjort basisk til pH 11. Denne opp-løsningen ble ekstrahert med etylacetat, tørket over natriumsulfat og så ga oppløsningsmiddelfjerning aminet som et urent faststoff som ble rekrystallisert fra etylacetat/heksan (44,78 g, 109 mmol, 100 %).

Til en oppløsning av fenylalanin-N-[4-(2-aminoetyl)-morfolin]amid (19,16 g, 69 mmol), 4-tert.-butyl-2R-isobutyl-succinat (18,57 g, 76 mmol), HOBT (10,27 g, 76 mmol) og NMM (7,69 g, 76 mmol) i DMF avkjølt til 0°C ble det tilsatt DCC (15,68 g, 76 mmol) i THF. Den resulterende blanding fikk varmes opp til værelsestemperatur og så ble omrøring fortsatt over weekenden. Det utfelte DCU ble samlet opp ved filtrering og så ble oppløsningsmidlene fjernet under redusert trykk. Den gjenværende olje ble oppløst i etylacetat, det ble vasket med 10 % natriumbikarbonat, vann og saltoppløsning, og så tørket over natriumsulfat. Oppløsningsmiddelfjerning under redusert trykk ga det urene produkt (36,5 g) som en olje. Dette materialet ble renset og diastereomerene separert ved forsiktig kolonnekromatografi under anvendelse av 0 - 5 % metanol i DCM som elueringsmiddel, hvorved man fikk den ønskede isomer svakt forurenset med en lavereløpende isomer (7,86 g, 15,6 mmol, 23 %).

[4-tert.-butoksy-2R-isobutyl-3S-metylsuccinyl]-L-fenylalanin-N-[4-(2-aminoetyl)-morfolin]amid (2,37 g, 4,7 mmol) ble oppløst i 5 % vann i TFA (15 ml) med avkjøling og omrøring i 90 minutter, hvoretter tic indikerte at av-beskyttelse var fullstendig. Fjerning av det overskytende TFA ble oppnådd ved azeotrop behandling med xylen, hvorved man fikk den urene syre som ble brukt uten ytterligere rensing. Dette produktet ble tatt opp i DCM/DMF (4:1, 20 ml) og pH ble regulert til 7 ved å tilsette NMM. Til dette ble det tilsatt HOBT (0,76 g, 5,6 mmol) og NMM (0,57 g, 5,6 .mmol) og så, etter avkjøling til 0°C, WSCDI (1,08 g, 5,6 mmol). Denne blandingen ble hensatt for omrøring ved værelsestemperatur i 45 minutter, avkjølt i et isbad og benzylhydroksylamin (0,90 g, 5,6 mmol) ble tilsatt og til sist ble det omrørt over natten. (a) [ 4- tert.- butoksy- 2( RS)- isobutylsuccinyl]-L-fenyl-alaninbenzylester
Til en oppløsning av fenylalaninbenzylester-tosin-syresalt (6,74 g, 17,6 mmol), 4-tert.-butyl-2(RS)-isobutyl-succinat (3,68 g, 16,0 mmol), HOBT (2,38 g, 17,6 mmol) og NMM (1,78 g, 17,6 mmol) i DMF (20 ml) ved 0°C ble det tilsatt en oppløsning av DCC (3,63 g, 17,6 mmol) i THF (20 ml). Den resulterende blanding fikk varmes opp til værelsestemperatur og ble så omrørt over natten. Det utfelte DGU ble fjernet ved filtrering og så ble oppløsningen oppkonsentrert under redusert trykk. Restoljen ble tatt opp i etylacetat og vasket i rekkefølge med 10 % natriumbikarbonat, fortynnet saltsyre, vann og endelig saltoppløsning. Oppløsningen ble tørket over natriumsulfat og etylacetatet fjernet under redusert trykk. Det således fremstilte, urene materialet ble renset ved kolonnekromatografi under anvendelse av gradienteluering med etylacetat i DCM (0-15 %), hvorved man fikk tittelforbindelsen (3,16 g, 6,7 mmol, 42 %).
(b) [ 4- tert.- butoksy- 2( RS)- isobutylsuccinyl]- L- fenylalanin
[4-tert.-butoksy-2(RS)-isobutylsuccinyl]-L-fenyl-alaninbenzylester (3,16 g, 6,7 mmol) ble oppløst i cykloheksen/etanol (10 % oppløsning, 50 ml), 10 % palladium-på-karbon (2,0 g) ble tilsatt og blandingen ble kokt under til-bakeløpskjøling i 10 minutter. Den avkjølte oppløsning ble filtrert gjennom celitt og oppløsningsmidlet fjernet, hvorved tittelforbindelsen ble tilbake (2,25 g, 6,0 mmol, 89 %).

a) 2R- brom- 5- metylpentansyre
D-leucin (100 g, 0,76 mol) og kaliumbromid (317,5 g,
2,67 mol) ble oppløst i vandig syre (150 ml konsentrert svovelsyre i 500 ml vann). Oppløsningen ble avkjølt til like under 0°C og natriumnitritt (69,6 g, 0,95 mol i vann) ble tilsatt i løpet av 1 time idet det ble passet på å holde temperaturen mellom -1 og -2°C. Etter at tilsetningen var fullført, ble blandingen holdt ved 0°C i en ytterligere time og så ble DCM tilsatt og blandingen omrørt i noen få minutter. Lagene ble separert og vannfasen ble vasket med ytterligere porsjoner av DCM (5 x 250 ml). b) tert.- butyl- 2R- brom- 5- metylpentanoat 2R-brom-5-metylpentansyre (123 g, 0,63 mol) ble opp-løst i DCM (400 ml) og oppløsningen ble avkjølt til -40°C mens isobuten ble kondensert inn for grovt sett å fordoble volumet. Mens temperaturen ble holdt ved -40°C ble konsentrert svovelsyre (4 ml) tilsatt dråpevis. Etter at tilsetningen var fullstendig, fikk reaksjonsblandingen varmes opp til værelsestemperatur over natten. Den resulterende oppløsning ble oppkonsentrert til det halve volum ved å fjerne oppløsningsmidlet ved redusert trykk, og så ble DCM vasket to ganger med et likt volum av 10 % natriumbikarbonatoppløsning. c) benzyl-( 2- benzyloksykarbonyl- 3R-( tert.- butoksy-karbonyl)- 5- metylheksanoat
Dibenzylmalonat (124,5 g, 0,44 mol) ble tatt opp i .tørt DMF og kalium-tert.-butoksid (49,2 g, 0,44 mol) ble tilsatt porsjonsvis med omrøring og avkjøling. Da den homogene oppløsning var blitt dannet, ble den avkjølt til 0°C og så ble tert.-butyl-2R-brom-5-metylpentanoat (110,0 g, 0,44 mol) i DMF (200 ml) tilsatt dråpevis i løpet av 1 time. Etter at tilsetningen var fullført ble reaksjonsblandingen overført til et kaldt rom ved <5°C og hensatt i 4 dager. Reaksjonsblandingen ble fordelt mellom etylacetat og mettet ammoniumklorid, og så ble vannlaget ekstrahert med ytterligere etylacetat (4 x 500 ml), tørking og oppløsningsmiddelfjerning etterlot en olje (228 g) som var svært forurenset med DMF. Denne oljen ble tatt opp i eter (1 liter) og vasket med saltoppløsning (2x1 liter), og så ble det organiske lag tørket (magnesiumsulfat), oppløsningsmidlet fjernet slik at det ønskede materialet (179 g) forurenset med en liten mengde dibenzylmalonat, ble tilbake.

d) r4-benzyloksy-3- benzyloksykarbonyl- 2R- isobutylsuccinyl]- L- fenylalanin- N-( 3- aminometylpyridin)- amid
Benzyl(2-benzyloksykarbonyl-5-metyl-3R-tert.-butoksy-karbonyl)heksanoat (40 g) ble tatt opp i 5 % vann i TFA
(105 ml) og fikk stå ved 5°C over natten. Etter dette tidsrom ble TFA avdampet under redusert trykk og så ble resten fordelt mellom DCM (250 ml) og saltoppløsning (50 ml). Oppløsnings-middelf jerning etterlot en olje som utkrystalliserte ved henstand (30 g).
Den urene syre fra denne reaksjonen ble oppløst i 250 ml DMF og så ble HOBT (13,5 g, 90 mmol), NMM (9,1 g, 90 mmol) og fenylalanin-N-(3-aminometylpyridin)-amid (23 g, 90 mmol) tilsatt ved værelsestemperatur. Blandingen ble avkjølt til 0°C før dråpevis tilsetning av DCC (18,5 g, 90 mmol) i THF (250 ml). Denne oppløsningen ble omrørt til værelsestemperatur i løpet av weekenden. Det utfelte DCU ble fjernet ved filtrering og så ble oppløsningsmidlene fjernet fra filtratet under redusert trykk, hvorved det ble tilbake en olje. Denne oljerest ble oppløst i etylacetat og så vasket med 10 % sitronsyre, 10 % natriumbikarbonat og mettet salt-oppløsning. Det organiske lag ble tørket (magnesiumsulfat), filtrert og oppløsningsmidlet ble så fjernet under redusert trykk, hvorved man fikk tittelforbindelsen som en olje (73 g). Dette materialet ble tilført en kolonne med silika under anvendelse av gradienteluering (0 - 50 % etylacetat i heksan) for å fjerne urenheter og fraskille en liten mengde av den underordnede diastereoisomer. Materialet fra kolonnen (29,0 g, 61 %) ble rekrystallisert fra etanol/DIPE, hvorved man fikk tittelforbindelsen som et hvitt, krystallinsk faststoff (17,1 g).
e) [ 4- hydroksy- 2R- isobutyl- 3- etenylsuccinyl]- L- fenylalanin- N- ( 3- aminometylpyridin)- amid
[4-benzyloksy-3-benzyloksykarbonyl-2R-isobutylsuccinyl]-L-fenylalanin-N-(3-aminometylpyridin)-amid (5 g, 7,9 mmol) ble tatt opp i etanol, ammoniumformiat (2,5 g, 40 mmol) ble tilsatt etterfulgt av 10 % palladium-på-karbon (1 g) som en oppslemming i isopropylalkohol. Etter 30 minutter ved værelsestemperatur ble katalysatoren fjernet ved filtrering og så ble det vasket med etanol, hvorved man fikk en oppløsning av den urene disyre. Til denne ble det tilsatt 1 g piperidin og blandingen ble omrørt ved værelsestemperatur i 15 minutter før tilsetning av vandig formaldehyd (40 % opp-løsning, 5 ml). Etter 18 timer ved værelsestemperatur ble blandingen kokt under tilbakeløpskjøling i 1 time. Opp-løsningsmidler ble fjernet ved rotasjonsinndamping og resten fordelt mellom etylacetat og sitronsyre. Det sure lag ble ekstrahert med ytterligere porsjoner av etylacetat (2 x 250 ml), de kombinerte organiske lag ble ekstrahert med kaliumkarbonat (3 x 200 ml). Disse baseekstraktene ble surgjort til pH 4 og reekstrahert med DCM, og så ble det organiske lag tørket over magnesiumsulfat. Oppløsningsmiddel-fjerning under redusert trykk ga det ønskede produkt som et hvitt faststoff (2,55 g, 77 %).
f) [ 4- hydroksy- 2R- isobutyl- 3S-( fenyltiometyl)- succinyl]-L-fenvlalanin-N-(3-ami nometylpyridin)- amid
[4-hydroksy-2R-isobutyl-3-etenylsuccinyl]-L-fenylalanin-N- (3-aminometylpyridin) -amid (lg, 2,4 -mmol) ble oppløst i 10 ml tiofenol og blandingen ble omrørt i mørket under nitrogen ved 60°C over natten. Eter ble tilsatt til den avkjølte reaksjonsblanding og det utfelte produkt samlet opp ved filtrering. Faststoffet ble vasket med store volumer eter og tørket under vakuum, hvorved man fikk tittelforbindelsen (650 mg, 51 %).
g) r 4-(N-hydroksyamino)-2R- isobutyl- 3S-( fenyltiometyl)-succinyl]- L- fenylalanin- N-( 3- aminometylpyridin)- amid
[4-hydroksy-2R-isobutyl-3S-fenyltiometyl-succinyl]-L-fenylalanin-N-(2-metylpyridyl)-amid (0,2 g, 0,4 mmol) og HOBT (0,07 g, 0,5 mmol) ble oppløst i 1:1 DCM/DMF og blandingen ble avkjølt til 0°C før tilsetning av WSDCI (0,09 g, 0,5 mmol) og NMM (0,05 g, 0,5 mmol). Blandingen ble omrørt ved 0°C i 1 time for å sikre fullstendig dannelse av den aktiverte ester. Hydroksylaminhydroklorid (0,04 g, 0,6 mmol) og NMM (0,06 g, 0,6 mmol) ble oppløst i DMF og så ble denne blandingen dråpe-
vis tilsatt til den avkjølte oppløsning av den aktiverte ester. Etter 1 time ble reaksjonsblandingen helt over i eter/ vann (1:1) hvoretter det ønskede produkt utfeltes som hvite krystaller. Disse ble samlet opp ved filtrering, videre vasket med eter og vann og så tørket under vakuum ved 50°C. Dette materialet ble rekrystallisert fra metanol/vann (1:1) for å fjerne et spor av den underordnede diastereomer (0,1 g, 0,2 mmol, 48 %).

Trinn 1. Bland bestanddelene nr. 1, nr. 2, nr. 3 og nr. 4 i en egnet blander.
Trinn 2. Tilsett tilstrekkelig vann porsjonsvis til blandingen fra trinn 1 med forsiktig blanding etter hver tilsetning. Slike tilsetninger av vann og blanding inntil massen har en konsistens som muliggjør dens omdannelse til våte granuler.
Trinn 3. Den våte masse omdannes til granuler ved å sende den gjennom en oscilleringsgranulator
under anvendelse av en sikt nr. 8 mesh (2,38). Trinn 4. De våte granuler tørkes så i en ovn ved 60°C inntil de er tørre.
