Description
Foreliggende oppfinnelse omfatter serumfritt medium som er
av en slik sammensetning som er angitt i krav 1, og som kan anvendes ved dyrking av celler som krever jern i vekstmediet. Oppfinnelsen kan brukes til å teste kvaliteten av andre vekstmedier i tillegg til at det muliggjør dyrking av celler i medium fri for serum.
Det har innenfor industri og forskning lenge vært et stort behov for serumfrie medier og vekstkulturer idet tilføringen av serum til slike medier gir oppløsningene, i tillegg til de stoffer som er nødvendige for celleveksten, også stoffer som innvirker uheldig på de fleste forsøk hvor nøyaktihet av resultatene er av stor betydning. Dette gjelder innenfor disipliner som cellulær immunologi, bioteknologi, "in vitro" fertilisering, organtransplantasjoner, kreftforskning og ved lagring og overføring av blod.
De medier og fysiologiske løsninger som idag benyttes i forskning, industri og ved klinisk arbeid baserer seg på inntil 100 år gamle resepter, og representerer således et "stand-still" i dette område av den biologiske forskning. De hittil anvendte media for cellekulturer består oftest av et såkalt "kommersielt basalmedium" som er tilsatt ca. 10% varmeinaktivert serum. Denne meget gamle metoden innebefatter følgende ulemper: 1) Varmebehandling som må til for å hindre serumets lytiske effekt på celler virker denaturerende på
viktige serum-komponenter.
2) Forskjellige serum-leveranser har forskjellige egenskaper ut fra deres oppfinnelse, noe som innfører uønskede variasjoner i cellekulturens oppførsel "in
vitro".
3) Serum inneholder mange ukjente stoffer med ukjent og ukontrollert innvirkning på celler. 4) Serum er en ufysiologisk væske for de aller fleste
celler idet cellene er tilpasset såkalte vevsvæsken i kroppen idet denne har et annet innhold av forskjellige stoffer enn
serum.
5) Antistoffer i serum kan binde seg til celler og innvirke på eksperimenter. 6) Serum hindrer studium av cellers syntese av serumfaktorer "in vitro" idet "bakgrunnsnivået" i serum for slike faktorer er såvidt høyt.
Ved anvendelse av det serumfrie medium ifølge oppfinnelsen og som inneholder de komponenter som er angitt i krav l's karakteriserende del, unngås de ovenfor angitte ulemper, og et slikt medium vil i tillegg være meget godt anvendelig for dyrkning og oppbevaring av celler og vev "in vitro". Dess- uten vil det fullstendige medium ifølge oppfinnelsen være nøyaktig definert, være serumfritt, proteinfritt bortsett fra evt. små mengder av et polypeptid sam f.eks. insulin som kreves av enkelte celler og som kan benyttes hvor dette er nødvendig, inneholde jern og sporelementer sam stabiliserte, chelaterte forbindelser uten transferrin og intet albumin eller lipid er tilstede. Mediet ifølge foreliggende oppfinnelse er også meget billig å produsere.
Hovedproblemet ved tidligere serum-fri definerte medium (se N.N. Iscove og F. Melchers, "Complete Replaoement of Serum by Albumin, Transferrin, and Soybean Lipid in cultures of Lipopolysaccharide-reactive B Lympho-cytes", J.Exp. Med., Vol. 147, s. 923-933 (1978); R.G.Ham, "Importance of the Basal Nutrient Medium in the Design of Hormonally Defined Media, Cold Spring Harbor Conference on Cell Proliferation, Vol.9, s.39-60 (1982); D.Barnes og G.Sato "Methods for Growth of Cultured Cells in Serum-Free Medium" Anal. Biochem. 102, s. 255-270 (1980) er at de ikke er lempelige ved anvendelse på en tilfeldig valgt cellekultur på grunn av to omstend-igheter: 1) Tilstedværelsen av fysiologiske megder bikarbonat og Ca<2+>/Mg<2+->ioner vil utfelle jern og trivalente sprometaller fra nesten ethvert kjent chelatorsystem ved 37'C og pH 7-8 (bortsett fra transferrin). Det eneste løsningen på dette problem er å fremskaffe en fullstendig stabil metallionbuffer som omfatter en balansert oppløsning av alle de nødvendige spormetaller, og således blir toksiske metaller absorbert i bufferen (den toksiske effekt blir balansert bort).
Idet transferrin er kostbart og spesifikt for hvert dyre-slag, ble mediet ifølge oppfinnelsen dannet, basert på en Fe/ sporelement-buffer av syntetiske ikke-protein chelatorer og hvor hovedproblemet, utfelling av metaller ved 37°C (se ovenfor) , bl.e løst ved tilsetning av bestemte chelatorer som i blanding har svært forskjellige og unike egenskaper sammenlignet med hver chelator alene.
Det andre problemet, nemlig presentasjon av jem til cellemembranen
og cellen, ble løst ved tilsetning av aurintrikarboksylsyre til mediet. Selv cm EDTA/citrat gjør en metalloppløsning fullstendig stabil, vil forskjellige animalske og humane celler ikke vokse på
grunn av jemmangel. Det er funnet at kombinasjonen EDTA/citrat/ aurintrikarboksylsyre gir enestående chelaterende cg jempresenterende egenskaper sam tidligere ikke er beskrevet. (I denne forbindelse bør det ikke være av betydning at EDTA alene tidligere har blitt birukt sam jern-chelator for vekst av planteceller ved pH 5,0 under betingelser hvor metallutfelling ikke representerer noe problem; se P.Kruse og M.K.Petterson,"Tissue Culture Methods and Applications", Acad.Press
(19873); J.R.Stein,"Handbook of Phycological Metods", Cambridge University Press (1973).)
I forhold til cellevekst er det funnet at et blandings-forhold mellom EDTA og sitronsyre (citrat) på 1:3 var mest fordelaktig. Det skal også kort nevnes at cellevekst i medier vanligvis foregår ved pH 7,4, men at pH i det endelige medium ifølge foreliggende oppfinnelse også kan være forskjellig fra dette, avhengig av de enkelte cellers vekstkrav. pH for det serumfrie vekstmedium ifølge oppfinnelsen vil generelt ligge innenfor 7,0-7,8.
Vekstmedium ifø?ge oppfinnelsen kan fremskaffes eksempelvis ved å tilsette en konsentrert oppløsning av de enkelte bestanddeler til rent vann (det er av stor viktighet å benytte vann av størst mulig renhet, som f.eks. Travenol® sterilt vann, ved fremstilling av mediet ifølge oppfinnelsen. Et eksempel på en slik konsentrert blanding er gitt nedenfor, hvor det fremstilles en blanding som kan benyttes i luuu x konsentrasjon:
pH i blandingen justeres fortrinnsvis til 4,5 - 5,0.
Eventuell tilsetning av spormetaller kan eksempelvis bli foretatt ved hjelp av en stamløsning som kan fremstilles som følger. Den angitte løsning vil ha 100 000 ganger konsentrasjon av det som anvendes i mediet. Ved dannelse av oppløsningen angitt nedenfor, bør Zn, Cu og chelatorer (EDTA og citrat) tilsettes før justering av pH til 5,0 med 5 - 10 N NaOH, hvoretter de ytterligere komponenter tilsettes. Den endelige pH bør ligge mellom 4,0 og 4,5. Det er selvfølgelig underforstått at en slik blanding av sporelementer kan inneholde andre elementer enn dem som er angitt i tabellen ovenfor, avhengig av de krav som stilles av de anvendte vevs- og celletyper. j
Mediet ifølge foreliggende oppfinnelse kan, i tillegg til
de bestanddeler som tidligere er angitt, også inneholde vekstfremmende og nødvendige forbindelser for cellevekst. Slike forbindelser kan omfatte f.eks. det o/erflateaktive middel Pluronic F6&<®> (20 mg/l), D-glukose (2,5 g/l), L-glutamin (2 mM), Na-pyruvat (1 mM), Insulin (0,5 mg/l) eller etanolamin (20 uM) . Alle i parantes angitte konsentrasjoner refererer seg til slutt-konsentrasjoner.
En tilsetningsblanding av de angitte forbindelser (med unntak av etanolamin som er flytende ved romtemperatur)
kan fremstilles som tørr-produkt, og som sådan kan dette lagres over lang tid, opp til flere år. Etanolamin kan om nødvendig tilsettes mediet i den ønskede konsentrasjon ved anvendelse av blandingen. Etanolamin brukes som oftest ved forsøk med hybridomceller under fremstilling av monoklonale antistoff.
Et foretrukket basalmedium hvori medietilsetningen ifølge foreliggende oppfinnelse kan tilsettes, er RPMI 1640 w/
2,5 g/l NaHC03 og 20 mM HEPES (N-2-hydroksyetylpiperazin-N-N'-2-etansulfonsyre).
Foretrukne cellekulturer hvor det serumfri mediet ifølge foreliggende oppfinnelse har vist seg spesiellt anvendelig er med L 929 muse fibroblast cellelinje, HELA human tumor cellelinje, forskjellige muse- og humane B hybridomceller, som alle danner monoklonale antistoff i mengder som tilsvarer vekst i serum-inneholdende kulturer og humane monocytter som differensierer til utviklede makrofager i løpet av 7 dager under opprettholdelse av sine komplement-reseptorer. En spesielt foretrukket anvendelse av foreliggende oppfinnelse utføres med hurtigvoksende celletyper for kvali-tetsprøving av andre typer medier. Ved å blande den serumfrie medietilsetning ifølge oppfinnelsen med PRMI 1640 og tilsette mediet som skal undersøkes i forholdet serumfri medietilsetning + RPMI 1640 : ukjent medium= 1 : 1, og deretter tilsette de hurtigvoksende celler i et antall på
ca. 30 000 celler/ml, vil det uten toksiske forbindelser tilstede i mediet som skal undersøkes kunne observeres en økning i celleantallet etter 4 dager på ca. 10 ganger flere celler enn det opprinnelige antallet. Dersom toksiske, forbindelser derimot er tilstede i det ukjente mediet, vil det ikke observeres cellevekst. Dette omfatter således en meget enkel kvalitetstest som er ekstremt følsom fordi den foretas uten serum, og falske resultater unngås fordi en unngår at serum binder og maskerer toksiske urenheter.
En spesiell type hurtigvoksende celler er en hybridom celletype laget ved fusjon av ^ 2^ 1 tumor med muse B-lymfocytter. Denne hybridom celletype er deponert hos European Collection of Animal Cell Cultures, ECACC, Storbritannia, 20 november 1986, ECACC 86112001. Denne cellelinje har vist seg spesielt anvendelig ved kvalitetstesting av andre vekstmedier som angitt ovenfor.
Den deponerte cellelinje ble fremstilt i henhold til myeloma hybrid teknikken introdusert av Kohler G. og Milstein C., 1975 i Nature (London) 256, 495.
Utgangscellene var ^ 2^ 1 meyloma og Balb/c B-lymfocytter, alle typer fra mus.
Hybridomceller er kjent for sine krav til mediumskvalitet. Cellelinjen 1E6 ble valgt ut som den raskeste voksende hybridom av 20 undersøkte linjer. Linjen ble brukt i et eksperimentelt overvåkningsforsøk av forskjellige kommersielt tilgjengelige media fra velkjente
forhandlere. Kun to av ni undersøkte medier ble funnet
å være av akseptabel kvalitet. Seks andre medier viste 60 - 100 % cytotoksisitet. Den serum-frie hybridom-
test er mer sensitiv enn andre bio-assays som innebefatter tilsetning av serum (detoksifiserende effekt) under dyrkning.
Foreliggende oppfinnelse kan også bli brukt for å identi-fisere toksisiteten til potensielt toksiske substanser ved å undersøke deres effekt på veksten av lE6-celler ifølge oppfinnelsen sammen med det serum-frie medium.
Kvaliteten av vekst-medier ble undersøkt i et sammen-ligningsforsøk med muse-embryo-testen. Muse-embryo-testen omfatter at to in vivo befruktede museembryoer dyrkes 4-5 dager i medium-prøvene. Dersom mer enn 80%
av embryoene utvikler seg til blastocytter, kan mediet aksepteres.
Celle-kultur-forsøket omfatter hybridom cellelinjen 1E6 og RPMI-16.4 0/SSR medium (RPMI/SSR) . Forsøket ble utført i henhold til følgende trinn:
Trinn 1: Bland RPMI/SSR og undersøk mediumprøven
1:1 (uten serum).
Trinn 2: Tilsett hybridomceller ved 1,5 x 10 celler/ml,
inkubér 4 dager.
Trinn 3: Tell cellene. Dersom ingen toksiske substanser er tilstede, bør tellingen være ca. 3 x IO<5 >celler/ml.
I et forsøk for å undersøke toksisiteten av lidokain, ble forbindelsen undersøkt for inhibering av cellevekst på cellelinjen 1E6. Resultatene er gitt nedenfor.
Det kan sluttes fra forsøkene at lidokain i en konsentrasjon på 0,01 mg/ml eller høyere ble funnet å være toksisk. Ingen toksisitet ble funnet ved must-embryo-testen.
Dette viser at testen som benytter hybridom og vekstmedium ifølge oppfinnelsen, er overlegen enhver test som vanligvis foretas idag.
Det kan også konkluderes at muse-embryo-testen er meget insensitiv sammenlignet med celle-hybridom-testen.

Vekstmediet ifølge foreliggende oppfinnelse sammen med nevnte hurtigvoksende hybridom celletype kan således inne-befattes i et test-kit omfattende f.eks. serumserstatningen ifølge oppfinnelsen, nedfrosne celler (f.eks. på tørris) samt eventuelt medium (RPMI 1640) og kulturutstyr av plast eller annet egnet materiale.
Claims (11)
1. Serumfritt vekstmedium bestående av et basalmedium for cellekulturer, såsom RPMI 1640/HEPES/NaHCO3, samt eventuelt sporstoffelementer som er nødvendige for cellevekst, såsom Zn, Cu, Mn, Ni, Al, Cr, Co og/eller Se,
hvilket vekstmedium er bufret i et pH-område innenfor 7,0-7,8, karakterisert ved at vekstmediet ytterligere består av et ikke-toksisk jernchelat hvori jern foreligger i konsentrasjonsområdet 1-10 uM, samt et overskudd av en eller fler i og for seg kjente, ikke-toksiske chelatorer og sitronsyre/citrat i en konsentrasjon som er minst tre ganger så stor som den totale konsentrasjon av chelator, samt aurintrikarboksylsyre i konsentrasjonsområdet 0,5-6 uM.
2. Serumfritt vekstmedium ifølge krav 1, karakterisert ved at aurintrikarboksylsyren foreligger i et konsentrasjonsintervall på 2,4 - 3,6 uM.
3. Serumfritt vekstmedium ifølge krav 1 eller 2, karakterisert ved at eventuelle sporstoffelementer foreligger i en mengde slik at konsentrasjonen av chelator/chelatorblanding er 300 - 300000 ganger større enn konsentrasjonen av de enkelte sporstoffelementer.
4. Serumfritt vekstmedium ifølge kravene 1-3, karakterisert ved at det inneholder ett eller flere overflateaktive midler.
5. Serumfritt vekstmedium ifølge ethvert av kravene 1-3, karakterisert ved at det inneholder ytterligere vekstfaktorer såsom D-glukose, L-glutamin, Na-pyruvat, insulin og etanolamin.
6. Serumfritt vekstmedium ifølge krav 5, karakterisert ved at det ytterligere inneholder vekst
faktorer såsom vitaminer og ikke-proteiniske hormoner.
7. Serumfritt vekstmedium ifølge kravene 1-6, karakterisert ved at det inneholder et basalmedium med konsentrasjonene 2,5 g/l NaHCO^ og 20 mM HEPES.
8. Serumfritt vekstmedium ifølge kravene 1-7, karakterisert ved at at den anvendte chelator er EDTA.
9. Konsentrat av serumfritt vekstmedium for anvendelse til fremstilling av vandig serumfritt vekstmedium ifølge kravene 1-6, karakterisert ved at det består av'et konsentrat av de enkelte bestanddeler, fortrinnsvis 1000 gangers konsentrat bestående av 2,4 - 3,6 mM Fe-EDTA og 0,2 - 1,2 mM Na2~EDTA samt 32 - 48 mM sitronsyre/citrat, 0,5 - 6 mM, fortrinnsvis 2,4 - 3,6 mM aurintrikarboksylsyre, samt eventuelt metallioner innenfor konsentrasjonsintervallene 0,08 - 0,12 mM Zn<2+>, 0,016 - 0,024 mM Cu<2+>, 0,0008 - 0,0012 mM Mn<2+>, 0,00016 - 0,00024 mM Ni2+, 0,0016 - 0,0024 mM Al3+, 0,0008 - 0,0012 mM Cr<3+>, 0,00016 - 0,00024 mM Co<2+> og 0,008 - 0,012 mM Se<4+>, hvilken blanding bufrer i pH-området 4,5 - 5,0.
10. Konsentrat av serumfritt vekstmedium ifølge krav 9, karakterisert ved at det foreligger som tørrstoffblanding.
11. Anvendelse av serumfritt vekstmedium ifølge kravene 1-7 for undersøkelse av vekstmediumkvalitet, fortrinnsvis ved bruk av cellelinjen ECACC 86112001, deponert ved The European Collection for Animal Cell Cultures den 20. november 1986.
