Abstract
Nye 8-(lavere alkyl)-bicyclo-. [4.2.0)-octanderivater som er anvendelige. for behandling av kardiovaskulære forstyrrelser og for inhibering av utskillelsen av magesyre. Derivatene er forbindelser med formlene. hvor Y er exo-(lavere alkyl) eller endo-(lavere alkyl), n er 2 eller 3, Ri er CHjOH, CHO, COR eller COH, Rer hydrogen eller methyl, og Rer rettkjedet eller forgrenet alkyl med 5-10 carbonatomer,som eventuelt kan være substituert med lavere alkyl, lavere alkoxy, trifluor-methyl eller halogen, hvor a er 0, 1 eller 2, b er 3-7, m er 1 eller 2, og. hvor hver Ruavhengig av de øvrige er hydrogen eller lavere alkyl med 1-6. carbonatomer,. og de i farmasøytisk henseende aksepterbare, ikke-toksiske salter og estere derav. Fremstillingen av de nye 8-(lavere alkyl)-bicyclo-[4.2.0]-octanderivater er beskrevet.
Description
Denne oppfinnelse angår fremstilling av nye, terapeutisk virksomme 8-(lavere alkyl)-bicyclo-[4.2.0]-octan-derivater og i farmasøytisk henseende aksepterbare, ikke-toksiske salter og estere derav, som er anvendelige for behandling av kardiovaskulære forstyrrelser og inhibering av utskillelsen av magesyre.
Bicyclo-[4.2.0]-oct-2-en-7-on lar seg lett fremstille ut fra 1-3-cyclohexadien (Tetrahedron, 27:615, 1971). Forbindelsen er blitt benyttet som et mellomprodukt ved en prostaglandinsyntese (Tetrahedron Lett., 3091, 1973).
Det er kjent flere prostaglandin-analoger som inneholder bisykliske skjeletter med utelukkende carbon i skjelettene. Carbacyclin inneholder et bicyclo-[3.3.0]-octanskjelett og er beskrevet i flere publikasjoner (J. Chem. Soc, Chem. Commun., 1067, 1978; Tetrahedron Lett., 3743, 1978; Tetrahedron Lett., 433, 1979, Tetrahedron Lett., 2807, 1979; J. Org. Chem. 44:2880, 1979) og i patentlittera-turen (belgisk patentskrift nr. 874 135, britisk patentskrift nr. 2 014 143, fransk patentskrift nr. 2 424 908, tysk off.skrift nr. 2 904 655, japansk patentskrift K 79 117 450, nederlandske patentsøknader nr. 7 901 076 og 8 003 579 og syd-afrikansk patentskrift nr. 79 00 176). Tallrike analoger av carbaprostacyclin er likeledes beskrevet (US patentskrift nr. 4 306 076, tysk off.skrift 3 146 278, tysk off.skrift nr. 3 204 443, Prostaglandins, Leukotrienes, Med., 9:307, 1982; J. Org. Chem. 48, 5341, 1983; Tetrahedron Lett. 3493, 1983; Biochem. Pharmacol. 32:2405, 1983; Prostaglandins, Leukotrienes, Med. 11:391, 1983).
Syntetiske prostaglandiner (homo-PGE2 og homo-PGF2a) er blitt fremstilt med hydroxylfunksjonen og en lavere sidekjede i trans-stilling i forhold til hverandre i en 6-leddet ring (Tetrahedron Lett., 3327, 1971).
De nye terapeutisk virksomme 8-(lavere alkyl)-bicyclo-[4.2.0]-octanderivater er forbindelser med de generelle formler (1), (2) og (3):

farmasøytisk henseende aksepterbare, ikke-toksiske salter og estere derav.
Forbindelsene fremstilles ved at en forbindelse med den generelle formel: hvor Y og R3 er som ovenfor angitt, omsettes med anionet (eller ylidet) av en fosfoniumforbindelse med den generelle formel:
hvor X, Y og Z er alifatiske eller aromatiske grupper, og n er 2 eller 3, eller et beskyttet derivat eller salt derav, for dannelse av en blanding av en forbindelse med formel (1) og en forbindelse med formel (2) eller en blanding av forbindelse (3) og den tilsvarende E-forbindelse, og den opp-
nådde blanding separeres, og at en syre med formel (1), (2) eller (3) eventuelt overføres til et i farmasøytisk henseende aksepterbart, ikke-toksisk salt eller ester derav.
Slik det her benyttes, menes med uttrykket "i farmasøytisk henseende aksepterbare, ikke-toksiske salter" carboxylsyresalter erholdt ved omsetning av COOH-gruppen i formel (1), (2) eller (3) med en egnet organisk eller uorganisk base. Spesifikke fremstillinger er beskrevet nedenfor.
De i farmasøytisk henseende aksepterbare carboxylsyreestere svarende til syrene med formler (1), (2) og (3) fremstilles etter konvensjonelle metoder ut fra syren, f.eks. ved omsetning med det passende diazoalkan eller ved omsetning av en alkohol eller fenol med et aktivert derivat av syren, eventuelt under anvendelse av et kondenseringsmiddel, såsom dicyclohexylcarbodiimid, ved omsetning av et salt med et egnet aktivt alkyleringsmiddel, eller ved omforestring av en foreliggende ester. Spesifikke fremstillinger er beskrevet i redegjørelsene og eksemplene nedenfor.
Med betegnelsen "cycloalkyl" menes et mettet mono-syklisk hydrocarbonradikal med 4-8 carbonatomer, såsom cyclobutyl, cyclopentyl, cyclohexyl, cycloheptyl og cyclo-octyl.
Betegnelsen "lavere alkyl" refererer seg til en forgrenet eller uforgrenet mettet hydrocarbonkjede med 1-4 carbonatomer, såsom f.eks. methyl, ethyl, n-propyl, i-butyl o.1.
Betegnelsen "behandling" er ment å skulle dekke enhver behandling av en sykdom hos mennesker og pattedyr, spesielt mennesker, og innbefatter: (i) å hindre sykdommen i å inntre hos en pasient som kan være predisponert for sykdommen, men som ennu ikke har fått stillet diagnosen, (ii) å hemme sykdommen, dvs. å hindre dens utvikling, eller (iii) å lindre sykdommen, dvs. å legge en demper på sykdommen eller holde den i sjakk.
Det nununereringssystem for bicyclo-[4.2.0]-octan-systemet som er vist i illustrasjonen nedenfor, benyttes ved benevningen av mellomproduktene og produktforbindelsene ifølge oppfinnelsen.
Den absolutte stereokjemi av carbonatomene 1,2,3 og 6 og 3' i sidekjeden bundet til carbonatomet i 2-stillingen er spesifisert i henhold til Cahn-Ingold-Prelog-R-S-systemet. Når forbindelsen er en ren enantiomer, er stereokjemien av hvert chiral-carbonatom spesifisert ved enten R eller S. Når en forbindelse er en racemisk blanding, spesifiseres stereokjemien for hvert chiral-carbonatom ved enten R<*> eller S<*>, ved henvisning til én enkelt enantiomer av racematet. På denne måte kan den relative stereokjemi angis utvetydig, og den racemiske modifikasjon angis ved stjernemerket "<*>".
Når substituenten ved carbonatomet i 8-stilling står i cis-stilling til protonene i ringforbindelsespunktene (ved carbonatomer 1 og 6), angis forbindelsen som en exo-isomer. Når substituenten ved carbonatomet i 8-stilling står i trans-stilling til protonene i ringforbindelsespunktene, angis forbindelsen som en endo-isomer.
En alternativ metode til å angi stereokjemien i en racemisk blanding går ut på å angi hvert chiral-carbon som RS eller SR under henvisning til én enkelt enantiomer. Således kan en ny forbindelse som betegnes (Z)-(3'S<*>,IS<*>,2S<*>,3R<*>,6S<*>)-4-[2-(3'-hydroxy-3'-cyclohexyl-prop-1'-ynyl)-3-hydroxy-8-exo-methylbicyclo-[4.2.0]-oct-7-yliden-smørsyre, med like stor nøyaktighet betegnes ( Z)-(3'SR,1SR,2SR,3RS,6SR)-4-[2-(3'-hydroxy-3'-cyclohexyl-prop-1'-ynyl)-3-hydroxy-8-exo-methylbicyclo-[4.2.0]-oct-7-yliden-smørsyre, mens en ny forbindelse som betegnes som (Z)-(3'S<*>,IS<*>,2S<*>,3R<*>,6S<*>)-4-[2-(3<1->hydroxy-3'-cyclopentyl-prop-1'-ynyl)-3-hydroxy-8-endo-methylbicyclo-[4.2.0]-oct-7-yliden-smørsyre, alternativt kan betegnes som (Z)-(3'SR,1SR,2SR,3RS,6SR)-4-[2-(3 *-hydroxy-3'-cyclopentyl-prop-1'-ynyl)-3-hydroxy-8-endo-methylbicyclo-[4.2.0]-oct-7-yliden-smørsyre.
Olefin-stereokjemien angis etter IUPAC-E-Z-systemet. I henhold til klassisk nomenklatur benevnes en forbindelse med en trippelbinding som alkynyl, mens betegnelsen -yliden benyttes når to bindinger går ut fra samme atom.

For å unngå enhver mis-forståelse skal det understrekes at denne skrivemåte er ment å skulle angi at gruppene CH og (CH2>b er bundet direkte til hverandre med to kovalente bindinger under dannelse av en cycloalkylgruppe med b+1 carbonatomer. Firkantparentesene er ikke ment å skulle angi eventuelle carbonatomer mellom gruppene CH og (CH2)t>. Formelen ville også kunne skrives på en slik måte at gruppene CH og (CH2)b forbindes med hverandre med buede linjer,

Foretrukne utførelsesformer av oppfinnelsen
En foretrukken underklasse av forbindelser ifølge oppfinnelsen innbefatter forbindelser med formler (1), (2) og (3), hvor n er 2. Fortrinnsvis er b 4 eller 5, aller helst 5. En helt spesielt foretrukken undergruppe utgjøres av de forbindelser som representeres med formel (1) hvor R3 er cyclohexyl. Det som ovenfor er sagt om de foretrukne utførelsesformer av oppfinnelsen, innbefatter de i farma-søytisk henseende aksepterbare salter og estere og likeledes de frie baser av de forbindelser som er omtalt ovenfor eller nevnt nedenfor.

FREMSTILLINGSMETODER
Forbindelsene med formler (1), (2) og (3) kan fremstilles i henhold til reaksjonsskjerna I. I den detaljerte beskrivelse svarer tallangivelsene (romertall) i parentes til forbindelsene vist i reaksjonsskjemaet. Det er bare fremstillingen av forbindelsene med formler (1) og (2) fra forbindelser med formel (XIV) og fremstillingen av forbindelser med formel (3) fra forbindelser med formel (XV) som foretas i henhold til analogifremgangsmåten ifølge oppfinnelsen. De øvrige, forutgående reaksjoner leder frem til de utgangsmaterialer som anvendes ved analogifremgangsmåten ifølge oppfinnelsen.
Syntesen av forbindelsene med formler (1), (2) og (3) begynner med omsetning av 1,3-cyclohexadien (I) med den passende forbindelse med formelen RoCHClC0Cl, hvor RQ er lavere alkyl, hvilke forbindelser er lett tilgjengelige i handelen. Eksempelvis gir omsetning av (I) med 2-klor-propionylklorid en blanding av 8-endo-klor-8-exo-methylbicyclo-[4.2.0]-oct-2-en-7-on (IIA) og 8-exo-klor-8-endo-methylbicyclo-[4.2.0]-oct-2-en-7-on (IIB) som isoleres og skilles fra hverandre på i og for seg konvensjonell måte.
8-exo-klor-8-endo-laverealkylbicyclo-[4.2.0]-oct-2-en-7-on (IIB) omsettes så med et dekloreringsmiddel, såsom zink-kobber-paret eller tri-n-butyltinnhydrid, hvorved det fåes et 8-endo-(lavere alkyl)-bicyclo-[4,2.0]-oct-2-en-7-on (III). Eksempelvis vil 8-exo-klor-8-endo-methylbicyclo-
[4,2.0]-oct-2-en-7-on gi 8-endo-methylbicyclo-[4.2.0]-oct-2-en-7-on.
Forbindelsen med formel (III) hvor Y er endo-(lavere alkyl), omsettes så med 1,2-bis-(trimethylsilyloxy)-ethan i nærvær av trimethylsilyltrifluormethansulfonat som en katalysator. Når for eksempel Y er endo-methyl og reaksjonen utføres ved en temperatur mellom -60°C og -80°C, fåes ketalet med formel (IVB), nemlig spiro-[8-endo-methylbicyclo- [4. 2.0] -oct-2-en-7, 2 ' - (1',3'-dioxolan)]. Dersom reaksjonen utføres ved en temperatur på 10-30°C, fåes ketalet med formel (IVA), nemlig spiro-[8-exo-methylbicyclo-[4.2.0]-oct-2-en-7,2'-(l',3'-dioxolan)].
Det ønskede ketal omsettes så med N-bromacetamid og vann i aceton, hvorved det fåes en blanding av brom-hydriner (V), som overføres til en blanding av epoxyacetaler (VI) og (VII) ved behandling med kaliumcarbonat.
De bisykliske epoxyacetaler (VI) og (VII) kan omsettes med et organolithiumreagens av den generelle struktur Li-R' (XXI) i nærvær av bortrifluoridetherat, hvorved man får de diastereomere alkoholer, (VIII) og (IX), hvor
hvor R3 er som ovenfor angitt og Pt er en
beskyttende gruppe for sidekjede-hydroxylfunksjonen.
Fremstillingen av organolithiumreagenser med formel XXI beskrives i forbindelse med det nedenstående Reaksjonsskjerna II. Egnede beskyttende grupper innbefatter, men er ikke begrenset til, silylethere, tertiære alkyl-ethere, f.eks. tert-butyl, eventuelt substituerte trifenyl-methylethere, acetaler, såsom tetrahydropyranylethere, og lignende. Vanligvis foretrekkes trialkylsilylethere, og spesielt foretrukne er tert-butyldimethylsilylderivatene. Disse ethere fremstilles ut fra de tilsvarende carbinoler ved hjelp av standardmetoder som vil være velkjente for en fagmann på området. Spesielt har tert-butyldimethylsilyl-etherne funnet utstrakt anvendelse i prostaglandinkjemien, og de lar seg lett fremstille ved omsetning av den passende carbinol med tert-butyldimethylsilylklorid i N,N-dimethyl-formamidoppløsning i nærvær av imidazol, som både tjener som spesifikk katalysator for silyleringen og som base for nøytralisering av den saltsyre som utgjør det annet reaksj onsprodukt.
Organolithiumreagenser av alkynyltypen fremstilles ut fra de tilsvarende l-alkyn-3-oler, som i sin tur lett kan fremstilles ved omsetning av et acetylenisk Grignard-reagens, nemlig ethynylmagnesiumhalogenid, eller lithiumacetylid, med aldehyder eller ketoner (1*21*30=0), hvorved det fåes
ethynylcarbinoler
(Fremstillingen av 1-alkyn-3-oler er beskrevet nærmere under Fremstilling 5.) Ved beskyttelse av hydroxylgruppen med en egnet beskyttende gruppe, slik som angitt ovenfor, gir de tilsvarende ethere Overføring til et organolithiumreagens kan så foretas, vanligvis ved en utvekslingsreaksjon med en støkiometrisk mengde av en mer reaktiv organolithiumforbindelse, f.eks. et alkyllithiumreagens R'Li, slik at man får det acetyleniske lithiumreagens

IX - (3'S,lR,2R,3S,6R)-spiro-[2-(3'-tert-butyl-dimethylsilyloxy)-3'-cyclohexylprop-1'-ynyl)-3-hydroxy-8-methylbicyclo-[4.2.0]-octan-7,2''-(1'',3''-dioxolan)].
Blandingen av diastéreoisomere 8-(lavere alkyl)-bicyclo-[4.2.0]-octanoler VIII og IX overføres til en blanding av dikobolthexacarbonylkompleksene X og XI ved omsetning med dikoboltoctacarbonyl i diethylether. Disse komplekser separeres kromatografisk i de separate komplekser X og XI, som hvert omsettes med ceriumammoniumnitrat for dannelse av forbindelsene med formler XII og XIII. Når for eksempel Y er methyl, fåes henholdsvis (3'S,IS,2S,3R,6S)-spiro-[2-(3'-tert-butyldimethylsilyloxy-3'-cyclohexylprop-1'-ynyl)-3-hydroxy-8-methylbicyclo-[4.2.0]-octan-7,2''-(1'',3''-dioxolan)] (XII) og (3'S,IR,2R,3S, 6R)-spiro-[2-(3'-tert-butyldimethylsilyloxy-3'-cyclohexylprop-1'-ynyl)-3-hydroxy-8-methylbicyclo-[4.2.0]-octan-7,2''-(1'',3''-dioxolan)] (XIII). Hver av disse omsettes med fortynnet svovelsyre for å oppnå ketalhydrolyse og likeledes hydrolyse av den beskyttende silylgruppe, hvorved forbindelsene med formler XIV og XV dannes. Når for eksempel Y er methyl, fåes henholdsvis (3'S,IS,2S,3R,6S)-2-(3'-hydroxy-3'-cyclohexyl-prop-1 '-ynyl)-3-hydroxy-8-methylbicyclo-[4.2.0]-octan-7-on (XIV) og (3'S,lR,2R,3S,6R)-2-(3'-hydroxy-3'-cyclohexylprop-1'-ynyl)-3-hydroxy-8-methylbicyclo-[4.2.0]-octan-7-on (XV).
Bicyclooctanonalkynolen med formel XIV omsettes så med anionet (eller ylidet) av en fosfoniumforbindelse med den generelle formel:
hvor X, Y og Z er alifatiske eller aromatiske grupper, og n er 2 eller 3, eller et beskyttet derivat eller salt derav, såsom trifenylfosforanylid med en carbonkjedelengde på 4-5 carbonatomer, i et aprotisk oppløsningsmiddel, fortrinnsvis dimethylsulfoxyd eller en blanding av dimethylsulfoxyd og tetrahydrofuran, ved en temperatur mellom 10 og 60°C i 1-24 timer, for dannelse av en blanding av Z- og E-alkansyrene (1) og (2), for eksempel, når Y er endo-methyl, Z- og E-(3'S,lS,2S,3R,6S)-2-(3<*->hydroxy-3'-cyclohexylprop-1' -ynyl)-3-hydroxy-8-endo-methylbicyclo-[4.2.0]-oct-7-ylidenalkansyrer. Denne blanding separeres kromatografisk, hvorved man får f.eks. Z-(3'S,IS,2S,3R,6S)-2-(3'-hydroxy-3'-cyclohexylprop-1'-ynyl)-3-hydroxy-8-endo-methylbicyclo-[4.2.0]-oct-7-ylidenalkansyre (1) og E-(3'S,IS,2S,3R,6S)-2-(3'-hydroxy-3'-hydroxy-3'-cyclohexylprop-1'-ynyl)-3-hydroxy-8-methylbicyclo-[4.2.0]-oct-7-ylidenalkansyre (2). På tilsvarende måte omsettes bicyclooctanonalkynol med formel XV med anionet (eller ylidet) av en fosfoniumforbindelse med den generelle formel:

Z-(3'S,IR,2R,3S,6R)-2-(3'-hydroxy-3'-cyclohexylprop-1-ynyl)-3-hydroxy-8-methylbicyclo-[4.2.0]-oct-7-ylidenalkansyre (3) og E- (3'S,IR,2R,3S,6R)-2-(3'-hydroxy-3'-cyclohexylprop-1'-ynyl)-3-hydroxy-8-methylbicyclo-[4.2.0]-oct-7-ylidenalkan-syre XVI.
For fremstilling av trifenylfosforanylidene som benyttes i Wittigreaksjonene som overfører XIV og XV til (1), (2) og (3), blir trifenylfosfoniumsaltene deprotonert med 2 ekvivalenter dimsylnatrium i dimethylsulfoxyd. Trifenylfosfoniumsaltene fremstilles ved kondensasjon av tri-fenylfosfin med en fl-halogencarboxylsyre som beskrevet i J. Org. Chem. 27, 3404 (1962). De urene fosfoniumsalter renses vanligvis ved vaskning med diethylether eller ved rekrystal-lisering fra acetonitril eller ethanol-diethylether.
De her beskrevne strukturer, inklusive de nye forbindelser som fremstilles i henhold til oppfinnelsen, har flere chiralsentere og er optisk aktive. Selv om det i illustrasjonsøyemed bare er beskrevet én optisk isomer, omfatter oppfinnelsen fremstilling av alle optiske isomerer og blandinger derav, idet blandingene innbefatter racemater og diastereomere blandinger i alle mengdeforhold. Dersom de nye forbindelser fremstilles ut fra optisk inaktive utgangsmaterialer og uten bruk av chiral-reagenser, vil produktene fåes som (optisk inaktive) racemiske blandinger.
De optisk aktive acetylener som benyttes i Reaksjonsskjerna I, fremstilles i henhold til Reaksjonsskjerna
II.
Som vist i Reaksjonsskjerna II blir en propargylalkohol med formel XVII, oxydert med Jones-reagens for dannelse av et propargylketon med formel XVIII. Dette keton reduseres med isopinokamfenyl-9-borabicyclo-[3.3.1]-nonan i henhold til den metode som er beskrevet i J. Amer. Chem. Soc., hvorved man får chiral-propargylalkoholene (XIX). Propargylalkoholene XIX overføres til 0-beskyttede derivater XX ved kondensasjon med trialkylsilylklorider og imidazol. Forbindelser XX omsettes med butyllithium for dannelse av lithiumacetylider XXI som anvendes i Reaksjonsskjerna I for å overføre epoxydet VI til VIII og IX.
De i Reaksjonsskjerna I benyttede optisk aktive acetylener kan også fremstilles i henhold til Reaksjons-skjema III. Reaksjonsskjerna III viser metoden ifølge Fried (Ann. N.Y. Acad. Sei., 180, 39 (1971)). Ved denne metode blir et hemifthalat av racemisk XVII, som dannes ved kondensasjon av XVII med fthalsyreanhydrid, overført til en blanding av diastéreoisomere salter XXII og XXIII under anvendelse av et egnet optisk aktivt amin. Blandingen av salter omkrystalliseres fra et passende oppløsningsmiddel, vanligvis acetonitril, for å danne et rent, diastereoisomert salt XXII. Dette salt behandles med fortynnet vandig saltsyre for å danne hemifthalat XXIV, som hydrolyseres med vandig base for dannelse av chiral-acetylenisk alkohol XXV.
Den foranstående redegjørelse av Reaksjonsskjemaer I, II og III beskriver delvis en fremgangsmåte for synteti-sering og separering av chiral-mellomproduktene med formel XII og XIII erholdt fra omsetningen av blandingen av epoxyder VI og VII med et chiral-lithiumacetylid med formel XXI. En alternativ metode for fremstilling av chiral-mellomproduktene ifølge Reaksjonsskjerna I under anvendelse av racemiske mellomprodukter med formel XXI er vist i Reaksjonsskjerna IV.
Utgangspunktet for Reaksjonsskjerna IV er en racemisk blanding angitt som d,l XII, som er en blanding av isomerene vist ved formler XII og XIIA. Blandingen d,l XII fåes ved omsetning av blandingen av epoxyder VI og VII med 2t racemisk lithiumacetylid med formel XXI. Reaksjonen gir Dgså den tilsvarende racemiske blanding d,l XIII, som ut-jjøres av forbindelsen med formel XIII og dennes enantiomer. 3e to diastereoisomerer d,l XII og d,l XIII skilles fra werandre ved hjelp av koboltprosedyren, som vist i Reaksjonsskjerna I ovenfor. Ved kondensasjon av d,l XII med
(R)-(-)-a-(1-nafthyl)-ethylisocyanat fåes en blanding av
diastereoisomerer (XXVI) og (XXVII). Disse skilles fra hverandre ved kromatografering, hvorved man får de individuelle diastereoisomerer (XXVI) og (XXVII). Forbindelser (XXVI) og (XXVII) omsettes hver for seg med lithiumaluminiumhydrid, hvorved man får de respektive enantiomerer (XIIA) og (XII).
Ved at man følger den ovenfor beskrevne prosedyre skilles på tilsvarende måte forbindelsen med formel (XIII) fra sin enantiomer.
De i farmasøytisk henseende aksepterbare ikke-toksiske salter av forbindelsene med formler (1), (2) og (3) fremstilles ved at de frie syrer behandles med en passende mengde av en i farmasøytisk henseende aksepterbar base. Representative baser som er aksepterbare i farmasøytisk henseende, er natriumhydroxyd, kaliumhydroxyd, lithium-hydroxyd, ammoniumhydroxyd, calciumhydroxyd, magnesium-hydroxyd, ferrohydroxyd, zinkhydroxyd, kobberhydroxyd, mangan(II)hydroxyd, aluminiumhydroxyd, ferrihydroxyd, mangan(III)hydroxyd, isopropylamin, trimethylamin, diethyl-amin, triethylamin, tripropylamin, ethanolamin, 2-dimethyl-aminoethanol, 2-diethylaminoethanol, lysin, arginin, histidin, procain, cholin, betain, ethylendiamin, glucos-amin, methylglucamin, theobromin, puriner, piperazin, piperidin, N-ethylpiperidin, polyaminharpikser o.l. For fremstilling av f.eks. énverdige kationsalter behandles den frie syre med formel (1), (2) eller (3) med en molekvivalent base i et egnet oppløsningsmiddel, såsom vann, methanol, ethanol, isopropanol, butanol, aceton, dioxan etter tetrahydrofuran. Molforholdet mellom forbindelsen med formel (1), (2) eller (3) og basen velges slik at det ønskede molforhold oppnås for det valgte salt. For fremstilling av f.eks. salter med toverdige kationer, såsom calcium- eller magnesiumsaltene, blir den som utgangsmateriale benyttede frie syre med formel (1), (2) eller (3) behandlet med minst 0,5 molekvivalent base for å danne et nøytralt salt. På tilsvarende måte vil man for salter med treverdig aluminium benytte minst 1/3 molekvivalent av aluminiumbasen, dersom det ønskes fremstilt et nøytralt salt.
De nye, frie carboxylsyrer (1), (2) og (3) som fremstilles i henhold til oppfinnelsen, kan på ny frigjøres fra deres respektive salter ved behandling av saltene med minst støkiometriske mengder av en sterk syre, fortrinnsvis en uorganisk syre, f.eks. saltsyre, svovelsyre o.l., ved temperaturer i området fra 0°C til 50°C, fortrinnsvis ved romtemperatur.
De i farmasøytisk henseende aksepterbare ikke-toksiske estere av de nye syrer (1), (2) og (3) som fremstilles i henhold til oppfinnelsen, kan fremstilles f.eks. ved forestring av de tilsvarende frie syrer med en opp-løsning av det passende diazoalkan i et egnet inert oppløsningsmiddel, såsom diethylether. En alternativ og generell metode for fremstilling av estrene av forbindelsene som fremstilles i henhold til oppfinnelsen, omfatter reaksjon mellom en benzenoppløsning av carboxylsyren og et alkylhalogenid i nærvær av den organiske base diazabicyclo-undecan (DBU) ved temperaturer fra 20 til 80°C i 1-12 timer. Disse betingelser er særlig anvendelige for forestring av syrer som oppviser labil funksjonalitet i molekylet, såsom prostaglandinene og deres syntetiske analoger, da de ikke omfatter bruk av syrekatalysatorer eller sterke reagenser.

Esterne kan også fremstilles under milde betingelser ved omsetning av de nye syrer (1), (2) eller (3) med en alkohol eller fenol i nærvær av et dehydratiseringsmiddel, f.eks. dicyclohexylcarbodiimid. Reaksjonen innebærer omsetning av ekvivalente mengder av syren og alkoholen eller fenolen i et egnet oppløsningsmiddel, f.eks. en blanding av methylenklorid og dimethylformamid, i nærvær av en katalyt-isk mengde 4-dimethylaminopyridin, og en ekvimolar mengde dicyclohexylcarbodiimid. Reaksjonen utføres ved en temperatur fra -10°C til 25°C, fortrinnsvis ved en temperatur på ca. 0°C, i 8-48 timer, fortrinnsvis i ca. 16 timer.
Typiske estere er de estere som avledes fra methylalkohol, ethylalkohol, propylalkohol, isopropyl-alkohol, butylalkohol, 2-butylalkohol, 2-pentylalkohol,
isopentylalkohol, 2-hexylalkohol o.l.
Alternativt kan alkylestrene fremstilles ved omforestring, katalysert av det tilsvarende alkoxyd, i henhold til i faget kjente metoder. Ved fremstilling av estrene ved omforestring foretrekkes det å gå fra en lavere ester til en høyere ester, f.eks. fra methylesteren til isoamylesteren. Ved bruk av et vesentlig overskudd av en lavere alkohol kan imidlertid en høyere ester omforestres til en lavere ester. Således kan man f.eks. ved å benytte et vesentlig overskudd av ethanol overføre hexylesteren til ethylesteren ved omforestring.
Salter av forbindelsene med formler (1), (2) og (3) kan utveksles ved at man utnytter ulikheter i oppløse-ligheten av saltene eller av flyktigheten eller aktiviteten av syrene, eller ved behandling med en passende ladet ione-bytteharpiks. Eksempelvis kan utvekslingen foretas ved at et salt av forbindelsene med formler (1), (2) og (3) omsettes med et svakt støkiometrisk overskudd av en syre med lavere pKa enn syrebestanddelen i det som utgangsmateriale benyttede salt. Denne omdannelse utføres ved en temperatur mellom 0°C og kokepunktet for det oppløsningsmiddel som benyttes som medium ved prosedyren.
De nye forbindelser er anvendelige for behandling av kardiovaskulære forstyrrelser. Spesielt er de virksomme som karutvidende midler, og de hemmer akkumuleringen av kolesterol i karveggen og i plasma. De er også kraftige hemmere av aggregeringen av blodplater og av frigjøringen fra disse av pro-coagulantfaktorer og pro-atherosclerotiske faktorer. Følgelig er disse forbindelser nyttige med hensyn til å behandle og forhindre kardiovaskulære forstyrrelser som involverer atherosclerosis, thrombotiske og vaso-spastiske betingelser. De er også nyttige antihypertensive midler og kolesterolsenkende midler.
De nye forbindelser oppviser det spektrum av akti-viteter som forbindes med prostacyclin. I motsetning til prostacyclin, hvis terapeutiske potensiale forringes alvor-lig som følge av dets ekstremt store kjemiske ustabilitet, har imidlertid de nye forbindelser som fremstilles i henhold til oppfinnelsen, en høy biologisk aktivitet, samtidig som de har en langt bedre kjemisk stabilitet. Denne kombinasjon av egenskaper peker dem ut som lovende midler for fore-byggende og/eller terapeutisk anvendelse, spesielt for behandling av kardiovaskulære forstyrrelser og sykdommer. Mange av disse forbindelser er selektive i sin antithrom-botiske virkning, og de utøver denne terapeutiske virkning uten i vesentlig grad å påvirke blodtrykket.
De nye forbindelser er også nyttige ved at de inhiberer utsondringen av magesyre.
De nye forbindelser er blitt testet med hensyn til deres hemmende virkning på blodplateaggregering, antihypertensive virkning og toksisitet.
Hemming av blodplateaggregering
Menneskelig veneblod fra drogefrie, friske fri-villige ble oppsamlet i 15 ml's vakuumbeholdere og anti-koagulert med 0,5 ml 11,4% natriumcitrat. Blodet ble sentri-fugert ved romtemperatur i 15 minutter ved 150 G i en Sorvall "GLC-28" sentrifuge, og topplaget bestående av blod-platerikt plasma (PRP) ble frasuget. Blodplate-fattig plasma (PPP) ble dannet ved sentrifugering av blodet fra hvilket PRP var blitt fjernet ved 12800 G i tre minutter i en Eppendorf-sentrifuge ved romtemperatur. Blodplateaggre-geringen ble foretatt etter metoden ifølge Born (J. Physio-logy, 168, 178 (1963)) i Payton aggregometere. Blodplate-aggregeringen ble innledet ved tilsetning av 2-5 nmol ADP til 1 ml PRP inneholdende 10 ul bærer inneholdende testforbindelser i en rekke forskjellige konsentrasjoner (eller ingen forbindelse) inkubert i aggregometerkuvetten ved 37°C i fem minutter med omrøringshastigheten innstilt på 500 rpm. Det ble så for hver testforbindelse trukket opp på halv-logaritmisk papir en kurve som viste den prosentvise hemning som funksjon av konsentrasjonen, og konsentrasjonen som svarte til 50% hemning ble uttrykt som IC50 for den angjeldende forbindelse. Samtlige testforbindelser (1-2 mg) ble blandet ut som 0,01M forrådsoppløsninger i 10% ethanol og 59 mM Na2C03. Påfølgende fortynninger ble foretatt med vann. Forsøksresultatene er oppført 1 den nedenstående tabell.
Antlhypertensiv aktivitet
De antihypertensive virkninger av de prosta-glandinlignende forbindelser ble bedømt på spontant hyper-tensive rotter (SHR/NCrlBR). Under etherbedøvelse ble lår-arterie- og lårvenekanyler innplantet, og rottene ble holdt ryggleie. Etter at rottene hadde kommet seg av bedøvelsen ble lidocain administrert. Blodtrykket ble registrert via lårarteriekanylen og skrevet ut på en Beckman "R611" polygraf. Grupper å fire rotter ble undersøkt for hver forbindelse. Bærer ble administrert ved forsøkets innled-ning, og en forbindelse ble deretter administrert intra-venøst med 30 minutters intervaller, i økende doser på 1, 3, 10, 30 og 100 ug/kg. Det midlere basislinje-arterieblodtrykk var blodtrykket umiddelbart før administreringen av den første dose av forbindelsen. ED2o-ver<3ier ble beregnet ut fra en lineær regresjon av den prosentvise minskning av det midlere blodtrykk som fulgte etter hver dose av forbindelsen.
Resultater
Toksisitet
Marsvin som veiet ca. 350-550 g, ble gitt 1 mg/kg av testforbindelsen (Z)-(3'S,IR,2R,3S,6R)-4-[2-(3'-hydroxy-3'-cyclohexylprop-1'-ynyl)-3-hydroxy-8-endo-methylbicyclo-[4.2.0]-oct-7-yliden]-smørsyre i form av en oppløsning én gang oralt. Det ble ikke iakttatt noen toksiske virkninger.
Heller ikke andre av de nye forbindelser som fremstilles i henhold til oppfinnelsen, har vist seg å oppvise toksiske virkninger.
Administrering av de aktive forbindelser i farma-søytiske preparater kan foretas ved hjelp av hvilke som helst av de aksepterte metoder for administrering av midler som påvirker det kardiovaskulære system. Disse metoder innbefatter oral, parenteral, topisk og annen systemisk administrering. Avhengig av metoden som benyttes, kan preparatet foreligge i fast, halvfast eller væskeformig doseringsform, f.eks. i form av tabletter, stikkpiller, piller, kapsler, pulvere, væsker, suspensjoner o.l., fortrinnsvis i enhets-doseformer egnet for administrering av presise doser.
Mengden av aktiv bestanddel som administreres, vil selvfølgelig være avhengig av pasienten som behandles, syk-domstilstanden, administreringsmetoden og legens bedømmelse av situasjonen. Imidlertid vil en effektiv dose være i området fra 0,001 til 15 mg/kg pr. dag, fortrinnsvis fra 0,01 til 3 mg/kg pr. dag. For et gjennomsnittsmenneske av vekt 70 kg vil dette svare til 0,07-1000 mg pr. dag, fortrinnsvis 0,7-210 mg pr. dag.
De følgende Fremstillinger illustrerer fremstil-lingstrinn som kan benyttes for fremstilling av de utgangsmaterialer som anvendes ved analogifremgangsmåten ifølge oppfinnelsen.

Fremstilling av 8-endo-klor-8-exo-methylbicyclo-[4.2.0]-oct-2-en-7-on og 8-exo-klor-8-endo-methyl-[4.2.0]-oct-2-en-7-on og beslektede forbindelser med formler IIA og IIB.
En blanding av 34,0 g cyclohexadien og 39,7 g 2-klorpropionylklorid i 300 ml diethylether ble under nitrogenatmosfære kokt med tilbakeløpskjøling, samtidig som det dråpevis og i løpet av tre timer ble tilsatt 31,5 g triethylamin i 300 ml diethylether. Blandingen ble så omrørt ved romtemperatur i 20 timer og filtrert. Filtratet ble vasket med saltoppløsning, IN saltsyre og mettet natriumbicarbonat, hvoretter den ble tørret over vannfritt natriumsulfat. Oppløsningsmidlet ble fjernet under redusert trykk, og residuet ble destillert, hvorved man fikk 32 g av en blanding av 8-exo-methyl- og 8-endo-methyl-isomerene. Isomerene ble skilt fra hverandre ved kromatografering på silicagel under eluering med hexan-methylenklorid (1:1), hvorved man fikk 22 g 8-éxo-klor-8-endo-methylbicyclo-[4.2.0]-oct-2-en-7-on og 6,4 g 8-endo-klor-8-exo-methylbicyclo-[4.2.0]-oct-2-en-7-ori.

Fremstilling av 8-endo-methylbicyclo-[4.2.0]-oct-2-en-7-on og beslektede forbindelser med formel IIIB
Til en blanding av 40 g 8-exo-klor-8-endo-methylbicyclo-[4.2.0]-oct-2-en-7-on, fremstilt som beskrevet i Fremstilling 1, og 60 g ammoniumklorid i 250 ml ethanol, som forelå ved romtemperatur, ble det porsjonsvis tilsatt 40 g av paret sink-kobber. Reaksjonsblandingen ble omrørt ved romtemperatur i tre timer og deretter fortynnet med diethylether og filtrert. Filtratet ble vasket med saltoppløsning og tørret over natriumsulfat, hvoretter oppløsningsmidlet ble fjernet under redusert trykk. Residuet ble renset ved kromatografering på silicagel under eluering med hexan-methylenklorid (1:1), hvorved man fikk 25,9 g 8-endo-methylbicyclo-[4.2.0]-oct-2-en-7-on.

Fremstilling av spiro-[8-endo-methylbicyclo-[4.2.0]-oct-2-en-7,2<1->(1',3'-dioxolan)] og beslektede forbindelser med formel IV
En blanding av 680 mg 8-endo-methylbicyclo-[4.2.0]-oct-2-en-7-on, fremstilt som beskrevet i Fremstilling 2, og 2,1 g 1,2-bis-(trimethylsilyloxy)-ethan ble kjølt til -78°C under nitrogen, og 40 ul trimethylsilyltrifluormethansulfonat ble tilsatt i fire porsjoner å 10 ul. Blandingen ble omrørt i tilsammen 16 timer ved -78°C, hvoretter reaksjonen ble stoppet ved tilsetning av overskudd av pyridin. Reaksjonsblandingen ble hellet over i en vandig, mettet natriumbicarbonatoppløsning og ekstrahert med ether. Oppløsningsmidlet ble fjernet fra ekstrakten under redusert trykk, og residuet ble kromatografert på silicagel, under eluering med 5% aceton i hexan, hvorved man fikk 800 ml spiro-[8-endo-methylbicyclo-[4.2.0]-oct-2-en-7, 2' - (1' , 3 ' - dioxolan)].

Fremstilling av (IS<*>,2S<*>,4R<*>,7S<*>)-spiro-[3-oxa-9-endo-methyltricyclo-[5.2.0.02'4]-nonan-8,2'- (1',3'-dioxolan)], (IS<*>,2R<*>,4S<*>,7S<*>)-spiro-[3-oxa-9-endo-methyltricyclo-[5.2.0.0<2>'<4>]-nonan-8,2'-(1',3'-dioxolan)] og beslektede forbindelser med formler VI og VII
Til en omrørt oppløsning av 5 g spiro-[8-endo-methylbicyclo- [4 . 2 . 0] -oct-2-en-7, 2 '-( 1 ' ,3'-dioxolan)], fremstilt i henhold til Fremstilling 3, i 40 ml aceton og 20 ml vann ved 0°C settes 4,76 g N-bromacetamid i løpet av én time. Denne blanding omrøres ved romtemperatur i 20 timer. Til oppløsningen settes 12,4 g kaliumcarbonat, og den resulterende blanding omrøres ved romtemperatur i to dager. Blandingen mettes med natriumklorid, og den resulterende blanding ekstraheres med fire 150 ml's porsjoner diethylether. De sammenslåtte organiske ekstrakter vaskes med 100 ml mettet natriumkloridoppløsning og tørres over natriumsulfat. Ved fjerning av oppløsningsmidlet i vakuum og rensning ved kromatografering av residuet på silicagel under eluering med 25% ethylacetat-hexan fåes 3,45 g av en blanding av (IS<*>,2S<*>,4R<*>,7S<*>)-spiro-[3-oxa-9-endo-methyl-tricyclo-[5.2.0.0<2>'<4>]-nonan-8,2'-(1',3'-dioxolan)] (VI) og (1S<*>,2R<*>,4S<*>,7S<*>)-spiro-[3-oxa-9-endo-methyltricyclo-[5.2.0.0<2>'<4>]-nonan-8,2'-(1',3'-dioxolan)] (VII) i et forhold på ca. 9:1.

En hurtig strøm av acetylen ble ledet gjennom
100 ml av en 2M oppløsning av methylmagnesiumbromid i THF,
inntil det ikke lenger ble observert noen methanutvikling.
10 g hexanal ble tilsatt ved 0°C, hvoretter blandingen ble
omrørt i 30 minutter og det ble tilsatt en mettet oppløsning av NH4CI. Det organiske produkt ble isolert ved ekstraksjon med ether. Etheroppløsningen ble vasket med vann og saltopp-løsning, tørret over MgS04 og inndampet til en væske som ble renset ved destillasjon, hvorved man fikk 3-hydroxyoct-l-yn.

Fremstilling av 3-cyclohexyl-l-propyn-3-on og beslektede forbindelser med formel XVIII
En oppløsning av kromsyre ble fremstilt ved opp-løsning av 106,88 g kromtrioxyd i 400 ml vann og påfølgende tilsetning av 92 ml konsentrert svovelsyre. Denne oppløsning dryppes i løpet av tre timer til en isavkjølt, omrørt opp-løsning av 120 g 3-cyclohexyl-l-propyn-3-ol i 175 ml aceton. Den resulterende blanding ble fortynnet med 500 ml vann, og produktet ble ekstrahert over ill diethylether. Etherekstrakten ble vasket med 250 ml mettet natriumbisulfitt-oppløsning og ble tørret over natriumsulfat. Diethyletheren ble fjernet ved destillasjon under nitrogenatmosfære, og det resulterende residuum ble renset ved Kugelrohr-destillasjon (65°C; 0,1 mm Hg), hvorved det ble erholdt 84,9 g 3-cyclohexyl-l-propyn-3-on i form av en olje. MS m/z = 136 (M+) Beregnet for CgH120 : C = 79,37; H = 8,88. Funnet: C = 79,24; H = 8,6.
Fremstilling 7
Fremstilling av (S)-3-cyclohexyl-l-propyn-3-ol og beslektede forbindelser med formel XIX
En blanding av 1,6 1 0,5 M 9-borabicyclo-[3.3.1]-nonan i tetrahydrofuran og 122,6 g (-)-a-pinen, av renhet 99+%, ble kokt med tilbakeløpskjøling under nitrogen i fire timer, på hvilket tidspunkt overskuddet av (-)-a-pinen og tetrahydrofuran ble fjernet under vakuum, slik at det ble tilbake en tykk olje. Kolbens innhold ble avkjølt til 0°C og 80 g 3-cyclohexyl-l-propyn-3-on, fremstilt i henhold til Fremstilling 6, ble tilsatt under omrøring. Den resulterende blanding ble tillatt å oppvarmes til 23°C, og den ble omrørt ved denne temperatur i 16 timer. Overskudd av S-alpin-boran ble ødelagt ved tilsetning av 44 ml propionaldehyd og omrøring ved 23°C i én time. Det frigjorte (-)-a-pinen ble fjernet ved vakuumdestillasjon. Den resulterende blanding ble fortynnet med 400 ml tetrahydrofuran, etterfulgt av 300 ml 3N natriumhydroxyd. Til denne omrørte blanding ble det tildryppet 300 ml 30% hydrogenperoxyd i løpet av én time. Blandingen ble oppvarmet ved 40°C i tre timer. Etter avkjøling ble blandingen ekstrahert med diethylether, og denne ekstrakt ble tørret over magnesiumsulfat. Ved avdampning av oppløsningsmidlet og rensning av residuet ved flash-kromatografering på silicagel under anvendelse av 5% ethylacetat-hexan ble det erholdt 56 g (S)-3-cyclohexyl-l-propyn-3-ol, som ved NMR-analyse viste seg å være 90% ren. Ved omkrystallisering fra hexan ble det erholdt 45 g av den rene S-isomer. Smeltepunkt 56-58°C. [a]<2>,<5> = -9,24° (C = 0,53, Et20).
Fremstilling 8
Fremstilling av (S)-3-cyclohexyl-l-propyn-3-ol og beslektede forbindelser med formler XIX og XXV
En blanding av 50 g racemisk 3-cyclohexyl-1-propyn-3-ol, fremstilt i henhold til Fremstilling 5, 53,3 g fthalsyreanhydrid og 100 ml pyridin oppvarmes ved 90°C i fire timer. Etter kjøling til °C settes denne blanding under omrøring til en blanding av 350 ml konsentrert saltsyre og 900 ml is. Det oljeaktige faste stoff som skilles ut, opp-løses i 600 ml diethylether. Denne oppløsning vaskes med mettet NaCl-oppløsning og tørres over natriumsulfat. Ved inndampning og omkrystallisering fra aceton/hexan fåes hemifthalatet, med smeltepunkt 136-138°C. 38,5 g av dette hemifthalat oppslemmes i 80 ml diklormethan, og en oppløsning av 16,2 g (-)-a-fenylethylamin i 250 ml diklormethan tilsettes under omrøring i løpet av 15 minutter. Blandingen filtreres etter én time, og filtratet inndampes, hvorved det fåes en blanding av diastéreoisomere salter. Denne blanding omkrystalliseres fem ganger fra acetonitril, hvorved det fåes 7 g av et rent diastereoisomert salt. Smeltepunkt 142-143°C.
[a]D = -36,7 (C = 1, CHCI3). 2 g av dette salt settes til en omrørt blanding av 25 ml 5% natriumbicarbonat og 25 ml diethylether. Etherskiktet hives, og det vandige skikt ekstraheres med tre ytterligere 25 ml's porsjoner diethylether. Det vandige skikt surgjøres med 4N HC1 og ekstraheres grundig med diethylether. Etherekstrakten tørres over natriumsulfat og inndampes til tørrhet, hvorved det fåes 1,37 g av hemifthalatet av (S)-3-cyclohexylprop-l-yn-3-ol. Smeltepunkt 70-74°C. [a]D = -35,8° (C = 1, CHCI3).
Hemifthalatet ble så omrørt med overskudd av vandig 2N kaliumhydroxyd i to timer ved 60°C, hvoretter blandingen ble kjølt til romtemperatur og ekstrahert med diethylether. Det organiske skikt ble fraskilt og tørret over natriumsulfat, hvoretter oppløsningsmidlet ble fjernet under redusert trykk. Residuet ble kromatografert på silicagel, under eluering med 10% ethylacetat i hexan, hvorved man fikk (S)-3-cyclohexylprop-l-yn-3-ol, sm.p. 56-58°C [a]<25> = -11,2° (C=0,6; Et20).

Fremstilling av (S)-3-tert-butyldimethylsilyloxyoct-l-yn og beslektede silylethere med formel XX
Til en oppløsning av 2,76 g (0,02 mol) (S)-3-cyclohexyl-l-propyn-3-ol (fremstilt i henhold til Fremstilling 7.A. og 8.A.) i 10 ml N,N-dimethylformamid (DMF), kjølt til 0°C, ble det satt 2,1 g imidazol og deretter 3,1 g (0,02 mol) tert-butyldimethylklorsilan. Blandingen ble omrørt i tre timer. 80 ml vann og 80 ml hexan ble tilsatt, og det organiske skikt ble fraskilt og slått sammen med 2 x 80 ml hexanekstrakter av det vandige skikt. Oppløsnings-midlet ble fjernet i vakuum etter tørring over natriumsulfat. Det ble erholdt 4,3 g av et urent residuum, som ble kromatografert på 80 g silicagel under eluering med ethylacetat-hexan i volumforholdet 2:1, hvorved man fikk 3-tert-butyldimethylsilyloxy-3-cyclohexylprop-l-yn.

(VIII) og (3'S,lR,2R,3S,6R)-spiro-[2-(3'-t-butyldimethyl-silyloxy-3'-cyclohexylprop-1'-ynyl)-3-hydroxy-8-endo-methylbicyclo-[4.2.0]-octan-7,2''-(1'',3<*>'-dioxolan)] (IX) og beslektede forbindelser med formler VIII og IX
Til en blanding av 6,45 g (S)-3-t-butyldimethyl-silyloxy-3-cyclohexyl-l-propyn fremstilt i henhold til Fremstilling 9, i 48 ml tetrahydrofuran ved 0°C og under argonatmosfære, ble det i løpet av 20 minutter satt 15,96 ml 1,37M n-butyllithium i hexan. Den resulterende oppløsning ble kjølt til -78°C, og en oppløsning av 3,6 g (IS<*>,2S<*>,4R,7S<*>)-spiro-[3-oxa-9-endo-methyltricyclo-[5.2.0.0.<2>'<4>]-nonan-8,2'-(l',3'-dioxolan i 18 ml tetrahydrofuran ble tilsatt. Til denne omrørte oppløsning ved -78°C ble det dryppet 2,2 ml bortrifluoridetherat i løpet av 25 minutter. Den resulterende blanding ble oppvarmet til romtemperatur, og den ble grundig ekstrahert med ethylacetat. Oppløsningen ble tørret over natriumsulfat og ble inndampet i vakuum, hvorved det ble erholdt et oljeaktig residuum, som ble renset ytterligere ved flash-kromatografering på silicagel under anvendelse av 2% aceton-diklormethan. Denne prosedyre ga 4,6 g av en blanding av de i overskriften angitte forbindelser i form av en olje.

Fremstilling av (3'S,IS,2S,3R,6S)-spiro-[2-(3'-t-butyl-dimethylsilyloxy-3'-cyclohexylprop-1'-ynyl)-3-hydroxy-8-endo-methylbicyclo-[4.2.0]-octan-7,2''-(1'',3''-dioxolan)] og (3'S,IR,2R,3S,6R)-spiro-[2-(3'-t-butyldimethylsilyloxy-3'-cyclohexylprop-1'-ynyl)-3-hydroxy-8-endo-methylbicyclo-[4.2.0]-octan-7,2''-(1'',3''-dioxolan)] og beslektede forbindelser med formler XII og XIII
Til en oppløsning av 4,5 g av den diastereomere blanding av de i overskriften angitte forbindelser, fremstilt som beskrevet i Fremstilling 10, i 200 ml diethylether, ble det satt 5,1 g dikoboltoctacarbonyl. Den resulterende oppløsning ble omrørt ved 23°C i to timer. Blandingen ble fortynnet med 200 ml diethylether, og filtrert. Filtratet ble konsentrert til en olje, som ble renset ved flash-kromatografering under anvendelse av 12% ethylacetat-hexan, hvorved man fikk to bestanddeler: A (høy Rf) og B (lav Rf). Bestanddel A (2,85 g) ble oppløst i
150 ml aceton-vann (9:1), hvortil det ble tilsatt 10,1 g ceriumammoniumnitrat. Etter to minutter ble denne blanding fortynnet med 200 ml vann. Produktet ble isolert ved ekstråksjon med diethylether. Etter tørring og inndampning ble det erholdt 1,5 g (3'S,IS,2S,3R,6S)-spiro-[2-(3'-t-butyldimethylsilyloxy-3'-cyclohexylprop-1'-ynyl)-3-hydroxy-8-endo-methylbicyclo-[4.2.0]-octan-7,2''-(1'',3''-dioxolan)] i form av en olje. På tilsvarende måte ble bestanddel B (3,26 mg) overført til (3'S,IR,2R,3S,6R)-spiro-[2-(3'-t-butyldimethylsilyloxy-3'-cyclohexylprop-1'-ynyl)-3-hydroxy-8-endo-methylbicyclo-[4.2.0]-octan-7,2''-(1'' ,3''-dioxolan)] som også ble erholdt i form av en olje.

En blanding av 2,25 g (3'S<*>,IS<*>,2S<*>,3R<*>,6S<*>)-spiro-[2-(3'-tert-butyldimethylsilyloxy-3'-cyclohexylprop-1'-ynyl)-3-hydroxy-8-endo-methylbicyclo-[4.2.0]-octan-7,2''-(1'',3''-dioxolan)], 20 ml ethyldiisopropylamin, 0,76 g 4-methylaminopyridin og 3,65 g (R)-(-)-a-(1-nafthyl)-ethyl-isocyanat omrøres ved 45°C i åtte timer. Ethyldiisopropyl-aminet fjernes ved vakuumfordampning, og residuet oppløses i 100 ml ethylacetat. Denne oppløsning vaskes med to 25 ml's porsjoner IN saltsyre og 25 ml vann. Etter tørring over natriumsulfat fjernes oppløsningsmidlet ved fordampning, hvorved det fåes tilbake et residuum som renses ytterligere ved flash-kromatografering på silicagel under anvendelse av ethylacetat-diklormethan-hexan (7,5:22,5:70). Til en omrørt blanding av 0,3 g lithiumaluminiumhydrid i 50 ml tetrahydrofuran ved 0°C settes en oppløsning av 1,25 g (3'S,IS,2S,3R,6S)-spiro-[2-(3'-tert-butyldimethyl-silyloxy-3 '-cyclohexylprop-1'-ynyl)-3-0-[(R)-a-(1-nafthyl) - ethylcarbamoyl]-8-endo-methylbicyclo-[4.2.0]-octan-7,2''-(1'',3''-dioxolan)] i 10 ml tetrahydrofuran. Denne blanding kokes med tilbakeløpskjøling i én time. Etter avkjøling til 23°C opparbeides blandingen ved dråpevis tilsetning av først 0,5 ml vann, deretter 0,5 ml 15% natriumhydroxydoppløsning og til slutt 1,5 ml vann. Det resulterende faste stoff fraskilles ved filtrering, og filtratet inndampes i vakuum. Residuet oppløses i 100 ml ethylacetat, og den erholdte oppløsning vaskes med to 20 ml's porsjoner vann og tørres deretter over natriumsulfat. Ved inndampning fåes et residuum som renses ved flash-kromatografering på silicagel under anvendelse av ethylacetat-hexan (15:85), hvorved man får (3'S,IS,2S,3R,6S)-spiro-[2-(3'-tert-butyldimethyl-silyloxy-3 '-cyclohexylprop-1'-ynyl)-3-hydroxy-8-endo-methylbicyclo-[4.2.0]-octan-7,2''-(1'',3''-dioxolan)].

Fremstilling av (3<1>S,IS,2S,3R,6S)-2-(3'-hydroxy-3'-cyclo-hexylprop-1 '-ynyl)-3-hydroxy-8-endo-methylbicyclo-[4.2.0]-octan-7-on med formler XIV og XV.
En oppløsning av 110 mg av (3'S,IS,2S,3R,6S)-cyclohexylpropynylketalproduktet fra Fremstilling 11 eller fra Fremstilling 12, hvor Y er endo-methyl, 3 ml acetonitril og 1,5 ml 2N svovelsyre ble omrørt ved omgivelsenes temperatur i 16 timer. Reaksjonen ble stoppet ved nøytrali-sering med vandig natriumbicarbonat, og blandingen ble ekstrahert med diethylether. Ekstraktene ble tørret med magnesiumsulfat og inndampet til tørrhet, og residuet ble renset ved kromatografering på en kort silicagelkolonne. Ved eluering med ethylacetat-hexan (7:3) ble det erholdt 58 mg (3'S,IS,2S,3R,6S)-2-(3'-hydroxy-3'-cyclohexylprop-1'-ynyl)-3-hydroxy-8-endo-methylbicyclo-[4.2.0]-octan-7-on.

Eksempel
Fremstilling av (Z)-(3'S,IS,2S,3R,6S)-4-[2-(3<1->hydroxy-3'-cyclohexylprop-1'-ynyl)-3-hydroxy-8-endo-methylbicyclo-[4.2.0]-oct-7-yliden]-butansyre og (E)-(3'S,IS,2S,3R,6S)-4-[2-(3'-hydroxy-3'-cyclohexylprop-1<1->ynyl)-3-hydroxy-8-endo-methylbicyclo- [4.2.0 ]-oct-7-yliden]-butansyre og beslektede forbindelser med formler (1), (2) og (3)
A. En forrådsoppløsning av dimsyl-natrium ble til-beredt ved oppløsning av 0,6 g natriumhydrid i 10 ml dimethylsulfoxyd ved 65°C under nitrogen. Til en omrørt oppslemning av 1,13 g 3-carboxypropyltrifenylfosfoniumbromid i 5 ml dimethylsulfoxyd under nitrogen ble det satt 4,0 ml av forrådsoppløsningen av dimsyl-natrium. Etter 20 minutter ved 23°C ble en oppløsning av 180 mg (3'S,IS,2S,3R,6S)-2-(3'-hydroxy-3'-cyclohexylprop-1'-ynyl)-3-hydroxy-8-endo-methylbicyclo-[4.2.0]-octan-7-on (Fremstilling 13) i 1 ml dimethylsulfoxyd tilsatt i én enkelt porsjon. Etter fire timer ved 23°C ble blandingen hellet over i 15 ml av en 5% natriumcarbonatoppløsning. Denne blanding ble vasket med to 30 ml's porsjoner ethylacetat og ble deretter surgjort med kons. HC1. Det vandige skikt ble ekstrahert tre ganger med 50 ml's porsjoner diethylether. De sammenslåtte ether-ekstrakter ble inndampet til 20 ml, og den inndampede ekstrakt ble holdt ved~-20°C i to timer. Den resulterende utfeining ble frafiltrert og hevet. Ved inndampning av filtratet ble det erholdt 300 mg av en olje. Dette materiale ble renset ved flash-kromatografering på silicagel under anvendelse av en oppløsningsmiddelblanding bestående av eddiksyre-ethylacetat-hexan (0,25:75:25), hvorved man fikk 230 mg av en olje. Ved ytterligere rensning ved flash-kromatografering på silicagel under anvendelse av en opp-løsningsmiddelblanding av eddiksyre-methanol-diklormethan (0,2:5,3:94,5) fikk man separert produktblandingen i de individuelle forbindelser av henholdsvis formel 2 og formel 1.
Den første forbindelse som ble eluert, var: (E)-(3'S,IS,2S,3R,6S)-4-[2-(3'-hydroxy-3'-cyclo-hexylprop-1'-ynyl)-3-hydroxy-8-endo-methylbicyclo-[4.2.0]-oct-7-yliden]-butansyre, sm.p. 43-50°C.
Den andre forbindelse som ble eluert, var: ( Z)-(3'S,IS,2S,3R,6S)-4-[2-(3'-hydroxy-3'-cyclo-hexylprop-1 ' -ynyl)-3-hydroxy-8-endo-methylbicyclo-[4.2.0]-oct-7-yliden]-butansyre, sm.p. 46-51°C.
B. Ved at man erstattet (3'S,IS,2S,3R,6S)-2-(3'-hydroxy-3'-cyclohexylprop-1'-ynyl)-3-hydroxy-8-endo-methyl-methylbicyclo-[4.2.0]-octan-7-on med andre passende forbindelser med formel XIV eller XV, fremstilt som beskrevet i Fremstilling 13, og benyttet 3-carboxypropyltrifenylfosfoniumbromid eller 4-carboxybutyltrifenylfosfoniumbromid, ble på tilsvarende måte de følgende forbindelser med formler (1), (2) og (3) fremstilt: (E)-(3'S,IR,2R,3S,6R)-4-[2-(3'-hydroxy-3'-cyclo-hexylprop-1 1-ynyl)-3-hydroxy-8-endo-methylbicyclo-[4.2.0]-oct-7-yliden] -butansyre, [ct]D = -93,8°-C=0,3698, (CHC13). (Z)-(3<1>S,IR,2R,3S,6R)-4-[2-(3'-hydroxy-3<1->cyclo-hexylprop-1 1-ynyl)-3-hydroxy-8-endo-methylbicyclo-[4.2.0]-oet-7-yliden]-butansyre, sm.p. 50-56°C. (E)-(3'S,IS,2S,3R,6S)-4-[2-(3'-hydroxy-3'-cyclo-hexylprop-1 '-ynyl)-3-hydroxy-8-endo-methylbicyclo-[4.2.0]-oct-7-yliden]-pentansyre, sm.p. 36-41°C. (Z)-(3'S,IS,2S,3R,6S)-4-[2-(3'-hydroxy-3 *-cyclo-hexylprop-1 '-ynyl)-3-hydroxy-8-endo-methylbicyclo-[4.2.0]-oct-7-yliden]-pentansyre, sm.p. 31-38°C. (E)-(3'S,IR,2R,3S,6R)-4-[2-(3'-hydroxy-3'-cyclo-hexylprop-1 '-ynyl)-3-hydroxy-8-endo-methylbicyclo-[4.2.0]-oct-7-yliden]-pentansyre, [a]D = -71,9° C=0,4183, (CHCI3).
(Z)-(3'S,IR,2R,3S,6R)-4-[2-(3'-hydroxy-3'-cyclo-hexylprop-1 1-ynyl)-3-hydroxy-8-endo-methylbicyclo-[4.2.0]-oct-7-yliden]-pentansyre, [a]D = -90,8° C=0,1928, (CHCI3).
Claims (3)
1. Analogifremgangsmåte for fremstilling av terapeutisk virksomme 8-(lavere alkyl)-bicyclo-[4.2.0]-octan-derivater med den generelle formel (1), (2) eller (3):
hvor Y er exo-(lavere alkyl) eller endo-(lavere alkyl).
n er 2 eller 3, og R3 er
hvor b er 3-7, og de i farmasøytisk henseende aksepterbare, ikke-toksiske salter og estere derav,
karakterisert ved at en forbindelse med den generelle formel:
hvor Y og R3 er som ovenfor angitt, omsettes med anionet (eller ylidet) av en fosfoniumforbindelse med den generelle formel:
hvor X, Y og Z er alifatiske eller aromatiske grupper, og n er 2 eller 3, eller et beskyttet derivat eller salt derav, for dannelse av en blanding av en forbindelse med formel (1) og en forbindelse med formel (2) eller en blanding av forbindelse (3) og den tilsvarende E-forbindelse, og den opp-nådde blanding separeres, og at én syre med formel (1), (2) eller (3) eventuelt overføres til et i farmasøytisk henseende aksepterbart, ikke-toksisk salt eller ester derav.
2. Analogifremgangsmåte ifølge krav 1, for fremstilling av (Z)-(3'S,IS,2S,3R,6S)-4-[2-(3'-hydroxy-3'-cyclo-hexylprop-1 '-ynyl)-3-hydroxy-8-endo-methylbicyclo-[4.2.0]-oct-7-yliden]-smørsyre, og de i farmasøytisk henseende aksepterbare, ikke-toksiske salter og estere derav, karakterisert ved at de tilsvarende utgangsmaterialer benyttes.
3. Analogifremgangsmåte ifølge krav 1, for fremstilling av (Z)-(3'S, IR, 2R,3S,6R)-4-(; 2-( 3'-hydroxy-3'-cyclo-hexylprop-1 '-ynyl)-3-hydroxy-8-endo-methylbicyclo-[4.2.0]-oct-7-yliden]-smørsyre, og de i farmasøytisk henseende aksepterbare, ikke-toksiske salter og estere derav, karakterisert ved at de tilsvarende utgangsmaterialer benyttes.
