Description
The present invention concerns the use of the fibrinolytic activity to predict the meat quality of a herd and for selection based on the meat quality.
For tiden anvendes halothantesten (Augustini et al.: Die Fleischwirtschaf t, 5_7, 1028 ( 1977)) og creatinkinase (CK)-aktivitetstesten (Beret et al.: Ållattenyésztés (in English: "Animal Breeding") 29, 445 (1980) i praksis for Currently, the halothane test (Augustini et al.: Die Fleischwirtschaf t, 5_7, 1028 (1977)) and the creatine kinase (CK) activity test (Beret et al.: Ållattenyésztés (in English: "Animal Breeding") 29, 445 (1980) are used in practice for
Briskey: Adv. Food Res. 13_, 89 ( 1964) eller DFD, dvs. mørk, fast, tørr, se ibid 13, 89 (1964 ). to predict the meat quality of the herd, i.e. to determine respectively the state of stress or the defective meat quality (using the terminology of animal husbandry experts: PSE, i.e. pale mild exudative; see E.J. Briskey: Adv. Food Res. 13_, 89 ( 1964) or DFD, i.e. dark, firm, dry, see ibid 13, 89 (1964).
De dyr som har falt i søvn The principle of the halothane test, which is used only for assessing the state of stress in pigs, consists in examining whether pigs that have been anesthetized with halothane sleep in a relaxed or spastic state. The animals that have fallen asleep
En ytterligere alvorlig ulempe er at denne metoden er farlig idet 4 til 5 av 100 svin dør i et forsøk. in a spastic state and thus are stress-sensitive, are gradually removed from further breeding. This method is applicable for investigating the sensitivity to stress, but meat quality cannot be predicted with certainty using this method. A further serious disadvantage is that this method is dangerous as 4 to 5 out of 100 pigs die in an attempt.
Prinsippet med CK-aktivitetstesten er å ta en blodprøve fra venen hos dyret, bestemme aktiviteten av creatinkinase The principle of the CK activity test is to take a blood sample from the vein of the animal, determine the activity of creatine kinase
Denne metoden er således ikke tilstrekkelig pålitelig. and from there find out the stress sensitivity and thus indirectly the meat quality of the animal. According to the authors of this test method, the correlation level between the CK activity and the meat quality is no better than 49% (ie, good meat quality can be predicted only for 49 out of 100 animals). This method is thus not sufficiently reliable.
Prinsippet for denne metoden er at nærværet av HAL 3-genet påvises, idet dette P. Sellier (Vågoållat-és husfördungs (in Norwegian: "Slaughter and meat production") 10, 42 (1980)) describes a method for predicting stress sensitivity in animals. The principle of this method is that the presence of the HAL 3 gene is detected, as this
er karakteristisk for stress-følsomme dyr. Denne metoden is characteristic of stress-sensitive animals. This method

stressfølsomhet, men kan ikke anvendes for å forutse kjøtt-kvaliteten! med høy sannsynlighet. stress sensitivity, but cannot be used to predict meat quality! with high probability.
For the sake of completeness, it should be noted that in industrial practice the pH determination is used in a defined muscle group to determine the quality of the meat in slaughtered animals. However, this method is only applicable for a technological meat selection and, obviously, it cannot be used for prediction.
For tiden anvendes bestemmelsen av den fibrinolyttiske aktiviten i den medisinske praksis for undersøkelse av meget uvanlige (spesielle) kliniske bilder, f.eks. fravær av en blodfaktor. Currently, the determination of the fibrinolytic activity is used in medical practice for the examination of very unusual (special) clinical pictures, e.g. absence of a blood factor.
It is the aim of the invention to develop a method using which the meat quality of a herd can be predicted and by which the animals that are suitable for further breeding could be selected instead of the stress-sensitive animals that give a deficient meat quality.
It has been established that, by measuring the fibrinolytic activity in blood taken from the herd and from there determining the lysis percentage, there exists a threshold value for this lysis percentage depending on the species and variety of the animal,
på basis av hvilken besetningen kan klassifiseres ifølge de ovenfor nevnte synspunktene, dvs. de dyr som gir en god kjøttkvalitet kan velges til fordel fra de som gir en util-strekkelig kvalitet og på denne måten kan kjøttkvaliteten forutsies. on the basis of which the herd can be classified according to the above-mentioned points of view, i.e. the animals that give a good meat quality can be selected in favor of those that give an insufficient quality and in this way the meat quality can be predicted.
The invention therefore concerns the application of a method J for predicting the meat quality of herds and selection on the basis of the meat quality.
<j>According to the invention, the fibrinolytic activity1 is determined in fresh blood taken from the herd, the lysis percentage is determined from this, then it is further bred on those animals that show a lysis percentage that is lower than the threshold value, depending on the species and variety of the animal, while those animals which show a light percentage higher than this threshold value are immediately or gradually excluded from breeding.
In practice, the method used according to the invention is carried out as follows: First of all, it should be noted that in principle the method used according to the invention is applicable to any of the herds, taking into account the biochemical basis for the invention. In practice, however, the method used according to the invention is particularly important when breeding pigs, cattle, horses, poultry, sheep and rabbits.
Blodprøven taes passende fra øret eller halsvenen The blood sample is conveniently taken from the ear or jugular vein
For å ta blodprøven anvendes passende en sprøyte, mens et medium inneholdende et anti-koaguleringsmiddel, f.eks. en 3,8%-ig trinatriumcitratløsning, anvendes for å motta blodprøven. in the herd under conditions which enable the blood sample to be free of any haemolysis. To take the blood sample, a syringe is suitably used, while a medium containing an anti-coagulant, e.g. a 3.8% trisodium citrate solution is used to receive the blood sample.
The fibrinolytic activity is determined in the blood sample taken as described above using a
i og for seg kjent metode, f.eks. ifølge Fearnley (Clin. per se known method, e.g. according to Fearnley (Clin.
Sei. 16, 646 (1957)). Pollock. 16, 646 (1957)). The light percentage characteristic of the activity is obtained by comparing the measured values with standard values.
Taking into account the variation in the biochemical state of the herd, it is appropriate to determine the lysis percentage in three repetitions from blood samples taken repeatedly for 3 days under a minimum stress condition.
The threshold value for the lyse percentage is in the range of 25 to 35% for pigs. It is characteristic of species and variety. However, it can vary depending on the bloodline.
Hereafter such areas of animal breeding and meat processing industry will be mentioned, where the method used according to the invention can be used effectively.
By using the method used according to the invention, the herd's meat quality can be predicted. This means that, after measuring the fibrinolytic activity by means of the method and after determining the lysis percentage from there, it can be predicted whether, after slaughtering the herd, a meat of PSE quality (with poor quality) or a meat with DFD is obtained -quality (i.e. a normal meat of good quality).
Metoden for å forutsi kvaliteten av kjøtt anvendt The method for predicting the quality of meat applied
ifølge oppfinnelsen viser på svin en korrelasjon på 71,1 % med den kjøttkvalitet som bestemmes ved hjelp av den tradisjonelle metoden basert på pH 24-verdi måling av kjøtt på slaktedyr (R. according to the invention shows on pigs a correlation of 71.1% with the meat quality determined using the traditional method based on pH 24 value measurement of meat on slaughter animals (R.
Ved bruk av metoden anvendt ifølge oppfinnelsen kan arbeidet med sortering ifølge kjøttdelene etter slakting unngås. hamm and R. Potthast: Die Fleischwirtschaft 52, 200 (1972)), while this correlation is no more than 49% using the CK activity test described above under the state of the art. It is clear that the quality of the meat can be significantly predicted in a more reliable way using the method used according to the invention, a fact that is of great importance in terms of slaughterhouse practice, because it makes it possible to sort out those animals that give a meat with poor quality and processing; that according to the meat quality. By using the method used according to the invention, the work of sorting according to the meat parts after slaughter can be avoided.
Et annet området for bruk av metoden anvendt ifølge oppfinnelsen består i den kontinuerlige forbedringen av kjøtt-kvaliteten i en definert populasjon i oppdrett av en stress- Another area for use of the method used according to the invention consists in the continuous improvement of the meat quality in a defined population in the breeding of a stressed
i in
Use of the method for continuous selection of breeding animals in a defined population will result in continuous improvement of meat quality. The halothane test mentioned above under the description of the state of the art is only applicable for sorting stress sensitivity and when using this test, the separation of animals with poor meat quality cannot be carried out, as poor meat quality can also be caused in stress-resistant animals animals as a result of enzyme-induced properties in the test.
If, by the method used according to the invention, a population with an improved meat quality has been established using selection based on the meat quality, it can be expected that other, important characteristics of this population, such as e.g. stress-resistance, utilization efficiency of the forage and even

The method used according to the invention is illustrated in detail by means of the following, non-limiting example.

From groups consisting of 100 pigs, each with a hybrid build from "Hungarian large white" and "Dutch common" marked for ham production in the slaughterhouse, 10 are selected
The fibrinolytic activity is determined on the samples thus obtained, using the above-mentioned method of Fearnley and at the same time, after slaughter, pH^- and pP^4~ measurements are carried out on the selected pigs using the method described in the above mentioned literature, or a GOF024~color determination is carried out (see: J. Dohy: Allattenyésztési Genetika (in Norwegian: "Animal breeding genetics") pages 133, 153 and 200, ed. Mezogazdasågi Kiado, Budapest (1979)) and the meat quality is determined on the basis of these measurements. The fibrinolytic activity is calculated by comparing the obtained value with the standard value and the correlation between the thus obtained lysis percentage and the meat quality determined using the traditional methods is established. The meat qualities determined on the basis of the lysis percentage show a correlation of 71.1% with the meat quality determined on the basis of the pl^ value, while this correlation is 67% when the meat quality is measured using pl^ and pH24 values of 68.5 % with the meat quality determined using the GOF024 value. The correlation is 56% with meat quality determined using the CK activity test method. It is clear that on the one hand the method used according to the invention is in good correlation with the known methods, the disadvantages of which are described above, and on the other hand the method used according to the invention is in itself more applicable for determining the meat quality.
