Oppfinnelsens område
Oppfinnelsen gjelder en fremgangsmåte for fremstilling
av naftalen-derivater som er nyttige til å forhindre visse dermatologiske tilstander og til å lindre virkningen av visse kroniske tilbakevendende papulosquamøse .dermatoser, for eksempel psoriasis.
Beslektede arbeider
Psoriasis er en hudsykdom karakterisert delvis ved usedvanlig sterk deling av cellene i epidermis, og disse for-blir sterkt fastsittende og bygges opp til et skjellet belegg som er typisk for sykdommen. For tiden tilgjengelige terapier, som ikke er helbredende, avhenger av kontroll av epidermcellenes delig ved bruk av hormonpreparater, såsom corticosteroider eller ved bruk av forbindelser beslektet med kreft-kjemoterapi såsom hydroksyurea, metotrexat og nitrogen sennepsgass-forbindelser.
Mens midlene ovenfor er effektive til en viss grad, forårsaker de tallrike alvorlige uønskede bivirkninger som omfatter nyreirritasjon, levertoksisitet og erytem.
Forbindelsen 2,3-dimetoksy-l,4-diacetyloksynaftalen er kjent og er rapportert å være nyttig som et utgangsstoff ved syntese, men ingen nyttig biologisk aktivitet har blitt tilskrevet det.
Se J. Chem. Res., Synop. 1980(4), 156-7 og An. Quim. 1976, 72(3):247-53. Visse naftokinoner er kjent å være nyttige ved behandling av psoriasis. Se for eksempel US-patent nr. 4 229 478 og britisk patent nr. 1 243 401. Men, disse forbindelsene har en eller flere ulemper såsom at de forårsaker hudirritasjon, farging av huden og sensibilisering av pasienten. Overraskende er det oppdaget at forbindelsene fremstilt ifølge den foreliggende oppfinnelse også er effektive anti-psoriasis-midler og er mindre irriterende, farger ikke huden og sensi-biliserer ikke når de brukes ved behandling av psoriasis.
Videre er forbindelsene fremstilt ifølge den foreliggende oppfinnelse mer stabile i de topiske preparater som vanligvis brukes.
Oppsummering
Den foreliggende oppfinnelse gjelder en fremgangsmåte
for fremstilling av en forbindelse med den følgende formel
hvori:
R<1> og R<2> er de samme og er alkoksy med ett til seks karbonatomer, eller fenoksy eventuelt substituert med halogen;
R<3> er alkyl med ett til seks karbonatomer, eller halogen, og m er 0, 1 eller 2; og
W er alkyl med ett til syv karbonatomer eller fenyl;
med det forbehold at m er 2 når R<1> og R<2> er C^-Cg alkoksy og W er en alkylgruppe med ett til syv karbonatomer.
Tilstanden av psoriasis hos et pattedyr kan lindres ved topisk å administrere til nevnte pattedyr en psoriasis-lindrende mengde av en forbindelse med formel (I).
Ifølge oppfinnelsen fremstilles forbindelser med formel
(I) ved omsetning av forbindelser med formel (XI) (infra) med
et syreanhydrid.
Alkylgrupper som inneholder ett til syv karbonatomer, inkluderte grupper med rette kjeder eller grupper med forgrenede kjeder. Illustrerende for slike grupper er for eksempel metyl, etyl, n-propyl, i-propyl, n-butyl, i-butyl, s-butyl, n-pentyl, i-pentyl, n-heksyl, 2-metylpentyl og n-heptyl. Eksempler på alkylgrupper med ett til seks -karbonatomer er metyl, etyl, n-propyl, i-propyl, n-butyl, i-butyl, s-butyl, 2,2-dimetylpropyl og t-heksyl.
Alkoksy med ett til seks karbonatomer er en alifatisk gruppe med rett eller forgrenet kjede bestående av ett til seks karbonatomer hvortil er bundet en oksygengruppe. Eksempler på slike er metoksy, etoksy, n-propoksy, i-propoksy, n-butoksy, i-butoksy, s-butoksy, t-^butoksy og n-pentyloksy.
Alkyltio med ett til seks karbonatomer er en alifatisk gruppe med rett eller forgrenet kjede på ett til seks karbonatomer hvortil det er bundet en svovelgruppe. Eksempler på slike er metyltio, etyltio, n-propyltio, i-butyltio og n-heksyltio.
Uttrykket "halogen" refererer til fluor, klor og brom.
Tillaging og administrering
Preparatene kan lages til for administrering på en hvilken som helst hensiktsmessig måte i analogi med andre topiske preparater som er tilpasset til bruk på pattedyr. Disse preparatene kan fremstilles til bruk på enhver hensiktsmessig måte med hjelp av noen i et vidt omfang av farmasøytiske'bære-stoffer eller hjelpestoffer.
Naftalene med formel (I) kan lages til med egnede farmasøytiske hjelpestoffer som er kjent innen faget, for å danne særlig effektive topiske preparater. En effektiv mengde av naftalenforbindelsen er omkring 0,001 % til omkring 10 %
av vekten av det totale tillagede preparat. Resten av det tillagede preparat vil være omkring 90 % til omkring 99,999 %
i vekt av en egnet eksipiens som kan omfatte et farmasøytisk godtagbart oppløsningsmiddel og andre farmasøytiske godtag-
bare tilsetningsstoffer slik at det dannes et topisk effektivt farmasøytisk preparat.
En fremgangsmåte for å lindre tilstanden av psoriasis
hos et pattedyr, omfatter topisk administrering av et preparat som inneholder en forbindelse med formel (I) hvori n, R 1 , R 2,
R 3, W, m og n er som definert ovenfor. Generelt bekjempes de psoriatiske utslag hos pattedyr, særlig mennesker, ved å
bringe de inflammerte områder i kontakt med en terapeutisk effektiv mengde av det naftalen-inneholdende preparat, det vil si, en mengde som resulterer i en minskning av delingen av epidermcellene (en anti-psoriasis-effekt). Fortrinnsvis lages naftalenene først til som et egnet farmasøytisk preparat,
og dette bringes deretter i kontakt med de rammede områder. En effektiv mengde av naftalenforbindelsen vil avhenge av den spesielle tilstand og pattedyret som får behandling, og vil variere mellom 0,001 % og 10 % i vekt av det farmasøytiske preparat og vil fortrinnsvis være mellom 0,01 % og 1 % i vekt av preparatet. Ved å benytte disse nivåer i preparatet, påføres en terapeutisk effektiv mengde som ikke gir bivirkninger, dvs. tilstrekkelig til å bevirke en anti-psoriasis-reaksjon, men ikke nok til å gi skadevirkninger på mottageren, på de rammede områder.
Oppfinnelsen gjelder også en fremgangsmåte for fremstilling av forbindelser med den ovenfor angitte formel (I), og den omfatter omsetning av en forbindelse med formel
hvori:
R1, R<2>, R<3> og m er som definert ovenfor, med et syreanhydrid.
FREMSTILLING
Forbindelsene med formel (I) kan fremstilles ut fra for-bindelense med formel (V) eller (VI).
Utgangsforbindelsene med formel (V) eller (VI) hvori m er 1, kan fremstilles ved reaksjonsrekkefølgen nedenfor.

Forbindelser med formel (III) fremstilles ved å starte
med et 2,3-dihalogennaftokinon, fortrinnsvis 2,3-diklor-l,4-naftokinon (forbindelse med formel (II)), som er'tilgjengelig fra bl.a. Aldrich Chemical Co., og direkte nitrering. Reaksjonen forløper på en måte som er kjent for polycykliske aroma-tiske forbindelser og gir en blanding av 5- og 6-nitro-2,3-diklor-1,4-naftokinoner, forbindelser med formel (III). Reaksjonen utføres typisk med konsentrert salpetersyre i et opp-løsnings-medium med lav pH, fortrinnsvis konsentrert svovel-syre, typisk ved 20°C til 100°C i et tidsrom som er tilstrekkelig til at reaksjonen blir fullstendig. Avhengig av reaksjonstemperaturen og reaksjonstiden, kan forholdene i 5-nitro-isomer: 6-nitro-isomer-blandingen variere fra 10:1 til 1:10, typisk 8:1.
Forbindelser med formel (IV) syntetiseres fra 5- og 6-nitro-2,3-diklor-l,4-naftokinon, forbindelsen med formel (III), ved å kondensere dem med et alkalimetallalkoksyd eller
-fenoksyd hvori alkoksy- eller fenoksy-delen er R 1 =R 2. Reaksjonen foregår fortrinnsvis i et inert organisk oppløsnings-middel såsom tetrahydrofuran, dimetylsulfoksyd, dimetylformamid og lignende, ved en temperatur fra rundt 20°C til rundt 100°C
i et tidsrom som er tilstrekkelig til å sikre at reaksjonen er fullstendig, for eksempel i rundt 2 timer til rundt 48 timer.
Forbindelser med formel (IV) hvori R^=R<2> og er C1 .-C6, alkyltio, fremstilles fra forbindelser med formel (III) på
den samme måte som for alkoksy- og fenoksy-forbindelser, bortsett fra at et alkalimetallsalt av en alkylmerkaptan brukes i stedet for et alkalimetall-alkoksyd eller
-fenoksyd.
Forbindelsene med formel (V) fremstilles fra forbindelser med formel (IV) ved katalytiske eller ikke-katalytiske reduksjons-prosesser som er kjent fra tidligere. Sammensetninger av metall og syre som reduksjonsmidler, såsom granulert jern og saltsyre, tinn og saltsyre og lignende eller nøytrale reduksjonsmiddel-sammensetninger såsom sinkstøv og vandig alkohol eller aluminium-amalgam og vandig alkohol samt organometalliske reduksjonsmidler såsom litium-aluminiumhydrid, natriumborhydrid og lignende, kan brukes ved denne reduksjonen. Fortrinnsvis ut-føres reduksjonen ved å behandle forbindelsene med formel (IV) med overskudd av hydrazin i nærvær av en katalytisk tilstrekkelig mengde av palladium, typisk på karbon. Reaksjonen skjer lett ved romtemperatur, reaksjonstiden styres ved tilsetnings-hastigheten av hydrazinet til reaksjonsblandingen, som typisk er rundt 1 til rundt 10 timer.
Forbindelser med formel (VI) hvori RJ er hydrogen
(dvs. m er 0), kan fremstilles ved å omsette forbindelsen
med formel (II) med et alkalimetall-alkoksyd eller -fenoksyd eller alkalimetallsalt av en merkaptan som beskrevet her ovenfor.
Forbindelser med formel (V) overføres til forbindelser
med formel (VI) der R 3 er halogen, ved å tilsette til forbindelsen med formel (V) i en surgjort vandig oppløsning en oppløsning av et alkalimetallnitritt. Denne utgangsreaksjonen danner diazoniumsaltet ved 5- eller 6-posisjonen på nafto-kinonringen. Saltet dekomponeres med en oppløsning av kobber(I)-halogenid dispergert eller løst opp i den tilsvarende halogen-syre (Sandmeyers reaksjon). Denne klassiske reaksjon er behandlet inngående i Bigelow, Org. Synthesis, Coll. Vol. I, 126-133 (1941).
En særlig foretrukket måte for å fremstille forbindelser med formel (VI) hvori R 3 er 6-halogen, er vist i den følgende reaksjonsrekkefølge.

Forbindelser med formel (VIII) fremstilles ifølge fremgangsmåten som er vist i J. Am. Chem. Soc, 70, 3165 (1948)
og det samme,'71, 3615 (1949). Halogensubstituert 1,3-butadien omsettes med 1,4-benzokinon i et oppløsningsmiddél såsom eddiksyre ved en temperatur på -10°C til 30°C, fortrinnsvis ved 25°C i 24 til 72 timer, fortrinnsvis fra 40 til 48 timer. 5,7-dihydrogen-forbindelsen med formel (VIII) samles og behandles med et oksydasjonsmiddel såsom natriumdikromat, natriumnitritt eller lignende, som beskrevet i artiklene ovenfor, slik at det dannes forbindelser med formel (VIII). Forbindelser med formel (IX) fremstilles ved å boble klorgass inn i en oppløsning av forbindelsen med formel (VIII) i et oppløsningsmiddél såsom iseddik, nitrobenzen, karbontetraklorid og lignende, fortrinnsvis iseddik ved romtemperatur. Denne forbindelsen som kan iso-leres ved kjente midler, løst opp i et oppløsningsmiddél såsom eddiksyre, behandles med klorgass og en egnet katalysator såsom natriumacetat, jod, jern(III)klorid, dimetylformamid eller alkoholer under oppvarming med tilbakeløp i 1/2 til 4 timer, fortrinnsvis i 1 til 2 1/2 timer, og gir forbindelser med formel (X). Forbindelsene med formel (VI) hvori R 3 er halogen, fremstilles ved å omsette forbindelser med formel (X) med et alkalimetallalkoksyd eller -fenoksyd såsom natriumalkoksyd eller -fenoksyd, for eksempel natriummetoksyd eller natriumfenoksyd i et vannfritt oppløsningsmiddél såsom metanol, dimetylformamid og lignende, oppløsningsmidlet blir valgt i henhold til lengden på alkylkjeden i alkoksygruppen. Reaksjonsblandingen varmes opp med tilbakeløp i 1/2 til 3 timer, fortrinnsvis
1/2 til 1 1/2 timer. Forbindelser med formel (VI) samles
på vanlige måter såsom ved krystallisering.
Forbindelsene med formel (VI) hvor R og R' er C^-Cg alkyltio, kan fremstilles ved fremgangsmåten som er beskrevet for alkoksy- eller fenoksy-forbindelsen, bortsett fra at alkalimetallalkoksydet erstattes med alkalimetallsaltet av alkylmerkaptan med oppløsningsmidlet som er dimetylformamid og lignende.
Utgangsforbindelsen, 2-klor-l,3-butadien (kloropren) er tilgjengelig fra bl.a. Pfaltz and Bauer Chemical Co.
2-brom-l,3-butadien og 2-fluor-1,3-butadien kan fremstilles ved fremgangsmåter som er velkjent innen faget, for eksempel ved fremgangsmåtene som er diskutert i henholdsvis J. Am. Chem. Soc, 55 786 (1933) og US-patent 2 401 850.
Utgangsforbindelsene med formel (VI) hvori R er C,-C, alkyl eller C^-Cg alkoksy, kan. fremstilles ved fremgangsmåter som er velkjent innen faget, såsom ved å omsette diazoniumsaltet av forbindelsen med formel (V) med en passende forbindelse så som en alkohol, for eksempel metanol, etanol, benzyl-alkohol og lignende. Disse utgangsforbindelsene kan også fremstilles ved fremgangsmåten som er vist i Reaksjonsrekkefølge (II) hvori 2-halogen-l,3-butadien er erstattet med det passende 1,3-butadien såsom 2-metyl-l,3-butadien(isopren), 2-etyl-l,3-butadien, 2,3-dimetyl-l,3-butadien og lignende.
Forbindelser med formel (I) fremstilles fra forbindelser med formel (VI) ved først å hydrogenere slik at det dannes forbindelser med formel (XI)
i en hydrogenatmosfære i nærvær av en katalysator slik som palladium på kull og deretter omsette den hydrogenerte forbindelsen med et passende anhydrid og pyridin såsom eddiksyreanhydrid, benzosyreanhydrid og lignende i et oppløsningsmiddél såsom tetrahydrofuran, dietyleter og lignende. Forbindelser med formel (I) samles ved krystallisering.
Syreanhydridene er kommersielt tilgjengelige fra bl.a.
Aldrich Chemical Co. eller hvis de ikke er tilgjengelige, kan de fremstilles ved å kondensere den rette syren i nærvær av eddiksyreanhydrid eller acetylklorid som inneholder et spor av fosforsyre. Anhydridet samles ved destillering eller krystallisering.
Den følgende spesifikke beskrivelse er gitt for å gjøre det mulig for dem med fagkunnskap å klarere forstå og prakti-sere den foreliggende oppfinnelse. Den bør ikke betraktes som noen begrensning av oppfinnelsens omfang men bare som en illustrasjon og representasjon av denne.
Fremstilling 1
2, 3- diklor- 5- og 6- nitro- l, 4- naftokinon (Fremstilling av forbindelser med formel (III))
Finpulverisert 2,3-diklor-l,4-naftokinon (50 g 0,22
mol) ble tilsatt til en rørt blanding av konsentrert svovel-syre (170 ml) og 90 % salpetersyre (102 ml) ved en hastighet slik at det eksoterme reaksjon hevet temperaturen til 60°C.
Den resulterende blandingen ble rørt ved 60°C i nok 2 timer. Det gule krystallinske faste stoffet ble filtrert fra, vasket grundig med vann og omkrystallisert fra kloroform, og det ga 22,9 g av 5-nitro-isomeren, smeltepunkt 156-157°C. Det sterkt sure filtratet ovenfor ble helt i isvann. Det resulterende faste stoff ble filtrert fra, vasket grundig med vann og tørret, og det ga 20,8 g av en blanding av 5- og 6-isomeren. Fraksjonert krystallisering av denne blandingen fra eddiksyre og kloroform:isopropanol ga 2,8 g av 6-nitro-isomeren, smeltepunkt 184-187°C.
Videre mengder av både 5- og 6-isomeren ble oppnådd fra omkrystalliserings-morluten ved kromatografering på en silika-gel-kolonne og utvasking med kloroform:cykloheksan-blandinger.
Fremstilling 2
2, 3- dimetoksy- 5- nitro- l, 4- naftokinon
(Fremstilling av forbindelsene med formel (IV)).
A. En oppløsning av 2,3-diklor-5-nitro-l,4-naftokinon
(2,72 g, 10 mmol) i vannfritt tetrahydrofuran (15 ml) ble tilsatt til en oppløsning av IN natriummetoksyd (2 5 ml, 2 5 mmol) og den resulterende oppløsningen ble satt hen ved 22° i 16 timer. Eddiksyre (1 ml) ble deretter tilsatt, oppløsningen ble konsentrert i vakuum og residuet skilt mellom vann (50 ml) og kloroform (100 ml). Vannfasen ble videre ekstrahert med kloroform (2 x 50 ml). De samlede kloroformekstrakter ble tørret med MgSO^ og konsentrert i vakuum. Residuet ble omkrystallisert fra metanol og det ga 1,99 g av 2,3-dimetoksy-5-nitro-l,4-naftokinon, smeltepunkt 156-157°C.
B. Tilsvarende, ved å bruke fremgangsmåten ovenfor, del A, med bruk av 2,3-diklor-6-nitro-l,4-naftokinon, hvor det er passende,i stedet for 2,3-diklor-5-nitro-l,4-naftokinon,og det passende natriumalkoksyd eller natriumfenoksyd i stedet for natriummetoksyd, fremstilles den følgende forbindelse: 2,3-dimetoksy-6-nitro-l,4-naftokinon, smeltepunkt 113-114°C;
Fremstilling 3
2, 3- dimetoksy- 5- amino- l, 4- naftokinon
(Fremstilling av forbindelser med formel (V))
Hydrazin (4,0 ml, 125 mmol av 97 %) ble i løpet av 2 timer tilsatt dråpevis under omrøring til en blanding av tittelforbindelsen i Fremstilling 2 (19,9 g, 75,6 mmol), 56 % palladium på karbon (10 g) og etanol (750 ml) i nitrogen-atmosfære. Katalysatoren ble filtrert fra gjennom en pute
av celite som ble vasket med varm etanol (2 x 300 ml). De samlede filtrater og vaskevæsken ble konsentrert til tørrhet i vakuum og residuet ble omkrystallisert fra vann:etanol (1,5:1) og det ga 14,6 g av 2,3-dimetoksy-5-amino-l,4-naftokinon, smeltepunkt 116-117°C.
Tilsvarende, ved å bruke forbindelsen fra Fremstilling 2 i stedet for 2,3-dimetoksy-5-nitro-l,4-naftokinon, fremstilles den følgende forbindelse: 2,3-dimetoksy-6-amino-l,4-naftokinon, smeltepunkt 196-197°C;
Fremstilling 4
5- klor- 2, 3- :dimetoksy- l, 4- naf tokinon
(Fremstilling av forbindelse med formel (VI) der R er 5-klor)
En oppløsning av natriumnitritt (0,69 g, 10 mmol) i vann (5 ml) ble tilsatt ved 0-5°C til en oppløsning av 5-amino-2,3-dimetoksy-1,4-naftokinon (1,17 g, 5 mmol) i 5:1 eddiksyre:vann (25 ml) som inneholder konsentrert saltsyre (1,7 ml). Nok en mengde natriumnitritt (0,69 g) ble déretter tilsatt til reaksjonsblandingen etter avkjøling til -5°C, etterfulgt av en oppløsning av kobber(I)klorid (0,6 g) i konsentrert saltsyre (5 ml). Blandingen fikk varmes opp til 22°C og fast kobber(I)klorid ble tilsatt porsjonsvis inntil blandingen antok en grønn farve. Fremstilling 5
(Fremstilling av forbindelser med formel (X) der R 3 er 6-klor).
Fremgangsmåte A: Inn i en oppløsning av 6-klor-l,4-naftokinon (193 g) i iseddik (1600 ml) ble det boblet klorgass inntil TLC av en alikvot viste at 6-klor-l,4-naftokinon var fullstendig forsvunnet. Det resulterende bunnfallet ble samlet opp ved filtrering og vasket med eddiksyre (200 ml) og heksan (2 x 300 ml) og lufttørret, og det ga 2,3,6-triklor-2,3-dihydrogen-l,4-naftokinon (157 g). Det faste stoffet ble overført til en kolbe utstyrt med en mekanisk rører og tilbake-løpskjøler. Natriumacetat (98,4 g) og eddiksyre (1,5 1) ble tilsatt, og inn i suspensjonen ble det boblet klorgass. Blandingen ble brakt til tilbakeløp og holdt der i 2 timer. Den av-kjølte blandingen ble helt i vann (3,5 1) og det resulterende bunnfall ble samlet ved filtrering og ble vasket med vann (2 x 500 ml), lufttørret og deretter vakuumtørret over fosfor-pentoksyd, og det ga 2,3,6-triklor-l,4-naftokinon (139 g), smeltepunkt 147,5-148,5°C.
Fremgangsmåte B: Inn i en suspensjon av 6-klor-l,4-naftokinon (100 g) i eddiksyre (800 ml) oppvarmet til 70°C, med mekanisk røring, ble det boblet klorgass. Oppvarmingen ble øket for å bringe blandingen til tilbakeløp, og ved det tidspunkt ble fast jod (13,2 g) tilsatt. Klortilsetning ved tilbakeløp fortsatte inntil TLC viste fullstendig overføring til produktet, 7-10 timer totalt. Reaksjonsblandingen ble avkjølt og ga et tykt bunnfall av 2,3,6-triklor-l,4-naftokinon som ble samlet ved filtrering. Ytterligere utbytte ble oppnådd ved konsentrering og/eller fortynning med vann. Totalt utbytte av tørret produkt var 121,2 g, smeltepunkt 147,5-148,5°C.

(Fremstilling av forbindelser med formel (VI) hvor R 3 er 6-klor)
Til en mekanisk rørt oppløsning av natriummetoksyd (55,5 g) i vannfri metanol (1,5 1) under et dekke av nitrogen, ble det tilsatt 2,3,6-triklorid-l,4-naftokinon fra Fremstilling 10
(130 g) så raskt som mulig. Temperaturen steg til 50°C i løpet av tilsetningen, og reaksjonsblandingen ble deretter varmet opp til tilbakeløp i 1 time. Blandingen ble avkjølt og gjort sur
med 6M saltsyre slik at den fikk en strålende gul farge. Etter tilsetningen av vann (300 ml), ble reaksjonsblandingen filtrert, og bunnfallet ble vasket med vandig metanol (4:1 vann-metanol) inntil filtratet var gulorange. Bunnfallet ble lufttørret og ga 6-klor-2,3-dimetoksy-l,4-naftokinon (102 g), smeltepunkt 125-126°C.
Fremstilling 7
Til en oppløsning av natrium-n-butoksyd (25,8 g) i tørr dimetylformamid (125 ml) ble det tilsatt 2,3,6-triklor-l,4-naftokinon (28 g) i én porsjon. Blandingen ble varmet med tilbakeløp i 2 timer, deretter avkjølt, surgjort med 6M saltsyre og dampet inn. Residuet ble kromatografert over silika-gel med bruk av diklormetan som vaskevæske, og det ga 2,3-di-n-butyloksy-6-klor-l,4-naftokinon (12,3 g) som et rødt, olje-aktig fast stoff.
Fremstilling 8
(Forbindelser med formel (VI) der R<3> er hydrogen)
Til en mekanisk rørt oppløsning av natriummetoksyd
(11,1 g) i vannfri metanol (200 ml) under et dekke av nitrogen, ble det tilsatt 2,3-diklor-l,4-naftokinon (22,7 g) så raskt som mulig. Temperaturen steg til 50°C i løpet av tilsetningen, og reaksjonsblandingen ble varmet til tilbakeløp i 1 time. Blandingen ble avkjølt og gjort sur med 6M saltsyre slik at den fikk en strålende gul farge. Etter tilsetning av vann (800 ml), ble reaksjonsblandingen filtrert, og bunnfallet ble vasket med vandig metanol (4:1 vann-metanol) inntil filtratet var gulorange. Bunnfallet ble lufttørret, og det ga 21,2 g av 2,3-dimetoksy-1,4-naftokinon, smeltepunkt 116-117°C.
Tilsvarende ved å bruke det rette natriumalkoksyd i stedet for natriummetoksyd, fremstilles de følgende forbindelser: 2,3-dietoksy-l,4-naftokinon;
2,3-di-n-propoksy-l,4-naftokinon.
Eksempel 1
A. En oppløsning av 6-klor-2,3-dimetoksy-l,4-naftokinon (50,5 g, 200 mmol) i tetrahydrofuran (500 ml) ble hydrogenert ved atmosfærisk trykk over palladium på trekull (10 %, 5,0 g) inntil oppløsningen var fargeløs, omtrent 4 timer.
B. Mens den fremdeles var under et dekke av hydrogen, ble en oppløsning av eddiksyreanhydrid (47 ml), pyridin (40 ml)
og DMAP (1,22 g) i tetrahydrofuran (50 ml) tilsatt til blandingen. Etter røring i 1 time, ble blandingen filtrert og dampet inn. Residuet ble løst opp i eter (500 ml) og ble vasket med IM HCl (3 x 2 50 ml) og med koksaltoppløsning (2 x 250 ml). Det organiske laget ble tørret over natrium-sulfat, filtrert og dampet inn til å gi en olje som krystalli-serte ved høyt vakuum. Omkrystallisering fra eter-petroleter ga 6-klor-l,4-diacetyloksy-2,3-dimetoksynaftalen, smeltepunkt 93-94°C.
Tilsvarende, ved å bruke fremgangsmåten ovenfor med bruk av den passende forbindelse med formel (VI) i stedet for 6-klor-2,3-dimetoksy-l,4-naftokinon der det måtte passe og det passende syreanhydrid i stedet for eddiksyreanhydrid, der det måtte passe, fremstilles de følgende forbindelser: 6,7-dimetyl-2,3-dimetoksy-l,4-di-acetyloksynaftalen, smeltepunkt 124-125°C:,
6,7-dimetyl-2,3-dimetoksy-l,4-di-n-propanoyloksynaftalen, smeltepunkt 108-109°C;
6,7-dimetyl-2,3-dimetoksy-l,4-di-n-butanoyloksynaftalen, smeltepunkt 69-70°C;
6,7-dimetyl-2,3-dimetoksy-l,4-di-i-butanoyloksynaftalen, smeltepunkt 98-99°c;
6,7-dimetyl-2,3-dimetoksy-l,4-di(2,2-dimetylpropanoyloksy)-naftalen, smeltepunkt 124-125°C;
6-klor-2,3-dimetoksy-l,4-dibenzoyloksynaftalen, smeltepunkt 161-162°C;
2,3-difenoksy-1,4-diacetyloksynaftalen, smeltepunkt 242-243°C;
2,3-difenoksy-1,4-di-n-propanoyloksynaftalen, smeltepunkt 164-165°C;
2,3-difenoksy-1,4-di-(2,2-dimetylpropanoyloksy)naftalen, smeltepunkt 218-219°C;
2,3-difenoksy-l,4-dibenzoyloksynaftalen, smeltepunkt 201-202°C;
6-klor-2,3-difenoksy-1,4-dibenzoyloksynaftalen, smeltepunkt 173-174°C;
6-klor-2,3-difenoksy-l,4-diacetyloksynaftalen, smeltepunkt 144-145°C;
6-klor-2,3-di(2-klorfenoksy)-1,4-dibenzoyloksynaftalen;
6-klor-2,3-difenyloksy-1,4-di-n-propanoyloksynaftalen, smeltepunkt 13 6-137°C;
6-klor-2,3-difenoksy-1,4-di-n-butanoyloksynaftalen, smeltepunkt 109-110°C;
6-klor-2,3-difenoksy-1,4-di(2,2-dimetylpropanoyloksy)-naftalen, smeltepunkt 164-165°C;
6-klor-2,3-difenoksy-l,4-di-i-butanoyloksynaftalen, smeltepunkt 116-117°C;

Materialer og fremgangsmåter
Swiss Webster hann- eller hunn-mus som veier 18-27 q ble virkårlig delt inn i behandlingsgrupper på 8 til 10 dyr, satt i bur sammen og gitt mat og vann etter ønske.
Testmaterialene ble fremstilt som oppløsninger eller suspensjoner i aceton av reagenskvalitet i et doseringsnivå
på 100 mg/ml, og arachidonsyre ble fremstilt som en oppløsning i aceton også i 100 mg/ml. Testmaterialene ble påført på det høyre øret til musene ved hjelp av en automatisk mikroliter-pipette slik at 10 vi av oppløsningen ble påført på hver av indre og ytre overflate. Hvert øre mottok derfor totalt 20 yl av oppløsningen og det inneholdt 2 mg testmateriale. En time etter påføringen av testmaterialet, ble arachidon-syreoppløs-ningen påført på samme måte. En time etter påføringen av
arachidon-syren ble dyrene avlivet ved cervikal dislokasjon og de høyre ørene ble fjernet. En 8 mm skive ble stanset ut fra hvert øre med en biopsi-stanse, og skivene ble veiet til nærmeste 0,1 mg. Med det formål å bestemme den antiinflamma-toriske virkning av testmaterialene, ble en negativ kontrollgruppe som bare fikk to påføringer av aceton, og en positiv kontrollgruppe som fikk bare aceton fulgt av arachidonsyre, tatt med i hvert eksperiment. Prosent hindring som skyldtes behandling med testmateriale.t, ble beregnet som følgende: % Hindring = sk- ivevekt ( pos . kontroll) - Skivevekt ( behandlet) x
